Abstract
Die vorliegende Erfindung betrifft die Verwendung von mindestens einer Verbindung, gewählt aus einer Verbindung der Formel (1a, 1b), einem physiologisch verträglichen Salz davon und einer stereoisomeren Form davon, worin R<1> H oder Alkyl, R<2>, H, COOH, COO-Alkyl oder CO-NH-R<5>, R<3> und R<4> jeweils unabhängig voneinander H oder OH, n 1,2 oder 3, R<5> H, Alkyl, einen Aminosäurerest, Dipeptidrest oder Tripeptidrest, und Alkyl einen Alkylrest mit 1 bis 4 Kohlenstoffatomen bedeuten, zur Stabilisierung von p53. Diese Verbindungen werden erfindungsgemäss üblicherweise in Form einer topischen Zusammensetzung verwendet.
Description
Verwendung von Ectoin oder Ectoin-Derivaten zur Stabilisierung von p53
Die vorliegende Erfindung betrifft die Verwendung von Ectoin oder Ectoin-Derivaten zur Stabilisierung von p53.
In den westlichen Industrieländern hat sich Krebs zu einer der am meisten gefürchteten Krankheiten entwickelt. Dies liegt zum einen daran, dass für manche Krebsarten noch keine wirksame Therapie gefunden wurde oder dass der jeweilige Krebs entweder gegen Chemotherapie oder Strahlentherapie resistent geworden ist. Es wird davon ausgegangen, dass Krebs die Todesursache von 1/5 der Menschen in den westlichen Industrienationen ist.
Der Prozeß der Entstehung von Krebs wird als Cancerogenese bezeichnet. Nach den gegenwärtigen Vorstellungen entstehen die Zellen eines Tumors aus einer gemeinsamen Stammzelle (Klonalität). Der Prozess, in dem eine Zelle zur Krebszelle entartet, d.h. eine maligne Transformation auftritt, wird auf die Umgehung oder Störung der normalen Zellwachstumskontrolle zurückgeführt. Bei fehlerhaften Zellteilungen kann es zur Umordnung von Chromosomen oder Chromosomenteilen ohne Wiederherstellung durch zelleigene Reparaturmechanismen kommen. Dies hat die Aktivierung von vorhandenen Genen mit zentraler Bedeutung für die Regulation von Zellaktivitäten, sogenannten On- cogenen, zur Folge. Diese Oncogene führen zur Störung der Wachstumskontrolle, z.B. durch die Produktion von Wachstumsfaktoren, die wieder auf die Zelle stimulierend rückwirken. Die unkontrollierte Steigerung der Zellvermehrung lässt biochemische und physikalische Veränderungen entstehen, die zum weiteren Verlust von Wachstumshemmung (Autonomie), zu zellulären und histologischen Abnormitäten und zur Entdiffe- renzierung (Anaplasie) sowie Streuung über den gesamten Organismus (Metastasen) führen.
Als ursächliche Bedingungen für die Krebsentstehung sieht man genetische Faktoren, ionisierende Strahlen, UV-Licht, Viren und die Einwirkung von Carzinogenen in Form von Tabakrauch, Nahrung, Medikamenten oder durch Arbeitsplatz- oder umweltbedingte
Aufnahme an. Auch die mangelnde Abwehr von Tumorzellen infolge von Störungen des Immunsystems trägt zur Entstehung von Krebserkrankungen bei. Hier liegt auch der Ansatzpunkt psychophysiologischer Theorien, die die Beteiligung von Stress und psychischen Faktoren berücksichtigen.
Die Verbindung zwischen Cancerogenese und Mutagenese wurde für drei Krebsfaktoren festgestellt:
- chemische Carzinogene, die einfache lokale Veränderungen in der DNA-Sequenz verursachen;
- ionisierende Strahlung, die Chromosomenbrüche herbeiführen und Translokationen verursachen kann; und
- Viren, die fremde DNA in die Zellen einführen.
Es wird davon ausgegangen, dass die Zelle Mechanismen besitzt, um zu verhindern, dass die Fremd-DNA der Viren bzw. Bakterien an ihre Tochterzellen weitergegeben wird. Dies führte dazu, dass sich Krebsforscher den Tumor- oder Wachstumssuppres- sorgenen, insbesondere dem p53-Gen, zuwandten.
Das p53-Gen befindet sich auf dem kurzen Arm des menschlichen Chromosoms 17, Bande 13 und ist ungefähr 20 Kilobasen (kb) lang. Dieses Gen ergibt ein 2,8 kb mRNA- Transkript und codiert für ein 53 kD Phosphorprotein, welches 393 Aminosäuren enthält.
