Abstract
Die vorliegende Erfindung betrifft neue und bekannte Pyrazolcarbonsäureamide der allgemeinen Formeln (I) und (Ia). Ausserdem wird die Herstellung der Pyrazolcarbonsäureamide entweder über Festphasenchemie oder über die neuen Pyrazolcarbonsäuren beschrieben. Die Pyrazolcarbonsäureamide werden als Arzneimittel, insb esondere als Medikamente zur Behandlung von Anämien verwendet.
Description
Verwendung von Pyrazol-Carbonsäureamiden
Die vorliegende Erfindung betrifft die Verwendung von Pyrazolcarbonsäureamiden zur Herstellung von Arzneimitteln zur Prophylaxe und/oder Behandlung von Anämien, neue Pyrazolcarbonsäureamide sowie Verfahren zu ihrer Herstellung.
Erythropoetin (EPO) ist ein Glykoprotein mit einem Molekulargewicht von ungefähr 34 000 Da. Über 90 % der EPO-Synthese finden in der Niere statt, und das dort pro- duzierte EPO wird ins Blut sezerniert. Die primäre physiologische Funktion von EPO ist die Regulation der Erythropoese im Knochenmark. Dort stimuliert EPO die Proliferation und Reifung der erythroiden Vorläuferzellen.
Die EPO-Spiegel im Blut sind normalerweise niedrig, sinkt aber der O2-Gehalt des Blutes ab, dann kommt es zu einem Anstieg der EPO-Synthese und dadurch auch zu einem Anstieg der EPO-Spiegel im Blut. Dies hat zur Folge, daß die Hämatopoese stimuliert wird und daß der Hämatokrit steigt. Hierdurch kommt es zu einer Steigerung der O2-Transportkapazität im Blut. Wenn die Erythrozytenzahl ausreicht, um genügend O2 zu transportieren, dann fällt die EPO-Blutkonzentration wieder ab. Eine mangelnde Sauerstoffversorgung (Hypoxie) kann eine Reihe von Ursachen haben, z.B. starker Blutverlust, längerer Aufenthalt in großen Höhen, aber auch Niereninsuffizienz oder Knochenmarksuppression.
Es ist bekannt, daß rekombinantes humanes (rh) EPO die Erythropoese stimuliert und somit in der Therapie von schweren Anämien Anwendung gefunden hat. Weiterhin wird rh EPO zur Vermehrung der körpereigenen Blutzellen eingesetzt, um die Notwendigkeit von Fremdbluttransfusionen zu vermindern.
Jedoch sind starke Nebenwirkungen, die bei der Gabe von rh EPO auftreten, bekannt. Dazu gehören die Entstehung und Verstärkung des Bluthochdrucks, Verursachung
einer Encephalopathie-ähnlichen Symptomatik bis hin zu tonisch-klonischen Krämpfen und cerebralem oder myocardialem Infarkt durch Thrombosen.
Ferner ist rh EPO nicht oral verfugbar und muß daher i.p., i.v. oder subcutan appli- ziert werden, wodurch die Anwendung auf die Therapie schwerer Anämie begrenzt ist.
Aus der Publikation WO 97/19039 sind substituierte Pyrazole bekannt. Die Synthese der Pyrazol-Derivate erfolgt an der festen Phase. Nach Abspaltung erhält man Pyra- zolphenyl-Carbonsäureamide. Die Verwendung als Arzneimittel ist nicht beschrieben.
Die vorliegende Erfindung betrifft nun die Verwendung von Pyrazolcarbonsäureamiden der allgemeinen Formel (I),
in welcher
A, D, E und G gleich oder verschieden sind und für Wasserstoff, Halogen, Trifluormethyl, Hydroxy oder für (C,-C6)-Alkyl oder für (C,-C6)-Alkoxy stehen,
R1 für Wasserstoff oder für (C,-C6)-Alkyl steht,
R2 für (C6-C)0)-Aryl oder einen 5- bis 6-gliedrigen aromatischen, gegebenenfalls benzokondensierten Heterocyclus mit bis zu 3 Heteroatomen aus der Reihe S, N und/oder O steht, wobei die oben aufgeführten aromatischen Ringsysteme gegebenenfalls bis zu 3 -fach gleich oder verschieden durch Halogen, Tri-
fluormethyl, Trifluormethoxy, (C,-C6)-Alkyl, (C,-C6)-Alkoxy oder (C,-C6)- Alkoxycarbonyl substituiert sein können,
und ihren Tautomeren und den jeweiligen Salzen,
als Arzneimittel, die sich zur Prophylaxe und/oder Behandlung von Erkrankungen, beispielsweise von Anämien, eignen.

