Claims (9)
1. Immunokromatografisk fremgangsmåde til detektering af et specifikt stof indeholdt i mælk, som omfatter: (1) trinnet at behandle mælken med et lytisk enzym eller et overfladeaktivt stof til lysering af en bakterie i mælken, (2) trinnet at bringe mælken fra trin (1) i kontakt med en teststrimmel med en første del, der indeholder et mærket første antistof rettet mod det specifikke stof, en andel del, der er anbragt nedstrøms for den første del, på hvilken et andet antistof rettet mod det specifikke stof er immobiliseret, og en tredje del, der er anbragt opstrøms for den første del eller den anden del, og som har hulrum, der muliggør fjernelse af mælkefedt-globuliner indeholdt i mælken, ved den tredje del eller en del, der befinder sig yderligere opstrøms, og (3) trinnet at lade mælken flyde op til den anden del eller en del, der befinder sig yderligere nedstrøms, til opnåelse af et detekterbart signal af mærket ved den anden del eller en del, der befinder sig yderligere nedstrøms, hvor den immunkromatografiske fremgangsmåde er en fremgangsmåde af typen med lateral strømning, det specifikke stof er en bestanddel af en bakterie eller et stof, der udskilles af en bakterie, og retentionspartikelstørrelsen af hulrummene af den tredje del er 1 til 3,5 pm.
2. Fremgangsmåde ifølge krav 1, hvor mindst et eller begge af det første antistof og det andet antistof er monoklonale antistoffer.
3. Fremgangsmåde ifølge krav 2, hvor det første antistof og det andet antistof er monoklonale antistoffer.
4. Fremgangsmåde ifølge et hvilket som helst af kravene 1 til 3, hvor den tredje del udgøres af to eller flere typer af elementer, som har hulrum, der kan fjerne mælkefedt-globuliner med forskellige partikelstørrelser.
5. Fremgangsmåde ifølge krav 4, hvor den tredje del udgøres af et første element, der er anbragt nedstrøms, og et andet element, der er anbragt opstrøms, og retentionspartikelstørrelsen af det andet element er større end retentionspartikelstørrelsen af det første element.
6. Fremgangsmåde ifølge krav 5, hvor retentionspartikelstørrelsen af det første element er 1,0 til 2,0 pm, og retentionspartikelstørrelsen af det andet element er 3,0 til 3,5 pm.
7. Fremgangsmåde ifølge et hvilket som helst af kravene 1 til 6, hvor det lytiske enzym er et autolysin.
8. Fremgangsmåde ifølge et hvilket som helst af kravene 1 til 6, hvor en stafylokok indeholdt i mælken detekteres ved anvendelse af lysostaphin som det lytiske enzym.
9. Detekteringskit bestående af en additivopløsning indeholdende et lytisk enzym eller et overfladeaktivt stof, der er egnet til lysis af en bakterie i mælk, og en immunkromatografisk indretning til detektering af et specifikt stof indeholdt i mælk, som er behandlet med et lytisk enzym eller et overfladeaktivt stof, som omfatter en teststrimmel med en første del, der indeholder et mærket første antistof rettet mod det specifikke stof, en andel del, der er anbragt nedstrøms for den første del, på hvilken et andet antistof rettet mod det specifikke stof er immobiliseret, og en tredje del, der er anbragt opstrøms for den første del eller den anden del, og som har hulrum, der muliggør fjernelse af mælkefedt-globu-liner indeholdt i mælken, hvor den immunkromatografiske indretning er en indretning af typen med lateral strømning, det specifikke stof er en bestanddel af en bakterie eller et stof, der udskilles af en bakterie, hvor detekteringen af det specifikke stof gør det muligt at skelne bakterien fra andre bakterier på arts- eller slægtsniveau; og retentionspartikelstørrelsen af hulrummene af den tredje del er 1 til 3,5 pm.