Das p53-Protein ist in der Lage, zu speziellen Sequenzen zu binden, und es wird angenommen, dass es ein Transkriptionsfaktor ist.
Das p53 reagiert auf DNA-Schäden oder abnorme Wachstumsbedingungen, indem es die Zelle in der G1 -Phase der DNA-Replikation (Ruhephase) arretieren kann. Durch DNA-Strangbrüche und AT-Genprodukte wird die p53-Proteinsynthese verstärkt. Die p53-Proteine verstärken ihrerseits die Bildung negativer Wachstumsfaktoren und hemmen die DNA-Replikation, positive Wachstumsfaktoren und GTP-Synthese via IMP- Dehydrogenase.
Ein weiterer Aspekt dieser Art der Regulierung beinhaltet die Kontrollfunktion von p53 in der Zelle, bei zu stark beschädigten Zellen den kontrollierten Zelltod oder Apoptosis herbeizuführen, bevor diese Zellen zu Tumoren heranwachsen können. Somit hat p53 eine bestimmte Rolle in der Zellantwort zur UV-Strahlung durch die Inhibition der DNA- Synthese, gefolgt von einer DNA-Schädigung.
Als am häufigsten zu beobachtende genetische Veränderung in menschlichen malignen Tumorzellen kommt es zu Veränderungen des p53-Gens und seines codierten Proteins. Insbesondere sind die Erbanlagen für p53 besonders häufig in Hauttumorzellen mutiert, wenn diese durch UV-Licht verursacht wurden. Eine übermäßige Sonnenstrahlung kann die Entstehung von Hauttumoren verursachen.
Eine Zelle, die p53-defizient ist oder mutiertes P53 exprimiert, tritt nicht in die G1- Arretierung oder die GO-Phase (Apoptosis) ein. Diese Unfähigkeit des mutierten p53 Apoptosis einzuleiten, kann ebenfalls erklären, warum die Strahlungstherapie in der Behandlung von verschiedenen Tumorzellen unwirksam ist.
Es gibt verschiedene Arten, wie die p53-Funktion deaktiviert werden kann. Dazu zählen Missense-Mutationen, Deletionen oder Nonsense-Mutationen des Gens, die dazu führen, dass das Protein nicht oligomerisiert oder tetramere Komplexe bilden kann, die zu speziellen DNA-Sequenzen binden können. Das p53-Gen besitzt mehrere "hot spots", d.h. Genbereiche, die schnell mutieren können. Weiterhin ist das p53-Gen sehr empfindlich gegenüber UV-Strahlen, welche leicht zu einer Mutation und dem damit verbundenen Funktionsverlust von p53 führen.
Da die Bedeutung von p53 insbesondere bei der Entstehung von Hauttumoren erkannt wurde, wurden bereits Vorschläge gemacht, wie die Wächterfunktion von p53 auch in Hautzellen funktionieren kann, in denen das p53-Gen durch starke Sonneneinstrahlung mutiert wurde. So wird in Bild der Wissenschaft 2/96, Seite 108 beschrieben, dass vorgeschlagen wurde, biotechnisch hergestelltes p53-Protein in eine Lipidhülle zu verpak- ken und so in die Hautzellen zu verfrachten. Dieses p53-Präparat in Form einer Creme soll potentielle Krebszellen ausmerzen und möglicherweise sogar vorhandenen Hautkrebs zurückbilden. Dieses Verfahren ist jedoch durch die Verwendung von p53 selbst sehr aufwendig und kostspielig.
Es ist daher die Aufgabe der vorliegenden Erfindung, eine Verbindung zur Verfügung zu stellen, die p53 auf DNA- und Protein-Ebene stabilisiert. Es soll somit eine Mutation und ein damit verbundener Funktionsverlust von p53 verringert werden, um die Apoptosis auch bei Tumorzellen einleiten zu können. Gleichzeitig soll die Synthese von p53 stimuliert werden, so dass unter Stressbedingungen, wie UV-Strahlung und chemischen Noxen, p53 in ausreichender Konzentration zur Verfügung steht.
Diese Aufgabe wird gelöst durch die Verwendung mindestens einer Verbindung, gewählt aus einer Verbindung der Formel 1a, 1b,

zur Stabilisierung von p53.
Die Verbindungen der Formeln 1a und 1b, die physiologisch verträglichen Salze der Verbindungen der Formeln 1a und 1b und die stereoisomere Form der Verbindungen der Formeln 1a und 1b werden nachstehend auch als „Ectoin oder Ectoin-Derivate" bezeichnet.
