Abstract
Данное изобретение относится к способам увеличения секреции представляющего интерес белка (POI) из эукариотической клетки, предусматривающим коэкспрессию POI и по меньшей мере одного белка, который усиливает секрецию белка, причем указанный усиливающий (энхансерный) белок выбран из группы, состоящей из ВМН2, BFR2, COG6, COY1, CUP5, IMH1, KIN2, SEC31, SSA4, SSE1. Это изобретение относится дополнительно к промоторной последовательности дрожжей, в частности промоторной последовательности гена РЕТ9 Р. pastoris, имеющей при сравнимых условиях увеличенную промоторную активность относительно промоторной последовательности белка GAP. Кроме того, это изобретение относится к экспрессирующему вектору, содержащему такую промоторную последовательность, и к применению такого экспрессируюшего вектора для экспрессии POI в клетке-хозяине. Это изобретение, относится также к новым промоторным последовательностям генов из Р. pastoris, которые применимы для экспрессии POI в дрожжах.
