Abstract
Die vorliegende Erfindung betrifft neue Verbindungen, die als Arzneimittel geeignet sind, Verfahren zu ihrer Herstellung und ihre Verwendung als Arzneimittel, insbesondere als antivirale Mittel.

SUBSTITUIERTE INDOLSULFONAMIDE ALS ANTIVIRALE MITTEL
Die vorliegende Erfindung betrifft neue Verbindungen, die als Arzneimittel geeignet sind, Verfahren zu ihrer Herstellung und ihre Verwendung als Arzneimittel, insbe- sondere als antivirale Mittel.
α,ß-Naphthyl verknüpfte Phenylsulfonamide sind überwiegend aus phototechnischen Publikationen bekannt [vgl. hierzu JP-06 122 669-A2, EP-684 515-A1; JP- 59 174 836-A2, DE-2 902 074, US-3 925 347, US-4 035 401, US-3 622 603, US- 3 482 971. EP-284 130].
Die WO 90/09 787 offenbart Sulfonamide als Radio- oder Chemosensibilisierungs- mittel und ihre Verwendung bei der Behandlung von Tumoren.
Außerdem ist die Verbindung N-[4-[[[5-(Dimethylamino)-l-naphthalenyl]sulfonyl]- amino]phenyl]-acetamid bekannt (J. Inst. Chem. (India) (1976), 48, Pt 6, 280-5).
Die Erfindung betrifft Verbindungen der allgemeinen Formel ( I )

