Compuesto y composición para agentes activos de
administración.
La presente invención relaciona a un compuesto
para agentes activos de administración, y particularmente agentes
activos biológicamente o químicamente. Estos compuestos son usados
como transportadores para facilitar la administración de un
cargamento a un objetivo. El compuesto transportador es bien
apropiado para formar mezclas no-covalentes con
agentes activos-biológicamente para administración
oral en animales.
Los medios convencionales para los agentes
activos de administración están siempre limitados severamente por
barreras biológicas, químicas y físicas. Típicamente, estas barreras
están impuestas por el ambiente en el cual ocurre el
administración, el ambiente del objetivo para la administración, o
el objetivo en si mismo. Los agentes activos biológicamente o
químicamente son particularmente vulnerables a tales barreras.
Por ejemplo, en los animales la administración
de agentes farmacológicos y terapéuticos biológicamente activos o
químicamente activos, las barreras son impuestas por el cuerpo.
Ejemplos de barreras físicas son la piel y varias membranas de los
órganos que deben ser atravesadas antes de alcanzar el objetivo. Las
barreras químicas incluyen, pero no están limitadas a, variaciones
del pH, bicapas de lípidos, y enzimas de degradación.
Estas barreras son de particular significado en
el diseño de sistemas de administración oral. La administración
oral de muchos agentes activos biológicamente o químicamente debería
ser la ruta elegida para la administración a animales si no fuera
por las barreras biológicas, químicas y físicas tal como variación
del pH en el tracto gastro-intestinal (GI), enzimas
digestivas poderosas, y membranas
gastro-intestinales impermeables al agente activo.
Entre los numerosos agentes que no son típicamente receptivos a la
administración oral están los péptidos activos biológicamente o
químicamente, tal como calcitonina e insulina; polisacáridos, y en
particular mucopolisacaridos incluyendo, pero no limitados a,
heparina; heparinoides; antibióticos; y otras sustancias orgánicas.
Estos agentes son rápidamente dejados inefectivos o son destruidos
en el tracto gastro-intestinal mediante hidrólisis
ácida, enzimas, o similares.
Los primeros métodos para la administración oral
de agentes farmacológicos vulnerables han dependido de la
co-administración de adyuvantes (e.g. resorcinoles y
surfactantes no iónicos tal como polioxietilen oleil éter y
n-hexadecilpolietilen éter) para aumentar
superficialmente la permeabilidad de las paredes intestinales, como
también la co-administración de inhibidores
enzimáticos (e.g. inhibidores de la tripsina pancreática,
diisopropilfluorofosfato (DFF) y trasilol) para inhibir la
degradación enzimatica.
Los liposomas también han sido descritos como
sistemas de administración de drogas para la insulina y la heparina.

(1979), Endocrinology Japan, Vol. 26, pg. 337.
Sin embargo, el uso de un espectro amplio de
tales sistemas de administración de drogas es evitado por: (1) los
sistemas requieren cantidades tóxicas de adyuvantes o inhibidores;
(2) los cargamentos de peso molecular bajo, i.e. agentes activos,
no están disponibles; (3) los sistemas exhiben una estabilidad pobre
y un tiempo de vida útil inadecuado; (4) los sistemas son difíciles
de fabricar; (5) los sistemas fallan en la protección del agente
activo (cargamento); (6) los sistemas adversamente alteran el agente
activo; o (7) los sistemas fallan en la promoción de la absorción
del agente activo.
Más recientemente, se han usado microesferas de
polímeros artificiales de aminoácidos mezclados (proteinoides) para
transportar los fármacos. Por ejemplo, la patente US 4,925,673
describe transportadores de microesfera de proteinoide que
contienen droga como también métodos para su preparación y uso.
Estas microesferas de proteinoide son útiles para la administración
de un número de agentes activos.
WO 96/12473 describe compuestos y composiciones
para agentes de administración.
Aun existe una necesidad en el arte de agentes
de administración simples y baratos. Por lo tanto la presente
invención provee el compuesto de la reivindicación 10 y la
composición farmacéutica de la reivindicación 1. Las modalidades
preferidas en las reivindicaciones 2 a 9 y 11 a 13.

Las composiciones que comprenden el
transportador como se definen en la reivindicación, y los agentes
activos son efectivos en la administración de agentes activos para
los sistemas biológicos seleccionados.