Bei Ectoin und den Ectoin-Derivaten handelt es sich um niedermolekulare, cyclische Aminosäurederivate, die aus verschiedenen halophilen Mikroorganismen gewonnen werden können. Sowohl Ectoin als auch Ectoin-Derivate besitzen den Vorteil, daß sie nicht in den Zellstoffwechsel eingreifen. Ectoin und Ectoin-Derivate werden bereits in der DE 43 42 560 als Feuchtigkeitsspender in Kosmetikprodukten beschrieben.
Die erfindungsgemäß verwendeten Verbindungen können in den topischen Zusammensetzungen als optische Isomere, Diastereomere, Racemate, Zwitterionen, Kationen oder als Gemisch derselben vorliegen.
Als erfindungsgemäß verwendete Verbindungen sind diejenigen bevorzugt, worin R1 H oder CH3, R2 H oder COOH, R3 und R4 jeweils unabhängig voneinander H oder OH und n 2 bedeuten. Von den erfindungsgemäß verwendeten Verbindungen sind (S)-1 ,4,5,6-Tetrahydro-2-methyl-4-pyrimidincarbonsäure (Ectoin) und (S,S)-1 ,4,5,6- Tetrahydro-5-hydroxy-2-methyl-4-pyrimidincarbonsäure (Hydroxyectoin) besonders bevorzugt.
Unter dem Begriff „Aminosäure" werden die stereoisomeren Formen, z.B. D- und L- Formen, folgender Verbindungen verstanden: Alanin, ß-Alanin, Arginin, Asparagin, As- paraginsäure, Cystein, Glutamin, Glutaminsäure, Glycin, Histidin, Isoleucin, Leucin, Ly- sin, Methionin, Phenylalanin, Serin, Threonin, Tryptophan, Tyrosin, Valin, γ-Aminobutyrat, Nε-Acetyllysin, Nδ-Acetylornithin, Nγ-Acetyldiaminobutyrat und Nα-Acetyldiaminobutyrat. L-Aminosäuren sind bevorzugt.
Aminosäurereste leiten sich von den entsprechenden Aminosäuren ab.
Die Reste folgender Aminosäuren sind bevorzugt: Alanin, ß-Alanin, Asparagin, Aspa- raginsäure, Glutamin, Glutaminsäure, Glycin, Serin, Threonin, Valin, γ-Aminobutyrat, Nε-Acetyllysin, Nδ-Acetylornithin, Nγ-Acetyldiaminobutyrat und Nα-Acetyldiaminobutyrat.
Die Di- und Tripeptidreste sind ihrer chemischen Natur nach Säureamide und zerfallen bei der Hydrolyse in zwei oder drei Aminosäuren. Die Aminosäuren in den Di- und Tri- peptidresten sind durch Amidbindungen miteinander verbunden. Bevorzugte Di- und Tripeptidreste sind aus den bevorzugten Aminosäuren aufgebaut.
Die Alkylgruppen umfassen die Methylgruppe CH3, die Ethylgruppe C2H5, die Propyl- gruppen CH2CH2CH3 und CH(CH3)2 sowie die Butylgruppen CH2CH2CH2CH3, H3CCHCH2CH3, CH2CH(CH3)2 und C(CH3)3. Die bevorzugte Alkylgruppe ist die Methylgruppe.
Bevorzugte physiologisch verträgliche Salze der erfindungsgemäß verwendeten Verbindungen sind beispielsweise Alkali-, Erdalkali- oder Ammoniumsalze, wie Na-, K-, Mg- oder Ca-Salze, sowie Salze, die von den organischen Basen Triethylamin oder Tris-(2- hydroxy-ethyl)amin abgeleitet sind. Weitere bevorzugte physiologisch verträgliche Salze der erfindungsgemäß verwendeten Verbindungen ergeben sich durch Umsetzung mit anorganischen Säuren, wie Salzsäure, Schwefelsäure und Phosphorsäure, oder mit organischen Carbon- oder Sulfonsäuren, wie Essigsäure, Citronensäure, Benzoesäure, Maleinsäure, Fumarsäure, Weinsäure und p-Toluolsulfonsäure.
Verbindungen der Formeln 1a und 1b, in denen basische und saure Gruppen, wie Car- boxyl- oder Aminogruppen, in gleicher Zahl vorliegen, bilden innere Salze.