Die Verbindungen können in Abhängigkeit von dem Substitutionsmuster in stereoisomeren Formen, die sich entweder wie Bild und Spiegelbild (Enantiomere), oder die sich nicht wie Bild und Spiegelbild (Diastereomere) verhalten, existieren. Die Erfindung betrifft sowohl die Enantiomeren oder Diastereomeren oder deren jeweilige Mischungen. Die Racemformen lassen sich ebenso wie die Diastereomeren in bekannter Weise in die stereoisomer einheitlichen Bestandteile trennen.
Physiologisch unbedenkliche Salze der Verbindungen können Salze der erfmdungs- gemäßen Stoffe mit Mineralsäuren, Carbonsäuren oder Sulfonsäuren sein. Besonders bevorzugt sind z.B. Salze mit Chlorwasserstoffsäure, Bromwasserstoffsäure, Schwefelsäure, Phosphorsäure, Methansulfonsäure, Ethansulfonsäure, Toluolsulfonsäure, Benzolsulfonsäure, Naphthalindisulfonsäure, Essigsäure, Propionsäure, Milchsäure, Weinsäure, Zitronensäure, Fumarsäure, Maleinsäure oder Benzoesäure.
Als Salze können Salze mit üblichen Basen genannt werden, wie beispielsweise Alkalimetallsalze (z.B. Natrium- oder Kaliumsalze), Erdalkalisalze (z.B. Calcium- oder Magnesiumsalze) oder Ammoniumsalze, abgeleitet von Ammoniak oder organischen Aminen wie beispielsweise Diethylamin, Triethylamin, Ethyldiisopropylamin, Prokain, Dibenzylamin, N-Methylmorpholin, Dihydroabietylamin, 1-Ephenamin oder Methyl- piperidin.
(C6-C10)-Aryl steht im allgemeinen für einen aromatischen Rest mit 6 bis 10 Kohlenstoffatomen. Bevorzugte Arylreste sind Phenyl und Naphthyl.
(C,-C6)-Alkyl steht für einen geradkettigen oder verzweigten Alkylrest mit 1 bis 6 Kohlenstoffatomen. Bevorzugt ist ein geradkettiger oder verzweigter Alkylrest mit 1 bis 4 Kohlenstoffatomen. Beispielsweise seien genannt: Methyl, Ethyl, n-Propyl, Isopropyl, tert.Butyl, n-Pentyl und n-Hexyl. Besonders bevorzugt ist ein geradkettiger oder verzweigter Alkylrest mit 1 bis 3 Kohlenstoffatomen.
(CrC6)-Alkoxy steht für einen geradkettigen oder verzweigten Alkoxyrest mit 1 bis 6 Kohlenstoffatomen. Bevorzugt ist ein geradkettiger oder verzweigter Alkoxyrest mit 1 bis 4 Kohlenstoffatomen. Beispielsweise seien genannt: Methoxy, Ethoxy, n-Propoxy, Isopropoxy, tert.Butoxy, n-Pentoxy und n-Hexoxy. Besonders bevorzugt ist ein geradkettiger oder verzweigter Alkoxyrest mit 1 bis 3 Kohlenstoffatomen.
(C i -Cfi)- Alkoxycarbonyl steht für einen geradkettigen oder verzweigten Alkoxycarb- onylrest mit 1 bis 6 Kohlenstoffatomen. Bevorzugt ist ein geradkettiger oder ver- zweigter Alkoxycarbonylrest mit 1 bis 4 Kohlenstoffatomen. Beispielsweise seien genannt: Methoxycarbonyl, Ethoxycarbonyl, n-Propoxycarbonyl, Isopropoxycarbonyl und tert.Butoxycarbonyl. Besonders bevorzugt ist ein geradkettiger oder verzweigter Alkoxycarbonylrest mit 1 bis 3 Kohlenstoffatomen.