Las composiciones específicas de la presente
invención incluyen un agente activo y un transportador. Estas
composiciones se pueden usar para administrar varios agentes activos
a través de varias barreras biológicas, químicas, y físicas y son
particularmente apropiadas para la administración de agentes activos
que están sujetos a degradación ambiental. Las composiciones
materia de esta invención son particularmente útiles para la
administración o suministro de agentes activos biológicamente o
químicamente a cualquiera de los animales tales como aves

y particularmente humanos; e insectos.
Otras ventajas de la presente invención incluyen

La coadministración subcutánea, sublingual e
intranasal de un agente activo, tal como, por ejemplo, hormona de
crecimiento humano recombinante (rhGH); calcitonina de salmón;
heparina, incluyendo, pero no limitado a, heparina de bajo peso
molecular; hormona paratiroides; y compuestos en composiciones como
se describen aquí resultan en un aumento de la biodisponibilidad
del agente activo comparado a la administración del agente activo
solo.
Los agentes activos apropiados para uso en la

péptidos, y particularmente péptidos pequeños; hormonas, y
particularmente hormonas que por si mismas no pasan o solamente una
fracción de la dosis administrada pasa a través de la mucosa
gastro-intestinal y/o son susceptibles a ruptura

Este compuesto de transporte puede ser usado
para administrar agentes activos incluyendo, pero no limitado a,
agentes activos biológicamente o químicamente tal como por ejemplo,
agentes farmacológicos y terapéuticos.
También pueden ser usadas sales tales como, por
ejemplo, sales de sodio de estos compuestos de transporte.
El compuesto de la presente invención puede ser
fácilmente preparado de aminoácidos.
Por ejemplo, el compuesto puede ser preparado
mediante la reacción del ácido simple con el agente apropiado que
reacciona con un intermedio amino libre presente en los aminoácidos
para formar amidas. Los grupos de protección se pueden usar para
evitar reacciones laterales no deseadas como deberán ser conocidas
por aquellos expertos en el arte.
El compuesto de transporte puede ser purificado
por recristalización o por fraccionamiento en soportes de columna
sólida. Sistemas solventes de recristalización apropiados incluyen
acetonitrilo, metanol y tetrahidrofurano. La fraccionamiento se
puede realizar sobre soportes de columna sólida apropiados tales
como alúmina, usando mezclas metanol/n-propanol
como fase móvil; soportes de columna de fase reversa usando mezclas
de ácido trifluoroacetico/acetonitrilo como fase móvil;
cromatografía de intercambio iónico usando agua como fase móvil. En
donde se realiza la cromatografía de intercambio aniónico, se
emplea preferiblemente un gradiente de cloruro de sodio
0-500 mM.

Las composiciones de la presente invención
pueden incluir uno o más agentes activos.
En una modalidad, el compuesto 67 o sus sales se
pueden usar directamente como un admistrador simplemente mezclando
el compuesto o sal, con el agente activo antes de la
administración.
Las mezclas de administración son preparadas
mezclando una solución acuosa del transportador con una solución
acuosa del ingrediente activo, justo antes de la administración.
Alternativamente, el transportador y el ingrediente activo
biológicamente o químicamente puede ser mezclado durante el proceso
de fabricación. Las soluciones pueden opcionalmente contener
aditivos tales como sales buffer de fosfato, ácido cítrico, ácido
acético, gelatina, y goma acacia.
Se pueden incorporar aditivos estabilizantes en
la solución de transporte. Con algunas drogas, la presencia de
tales aditivos promueve la estabilidad y dispersabilidad del agente
en la solución.
Los aditivos estabilizantes se pueden emplear a
un rango de concentración entre cerca de 0.1 y 5% (w/v),
preferiblemente cerca de 0.5% (w/v). Ejemplos apropiados, pero no
limitantes, de aditivos estabilizantes incluyen goma acacia,
gelatina, metil celulosa, polietilen glicol, ácidos carboxílicos y
sales de ellos, y polilisina. Los aditivos estabilizantes
preferidos son goma acacia, gelatina y metil celulosa.
La cantidad de agente activo es una cantidad
efectiva para lograr el propósito del agente activo en particular.