Die Herstellung der erfindungsgemäß verwendeten Verbindungen wird in der
DE 4342 560 beschrieben. (S)-1 ,4,5,6-Tetrahydro-2-methyl-4-pyrimidin-carbonsäure oder (S,S)-1,4,5,6-Tetrahydro-5-hydroxy-2-methyl-4-pyrimidincarbonsäure können auch mikrobiologisch gewonnen werden (Severin et al., J. Gen. Microb. 138 (1992)
1629-1638).
Ectoin oder Ectoin-Derivate werden erfindungsgemäß üblicherweise in Form einer topischen Zusammensetzung verwendet. Möglich ist auch deren Verwendung im pharmazeutischen Bereich und/oder im Nahrungsmittelbereich.
Die Herstellung der topischen Zusammensetzung erfolgt, indem mindestens eine der erfindungsgemäß verwendeten Verbindungen, gegebenenfalls mit Hilfs- und/oder Trägerstoffen, in eine geeignete Formulierungsform gebracht werden. Die Hilfs- und Trägerstoffe stammen aus der Gruppe der Trägermittel, Konservierungsstoffe und anderer üblicher Hilfsstoffe.
Die topischen Zusammensetzung auf der Grundlage mindestens einer erfindungsgemäß verwendeten Verbindung wird äußerlich auf der Haut oder den Hautadnexen angewendet.
Als Anwendungsform seien z.B. genannt: Lösungen, Suspensionen, Emulsionen, Pasten, Salben, Gele, Cremes, Lotionen, Puder, Seifen, tensidhaltige Reinigungspräparate, Öle und Sprays. Zusätzlich zu einer oder mehreren erfindungsgemäß verwendeten Verbindungen werden der Zusammensetzung beliebige übliche Trägerstoffe, Hilfsstoffe und gegebenenfalls weitere Wirkstoffe zugesetzt.
Bevorzugte Hilfsstoffe stammen aus der Gruppe der Konservierungsstoffe, Antioxidan- tien, Stabilisatoren, Lösungsvermittler, Vitamine, Färbemittel und Geruchsverbesserer. Salben, Pasten, Cremes und Gele können neben einer oder mehreren erfindungsgemäß verwendeten Verbindungen die üblichen Trägerstoffe enthalten, z.B. tierische und pflanzliche Fette, Wachse, Paraffine, Stärke, Traganth, Cellulosederivate, Polyethylen- glykole, Silicone, Bentonite, Kieselsäure, Talkum und Zinkoxid oder Gemische dieser Stoffe.
Puder und Sprays können neben einer oder mehreren erfindungsgemäß verwendeten Verbindungen die üblichen Trägerstoffe enthalten, z.B. Milchzucker, Talkum, Kieselsäure, Aluminiumhydroxid, Calciumsilikat und Polyamid-Pulver oder Gemische dieser Stoffe. Sprays können zusätzlich die üblichen Treibmittel, z.B. Chlorfluorkohlenwasserstoffe, Propan/Butan oder Dimethylether.
Lösungen und Emulsionen können neben einer oder mehreren erfindungsgemäß verwendeten Verbindungen die üblichen Trägerstoffe, wie Lösungsmittel, Lösungsvermittler und Emulgatoren, z.B. Wasser, Ethanol, Isopropanol, Ethylcarbonat, Ethylacetat, Ben- zylalkohol, Benzylbenzoat, Propylenglykol, 1,3-Butylglykol, Öle, insbesondere Baumwollsaatöle, Erdnußöl, Maiskeimöl, Olivenöl, Rizinusöl und Sesamöl, Glycerinfettsäure- ester, Polyethylenglykole und Fettsäureester des Sorbitans oder Gemische dieser Stoffe, enthalten.
Suspensionen können neben einer oder mehreren erfindungsgemäß verwendeten Verbindungen die üblichen Trägerstoffe, wie flüssige Verdünnungsmittel, z.B. Wasser, Ethanol oder Propylenglykol, Suspendiermittel, z.B. ethoxylierte Isostearylalkohole, Polyoxyethylensorbitester und Polyoxyethylensorbitanester, mikrokristalline Cellulose, Aluminiummetahydroxid, Bentonit, Agar-Agar und Traganth oder Gemische dieser Stoffe, enthalten.
Seifen können neben einer oder mehreren erfindungsgemäß verwendeten Verbindungen die üblichen Trägerstoffe, wie Alkalisalze von Fettsäuren, Salze von Fettsäurehalbestern, Fettsäureeiweißhydrolysaten, Isothionate, Lanolin, Fettalkohol, Pflanzenöle, Pflanzenextrakte, Glycerin, Zucker oder Gemische dieser Stoffe, enthalten.