Ein 5- bis 6-gliedriger aromatischer Heterocyclus mit bis zu 3 Heteroatomen aus der
Reihe S, O und/oder N steht beispielsweise für Pyridyl, Pyrimidyl, Pyridazinyl,
Thienyl, Furyl, Pyrrolyl, Thiazolyl, Oxazolyl oder Imidazolyl. Bevorzugt sind Pyridyl, Pyrimidyl, Pyridazinyl, Furyl und Thienyl.
Ein 5- bis 6-gliedriger aromatischer benzokondensierter Heterocyclus mit bis zu 3 Heteroatomen aus der Reihe S, O und/oder N steht beispielsweise für Benzothiophen,

Als Lösemittel für die zuvor beschriebenen Verfahren eignen sich hierbei inerte organische Lösemittel, die sich unter den Reaktionsbedingungen nicht verändern.
Hierzu gehören Halogenkohlenwasserstoffe wie Dichlormethan, Trichlormethan,
Tetrachlormethan, 1 ,2-Dichlorethan, Trichlorethan, Tetrachlorethan, 1,2-Dichlor-
ethylen oder Trichlorethylen, Kohlenwasserstoff wie Benzol, Xylol, Toluol, Hexan oder Cyclohexan, Dimethylformamid, Dimethylacetamid, Acetonitril oder DMSO sowie Ether wie beispielsweise Diethylether, Dioxan oder Tetrahydrofuran.. Ebenso ist es möglich, Gemische der Lösemittel einzusetzen. Besonders bevorzugt sind Dimethylformamid und Dichlormethan.
Die zuvor beschriebenen Reaktionen erfolgen im allgemeinen in einem Temperaturbereich von -78°C bis zu Rückflußtemperatur, bevorzugt von 0°C bis Siedepunkt des benutzten Lösemittels.
Die zuvor beschriebenen Umsetzungen können bei normalem, erhöhtem oder bei erniedrigtem Druck durchgeführt werden (z.B. von 0,5 bis 5 bar). Im allgemeinen arbeitet man bei Normaldruck.
Als Dehydratisierungsreagenzien für das Verfahren [B] eignen sich Carbodiimide wie beispielsweise Diiso-propylcarbodiimid, Dicyclohexylcarbodiimid oder N-(3- Dimethylaminopropyl)-N'-ethylcarbodiimid-Hydrochlorid oder Carbonylverbindun- gen wie Carbonyldiimidazol oder 1 ,2-Oxazoliumverbindungen wie 2-Ethyl-5-phe- nyl-l,2-oxazolium-3-sulfonat oder Propanphosphorsäureanhydrid oder Isobutylchlo- roformat oder Benzotriazolyloxy-tris-(dimethylamino)phosphonium-hexyfluorophos- phat oder Phosphonsäurediphenylesteramid oder Methansulfonsäurechlorid, gegebenenfalls in Anwesenheit von Basen wie Triethylamin oder N-Ethylmorpholin oder N- Methylpiperidin oder Dicyclohexylcarbodiimid und N-Hydroxysuccinimid. Bevorzugt ist Carbonyldiimidazol (CDI).
Als feste Phase werden die üblichen aminofunktionalisierten Polystyrole eingesetzt. Bevorzugt sind aminofunktionalisierte Polystyrol-Polyethylen Copolymere, die mit einer Polyethylenglycolkette modifiziert sind. Besonders bevorzugt sind sogenannte SAM-Harze (Abkürzung für Standard-Amid-Harz) und RAM-Harze (Abkürzung für Rink-Amid-Harz). Beispiele für erfindungsgemäß verwendbare, aminofunktionali-

Als Lösemittel für die Verfahren [C] und [D] eignen sich hierbei inerte organische Lösemittel, die sich unter den Reaktionsbedingungen nicht verändern. Ebenso ist es möglich, Gemische der Lösemittel einzusetzen. Besonders bevorzugt sind Ethanol und DMSO.
Für die unter [A] beschriebene Umsetzung des Aldehyds mit den Acetophenonen kommen als Katalysator die üblichen sauren und basischen Katalysatoren in Betracht, vorzugsweise Säure (z.B. para-Toluolsulfonsäurechlorid).
Die zuvor beschriebenen Reaktionen erfolgen im allgemeinen in einem Temperaturbereich von -78°C bis zu Rückflußtemperatur, bevorzugt von 0°C bis Siedepunkt des benutzten Lösemittels.
