Abstract
Das neue Verfahren ermöglicht die Oxidation von organischen Verbindungen mit Peroxycarbonsäuren, die in situ in Gegenwart von Enzymen durch Umsetzung von Wasserstoffperoxid mit gesättigten aliphatischen Carbonsäureestern und/oder Mischungen der korrespondierenden Carbonsäuren und Alkohole erzeugt werden, wobei kurzkettige Carbonsäuren und längerkettige Alkohole eingesetzt werden und das bei der Umsetzung entstehende und/oder zugeführte Wasser ganz oder teilweise entfernt wird.
Description
Verfahren zur Oxidation von organischen Verbindungen
Die Erfindung betrifft ein Verfahren zur Oxidation von organischen Verbindungen mit Peroxycarbonsäuren, die in situ mit Wasserstoffperoxid aus gesättigten aliphati- schen Carbonsäureestern und/oder Mischungen der korrespondierenden Carbonsauren und Alkohole in Gegenwart von Enzymen als Katalysatoren erzeugt werden.
Die Oxidation von organischen Verbindungen unter Einsatz von Peroxycarbon- säuren, die in situ aus Carbonsäuren mit Wasserstoffperoxid in Gegenwart von
Enzym-Katalysatoren hergestellt werden, ist prinzipiell bekannt. Beispiele für Reaktionen, bei denen Peroxycarbonsäuren als Oxidationsmittel verwendet werden, sind: die Epoxidation von Olefmen - die Baeyer-Villiger-Oxidation von Ketonen zu Estern die Oxidation von Aminen zu Aminoxiden, Nitroso- und Nitroverbin- dungen die Oxidation aliphatischer Aldehyde zu Carbonsäuren die Oxidation aromatischer Aldehyde zu Phenolen
Für die Verwendung von Peroxycarbonsäuren in solchen Oxidationsreaktionen spricht, daß sie eine deutlich größere und selektivere Oxidationskraft als Wasserstoffperoxid selber besitzen. Obgleich einige Peroxycarbonsäuren nützliche und kommerziell verfügbare Reagenzien sind, ist ihre Verwendung wegen ihrer relativ hohen Kosten und der Risiken (insbesondere der Explosionsgefahr), die die Handhabung der Reagenzien insbesondere im produktionstechnischen Maßstab mit sich bringt, beschränkt. Durch den Einsatz von Enzymen zur Herstellung der Peroxycarbonsäuren können deutlich mildere Reaktionsbedingungen gewählt werden. Hierdurch werden vor allem die Risiken verringert, die im Umgang mit den Peroxy- carbonsäuren bestehen. Zum anderen wird es möglich, auch thermisch empfindlichere Carbonsäuren zu den entsprechenden Peroxycarbonsäuren umzusetzen.
So wird in Tetrahedron 48 ( 1992), 4587-92 die Lipase-katalysierte Herstellung von langkettigen C(,-C| (.-Peroxycarbonsäuren durch Umsetzung der entsprechenden Carbonsauren mit Wasserstoffperoxid beschπeben, und die in situ Verwendung dieser Peroxycarbonsäuren zur Oxidation von Alkenen bzw. Sulfiden Diese in-situ Herstellung der Peroxy carbonsäuren sowie die Oxidation der Alkene bzw. Sulfide erfolgt dabei entw eder in einem 2-Phasen-System aus einem organischen Lösungsmittel und Wasser oder aber nur in Gegenwart von Wasser ohne weiteres Lösungsmittel.
WO-A-91/04333 beschreibt ebenfalls ein Verfahren zur Herstellung von Peroxycarbonsäuren durch Umsetzung der entsprechenden Carbonsäuren mit Wasserstoffperoxid in Gegenwart von Enzymen als Katalysatoren. Das Verfahren ermöglicht die Herstellung von Peroxycarbonsäuren RCOOOH, wobei R em organischer Rest ist, insbesondere em linearer oder verzweigter, gesättigter oder ungesättigter
Alkylrest, ein Arylrest oder ein Alkylarylrest, die jeweils gegebenenfalls durch unterschiedlichste Gruppen und Reste substituiert sein können. Der Rest R kann z.B. für einen C1-C30- Alkylrest stehen. Der eindeutige Schwerpunkt der WO-A-91/04333 liegt dabei auf den in den Beispielen hergestellten längerkettigen Peroxycarbon- säuren mit C6-Cι8-Alkylresten Als Enzyme werden bevorzugt Hydrolasen wie
Proteasen oder Lipasen eingesetzt. Die Herstellung der Peroxycarbonsäuren kann in der Lösung der zugrundeliegenden Carbonsäure selber oder aber in einem Lösungsmittel vorgenommen. Als Lösungsmittel werden explizit Wasser, wäßπge Pufferlösungen oder aber organische Losungsmittel, z.B. Kohlenwasserstoffe wie Hexan, Cylcohexan, Heptan, Benzol, Toluol, Methylenchlorid, Hexachlorethan, Acetonitπl,
DMF, Dioxan oder THF genannt
WO- A-91/04333 beschreibt ferner die Oxidation organischer Verbindungen unter
Einsatz der in-situ hergestellten Peroxycarbonsäuren Experimentell beschπeben wird die Oxidation von Alkenen zu Oxiranen, von Ketonen zu Estern und von
Sulfiden zu Sulfoxiden in Gegenwart von Wasserstoffperoxid und men sowie
den längerkettigen Carbonsäuren Octansäure oder Myristinsäure. Der Bedarf an Wasserstoffperoxid ist dabei mit 1 ,5 bis 5 mol pro mol Edukt deutlich überstöchio- metrisch, und in vielen Fällen muß hochkonzentriertes 60 %iges Wasserstoffperoxid eingesetzt werden. Auch das immobilisierte Enzym wird in sehr großen Mengen eingesetzt. Trotzdem werden bei langen Reaktionszeiten zwischen 4 und 24 h im allgemeinen keine vollständigen, bisweilen sogar nur geringe Umsätze bzw. Ausbeuten von ungefähr 10 bis 60 % erzielt.