Tensidhaltige Reinigungsprodukte können neben einer oder mehreren erfindungsgemäß verwendeten Verbindungen die üblichen Trägerstoffe, wie Salze von Fettalkoholsulfaten, Fettalkoholethersulfaten, Sulfobernsteinsäurehalbestern, Fettsäureeiweißhydrolysaten, Isothionaten, Imidazoliniumderivate, Methyltaurate, Sarkosinate, Fettsäureamidether- sulfate, Alkylamidobetaine, Fettalkohole, Fettsäureglyceride, Fettsäurediethanolamide, pflanzliche und synthetische Öle, Lanolinderivate, ethoxylierte Glycerinfettsäureester oder Gemische dieser Stoffe, enthalten.
Gesichts- und Körperöle können neben einer oder mehreren erfindungsgemäß verwendeten Verbindungen die üblichen Trägerstoffe, wie synthetische Öle, wie Fettsäureester, Fettalkohole, Silikonöle, natürliche Öle, wie Pflanzenöle und ölige Pflanzenauszüge, Paraffinöle, Lanolinöle oder Gemische dieser Stoffe, enthalten.
Weitere typisch kosmetische Anwendungsformen sind auch Lippenstifte, Lippenpflege- stifte, Mascara, Eyeliner, Lidschatten, Rouge, Puder-, Emulsions- und Wachs-Make up sowie Sonnenschutz-, Prä-Sun- und After-Sun-Präparate.
Mindestens eine erfindungsgemäß verwendete Verbindung liegt in der topischen Zusammensetzung in einer Menge von vorzugsweise 0,0001 bis 50 Gew.-%, besonders bevorzugt 0,001 bis 10 Gew.-%, insbesondere bevorzugt 0,1 bis 1 Gew.%, bezogen auf die Zusammensetzung, vor.
Vorzugsweise werden neben Ectoin oder den Ectoin-Derivaten zusätzlich mindestens ein Antioxidationsmittel und/oder UV-Filter verwendet.
Es können erfindungsgemäß die aus der Fachliteratur bekannten Antioxidationsmittel verwendet werden, z.B. Flavonoide, Coumaranone, Aminosäuren (z.B. Glycin, Histidin, Tyrosin, Tryptophan) und deren Derivate, Imidazole, (z.B. Urocaninsäure) und deren Derivate, Peptide, wie D,L-Carnosin, D-Carnosin, L-Carnosin und deren Derivate (z.B. Anserin), Carotinoide, Carotine (z.B. α-Carotin, ß-Carotin, Lycopin) und deren Derivate, Chlorogensäure und deren Derivate, Liponsäure und deren Derivate (z.B. Dihydrolipon- säure), Aurothioglucose, Propylthiouracil und andere Thiole (z.B. Thioredoxin, Glutathion, Cystein, Cystin, Cystamin und deren Glycosyl-, N-Acetyl-, Methyl-, Ethyl-, Propyl-, Amyl-, Butyl- und Lauryl-, Palmitoyl-, Oleyl-, γ-Linoleyl, Cholesteryl- und Glyce- rylester) sowie deren Salze, Diaurylthiodipropionat, Distearylthiodipropionat, Thiodipro- piosäure und deren Derivate (Ester, Ether, Peptide, Lipide, Nukleotide, Nukleoside und Salze) sowie Sulfoximinverbindungen (z.B. Buthioninsulfoximine, Homocysteinsulfoxi- min, Buthioninsulfone, Penta-, Hexa-, Heptathioninsulfoximin), ferner (Metall-) Chelato- ren (z.B. -Hydroxyfettsäuren, Palmitinsäure, Phytinsäure, Lactoferrin), α-Hydroxy- säuren (z.B. Citronensäure, Milchsäure, Äpfelsäure), Huminsäure, Gallensäure, Gallenextrakte, Bilirubin, Biliverdin, EDTA, EGTA und deren Derivate, ungesättigte Fettsäuren und deren Derivate, Vitamin C und Derivate (z.B. Ascorbylpalmitat, Magnesium- Ascorbyl-phosphat, Ascorbylacetat) sowie Koniferylbenzoat des Benzoeharzes, Rutin- säure und deren Derivate, -Glycosylrutin, Ferulasäure, Furfurylidenglucitol, Camosin, Butylhydroxyltoluol (BHT), Butylhydroxyanisol, Nordohydroguajaretsäure, Trihydroxybu- tyrophenon, Harnsäure und deren Derivate, Mannose und deren Derivate, Zink und
dessen Derivate (z.B. ZnO, ZnSO4), Selen und dessen Derivate (z.B. Selenmethionin), Stilbene und deren Derivate (z.B. Stilbenoxid, trans-Stilbenoxid).