Die zuvor beschriebenen Umsetzungen können bei normalem, erhöhtem oder bei erniedrigtem Druck durchgeführt werden (z.B. von 0,5 bis 5 bar). Im allgemeinen arbeitet man bei Normaldruck.
Die Verseifung der Carbonsäureester erfolgt nach üblichen Methoden, indem man die Ester in inerten Lösemitteln mit üblichen Basen oder Säuren behandelt, wobei im Fall der basischen Verseifung die zunächst entstehenden Salze durch Behandeln mit
Säure in die freien Carbonsäuren überführt werden können.
Als Basen eignen sich für die Verseifung die üblichen anorganischen Basen. Hierzu gehören bevorzugt Alkalihydroxide oder Erdalkalihydroxide wie beispielsweise Natriumhydroxid, Lithiumhydroxid, Kaliumhydroxid oder Bariumhydroxid, oder
Alkalicarbonate wie Natrium- oder Kaliumcarbonat oder Natriumhydrogencarbonat. Besonders bevorzugt werden Natriumhydroxid oder Lithiumhydroxid eingesetzt.
Als Lösemittel eignen sich für die Verseifung Wasser oder die für eine Verseifung üblichen organischen Lösemittel. Hierzu gehören bevorzugt Alkohole wie Methanol,
Ethanol, Propanol, Isopropanol oder Butanol, oder Ether wie Tetrahydrofuran oder Dioxan, oder Dimethylformamid oder Dimethylsulfoxid. Besonders bevorzugt werden Alkohole wie Methanol, Ethanol, Propanol oder Isopropanol verwendet. Ebenso ist es möglich, Gemische der genannten Lösemittel einzusetzen. Bevorzugt ist Wasser/Tetrahydrofuran oder Wasser/Ethanol.
Die Verseifung wird im allgemeinen in einem Temperaturbereich von 0°C bis +100°C, bevorzugt von 0°C bis +80°C durchgeführt.
Im allgemeinen wird die Verseifung bei Normaldruck durchgeführt. Es ist aber auch möglich, bei Unterdruck oder bei Überdruck zu arbeiten (z.B. von 0,5 bis 5 bar).
Bei der Durchführung der Verseifung wird die Base oder die Säure im allgemeinen in einer Menge von 1 bis 3 mol, bevorzugt von 1 bis 2,5 mol bezogen auf 1 mol des Esters eingesetzt. Besonders bevorzugt verwendet man molare Mengen der Reaktan- den.
Die Verbindungen der allgemeinen Formeln (VI) und (XI) sind an sich bekannt oder nach üblichen Methoden herstellbar [vgl. Erne, D. et al., Helv. Chim. Acta 62(1979), 994-1006; Hasegawa, E. et al. J. Org. Chem. 56 (1991), 1631-1635; Watanabe, Ken- ichi et al., Bull. Chem. Soc. Jpn. 55 (1988) 3208-3211].
Die Verbindungen der allgemeinen Formel (VII), (VIII), (IX), (X) und (XII) sind teilweise neu oder bekannt und können wie oben beschrieben hergestellt werden.
Die erfindungsgemäß verwendeten Verbindungen der allgemeinen Formel (I) und die neuen Stoffe der allgemeinen Formel (la) zeigen ein nicht vorhersehbares, wertvolles pharmakologisches Wirkspektrum und sind daher zur Behandlung und Prophylaxe von Erkrankungen geeignet.
Sie können bevorzugt in Arzneimitteln eingesetzt werden zur Behandlung und Prophylaxe von Anämien, wie beispielsweise bei Frühgeborenen- Anämie, Anämie bei chronischer Niereninsuffizienz, Anämie nach einer Chemotherapie und der Anämie bei HIV-Patienten, somit auch zur Behandlung von schweren Anämien.
Auch bei völlig intakter endogener EPO-Produktion kann durch die Gabe der erfindungsgemäßen Verbindungen eine zusätzliche Stimulation der Erythropoese induziert werden, was insbesondere bei Eigenblutspendern ausgenutzt werden kann.
