Description

  • Die Erfindung bezieht sich auf eine Dispensiereinrichtung für Flüssigkeiten, insbesondere für das Beschicken von Kavitäten von Titerplatten, mit einem über eine Leitung an eine Pumpe angeschlossenen horizontalen Verteilerrohr, von dem mindestens zwei, vorzugsweise acht, Füllrohre radial wegführen.
  • In der medizinischen Labortechnik wird beispielsweise zur Messung von Antigenen und/oder Antikörpern das Elisa-Verfahren angewendet (Enzym linked immunosorbent assay), bzw. die verschiedenen Verfahren dieses Systems. Es werden beispielsweise zur Messung von Antigenen in einem ersten Verfahrensschritt Antikörper an der Titerplatte absorbiert, anschließend wird die Titerplatte gewaschen, dann erfolgt in einem weiteren Verfahrensschritt die Beimengung einer Restlösung, welche das Antigen enthält, darauf erfolgt wiederum eine Waschung, worauf enzymmarkierte spezifische Antikörper auf die Titerplatte gegeben werden, dann erfolgt wieder ein Waschen der Titerplatte, worauf die Beimengung des Enzymsubstrates erfolgt.
  • Die eigentliche Messung erfolgt in einem Photometer.
  • Die derzeit in Gebrauch befindlichen Mikrotiterplatten weisen 96 Kavitäten auf, von denen jede praktisch einer Küvette entspricht.
  • Mit den bekannten Dispensiereinrichtungen werden jeweils acht Kavitäten auf einmal gefüllt.
  • Eines der wesentlichen Probleme, die dabei auftreten, ist, daß die Füllmenge für jede Kavität möglichst gleich bleiben soll und daß weiters sowohl die Bildung von Luftblasen als auch ein Nachtropfen absolut sicher verhindert werden muß.
  • Das Problem der gleichmäßigen Abgabe wird noch dadurch erschwert, daß mit ein und derselben Vorrichtung bei verschiedenen Testvorgängen Flüssigkeiten unterschiedlicher Viskosität abgegeben werden müssen.
  • Eine Dispensiereinrichtung der eingangs erwähnten Art ist in der US-A-3357606 beschrieben. Dabei ist ein Block an eine Zuführleitung angeschlossen und von diesem Block führen Füllröhrchen weg. In jedem Füllrohr ist ein Kugelventil vorgesehen, und es wurde versucht, die Abgabeegalität durch genaue Einstellung dieser Ventile zu erreichen.
  • Mit den herkömmlichen Kolbenhub- oder Membranpumpen wird eine auf 5 % genaue Abgabeegalität erreicht. Aufgabe der Erfindung ist es, die Abgabeegalität wesentlich zu verbessern. Dabei sollen keine besonderen Einstellarbeiten notwendig sein.
  • Dies wird erfindungsgemäß dadurch erreicht, daß am oberen Ende des nach oben führenden Abschnittes der Füllrohre ein parallel zum Verteilerrohr ausgerichtetes Ausgleichsrohr ist, an das die Umleitungsbereiche der Füllrohre anschließen.
  • Mit der erfindungsgemäßen Einrichtung werden Volumensunterschiede bei den Füllungen der Kavitäten von nur ± 1 % erzielt.
  • In diesem Zusammenhang ist zu beachten, daß nicht die Gesamtmenge der in eine Titerplatte abgefüllten Flüssigkeitsmenge von Bedeutung ist und diesbezügliche Volumensschwankungen, sondern das Volumensverhältnis der Flüssigkeiten in den einzelnen Kavitäten zueinander.
  • Von den herkömmlichen Kolbenhubpumpen wird pro Pumpenzyklus eine Menge von 0,4 ml abgegeben, so daß bei einer Dispensiereinrichtung mit acht Füllrohren 50 111 Testflüssigkeit pro Kavität der Mikrotiterplatte abgegeben werden. Es hat sich dabei als vorteilhaft für die Abgabegenauigkeit erwiesen, wenn die Leitung zwischen dem Verteilerrohr und der Pumpe aus elastischen Material ist, um die relativ harten Pumpenstöße etwas ausgleichen zu können. Es ist noch darauf hinzuweisen, daß bei der erfindungsgemäßen Dispensiereinrichtung am Ende jedes Pumpvorganges die Flüssigkeit in den Füllrohren um etwa 1 - 2 mm zurückgezogen wird, wodurch ein Nachtropfen sicher verhindert wird.
  • Ein Ausführungsbeispiel der Erfindung sieht vor, daß die Umleitungsbereiche der Füllrohre horizontal geradlinig sind.
  • Ein besonders vorteilhaftes Ausführungsbeispiel der Erfindung ist dadurch gekennzeichnet, daß zwischen dem Verteilerrohr und dem Ausgleichsrohr zwei nach oben führende Abschnitte der Füllrohre sind und vom Ausgleichsrohr mehr als zwei Umleitungsbereiche mit den anschließenden Abflußbereichen der Füllrohre wegführen.
  • Die Abflußbereiche der Füllrohre werden vorteilhaft von metallischen Nadeln gebildet.
  • Ein weiteres Ausführungsbeispiel sieht vor, daß das Verteilerrohr, die nach oben führenden Abschnitte und die Ableitungsbereiche der Füllrohre und gegebenenfalls das Ausgleichsrohr in einem gemeinsamen Block eingearbeitet sind, in dem die Nadeln gehalten sind. Der Block kann sowohl aus Kunststoff als auch aus Edelstahl gefertigt sein.
  • Vorteilhaft ist der Block zweiteilig gearbeitet, wobei die Trennungsebene zwischen den Blockteilen gleich ein Schnitt durch die Rohre ist.
  • Vorteilhaft ist vorgesehen, daß der Block eine untere Halteleiste für die Nadel aufweist.
  • Nachfolgend wird ein Ausführungsbeispiel der Erfindung anhand der Figuren der beiliegenden Zeichnung beschrieben. Fig. 1 zeigt einen Schnitt durch einen erfindungsgemäßen Block mit Verteilerrohr, Ausgleichsrohr und Füllrohr, die Fig. 2 zeigt einen Schnitt nach der Linie 111 - 111 der Fig. 1, die Fig. 3 zeigt einen Schnitt nach der Linie IV - IV der Fig. 1.
  • In den Figuren der Zeichnung sind die restlichen Teile der Abgabevorrichtung, wie Pumpe, Steuereinrichtung und Flüssigkeitsbehälter nicht besonders gezeigt, sie sind nach dem herkömmlichen Stand der Technik gefertigt.
  • Von der Pumpe 1 führt eine Leitung 2 zum Anschluß 3 im erfindungsgemäßen Block 4 und in diesem wird die Flüssigkeit zuerst in ein horizontal ausgerichtetes Verteilerrohr 5 geführt.
  • Im gezeigten Ausführungsbeispiel führen innerhalb des Blockes 4 vom Verteilerrohr zwei Füllrohre 6 nach oben in ein Ausgleichsrohr 7.
  • Vom Ausgleichsrohr 7 führen acht Füllrohre 6 weg, und zwar zuerst mit horizontalen Umleitungsbereichen 8, schließlich mit vertikal nach unten führenden Abflußbereichen 9.
  • Die Umleitungsbereiche 8 sind, wie aus der Fig. 1 ersichtlich, im Block 4 eingearbeitet. Die Abflußbereiche 9 der Füllrohre 6 werden im Ausführungsbeispiel von separaten Nadeln gebildet.
  • Wie aus der Fig. 1 ersichtlich, ist der Block 4 zweiteilig ausgebildet, wobei in einem Teil 4' das Verteilerrohr 5, das Ausgleichsrohr 7 und die beiden nach oben führenden Abschnitte der Füllrohre 6 ausgebildet sind, während im zweiten Teil 4" Sich die horizontalen Umleitungsbereiche 8 der Füllrohre 6 befinden. Die Nadeln, die die Abflußbereiche 9 bilden, sind ebenfalls im zweiten Teil 4" des Blockes 4 gelagert, der oben und unten mit einer Halteleiste 10 für die Nadeln versehen ist.
  • Ergänzend sei noch hinzugefügt, daß die Nadeln im gezeigten Ausführungsbeispiel einen Durchmesser von 0,6 mm und eine Länge von 21 mm haben. Die Geschwindigkeit der Flüssigkeit in den Nadeln beträgt ungefähr 0,7 m/sec.