Die erfindungsgemäßen Stoffe können auch als Salze vorliegen. Im Rahmen der Erfindung sind physiologisch unbedenkliche Salze bevorzugt.
Physiologisch unbedenkliche Salze können Salze der erfindungsgemäßen Verbindungen mit anorganischen oder organischen Säuren sein. Bevorzugt werden Salze
mit anorganischen Säuren wie beispielsweise Salzsäure, Bromwasserstoffsäure, Phosphorsäure oder Schwefelsäure, oder Salze mit organischen Carbon- oder Sulfon- säuren wie beispielsweise Essigsäure, Maleinsäure, Fumarsäure, Äpfelsäure, Zitronensäure, Weinsäure, Milchsäure, Benzoesäure, oder Methansulfonsäure, Ethansul- fonsäure, Phenylsulfonsäure, Toluolsulfonsäure oder Naphthalindisulfonsäure.
Physiologisch unbedenkliche Salze können ebenso Metall- oder Ammoniumsalze der erfindungsgemäßen Verbindungen sein. Besonders bevorzugt sind z.B. Natrium-, Kalium-, Magnesium- oder Calciumsalze, sowie Ammoniumsalze, die abgeleitet sind von Ammoniak, oder organischen Aminen, wie beispielsweise Ethylamin, Di-bzw.
Triethylamin, Di- bzw. Triethanolamin, Dicyclohexylamin, Dimethylaminoethanol, Arginin, Lysin, Ethylendiamin oder 2-Phenylethylamin.
Die erfϊndungsgemäßen Verbindungen der allgemeinen Formel (I) können in ver- schiedenen stereochemischen Formen auftreten, die sich entweder wie Bild und
Spiegelbild (Enantiomere), oder die sich nicht wie Bild und Spiegelbild (Diastereo- mere) verhalten. Die Erfindung betrifft sowohl die Antipoden als auch die Racem- formen sowie die Diastereomerengemische. Die Racemformen lassen sich ebenso wie die Diastereomeren in bekannter Weise in die stereoisomer einheitlichen Bestandteile trennen.
Weiterhin können bestimmte Verbindungen in tautomeren Formen vorliegen. Dies ist dem Fachmann bekannt, und derartige Verbindungen sind ebenfalls vom Umfang der Erfindung umfaßt.
(Cι-Cg)Alkyl steht im Rahmen der Erfindung im allgemeinen für geradkettige oder verzweigtkettige Kohlenwasserstoffreste mit 1 bis 6 Kohlenstoffatomen. Entsprechend stehen (Cj-C4)Alkyl bzw. (Cj-C3)Alkyl im Rahmen der Erfindung im allgemeinen für geradkettige oder verzweigtkettige Kohlenwasserstoffreste mit 1 bis 4, bzw. 1 bis 3 Kohlenstoffatomen Es seien beispielsweise genannt: Methyl, Ethyl, Pro-
pyl, Isopropyl, Butyl, Isobutyl, sek.-Butyl, tert.-Butyl, Pentyl, Isopentyl, Hexyl und Isohexyl.
(C3-C6)Cycloalkyl steht für Cycloalkylgruppe mit 3 bis 6 Kohlenstoffatomen Schließt beispielsweise ein: Cyclopropyl, Cyclopentyl und Cyclohexyl. Bevorzugt ist Cyclo- propyl.
Die (Cj-C6)Alkoxygruppe, wie sie in der vorliegenden Erfindung verwendet wird, und wie sie auch in den Definitionen (Cι-C6)Alkoxycarbonyl verwendet wird, schließt beispielsweise geradkettige oder verzweigtkettige Alkoxygruppen mit 1 bis
6 Kohlenstoffatomen ein, besonders bevorzugt Alkoxygruppen mit 1 bis 4 Kohlenstoffatomen ((Cj-C4)Alkoxy), noch bevorzugter Alkoxygruppen mit 1 bis 3 Kohlenstoffatomen ((Cj-C3)Alkoxy). Beispielsweise können erwähnt werden Methoxy, Ethoxy, Propoxy, Isopropoxy, Butoxy, Isobutoxy, Pentoxy, Isopentoxy, Hexoxy und Isohexoxy. Bevorzugt ist Methoxy, Ethoxy und Propoxy.
Mono- oder Di(Cι-Cg)Alkylamino schließt im Rahmen der Erfindung solche ein, deren Alkylgruppen 1 bis 6 Kohlenstoffatome aufweisen. Dabei kann es sich um symmetrische oder unsymmetrische Alkylaminogruppen handeln, wie beispielsweise Dimethylamino, Diethylamino, Methylethylamino usw. Dies gilt auch für den Mono- oder Di(Cι-C6)Alkylamino-Teil in der Mono- oder Di(Cj-C6)-Alkylaminocarbonyl- Gruppe.
(C6-Cι0)Aryl steht im Rahmen der Erfindung für einen aromatischen Rest mit 6 bis 10 Kohlenstoffatomen. Bevorzugte Arylreste sind Phenyl und Naphthyl.
5- bis 10-gliedriges Heteroaryl steht im Rahmen der Erfindung für 5- bis 10-gliedrige Heteroatome enthaltende Ringe, die 1 bis 8 Heteroatome im Ring enthalten können, die ausgewählt werden aus O, S und N und schließen beispielsweise ein Pyridyl. Furyl, Thienyl, Pyrrolyl, Imidazolyl, Oxazolyl, Thiazolyl, Isoxazolyl, Triazolyl.
Thiadiazolyl, Pyridazinyl, Pyrazinyl, Pyrimidinyl, Indolicenyl, IndolyL
Benzo[b]thienyl, Benzimdiazolyl, Pyridoimidazolyl, Indazolyl, Chinolyl, Isochinolyl, Naphthyridinyl, Chinazolinyl, etc.
5- bis 6-gliedrige stickstoffhaltige Heterocyclen schließen beispielsweise ein: Pyrro- lidin, Piperidin, Piperazin, Morpholin, Pyridyl, Furyl, Thienyl, Pyrolyl, Imidazolyl,
Pyrazolyl, Pyrazinyl, Pyrimidinyl, Pyridazinyl, etc.
Halogen schließt im Rahmen der Erfindung Fluor, Chlor, Brom und Iod ein. Bevorzugt sind Chlor oder Fluor.
Bezüglich (C6-Cjo)Arylsulfonyl und -carbonyl sei auf die oben erwähnten Definitionen für (C6-Cjo)Aryl verwiesen.
(Ci-Cg) Alkanoyl sowie (C]-C6) Alkanoyl in der Definition (Cι-Cg)Alkanoyloxy und (Cj-Cg)Alkanoylamino steht im Rahmen der Erfindung für geradkettiges oder ver- zweigtkettiges Alkanoyl mit 1 bis 6 Kohlenstoffatomen. Beispielsweise seien erwähnt: Formyl, Acetyl, Propanoyl, Butanoyl,, Pentanoyl, Pivaloyl und Hexanoyl.
Mit dem Begriff "Alkandiylgruppe mit bis zu 6 Kohlenstoffatomen" werden hier geradkettige oder verzweigtkettige Kohlenwasserstoff-Gruppen bezeichnet, die an zwei Positionen mit weiteren Resten verknüpft sind. Beispiele für Alkandiylgruppen sind: -CH2-CH2-, -CH2-CH2-CH2-, -C(CH3)2-CH2-, -CH(CH3)-CH2-, -C(CH3)2- CH2-CH2-, -CH(CH3)-CH2-CH2- etc.
Aminoschutzgruppen im Rahmen der Erfindung sind die üblichen in der Peptid—
Chemie verwendeten Aminoschutzgruppen.
Hierzu gehören bevorzugt: Benzyloxycarbonyl, 3,4-Dimethoxybenzyloxycarbonyl,
3,5-Dimethoxybenzyloxycarbonyl, 2,4-Dimethoxybenzyloxycarbonyl, 4-Methoxy- benzyloxycarbonyl, 4-Nitrobenzyloxycarbonyl, 2-Nitrobenzyloxycarbonyl, 2-Nitro-
4,5-dimethoxybenzyloxycarbonyl, Methoxycarbonyl, Ethoxycarbonyl, Propoxy-
carbonyl, Isopropoxycarbonyl, Butoxycarbonyl, Isobutoxycarbonyl, tert.Butoxy- carbonyl, Allyloxycarbonyl, Vinyloxycarbonyl, 2-Nitrobenzyloxycarbonyl, 3,4,5- Trimethoxybenzyloxycarbonyl, Cyclohexoxycarbonyl, 1 , 1 -Dimethylethoxycarbonyl, Adamantylcarbonyl, Phthaloyl, 2,2,2-Trichlorethoxycarbonyl, 2,2,2-Trichlor-tert- butoxycarbonyl, Menthyloxycarbonyl, Phenoxycarbonyl, 4-Nitrophenoxycarbonyl, Fluorenyl-9-methoxycarbonyl, Formyl, Acetyl, Propionyl, Pivaloyl, 2-Chloracetyl, 2-Bromacetyl, 2,2,2-Trifluoracetyl, 2,2,2-Trichloracetyl, Benzoyl, 4-Chlorbenzoyl, 4-Brombenzoyl, 4-Nitrobenzoyl, Phthalimido, Isovaleroyl oder Benzyloxymethylen, 4-Nitrobenzyl, 2,4-Dinitrobenzyl oder 4-Nitrophenyl.
In einer bevorzugten Ausführungform schließt die Erfindung Verbindungen der Formeln ein
worin R1 und R2 die oben angegebene Bedeutung haben.
In einer weiteren bevorzugten Ausführungform schließt die Erfindung Verbindungen der allgemeinen Formel ( I ) ein, worin:

In einer bevorzugten Ausführungform schließt die Erfindung Verbindungen der allgemeinen Formel ( I ) ein, worin:

In einer bevorzugten Ausführungform schließt die Erfindung Verbindungen der all- gemeinen Formel (I) ein, worin R6 tert.-Butyl ist, das gegebenenfalls durch Halogen oder Hydroxy substituiert ist, und deren Salze.
Die Erfindung betrifft weiterhin Verfahren zur Herstellung der Verbindungen der Formel (I).
Im Verfahren (A) setzt man Verbindungen der allgemeinen Formel (II)
worin R1 wie oben definiert ist, mit Verbindungen der allgemeinen Formel (III)
worin R2 wie oben definiert ist, zu Verbindungen der allgemeinen Formel (I) um.
Die Reaktion wird bevorzugt in Gegenwart von Basen, wie Pyridin, Triethylamin und Hünig Base etc. durchgeführt.
Die Reaktion wird bevorzugt in einem Lösungsmittel wie Tetrahydrofuran, 1,4- Dioxan, Dichlormethan, etc. durchgeführt.
Die Reaktion wird bevorzugt in einem Temperaturbereich von -10°C bis 70°C durchgeführt.
Die Reaktion wird bevorzugt bei Normaldruck durchgeführt.
Im Verfahren (B) setzt man Verbindungen der allgemeinen Formel (Ia):
worin R2 wie oben definiert ist, und
R a für eine Gruppe steht, die aus den folgenden Formeln ausgewählt wird:

worin R3 wie oben definiert ist und A eine übliche Abgangsgruppe ist, in an sich bekannter Weise in Gegenwart einer Base zu Verbindungen der allgemeinen Formel (Ib) um:

A stellt dabei eine übliche, in nukleophilen Substitutionsreaktionen verwendete Abgangsgruppe dar, wie z.B. Halogen (z. B. Chlor, Brom, Jod), OTs (Ts = Tosyl) und OMes (Mes = Mesyl).
In der Reaktion bevorzugte Basen sind tertiäre Amine, wie Pyridin, Hünig Base etc., Alkalihydroxid und Alkalicarbonat.
Die Reaktion wird bevorzugt in inerten Lösungsmittel wie Tetrahydrofuran, 1,4- Dioxan, Dichlormethan, Dimethylformamid etc. durchgeführt.
Die Reaktion wird bevorzugt in einem Temperaturbereich von -10°C bis 100°C durchgeführt.
Die Reaktion wird bevorzugt bei Normaldruck durchgeführt.
Im Verfahren (C) setzt man Verbindungen der allgemeinen Formel (Ic)
worin R2 wie oben definiert ist und R1c für eine Gruppe steht, die aus den folgenden Formeln ausgewählt wird:
worin R3 wie oben definiert ist, durch Oxidation mit DDQ (2,3-Dichlor-5,6-dicyano- para-benzochinon) in an sich bekannter Weise zu Verbindungen der allgemeinen Formel (Id) um:

Die Reaktion wird bevorzugt in einem Lösungsmittel wie 1,4-Dioxan oder 1,2-
Dichlorethan durchgeführt.
Die Reaktion wird bevorzugt in einem Temperaturbereich von Raumtemperatur bis zum Siedepunkt des jeweiligen Lösemittels bei Normaldruck durchgeführt.

Im Verfahren (D) setzt man Verbindungen der allgemeinen Formel (Ie)
worin R2 wie oben definiert ist und R1e die folgende Formeln aufweist:

Alkalimetallhydroxiden schließen dabei beispielsweise Lithiumhydroxid, Natrium- hydroxid, Kaliumhydroxid, etc. ein, wobei Lithiumhydroxid bevorzugt ist.
Die Reaktion wird bevorzugt in homogenen wässrigen Lösemittelsystemen durchgeführt.
Die Reaktion wird bevorzugt in einem Temperaturbereich von Raumtemperatur bis
70°C durchgeführt.
Die Reaktion wird bevorzugt bei Normaldruck durchgeführt.
Im Verfahren (E) setzt man Verbindungen der allgemeinen Formel (If)
worin R2 wie oben definiert ist und R1f die folgende Formeln aufweist:
worin R3 für (C C6)Alkanoyl steht, in an sich bekannter Weise mit komplexen Metallhydriden zu Verbindungen der allgemeinen Formel (Ig) um:

In der Reaktion bevorzugt verwendete komplexe Metallhydride sind Lithiumaluminiumhydrid, Diisobutylaluminiumhydrid, etc.
Die Reaktion wird bevorzugt in einem Lösungsmittel wie Tetrahydrofuran, 1 ,4- Dioxan etc. durchgeführt.
Die Reaktion wird bevorzugt in einem Temperaturbereich von — 50°C bis 40°C durchgeführt.
Die Reaktion wird bevorzugt bei Normaldruck durchgeführt.
Die erfindungsgemäßen Verfahren können durch folgende Reaktionschemata illustriert werden.
Die Darstellung der Indol- und Indolin-Verbindungen kann wie folgt erfolgen:
Aus den unsubstituierten Indol- und Indolinverbindungen (R3 = Wasserstoff) sind dann durch Umsetzung mit R3-A die Verbindungen der allgemeinen Formel (I) wie oben beschrieben erhältlich.
Die Isoindolinverbindungen sind z.B. nach folgendem Schema erhältlich:
Aus den unsubstituierten Isoindolinverbindungen (R3 = Wasserstoff) sind dann durch Umsetzung mit R3-A die Verbindungen der allgemeinen Formel (I) wie oben beschrieben erhältlich.
Die Herstellung der Sulfonylchlorid-Ausgangsverbindungen der Formel (II) wird durch folgendes Reaktionsschema illustriert:

Dabei erfolgt die Herstellung des Sulfonylchlorids 1 z.B. gemäß A. L.Borrer, E. Chinoporos, M. Filosa, S. R. Herrchen, C. R. Petersen, C. A. Stern, J Org. Chem. 53, 2047 (1988).
Die Herstellung des Sulfonylchlorids 3 kann in Analogie zur obigen Umsetzung erfolgen.
Die Herstellung des Sulfonylchlorids 2 erfolgt z.B. gemäß P. R. Carlier, M. P. Lockshin, M. P. Filosa, J. Org. Chem. 59, 3232 (1994).
Aus diesen Verbindungen sind dann nach Umsetzung mit den Aminen der Formel (III), Verseifen der Acetylgruppe z.B. mit LiΟH/H2Ο und anschließender Umsetzung mit R3-A dieVerbindungen der allgemeinen Formel (I) erhältlich.
Die Herstellung der Verbindungen der allgemeinen Formel (III) wird beispielsweise anhand folgenden Reaktionsschemas erläutert:

Die Herstellung des Anilins 4 erfolgt z.B. gemäß US-Patent Nr. 3979202.
Die Herstellung des Anilins 6 erfolgt z.B. gemäß S. Rajappa, R. Sreenivasan, A.
Khalwadekar, J. Chem. Res. Miniprint 5, 1657 (1986).
Die Herstellung des Anilins 7 erfolgt z.B. gemäß WO 9631462.
Die Herstellung des Anilins 8 erfolgt z.B. gemäß R. W. Hartmann, M. Reichert, S. Goehring, Ewr. J Med. Chem Chim. Ther. 29, 807 (1994).
Die Herstellung der Aniline 5 und 9 erfolgt in analoger Weise.
Bezüglich der genauen Reaktionsbedingungen sei auf die Beispiele und Ausgangsbeispiele verwiesen.
Die Erfindung betrifft weiterhin Verbindungen der Formel (I) zur Verwendung als Arzneimittel.
Die Erfindung betrifft weiterhin eine pharmazeutische Zusammensetzung, die eine Verbindung der allgemeinen Formel (I) in Mischung mit mindestens einem pharmazeutisch verträglichen Träger oder Exzipienten umfaßt.
Die Erfindung betrifft weiterhin die Verwendung der Verbindung der allgemeinen Formel (I) zur Herstellung eines Arzneimittels.
Die Erfindung betrifft weiterhin dieVerwendung einer Verbindung der allgemeinen Formel (I) zur Herstellung eines Arzneimittels zur Behandlung von viralen Infektionen, insbesondere Infektionen durch Cytomegalieviren.
Die erfindungsgemäßen Verbindungen der allgemeinen Formeln (I) zeigen ein nicht vorhersehbares überraschendes Wirkspektrum. Sie zeigen eine antivirale Wirkung gegenüber Vertretern der Gruppe der Herpes viridae, besonders gegenüber dem humanen Cytomegalievirus (HCMV). Sie eignen sich somit zur Behandlung und Prophylaxe von Erkrankungen, die durch Herpes- Viren, insbesondere Erkrankungen, die durch humanes Cytomegalievirus (HCMV) hervorgerufen werden.
Die Anti-HCMV-Wirkung wurde in einem Screening-Testsystem in 96-Well-Mikro- titerplatten unter Zuhilfenahme von humanen embryonalen Lungenfibroblasten
(HELF)-Zellkulturen bestimmt. Der Einfluß der Substanzen auf die Ausbreitung des cytopathogenen Effektes wurde im Vergleich zu der Referenzsubstanz Ganciclovir (CymeveneR-Natrium), einem klinisch zugelassenen anti-HCMV-Chemotherapeuti- kum, bestimmt (EC50, entsprechend der effektiven Konzentration, bei der eine 50 %- ige Inhibierung der Virusaktivität erreicht wird).
Die in DMSO (Dimethylsulfoxid) gelösten Substanzen (50 mM) werden auf Mikro- titerplatten (96-Well) in Doppelbestimmungen (4 Substanzen/Platte) untersucht. Toxische und cytostatische Substanzwirkungen werden dabei miterfaßt (CC50, entsprechend der Konzentration, bei der die Hälfte der Zellen aufgrund der Substanzgabe zerstört werden). Nach den entsprechenden Substanzverdünnungen (1 :2) auf der Mikrotiterplatte wird eine Suspension von 50 - 100 HCMV-infizierten HELF-Zellen und 30 x 105 nichtinfizierten HELF-Zellen in Eagle's MEM mit 10% fötalem Kälberserum in jedes Näpfchen gegeben, und die Platten bei 37°C in einem CO2-Brutschrank über mehrere Tage inkubiert. Nach dieser Zeit ist der Zellrasen in den substanzfreien Viruskontrollen, ausgehend von 50 - 100 infektiösen Zentren, durch den cytopathogenen Effekt des HCMV völlig zerstört (100% CPE). Nach einer Anfärbung mit Neutralrot und Fixierung mit Formalin/Methanol werden die Platten mit Hilfe eines Projektions-Mikroskopes (Plaque- Viewer) ausgewertet.
Im folgenden werden die Ergebisse für zwei beispielhafte Verbindungen gegeben:
Die erfindungsgemäßen Verbindungen stellen somit wertvolle Wirkstoffe zur Behandlung und Prophylaxe von Erkrankungen dar, die durch humanes Cytomegalievirus ausgelöst werden. Als Indikationsgebiete können beispielsweise genannt werden:
1) Behandlung und Prophylaxe von HCMV-Infektionen bei AIDS-Patienten (Retinitis, Pneumonitis, gastrointestinale Infektionen).
2) Behandlung und Prophylaxe von Cytomegalievirus-Infektionen bei Knochenmark- und Organtransplantationspatienten, die an einer HCMV-Pneumonitis, -Enzephalitis, sowie an gastrointestinalen und systemischen HCMV-Infektionen oft lebensbedrohlich erkranken.
3) Behandlung und Prophylaxe von HCMV-Infektionen bei Neugeborenen und
Kleinkindern.
4) Behandlung einer akuten HCMV-Infektion bei Schwangeren.
In vivo-Wirkung
Tiere
5 Wochen alte männliche Mäuse, Stamm NOD/LtSz-Prkdc(scid)/J, wurden von einem kommerziellen Züchter (The Jackson Lab., Bar Harbor) bezogen. Die Tiere wurden unter sterilen Bedingungen (einschließlich Einstreu und Futter) in Isolatoren gehalten.
