Abstract
Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung von Verbindungen der allgemeinen Formel (I) in der R1 und R2 wie in der Beschreibung erwähnt definiert sind, deren pharmazeutisch verträglichen Salze und den Solvaten davon, die ausgehend von Verbindungen der allgemeinen Formel (II) in der R1 wie in der Beschreibung erwähnt definiert ist, hergestellt werden können.

NEUES HERSTELLVERFAHREN
Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung von Verbindungen der allgemeinen Formel I
in der R1 und R2 wie in Anspruch 1 definiert sind, deren pharmazeutisch verträglichen Salze und den Solvaten davon.
HINTERGRUND DER ERFINDUNG
TECHNISCHES GEBIET
Die vorliegende Erfindung betrifft ein technisches Verfahren zur Herstellung von Verbindungen der allgemeinen Formel I, welche CGRP-antagonistische Eigenschaften besitzen. Weiterhin betrifft die Erfindung die Verbindungen der allgemeinen Formeln V und VI per se, da diese in besonderer Weise zur Herstellung der Verbindungen der allgemeinen Formel I geeignet sind.
DETAILLIERTE BESCHREIBUNG DER ERFINDUNG
Die Verbindungen der allgemeinen Formeln V und VI stellen wertvolle Ausgangsstoffe zur Synthese der Verbindungen der allgemeinen Formel I dar, welche CGRP-antagonistische Eigenschaften besitzen.
Die isolierten Zwischenstufen fallen als kristalline Feststoffe an, was für die Aufreinigung sowie die Trennung eventuell anfallender Enantiomerengemische von großem Vorteil ist.
Ein erster Gegenstand der vorliegenden Erfindung betrifft ein Verfahren zur Herstellung von Verbindungen der allgemeinen Formel I
in der
R1 H, d-s-Alkyl, C(O)-O-Benzyl, C(O)-O-te/f-Butyl oder Benzyl, vorzugsweise H oder Benzyl, darstellt und
R2 ein sekundäres Amin -NR2 1R22 bedeutet, wobei
R2 1 und R22 unabhängig voneinander ausgewählt sein können aus der Gruppe bestehend aus d-3-Alkyl und Benzyl, oder
die Gruppe -NR2 1R22 zusammen ein cyclisches Amin bildet, das ausgewählt sein kann aus der Gruppe bestehend aus Morpholin-4-yl-, 1-Methylpiperazin-4-yl-, 1-Benzylpiperazin-4-yl-, 1-(Ci_3-Alkylcarbonyl)-piperazin-4-yl, 1-(te/f-Butyloxycarbonyl)- piperazin-4-yl, 1-(Benzyloxycarbonyl)-piperazin-4-yl, Piperidin-1-yl- und Pyrrolidin-1-yl-, vorzugsweise Morpholin-4-yl-,
deren Salze und Solvate, umfassend die Schritte:
(a) Umsetzung eines Glykolsäureethylester der allgemeinen Formel Il o

(f) Abspalten einer Schutzgruppe PG aus einer unter (e) erhaltenen Verbindung der allgemeinen Formel VII
in der PG und R1 wie voranstehend erwähnt definiert sind;
(g) Reduktion einer unter (f) erhaltenen Verbindung der allgemeinen Formel VIII
in der R1 wie voranstehend erwähnt definiert ist, in Gegenwart eines Reduktionsmittels und gegebenenfalls zusätzlich in Gegenwart einer Base zu einer Verbindung der allgemeinen Formel IX
in der R1 wie voranstehend erwähnt definiert ist;
(h) Isolieren einer unter (g) erhaltenen Verbindung der allgemeinen Formel IX in Form eines Alkalisalzes der allgemeinen Formel X

(i) Kupplung einer unter (h) erhaltenen Verbindung der allgemeinen Formel X mit einer Verbindung der allgemeinen Formel Xl

