Abstract
Die Erfindung betrifft ein dünnschichtchromatographisches Verfahren zum Nachweis von Antibiotika. Durch die Verwendung einer Bakteriensuspension, die ein bakterielles Polysaccharid als Geliermittel enthält, kann eine entwickelte Dünnschichtchromatographie-Platte problemlos direkt in eine Bakteriensuspension getaucht werden. Die nachfolgende Inkubation und Färbung erfolgt ebenso auf der DC-Platte.

Die Erfindung betrifft ein dünnschichtchromatographisches Verfahren zum Nachweis von Antibiotika.
Der Nachweis geringster Mengen an Antibiotika spielt in vielen Bereichen eine wichtige Rolle. Beispiele sind die Untersuchung von Lebens- und Futtermitteln auf verbotene Antibiotika bzw. Leistungsförderer, der Nachweis von Antibiotika in Trink- und Abwasser, die Suche und Forschung nach neuen Antibiotika oder auch das Testen von Antibiotikapräparaten in der Qualitätskontrolle.
Antibiotika werden zumeist direkt oder indirekt über ihre abtötende oder wachstumshemmende Wirkung auf Bakterien nachgewiesen. Zum Nachweis von Antibiotika in komplexen Proben eignet sich besonders eine vorherige chromatographische Auftrennung der Probe beispielsweise auf einer Dünnschichtchromatographie-Platte (DC-Platte). Auf diese Weise kann nicht nur die antibiotische Wirkung der gesamten Probe, sondern die Wirkung einzelner Antibiotika eventuell unterschiedlicher Konzentration festgestellt werden.
Bekannte Methoden beschreiben daher zunächst die chromatographische Auftrennung einer Probe. Anschließend werden die abgetrennten potentiellen Antibiotika von der DC-Platte abgekratzt, das Antibiotikum herausgelöst und die Lösung auf eine Agarplatte gegeben. Die Agarplatte wurde vorher mit einem Bakterium angeimpft. Nach längerer Inkubationszeit können auf dem Bakterienrasen dort wo Antibiotika aufgegeben wurden freie Stellen, sogenannte Hemmhöfe, nachgewiesen werden.
Ein weiteres Verfahren ist das Agar-Overlay Verfahren. Hierbei wird die entwickelte DC-Platte mit einer mit Bakterien angeimpften Agarschicht blasenfrei Übergossen. Nach einer bestimmten Inkubationszeit wird die
Agarschicht mit oder ohne DC-Platte und in eine Färbelösung gelegt. In der Färbelösung werden bei Anwesenheit von Antibiotika helle Hemmhöfe vor einem farbigen Hintergrund sichtbar.
Nachteil dieser bekannten Verfahren ist ihre mehrstufige, aufwendige
Durchführung. Da der Nachweis der Antibiotika über das Absterben oder die Wachstumshemmung von Bakterien erfolgt, müssen die Antibiotika nach ihrer Auftrennung mit den Bakterien in Kontakt gebracht werden. Bakterien zeigen jedoch nur unter bestimmten Bedingungen ein ausreichend stabiles und gleichmäßiges Wachstum. Beispielsweise benötigen sie einen stabilen pH-Wert und dürfen nicht austrocknen. Aus diesem Grund ist es notwendig, die aufgetrennten Antibiotika durch Herauslösen oder Herausdiffundieren aus der DC-Platte in ein für Bakterien geeignetes Wachstumsmedium zu überführen. Bei diesem Vorgang muß ein möglichst großer Teil der Antibiotika mit der Bakterienkultur in Kontakt kommen, um eine gute Nachweisgrenze zu erreichen. Bei dem Overlay- Verfahren wird die DC-Platte dazu mit einer Agar-Schicht übergössen. Dieser Schritt muß von Fachleuten durchgeführt werden, da Agarlösungen nur in einem bestimmten Temperaturbereich fließfähig sind, gleichzeitig aber nicht zu heiß sein dürfen, damit die darin enthaltenen Bakterien lebensfähig bleiben. Dadurch kommt es häufig zu Verklumpungen des Agars, Blasenbildung oder unregelmäßigem Ausgießen auf der DC-Platte.
Aufgabe der vorliegenden Erfindung war es daher, ein Verfahren zum dünnschichtchromatographischen Nachweis von Antibiotika bereitzustellen, das einfach in der Handhabung ist und vollständig auf der DC-Platte durchgeführt werden kann. Dadurch wird ein schnellerer, einfacherer und genauerer Nachweis ermöglicht.