WO-A-98/36058 beschreibt die kontinuierliche Entfernung von Reaktionswasser, welches bei enzymatisch katalysierten Reaktionen gebildet wird, durch Druck- permeation an einer speziellen nicht-porösen Membran. Auf dieser nicht-porösen Membran ist dabei gleichzeitig auch der Enzym-Katalysator fixiert. Als derart katalysierte Reaktionen werden die Veretherung von Monosacchariden zu Poly- sacchariden in Gegenwart von Carbohydrasen genannt, ferner die Veresterung von Carbonsäuren mit Alkoholen in Gegenwart von Lipasen und die Bildung von
Amiden aus Aminosäuren in Gegenwart von Proteasen. Die beschriebenen Membransysteme sind kompliziert und werden nicht für die Herstellung von Peroxycarbonsäuren oder die Oxidation organischer Verbindungen eingesetzt.
EP-A-0 310 952 beschreibt ein Verfahren zur Herstellung verdünnter wäßriger
Peroxycarbonsäurelösungen zum Einsatz als Bleich- und Desinfektionsmittel. Das Verfahren umfaßt die Umsetzung von Carbonsäureestern mit Wasserstoffperoxid in Gegenwart von Hydrolasen. Für das Erreichen einer vernünftigen Selektivität ist es hierbei essentiell, daß es sich bei den Hydrolasen um Proteasen handelt und gege- benenfalls in Anwesenheit von Tensiden und im Alkalischen gearbeitet wird. Als
Ausgangsstoffe für die Percarbonsäuren werden Ester von Monocarbonsäuren verwendet. Als prinzipiell einsetzbar werden Ester von Monocarbonsäuren mit 1 bis 24 C- Atomen im Säureteil genannt, wobei die Konzentration der Carbonsäureester bevorzugt zwischen 1 und 10 Gew.-%, bezogen auf die Gesamtlösung, liegt. Der Schwerpunkt der EP-A-0 310 952 liegt auf längerkettigen Monocarbonsäuren mit 4 bis 10 C-Atomen im Säureteil und auf kurzkettigen Alkoholresten mit 1 bis 4 C-Ato-
men. Besonderer Augenmerk ist auf die Herstellung von Percarbonsäuren mit 8 C- Atomen im Säureteil gerichtet, da solche Percarbonsäuren beim beabsichtigten Einsatz als Bleich- und Desinfektionsmittel besonders günstige Eigenschaften besitzen.
Auch aus der DE-OS-2 240 605 sind Zusammensetzungen bekannt, die beim Waschen von Textilien in wäßrigem Medium zu Haushalts- oder industriellen Zwecken als Bleichmittel eingesetzt werden. Diese Zusammensetzungen enthalten Acylalkylester mit jeweils 1 bis 10 C-Atomen im Säure- und Alkoholrest sowie eine Hydrolase. Durch Temperaturerhöhung kommt es in der Waschlauge zur Bildung der entsprechenden Persäuren, die aufgrund ihrer bleichenden Wirkung ein breites Spektrum von Flecken und Verschmutzungen entfernen. Auch in EP-A-0 268 456 und EP-A-0 253 487 werden ähnliche Bleichmittelzusammensetzungen für das Waschen von Textilien beschrieben.
Aus DE- 197 38 442-A1 ist ferner die Herstellung von Perkohlensäurehalbestern der Formel ROC(O)OOH durch katalytische Perhydrolyse von Kohlensäurediestern mit Wasserstoffperoxid beschrieben. Die so erhaltenen Perkohlensäurehalbester können in situ als Oxidationsmittel eingesetzt werden.