La cantidad en la composición típicamente es una cantidad efectiva
farmacológicamente, biológicamente, terapéuticamente, o
químicamente. Sin embargo, la cantidad puede ser menor que una
cantidad efectiva farmacológicamente, biológicamente,
terapéuticamente, o químicamente cuando la composición es usada en
forma de dosis unitaria, tal como una cápsula, una tableta o un
líquido, porque la forma de dosis unitaria puede contener un
múltiplo de las composiciones del transportador/agente activo
biológicamente o químicamente o puede contener una cantidad
divisoria efectiva farmacológicamente, biológicamente,
terapéuticamente, o químicamente. Las cantidades totales efectivas
pueden entonces ser administradas en unidades acumulativas que
contienen, en total, las cantidades activas farmacológicamente,
biológicamente, terapéuticamente, o químicamente del agente activo
biológicamente o farmacológicamente.
La cantidad total de agente activo, y
particularmente del agente activo biológicamente o químicamente, a
ser usada puede ser determinada por aquellos expertos en el arte.
Sin embargo, sorprendentemente se ha encontrado que con algunos
agentes activos biológicamente o químicamente, el uso de los
transportadores presentes descritos proveen una administración
extremadamente eficiente, particularmente en sistemas orales,
intranasales, sublinguales, intraduodenales, réctales, vaginales,
bucales, oftálmicos, o subcutáneos como también sistemas para
atravesar la barrera sangre/cerebro. Por lo tanto, se pueden
administrar cantidades menores del agente activo biológicamente o
químicamente a aquellas usadas en las formas de dosis unitaria o los
sistemas de administración al sujeto, logrando los mismos niveles
en la sangre y efectos terapéuticos.
La cantidad de transportador en la presente
composición es una cantidad para administración efectiva y se puede
determinar para cualquier transportador o agente activo
biológicamente o químicamente en particular por los métodos
conocidos por aquellos expertos en el arte.
Las formas de dosis unitaria pueden incluir
también cualquier tipo de excipientes; diluyentes; desintegradores;
lubricantes; plastificantes; colorantes; y vehículos de
dosificación, incluyendo, pero no limitado al agua,
1,2-propano diol, etanol, aceite de oliva, o
cualquier combinación de ellos.
La administración de las presentes composiciones
o de las formas de dosis unitarias preferiblemente es oral o por
inyección intraduodenal.
Las composiciones de administración de la
presente invención pueden también incluir uno o más inhibidores de
enzimas. Tales inhibidores de enzima incluyen, pero no están
limitados a, compuestos tales como actinonina o epiactinonina y
derivados de ellos. Estos compuestos tienen las formulas de

Otros inhibidores de enzimas incluyen, pero no están limitados a,
aprotinina (Trasylol) e inhibidor de
Bowman-Birk.
El compuesto y las composiciones del sujeto de
la invención son útiles para la administración de agentes activos
biológicamente o químicamente a cualquier tipo de animales tales
como aves; mamíferos, tal como primates y particularmente humanos;
e insectos. El sistema es particularmente ventajoso para la
administración de agentes activos biológicamente o químicamente que
podrían ser de otra forma destruidos o dejados menos efectivos por
las condiciones encontradas antes de que el agente activo esté en la
zona objetivo (i.e. el área en la que el agente activo o la
composición de la administración será liberada) y dentro del cuerpo
del animal al que ellos serán administrados. Particularmente, los
compuestos y composiciones de la presente invención son útiles en
la administración oral de agentes activos, especialmente aquellos
que ordinariamente no son deseables por vía oral.
Ejemplo
1
Preparaciones generales de los
transportadores. Los siguientes procedimientos fueron usados
para preparar los compuestos de referencia descritos aquí. Muchos
de los compuestos fueron preparados mediante la reacción del amino
ácido apropiado con el ácido clorhídrico apropiado. La preparación
del compuesto 79 se da como un ejemplo representativo de los
compuestos preparados de esta manera.
Preparación del compuesto 79. Método A.
Un matraz de base redonda con un agitador magnético fue cargado con
ácido 3-(4-aminofenil)propiónico (46.3 g,
0.28 moles, 1.17 equiv.) y una solución acuosa 2 M de hidróxido de
sodio (300 ml). El 2,3-dimetoxibenzoilcloruro (48.0
g, 0.24 moles, 1.00 equiv.) fue adicionado a porciones a través de
1 hora a la solución en agitación. Después de la adición, la
reacción fue agitada por 2.5 h a temperatura ambiente, y el pH de
la solución fue mantenido a arriba de 10 mediante la adición de NaOH
10 M. La solución fue luego acidificada con HCl 1 M (3 x 100 ml),
agua (100 ml), y aire seco. Fue redisuelto en acetona hirviendo (ca
500 ml), decolorado con carbón activado (3 g), y filtrado. Fue
adicionada agua (1.5 L) al filtrado para inducir la formación de un
aceite marrón. El aceite marrón solidificado fue agitado a
temperatura ambiente por 10 min. El sólido crudo fue colectado
mediante filtración y recristalizado de metanol-agua
(v/v) para obtener el compuesto 79 como un sólido bronceado (39.5
g, 50%).