Es wurde gefunden, daß der Einsatz bestimmter Bakterien-Nährmedien, insbesondere bestimmter Geliermittel, die Versuchsdurchführung stark vereinfacht. Es ist möglich, den gesamten Nachweis auf der DC-Platte
durchzuführen. Dazu wird eine entwickelte DC-Platte direkt in eine Suspension der Bakterien in einem solchen Nährmedium eingetaucht. Man läßt die Bakterien auf der DC-Platte wachsen und führt auch die abschließende Anfärbung direkt auf der DC-Platte durch. Auf diese Weise wird die Durchführung des Nachweises stark vereinfacht. Durch die besondere Konsistenz des Nährmediums bzw. der Bakteriensuspension kann die Suspension bei Raumtemperatur verwendet werden. Es kann weder zu Verklumpungen noch zum vorzeitigen Absterben von Bakterien durch zu hohe Temperaturen kommen. Zusätzlich können bessere Nachweisgrenzen erreicht werden, da die Bakteriensuspension durch ihre guten Fließeigenschaften auch in die Poren der DC-Platte eindringen kann und so der Kontakt zwischen Antibiotikum und Bakterien intensiviert wird.
Gegenstand der vorliegenden Erfindung ist daher ein Verfahren zum dünnschichtchromatographischen Nachweis von Antibiotika mit folgenden Reaktionsschritten: a) Durchführung einer dünnschichtchromatographischen Trennung einer Probe auf einer DC-Platte; b) Herstellung einer Bakteriensuspension aus einer lagerfähigen Form eines Bakterien-Teststammes und einem Nährmedium enthaltend zwischen 1 und 3 Gewichts%, bezogen auf die Trockenmasse, eines Polysaccharides als Geliermittel; c) Eintauchen oder Benetzen der DC-Platte mit der Bakteriensuspension; d) Inkubation der DC-Platte e) Anfärben der DC-Platte mittels eines Färbereagenzes. Schritt a) und b) des erfindungsgemäßen Verfahrens können parallel oder nacheinander durchgeführt werden.
Für bevorzugte Ausführungsformen des erfindungsgemäßen Verfahrens wird in Schritt b) Gelrite® als Geliermittel bzw. als Teststamm eine Bacillus Subtilis Sporensuspension verwendet.
Bevorzugte Ausführungsformen des erfindungsgemäßen Verfahrens umfassen den Einsatz von MTT (3-(4,5-Dimethyl-2-thiazolyl)-2,5-diphenyl- 2H-tetrazoliumbromid) als Färbereagenz in Schritt e).
Gegenstand der vorliegenden Erfindung ist zudem ein Testkit zur Durchführung des erfindungsgemäßen Verfahrens, der zumindest ein Nährmedium, das zwischen 1 und 3 Gewichts%, bezogen auf die Trockenmasse, eines Polysaccharides als Geliermittel enthält, einen lagerbeständigen Bakterien-Teststamm und ein Färbereagenz enthält.
Abbildung 1 und 2 zeigen schematisch das Ergebnis zweier Antibiotika- Nachweise und sind in Beispiel 4 und 5 näher erläutert.
Die Durchführung einer dünnschichtchromatographischen Trennung einer Probe auf einer DC-Platte, das heißt die Entwicklung der DC-Platte, erfolgt unter bekannten Bedingungen. Zumeist werden hierfür DC-Platten mit einer Sorbensschicht auf Basis von Si02 verwendet. Geeignete Fließmittel, beispielsweise Acetonitril, Gemische aus Acetonitril und Dichlormethan oder Gemische aus Ethylacetat, Methanol und Ammoniak, sind dem Fachmann bekannt. Der Fachmann ist weiterhin in der Lage, die Fließmittel dem jeweiligen Trennproblem anzupassen.
Nach der Entwicklung wird die DC-Platte sorgfältig getrocknet, um Rückstände des Fließmittels zu entfernen. Anschließend wird die entwickelte DC-Platte in eine Bakteriensuspension getaucht. Man läßt die Bakterien über Nacht wachsen und besprüht den Bakterienrasen dann mit einem geeigneten Färbereagenz. Auf diese Weise werden die Hemmhöfe der Antibiotika auf dem Bakterienrasen sichtbar.