Aus Proc. World Congr. Int. Soc. Fat Res., 21st (1996), 3, 469-471 ist es ebenfalls bekannt, die Oxidation von Alkenen in Gegenwart von Peroxycarbonsäuren durchzuführen, die in-situ durch Umsetzung von Carbonsäuren und Wasserstoffperoxid hergestellt werden. Eingesetzt wird ein breites Spektrum von Carbonsäuren; das besondere Augenmerk liegt dabei widerum auf den langkettigen Carbonsäuren/Fettsäuren mit bis zu 22 C-Atomen, da diese bei der Oxidation von 1-Okten zu Ausbeuten im Bereich um 60 % führen. Der Einsatz von kurzkettigen Carbonsäuren wie Propionsäure oder Isobutyrsäure liefert dagegen nur Ausbeuten von 41 % bzw. sogar nur 25 % 1,2-Epoxyoktan. Beschrieben wird ferner der Einsatz von Estern langkettiger Carbonsäuren mit kurzkettigen Alkoholen, die in Gegenwart von
Lipasen unter Einwirkung des Wasserstoffperoxids ebenfalls zu Peroxycarbonsäuren
gespalten werden und dann die Alken-Oxidation katalysieren Diese Carbonsaure- ester führen jedoch bei der Oxidation von 1 -Okten immer zu deutlich schlechteren Ausbeuten als die entsprechenden freien Carbonsauren So w erden bei Verwendung des Methyl-, Butyl- oder Vιn\ lcsters von Dodecansaure nur Ausbeuten zwischen 13 und 33 % erhalten, w ahrend die freie Saure eine Ausbeute von 59 % liefert Vor diesem Hintergrund deutlich schlechterer Ausbeuten geben die Autoren als einzigen Grund für den Einsatz \ o Carbonsaureestern den Vorteil an, daß medπger konzentrierte Wasserstoffperoxid-Losungen verwendet werden können Nur bei Einsatz des Tπfluorethyl esters der Dodecansaure wird eine zur freien Dodecansaure vergleich- bare gute Ausbeute von 62 % erreicht, was die Autoren aber auf den Tπfluorethylrest als gute Abgangsgruppe zurückführen
Da Oxidationsreaktionen zu den grundlegenden und häufigsten Reaktionen der organischen Chemie zahlen, bestand die Aufgabe der vorliegenden Erfindung dann, ein Oxidationsverfahren bereitzustellen, welches es erlaubt, mit einfachen technischen Maßnahmen und unter milden Reaktionsbedingungen höhere Umsätze an Edukt zu erzielen
Gegenstand der Erfindung ist em Verfahren zur Oxidation von organischen Ver- bmdungen mit Peroxycarbonsäuren, die in situ m Gegenwart von Enzymen durch
Umsetzung von Wasserstoffperoxid mit gesattigten ahphatischen Carbonsaureestern und/oder Mischungen der korrespondierenden Carbonsauren und Alkohole erzeugt werden, dadurch gekennzeichnet, daß die gesattigten ahphatischen Carbonsauren geradkettig oder verzweigt sind und 1 bis 4 C-Atome aufweisen und die Alkohole die Formel
R-OH besitzen, wobei
R fui einen geradkettigen oder verzweigten C -Cis-Alkvlrest steht, der gege- benenfalls durch ein oder zwei Reste OR' substituiert ist wobei R' unabhängig voneinander Wasserstoff oder ein C?-C4-Acylrest bedeuten, und
dessen Alkylkette gegebenenfalls durch ein oder mehrere Sauerstoffatome unterbrochen ist, und
daß das bei der Umsetzung entstehende und/oder zugeführte Wasser ganz oder teil- weise entfernt wird.
Im erfindungsgemäßen Verfahren werden zur in-situ Herstellung der Peroxycarbon- säureester gesättigte aliphatische Carbonsäureester und/oder Mischungen der korrespondierenden Carbonsäuren und Alkohole eingesetzt, wobei die Carbonsäuren 1 bis 4 C-Atome, bevorzugt 2 oder 3 C-Atome aufweisen.
Bei den Alkoholen der Formel ROH sind solche bevorzugt, bei denen R für einen linearen oder verzweigten C4-C8-Alkylrest oder einen Monohydroxy-substituierten C3-C6-Alkylrest, insbesondere einen ω-Hydroxy-C3-C6- Alkylrest steht. Geeignet sind auch solche Alkohole, bei denen R für einen C3-C6- Alkylrest steht, der durch ein oder zwei Hydroxy- oder O-(C2-C4)-Acylreste substituiert ist. Hierbei ist insbesondere ein Glycerinrest bevorzugt, bei dem 1 oder 2 OH Gruppen durch einen O-(C -C4)-Acylrest verestert sind Besonders bevorzugt werden im erfindungsgemäßen Verfahren Butylacetat als ahphatischer Carbonsäureester und/oder Essigsaure-Butanol-Mischungen eingesetzt.
Als Enzyme können im erfindungsgemäßen Verfahren Hydrolasen wie Esterasen oder Proteasen eingesetzt werden. Bevorzugt werden Lipasen, Proteasen oder Pepti- dasen eingesetzt. Die Eignung eines gegebenen Enzyms zur Verwendung im vor- liegenden Verfahren kann leicht getestet werden, indem em Carbonsäureester-Substrat in Gegenwart des Enzyms Wasserstoffperoxid oder einer Wasserstoffperoxid- Vorstufe ausgesetzt wird und die Erzeugung von Peroxycarbonsaure aus der Reaktion überwacht wird Das Enzym kann als solches eingesetzt werden, als Losung, in lyophilisierter Form, in chemisch modifizierter Form oder aber immobilisiert auf einem Träger, um seine Stabilität und seine Aktivität gegenüber dem fraglichen
Substrat zu erhöhen. Bevorzugt werden durch Immobilisierung auf Trägem leicht handhabbare und stabilisierte Enzyme eingesetzt.
Lipasen, die im vorliegenden Verfahren eingesetzt werden können, können mikro- bielle Lipasen sein, die z.B von Stämmen von Aspergillus, Enterobacterium,
Chromobacterium, Geotricium oder Penicillium produziert werden. Bevorzugte Lipasen zur Verwendung gemäß der Erfindung sind diejenigen, die von Spezies von Mucor, Humicola, Pseudomonas oder Candida produziert werden.