Preparación del compuesto 79. Método B.
Un matraz redondo de 3 cuellos con un agitador magnético y 2 embudos
de adición bajo una atmósfera de argón. Fue adicionada en el matraz
una suspensión de ácido
3-(4-aminofenil)propiónico (46.3 g, 0.28
moles, 1.17 equiv.) en acetato de etilo (700 ml). Fue cargada una
solución de 2.3-dimetoxibenzoilcloruro (48.0 g,
0.24 moles, 1.00 equiv.) en acetato de etilo a uno de los embudos de
adición y adicionado a gotas durante una hora. La trietilamina
(28.20 g, 0.28 moles, 1.00 equiv.) fue posteriormente cargada por el
segundo embudo y adicionada a gotas durante 15 minutos. La reacción
fue agitada a temperatura ambiente por 3 horas, y el solvente fue
evaporado al vacío dando un aceite marrón residual. Fue adicionada
agua (600 ml) al residuo seguido por una solución de NaOH (2 M, 500
ml), y la mezcla fue agitada a temperatura ambiente por 3 horas. La
solución marrón resultante fue acidificada con HCl 2 M (ca 1 L).
Después de enfriada la mezcla en un baño de hielo por 1 hora,
formándose un sólido amarillo que fue colectado por filtración. El
sólido fue lavado con agua (3 x 1.5 L) y recristalizado de
etanol-agua (v/v) para dar el compuesto 79 como un
sólido bronceado (59.2 g, 68%).
Preparación del compuesto 79. Método C.
Un matraz de base redonda de 2 L equipado con un agitador magnético
y un condensador de reflujo fue cargado con una suspensión de ácido
3-(4-aminofenil)propiónico (46.3 g, 0.28
moles, 1.17 equiv.) en diclorometano (560 ml). Fue adicionado
Clorotrimetilsilano (62.36 g, 0.57 moles, 2.05 equiv.) en una
porción, y la mezcla fue calentada a reflujo durante 1 hora bajo
argón. Se permitió el enfriamiento de la reacción a temperatura
ambiente y fue colocada en un baño de hielo (temperatura interna
< 10ºC). El condensador de reflujo fue reemplazado con un embudo
de adición con trietilamina (42.50 g, 0.42 moles, 1.50 equiv.). La
trietilamina fue adicionada a gotas durante 15 minutos, y se formó
un sólido amarillo durante la adición. El embudo fue reemplazado
por otro embudo de adición que contenía una solución de
2,3-dimetoxibenzoilcloruro (48.0 g, 0.24 moles,
1.00 equiv.) en diclorometano (100 ml). La solución fue adicionada
gota a gota durante 30 min. La reacción fue agitada en un baño de
hielo por otros 30 minutos y a temperatura ambiente por 1 hora. El
diclorometano fue evaporado al vacío para dar un aceite marrón., El
aceite marrón fue enfriado en un baño de hielo y fue adicionada una
solución enfriada en hielo de NaOH 2 M (700 ml). El baño de hielo
fue removido, y la reacción fue agitada por 2 h para lograr una
solución marrón clara. La solución fue acidificada con ácido
sulfúrico 2 M (400 ml) y guardada arriba de (ca) 5ºC por 1 hora. Se
formó un sólido amarillo que fue colectado por filtración. El
sólido fue lavado con agua (3 x 100 ml) y recristalizado de 50%
etanol-agua (v/v) para obtener el compuesto 79 como
agujas bronceadas (64.7 g, 82%).
El compuesto 67, fue también preparado por este
proceso.
Preparación del compuesto 35. Una
solución de O-acetilsaliciloil cloruro (24.68 g, 124
mmol, 1 equiv.) fue enfriado en tetrahidrofurano (300 ml) en un
baño de hielo. Fue adicionada a gotas trietilamina (25 g, 249 mmol,
2 equiv.) mediante un embudo adicional.
