Abstract
L'invention concerne des cellules embryonnaires du muscle cardiaque (cardiomyocytaires et cardiomyoblastaires), leur préparation et leur utilisation, notamment pour la transplantation génique à médiation cellulaire. Les domaines d'application de cette invention sont la médecine et le génie génétique. Les cellules embryonnaires obtenues selon l'invention contiennent deux produits de recombinaison génique comprenant a) une séquence d'ADN régulatrice de 2,1 kb de long du promoteur spécifique du ventricule myosine-chaîne légère-2(MLC-2v), le gène marqueur sélectionnable β-galactosidase en fusion avec le gène rapporteur néomycine et b) une séquence d'ADN régulatrice du promoteur de la HSV (virus de l'herpès simplex) thymidine kinase et le gène marqueur sélectionnable hygromycine, ainsi qu'éventuellement des gènes d'immortalisation et/ou des gènes inactivés par recombinaison homologue et/ou éventuellement un ou plusieurs gène(s) thérapeutique(s).