Virus/Infektion
Murines Cytomegalievirus (MCMV), Stamm Smith, wurde in vivo (BALB/c) passagiert und über eine fraktionierte Zentrifugation aufgereinigt. Der Titer wurde mit Hilfe eines Plaqueassays auf primären embryonalen Mäusefibroblasten untersucht.
Die Infektion der Mäuse erfolgte mit einer Dosis von 5xl05 pfu in einem Gesamtvolumen von 0,2 ml intraperitoneal. Diese Dosis führt bei 100% der infizierten Tiere nach ca. 1 1 Tagen zum Tode.
Behandlung/ Auswertung
24 Stunden nach der Infektion wurden die Mäuse über einen Zeitraum von 8 Tagen zweimal täglich (morgens und abends) per os mit Substanz behandelt. Die Dosis betrug 25 mg/kg Körpermasse, das Applikationsvolumen 10 ml/kg Körpermasse. Die Formulierung der Substanzen erfolgte in Form einer 0,5%igen Tylosesuspension. 16
Stunden nach der letzten Substanzapplikation wurden die Tiere schmerzlos getötet und Speicheldrüse, Leber und Niere entnommen.
Aus 25 mg der Gewebe wurde über Phenol/Chloroform-Extraktion genomische DNA aufgereinigt. Die Quantifizierung der DNA erfolgte photometrisch und mit Hilfe der
Formel OD260x50=mg/ml.
Die Reinheit der DNA wurde über den Quotienten OD260/OD280 kontrolliert und die DNA anschließend mit Tris-EDTA pH = 8,0 eingestellt.
Die Quantifizierung der MCMV-DNA erfolgte mittels DNA-Dot-Blot-Hybridisie- rung. Als Sonde wurde ein Digoxygenin-gelabeltes (Boehringer-Mannheim, ebenfalls aufgeführte Puffer, wenn nicht anders beschrieben) 1,2 kb Fragment aus dem Bereich MCMV, Smith, Hindlll J, verwendet. Die Detektion der Signale erfolgte mittels Chemolumineszenz. Dafür wurde die Membran für 3 Minuten in 1 x Digoxy- genin- Waschpuffer 1 gewaschen. Im Anschluß wurden die Filter für 30 Minuten bei Raumtemperatur unter Schütteln in 1 x Digoxygenin Blockierungslösung inkubiert. Die Filter wurden danach für 30 Minuten in 20 ml/100 cm2 Membran mit der Anti- DIG-Alkalische-Phosphatase-Konjugatlösung (1 :20000 in 1 x Digoxygenin Blockie- rungslösung) inkubiert. 2 je 15 Minuten dauernde Waschschritte mit 1 x Digoxygenin- Waschpuffer schlössen sich an. Es folgten 5 Minuten Aquilibrierung der Filter in 1 x Digoxygenin-Detektionspuffer und die Detektion mittels 1 ml / 100 cm2 Membranfläche 1 :100 verdünnte CDP-Star-Lösung. Nach Ausstreichen der CDP-Star- Lösung und 5 minütiger Inkubation in einer dunklen Box erfolgte der Nachweis der Chemolumineszenz bzw. die Auswertung mittels Röntgenfilm (Kodak) oder
Lumilmager (Boehringer Mannheim).
Alle Ergebnisse wurden statistisch gesichert (Varianzanalyse mittels Statistika; StatSoft Inc.).
Die neuen Wirkstoffe können in bekannter Weise in die üblichen Formulierungen überfuhrt werden, wie Tabletten, Dragees, Pillen, Granulate, Aerosole, Sirupe, Emulsionen, Suspensionen und Lösungen, unter Verwendung inerter, nicht-toxischer, pharmazeutisch geeigneter Trägerstoffe oder Lösemittel. Hierbei soll die therapeutisch wirksame Verbindung jeweils in einer Konzentration von etwa 0,5 bis 90 Gew.-% der Gesamtmischung vorhanden sein, d.h. in Mengen, die ausreichend sind, um den angegebenen Dosierungsspielraum zu erreichen.
Die Formulierungen werden beispielsweise hergestellt durch Verstrecken der Wirkstoffe mit Lösemitteln und/oder Trägerstoffen, gegebenenfalls unter Verwendung von Emulgiermitteln und/oder Dispergiermitteln, wobei z.B. im Fall der Benutzung von Wasser als Verdünnungsmittel gegebenenfalls organische Lösemittel als Hilfslösemittel verwendet werden können.
Die Applikation erfolgt in üblicher Weise, vorzugsweise oral, parenteral oder topisch, insbesondere perlingual, intravenös oder intravital gegebenenfalls als Depot in einem Implantat.
Für den Fall der parenteralen Anwendung können Lösungen der Wirkstoffe unter Verwendung geeigneter flüssiger Trägermaterialien eingesetzt werden.
Im allgemeinen hat es sich als vorteilhaft erwiesen, bei intravenöser Applikation Mengen von etwa 0,001 bis 10 mg/kg, vorzugsweise etwa 0,01 bis 5 mg/kg Körpergewicht zur Erzielung wirksamer Ergebnisse zu verabreichen, und bei oraler Applikation beträgt die Dosierung etwa 0,01 bis 25 mg/kg, vorzugsweise 0,1 bis 10 mg/kg Körpergewicht.
Trotzdem kann es gegebenenfalls erforderlich sein, von den genannten Mengen abzuweichen, und zwar in Abhängigkeit vom Körpergewicht bzw. der Art des Applikationsweges, vom individuellen Verhalten gegenüber dem Medikament, der Art von dessen Formulierung und dem Zeitpunkt bzw. Intervall, zu welchem die Verabreichung erfolgt. So kann es in einigen Fällen ausreichend sein, mit weniger als der vorgenannten Mindestmenge auszukommen, während in anderen Fällen die genannnte obere Grenze überschrittten werden muß. Im Falle der Applikation größerer Mengen kann es empfehlenswert sein, diese in mehreren Einzelgaben über den Tag zu verteilen.
Gegebenenfalls kann es sinnvoll sein, die erfindungsgemäßen Verbindungen mit anderen Wirkstoffen zu kombinieren.