El metil 9-aminononanoato
hidrocloruro fue disuelto en DMF (190 ml, ligeramente tibio para
disolver), cargado a un embudo de adición y adicionado a gotas por
encima de la mezcla. La reacción fue agitada en el baño de hielo
por 20 minutos a temperatura ambiente por 2 horas. La evaporación
del THF bajo presión reducida da una solución DMF rosada. La
solución rosada fue enfriada en un baño de hielo, y fue adicionada
una solución 2 M de NaOH (300 ml). Después de ser agitado a
temperatura ambiente por 12 horas, la mezcla fue acidificada con
HCl 2 M (500 ml). La solución fue enfriada en un baño de hielo, y
se formó un sólido. El sólido fue colectado por filtración y fue
recristalizado de 50% etanol-agua para dar el
compuesto 35 (32 g, 87%) como un sólido de un color crudo.
Preparación del compuesto 49. Se colocó
1-(2-hidroxifenil)-3-(4-metil
benzoato)-1,3-propano diona (3.00 g,
0.0101 mil.) en un matraz de base redonda colocado con purga de
argón, una barra de agitación magnética y un condensador de agua
fría. Fueron adicionados ácido acético glacial (20 mls) y ácido
sulfurico concentrado (5 mls), y el calentamiento de la mezcla de
reacción fue iniciado. Se permitió el calentamiento a reflujo por 6
horas antes de que el calentamiento fuera suspendido. Se permitió a
la mezcla de reacción alcanzar la temperatura ambiente, y luego fue
puesta dentro de 100 mls de hielo/agua. Esto fue agitado por
aproximadamente ½ hora antes de que la mezcla fuera filtrada, y fue
aislado un sólido marrón. El sólido marrón fue recristalizado 2
veces de ácido acético, produciendo el compuesto 49 como un sólido
bronceado (1.44 g, 53.8%).
Preparación del compuesto 167. Fue
disuelta 2-cumarona (4.21 g, 0.0314 mol) con
agitación magnética, en acetonitrílo (75 mls) en un matraz de base
redonda de 250 ml, purgado con argón y condensador de agua fría.
Fueron adicionados Trietanolamina (3.18 g, 0.0314 mol) y ácido
8-aminocaprílico (5.00 g, 0.0314 mol) y se formó una
pasta bronceada. Se comenzó el calentamiento, y se dejó toda la
noche la mezcla de reacción en reflujo. Después del calentamiento
en la noche, una cromatografía en capa delgada de la mezcla de
reacción (50% acetato de etilo/50% hexano) indicó que la reacción
se completó. Se paró el calentamiento, se permitió el enfriamiento
de la mezcla de reacción a temperatura ambiente, y fue concentrada
al vacío. El residuo resultante fue extraído con cloruro de
metileno, y fue lavado con 2, porciones de 100 ml de HCl 1N. La capa
de cloruro de metileno fue secada con sulfato de sodio y fue
concentrada al vacío. El sólido bronceado resultante fue secado al
vacío durante toda la noche, obteniendo el compuesto 167 como un
sólido bronceado (8.35 g, 70.4%).
Preparación del compuesto 171. Fue
disuelta
1,4-benzodioxano-2-ona
(3.93 g, 0.0262 mol), con agitación magnética, en un matraz de
fondo redondo de 250 ml y con un condensador de agua fría y con
acetonitrílo (70 mls) y purgado con argón. Fueron adicionados
Trietilamina (2.64 g, 0.0262 mol) y ácido
8-aminocaprílico (500 g, 0.0262 mol) y se formó una
pasta bronceada. Se comenzó el calentamiento, y se permitió el
reflujo de la mezcla de reacción por aproximadamente 3 horas. En
este momento, una cromatografía de capa delgada de la mezcla de
reacción (50% acetato de etilo/50% hexano) indicó que la reacción
se había completado. El calentamiento fue suspendido, y se permitió
el enfriamiento a temperatura ambiente y se concentró al vacío. El
residuo resultante se extrajo con cloruro de metileno y se lavó con
una porción de 100 ml de HCl 1N. En este momento, se notó la
precipitación de un sólido bronceado, el cual fue aislado por
filtración. Este sólido bronceado fue después lavado con una
porción adicional de 100 ml de HCl 1N, y luego con 100 ml de agua.
El sólido bronceado resultante se secó al vacío durante la noche
obteniéndose el compuesto 171 (7.73 g, 95.6%).