Diese erfindungsgemäße Durchführung wird durch die spezielle Zusammensetzung der Bakteriensuspension ermöglicht. Die Bakteriensuspension wird hergestellt durch Mischung einer lagerstabilen
Form eines geeigneten bakteriellen Teststamms mit einem speziellen Nährmedium. Besonders geeignet sind gepufferte Nährmedien, pH-Bereich 7,0 - 7,5. Ein erfindungsgemäß geeignetes Nährmedium enthält neben den üblichen Bestandteilen, wie Salzen, Peptonen, Hefeextrakt, sowie bevorzugterweise Mueller-Hinton-Boullion, zusätzlich zwischen 1 und 3
Gewichts%, bevorzugt ca. 1 ,5 %, bezogen auf die Trockenmasse eines Polysaccharides als Geliermittel. Bevorzugt sind dies gelierende lineare Heteropolysaccharide, die typischerweise bakteriellen Ursprungs sind, wie Gelrite® der Firma Roth, Deutschland. Derartige Materialien bilden typischerweise in Anwesenheit von löslichen Salzen stabile Gele.
Erfindungsgemäß geeignete Teststämme sind antibiotikaempfindliche, bevorzugterweise nicht pathogene, Bakterien, die in lagerfähiger Form vorliegen. Auswahlkriterien für geeignete Teststämme sind dem Fachmann bekannt. Beispiele sind Eche chia Coli oder, besonders bevorzugt, Bacillus subtilis. Lagerfähige Formen können Sporenpräparationen oder lyophilisierte vegetative Bakterien sein.
Natürlich kann die Bakteriensuspension zum Eintauchen der DC-Platte erst eine definierte Zeit vor dem Eintauchen zusammengestellt werden. Dies geschieht typischerweise durch Mischen des Nährmediums mit einer lagerfähigen Form eines Teststamms. Bei der Verwendung von Sporenpräparationen wird diese Mischung für einen bestimmten Zeitraum, zumeist zwischen 2 und 5 Stunden vorinkubiert. Erst dann kann die DC-Platte in die Mischung aus Nährmedium und Sporensuspension, erfindungsgemäß Bakteriensuspension genannt, getaucht werden.
Sporensuspensionen für das erfindungsgemäße Verfahren enthalten das Teststamm-Bakterium typischerweise in einer Sporenzahl zwischen 106 und 107.
In einer zeitsparenden Version des erfindungsgemäßen Verfahrens wird statt der Kombination von Nährmedium und Sporensuspension, die zunächst zur Herstellung der Bakteriensuspension gemischt und vorinkubiert werden müssen, das Nährmedium mit einem Teststamm- Lyophilisat gemischt. Dieses Gemisch kann typischerweise kurz vor dem
Tauchvorgang hergestellt werden, auf 35 °C erwärmt werden und steht dann nach Abkühlen auf Raumtemperatur direkt zum Tauchvorgang zur Verfügung.
Nach dem Bereitstellen der Bakteriensuspension kann die entwickelte DC- Platte eingetaucht werden. Dazu wird die DC-Platte sehr kurz vollständig in die Bakteriensuspension getaucht und überschüssige Suspension an der Unterseite entfernt. Alternativ kann die DC-Platte mit der Bakteriensuspension besprüht werden.
Die Inkubation der beschichteten DC-Platte erfolgt bevorzugt in einer Aufbewahrungsbox, die zur Schaffung einer ausreichend feuchten Atmosphäre beispielsweise mit feuchtem Papier ausgelegt ist. Die Inkubationstemperatur beträgt typischerweise 23 bis 35 °C. Nach einer Inkubationszeit von 15 bis 25 Stunden, bevorzugt 17 bis 20 Stunden, wird die DC-Platte entnommen und angefärbt.
Geeignete Färbereagenzien, die eine Unterscheidung zwischen lebenden und wachstumsgehemmten oder bevorzugt lebenden und toten Bakterien ermöglichen, sind dem Fachmann bekannt. Besonders bevorzugt sind dies Tetrazolium-Farbstoffe, wie MTT (3-(4,5-Dimethyl-2-thiazolyl)-2,5-diphenyl- 2H-tetrazoliumbromid). Die DC-Platte kann mit diesen Reagenzien besprüht werden oder in eine Lösung der Reagenzien getaucht werden. Besonders bevorzugt wird die DC-Platte erfindungsgemäß mit einer Lösung von MTT (3-(4,5-Dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazoliumbromid) besprüht. Nach einer Einwirkzeit von ca. 5 bis 30 min. treten dann die gelb gefärbten Hemmhöfe vor einem blau-violetten Hintergrund deutlich hervor.
Die Auswertung der Färbung erfolgt bevorzugt visuell und kann zusätzlich videodokumentarisch, fotographisch oder zeichnerisch festgehalten werden. Die Größe der Hemmhöfe ermöglicht Aussagen über die Auftragemenge und die Wirkungsspezifität der Substanzen.