Besonders bevorzugte Lipasen sind diejenigen, die von folgenden Mikroorganismenstämmen produziert werden, die alle in der Deutschen Sammlung von Mikroorganismen gemäß den Bestimmungen des Budapester Vertrages über die Internationale Anerkennung der Hinterlegung von Mikroorganismen für die Zwecke von Patentverfahren hinterlegt worden sind:
Candida antaretica, hinterlegt am 29. September 1986, mit der Nummer DSM 3855, und am 8. Dezember 1986, mit den Nummern DSM 3908 und DSM 3909.
Pseudomonas cephacia, hinterlegt am 30. Januar 1987, mit der Nummber 3959.
Humicola lanuginosa, hinterlegt am 13 August 1986 und am 4. Mai 1987 mit den Hinterlegungsnummem 3819 bzw. 4109.
Humicola brevispora, hinterlegt am 4. Mai 1987, mit der Hinterlegungsnummer DSM 41 10.
Humicola brevis var. thermoidea, hinterlegt am 4. Mai 1987, mit der Hinterlegungsnummer DSM 41 1 1 , und

Gegenwärtig bevorzugte Lipasen sind diejenigen, die von Candida antarctica, DSM 3855, DSM 3908 und DSM 3909, produziert werden. Diese Enzyme können mit dem in WO 88/02775 offenbarten Verfahren hergestellt werden. Die fraglichen Candida-Stämme können dabei unter aeroben Bedingungen in einem Nährstoffmedium kultiviert werden, das assimilierbare Kohlenstoff- und Stickstoffquellen sowie essentielle Mineralien, Spurenelemente etc. enthält, wobei das Medium gemäß etablierter Praxis zusammengesetzt ist. Nach Kultivierung können flüssige Enzymkonzentrate hergestellt werden, indem unlösliche Materialien, z.B. durch Filtration oder Zentrifugation, entfernt werden, wonach die Kulturbrühe durch Eindampfen oder Umkehrosmose konzentriert werden kann. Feste Enzymzubereitungen können aus dem Konzentrat durch Ausfällung mit Salzen oder wasser-mischbaren Lösungsmitteln, z.B. Ethanol, oder durch Trocknung, wie etwa Sprühtrocknung, gemäß gut bekannten Verfahren hergestellt werden.
Zusätzliche Lipasen können aus den folgenden Stämmen erhalten werden, die ohne Beschränkung vom Centraalbureau voor Schimmelculturen (CBS), von der American Type Culture Collection (ATCC), von der Agricultural Research Culture Collection (NRRL) und vom Institute of Fermentation, Osaka (IFO) öffentlich zugänglich sind, mit den folgenden Hinterlegungsnummern: Candida antarctica, CBS 5955, ATCC
34888, NRRL Y-8295, CBS 6678, ATCC 28323, CBS 6821 und NRRL Y-7954; Candida tsukubaensis, CBS 6389, ATCC 24555 und NRRL Y-7795; Candida auriculariae, CBS 6379, ATTC 24121 und IFO 1580; Candida humicola, CBS 571, ATCC 14438, IFO 0760, CBS 2041, ATTC 9949, NRRL Y-1266, IFO 0753 und IFO 1527; und Candida foliorum, CBS 5234 und ATCC 18820.
Es ist bekannt, Lipase durch rekombinante DNA-Techniken herzustellen, vgl. z.B. EP-A-0 238 023. Rekombinante Lipasen können auch für den vorliegenden Zweck eingesetzt werden.
Wenn es im Verfahren der Erfindung eingesetzt wird, kann das Enzym in einem löslichen Zustand vorliegen. Es ist jedoch bevorzugt, das Enzym zu immobilisieren, um die Gewinnung der Peroxycarbonsäuren, die mit dem vorliegenden Verfahren hergestellt werden, zu erleichte . Immobilisierungsverfahren sind gut bekannt und schließen Vernetzung von Zellhomogenaten, kovalente Kopplung an unlösliche organische oder anorganische Träger, Einschluß in Gele und Adsorption an Ionen- austauschharze oder andere adsorbierende Materialien ein. Aufbringen auf einen teil- chenförmigen Träger kann ebenfalls eingesetzt werden (z.B. A.R. Macrac und R.C. Hammond, Biotechnology and Genetic Engineering Reviews, 3, 1985, S. 193). Geeignete Trägermaterialien für das immobilisierte Enzym sind z.B. Kunststoffe
(z.B. Polypropylen, Polystyrol, Polyvinylchlorid, Polyurethan, Latex. Nylon, Teflon, Dacron, Polyvinylacetat, Polyvinylalkohol oder jedes geeignete Copolymer derselben), Polysaccharide (z.B. Agarose oder Dextran). Ionenaustauscherharze (sowohl Kationen- als auch Anionen- Austauschharze), Sihciumpolymere (z.B. Siloxan) oder Silikate (z.B. Glas).