in der R5 einen Imidazol- oder Triazol-Rest, vorzugsweise einen Imidazol-Rest, darstellt, der über ein Stickstoffatom gebunden ist;
(j) Umsetzung eines in Schritt (i) entstandenen Produkts der allgemeinen Formel XII
in der R1 wie voranstehend erwähnt definiert ist, mit einer Verbindung der allgemeinen Formeln XIII
R2 ( NH x n HCI w , (XIII) in der R2 wie voranstehend erwähnt definiert ist und n eine der Ziffern 0, 1 , 2 oder 3 bedeutet;
(k) zur Herstellung von Verbindungen der allgemeinen Formel I, in der R1 ein Wasserstoffatom bedeutet, gegebenenfalls anschließend Abspaltung einer vorhandenen Schutzgruppe aus einer Verbindung der allgemeinen Formel I, in der R1 C(O)-O-Benzyl, C(O)-O-te/f-Butyl oder einen Benzylrest darstellt; und
(I) gegebenenfalls Umsetzung einer unter (k) erhaltenen Verbindung der allgemeinen Formel I, in der R1 ein Wasserstoffatom darstellt, mit einer physiologisch verträglichen Säure in einem polaren Lösungsmittel zu einem entsprechenden Salz, Auskristallisieren und Isolieren des entsprechenden Salzes.
Unter einer voranstehend erwähnten Schutzgruppe PG wird allgemein eine Schutzgruppe für eine Hydroxyfunktion verstanden. Beispielsweise kommt als Schutzrest für eine Hydroxygruppe die Trimethylsilyl-, Trimethylsilylethyl-, te/f-Butyldimethylsilyl-, Methoxymethyl-, 2-Methoxyethoxymethyl-, te/f-Butyloxycarbonyl-, Acetyl-, Benzoyl-, Methyl-, Ethyl-, te/f-Butyl-, Trityl-, Benzyl- oder Tetrahydropyranylgruppe in Frage.
Bei der Umsetzung in Schritt (a) können 1.0 Äquivalente Glykolsäureethylester mit 1.0 bis 1.3 Äquivalenten eines Reagenzes zum Einbringen einer Schutzgruppe, beispielsweise
3,4-Dihydro-2/-/-pyran oder Benzylchlorid, umgesetzt werden. Das aprotische Lösungsmittel kann ausgewählt sein aus der Gruppe bestehend aus Methylenchlorid, Toluol, o-Xylol, m-Xylol und p-Xylol sowie entsprechenden Gemischen aus diesen Lösungsmitteln. In einer weiteren Ausführungsform können 0.1 bis 0.5 L Lösungsmittel/mol eingesetztem Glykolsäureethylester verwendet werden.
Die in Schritt (a) verwendete Säure kann ausgewählt sein aus der Gruppe bestehend aus p-Toluolsulfonsäure, Methansulfonsäure, Schwefelsäure und Benzolsulfonsäure.
Die Umsetzung in Schritt (b) kann in einem Lösungsmittel durchgeführt werden, das ausgewählt ist aus der Gruppe bestehend aus Tetrahydrofuran, 2-Methyltetrahydrofuran, Toluol, te/f-Butylmethylether, Dioxan, Mono-, Di-, Tri- und Polyethylenglykolether. Die bei der Umsetzung verwendete starke Base kann ausgewählt sein aus der Gruppe bestehend aus Kalium-te/f-butylat, Kalium-te/f-amylat, Natrium-te/f-butylat, Natrium-te/f-amylat und Lithium-te/f-butylat. Bei der Umsetzung in Schritt (b) können 1.0 Äquivalente einer Verbindung der allgemeinen Formel IV mit 0.5 bis 2.0 Äquivalenten einer Verbindung der allgemeinen Formel III umgesetzt werden. In einer weiteren Ausführungsform werden 0.5 bis 1.0 L Lösungsmittel/mol Verbindung der allgemeinen Formel IV verwendet.
Die voranstehend unter schritt (c) beschriebene Umsetzung kann in Methanol, Ethanol, Propanol, Isopropanol, te/f-Amylalkohol oder te/f-Butanol oder in einem Gemisch dieser
Lösungsmittel durchgeführt werden. Es können 1.0 bis 2.0 L Lösungsmittel/mol
Verbindung der allgemeinen Formel VI verwendet werden.
Die starke anorganische Base kann dabei ausgewählt sein aus der Gruppe bestehend aus Lithiumhydroxid, Kaliumhydroxid und Natriumhydroxid. Vorzugsweise wird die Base in einer Menge von 1.0 bis 1.2 mol pro Mol eingesetzter Verbindung der allgemeinen Formel
V zugegeben.
Die voranstehend unter Schritt (e) beschriebene Umsetzung kann in Toluol, te/f-Butyl- methylether, Dioxan, Tetrahydrofuran oder 2-Methyltetrahydrofuran als Lösungsmittel durchgeführt werden. Es können 1.5 bis 5.0 L Lösungsmittel/mol Verbindung der allgemeinen Formel VI verwendet werden.
Die verwendete Säure kann ausgewählt sein aus der Gruppe bestehend aus Salzsäure, Bromwasserstoffsäure, lodwasserstoffsäure, Phosphorsäure, p-Toluolsulfonsäure, Methansulfonsäure, Schwefelsäure und Benzolsulfonsäure; Salzsäure und Methansulfon-
säure kommen erfindungsgemäß eine besondere Bedeutung zu. Es können 1.5 bis 4.0 Äquivalente Säure pro Mol eingesetzter Verbindung der allgemeinen Formel VI zugegeben werden. Die Reaktion kann bei einer Temperatur von -20 bis 25°C durchgeführt werden.
Die voranstehend unter Schritt (f) erwähnte Abspaltung der Schutzgruppe PG kann nach literaturbekannten Methoden erfolgen, wie sie beispielsweise in "Protective Groups in Organic Synthesis" (Theodora W. Greene, Peter G. M. Wuts, Third Edition, Wiley Interscience) beschrieben sind. Beispielsweise kann die voranstehend unter Schritt (f) beschriebene Abspaltung der Schutzgruppe PG in Toluol, te/f-Butylmethylether, Dioxan, Tetrahydrofuran oder 2-Methyltetrahydrofuran als Lösungsmittel durchgeführt werden. Es können 1.5 bis 5.0 L Lösungsmittel/mol Verbindung der allgemeinen Formel VII verwendet werden. Die verwendete Säure kann beispielsweise ausgewählt sein aus der Gruppe bestehend aus Salzsäure, Bromwasserstoffsäure, lodwasserstoffsäure, Phosphorsäure, p-Toluol- sulfonsäure, Methansulfonsäure, Schwefelsäure und Benzolsulfonsäure; Salzsäure und Methansulfonsäure kommen erfindungsgemäß eine besondere Bedeutung zu. Es können 1.5 bis 4.0 Äquivalente Säure pro Mol eingesetzter Verbindung der allgemeinen Formel VII zugegeben werden. Die Reaktion kann bei einer Temperatur von -20 bis 25°C durchgeführt werden.
In einer weiteren Ausführungsform kann die voranstehend unter Schritt (f) erwähnte Abspaltung der Schutzgruppe PG in Dioxan oder Toluol als Lösungsmittel und unter Zugabe von Salzsäure erfolgen.
Die voranstehend unter Schritt (g) erwähnte Base kann ausgewählt sein aus der Gruppe bestehend aus Triethylamin, Diisopropylethylamin, Λ/-Methylmorpholin, Λ/-Methylpyrrolidin und Pyridin. Es können 2.0 bis 2.6 Äquivalente Base pro Mol eingesetzter Verbindung der allgemeinen Formel VIII zugegeben werden. Das ebenfalls unter Schritt (g) beschriebene Reduktionsmittel kann ausgewählt sein aus der Gruppe bestehend aus ß-Chlordiisopinocampheylboran, Alpine-Boran und Methyl- CBS-oxazaborolidin. Es können 1.0 bis 1.6 Äquivalente Reduktionsmittel pro Mol eingesetzter Verbindung der allgemeinen Formel VIII zugegeben werden.
Die voranstehend unter Schritt (h) beschriebene Isolierung eines Alkalisalzes erfolgt durch Zugabe einer entsprechenden anorganischen Lauge. Diese kann ausgewählt sein aus der Gruppe bestehend aus Lithiumhydroxid, Natriumhydroxid und Kaliumhydroxid, vorzugsweise in Form einer wässrigen Lösung. Es können 1.0. bis 1.5 mol Lauge pro Mol eingesetzter Verbindung der allgemeinen Formel IX zugegeben werden.
Bei der Kupplung in Schritt (i) können 1.0 Äquivalente einer Verbindung der allgemeinen Formel X und 1.0 bis 1.5 Äquivalente einer Verbindung der allgemeinen Formel Xl in einem polaren Lösungsmittel suspendiert und bei erhöhter Temperatur in Gegenwart einer starken Base umgesetzt werden.
Als polare Lösungsmittel können erfindungsgemäß te/f-Butanol, te/f-Amylalkohol, Dimethylformamid, Λ/-Methylpyrrolidon oder Tetrahydrofuran verwendet werden. Vorzugsweise werden 3.0 bis 6.0 L Lösungsmittel pro Mol eingesetzter Verbindung der allgemeinen Formel X verwendet. Die verwendete Base kann ausgewählt sein aus der Gruppe bestehend aus Kalium- te/f-butylat, Natrium-te/f-butylat, Lithium-te/f-butylat, Kalium-te/f-amylat und Natrium- te/f-amylat. Die Reaktion kann bei einer Temperatur zwischen 40 und 900C durchgeführt werden.
Die voranstehend unter Schritt (j) beschriebene Umsetzung kann bei niedriger
Temperatur in Gegenwart eines Amins und eines Kondensationsmittels in einem polaren, aprotischen Lösungsmittel durchgeführt werden. Vorzugsweise werden 1.0 bis 1.5 Äquivalente einer Verbindung der allgemeinen Formel XIII pro Mol eingesetzter Verbindung der allgemeinen Formel XII verwendet. Das verwendete Amin kann ausgewählt sein aus der Gruppe bestehend aus Triethylamin, Diisopropylethylamin, Ethyldiisopropylamin und Tributylamin. Es kann in einer Menge von 4.0 bis 6.0 Äquivalenten pro Mol eingesetzter Verbindung der allgemeinen Formel XII verwendet werden. Das Kondensationsmittel kann ausgewählt sein aus der Gruppe bestehend aus Propan- phosphonsäureanhydrid, Dicyclohexylcarbodiimid, Carbonyldiimidazol, Carbonylditriazol, 2-(1 /-/-Benzotriazol-1-yl)-1 ,1 ,3,3-tetramethyluronium-tetrafluorborat, 1-Ethyl-3-(3'-dimethyl- amino-propyl)-carbodiimid und Chlordimethoxy-triazin, gegebenenfalls in Gegenwart von Hydroxysuccinimid, Hydroxybenzotriazol, p-Nitrophenol oder Pentafluorphenol. Es wird
vorzugsweise in einer Menge von 1.5 bis 2.0 Äquivalenten pro Mol eingesetzter Verbindung der allgemeinen Formel XII verwendet.
Als polares, aprotisches Lösungsmittel können Tetrahydrofuran, 2-Methyltetrahydrofuran, Toluol oder Ethylacetat verwendet werden. Es wird vorzugsweise in einer Menge von 4.0 bis 6.0 L pro Mol eingesetzter Verbindung der allgemeinen Formel XII verwendet.
Erfindungsgemäß bevorzugt wird die Reaktion bei einer Temperatur zwischen 0 und 500C durchgeführt.
Zur optionalen Abspaltung einer Schutzgruppe in Schritt (k) wird eine in Schritt (j) erhaltene Verbindung der allgemeinen Formel I, in der R1 C(O)-O-Benzyl oder einen Benzylrest darstellt, in einem polaren Lösungsmittel, wie beispielsweise Methanol, Ethanol, Wasser, Aceton, Tetrahydrofuran, Dimethylformamid, Toluol oder Propanol, gelöst, und in einem Druckreaktor hydriert. Als Hydrierungsmittel können beispielsweise Pd/C oder Pd(OH)2 verwendet werden. Vorteilhafte Bedingungen für die Hydrierung sind Temperaturen von 40 bis 800C und ein Wasserstoffüberdruck von maximal 3 bar. Nach dem Abfiltrieren des Katalysators kann die Verbindung der allgemeinen Formel I, in der R1 ein Wasserstoffatom darstellt, durch Aufkonzentrierung des Lösungsmittels unter Zugabe eines weiteren polaren Lösungsmittels, vorzugsweise Ethanol, erhalten werden.
Die voranstehend unter Schritt (I) beschriebene Umsetzung kann in Methanol, Ethanol, Propanol, Isopropanol oder Wasser oder in einem Gemisch dieser Lösungsmittel erfolgen. Die physiologisch verträgliche Säure kann dabei ausgewählt sein aus der Gruppe bestehend aus Bromwasserstoffsäure, Phosphorsäure, Salpetersäure, Salzsäure, Schwefelsäure, Methansulfonsäure, Ethansulfonsäure, Benzolsulfonsäure, p-Toluolsulfon- säure, Äpfelsäure, Bernsteinsäure, Essigsäure, Fumarsäure, Maleinsäure, Mandelsäure, Milchsäure, Weinsäure und Zitronensäure.
Die Verbindungen der allgemeinen Formel Xl
■ (XI)
in der R5 einen Imidazol- oder Triazol-Rest, vorzugsweise einen Imidazol-Rest, der über ein Stickstoffatom gebunden ist, bedeutet, werden nach einem Verfahren hergestellt, umfassend die Schritte:
(a) Lösen von Carbonyldiimidazol oder Carbonylditriazol, vorzugsweise Carbonyldi- imidazol, in einem polaren, aprotischen Lösungsmittel und Umsetzung bei erhöhter Temperatur mit 1 ,3,4,5-Tetrahydro-3-(4-piperidinyl)-2/-/-1 ,3-benzodi- azepin-2-on; und
(b) Auskristallisieren eines in Schritt (a) entstandenen Rohprodukts durch Zugabe eines weiteren polaren, aprotischen Lösungsmittels, wenn R5 eine Imidazol- Gruppe darstellt.
Das voranstehend unter Schritt (a) erwähnte Lösungsmittel kann ausgewählt sein aus der Gruppe bestehend aus Aceton, Acetonitril, te/f-Butylmethylether, Λ/,Λ/-Dimethylacetamid, Dimethylformamid, Dimethylsulfoxid, Pyridin und Λ/-Methylpyrrolidon.
Das voranstehend unter Schritt (b) erwähnte polare, aprotische Lösungsmittel kann ausgewählt sein aus der Gruppe bestehend aus te/f-Butylmethylether, Dimethylformamid, Tetrahydrofuran, Toluol und 2-Methyltetrahydrofuran.
Ein Verfahren zur Herstellung von 1 ,3,4,5-Tetrahydro-3-(4-piperidinyl)-2/-/-1 ,3-benzodi- azepin-2-on wird in der europäischen Patentanmeldung Nr. 04017424.5 beschrieben.
Ein zweiter Gegenstand der vorliegenden Erfindung betrifft die Verbindungen der allgemeinen Formel V
in der
PG eine Schutzgruppe, vorzugsweise eine Gruppe ausgewählt aus
R1 H, d-s-Alkyl, C(O)-O-Benzyl, C(O)-O-te/f-Butyl oder Benzyl, vorzugsweise Benzyl, und
R3 C-ι-6-Alkyl, vorzugsweise Ethyl,
bedeutet.
Ein bevorzugter zweiter Gegenstand umfasst die folgenden Verbindungen der Formeln Va bis Vd:

Ein weiter bevorzugter zweiter Gegenstand betrifft die Verbindung (3Z)-3-(4-Benzyloxy- 3,5-dimethyl-phenyl)-2-tetrahydropyran-2-yloxy)-acrylsäureethylester der Formel Va
welche kristallin anfällt und durch ein hohes Maß an Stabilität gekennzeichnet ist.
Die kristalline Verbindung der Formel Va ist gekennzeichnet durch einen charakteristischen Schmelzpunkt von T = 123 ± 3°C. Der aufgeführte Wert wurde mittels Differential Scanning Calorimetry (DSC: Auswertung über Onset, Heizrate: 10°C/min) bestimmt (Netzsch STA Jupiter).
Ein weiterer Gegenstand der vorliegenden Erfindung betrifft die Verwendung der voranstehend genannten Verbindungen der allgemeinen Formel V als Zwischenprodukte zur Herstellung von Verbindungen der allgemeinen Formel I nach einem voranstehend unter der ersten Ausführungsform beschriebenen Verfahren.
Ein dritter Gegenstand der vorliegenden Erfindung betrifft ein Verfahren zur Herstellung von Verbindungen der allgemeinen Formel V

Bei der Umsetzung in Schritt (a) können 1.0 Äquivalente Glykolsäureethylester mit 1.0 bis 1.1 Äquivalenten eines Reagenzes zum Einbringen einer Schutzgruppe, beispielsweise 3,4-Dihydro-2/-/-pyran oder Benzylchlorid, umgesetzt werden. Das unpolare, aprotische Lösungsmittel kann ausgewählt sein aus der Gruppe bestehend aus Toluol, o-Xylol, m-Xylol und p-Xylol sowie entsprechenden Gemischen aus diesen Lösungsmitteln. Es können 0.1 bis 0.5 L Lösungsmittel/mol Glykolsäureethylester verwendet werden. Das in Schritt (a) verwendete Benzylhalogenid kann ausgewählt sein aus der Gruppe bestehend aus Benzylchlorid, Benzylbromid und Benzyliodid. Die in Schritt (a) verwendete Säure kann ausgewählt sein aus der Gruppe bestehend aus p-Toluolsulfonsäure, Methansulfonsäure, Schwefelsäure und Benzolsulfonsäure. Die in Schritt (a) verwendete Base kann ausgewählt sein aus der Gruppe bestehend aus anorganischen Basen oder organischen Basen. Als anorganische Basen können Alkali- metallhydroxide, beispielsweise Natriunhydroxid oder Kaliumhydroxid, oder Silberoxid verwendet werden. Als organische Basen können tertiäre Amine, beispielsweise Triethyl- amin oder Hünig-Base, verwendet werden.
Die Umsetzung in Schritt (b) kann in einem Lösungsmittel erfolgen, das ausgewählt ist aus der Gruppe bestehend aus 2-Methyltetrahydrofuran, Toluol, Tetrahydrofuran, te/f-Butylmethylether, Dioxan, Mono-, Di-, Tri- und Polyethylenglykolether. Die bei der Umsetzung verwendete starke Base kann ausgewählt sein aus der Gruppe bestehend aus 1 ,4-Diazabicyclo[2,2,2]octan (DABCO), Kalium-te/f-butylat, Kalium-te/f-amylat, Natrium-te/f-butylat, Natrium-te/f-amylat, Lithium-te/f-butylat. Sofern die Verbindungen der allgemeinen Formel V kristallin sind, können sie wie voranstehend unter Schritt (c) beschrieben umkristallisiert werden.
Ein vierter Gegenstand der vorliegenden Erfindung betrifft die Verbindungen der allgemeinen Formel VI
in der
PG eine Schutzgruppe, vorzugsweise eine Gruppe ausgewählt aus

Die kristalline Verbindung der Formel VIa ist gekennzeichnet durch einen charakteristischen Schmelzpunkt von T = 89 ± 3°C. Der aufgeführte Wert wurde mittels Differential
Scanning Calorimetry (DSC: Auswertung über Onset, Heizrate: 10°C/min) bestimmt (Netzsch STA Jupiter).
Ein weiter bevorzugter vierter Gegenstand betrifft die Verbindung (3Z)-3-(4-te/f-Butyl- oxycarbonyloxy-S.δ-dimethyl-phenyl^-tetrahydro-pyran^-yloxy^acrylsäure-monodicyclo- hexylaminsalz der Formel VIe
welche kristallin anfällt und durch ein hohes Maß an Stabilität gekennzeichnet ist.
Die kristalline Verbindung der Formel VIe ist gekennzeichnet durch einen charakteristischen Schmelzpunkt von T = 140 ± 3°C. Der aufgeführte Wert wurde mittels Differential Scanning Calorimetry (DSC: Auswertung über Onset, Heizrate: 10°C/min) bestimmt (Netzsch STA Jupiter).
Ein weiterer Gegenstand der vorliegenden Erfindung betrifft die Verwendung der voranstehend genanten Verbindungen der allgemeinen Formel VI als Zwischenprodukte zur Herstellung von Verbindungen der allgemeinen Formel I nach einem voranstehend unter der ersten Ausführungsform beschriebenen Verfahren.
Ein fünfter Gegenstand der vorliegenden Erfindung betrifft ein Verfahren zur Herstellung von Verbindungen der allgemeinen Formel VI
in der
PG eine Schutzgruppe, vorzugsweise eine Gruppe ausgewählt aus

M+ ein Metallkation ausgewählt aus Na+, K+ und Li+, bevorzugt K+,

(a) eine Verbindung der allgemeinen Formel V
in der PG, R1 und R3 wie voranstehend erwähnt definiert sind, mit einem polaren Lösungsmittel gemischt und eine starke anorganische Base zugegeben wird; und
(b) eine unter (a) erhaltene Verbindung der allgemeinen Formel VI
in der PG, R1 und R4 wie voranstehend erwähnt definiert sind, anschließend isoliert wird.
Die voranstehend unter Schritt (a) beschriebene Umsetzung kann in Methanol, Ethanol, Propanol, Isopropanol, te/f-Amylalkohol oder te/f-Butanol oder in einem Gemisch dieser polaren Lösungsmittel erfolgen.
Die starke anorganische Base kann dabei ausgewählt sein aus der Gruppe bestehend aus Lithiumhydroxid, Kaliumhydroxid und Natriumhydroxid.
Die Herstellung einer Ausgangsverbindung der allgemeinen Formel V erfolgt wie voranstehend unter der dritten Ausführungsform beschrieben.
Ein sechster Gegenstand der vorliegenden Erfindung betrifft ein Verfahren zur Herstellung von Verbindungen der allgemeinen Formel XIII

die Gruppe -NR2 1R22 zusammen ein cyclisches Amin bildet, das ausgewählt sein kann aus der Gruppe bestehend aus Morpholin-4-yl-, 1-Methylpiperazin-4-yl-, 1 -Benzylpiperazin-4-yl-, 1 -(Ci_3-Alkylcarbonyl)-piperazin-4-yl, 1 -(te/f-Butyloxycarbonyl)- piperazin-4-yl, 1-(Benzyloxycarbonyl)-piperazin-4-yl, Piperidin-1-yl- und Pyrrolidin-1-yl-, vorzugsweise Morpholin-4-yl,
umfassend die Schritte:
(a) Umsetzung von 1-Benzylpiperidon mit einem Amin der allgemeinen Formel XIV

(b) Reduzieren des unter (a) entstandenen Enamins unter Druck und in Gegenwart eines Katalysators und Isolieren des erhaltenen Produkts der allgemeinen Formel XV
in der n, R2 1 und R22 wie voranstehend erwähnt definiert sind, durch Zugabe von Salzsäure;
(c) Entfernen der Benzyl-Schutzgruppe aus einer unter (b) erhaltenen Verbindung der allgemeinen Formel XV und Isolieren einer Verbindung der allgemeinen Formel
XVI
in der n, R2 1 und R22 wie voranstehend erwähnt definiert sind.
Bei der Umsetzung in Schritt (a) können 1.0 Äquivalente 1-Benzylpiperidon mit 1.8 bis 2.2 Äquivalenten, vorzugsweise 2.0 Äquivalenten, eines Amins der allgemeinen Formel XIV umgesetzt werden.
Das verwendete Lösungsmittel kann ausgewählt sein aus der Gruppe bestehend aus Ethylacetat, Isopropylacetat, Toluol und Methyltetrahydrofuran, wobei Isopropylacetat bevorzugt verwendet wird. Vorzugsweise werden 0.5 bis 1.0 L Lösungsmittel/mol 1-Benzylpiperidon verwendet.
Die Reduktion in Schritt (b) wird in Gegenwart eines Katalysators durchgeführt, der ausgewählt sein kann aus der Gruppe bestehend aus Raney-Nickel, Platin/Kohle und Platindioxid; bevorzugt wird Raney-Nickel verwendet. Vorteilhafte Bedingungen für die Hydrierung sind Temperaturen von 20 bis 700C und ein Wasserstoffüberdruck von maximal 5 bar. Nach dem Abfiltrieren des Katalysators kann das Hydrierungsprodukt durch Abdestillieren des Lösungsmittels aufkonzentriert werden.
Zur Isolierung eines Hydrochlorids der allgemeinen Formel XV werden 2.0 bis 2.5 Äquivalente Salzsäure zugegeben, bezogen auf die Menge an eingesetztem 1-Benzylpiperidon.
Die unter Schritt (c) beschriebene Abspaltung einer Benzyl-Schutzgruppe aus einer Verbindung der allgemeinen Formel XV, kann mittels Hydrierung in einem polaren Lösungsmittel, wie beispielsweise Methanol, Ethanol, Propanol, te/f-Butanol, Wasser, Aceton, Tetrahydrofuran, Dimethylformamid oder Mischungen dieser Lösungsmittel, erfolgen. Dabei kann das Lösungsmittel in einer Menge von 0.5 bis 2.5 L/mol eingesetzter Verbindung der allgemeinen Formel XV, vorzugsweise von 0.7 bis 2.2 L/mol eingesetzter Verbindung der allgemeinen Formel XV, bevorzugt von 0.7 bis 1.5 L/mol eingesetzter Verbindung der allgemeinen Formel XV, besonders bevorzugt von 1.0 L/mol eingesetzter Verbindung der allgemeinen Formel XV, zugegeben werden. Die Hydrierung wird in Gegenwart eines Katalysators durchgeführt, der ausgewählt sein kann aus der Gruppe bestehend aus Palladium/Kohle und Palladiumhydroxid; bevorzugt wird Palladium/Kohle verwendet.
Vorteilhafte Bedingungen für die Hydrierung sind Temperaturen von 40 bis 800C und ein Wasserstoffüberdruck von maximal 5 bar. Das Isolieren einer Verbindung der allgemeinen Formel XIII kann beispielsweise mittels Kristallisation erfolgen.
Alternativ kann eine Verbindung der allgemeinen Formel XIII, in der n und R2 wie voranstehend erwähnt definiert sind, auch nach einem Verfahren hergestellt werden, umfassend die Schritte:
(a) Umsetzung von 1-Benzylpiperidon mit einem Amin der allgemeinen Formel XIV