Für die Applikation der erfindungsmäßen und erfindungsgemäß verwendeten Verbindungen kommen alle üblichen Applikationsformen in Betracht. Vorzugsweise erfolgt die Applikation oral, transdermal oder parenteral. Ganz besonders bevorzugt ist die orale Applikation, worin ein weiterer Vorteil gegenüber der aus dem Stand der Technik bekannten Therapie von Anämien mit rh-EPO liegt.
Die erfindungsgemäßen Verbindungen wirken insbesondere als Erythropoetin-Sensi- tizer.
Als Erythropoetin-Sensitizer werden Verbindungen bezeichnet, die in der Lage sind, die Wirkung des im Körper vorhandenen EPO so effizient zu beeinflussen, daß die
Erythropoese gesteigert wird, insbesondere daß die Sauerstoffversorgung verbessert wird. Sie sind überaschenderweise oral wirksam, wodurch die therapeutische Anwendung unter Ausschluß oder Reduktion der bekannten Nebenwirkungen wesentlich verbessert und gleichzeitig vereinfacht wird.
Gegenstand der vorliegenden Erfindung ist somit auch die Verwendung von EPO- Sensitizern zur Stimulation der Erythropoese, insbesondere zur Prophylaxe und/oder Behandlung von Anämien, vorzugsweise schweren Anämien wie beispielsweise Frühgeborenen-Anämie, Anämie bei chronischer Niereninsuffizienz, Anämie nach Chemotherapie oder auch Anämie bei HIV-Patienten. Daneben kommt auch die
Gabe von EPO-Sensitizern bei völlig intakter endogener EPO-Produktion zur zusätzlichen Stimulation der Erythropoese in Betracht, was insbesondere bei Eigenblutspendern ausgenutzt werden kann.
Besonders bevorzugt ist die orale Applikation dieser sogenannten EPO-Sensitizer für die zuvor genannten Zwecke.
Somit ermöglichen die erfindungsgemäßen Verbindungen eine effiziente Stimulation der Erythropoese und folglich eine Prophylaxe bzw. Therapie von Anämien, die noch vor dem Stadium eingreift, in welchem die herkömmlichen Behandlungsmethoden mit EPO einsetzen. Denn die erfindungsgemäßen Verbindungen erlauben eine wirksame Beeinflussung des körpereigenen EPO, woduch die direkte Gabe von EPO mit den damit verbundenen Nachteilen vermieden werden kann.
Weiterer Gegenstand der vorliegenden Erfindung sind also Arzneimittel und pharmazeutische Zusammensetzungen, die mindestens eine Verbindung der allgemeinen Formel (I) zusammen mit einem oder mehreren pharmakologisch unbedenklichen Hilfs- oder Trägerstoffen enthalten, sowie deren Verwendung zur Stimulation der Erythropoese, insbesondere zu Zwecken der Prophylaxe und/oder Behandlung von Anämien, wie z.B. Frühgeborenenanämien, Anämien bei chronischer Niereninsuffizienz, Anämien nach einer Chemotherapie oder Anämien bei HIV-Patienten.
Testbeschreibung (in vitro)
Zeilproliferation von humanen erythroiden Norläuferzellen
20 ml Heparin-Blut wurden mit 20 ml PBS verdünnt und für 20 min (220xg) zentrifugiert. Der Überstand wurde verworfen, die Zellen wurden in 30 ml PBS resuspendiert und auf 17 ml Ficoll Paque (d=1.077g/ml, Pharmacia) in einem 50 ml Röhrchen pipettiert. Die Proben wurden für 20 min bei 800xg zentrifugiert. Die mononukleären Zellen an der Grenzschicht wurden in ein neues Zentrifugenröhrchen überführt, mit dem 3-fachem Volumen mit PBS verdünnt und für 5 min bei 300xg zentrifugiert.
Die CD34-positiven Zellen aus dieser Zellfraktion wurden mittels eines kommerziellen Aufreinigungsverfahrens (CD34 Multisort Kit von Miyltenyi) isoliert.
CD34-positive Zellen (6000-10000 Zellen/ml) wurden in Stammzellmedium (0.9%
Methylzellulose, 30% Kälberserum, 1% Albumin (Rind), lOOμM 2-Mercaptoethanol und 2 mM L-Glutamin) von StemCell Technologies Inc. resuspendiert. 10 mU/ml humanes Erythropoietin, 10 ng/ml humanes IL-3 und 0-1 OμM Testsubstanz wurden zugesetzt. 500 μl ertiefung (24- Wellplatten) wurden für 14 Tage bei 37°C in 5% CO2, 95% Luft kultiviert.