Es ist bevorzugt, das Enzym auf einem Ionenaustauschharz zu immobilisieren, indem das Enzym an das Harz adsorbiert wird oder indem es mit Hilfe von Glutaraldehyd oder einem anderen Vernetzungsmittel in einer per se bekannten Art und Weise mit dem Harz vernetzt wird. Ein besonders bevorzugtes Harz ist ein schwachbasisches
Anionen-Austauschharz, das ein Harz vom Polystyrol-, Acryl- oder Phenol-Formaldehyd-Typ sein kann. Beispiele kommerziell verfügbarer Harze vom Polyacryl-Typ sind Lewatit® E 1999/85 (eingetragene Marke von Bayer, Bundesrepublik Deutschland) und Duolite® ES-568 (eingetragene Marke von Rohm & Haas, Bundesrepublik Deutschland). Immobilisierung von Enzymen an diesen Harztyp kann gemäß EP-A-0
140 542 durchgeführt werden. Immobilisierung an Harze vom Phenyl-Formaldehyd- Typ kann gemäß DK 85/878 durchgeführt werden. Ein Beispiel eines kommerziell verfügbaren Harzes vom Acryl-Typ ist Lewatit" E 2001/85 (eingetragene Marke von
Bayer, Bundesrepublik Deutschland).
Ein anderes geeignetes Material zur Immobilisierung von Enzymen ist ein anorganischer Trager, w ie etwa ein Silikat Das Enzym kann an den Trager durch Adsorption oder durch kovalente Kopplung gebunden werden
Auch Mischungen von Enzymen können Verwendung finden
Das erfindungsgemaße Verfahren wird bei Temperaturen von 10 bis 1 10°C, bevorzugt 20 bis 90°C, besonders bevorzugt 40 bis 60°C durchgeführt
Das als Oxidationsmittel verwendete Wasserstoffperoxid wird üblicherweise in Form einer 10 bis 70 %-ιgen, bevorzugt in Form einer 20 bis 40 %-ιgen wäßrigen Losung eingesetzt, so daß 1 bis 100 mol, bevorzugt 1,1 bis 10 mol und insbesondere 1,2 bis 2 mol Wasserstofφeroxid pro Oxidationsaquivalent der organischen Verbindung vorliegen Die Dosierung kann dabei entweder diskontinuierlich in Form einer ein- maligen Zugabe erfolgen oder in mehreren Portionen oder aber auch kontinuierlich mit einer bestimmten gewünschten Geschwindigkeit Alternativ ist es möglich, eine Vorstufe von Wasserstofφeroxid einzusetzen, die unter den Reaktionsbedingungen in situ Wasserstofφeroxid freisetzt, z B Percarbonate oder Perborate jeweils m Form ihrer Alkali- oder Erdalkahmetallsalze
Ein wesentliches Merkmal des erfindungsgemaßen Verfahrens besteht dann, daß das im erfindungsgemaßen Verfahren entstehende und/oder dem Reaktionssystem zugeführte Wasser aus dem Reaktionssystem entfernt wird Diese Wasserabtrennung kann zum einen direkt im Reaktor wahrend der Umsetzung erfolgen, d h durch Ver- dampfung, gegebenenfalls durch Pervaporation oder Dampf-Pervaporation Bevorzugt wird eine einfache Destillation duchgeführt Alternativ dazu kann ein Teilstrom des flussigen Reaktionsgemisches, das mit dem Enzym im Kontakt steht, aus dem Reaktor entfernt werden und dann räumlich getrennt vom Enzym das Wasser außerhalb des Reaktors z B durch Verdampfung entfernt werden und der von Wasser befreite Strom der Reaktion wieder zugeführt zu werden Es hat sich bewahrt, das
Wasser soweit zu entfernen, daß der Wasseranteil im Reaktionsgemisch bei 0,001 bis 10 Gew -%, bevorzugt 0,01 bis 3 Gew -% und insbesondere 0,1 bis 2 Gew -% hegt
Erfolgt die Wasserabtrennung aus der Reaktionsmischung räumlich getrennt \ om Enzym, so werden bei der Wasserabtrennung Temperaturen im Bereich von 50 bis 200°C, be\ orzugt 80 bis 160°C eingestellt
Die Wasserabtrennung erfolgt bei einem Druck im Bereich von 0,001 bis 10 bar, bevorzugt 0,01 bis 1 bar
Im erfindungsgemaßen Verfahren dient der Carbonsaureester im allgemeinen gleichzeitig als Losungsmittel Es hat sich aber auch bewahrt, zusätzlich em oder mehrere inerte organische Losungsmittel zu benutzen Einige bevorzugte organische Losungsmittel sind Kohlenwasserstoffe wie Hexan, Cyclohexan, Heptan, Benzol, Toluol, die isomeren Xylole und deren Mischungen, Chlorbenzol, Dichlorbenzol, Methylen- chlond, Hexachlorethan, Acetonitnl, Dimethylformamid, Dioxan und Tetrahydro- furan Die Verwendung eines solchen Losungsmittels ist besonders dann vorteilhaft, wenn es mit dem zu entfernenden Wasser ein Azeotrop bildet und so die Abtrennung des Wassers vereinfacht Besonders bevorzugt ist die Verwendung solcher Losungs- mittel, die mit Wasser ein Heteroazeotrop bilden, damit die einfache Ruckführung des Losungsmittels gewährleistet ist Alternativ zur Zugabe organischer Losungsmittel kann auch die zu oxidierende Verbindung gleichzeitig als Losungsmittel dienen
Die Reaktionszeiten hegen üblicherweise bei 0,5 bis 24 Stunden, bevorzugt bei 2 bis
12 Stunden und besonders bevorzugt bei 4 bis 8 Stunden Die Wτasserabtrennung kann wahrend der gesamten Reaktionszeit oder in bestimmten Zeitabschnitten erfolgen
Das ei findungsgemaße V erfahren ist besonders geeignet, um Pyndme zu Pvπdιn-\- oxiden, Olefine zu Oxiranen, Hydrosulfide zu Disulfiden, Sulfide zu Sulfoxiden und
Sulfonen und Ketone zu Estern zu oxidieren. Besonders bevorzugt werden Pyridine und Olefine oxidicrt, ganz besonders bevorzugt Pyridine in die entsprechenden N- Oxide überführt.
