Abstract
Una cepa de levadura transformada para reducir la represión por catabolito nitrogenado de una actividad enzimática degradadora de urea expresada por la cepa de levadura bajo condiciones de fermentación.
Claims (20)
1. Una cepa de levadura transformada para reducir
la represión por catabolito nitrogenado de una actividad enzimática
degradadora de urea expresada por la cepa de levadura bajo
condiciones de fermentación.
2. La cepa de levadura de la reivindicación 1, en
donde la actividad enzimática degradadora de urea es una actividad
urea carboxilasa o alofanato hidrolasa.
3. La cepa de levadura de la reivindicación 1 ó
2, en donde la cepa es transformada con un ácido nucléico
recombinante que comprende una secuencia codificante que codifica
para la actividad enzimática degradadora de urea.
4. La cepa de levadura de la reivindicación 1 ó
2, en donde la cepa es transformada con un ácido nucléico
recombinante que comprende un promotor adaptado para regular la
expresión de la actividad enzimática degradadora de urea bajo
condiciones de fermentación.
5. La cepa de levadura de la reivindicación 3, en
donde la secuencia codificante comprende un marco abierto de lectura
homólogo al marco de lectura abierto de DUR1,2.
6. La cepa de levadura de cualquiera de las
reivindicaciones 1 a 5, en donde bajo las condiciones de
fermentación la cepa de levadura fermenta un carbohidrato para
producir alcohol etílico.
7. La cepa de levadura de cualquiera de la
reivindicaciones 1 a 6, en donde la cepa de levadura es una cepa de
S. cerevisiae.
8. La cepa de levadura de cualquiera de las
reivindicaciones 1 a 7, en donde bajo las condiciones de
fermentación la cepa de levadura produce un vino que posee una
concentración de carbamato de etilo de menos de 30 ppb.
9. Una cepa de levadura substancialmente tal como
la que aquí se describe y con referencia a los ejemplos y
dibujos.
10. El uso de la cepa de levadura de cualquiera
de las reivindicaciones 1 a 9 para producir una bebida alcohólica
fermentada.
11. Un método de fabricar una bebida alcohólica
fermentada que comprende mantener la cepa de levadura de cualquiera
de las reivindicaciones 1 a 9 bajo condiciones de fermentación.
12. La bebida alcohólica fermentada de la
reivindicación 10 o de la reivindicación 11.
13. La bebida alcohólica fermentada de la
reivindicación 12, en donde la bebida es un vino o una bebida
alcohólica destilada.
14. La bebida alcohólica fermentada de la
reivindicación 13, en donde la bebida es vino, y el vino tiene una
concentración de carbamato de etilo de menos de 30 ppb.
15. El uso de la cepa de levadura de cualquiera
de las reivindicaciones 1 a 9 para producir un producto
fermentado.
16. Un método de fabricar un producto fermentado
que comprende mantener la cepa de levadura de cualquiera de las
reivindicaciones 1 a 9 bajo condiciones de fermentación.
17. El producto fermentado de la reivindicación
15 o de la reivindicación 16.
18. Un método de modificar una cepa de levadura,
que comprende transformar la cepa de levadura para reducir la
represión por catabolito nitrogenado de una actividad enzimática
degradadora de urea expresada por la cepa de levadura bajo las
condiciones de fermentación.
19. Un ácido nucléico aislado que comprende una
secuencia promotora que es 95% idéntica cuando es óptimamente
alineada a una secuencia promotora DUR1,2 nativa, en donde la
secuencia promotora no regula la represión por catabolito
nitrogenado de una secuencia codificante operativamente unida bajo
condiciones de fermentación en una célula hospedadora.
20. Una secuencia de ácido nucléico aislada que
comprende una secuencia codificante que es 80% idéntica cuando es
óptimamente alineada a una secuencia codificante de DUR1,2 nativa,
en donde la secuencia codificante está operativamente unida a una
secuencia promotora que no regula la represión por catabolito
nitrogenado de la secuencia codificante bajo condiciones de
fermentación en una célula hospedadora.