Preparación del compuesto 120. Una
solución de 3.00 g (18.3 mmol) de
2-nitrofenilisocianato y 5 ml de tetrahidrofurano
fueron adicionados gota a gota durante 10 minutos a una solución
enfriada en un baño de hielo de 2.08 g (13.1 mmol) de ácido
aminocaprílico, 1.40 ml de NaOH 10N y 40 ml de agua. La mezcla de
reacción fue agitada por 30 minutos adicionales, calentada a 25°C y
tratada con HCl al 3% hasta obtener un pH de 5. El precipitado
amarillo fue filtrado y lavado con 100 ml de agua. El sólido
amarillo fue recristalizado en 2-propanol y agua
para dar 3.7 g del compuesto 120 en forma de cristales amarillos
pálidos.
Preparación del compuesto 133. Se calentó
a 140°C una suspensión de 2.40 g (16.3 mmol) y 2.80 g (15.6 mmol) y
ácido 4-(4aminofenil)butírico en 20 ml de propilen glicol,
2.40 ml (1.74 g, 17.3 mmol) de trietilamina y 10 mg (0.08 mmol) de
dimetilaminopiridina. La mezcla se comienza a aclarar después de 5
minutos a 140°C. Después del agitado por 330 minutos, la mezcla de
reacción fue enfriada a 25°C y diluida con 20 ml de agua. La
ftalamida sólida que se formó fue filtrada. El filtrado fue
acidificado con HCl al 3%. El sólido resultante fue filtrado y
recristalizado de 2-propanol y agua para dar 0.62 g
del compuesto 133 como un sólido bronceado.
Preparación del compuesto 138. Una
solución de 1.73 g (12.9 mmol) de dialdehído ftalico, 2.04 g de
ácido 8-aminocaprílico y 20 ml de ácido acético fue
calentada al reflujo por 10 minutos. La mezcla de reacción fue
enfriada a 40C, diluida con agua y extraída con CH_{2}Cl_{2} (2
X 20 ml). La fase orgánica fue lavada con agua y salmuera, secada
sobre Na_{2}SO_{4} y evaporada. El residuo fue disuelto en éter
y extraído con NaOH 2N. Las capas fueron separadas. La capa acuosa
fue acidificada con HCl al 3% y extraída con CH_{2}Cl_{2}. La
fase orgánica fue secada sobre Na_{2}SO_{4} y evaporada. El
residuo amarillo fue recristalizado de acetonitrilo y agua para dar
1.25 g del compuesto 138 como un sólido amarillo.
Preparación del compuesto 140. Una mezcla
de 1.40 g (9.48 mmol) de anhídrido ftalico y 1.51 g (9.48 mmol) de
ácido 8-aminocaprílico fue calentada a 150°C por 5
minutos. Después del enfriamiento, se obtuvieron 2.61 g del
compuesto 140 sólido.
Preparación del compuesto 145. Una
suspensión de 2.11 g (10.1 mmol) de ácido etil carbamoilantranílico
y 5 ml de CH_{2}Cl_{2} fue tratada con 2.20 ml de oxalil
cloruro. Después de agitado por 1 hora los volátiles fueron
descartados. Al mismo tiempo, una suspensión de 1.60 g (10.1 mmol)
de ácido 8-aminocaprílico y 15 ml de
CH_{2}Cl_{2} fue tratado con 2.60 ml (2.23 g, 20.5 mmol) de
TMSCl. Esta mezcla fue calentada a reflujo por 90 minutos, enfriada
en un baño de hielo y tratada con 4.30 ml (3.12 g, 30.9 mmol) de
trietilamina. Cinco minutos mas tarde, una pasta del residuo de la
reacción del oxalil cloruro en 20 ml de CH_{2}Cl_{2} fue
adicionado. La mezcla de reacción fue calentada a 25°C y agitada
durante la noche. Luego de la acidificación de la mezcla con HCl al
3% se formó un sólido blanco. El sólido fue filtrado y
recristalizado de EtOH y agua para dar 1.88 g del compuesto
145.
Preparación del compuesto 154. Una
suspensión de 4.02 g (25.6 mmol) de ácido
trans-4-aminometilciclohexanocarboxílico,
4.18 g (25.6 mmol) de anhídrido isatóico, 20 ml de
CH_{2}Cl_{2}, 20 ml de dioxano, y 4 ml de agua fue calentada a
reflujo por 12 horas. La solución fue enfriada a 25°C y extraída con
éter (4 x 20 ml). La capa orgánica fue secada sobre
Na_{2}SO_{4} y concentrada. El sólido resultante fue
recristalizado de EtOH y agua para dar 4.95 g del compuesto
154.