Im erfindungsgem ßen Verfahren können Pyridine der Formel la zu Pyridin-N- oxiden der Fomiel 1 b oxidicrt werden.

R und R gleich oder verschieden sind und H, G-C6-Alkyl, C3-C6-Cycloalkyl, oder Phenyl bedeuten oder aber beide Substituenten R und R10 gemeinsam einen
Ci-Go-Alkylen- oder C3-Cιo-Alkylidenrest bilden, wobei diese Alkylen- und
Alkylidenreste ein oder mehrfach durch O, COO oder CO unterbrochen sein können.
Das molare Verhältnis zwischen der zu oxidierenden organischen Verbindung und dem Carbonsäureester bzw. der Carbonsäure beträgt im erfindungsgemäßen Verfahren 0, 1 : 1 bis 1000: 1 , bevorzugt 0,5 : 1 bis 500: 1.
Als Reaktoren für das erfindungsgemäße Verfahren sind alle Reaktoren für die
Umsetzung flüssiger Reaktionsmischungen geeignet, wie sie aus dem Stand der Technik bekannt sind. Im bevorzugten Fall der Verwendung von auf Trägem immobilisierten Enzyme werden die Katalysatorpartikel in der Flüssigkeit schwimmend eingesetzt oder können als feste stationäre Katalysatorschüttungen vom Reaktions- gemisch durchströmt werden.
Als Reaktoren sind z.B. Rührkessel geeignet, bevorzugt solche mit Kolonne und Wasserabscheider, ferner Blasensäulen, Schlaufenreaktoren mit oder ohne stationäre Katalysatorschüttung, Rohrreaktoren und Rohrbündelreaktoren mit stationärer Kata- lysatorschüttung.
Das erfindungsgemäße Verfahren zeichnet sich dadurch aus, daß die Oxidation eines breiten Spektrums organischer Verbindungen unter Verwendung nur geringer Überschüsse an Wasserstofφeroxid und geringer Mengen an Enzym-Katalysator möglich ist. Hervorzuheben sind femer die milden Reaktionsbedingungen, die kurzen Reaktionszeiten und die hohen erzielbaren Umsätze.
Beispiele
Beschreibung des Versuchsaufbaus
Alle Reaktionen werden in einem 500 ml Dreihalskolben durchgeführt, der mit einem Wasserabscheider, einem Rührer mit Magnetkupplung und einem Tropftrichter ausgestattet ist. Über den Kühler des Wasserabscheiders wird eine Vakuumpumpe angeschlossen.
Alle Versuche werden gaschromatographisch oder über HPLC hinsichtlich des Umsatzes und der Selektivität mit gegen Reinsubstanzen der Ausgangs- und Endprodukte kalibrierten Methoden ausgewertet.
Beispiel la
Oxidation von Lutidin zu Lutidin-N-oxid
26,8 g Lutidin und 1 g Novozym 435 (eingetragene Marke der Firma Novo Nor- disk) werden in der oben beschriebenen Versuchsapparatur zusammen mit 400 ml Butylacetat vorgelegt. Unter Rühren bei 45°C und vermindertem Druck von 40 mbar läßt man 30 g Wasserstofφeroxid in Form einer 35 %-igen wäßrigen Lösung innerhalb von 2 Stunden zutropfen. Anschließend wird 4 Stunden bei 45°C nachgerührt. Während der gesamten Zeit wird das bei der Reaktion freiwerdende und in die Reaktion eingetragene Wasser kontinuierlich über den Wasserabscheider abgetrennt. Luti- din wird mit einem Umsatz von 100 % und einer Selektivität von 96 % zu Lutidin-N- oxid umgesetzt.
Beispiel lb
(Vergleichsbeispiel zu Beispiel la ohne Wasserabtrennung)
26,8 g Lutidin und 2,6 g Novozym 435® werden in der oben beschriebenen Versuchsapparatur zusammen mit 400 ml Butylacetat vorgelegt. Unter Rühren bei 45°C läßt man 34 g Wasserstofφeroxid in Form einer 35 %-igen wäßrigen Lösung innerhalb von 2 Stunden zutropfen. Anschließend wird 4 Stunden bei 45°C nachgerührt. Lutidin wird mit einem Umsatz von 62 % und einer Selektivität von 95 % zu Lutidin-N-oxid umgesetzt.
Trotz 2,6-facher Katalysatormenge wird, relativ zum Beispiel la, welches unter erfindungsgemäßen Bedingungen durchgeführt wird, in der gleichen Reaktionszeit nur ein sehr viel geringerer Umsatz erzielt.