Mischungen von Antioxidationsmitteln sind ebenfalls geeignet. Bekannte und käufliche Mischungen sind beispielsweise Mischungen, enthaltend als aktive Inhaltsstoffe Lecithin, L-(+)-Ascorbylpalmitat und Zitronensäure (z.B. Oxynex® AP), natürliche Tocophe- role, L-(+)-Ascorbylpalmitat, L-(+)-Ascorbinsäure und Zitronensäure (z.B. Oxynex® K LIQUID), Tocopherolextrakte aus natürlichen Quellen, L-(+)-Ascorbylpalmitat, L-(+)- Ascorbinsäure und Zitronensäure (z.B. Oxynex® L LIQUID), DL-α-Tocopherol, L-(+)- Ascorbylpalmitat, Zitronensäure und Lecithin (z.B. Oxynex® LM) oder Butylhydroxytoluol (BHT), L-(+)-Ascorbylpalmitat und Zitronensäure (z.B. Oxynex® 2004).
In einer bevorzugten Ausführungsform der Erfindung wird als Antioxidationsmittel Butylhydroxytoluol verwendet. In einer weiteren bevorzugten Ausführungsform wird als Antioxidationsmittel eine oder mehrere Verbindungen, ausgewählt aus Flavonoiden und/oder Courmaranonen, verwendet.
Als Flavanoide werden die Glycoside von Flavanonen, Flavonen, 3-Hydroxyflavonen (= Flavanolen), Auronen, Isoflavonen und Rotenoiden aufgefaßt (Römpp Chemie Lexikon, Band 9, 1993). Im Rahmen der vorliegenden Erfindung werden hierunter jedoch auch die Aglykone, d.h. die zuckerfreien Bestandteile, und die Derivate der Flavonoide und der Aglykone verstanden. Im Rahmen der vorliegenden Erfindung werden unter Coumaranonen auch deren Derivate verstanden.
Bevorzugte Flavonoide leiten sich von Flavanonen, Flavonen, 3-Hydroxyflavonen, Auronen und Isoflavonen, insbesondere von Flavanonen, Flavonen, 3-Hydroxyflavonen und Auronen, ab.

Die Hydroxyalkoxygruppen sind vorzugsweise linear und besitzen 2 bis 12, vorzugsweise 2 bis 8 C-Atome. Diese Gruppen entsprechen somit der Formel -O-(CH2)n-OH, wobei n 2,3,4,5,6,7 oder 8, insbesondere 2 bis 5 und besonders bevorzugt 2 bedeutet.
Die Mono- und Oligoglycosidreste sind vorzugsweise aus 1 bis 3 Glycosideinheiten aufgebaut. Vorzugsweise werden diese Einheiten ausgewählt aus der Gruppe der Hexosyl- reste, insbesondere der Rhamnosylreste und Glucosylreste. Aber auch andere Hexosyl- reste, beispielsweise Allosyl, Altrosyl, Galactosyl, Gulosyl, Idosyl, Mannosyl und Talosyl, sind gegebenenfalls vorteilhaft zu verwenden. Es kann auch erfindungsgemäß vorteilhaft sein, Pentosylreste zu verwenden.

Methoxy, Ethoxy oder 2-Hydroxyethoxy.
In einer weiteren bevorzugten Ausführungsform, insbesondere, wenn die Wasserlöslichkeit der Flavonoide und Coumaranone gesteigert werden soll, ist an die Hydroxyguppen eine Sulfat- oder Phosphatgruppe gebunden. Geeignete Gegenionen sind beispielsweise die Ionen der Alkali- oder Erdalkalimetalle, wobei diese z.B. aus Natrium oder Kalium ausgewählt werden.
In einer weiteren bevorzugten Ausführungsform werden die Flavonoide ausgewählt aus folgenden Verbindungen: 4,6,3',4'-Tetrahydroxyauron, Quercetin, Rutin, Isoquercetin, Anthocyanidin (Cyanidin), Eriodictyol, Taxifolin, Luteolin, Trishydroxyethylquercetin (Troxequercetin), Trishydroxyethylrutin (Troxerutin), Trishydroxyethylisoquercetin (Troxeisoquercetin), Trishydroxyethylluteolin (Troxeluteolin) sowie deren Sulfaten und Phosphaten.