in der R2 1 und R22 wie voranstehend erwähnt definiert sind, in einem Lösungsmittel unter Wasserabspaltung;
(b) Reduzieren des unter (a) entstandenen Enamins unter Druck und in Gegenwart eines Katalysators und Isolieren des erhaltenen Produkts der allgemeinen Formel XV
in der n, R2 1 und R22 wie voranstehend erwähnt definiert sind, durch Zugabe von Salzsäure;
(c) Umsetzung einer unter (b) erhaltenen Verbindung der allgemeinen Formel XV, in der n, R2 1 und R22 wie voranstehend erwähnt definiert sind, mit einer Base in einem polaren Lösungsmittel, wodurch eine Verbindung der allgemeinen Formel XXVV eennttsstteehhtt,, iinn d der R2 1 und R22 wie voranstehend erwähnt definiert sind und n die Ziffer 0 darstellt;
(d) Entfernen der Benzyl-Schutzgruppe aus einer unter (c) erhaltenen Verbindung der allgemeinen Formel XV und Isolieren einer Verbindung der allgemeinen Formel XVI

(e) Umsetzung einer unter (d) erhaltenen Verbindung der allgemeinen Formel XVI, in der n die Ziffer 0 darstellt, durch Zugabe von Salzsäure in einem Lösungsmittel zu einer Verbindung der allgemeinen Formel XVI, in der n eine der Ziffern 1 , 2 oder 3 darstellt.
Bei der Umsetzung in Schritt (a) können 1.0 Äquivalente 1-Benzylpiperidon mit 1.8 bis 2.2 Äquivalenten, vorzugsweise 2.0 Äquivalenten, eines Amins der allgemeinen Formel XIV umgesetzt werden.
Das verwendete Lösungsmittel kann ausgewählt sein aus der Gruppe bestehend aus Ethylacetat, Isopropylacetat, Toluol und Methyltetrahydrofuran, wobei Isopropylacetat
bevorzugt verwendet wird. Vorzugsweise werden 0.5 bis 1.0 L Lösungsmittel/mol 1-Benzylpiperidon verwendet.
Die Reduktion in Schritt (b) wird in Gegenwart eines Katalysators durchgeführt, der ausgewählt sein kann aus der Gruppe bestehend aus Raney-Nickel, Platin/Kohle und
Platindioxid; bevorzugt wird Raney-Nickel verwendet. Vorteilhafte Bedingungen für die
Hydrierung sind Temperaturen von 20 bis 700C und ein Wasserstoffüberdruck von maximal 5 bar. Nach dem Abfiltrieren des Katalysators kann das Hydrierungsprodukt durch Abdestillieren des Lösungsmittels aufkonzentriert werden. Zur Isolierung eines Hydrochlorids der allgemeinen Formel XV werden 2.0 bis 2.5
Äquivalente Salzsäure zugegeben, bezogen auf die Menge an eingesetztem
1-Benzylpiperidon.
Die unter Schritt (c) beschriebene Umsetzung kann in einem polaren Lösungsmittel, das ausgewählt ist aus der Gruppe bestehend aus Methanol, Ethanol, Propanol, Isopropanol,
Butanol, Wasser, te/f-Butanol oder Mischungen dieser Lösungsmittel, erfolgen. Dabei kann das Lösungsmittel in einer Menge von 1.0 bis 3.0 L/mol eingesetzter Verbindung der allgemeinen Formel XV verwendet werden.
Als Base kann ein Alkalimetallhydroxid oder ein Alkalicarbonat, beispielsweise Natriumhydroxid, Kaliumhydroxid, Natriumcarbonat oder Kaliumcarbonat, verwendet werden.
Die unter Schritt (d) beschriebene Abspaltung einer Benzyl-Schutzgruppe aus einer Verbindung der allgemeinen Formel XV, kann mittels Hydrierung in einem polaren Lösungsmittel, wie beispielsweise Methanol, Ethanol, Propanol, te/f-Butanol, Wasser, Aceton, Tetrahydrofuran, Dimethylformamid oder Mischungen dieser Lösungsmittel, erfolgen. Dabei kann das Lösungsmittel in einer Menge von 0.5 bis 2.5 L/mol eingesetzter Verbindung der allgemeinen Formel XV, vorzugsweise von 0.7 bis 1.5 L/mol eingesetzter Verbindung der allgemeinen Formel XV, besonders bevorzugt von 1.3 L/mol eingesetzter Verbindung der allgemeinen Formel XV, zugegeben werden.
Die Hydrierung wird in Gegenwart eines Katalysators durchgeführt, der ausgewählt sein kann aus der Gruppe bestehend aus Palladium/Kohle und Palladiumhydroxid; bevorzugt wird Palladium/Kohle verwendet.
Vorteilhafte Bedingungen für die Hydrierung sind Temperaturen von 40 bis 800C und ein Wasserstoffüberdruck von maximal 5 bar.
Das Isolieren einer Verbindung der allgemeinen Formel XIII kann beispielsweise mittels Kristallisation erfolgen.
Die unter schritt (e) beschriebene Umsetzung zu einer Verbindung der allgemeinen Formel XVI, in der n die Ziffer 1 , 2 oder 3 darstellt, erfolgt durch Zugabe von 1.8 bis 2.5 Äquivalenten, vorzugsweise 2.0 bis 2.2 Äquivalenten Salzsäure, jeweils bezogen auf die Menge an eingesetzter Verbindung der allgemeinen Formel XVI, in der n die Ziffer 0 darstellt.
Als Lösungsmittel können Methanol oder Ethanol verwendet werden. Das Lösungsmittel wird in einer Menge von 1.0 bis 7.0 L/mol eingesetzter Verbindung der allgemeinen Formel XVI zugegeben, in der n die Ziffer 0 darstellt.
Ein weiterer Gegenstand der vorliegenden Erfindung betrifft die Verwendung der voranstehend genannten Verbindungen der allgemeinen Formel XIII als Zwischenprodukte zur Herstellung von Verbindungen der allgemeinen Formel I nach einem voranstehend unter der ersten Ausführungsform beschriebenen Verfahren.
VERWENDETE BEGRIFFE UND DEFINITIONEN
Unter dem Begriff "sekundäres Amin" wird eine Aminogruppe der allgemeinen Formel -NR2 1R2 2 verstanden, wobei die Reste R2 1 und R22 unabhängig voneinander ausgewählt sein können aus der Gruppe bestehend aus Ci-3-Alkyl und Benzyl, oder die Gruppe -NR2 1R2 2 zusammen ein cyclisches Amin bildet, das ausgewählt sein kann aus der