Beispiel 2
Oxidation von 4-Cyanpyridin zu 4-Cyanpyridin-N-oxid
26,0 g 4-Cyanpyridin und 1 g Novozym 435® werden in der oben beschriebenen Versuchsapparatur zusammen mit 400 ml Butylacetat vorgelegt. Unter Rühren bei 45°C und vermindertem Druck von 40 mbar läßt man 30 g Wasserstofφeroxid in Form einer 35 %-igen wäßrigen Lösung innerhalb von 2 Stunden zutropfen. Anschließend wird 3 Stunden bei 45 °C nachgerührt. Während der gesamten Zeit wird das bei der Reaktion freiwerdende und das in die Reaktion eingetragene Wasser kontinuierlich über den Wasserabscheider abgetrennt. 4-Cyanpyridin wird mit einem Umsatz von 67 % und einer Selektivität von 95 % zu 4-Cyanpyridin-N-oxid umgesetzt.
Beispiel 3
Oxidation von Cyclohexen zu Cyclohexenoxid
20,6 g Cyclohexen und 1 g Novozym 435 werden in der oben beschriebenen Versuchsapparatur zusammen mit 400 ml Butylacetat vorgelegt. Unter Rühren bei 45°C und vermindertem Druck von 70 mbar läßt man 30 g Wasserstoffperoxid in Form einer 35 %-igen wäßrigen Lösung innerhalb von 2 Stunden zutropfen. Anschließend wird 3 Stunden bei 48°C nachgerührt. Während der gesamten Zeit wird das bei der Reaktion freiwerdende und das in die Reaktion eingetragene Wasser kontinuierlich über den Wasserabscheider abgetrennt. Cyclohexen wird mit einem Umsatz von 100 % und einer Selektivität von 97 % zu Cyclohexenoxid umgesetzt.
Beispiel 4
Oxidation von Styrol zu Styroloxid
26,3 g Styrol und 1 g Novozym 435 werden in der oben beschriebenen Versuchsapparatur zusammen mit 400 ml Butylacetat vorgelegt. Unter Rühren bei 45°C und vermindertem Druck von 40 mbar läßt man 30 g Wasserstofφeroxid in Form einer 35 %-igen wäßrigen Lösung innerhalb von 2 Stunden zutropfen. Anschließend wird 3 Stunden bei 45°C nachgerührt. Während der gesamten Zeit wird das bei der Reaktion freiwerdende und das in die Reaktion eingetragene Wasser kontinuierlich über den Wasserabscheider abgetrennt. Styrol wird mit einem Umsatz von 70 % und einer Selektivität vom 99 % zu Styroloxid umgesetzt.
Beispiel 5
Oxidation von Cyclohexanon zu Caprolacton
24,6 g Cyclohexanon und 1 g Novozym 435® werden in der oben beschriebenen Versuchsapparatur zusammen mit 400 ml Butylacetat vorgelegt. Unter Rühren bei 45°C und vermindertem Druck von 40 mbar läßt man 30 g Wasserstofφeroxid in Form einer 35 %-igen wäßrigen Lösung innerhalb von 2 Stunden zutropfen. Anschließend wird 4 Stunden bei 40°C nachgerührt. Während der gesamten Zeit wird das bei der Reaktion freiwerdende und das in die Reaktion eingetragene Wasser kontinuierlich über den Wasserabscheider abgetrennt. Cyclohexanon wird mit einem Umsatz von 45 % und einer Selektivität von 82 % zu Caprolacton umgesetzt.
Beispiel 6
Oxidation von Thioanisol zu Methylphenylsulfoxid
18,8 g Thioanisol und 1 g Novozym 435 werden in der oben beschriebenen Versuchsapparatur zusammen mit 400 ml Butylacetat vorgelegt. Unter Rühren bei 45°C und vermindertem Druck von 40 mbar läßt man 18,5 g Wasserstofφeroxid in Form einer 35 %>-igen wäßrigen Lösung innerhalb von 2 Stunden zutropfen. Anschließend wird 4 Stunden bei 40°C nachgerührt. Während der gesamten Zeit wird das bei der Reaktion freiwerdende und das in die Reaktion eingetragene Wasser kontinuierlich über den Wasserabscheider abgetrennt. Thioanisol wird mit einem Umsatz von 93 % und einer Selektivität von 93 % zu Methylphenylsulfoxid umgesetzt.
Claims
1. Verfahren zur Oxidation von organischen Verbindungen mit Peroxycarbonsäuren, die in situ in Gegenwart von Enzymen durch Umsetzung von Wasser- stoffperoxid mit gesättigten ahphatischen Carbonsaureestern und/oder Mischungen der korrespondierenden Carbonsäuren und Alkohole erzeugt werden, dadurch gekennzeichnet, daß die gesättigten aliphatischen Carbonsäuren geradkettig oder verzweigt sind und 1 bis 4 C-Atome aufweisen und die Alkohole die Formel R-OH besitzen, wobei
R für einen geradkettigen oder verzweigten C3-Cι8- Alkylrest steht, der gegebenenfalls durch ein oder zwei Reste OR' substituiert ist, wobei R' unabhängig voneinander Wasserstoff oder einen C2-C4-Acylrest bedeuten, und dessen C3-Cι8-Alkylkette gegebenenfalls durch ein oder mehrere Sauerstoffatome unterbrochen ist, und
daß das bei der Umsetzung entstehende und/oder zugeführte Wasser ganz oder teilweise entfernt wird.