Unter den Flavonoiden sind insbesondere Rutin und Troxerutin bevorzugt. Besonders bevorzugt ist Troxerutin.
Unter den Coumaranonen ist 4,6,3',4'-Tetrahydroxybenzylcoumaranon-3 bevorzugt.
Die Antioxidationsmittel werden erfindungsgemäß in üblichen Mengen in der topischen Zusammensetzung verwendet.
Weiterhin können erfindungsgemäß die aus der Fachliteratur bekannten UV-Filter verwendet werden.
Als geeignete organische UV-Filter kommen alle dem Fachmann bekannten UVA- als auch UVB-Filter in Frage. Für beide UV-Bereiche gibt es viele aus der Fachliteratur bekannte und bewährte Substanzen, z.B. Benzylidenkampferderivate, wie
- 3-(4'-Methylbenzyliden)-dl-kampfer (z.B. Eusolex® 6300),
- 3-Benzylidenkampfer (z.B. Mexoryl® SD),

Diese organischen UV-Filter werden in der Regel in einer Menge von 0,5 bis 10 Gew.%, vorzugsweise 1 bis 8 Gew.%, in der erfindungsgemäß verwendeten topischen Zusammensetzung eingesetzt.
Weitere geeignete organische UV-Filter sind z.B.
- 2-(2H-Benzotriazol-2-yl)-4-methyl-6-(2-methyl-3-(1 ,3,3,3-tetramethyl-1- (trimethylsilyloxy)disiloxanyl)propyl)phenol (z.B. Silatrizole®),
- 4,4'-[(6-[4-((1 , 1 -Dimethylethyl)aminocarbonyl)phenylamino]-1 ,3,5-triazin- 2,4-diyl)diimino]bis(benzoesäure-2-ethylhexylester) (z.B. Uvasorb® HEB),
- -(Trimethylsilyl)-ω[trimethylsilyl)oxy]poly[oxy(dimethyl] [und ca. 6% methyl[2-[p-[2,2-bis(ethoxycarbonyl]vinyl]phenoxy]-1-methylenethyl] und ca. 1 ,5% methyl[3-[p-[2,2-bis(ethoxycarbonyl)vinyl)phenoxy)- propenyl) und 0,1 bis 0,4% (methylhydrogenjsilylen]] (n«60) (z.B. Parsol® SLX,
- 2,2'-Methylen-bis-(6-(2H-benzotriazol-2-yl)-4-(1 ,1 ,33-tetramethyl- butyl)phenol (z.B. Tinosorb® M),
- 2,2'-(1 ,4-Phenylen)bis-(1 H-benzimidazol-4,6-disulfonsäure, Mononatriumsalz,
- 2,2'-(1 ,4-Phenylen)bis-(1 H-benzimidazol-5-sulfonsäure, Mononatriumsalz,
- 2,2'-(1 ,4-Phenylen)bis-(1 H-benzimidazol-5-sulfonsäure,
- Monokaliumsalz und
2,4-bis-ι1 [4-(2-Ethyl-hexyloxy)-2-hydroxyl]-phenyl !>-6-(4-rτιethoxyphenyl)-1 ,3,5-triazin (z.B. Tinosorb® S).
Diese organischen Filter werden in der Regel in einer Menge von 0,5 bis 20 Gew.%, vorzugsweise 1 bis 15 Gew.%, in der erfindungsgemäß verwendeten topischen Zusammensetzung eingesetzt.
Als anorganische UV-Filter sind solche aus der Gruppe der Titandioxide, z.B. gecoatetes Titandioxid (z.B. Eusolex® T-2000 oder Eusolex® T-Aqua), Zinkoxide (z.B. Sachtotec®), Eisenoxide oder auch Ceroxide denkbar. Diese anorganischen UV-Filter werden in der Regel in einer Menge von 0,5 bis 20 Gew.%, vorzugsweise 2 bis 10 Gew.%, in der erfindungsgemäß verwendeten topischen Zusammensetzung eingesetzt.
Bevorzugte UV-Filter sind Zinkoxid, Titandioxid, 3-(4'-Methylbenzyliden)-dl-kampfer, 1 -(4-tert-Butylphenyl)-3-(4-methoxyphenyl)propan-1 ,3-dion, 4-lsopropyldibenzoyl- methan, 2-Hydroxy-4-methoxybenzophenon, Methoxyzimtsäureoctylester, 3,3,5-Trimethylcyclohexylsalicylat, 4-(Dimethylamino)benzoesäure-2-ethylhexylester, 2-Cyano-3,3-diphenylacrylsäure-2-ethylhexylester, 2-Phenylbenzimidazol-5-sulfonsäure sowie ihre Kalium-, Natrium- und Triethanolaminsalze.