Ratas macho Sprague-Dawley que
pesaban entre 200 - 250 g fueron puestas en ayunas por 24 horas y
fueron administradas con ketamina (44 mg/Kg) y clorpromazina (1.5
mg/Kg) 15 minutos antes de la dosificación. Las ratas fueron
administradas con una de las soluciones de dosificación del ejemplo
4 ya sea por administración oral con sonda o por instilación
intracolónica. Las muestras de sangre fueron colectadas serialmente
de la arteria del rabo para la determinación de las concentraciones
del rHGH en el suero. Las concentraciones del rHGH en el suero
fueron cuantificadas mediante un kit de prueba de inmunoensayo.

Claims (13)
1. Una composición farmacéutica o una forma de
dosis unitaria que comprende la composición farmacéutica que
comprende:
- A.
- Al menos un agente activo; y
- B.
- Un compuesto que tiene la siguiente fórmula o una sal de él
2. La composición farmacéutica o forma de dosis
unitaria de la reivindicación 1, en donde el agente activo es
seleccionado del grupo que consiste de agentes activos
biológicamente, y cualquier combinación de ellos.
3. La composición farmacéutica o forma de dosis
unitaria de la reivindicación 2, en donde el agente activo
biológicamente comprende al menos un péptido, mucopolisacárido,
carbohidrato o lípido.
4. La composición farmacéutica o forma de dosis
unitaria de la reivindicación 2, en donde el agente activo
biológicamente es seleccionado del grupo que consiste de hormonas de
crecimiento humano, hormonas de crecimiento bovino, hormonas de
liberación de crecimiento, interferones,
interleuquina-1, interleuquina-II,
insulina, heparina, heparina de bajo peso molecular, calcitonina,
eritropoietina, factor naturetico atrial, antígenos, anticuerpos
monoclonales, somatostatina, adreno-corticotropina,
hormona de liberación de gonadotropina, oxitocina, vasopresina,
cromolin sodio, vancomicina, desferrioxamina, hormona paratiroide, y
cualquier combinación de ellos.
5. La composición farmacéutica o forma de dosis
unitaria de la reivindicación 4, en donde el agente activo
biológicamente comprende interferón,
interleuquina-1, interleuquina II, insulina,
heparina, heparina de bajo peso molecular, calcitonina, oxitocina,
vasopresina, vancomicina, desferrioxamina, hormona paratiroide, o
cualquier combinación de ellos.
6. Una forma de dosis unitaria que
comprende
- A.
- La composición farmacéutica de cualquiera de las reivindicaciones 1 a 5; y
- B.
- a)
- un excipiente,
- b)
- un diluyente
- c)
- un desintegrante
- d)
- un lubricante
- e)
- un plastificante
- f)
- un colorante
- g)
- un vehículo de dosis, o
- h)
- cualquier combinación de ellos.
7. La forma de dosis unitaria de la
reivindicación 6, que comprende una tableta, una cápsula o un
líquido.
8. La forma de dosis unitaria de cualquiera de
las reivindicaciones 6 y 7, en donde dicho vehículo de dosificación
es seleccionado del grupo que consiste de agua,
1,2-propano diol, etanol, aceite de oliva y
cualquier combinación de ellos.
9. La composición farmacéutica o forma de dosis
unitaria como es definida en las reivindicaciones 1 a 5 para ser
administrada oralmente, intranasalmente, sublingualmente,
intraduodenalmente, subcutáneamente, rectalmente, vaginalmente,
bucalmente u oftalmológicamente.
10. Un compuesto que tiene la fórmula
o una sal de
el.
11. Un método para la preparación de una
composición, dicho método comprende mezclado de:
- A.
- al menos un agente activo;
- B.
- el compuesto de la reivindicación 10; y
- C.
- opcionalmente, un vehículo de dosificación.
12. La composición farmacéutica de cualquiera de
las reivindicaciones 1 a 5 capaz de pasar un agente activo
biológicamente a través de la barrera sangre/cerebro de un
animal.
13. La composición farmacéutica de acuerdo a la
reivindicación 1, en donde al menos un agente activo es hormona de
crecimiento humano.