Ausgangsverbindungen
A) Sulfonylchloride
N-Acetyl-indolin-5-sulfonylchlorid wurde nach Literaturverfahren (A.L. Borrer, E. Chinoporos, M. Filosa, S.R. Herrchen, C.R. Petersen, CA. Stern, J. Org. Chem. 53, 2047 (1988)) ebenso wie N-Acetyl-indolin-6- sulfonylchlorid (P.R. Carlier, M.P. Lockshin, M.P. Filosa, J. Org. Chem. 59,
3232 (1994)). N-Acetyl-isoindolin-5-sulfonylchlorid wurde aus N- Acetylisoindolin und Chlorsulfonsäure in Analogie zu N-Acetyl-indolin-5- sulfonylchlorid hergestellt.
B) Anüine
Die erforderlichen Aniline wurden nach Literaturverfahren hergestellt (Monsanto, US-A 3 979 202, S. Rajappa, R. Sreenivasan, A. Khalwadekar, J. Chem. Res. Miniprint 5, 1657 (1986), WO 96/31462, R.W. Hartmann, M. Reichert, S. Göring, Eur. J. Med. Chem. Chim. Ther. 29, 807 (1994)) wie in den obigen Schemata dargestellt.
Beispiele
A) N-Acetyl-(iso)indolin-suIfonamide
Beispiel 26
N-(N-Acetylindolin-5-sulfonyl)-N'-(3-fluor-2,2-dimethylpropanoyl)-l,3-diaminoben- zol
Eine Lösung von 3,50 g (16,65 mmol) N-3-Fluor-2,2-dimethylpropanoyl)-l,3-diami- nobenzol in 10 ml trockenem THF wurde tropfenweise zu einer Lösung von 3,93 g (15,13 mmol) N-Acetyl-indolin-5-sulfonylchlorid und 5,99 g (75,67 mmol) Pyridin in 50 ml trockenem THF bei 0°C gegeben. Das Eisbad wurde entfernt, und das Rühren wurde bei Raumtemperatur für 24 Stunden fortgesetzt. Dann wurde das
Lösungsmittel und überschüssiges Pyridin im Vakuum entfernt. Die resultierende Aufschlämmung wurde mit einer Mischung von 25 ml Ether, 5 ml Ethylacetat und 2- molarer wäßriger Salzsäure behandelt. Das kristalline Produkt wurde isoliert und nacheinander mit Wasser und Ether gewaschen. 4,90 g (11,30 mmol, 75 % Ausbeute) eines schwach blaß rosa-farbenen Feststoffes wurden erhalten.
Rf: 0.40 (Ethylacetat).