Unter dem Begriff "C1-3-Alkyl" (auch soweit sie Bestandteil anderer Reste sind) werden verzweigte und unverzweigte Alkylgruppen mit 1 bis 3 Kohlenstoffatomen und unter dem Begriff "C1-6-Alkyl" verzweigte und unverzweigte Alkylgruppen mit 1 bis 6 Kohlenstoff- atomen verstanden. Beispielsweise werden hierfür genannt: Methyl, Ethyl, n-Propyl, iso- Propyl, n-Butyl, /so-Butyl, te/f-Butyl, n-Pentyl, te/f-Amyl oder n-Hexyl. Gegebenenfalls werden für vorstehend genannten Gruppen auch die Abkürzungen Me, Et, n-Pr, /-Pr etc. verwendet.
Unter dem Begriff "Cs-β-Cycloalkyl" (auch soweit sie Bestandteil anderer Reste sind) werden cyclische Alkylgruppen mit 3 bis 6 Kohlenstoffatomen verstanden. Beispielsweise werden hierfür genannt: Cyclopropyl, Cyclobutyl, Cyclopentyl oder Cyclohexyl. Soweit nicht anders beschrieben, können die cyclischen Alkylgruppen substituiert sein mit einem oder mehreren Resten ausgewählt aus der Gruppe bestehend aus Methyl, Ethyl, iso- Propyl, te/f-Butyl, Hydroxy, Fluor, Chlor, Brom und Jod.
Die Verbindungen der allgemeinen Formel I können basische Gruppen wie z.B. Amino- funktionen besitzen. Sie können daher als innere Salze, als Salze mit pharmazeutisch verwendbaren anorganischen Säuren wie beispielsweise Bromwasserstoffsäure, Phosphorsäure, Salpetersäure, Salzsäure, Schwefelsäure, Methansulfonsäure, Ethan- sulfonsäure, Benzolsulfonsäure, p-Toluolsulfonsäure oder organischen Säuren wie beispielsweise Äpfelsäure, Bernsteinsäure, Essigsäure, Fumarsäure, Maleinsäure, Mandelsäure, Milchsäure, Weinsäure oder Zitronensäure vorliegen.
Gegenstand der Erfindung sind die jeweiligen Verbindungen gegebenenfalls in Form der einzelnen optischen Isomeren, Mischungen der einzelnen Enantiomeren oder Racemate, in Form der Tautomere sowie in Form der freien Basen oder der entsprechenden Säureadditionssalze mit pharmakologisch unbedenklichen Säuren.
EXPERIMENTELLER TEIL
Beispiel 1.1 : (Tetrahydropyran-2-yloxy)-essigsäureethylester (B)
(A) (B)
20.00 kg (182.50 mol) Glykolsäureethylester (A) wurden in 60.0 L Toluol gelöst und 71.04 g (0.366 mol) 4-Toluolsulfonsäure Monohydrat zugegeben. Zur erhaltenen Reaktionsmischung wurde bei 200C eine Lösung aus 15.83 kg (182.50 mol) 3,4-Dihydro-2/-/-pyran in 40.0 L Toluol zugetropft. Anschließend wurde die Reaktionsmischung 1 Stunde bei 200C gerührt und nach vollständiger Umsetzung mit 37.0 L Wasser und 3.69 L (49.28 mol) Ammoniaklösung (25%) versetzt. Nach Phasentrennung wurde die organische Phase mit 40.0 L Wasser gewaschen und anschließend das Lösungsmittel im Vakuum vollständig abdestilliert. Ausbeute: 32.0 kg (88% der Theorie)
Beispiel 1.2: (Tetrahydropyran-2-yloxy)-essigsäureethylester (B)
(A) (ß)
100.00 kg (960.52 mol) Glykolsäureethylester (A) wurden in 180.0 L Toluol gelöst und 365.22 g (1.92 mol) 4-Toluolsulfonsäure Monohydrat zugegeben. Zur erhaltenen Reaktionsmischung wurde bei 200C eine Lösung aus 80.80 kg (960.52 mol) 3,4-Dihydro- 2/-/-pyran zugetropft und anschließend mit 20.0 L Toluol nachgewaschen. Die Reaktions- mischung wurde 1 Stunde bei 200C gerührt und nach vollständiger Umsetzung mit
100.0 L Wasser und 6.53 kg (96.05 mol) Ammoniaklösung (25%) versetzt. Nach Phasentrennung wurde die organische Phase mit 100.0 L Wasser gewaschen und anschließend das Lösungsmittel im Vakuum vollständig abdestilliert. Ausbeute: 188.4 kg (94.7% der Theorie)
Beispiel 2.1 : (3Z)-3-(4-Benzyloxy-3,5-dimethyl-phenyl)-2-tetrahydro-pyran-2-yloxy)- acrylsäureethylester (D)
20.00 kg (83.23 mol) 4-Benzyloxy-3,5-dimethylbenzaldehyd (C) wurden in 50.0 L Tetra- hydrofuran gelöst und 31.68 kg (149.81 mol) (Tetrahydropyran-2-yloxy)-essigsäure- ethylester (B) zugegeben. Bei 20°C wurden 25.68 kg (91.55 mol) Kalium-te/f-amylat (45%ig in Tetrahydrofuran) zugetropft. Anschließend wurde die erhaltene Reaktionsmischung 3.5 Stunden bei 200C gerührt und nach vollständiger Umsetzung mit 120.0 L Wasser versetzt. Die erhaltene Suspension wurde auf 00C abgekühlt und 1 Stunde bei dieser Temperatur gerührt. Anschließend wurde das Produkt abzentrifugiert und getrocknet.
Ausbeute: 25.04 kg (73% der Theorie)
Chemische Reinheit (HPLC): 99.8% Schmelzpunkt: 122.6°C
Beispiel 2.2: (3Z)-3-(4-Benzyloxy-3,5-dimethyl-phenyl)-2-tetrahydro-pyran-2-yloxy)- acrylsäureethylester (D)
100.0 kg (416.15 mol) 4-Benzyloxy-3,5-dimethylbenzaldehyd (C) wurden in 200.0 L Tetra- hydrofuran gelöst und 141.41 kg (749.06 mol) (Tetrahydropyran-2-yloxy)-essigsäureethyl- ester (B) zugegeben. Bei 200C wurden 128.42 kg (457.76 mol) Kalium-te/f-amylat (45%ig in Tetrahydrofuran) zugetropft. Anschließend wurde die erhaltene Reaktionsmischung 2 Stunden bei 25°C gerührt und nach vollständiger Umsetzung mit 300.0 L Wasser versetzt. Die erhaltene Suspension wurde 1 Stunde bei 25°C gerührt. Anschließend wurde das Produkt abzentrifugiert und getrocknet. Ausbeute: 118.9 kg (69.6% der Theorie)

Beispiel 3.1 : (3Z)-3-(4-Benzyloxy-3,5-dimethyl-phenyl)-2-tetrahydro-pyran-2-yloxy)- acrylsäure-Monokaliumsalz (E)
35.00 kg (85.26 mol) (3Z)-3-(4-Benzyloxy-3,5-dimethyl-phenyl)-2-tetrahydro-pyran-2-yl- oxy)-acrylsäureethylester (D) wurden in 157 L te/f-Butanol suspendiert und 11.69 kg (93.79 mol) einer 45%igen Kaliumhydroxid-Lösung zudosiert. Nach Spülen mit 17.5 L tert- Butanol wurde die Reaktionsmischung zum Rückfluss erhitzt und 1 Stunde gerührt. Anschließend wurde die Lösung auf 600C abgekühlt und 140.0 L te/f-Butylmethylether zugegeben. Danach wurde die Lösung weiter abgekühlt und bei 200C angeimpft. Nach 1.5 Stunden Rühren bei 200C wurde das erhaltene Produkt abzentrifugiert und getrocknet. Ausbeute: 35.50 kg (99% der Theorie)

Beispiel 3.2: (3Z)-3-(4-Benzyloxy-3,5-dimethyl-phenyl)-2-tetrahydro-pyran-2-yloxy)- acrylsäure-Monokaliumsalz (E)
115.0 kg (280.15 mol) (3Z)-3-(4-Benzyloxy-3,5-dimethyl-phenyl)-2-tetrahydro-pyran-2-yl- oxy)-acrylsäureethylester (D) wurden in 430 L te/f-Amylalkohol suspendiert und 38.42 kg (308.16 mol) einer 45%igen Kaliumhydroxid-Lösung zudosiert. Nach Spülen mit 30.0 L te/f-Amylalkohol wurde die Reaktionsmischung zum Rückfluss erhitzt und 1 Stunde gerührt. Anschließend wurde die Lösung auf 600C abgekühlt und 230.0 L te/f-Butyl- methylether zugegeben. Danach wurde die Lösung weiter abgekühlt und bei 400C angeimpft. Nach 1.0 Stunden Rühren bei 200C wurde das erhaltene Produkt abzentri- fugiert und getrocknet. Ausbeute: 1 16.6 kg (99% der Theorie) Chemische Reinheit (HPLC): 99.8% Schmelzpunkt: 89°C
Beispiel 4.1 : (α/?)-α-Hydroxy-3,5-dimethyl-4-(phenylmethoxy)-phenylpropionsäure- Mononatriumsalz (H)
24.00 kg (57.07 mol) (3Z)-3-(4-Benzyloxy-3,5-dimethyl-phenyl)-2-tetrahydro-pyran-2-yl- oxy)-acrylsäure-Monokaliumsalz (E) wurden in 240.0 L 2-Methyltetrahydrofuran suspendiert. Bei 0 bis 5°C wurden 12.07 kg (125.55 mol) Methansulfonsäure zugegeben. Die erhaltene Suspension wurde bei 00C 30 Minuten gerührt. Nach Abkühlen der erhaltenen Lösung auf -15°C wurden 13.86 kg (136.97 mol) Triethylamin zugegeben. Die
erhaltene Suspension wurde anschließend bei einer Temperatur von -15°C mit 42.24 kg (85.60 mol) mit Diisopinocampheylborchlorid (65%ig in Heptan) versetzt. Anschließend ließ man die Temperatur der Reaktionsmischung innerhalb von 1 Stunde auf 110C ansteigen und tropfte 48.0 L Wasser zu. Die Temperatur des 2-Phasengemisches wurde auf 25°C eingestellt und die wässrige Phase abgetrennt. Die organische Phase wurde mit 48.0 L Wasser gewaschen. Anschließend wurden im Vakuum 144 L Lösungsmittel abdestilliert. Der erhaltene Rückstand wurde durch Zugabe von 168.0 L Tetrahydrofuran verdünnt und auf 3°C abgekühlt. Die erhaltene Reaktionslösung wurde bei 3°C angeimpft und eine Mischung aus 4.79 kg (59.92 mol) Natriumhydroxid-Lösung (50%ig) in 20 L Wasser zudosiert. Nach 45 Minuten Rühren bei 0 bis 5°C wurde das erhaltene Produkt abzentrifugiert und getrocknet.
Ausbeute: 9.80 kg (53% der Theorie) ee-Wert: 98%