Durch das erfindungsgemäße Verfahren wird somit ein einfacher, leicht durchführbarer und empfindlicher Test zum Nachweis von Antibiotika zur Verfügung gestellt. Durch die einfache Handhabung kann das erfindungsgemäße Verfahren auch von Personen durchgeführt werden, die keine spezifische Ausbildung für die Kultivierung von Bakterien besitzen. Weiterhin können alle wesentlichen Schritte des Verfahrens in einem einfachen chemischen Labor ohne besondere Sterilitätsanforderungen durchgeführt werden.
Mit dem erfindungsgemäßen Verfahren können beispielsweise bis zu 1 ng von Penicillin V nachgewiesen werden.
Zur anwenderfreundlichen Durchführung des Verfahrens wird erfindungsgemäß weiterhin ein Testkit zur Verfügung gestellt, der in entsprechend ausgerüsteten Labors sehr leicht durchgeführt werden kann. Der Testkit enthält Nährmedium, bevorzugt in fester Form zum Lösen in Wasser, einen geeigneten Teststamm, entweder als Lyophilisat oder als Sporenpräparation, und ein Färbereagenz, ebenso bevorzugt in fester Form zum Auflösen in Wasser.
Auch ohne weitere Ausführungen wird davon ausgegangen, daß ein Fachmann die obige Beschreibung im weitesten Umfang nutzen kann. Die bevorzugten Ausführungsformen und Beispiele sind deswegen lediglich als beschreibende, keineswegs als in irgendeiner Weise limitierende Offen- barung aufzufassen.
Die vollständige Offenbarung aller vor- und nachstehend aufgeführten Anmeldungen, Patente und Veröffentlichungen insbesondere der korrespondierenden Anmeldung DE 199 57 368, eingereicht am 30.11.1999, ist durch Bezugnahme in diese Anmeldung eingeführt.
Beispiele
1. Dünnschichtchromatoqraphische Trennung
Allgemeine Arbeitshinweise:
Zur Auftrennung von Antibiotika eignen sich insbesondere HPTLC-Platten KG 60, WRF254S der Firma Merck KGaA.
Als Fließmittel können je nach Art der Probe beispielsweise folgende Mischungen für eine Trennstrecke zwischen 40 und 60 mm eingesetzt werden:
a) Acetonitril b) Acetonitril / Dichlormethan, 98 / 2 (v/v) c) Ethylacetat / Methanol / Ammoniak 25%, 60 / 20 / 2 (v/v/v)
2. Vorkultur von Bacillus subtilis
Ein Nährmedium bestehend aus Peptonwasser, Mueller-Hinton-Boullion und 1 ,5 Gew.% Gelrite® wird mit 200 ml deionisiertem Wasser (VE-Wasser) in einen 300 ml Erlenmeyerkolben gegeben, ca. 10 min. zum Sieden erhitzt, bis sich eine klare Lösung bildet. Bei der Verwendung von sauren Fließmitteln bei der DC-T rennung sollte die Nährlösung statt mit reinem VE-Wasser mit 0,5 M HEPES oder 0,5 M Tris-Puffer (pH 7,2) angesetzt werden.
Nach dem Aufkochen wird das Nährmedium auf Raumtemperatur abgekühlt (25°C ± 3°C) und mit der Sporenlösung (106 - 107 Sporen Bacillus subtilis (BGA)) angeimpft. Dies sollte unter keimarmen oder sterilen Bedingungen erfolgen. Anschließend wird die Suspension 2 h im Brutschrank bei 35°C kultiviert. Anschließend wird die Vorkultur auf 20 bis 25°C Tauchtemperatur abgekühlt.
3. Beschichtunq der DC-Platte und Anfärben
Die entwickelte DC-Platte wird kurz (ca. 1 sec.) ganzflächig in die Bakteriensuspension getaucht. Anschließend hält man die DC-Platte waagerecht und entfernt die überschüssige Bakteriensuspension an der Unterseite.
Sofort nach dem Tauchvorgang wird die DC-Platte in einer Aufbewahrungsbox über Nacht (19 h) bei 23-35°C inkubiert. Die Aufbewahrungsbox wird 1 h vorher am Rand mit feuchtem Kammersättigungspapier (3 Streifen a 300x70 mm) oder mit 2 feuchten Zellstofftüchern ausgelegt. Zur Lagerung der DC-Platte in der Box wird zusätzlich eine Dose hineingestellt, auf die die DC-Platte gelegt werden kann. Weiterhin sollte die DC-Platte leicht schräg gelagert werden, so daß die überschüssige Bakteriensuspension in den Bereich oberhalb der Lösungsmittelfront am oberen Rand abfließen kann.