Besonders bevorzugte UV-Filter sind Zinkoxid und Titandioxid.
Wird Titandioxid erfindungsgemäß verwendet, ist es bevorzugt, daß neben Titandioxid zusätzlich ein oder mehrere weitere UV-Filter, ausgewählt aus 3-(4'-Methylbenzyliden)- dl-kampfer, 1 -(4-tert-Butylphenyl)-3-(4-methoxyphenyl)propan-1 ,3-dion, 4-lsopropyldibenzoyl-methan, 2-Hydroxy-4-methoxybenzophenon, Methoxyzimtsäureoctylester, 3,3,5-Trimethylcyclohexylsalicylat, 4-(Dimethylamino)benzoesäure-2- ethylhexylester, 2-Cyano-3,3-diphenylacrylsäure-2-ethylhexylester, 2-Phenylbenz- imidazol-5-sulfonsäure sowie ihre Kalium-, Natrium- und Triethanolaminsalze, verwendet werden.
Es ist insbesondere bevorzugt, daß neben Titandioxid zusätzlich die UV-Filter 2-Hydroxy-4-methoxybenzophenon und/oder p-Methoxyzimtsäure-2-ethylhexylester verwendet werden.
Ectoin oder Ectoin-Derivate können erfindungsgemäß als Arzneimittel zur Stabilisierung von p53 verwendet werden. Zum einen kommen eine prophylaktische Anwendung, d.h. eine Anwendung vor einer Stressbelastung, wie UV-Licht oder chemischen Noxen, oder eine therapeutische Anwendung infolge dieser Stressbelastung, z.B. als "After-Sun- Präparat", in Frage. Eine kosmetische Verwendung und eine Verwendung im Nah-
rungsmitelbereich ist ebenfalls möglich. Die erfindungsgemäße Verwendung von Ectoin oder Ectoin-Derivaten führt dabei zu einer Stabilisierung von p53 auf DNA- und Proteinebene in den Zellen, so dass die natürliche Reparatur und die Schutzmechanismen der Haut und anderer Gewebe verbessert werden. Die Mutation des p53-Gens durch UV- Licht oder chemische Noxen kann durch Ectoin oder Ectoin-Derivate weitgehend verhindert werden, so dass krebsgeschädigte Zellen durch p53 abgetötet werden können, bevor sie zu Krebsherden heranwachsen. Desweiteren führen Ectoin oder Ectoin- Derivate zu einer höheren Konzentration an p53 unter Stressbedingungen, da Ectoin die Synthese von p53 stimuliert. Das p53-Protein wird durch Ectoin oder Ectoin-Derivate geschützt, indem der Wirkstoff eine Hydrathülle um das Protein bildet. Dies führt dazu, dass Wassermoleküle aus der Proteinstruktur von p53 nicht entfernt werden können, so dass die 3D-Struktur des p53-Proteins konserviert wird. Insgesamt kommt es damit zu einer Verbesserung des Abwehrstatus der Zellen, insbesondere der Hautzellen.
Die folgenden Formulierungsbeispiele erläutern die vorliegende Erfindung. Alle Verbindungen oder Komponenten, die in den kosmetischen Formulierungen verwendet werden können, sind entweder bekannt und käuflich erhältlich oder können nach bekannten Methoden synthetisiert werden.

3. Verwendung nach Anspruch 1, wobei p53 als Protein vorliegt.
4. Verwendung nach einem der Ansprüche 1 bis 3 in Form einer topischen Zusammensetzung.
5. Verwendung nach einem der Ansprüche 1 bis 3 in Form einer pharmazeutischen Zusammmensetzung.
6. Verwendung nach einem der Ansprüche 1 bis 3 in Form einer Nahrungsmittelzusammensetzung.
7. Verwendung nach einem der Ansprüche 1 oder 4, dadurch gekennzeichnet, daß mindestens eine gemäß Anspruch 1 verwendete Verbindung in einer topischen Zusammensetzung in einer Menge von 0,0001 bis 50 Gew.-%, bezogen auf die Zusammensetzung, vorliegt.
8. Verwendung nach einem der Ansprüche 1 bis 7, dadurch gekennzeichnet, daß (S)-1,4,5,6-Tetrahydro-2-methyl-4-pyrimidincarbonsäure und/oder (S, S)-1 ,4,5,6- Tetrahydro-5-hydroxy-2-methyl-4-pyrimidincarbonsäure verwendet werden.
