Abstract
Un método de identificación de enzimas [2Fe-2S] DHAD que comprende: a) consultar una o más secuencias de aminoácidos con un perfil modelo oculto de Markov preparado usando las proteínas de SEC ID Nº: 164, 168, 230, 232, 298, 310, 344 y 346, en el que una coincidencia con un valor de E inferior a 10-5 proporciona un primer subconjunto de secuencias, por el cual dicho primer subconjunto de secuencias se corresponde con una o más proteínas relacionadas con DHAD; b) analizar el primer subconjunto de secuencias que se corresponde con una o más proteínas relacionadas con DHAD de la etapa (a) para la presencia de tres cisteínas conservadas que se corresponden con las posiciones 56, 129 y 201 en la secuencia de aminoácidos de dihidroxi-ácido deshidratasa de Streptococcus mutans (SEC ID Nº: 168), por el cual se identifican un segundo subconjunto de secuencias que codifican enzimas [2Fe-2S] DHAD; y c) analizar el segundo subconjunto de secuencias de la etapa (b) para la presencia de aminoácidos conservados distintivos en las posiciones correspondientes a las posiciones en la secuencia de aminoácidos de DHAD de Streptococcus mutans (SEC ID Nº: 168) que son ácido aspártico en la posición 88, arginina o asparagina en la posición 142, asparagina en la posición 208 y leucina en la posición 454, por el cual se identifican adicionalmente un tercer subconjunto de secuencias que codifica enzimas [2Fe-2S] DHAD.