In der gleichen Weise wurden die anderen N-Acetyl-indolin- und isoindolin- sulfonamide der Beispiele 25, 28, 32, 33, 18, 36, 56, 50, 49, 63, 61 und 59, wie in der Tabelle unten gezeigt, hergestellt.
B) N-Ethyl(iso)indolin-sulfonamide
Beispiel 19
N-(N-Ethylindolin-5-sulfonyl)-N'-(3-fluor-2,2-dimethylpropanoyl)-l,3-diaminoben- zol
Bei 0°C wurde eine Lösung von 1,0 g (2,31 mmol) N-(yV-Acetylindolin-5-sulfonyl)- N'-(3-fluor-2,2-dimethylpropanoyl)-l,3-diaminobenzol in 30 ml trockenem THF mit
2,80 ml (2,77 mmol) einer 1 -molaren Lösung von Lithiumaluminiumhydrid in THF
umgesetzt. Das Eiswasserbad wurde entfernt und das Rühren bei Raumtemperatur für 2 Stunden fortgesetzt. Die Reaktion wurde durch Zugabe von Methanol beendet. Die Mischung wurde mit Ethylacetat verdünnt und nacheinander mit wäßriger Seignette-Salzlösung, wäßriger (5 %iger) Natriumhydrogenphosphatlösung, Wasser und Kochsalzlösung gewaschen und über wasserfreiem Natriumsulfat getrocknet.
Das Produkt wurde durch präparative HPLC gereinigt. 105 mg (0,25 mmol, 11 % Ausbeute) eines weißen Feststoffes wurden erhalten.
Rj: 0,17 (Cyclohexan/Ethylacetat, 1:1). 'H-NMR (300 MHz, DMSO-d6, δ/ppm): 9.87 (1H, s), 9.31 (1H, s), 7.51 (1H, t), 7.42

In der gleichen Weise wurden die anderen N-Ethylindolin- und -isoindolinsulfon- amide der Beispiele 15, 20, 35, 30, 34, 8, 11, 27, 46, 41, 40, 62, 60 und 57 der folgenden Tabelle erhalten.
C) (Iso)Indolin-sulfonamide
Beispiel 16
4-[ N-(5-Indolinsulfonyl)]amino-N-(tert. -butyl)benzoesäureamid
300 mg (0,72 mmol) [N-(N-Acetylindolin-5-sulfonyl)]amino-N-tert.-butyl)benzoe- säureamid wurden in 21 ml wäßriger (5 %ig) Lithiumhydroxidlösung gelöst. Die Mischung wurde bei 60°C für 24 Stunden gehalten. Nach Abkühlen wurde wäßrige (5 %ige) Natriumhydrogenphosphatlösung hinzugegeben. Die wäßrige Schicht wurde mit Ethylacetat extrahiert. Die vereinigten organischen Extrakte wurden mit Wasser und Kochsalzlösung nacheinander gewaschen und über wasserfreiem
Natriumsulfat getrocknet. Das Produkt wurde aus Ether umkristallisiert. 230 mg (0,62 mmol, Ausbeute 85 %) eines weißen Feststoffes wurden erhalten.
Rf: 0.56 (Ethylacetat). 'H-NMR (400 MHz, DMSO-d6, δ/ppm): 10.18 (1H, s), 7.63 (2H, d), 7.53 (1H, s),

In der gleichen Weise wurden die anderen Indolin und Isoindolinsulfonamide der Beispiele 24, 38, 23, 37, 31, 55, 54, 53, 58, 65 und 64 der folgenden Tabelle erhalten.
D) N-Acetylindol-sulfonamide
Beispiel 2
4- [N- Acetylindol-5 -sulfonyl] amino-N-(tert. -butyl)benzoesäureamid
Eine Mischung von 200 mg (0,481 mmol) 4-[N-Acetylindolin-5-sulfonyl]amino-N- tert.-butyl)benzoesäureamid und 164 mg (0,722 mmol) DDQ in 10 ml trockenem 1,4-Dioxan wurde für 48 Stunden unter Rückfluß erhitzt. Nach 6 und 24 Stunden wurden 80 mg (0,352 mmol) DDQ jeweils hinzugegeben. Das Produkt wurde durch Flash-Chromatographie (Silicagel, Ethylacetat/Cyclohexan, 4:1) isoliert. 85 mg (0,206 mmol, 42 % Ausbeute) eines weißen Feststoffes wurden erhalten.
1^: 0,62 (Ethylacetat).

In der gleichen Weise wurden die anderen N-Acetylindolsulfonamide der Beispiele
4, 7, 21, 29, 13, 43, 47 und 48 der folgenden Tabelle erhalten.
E) N-Ethylindolsulfonamide
Beispiel 1
N-[(N-Ethylindol)-5-sulfonyl]-N'-(3-fluor-2,2-dimethylpropanoyl)-l,3-diaminoben- zol und 27 mg (0,25 mmol) DDQ in 7 ml trockenem 1,4-Dioxan wurde für 3 Stunden unter Rückfluß erhitzt. Nach Eindampfen des Lösungsmittels wurde das Produkt durch Flash-Chromatographie (Silicagel, Cyclohexan/Ethylacetat, 1 : 1) isoliert. 55 mg (0,132 mmol, 79 % Ausbeute) eines weißen Feststoffes wurden erhalten.
Rf: 0,30 (Cyclohexan/Ethylacetat, 1 :1).

In der gleichen Weise wurden die anderen N-Ethylindolsulfonamide der Beispiele 9, 22, 3, 5, 10, 44, 51 und 45 der folgenden Tabelle erhalten.
F) Indolsulfonamide
Beispiel 6
4-[N-(Indol-5-sulfonyl)]amino-N-(tert. -butyl)benzoesäureamid
Eine Mischung von 230 mg 4-(Indolin-5-sulfonyl)amino-N-(tert.-butyl)benzoesäure- amid und 210 mg (0,924 mmol) DDQ in 10 ml trockenem 1,4-Dioxan wurde für 4
Stunden unter Rückfluß erhitzt. Nach Eindampfen des Lösungsmittels wurde das Produkt durch Flash-Chromatographie (Silicagel, Cyclohexan/Ethylacetat, 1 : 1)
isoliert. 78 mg (0,21 mmol, 34 % Ausbeute) eines weißen Feststoffes wurden erhalten.
Rf: 0,30 (Dichlormethan/Methanol, 100:5). Η-NMR (400 MHz, DMSO-d6, δ/ppm): 10.58 (IH, s), 10.40 (IH, s), 8.08 (IH, s),
7.60 (2H, d), 7.52-7.50 (4H, m), 7.12 (2H, d), 6.60 (IH, d), 1.30 (9H, s). MS (CI, NH3, m/z): 743 (2M+H+), 389 (M+NH4 +), 372 (M+H+).
In der gleichen Weise wurden die anderen Indolsulfonamide der Beispiele 12, 17, 42, 52 und 39 der folgenden Tabelle erhalten.