Beispiel 4.2: (α/?)-α-Hydroxy-3,5-dimethyl-4-(phenylmethoxy)-phenylpropionsäure- Mononatriumsalz (H)
48.00 kg (1 14.14 mol) (3Z)-3-(4-Benzyloxy-3,5-dimethyl-phenyl)-2-tetrahydro-pyran-2-yl- oxy)-acrylsäure-Monokaliumsalz (E) wurden in 288.0 L Toluol suspendiert. Bei 0 bis 5°C wurden 23.04 kg (239.69 mol) Methansulfonsäure zugegeben. Die erhaltene Suspension wurde bei -5°C 30 Minuten gerührt. Nach vollständiger Reaktion wurden 26.56 kg (262.52 mol) Triethylamin zugegeben. Die Suspension wurde anschließend bei einer Temperatur von -5°C mit 84.49 kg (171.21 mol) mit Diisopinocampheylborchlorid (65%ig in Heptan) versetzt. Anschließend ließ man die Temperatur der Reaktionsmischung innerhalb von 1 Stunde auf 200C ansteigen und tropfte 192.0 L Wasser zu. Die Temperatur des 2-Phasengemisches wurde auf 25°C eingestellt und die wässrige Phase abgetrennt. Die organische Phase wurde mit 308.0 L Wasser und 10.04 kg (125.55 mol) Natriumhydroxid- Lösung (50%ig) versetzt. Das 2-Phasengemisch wurde auf 700C erhitzt und die wässrige
Phase (Produktphase) abgetrennt. Anschließend wurden weitere 144 L Wasser zugegeben und im Vakuum 48.0 L Lösungsmittel abdestilliert. Die Lösung wurde auf 45°C abgekühlt und angeimpft. Danach wurde weiter auf 200C abgekühlt. Nach 30 Minuten Rühren bei 200C wurde das erhaltene Produkt abzentrifugiert und getrocknet. Ausbeute: 23.9 kg (65% der Theorie) ee-Wert: 99.6%

Beispiel 4.3: (α/?)-α-Hydroxy-3,5-dimethyl-4-(phenylmethoxy)-phenylpropionsäure- Mononatriumsalz (H)
61.9 kg (147.4 mol) (3Z)-3-(4-Benzyloxy-3,5-dimethyl-phenyl)-2-tetrahydro-pyran-2-yloxy)- acrylsäure-Monokaliumsalz (E) wurden in 300 kg Toluol suspendiert und bei 0 C 27.0 L (189 mol) dioxanische HCl (7.0 mol) zudosiert sowie 22 kg Toluol nachgegeben. Die erhaltene Suspension wurde bei gleicher Temperatur für 30 Minuten gerührt. Die
Suspension wurde anschließend bei einer Temperatur von 200C innerhalb von 30 min mit 87.3 kg (177.3 mol) Diisopinocampheylborchlorid (65%ig in Heptan) versetzt und anschließend 30 min bei gleicher Temperatur nachgerührt. Nachfolgend wurde die entstandene Lösung bei gleicher Temperatur mit 309 L Wasser extrahiert. Die organische Phase wurde mit 319 L Wasser und 14.4 kg (162 mol) Natriumhydroxid-Lösung (45%ig) versetzt. Das 2-Phasengemisch wurde auf 70°C erhitzt und die wässrige Phase (Produktphase) abgetrennt. Nach Verdünnung dieser Phase mit 124 L Wasser wurde die Lösung auf 400C abgekühlt, mit 240 g (H) angeimpft, auf 25°C abgekühlt und mind. 30 min nachgerührt. Das erhaltene Produkt wurde abfiltriert, mit 20 ml Wasser gewaschen und getrocknet.
Ausbeute: 26.7 kg (56% der Theorie) ee-Wert: 98.6%

Beispiel 4.4: (α/?)-α-Hydroxy-3,5-dimethyl-4-(phenylmethoxy)-phenylpropionsäure- Mononatriumsalz (H)
20 g (47.6 mmol) (3Z)-3-(4-Benzyloxy-3,5-dimethyl-phenyl)-2-tetrahydro-pyran-2-yloxy)- acrylsäure-Monokaliumsalz (E) wurden in 60 ml_ Toluol suspendiert. Bei 0 bis 5°C wurden 12 ml_ (144.9 mmol) 37 %ige wässrige Salzsäure zugegeben. Die erhaltene Suspension wurde bei 5 - 100C 30 Minuten gerührt. Die Suspension wurde anschließend bei einer Temperatur von 200C mit 28.2 g (57.2 mmol) Diisopinocampheylborchlorid (65%ig in Heptan) versetzt und anschließend 30 min bei gleicher Temperatur nachgerührt. Nachfolgend wurde die entstandene Lösung bei gleicher Temperatur mit 60 ml_ Wasser extrahiert. Die organische Phase wurde mit 60 ml_ Wasser und 4.2 g (52.5 mmol) Natriumhydroxid-Lösung (50%ig) versetzt. Das 2-Phasengemisch wurde auf 700C erhitzt und die wässrige Phase (Produktphase) abgetrennt. Nach Verdünnung mit 80 mL Wasser wurde die Lösung auf 400C abgekühlt, angeimpft und weitere 10 min bei Animpftemperatur gerührt. Danach wurde weiter auf 20°C abgekühlt und 2 h nachgerührt. Das erhaltene Produkt wurde abfiltriert, mit 20 ml Wasser gewaschen und getrocknet. Ausbeute: 9.2 g (60% der Theorie) ee-Wert: >99.9%

Beispiel 5.1 : (1/?)-4-(1 ,2,4,5-Tetrahydro-2-oxo-3H-1,3-benzodiazepin-3-yl)-1 -car- boxy-2-[3,5-dimethyl-4-(phenylmethoxy)phenyl]-1 -piperidincarbon- säureethylester (J)
10.00 kg (31.02 mol) ) (α/?)-α-Hydroxy-3,5-dimethyl-4-(phenylmethoxy)-phenylpropion- säure-Mononatriumsalz (H) wurden vorgelegt, 12.11 kg (35.68 mol) 1 -(1 H-lmidazol-1 -yl- carbonyl)-4-(1 ,2,4,5-tetrahydro-2-oxo-3/-/-1 ,3-benzodiazepin-3-yl)-piperidin (I) zugegeben und diese Mischung in 160.0 L te/f-Amylalkohol suspendiert. 30.0 L Lösungsmittel wurden anschließend bei Normaldruck abdestilliert. Danach wurden zur Reaktionsmischung bei 82°C 15.96 kg (34.13 mol) Kalium-te/f-butylat-Lösung in Tetrahydrofuran zugegeben und mit 10.0 L te/f-Amylalkohol spülen. Nach einer Stunde Rühren bei 82°C wurden 16.59 kg (136.51 mol) Salzsäure (30%) zugeben. Nach beginnender Kristallisation ließ man 23 L Wasser zulaufen und kühlte die Suspension auf 22°C ab. Anschließend wurde die Suspension 16 Stunden bei 22°C gerührt, das erhaltene Produkt abzentrifugiert und getrocknet.
Ausbeute: 17.70 kg (93% der Theorie) ee-Wert: 99.9%

Beispiel 5.2: (1 /?)-4-(1 ,2,4,5-Tetrahydro-2-oxo-3H-1 ,3-benzodiazepin-3-yl)-1 -car- boxy-2-[3,5-dimethyl-4-(phenylmethoxy)phenyl]-1 -piperidincarbon- säureethylester (J)
30.00 kg (93.07 mol) ) (α/?)-α-Hydroxy-3,5-dimethyl-4-(phenylmethoxy)-phenylpropion- säure-Mononatriumsalz (H) wurden vorgelegt, 33.17 kg (97.72 mol) 1-(1 H-lmidazol-1-yl- carbonyl)-4-(1 ,2,4,5-tetrahydro-2-oxo-3/-/-1 ,3-benzodiazepin-3-yl)-piperidin (I) zugegeben und diese Mischung in 510.0 L te/f-Amylalkohol suspendiert. 120.0 L Lösungsmittel wurden anschließend bei Normaldruck abdestilliert. Danach wurden zur Reaktionsmischung bei 82°C 57.44 kg (102.38 mol) Kalium-te/f-butylat-Lösung in Tetrahydrofuran zugegeben und mit 30.0 L te/f-Amylalkohol spülen. Nach 1.5 Stunden Rühren bei 82°C wurden 49.77 kg (409.52 mol) Salzsäure (30%) zugeben. Nach beginnender Kristallisation ließ man 69.0 L Wasser zulaufen und kühlte die Suspension auf 22°C ab.
Anschließend wurde die Suspension mindestens 2 Stunden bei 22°C gerührt, das erhaltene Produkt abzentrifugiert und getrocknet. Ausbeute: 47.88 kg (90% der Theorie) ee-Wert: 99.9% Chemische Reinheit (HPLC): 98.7% Schmelzpunkt: 132°C
Beispiel 5.3: (1 /?)-4-(1 ,2,4,5-Tetrahydro-2-oxo-3H-1 ,3-benzodiazepin-3-yl)-1 -car- boxy-2-[3,5-dimethyl-4-(phenylmethoxy)phenyl]-1 -piperidincarbon- säureethylester (J)
55.4 kg (172.0 mol) ) (αR)-α-Hydroxy-3,5-dimethyl-4-(phenylmethoxy)-phenylpropion- säure-Mononatriumsalz (H) wurden vorgelegt, 64.2 kg (189.4 mol) 1 -(1 H-lmidazol-1 -yl- carbonyl)-4-(1 ,2,4,5-tetrahydro-2-oxo-3/-/-1 ,3-benzodiazepin-3-yl)-piperidin (I) zugegeben und diese Mischung in 149 kg Λ/,Λ/-Dimethylformamid suspendiert. Danach wurden bei 700C zur Reaktionsmischung 11 1 kg (198.2 mol) einer 20%igen Kalium-te/f-butylat- Lösung in Tetrahydrofuran zugegeben. Die entstandene Lösung wurde 30 min nachgerührt und anschließend sukzessive mit 23 kg Λ/,Λ/-Dimethylformamid, 192 L Wasser und 100 kg Isopropanol verdünnt. Anschließend wurden bei 65 0C 66.2 kg (653.6 mol) Salzsäure (36%) zugeben. Nach Animpfung bei gleicher Temperatur wurde die Mischung innerhalb von 2.5 h schrittweise auf 200C abgekühlt und weitere 60 min gerührt. Das erhaltene Produkt wurde abzentrifugiert und getrocknet. Ausbeute: 96.6 kg (98% der Theorie) ee-Wert: 99.7% Chemische Reinheit (HPLC): 99.5%
Beispiel 6.1 : 4-[1 -(Phenylmethyl)-4-piperidinyl]-morpholin-Dihydrochlorid (M)
(K) (L) (M)
10.00 kg (52.84 mol) 1-Benzylpiperidon (K) wurden vorgelegt und 9.21 kg (105.68 mol) Morpholin (L) sowie 30.0 L Isopropylacetat zugegeben. Anschließend wurde die erhaltene Reaktionsmischung am Wasserabscheider auf Rückflusstemperatur erhitzen (95°C) und solange gerührt, bis sich kein Wasser mehr abtrennte. Anschließend wurde bei 4 bar und einer Innentemperatur von 600C in Gegenwart von 0.50 kg Raney-Nickel Katalysator hydriert. Danach wurde das Lösungsmittel im Vakuum vollständig abdestilliert. Anschließend wurden 45.0 L Ethanol zugesetzt und 10.87 kg (116.25 mol) ethanolische 10-molare Salzsäure zudosiert. Die entstandene Suspension wurde auf 200C abgekühlt und 30 Minuten bei 20°C gerührt. Das Produkt wurde abzentrifugiert und getrocknet. Ausbeute: 15.1 kg (86% der Theorie)