2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, daß gesättigte alipha- tische Carbonsäureester und/oder Mischungen der korrespondierenden Carbonsäuren und Alkohole eingesetzt werden, bei denen die Carbonsäuren 2 oder 3 C-Atome aufweisen.
3. Verfahren nach Anspruch 1 oder 2, dadurch gekennzeichnet, daß gesättigte aliphatische Carbonsäureester und/oder Mischungen der korrespondierenden Carbonsäuren und Alkohole eingesetzt werden, bei denen R für einen geradkettigen oder verzweigten C4-C8-Alkylrest, einen
Monohydroxy-substituierten C3-C6-Alkylrest, insbesondere einen ω- Hydroxy-CvC(,-Alkylrest, oder einen C3-C6-Alkylrest steht, der durch zwei Hydroxy- oder O-(C2-C4)-Acylreste substituiert ist.
4. Verfahren nach Anspruch 3, dadurch gekennzeichnet, daß Butylacetat und/oder Essigsäure-Butanol-Mischungen eingesetzt werden.
5. Verfahren nach einem oder mehreren der Ansprüche 1 bis 4, dadurch gekenn- zeichnet, daß als Enzyme Hydrolasen, bevorzugt Esterasen, Lipasen oder
Proteasen eingesetzt werden.
6. Verfahren nach Anspmch 5, dadurch gekennzeichnet, daß als Enzym Candida antarctica eingesetzt wird.
7. Verfahren nach einem oder mehreren der Ansprüche 1 bis 7, dadurch gekennzeichnet, daß das bei der Umsetzung entstehende und/oder dem Reaktionssystem zugeführte Wasser aus dem Reaktionssystem entfernt wird, so daß der Wassergehalt des Reaktionssystems bei 0,001 bis 10 Gew.-%, bevorzugt 0,01 bis 3 Gew.-% und insbesondere 0,1 bis 2 Gew.-% liegt.
8. Verfahren nach einem oder mehreren der Ansprüche 1 bis 6, dadurch gekennzeichnet, daß Pyridine zu Pyridin-N-oxiden, Olefine zu Oxiranen, Sulfide zu Sulfoxiden und Sulfonen oder Ketone zu Estern oxidiert werden.
9. Verfahren nach Anspruch 8, dadurch gekennzeichnet, daß Pyridine der Formel la zu Pyridin-N-oxiden der Fomiel I b oxidiert werden,

R2, R3, R4 und R5 unabhängig voneinander H, Ci-Cio-Alkyl, bevorzugt Cι-C6- Alkyl C3-Go-Cycloalkyl, bevorzugt C3-C6-Cycloalkyl, Cö-C^-Aryl, bevorzugt Phenyl, OH, Ci-Qo-Alkoxy, bevorzugt Cι-C6-Alkoxy, C(=O)-C1-C10-Alkyl, bevorzugt C(=O)-C,-C6-Alkyl, C(=O)-O-C,- Cio-Alkyl, bevorzugt C(O)-0-C,-C6-Alkyl, CN, NO2, F, Cl, Br, C(=O)N(R')2, wobei R' gleich oder verschieden sind und für Wasserstoff oder einen Ci-Cio- Alkylrest, bevorzugt Cι-C6- Alkylrest, stehen, NH-C(=0)R', wobei R' die zuvor genannte Bedeutung hat, bedeuten, oder aber jeweils zwei benachbarte Reste aus der Gruppe von R2"5 gemeinsam einen C2-C20- Alkylen- oder C2-C2o-Alkylidenrest, bevorzugt einen C3-Cιo-Alkylen- oder C3-Cιo-Alkylidenrest, bilden, wobei diese Alkylen- und Alkylidenreste ein- oder mehrfach durch O, COO oder CO unterbrochen sein können.
11. Verfahren nach Anspmch 8, dadurch gekennzeichnet, daß Sulfide der Formel lila zu Sulfoxiden der Formel Illb und zu Sulfonen der Formel IIIc oxidiert werden,

Ci2-Aryl, bevorzugt Phenyl, bedeuten oder aber beide Substituenten
R7 und R8 gemeinsam einen C2-C2o-Alkylen- oder C2-C20- Alkylidenrest, bevorzugt C4-Cιo-Alkylen- oder C4-Cιo-Alkylidenrest bilden, wobei diese Alkylen- und Alkylidenreste ein- oder mehrfach durch O, COO oder CO unterbrochen sein können.
12. Verfahren nach Anspmch 8, dadurch gekennzeichnet, daß Ketone der Formel IVa zu Estern der Fomiel IVb oxidiert werden,

C6-Alkyl, C3-CiQ-Cycloalkyl, bevorzugt C3-C6-Cycloalkyl, oder C6- Ci2-Aryl, bevorzugt Phenyl, bedeuten oder aber beide Substituenten R9 und R10 gemeinsam einen C2-C2o-Alkylen- oder C2-C20- Alkylidenrest, bevorzugt C3-C1Q- Alkylen- oder C3-Cιo-Aιkylidenrest bilden, wobei diese Alkylen- und Alkylidenreste ein- oder mehrfach durch O, COO oder CO unterbrochen sein können.