Amino, (C1-C4)Alkylamino, Hydroxy, Cyano, Halogen, Azido, Nitro, Trifluormethyl, Carboxyl oder Phenyl substituiert ist, wobei Phenyl seinerseits bis zu zweifach, gleich oder verschieden durch Nitro, Halogen, Hydroxy, (C1-C4)Alkyl oder (C C4)Alkoxy substituiert sein kann, oder
R6 für Reste der Formeln oder -L-O-CO-Q steht,
worin
für eine geradkettige oder verzweigte Alkandiylgruppe mit bis zu 6 Kohlenstoffatomen steht,
für (C1-C6)Alkyl steht, das gegebenenfalls durch Carboxyl substituiert ist, oder
10 für Reste der Formeln

die Zahl 1 oder 2 bedeutet,

R14 Hydroxy, Benzyloxy, (C1-C6)Alkoxy oder die oben aufgeführte Gruppe 10 -NR,2R13 bedeutet,
oder das (CrC8)Alkyl gegebenenfalls durch (C3- C8)-Cycloalkyl oder durch (C6-C10)Aryl substituiert ist, das seinerseits durch Hydroxy, Halo- 15 gen, Nitro, (C1-C8)-Alkoxy oder durch die
Gruppe -NR12R13 substituiert ist,
worin R12 und R13 die oben angegebene Bedeutung haben,
20 oder das (C1-C8)Alkyl gegebenenfalls durch einen 5- bis 6-gliedrigen stickstoffhaltigen Heterocyclus oder durch Indolyl substituiert ist, worin die entsprechenden -NH-Funktionen
25 gegebenenfalls durch (C1-C6)-Alkyl substituiert oder durch eine Aminoschutzgruppe geschützt sind,
R'° und R" gleich oder verschieden sind und Wasser- 30 stoff oder eine Aminoschutzgruppe bedeuten, R7 für Wasserstoff oder für einen Rest der Formel
steht- worin
R8 , R9 , R'°'und R11 die oben angegebene Bedeutung von R8, R9, R10 und R" haben und mit dieser gleich oder verschieden sind,
und deren Salze.
2. Verbindungen nach Anspruch 1 der Formeln
worin R1 und R2 die oben angegebene Bedeutung haben.
3. Verbindungen der allgemeinen Formel ( I ) nach Anspruch 1 oder 2, worin
R1 für eine Gruppe steht, die aus den Formeln ausgewählt wird:

— für eine Einfach- oder eine Doppelbindung steht, und
R3 die oben angegebene Bedeutung besitzt, und deren Salze.
4. Verbindungen der allgemeinen Formel ( I ) nach irgendeinem der Ansprüche 1 bis 3, worin
R3 für Wasserstoff, (CrC6)Alkyl oder (CrC6)Alkanoyl steht, und
R2 für
R' Rb "N-C°-R6 oder -CO-N-R7 steh worin
R6 (C1-C8)Alkyl ist, das gegebenenfalls durch Halogen oder Hydroxy substituiert ist, und
R7 Wasserstoff ist,
und deren Salze.
5. Verbindungen der allgemeinen Formel ( I ) nach irgendeinem der Ansprüche
1 bis 4, worin R6 tert.-Butyl ist, das gegebenenfalls durch Halogen oder Hydroxy substituiert ist, und deren Salze.
6. Verfahren zur Herstellung der Verbindungen der Formel (I) nach Anspruch 1, dadurch gekennzeichnet, daß man
(A) Verbindungen der allgemeinen Formel (II) worin R1 wie oben definiert ist, mit Verbindungen der allgemeinen
Formel (III)
worin R2 wie oben definiert ist, zu Verbindungen der allgemeinen Formel (I) umsetzt,
(B) Verbindungen der allgemeinen Formel (Ia)
worin R2 wie oben definiert ist, und
R1a für eine Gruppe steht, die aus den folgenden Formeln ausgewählt wird:
mit Verbindungen der Formel (IV)
R3-A (IV) worin R3 wie oben definiert ist und A eine übliche Abgangsgruppe ist, in an sich bekannter Weise in Gegenwart einer Base zu Verbindungen der allgemeinen Formel (Ib) umsetzt:

7. Verbindungen nach Anspruch 1, zur Verwendung als Arzneimittel.
8. Pharmazeutische Zusammensetzung, die eine Verbindung der allgemeinen Formel (I) nach Anspruch 1 in Mischung mit mindestens einem pharmazeutisch verträglichen Träger oder Exzipienten umfaßt.
9. Verwendung der Verbindung der allgemeinen Formel (I) nach Anspruch 1 zur Herstellung eines Arzneimittels.
10. Verwendung einer Verbindung der allgemeinen Formel (I) nach Anspruch 1 zur Herstellung eines Arzneimittels zur Behandlung von viralen Infektionen, insbesondere Infektionen durch Cytomegalieviren.
