Abstract

Un procedimiento enzimático para preparar ácido 7 b -(carboxibutanamido) cefalosporánico utilizando la enzima D-aminoácido oxidasa de trigonopsis variabilis modificada producida en Escherichia coli. El procedimiento de expresión de la enzima comprende: para la enzima D-aminoácido oxidasa; (II) eliminar el intrón que contiene dicho gen; (III) insertar el fragmento de ADN obtenido en un plásmido que sea capaz de replicarse en Escherichia coli (IV) fusionar en el extremo 5"'' de la región estructural del gen un ensamblador sintético que contiene una secuencia de nucleótidos que codifica para seis histidinas; (V) transformar una cepa de Escherichia coli con el plásmido recombinante resultante; (VI) cultivar las células de Escherichia coli transformadas y (VII) recuperar la enzima D-aminoácido oxidasa del cultivo de la operación anterior mediante cromatografía de afinidad.