Beispiel 6.2: 4-[1 -(Phenylmethyl)-4-piperidinyl]-morpholin-Dihydrochlorid (M)

10.00 kg (52.84 mol) 1-Benzylpiperidon (K) wurden vorgelegt und 9.21 kg (105.68 mol) Morpholin (L) sowie 50.0 L Toluol zugegeben. Anschließend wurde die erhaltene Reaktionsmischung am Wasserabscheider auf Rückflusstemperatur erhitzen (1100C) und solange gerührt, bis sich kein Wasser mehr abtrennte. Anschließend wurde bei 4 bar und einer Innentemperatur von 600C in Gegenwart von 0.50 kg Raney-Nickel Katalysator in 6 L Ethanol hydriert. Der Katalysator wurde abgetrennt und mit 20 L Ethanol gewaschen. Danach wurde das Lösungsmittel im Vakuum vollständig abdestilliert. Anschließend wurden 80.0 L Ethanol zugesetzt und bei 50°C wurden 16.05 kg (132.10 mol) Salzsäure (30%) zudosiert. Die entstandene Suspension wurde auf 20°C abgekühlt und 30 Minuten bei 200C gerührt. Das Produkt wurde abzentrifugiert und getrocknet. Ausbeute: 15.5 kg (88% der Theorie)

Beispiel 7: 4-(4-Piperidinyl)-morpholin (N)

10.00 kg (30.00 mol) 4-[1-(Phenylmethyl)-4-piperidinyl]-morpholin-Dihydrochlorid (M) wurden in 50.0 L te/f-Butanol und 10.0 L Wasser suspendiert und auf 45°C erwärmt. Bei 45°C wurde eine Lösung aus 10.37 kg (75.01 mol) Kaliumcarbonat in 24.0 L Wasser zudosiert. Anschließend wurde bei 45°C die wässrige Phase abgetrennt und im Vakuum 20.0 L Lösungsmittel abdestilliert. Danach wurde bei einem Druck von 4 bar und einer Innentemperatur von 600C solange hydriert, bis keine Wasserstoffaufnahme mehr zu verzeichnen war. Nach vollständiger Reaktion wurde der Katalysator abfiltriert und mit 20.0 L te/f-Butanol gewaschen. Die erhaltene Hydrierlösung wurde anschließend in einen Reaktor übergeführt und im Vakuum das Lösungsmittel vollständig abdestilliert. Zurück bleibt ein Öl, das nach kurzer Zeit kristallisiert.
Ausbeute: 5.53 kg (88% der Theorie)

Die entstandene Suspension wurde auf 200C abgekühlt und 30 Minuten bei 20°C nachgerührt. Das Produkt wurde abzentrifugiert und getrocknet. Ausbeute: 1 1.46 kg (80% der Theorie)
Beispiel 9: 4-(4-Piperidinyl)-morpholin Hydrochlorid (O)

30.00 kg (90.01 mol) 4-[1-(Phenylmethyl)-4-piperidinyl]-morpholin-Dihydrochlorid (M) wurden in 67.5 L Methanol und 20.0 L Wasser gelöst. Danach wurde bei einem Druck von 4 bar und einer Innentemperatur von 500C in Gegenwart von 1.80 kg Palladium auf Kohle solange hydriert, bis keine Wasserstoffaufnahme mehr zu verzeichnen war. Nach
vollständiger Reaktion wurde der Katalysator abfiltriert und mit einer Mischung aus 18.0 L Methanol und 2.0 L Wasser gewaschen. Die erhaltene Hydrierlösung wurde anschließend in einen Reaktor übergeführt und auf 600C erhitzt. Bei 600C wurden 540.0 L Ethanol zudosiert. Die entstandene Suspension wurde auf 5°C abgekühlt. Das Produkt wurde abzentrifugiert und getrocknet.
Ausbeute: 19.69 kg (90% der Theorie) Chemische Reinheit (GC): 99.8%
Beispiel 10.1 : (1 /?)-4-(1 ,2,4,5-Tetrahydro-2-oxo-3H-1 ,3-benzodiazepin-3-yl)-1 -[[3,5-di- methyl-4-hydroxy)phenyl]methyl]-2-[4-(4-morpholinyl)-1 -piperidinyl]-
1 -piperidincarbonsäure-2-oxoethylester-Tartrat (P)
10.00 kg (17.49 mol) (1 R)-4-(1 !2,4!5-Tetrahydro-2-oxo-3/-/-1 ,3-benzodiazepin-3-yl)-1-car- boxy-2-[3,5-dimethyl-4-(phenylmethoxy)phenyl]-1 -piperidincarbonsäureethylester (J) wurden vorgelegt und 5.10 kg (20.99 mol) 4-(4-Piperidinyl)-morpholin-Dihydrochlorid (O) sowie 85.0 L Ethylacetat zugeben. Bei 200C wurden der erhaltenen Reaktionsmischung 8.85 kg (87.45 mol) Triethylamin zugesetzt und anschließend mit 5.0 L Ethylacetat gespült. Innerhalb von 30 Minuten wurden 17.81 kg (27.99 mol) 50%iges Propan- phosphonsäureanhydrid zugetropft. Nach weiteren 30 Minuten Reaktionszeit wurden 25.0 L Wasser zugegeben. Die wässrige Phase wurde abtrennt und eine Lösung aus 3.63 kg (26.24 mol) Kaliumcarbonat in 25.0 L Wasser zugegeben. Die wässrige Phase wurde abgetrennt und bei Normaldruck 85.0 L Lösungsmittel abdestilliert. Anschließend wurden 1 18.0 L Ethanol und 0.65 kg Palladium/Kohle (10%) zugesetzt. Anschließend wurde bei einem Druck von 3 bar und einer Innentemperatur von 40°C solange hydriert, bis keine Wasserstoffaufnahme mehr zu verzeichnen war. Nach vollständiger Reaktion wurde der Katalysator abfiltriert und mit 20.0 L Ethanol gewaschen. Bei Normaldruck wurden 43.0 L Lösungsmittel abdestilliert und anschließend bei 75°C 35.0 L Isopropylalkohol zugeben. Die Lösung wurde über einen Druckfilter klarfiltriert, mit
10.0 L Isopropylalkohol gespült und die Reaktionsmischung dann auf 65°C abgekühlt. Die Lösung wurde mit 10 g Impfkristallen versetzen. Anschließend wurde eine Lösung aus 2.89 kg (19.24 mol) L (+)-Weinsäure in 3.0 L Wasser zugegeben und mit 5.0 L Isopropylalkohol nachgespült. Nach 1 Stunde Rühren bei 65°C wurde die Suspension auf Raumtemperatur abgekühlt, das Produkt filtriert und getrocknet. Ausbeute: 10.97 kg (80% der Theorie)

Beispiel 10.2: (1 /?)-4-(1 ,2,4,5-Tetrahydro-2-oxo-3H-1 ,3-benzodiazepin-3-yl)-1 -[[3,5-di- methyl-4-hydroxy)phenyl]methyl]-2-[4-(4-morpholinyl)-1 -piperidinyl]- 1 -piperidincarbonsäure-2-oxoethylester-Tartrat (P)
30.00 kg (52.48 mol) (1 R)-4-(1 !2,4!5-Tetrahydro-2-oxo-3/-/-1 ,3-benzodiazepin-3-yl)-1-car- boxy-2-[3,5-dimethyl-4-(phenylmethoxy)phenyl]-1-piperidincarbonsäureethylester (J) wurden vorgelegt und 15.31 kg (62.97 mol) 4-(4-Piperidinyl)-morpholin-Dihydrochlorid (O) sowie 210.0 L Ethylacetat zugeben. Bei 400C wurden der erhaltenen Reaktionsmischung 26.55 kg (262.39 mol) Triethylamin zugesetzt und anschließend mit 15.0 L Ethylacetat gespült. Innerhalb von 30 Minuten wurden bei 400C 53.43 kg (83.97 mol) 50%iges Propanphosphonsäureanhydrid zugetropft. Nach weiteren 2 Stunden Reaktionszeit wurden 60.0 L Wasser zugegeben. Die wässrige Phase wurde abtrennt und eine Lösung aus 10.88 kg (78.72 mol) Kaliumcarbonat in 60.0 L Wasser zugegeben. Die wässrige Phase wurde abgetrennt und die organische Phase nochmals mit 60 L Wasser gewaschen. Danach wurde im Vakuum 210.0 L Lösungsmittel abdestilliert. Anschließend wurden 90.0 L Ethanol und 1.95 kg Palladium/Kohle (10%) zugesetzt. Anschließend wurde bei einem Druck von 3 bar und einer Innentemperatur von 400C solange hydriert,
bis keine Wasserstoffaufnahme mehr zu verzeichnen war. Nach vollständiger Reaktion wurde der Katalysator abfiltriert und mit 35.0 L Isopropanol gewaschen. Die Lösung wurde über einen Druckfilter klarfiltriert, mit 10.0 L Isopropylalkohol gespült und die Reaktionsmischung dann auf 65°C erwärmt. Die Lösung wurde mit 30 g Impf- kristallen versetzen. Anschließend wurde eine Lösung aus 9.06 kg (60.35 mol) L (+)- Weinsäure in 9.0 L Wasser zugegeben und mit 7.5 L Isopropylalkohol nachgespült. Nach 1 Stunde Rühren bei 65°C wurde die Suspension innerhalb von 1.5 Stunden auf 5°C abgekühlt, 1 Stunde nachgerührt, das Produkt filtriert und getrocknet. Ausbeute: 34.80 kg (85% der Theorie) Chemische Reinheit (HPLC): 99.4% ee-Wert: 99.6%