Kulturen wurden mit 20 ml 0.9%w/v ΝaCl-Lösung verdünnt, für 15 min bei 600xg zentrifugiert und in 200 μl 0,9%w/v ΝaCl resuspendiert. Zur Bestimmung der Zahl der erythroiden Zellen, wurden 50μl der Zellsuspension zu lOμl Benzidin-Färbe- lösung (20μg Benzidin in 500 μl DMSO, 30μl H2O2 und 60 μl konz. Essigsäure) pipettiert. Die Zahl der blauen Zellen wurde mikroskopisch ausgezählt.
Testbeschreibung Hämatokrit-Maus
Normale Mäuse werden mit Testsubstanzen über mehrere Tage behandelt. Die Applikation erfolgt intraperitoneal, subkutan oder per os. Bevorzugte Lösungsmittel sind Solutol/DMSO/Sacharose/NaCl-Lösung oder Glycofurol.
Vom Tag 0 (vor der ersten Applikation) bis zu ca. 3 Tagen nach der letzten Applikation werden mehrfach ca. 70 μl Blut durch Punktion des retroorbitalen Venenplexus mit einer Hämatokritkapillare entnommen. Die Proben werden zentrifugiert und der Hämatokrit durch manuelle Ablesung bestimmt. Primärer Parameter ist der Häma- tokritanstieg gegenüber dem Ausgangswert der behandelten Tiere im Vergleich zur Veränderung des Hämatokrit in der Placebo-Kontrolle (zweifach normierter Wert).
Die neuen Wirkstoffe können in bekannter Weise in die üblichen Formulierungen überführt werden, wie Tabletten, Dragees, Pillen, Granulate, Aerosole, Sirupe, Emulsionen, Suspensionen und Lösungen, unter Verwendung inerter, nicht toxischer, pharmazeutisch geeigneter Trägerstoffe oder Lösungsmittel. Hierbei soll die therapeutisch wirksame Verbindung jeweils in einer Konzentration von etwa 0,5 bis 90- Gew.-% der Gesamtmischung vorhanden sein, d.h. in Mengen, die ausreichend sind, um den angegebenen Dosierungsspielraum zu erreichen.
Die Formulierungen werden beispielsweise hergestellt durch Verstrecken der Wirkstoffe mit Lösungsmitteln und/oder Trägerstoffen, gegebenenfalls unter Verwendung von Emulgiermitteln und/oder Dispergiermitteln, wobei z.B. im Fall der Benutzung von Wasser als Verdünnungsmittel gegebenenfalls organische Lösungsmittel als
Hilfslösungsmittel verwendet werden können.
Die Applikation erfolgt in üblicher Weise, vorzugsweise oral, transdermal oder parenteral, insbesondere perlingual oder intravenös.
Im allgemeinen hat es sich als vorteilhaft erwiesen, bei intravenöser Applikation Mengen von etwa 0,01 bis 10 mg/kg, vorzugsweise etwa 0,1 bis 10 mg/kg Körpergewicht zur Erzielung wirksamer Ergebnisse zu verabreichen.
Trotzdem kann es gegebenenfalls erforderlich sein, von den genannten Mengen abzuweichen, und zwar in Abhängigkeit vom Körpergewicht bzw. der Art des Applikationsweges, vom individuellen Verhalten gegenüber dem Medikament, der Art von dessen Formulierung und dem Zeitpunkt bzw. Intervall, zu welchen die Verabreichung erfolgt. So kann es in einigen Fällen ausreichend sein, mit weniger als der vorgenannten Mindestmenge auszukommen, während in anderen Fällen die genannte obere Grenze überschritten werden muß. Im Falle der Applikation größerer Mengen kann es empfehlenswert sein, diese in mehreren Einzelgaben über den Tag zu verteilen.

