Claims (14)
- EP2807264 FREMGANGSMÅDE TIL FREMSTILLING AF EN SAMMENSÆTNING INDEHOLDENDE GALACTO-OLIGOSACCHARIDER1. Fremgangsmåde til fremstilling af en sammensætning omfattende et eller flere galacto-oligosaccharid(er), hvilken fremgangsmåde omfatter trinnene: a) tilvejebringelse af en blanding omfattende - en galactosyldonor omfattende en galactosylgruppe, der er bundet til en fraspaltelig gruppe, hvilken galactosyldonor har en molvægt på højst 350 g/mol, - en galactosylacceptor, som er forskellig fra galactosyldonoren, hvilken galactosylacceptor er et saccharid eller en sukkeralkohol, og hvor molforholdet mellem galactosylacceptoren og galactosyldonoren er mindst 1:10, og hvor blandingen omfatter mindst 0,05 mol/L af galactosylacceptoren, b) tilvejebringelse af et første enzym, hvor det første enzym har beta-galactosidaseaktivitet og transgalactosylerende aktivitet, hvor det første enzym kommer i kontakt med blandingen, og c) inkubation af blandingen og det første enzym, hvorved det bliver muligt for det første enzym at frigive den fraspaltelige gruppe af galactosyldonoren og overføre galactosylgruppen af galactosyldonoren til galactosylacceptoren, hvorved galacto-oligosaccharidet dannes, trin c) endvidere omfattende fjernelse af en fraspaltelig gruppe, der er frigivet fra galactosyldonoren, fra inkubationsblandingen, og hvor fjernelsen af den fraspaltelige gruppe, der er frigivet fra galactosyldonoren, indbefatter fysisk fjernelse af frie fraspaltelige grupper fra inkubationsblandingen og/eller omdannelse af frie fraspaltelige grupper til en eller flere andre kemiske ar ter, der stadig kan være til stede i inkubationsblandingen, hvorved sammensætningen, der omfatter det ene eller flere galacto-oligosaccharid(er), opnås.
- 2. Fremgangsmåde ifølge krav 1, hvor den fraspaltelige gruppe af galacto-syldonoren er en glycosylgruppe
- 3. Fremgangsmåde ifølge krav 1 eller 2, hvor galactosyldonoren er lactose eller lactitol.
- 4. Fremgangsmåde ifølge et hvilket som helst af de foregående krav, hvilken fremgangsmåde endvidere omfatter at tilvejebringe en mikroorganisme, som kan omdanne frie fraspaltelige grupper, der er frigivet fra galactosyldonoren, og gøre det muligt for mikroorganismen under inkubationen at fjerne en fraspaltelig gruppe, der er frigivet fra galactosyldonoren.
- 5. Fremgangsmåde ifølge et hvilket som helst af de foregående krav, hvilken fremgangsmåde endvidere omfatter at tilvejebringe et andet enzym, som kan omdanne frie fraspaltelige grupper, der er frigivet fra galactosyldonoren, og gøre det muligt for det andet enzym under inkubationen at omdanne en fraspaltelig gruppe, der er frigivet fra galactosyldonoren.
- 6. Fremgangsmåde ifølge krav 5, hvor det andet enzym har glucoseoxidaseaktivitet.
- 7. Fremgangsmåde ifølge et hvilket som helst af kravene 5-6, hvor a) specificitetskonstanten af det andet enzym i forhold til den frie fraspaltelige gruppe er mindst 10 gange højere end dets specificitetskonstant i forhold til galactosylacceptoren, b) specificitetskonstanten af det andet enzym i forhold til den frie fraspaltelige gruppe er mindst 10 gange højere end dets specificitetskonstant i forhold til galactosyldonoren, eller c) specificitetskonstanten af det andet enzym i forhold til den frie fraspaltelige gruppe er mindst 10 gange højere end dets specificitetskonstant i forhold til den mono-galactosylerede galactosylacceptor.
- 8. Fremgangsmåde ifølge et hvilket som helst af kravene 6-7, hvor fremgangsmåden endvidere omfatter tilvejebringelse af et enzym, som har ka-talaseaktivtet, hvilket enzym kommer i kontakt med inkubationsblandingen.
- 9. Fremgangsmåde ifølge et hvilket som helst af kravene 5-8, hvor fremgangsmåden endvidere omfatter at tilvejebringe et fjernelsesmiddel, som kan fjerne mindst en del af omdannelsesproduktet, der opnås ved omdannelse af den fraspaltelige gruppe med det andet enzym, og gøre det muligt for fjernelsesmidlet under inkubationen at fjerne mindst en del af omdannelsesproduktet.
- 10. Fremgangsmåde ifølge krav 9, hvor fjernelsesmidlet omfatter et salt af en divalent eller trivalent metalion.
- 11. Fremgangsmåde ifølge krav 9, hvor fjernelsesmidlet omfatter eller endda består af et anionbyttermateriale.
- 12. Fremgangsmåde ifølge et hvilket som helst af de foregående krav, hvor trin c) omfatter tilsætning af yderligere galactosyldonor.
- 13. Fremgangsmåde ifølge et hvilket som helst af de foregående krav, hvor koncentrationen af galactosyldonor af blandingen i trin c) bibeholdes ved en koncentration i området af 0,01-1 mol/L.
- 14. Fremgangsmåde ifølge et hvilket som helst af de foregående krav, hvilken fremgangsmåde endvidere omfatter trinnet: d) berigning af galacto-oligosaccharidet af sammensætningen i trin c).