Claims (5)

1. Dispensiereinrichtung für Flüssigkeiten, insbesondere für das Beschicken der Kavitäten von Titerplatten, mit einem über eine Leitung (2) an eine Pumpe (1) angeschlossenen horizontalen Verteilerrohr (5), von dem mindestens zwei, vorzugsweise acht, Füllrohre (6) radial wegführen, die vom Verteilerrohr (5) zuerst nach oben und dann nach einem Umleitungsbereich (8) in einen Abflußbereich (9) nach unten führen, dadurch gekennzeichnet, daß am oberen Ende des nach oben führenden Abschnittes der Füllrohre (6) ein parallel zum Verteilerrohr (5) ausgerichtetes Ausgleichsrohr (7) ist, an das die Umleitungsbereiche (8) der Füllrohre (6) anschließen.

Dispensiereinrichtung nach Anspruch 1, dadurch gekennzeichnet, daß zwischen dem Verteilerrohr (5) und dem Ausgleichsrohr (7) zwei nach oben führende Abschnitte der Füllrohre (6) sind und vom Ausgleichsrohr (7) mehr als zwei Umleitungsbereiche (8) mit den anschließenden Abflußbereichen (9) der Füllrohre (6) wegführen.
3. Dispensiereinrichtung nach Anspruch 2, dadurch gekennzeichnet, daß das Verteilerrohr (5), die nach oben führenden Abschnitte und die Umleitungsbereiche (8) der Füllrohre (6) und gegebenenfalls das Ausgleichsrohr (7) in einem gemeinsamen Block (4) eingearbeitet sind.
4. Dispensiereinrichtung nach Anspruch 3, dadurch gekennzeichnet, daß der Block (4) zweiteilig ist.
5. Dispensiereinrichtung nach Anspruch 3, dadurch gekennzeichnet, daß der Block (4) eine untere Halteleiste (10) für die Abflußbereiche (9) bildende Nadeln aufweist.