Herstellungsbeispiele
Beispiel 1
3-(4-Aminocarbonylphenyl)-5-(4-bromphenyl)- 1 -methyl-pyrazol
Tentagel SAM Harz (100 mg, Beladung 0.22 mmol/g) wird in Dimethylformamid (10 ml) suspendiert, mit Acetophenon-4-carbonsäure (144 mg, 0.88 mmol), TBTU (TBTU = o-(Benztriazol-l-yl)-N,N,N',N'-tetramethyl-uronium-tetrafluoroborat) (282 mg, 0.88 mmol) und DIEA (DIEA = Diisopropylethylamid) (16 mg, 0.12 mmol) versetzt und über Nacht bei Raumtemperatur geschüttelt. Anschließend wird die flüssige Phase abgesaugt und das Harz mit Dimethylformamid, Methanol, Methylenchlorid und Diethylether (je zweimal 5 ml)gewaschen.
Für die Claisen-Kondensation wird das Harz (100 mg, 0.21 mmol) unter Argon in
Dimethylacetamid (10 ml) suspendiert, mit 4-Brom-benzoesäuremethylester (812 mg, 3.78 mmol) versetzt und 10 min bei Raumtemperatur geschüttelt. Dann wird NaH (Disp. in Mineralöl, 60%ig, 96.6 mg, 2.52 mmol) zugegeben und die Mischung 1 h bei 90°C geschüttelt. Anschließend wird die flüssige Phase abgesaugt und das Harz mit Dimethylformamid, Methanol, Methylenchlorid und Diethylether (je zweimal 5 ml) gewaschen.
Für die Kondensation wird das Harz (100 mg, 0.2 mmol) in Dimethylacetamid suspendiert und mit 2 ml einer 1.5 M Lösung von Methylhydrazin in Dimethylacetamid versetzt. Die Mischung wird 48h bei 70°C gerührt. Anschließend wird die flüssige
Phase abgesaugt und das Harz mit Dimethylformamid, Methanol, Methylenchlorid und Diethylether (je zweimal 5 ml) gewaschen.
Zur Abspaltung wird das Harz (100 mg) mit 2 ml einer l : l-(v:v)-Mischung von Methylenchlorid und Trifluoressigsäure versetzt und 1 h bei Raumtemperatur geschüttelt. Die flüssige Phase wird abgetrennt, die feste Phase mit Methylenchlorid nachgewaschen und die vereinigten flüssigen Phasen werden eingedampft. Das Produkt fällt als Regioisomerengemisch in einer Ausbeute von 4 mg, und mittels präpa- rativer HPLC wird das gewünschte Isomer abgetrennt.

In Analogie zu der aufgeführten Vorschrift des Beispiels 1 werden die in der folgenden Tabelle aufgeführten Beispiele hergestellt:

Claims
1. Verwendung von Pyrazolcarbonsäureamiden der allgemeinen Formel (I),
in welcher
A, D, E und G gleich oder verschieden sind und für Wasserstoff, Halogen, Trifluormethyl, Hydroxy oder für (C,-C6)- Alkyl oder für (C,-C6)-Alkoxy stehen,
R1 für Wasserstoff oder für (C,-C6)- Alkyl steht,
R2 für (C6-Cl0)-Aryl oder einen 5- bis 6-gliedrigen aromatischen, gegebe- nenfalls benzokondensierten Heterocyclus mit bis zu 3 Heteroatomen aus der Reihe S, N und/oder O steht, wobei die oben aufgeführten aromatischen Ringsysteme gegebenenfalls bis zu 3 -fach gleich oder verschieden durch Halogen, Trifluormethyl, Trifluormethoxy, (C,-C6)- Alkyl, (C,-C6)-Alkoxy oder (C,-C6)-Alkoxycarbonyl substituiert sein können,
und ihren Tautomeren sowie den jeweiligen Salzen
zur Herstellung von Arzneimitteln zur Prophylaxe und/oder Behandlung von Erkrankungen, insbesondere Anämien.
2. Verwendung gemäß Anspruch 1 , wobei in der allgemeinen Formel (I) A, D, E und G gleich oder verschieden sind und für Wasserstoff, Fluor, Chlor, Brom oder Trifluormethyl stehen,
R! für Wasserstoff oder Methyl steht,
R2 für Phenyl, Furyl, Thienyl, Benzothiophen oder Pyridyl steht, wobei die oben aufgeführten aromatischen Ringsysteme gegebenenfalls bis zu 2-fach gleich oder verschieden durch Fluor, Chlor, Brom, Trifluormethyl, Trifluormethoxy, (C,-C4)-Alkyl oder (C,-C4)-Alkoxy oder (C,-C4)-Alkoxycarbonyl substituiert sein können.