Beispiel 10.3: (1 /?)-4-(1 ,2,4,5-Tetrahydro-2-oxo-3H-1 ,3-benzodiazepin-3-yl)-1 -[[3,5-di- methyl-4-hydroxy)phenyl]methyl]-2-[4-(4-morpholinyl)-1 -piperidinyl]-
1 -piperidincarbonsäure-2-oxoethylester-Tartrat (P)
32.6 kg (133.9 mol) 4-(4-Piperidinyl)-morpholin-Dihydrochlorid (O) wurden in 290 kg Toluol vorgelegt, mit 56.4 kg (557.4 mol) Triethylamin versetzt und bei 85°C für 60 min gerührt. Die Suspension wurde auf 300C abgekühlt und mit 61.2 kg (107.1 mol) (1 R)-4- (1 ,2,4,5-Tetrahydro-2-oxo-3/-/-1 ,3-benzodiazepin-3-yl)-1 -carboxy-2-[3,5-dimethyl-4- (phenylmethoxy)phenyl]-1-piperidincarbonsäureethylester (J) versetzt. Anschließend wurden 105.4 kg (165.6 mol) 50%iges Propanphosphonsäureanhydrid in Ethylacetat innerhalb von 30 Minuten bei 400C zugetropft und mit 26 kg Toluol nachgespült. Nach weiteren 3 Stunden Reaktionszeit wurden sukzessive 245 L Wasser und 98 kg 2-Butanol zugegeben. Die wässrige Phase wurde bei 50 °C abtrennt und die organische Phase mit 18.4 kg (183.6 mol) Kaliumhydrogencarbonat sowie 245 L Wasser versetzt. Nach erneuter Phasentrennung bei 50 0C und einer weiteren Extraktion mit 184 L Wasser
wurde die organische Phase mit 3.1 kg Norit SX Ultra und 33 L Ethanol versetzt und anschließend filtriert. Der Lösung wurden 33 L Ethanol und 3.1 kg Palladium/Kohle (10%) zugesetzt. Anschließend wurde bei einem Druck von 4 bar und einer Innentemperatur von max. 500C solange hydriert, bis keine Wasserstoffaufnahme mehr zu verzeichnen war. Nach vollständiger Reaktion wurde der Katalysator abfiltriert und mit 44 L Ethanol gewaschen. Die Lösung wurde bei 60 0C im Vakuum bis zu einem Drittel des Ursprungsvolumens aufkonzentriert, mit 98 kg Ethanol versetzt und über einen Druckfilter klarfiltriert. Das Filtrat wurde bei 65 0C mit 242 kg Isopropylalkohol verdünnt. Anschließend wurde eine Lösung von 18.7 kg (124.5 mol) L (+)-Weinsäure in 22 L Wasser zugesetzt und mit 49 kg Ethanol nachgespült. Die Reaktionsmischung wurde bei 65 0C angeimpft, 45 min bei dieser Temperatur und anschließend bei 20 0C weitere 2 h gerührt. Abschließend wurde das Produkt filtriert, getrocknet und mittels befeuchtetem Stickstoffstrom auf einen definierten Wassergehalt äquilibriert. Ausbeute: 80.8 kg (94 % der Theorie)

Beispiel 11 : (3Z)-3-(4-terf-Butyloxycarbonyloxy-3,5-dimethyl-phenyl)-2-tetrahydro- pyran-2-yloxy)-acrylsäure-monodicyclohexylaminsalz (S)
20.00 g (79.91 mmol) 4-te/f-Butyloxycarbonylyloxy-3,5-dimethylbenzaldehyd (R) wurden in 120.0 ml 2-Methyltetrahydrofuran gelöst und 31.04 g (194.23 mmol) Benzyloxy-essig- säureethylester (Q) zugegeben. Bei 200C wurden 26.9 g (95.90 mol) Kalium-te/f-amylat (45%ig in Tetrahydrofuran) zugetropft. Anschließend wurde die erhaltene Reaktionsmischung 2 Stunden bei 20°C gerührt und nach vollständiger Umsetzung mit 30 ml Wasser und 16.0 g Natriumhydroxid-Lösung (50%) (199.8 mmol) versetzt. Die Reaktionsmischung wurde zum Rückfluss erhitzt und 1 Stunde gerührt. Nach Neutralisation mit
Essigsäure und Phasentrennung wurde die organische Phase im Vakuum eingeengt. Der Rückstand wurde mit 80 ml Toluol und 250 ml n-Heptan versetzt. Nach Zugabe von 30.0 g (165.45 mmol) Dicyclohexylamin wurde die erhaltene Suspension 20 Stunden bei Raumtemperatur gerührt. Anschließend wurde das Produkt filtriert und getrocknet. Ausbeute: 1 1.47 g (24.8% der Theorie)

1. Verfahren zur Herstellung von Verbindungen der allgemeinen Formel I
in der
R1 H, d-3-Alkyl, C(O)-O-Benzyl, C(O)-O-terf-Butyl oder Benzyl darstellt und
R2 ein sekundäres Amin -NR2 1R22 bedeutet, wobei
R2 1 und R22 unabhängig voneinander ausgewählt sein können aus der Gruppe bestehend aus d-3-Alkyl und Benzyl, oder
die Gruppe -NR2 1R22 zusammen ein cyclisches Amin bildet, das ausgewählt sein kann aus der Gruppe bestehend aus Morpholin-4-yl-, 1-Methylpiperazin-4-yl-, 1 -Benzylpiperazin-4-yl-, 1 -(Ci_3-Alkylcarbonyl)-piperazin-4-yl, 1 -(te/f-Butyloxycarbonyl)- piperazin-4-yl, 1-(Benzyloxycarbonyl)-piperazin-4-yl, Piperidin-1-yl- und Pyrrolidin-1-yl-,
umfassend die Schritte:
(a) Umsetzung eines Glykolsäureethylester der allgemeinen Formel Il o

2. Verfahren gemäß Anspruch 1 , dadurch gekennzeichnet, dass R1 ein Wasserstoffatom oder eine Benzylgruppe bedeutet.
3. Verfahren gemäß Anspruch 1 , dadurch gekennzeichnet, dass R2 ein sekundäres Amin -NR2 1R22 bedeutet, wobei die Gruppe -NR21R22 zusammen Morpholin-4-yl darstellt.
4. Verfahren gemäß Anspruch 1 , dadurch gekennzeichnet, dass R3 eine Ethylgruppe darstellt.

15. Verfahren zur Herstellung von Verbindungen der allgemeinen Formel XIII
R2 ( NH x n HCl w , (XIII) in der n eine der Ziffern 1 , 2 oder 3 und
R2 ein sekundäres Amin -NR2 1R22 bedeutet, wobei
R2 1 und R22 unabhängig voneinander ausgewählt sein können aus der Gruppe bestehend aus d-3-Alkyl und Benzyl, oder
die Gruppe -NR2 1R22 zusammen ein cyclisches Amin bildet, das ausgewählt sein kann aus der Gruppe bestehend aus Morpholin-4-yl-, 1-Methylpiperazin-4-yl-, 1-Benzylpiperazin-4-yl-, 1-(Ci-3-Alkylcarbonyl)-piperazin-4-yl, 1-(te/f-Butyloxycarbonyl)- piperazin-4-yl, 1-(Benzyloxycarbonyl)-piperazin-4-yl, Piperidin-1-yl- und Pyrrolidin-1-yl-,
umfassend die Schritte:
(a) Umsetzung von 1-Benzylpiperidon mit einem Amin der allgemeinen Formel XIV
H-NR2 1R2 2 , (XIV)
in der R2 1 und R22 wie voranstehend erwähnt definiert sind, in einem Lösungsmittel unter Wasserabspaltung;
(b) Reduzieren des unter (a) entstandenen Enamins unter Druck und in Gegenwart eines Katalysators und Isolieren des erhaltenen Produkts der allgemeinen Formel XV
in der n, R2 1 und R22 wie voranstehend erwähnt definiert sind, durch Zugabe von Salzsäure;
(c) Umsetzung einer unter (b) erhaltenen Verbindung der allgemeinen Formel XV, in der n, R2 1 und R22 wie voranstehend erwähnt definiert sind, mit einer Base in einem polaren Lösungsmittel, wodurch eine Verbindung der allgemeinen Formel XV entsteht, in der R2 1 und R22 wie voranstehend erwähnt definiert sind und n die Ziffer 0 darstellt;
(d) Entfernen der Benzyl-Schutzgruppe aus einer unter (c) erhaltenen Verbindung der allgemeinen Formel XV und Isolieren einer Verbindung der allgemeinen Formel XVI

in der R2 1 und R22 wie voranstehend erwähnt definiert sind und n die Ziffer 0 darstellt; und
(e) Umsetzung einer unter (d) erhaltenen Verbindung der allgemeinen Formel XVI, in der n die Ziffer 0 darstellt, durch Zugabe von Salzsäure in einem Lösungsmittel zu einer Verbindung der allgemeinen Formel XVI, in der n eine der Ziffern 1 , 2 oder 3 darstellt.