Nach 19 h wird die überschüssige Suspension am oberen Rand mit einem Tuch abgetupft und die DC-Platte mit MTT-Lösung (3 mg/ml) aus einer Entfernung von ca. 10 cm besprüht, bis eine leichte Gelbfärbung der Oberfläche zu erkennen ist. Die DC-Platte wird erneut für ca. 5 bis 30 min. jn die Aufbewahrungsbox gelegt, anschließend bei Raumtemperatur getrocknet und ausgewertet.
4. Anwendunqsbeispiel A
20 ng Chloramphenicol, 1 ng Penicillin und 4 ng Oxatetracyclin (jeweils aus Lösungen von 10 mg in 100 ml Methanol) werden auf eine DC-Platte aufgetragen. Die Entwicklung erfolgt in Ethylacetat / Methanol / Ammoniak
25%, 60/20/2 (v/v/v). Die Bakteriensuspension wird 3,5 h bei 35°C vorinkubiert und dann auf Tauchtemperatur abgekühlt.
Nach dem Tauchvorgang wird die DC-Platte 19 h bei RT inkubiert. Die Anfärbung erfolgt mit 3 mg/ml MTT in wässriger Lösung. Nach 30 min erhält man die in Abbildung 1 schematisch dargestellte gefärbte DC-Platte.
Auf der DC-Platte sind aufgetragen:
Bahn 1 Gemisch aller 3 Antibiotika Bahn 2 Oxatetracyclin
Bahn 3 Penicillin
Bahn 4 Chloramphenicol
Die Entwicklung erfolgte von unten nach oben. Die Hemmhöfe der Antibiotika sind jeweils als gelbe Flecken auf blau/violettem Untergrund sichtbar.
5. Anwendunqsbeispiel B
5 ng Chlortetracyclin, 2 ng Penicillin G-Procain und (jeweils aus Lösungen von 10 mg in 100 ml Methanol) und 50 nl eines Extraktes aus Futtermittel werden auf eine DC-Platte aufgetragen. Die Entwicklung erfolgt in Acetonitril. Die Bakteriensuspension wird 4 h bei 25°C vorinkubiert und dann auf Tauchtemperatur abgekühlt. Nach dem Tauchvorgang wird die DC-Platte 19 h bei RT inkubiert. Die Anfärbung erfolgt mit 3 mg/ml MTT in wässriger Lösung. Nach 30 min erhält man die in Abbildung 2 schematisch dargestellte gefärbte DC-Platte.
Auf der Platte sind aufgetragen: Bahn 1 : Extrakt
Bahn 2: Chlortetracyclin Bahn3: Penicillin G-Procain
Die Hemmhöfe der Antibiotika sind jeweils als gelbe Flecken auf blau/violettem Untergrund sichtbar.
Claims
1. Verfahren zum dünnschichtchromatographischen Nachweis von Antibiotika, gekennzeichnet durch folgende Verfahrensschritte: a) Durchführung einer dünnschichtchromatographischen Trennung einer Probe auf einer DC-Platte; b) Herstellung einer Bakteriensuspension aus einem Teststamm und einem Nährmedium enthaltend zwischen 1 und 3 Gewichts%, bezogen auf die Trockenmasse, eines bakteriellen Polysaccharides als Geliermittel; c) Eintauchen oder Besprühen der DC-Platte mit der Bakteriensuspension; d) Inkubation der DC-Platte; e) Anfärben der DC-Platte mittels eines Färbereagenzes.
2. Verfahren zum dünnschichtchromatographischen Nachweis von Antibiotika entsprechend Anspruch 1 , dadurch gekennzeichnet, daß als Geliermittel in Schritt b) Gelrite® verwendet wird.
3. Verfahren zum dünnschichtchromatographischen Nachweis von
Antibiotika entsprechend Anspruch 1 oder 2, dadurch gekennzeichnet, daß als Teststamm in Schritt b) eine Bacillus Subtilis Sporensuspension eingesetzt wird.
4. Verfahren zum dünnschichtchromatographischen Nachweis von
Antibiotika entsprechend einem der Ansprüche 1 bis 3, dadurch gekennzeichnet, daß als Färbereagenz in Schritt e) MTT (3-(4,5-
Dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazoliumbromid) verwendet wird. Testkit zur Durchführung des Verfahrens nach einem der Ansprüche 1 bis 4, zumindest enthaltend ein Nährmedium, das zwischen 1 und 3 Gewichts%, bezogen auf die Trockenmasse, eines bakteriellen Polysaccharides als Geliermittel enthält, einen lagerstabilen Teststamm und ein Färbereagenz.