3. Verwendung gemäß Anspruch 1, wobei in der allgemeinen Formel (I)
A, D, E und G für Wasserstoff stehen,
R1 für Wasserstoff oder Methyl steht
und
R2 für Phenyl, Thienyl, Benzothiophen oder Pyridyl steht, wobei die oben aufgeführten aromatischen Ringsysteme gegebenenfalls durch Fluor, Chlor, Brom, Trifluormethyl, Trifluormethoxy, Methyl oder Methoxy substituiert sein können.
4. Verwendung von Pyrazolcarbonsäureamiden gemäß Anspruch 1, wobei die
Verbindungen der allgemeinen Formel (I) die folgenden Strukturen aufweisen:

in welcher
A', D', E', G' und R1 die oben angegebene Bedeutung von A, D, E, G und R1 haben,
und
L für CF, oder OCF, steht.
6. Pyrazolcarbonsäureamid der allgemeinen Formel (la) gemäß Anspruch 5 mit folgender Struktur:
7. Verfahren zur Herstellung von Pyrazolcarbonsäureamiden der allgemeinen Formel (I) gemäß Ansprüchen 5 bis 6, dadurch gekennzeichnet, daß man
[A] Verbindungen der allgemeinen Formel (II)
in welcher
A, D, E und G die oben angegebene Bedeutung haben,
an ein aminofunktionalisiertes Harz kuppelt und dann mit Verbindungen der allgemeinen Formel (III)
R3OOC-R2 (III), in welcher
R2 die oben angegebene Bedeutung hat
und R3 für einen (C,-C4)-Alkylrest steht,
an der Festphase umsetzt und anschließend mit Verbindungen der allgemeinen Formel (IV)
H_N^N-R (IV),
H in welcher
R1 die oben angegebene Bedeutung hat,
umsetzt, die Verbindungen anschließend vom Harz abspaltet und gegebenenfalls (nämlich für R1 ≠ H) in die Isomeren trennt,
oder
[B] Pyrazolcarbonsäuren der allgemeinen Formel (V)
in welcher
R , R , A; D; E und G die oben angegebene Bedeutung haben,
gegebenenfalls in Gegenwart eines Dehydratisierungsmittels oder über ein aktiviertes Carbonsäure-Derivat mit Ammoniak umsetzt.
8. Arzneimittel, enthaltend mindestens ein Pyrazolcarbonsäureamid gemäß Ansprüchen 1 bis 6 sowie pharmakologisch unbedenkliche Formulierungsmittel.
9. Arzneimittel gemäß Ansprüchen 1 bis 6 zur Prophylaxe und/oder Behandlung von Anämien.
10. Arzneimittel gemäß Ansprüchen 1 bis 6 zur Behandlung von bei Frühgeborenen-Anämie, Anämie bei chronischer Niereninsuffizienz, Anämie nach einer Chemotherapie und der Anämie bei HIV-Patienten.
11. Arzneimittel nach Anspruch 8 zur Stimulation der Erythropoese von Eigenblutspendern.
12. Pyrazolcarbonsäureamide gemäß Anprüchen 5 und 6 zur Behandlung und
Prophylaxe von Erkrankungen
13. Verwendung von Erythropoetin- Sensitizern zur Herstellung von Arzneimitteln oder pharmazeutischen Zusammensetzungen zur Prophylaxe und/oder Behandlung von Anämien.
14. Verwendung nach Anspruch 13 zur Herstellung von Arzneimitteln oder pharmazeutischen Zusammensetzungen zur Prophylaxe und/oder Behandlung von Frühgeborenen-Anämien, Anämien bei chronischer Niereninsuffizienz, Anämien nach einer Chemotherapie und Anämien bei HIV-Patienten.
15. Verwendung von Erythropoetin-Sensitizern zur Herstellung von Arzneimitteln oder pharmazeutischen Zusammensetzungen zur Stimulation der Erythropoese von Eigenblutspendern.
6. Verwendung nach einem der Ansprüche 13 bis 15, dadurch gekennzeichnet, dass die Erythropoetin-Sensitizer peroral appliziert werden.
