Abstract
Un polipéptido modificado genéticamente que comprende: una secuencia de polipéptido de dominio de unión a albúmina (ABD) que comprende la secuencia de aminoácidos expuesta en SEC ID Nº: 35, y una primera secuencia de dominio de hormona peptídica (HD1) que comprende la secuencia de aminoácidos expuesta en SEC ID Nº: 3.
Description
Se contemplan específicamente compuestos de las secuencias anteriores en los que está ausente cualquier metionina N terminal. La metionina N terminal puede estar presente principalmente como una conveniencia para expresión bacteriana. Sin embargo, los péptidos modificados genéticamente de la presente invención pueden expresarse en una célula hospedadora eucariota (por ejemplo levadura (por ejemplo Pichia), mamífero, baculovirus)
5 u otra célula hospedadora que tiene procesamiento proteolítico N terminal postraduccional para producir un aminoácido N terminal como se encuentra en un homólogo peptídico maduro de origen natural de la secuencia de hormona o ABD deseada, es decir sin la metionina añadida u otra secuencia líder. Como alternativa, una secuencia N terminal usada para expresión y/o secreción (e incluso purificación) puede ser una que pueda retirarse postraduccionalmente, por ejemplo mediante el uso de una proteasa tal como TEV.
10 Los compuestos polipeptídicos modificados genéticamente adicionales contemplados en el presente documento, que tienen una diversidad de componentes HD1, L1 y ABD, incluyen los compuestos que tienen la estructura de cualquiera de los polipéptidos modificados genéticamente de las tablas y los listados del presente documento, incluyendo los desvelados en la Tabla 3B a continuación.
