Den foreliggende opfindelse angår forbindelser med biologisk aktivitet som cholecystokinin-ligander, samt angår farmaceutiske præparater indeholdende disse forbindelser.
5 Nærmere angivet angår opfindelsen 2- og N-substi- tuerede-3-[indol-3-yl]propanamider med påvist aktivitet ved den centrale cholecystokinin-receptor, samt farmaceutiske præparater indeholdende disse forbindelser.
Cholecystokinin(CCK)-peptiderne er meget udbredt 10 i forskellige organer i legemet inkluderende mavetarmkanalen og endocrine kirtler, og i nerverne i de perifere og centrale nervesystemer. Forskellige biologisk aktive former er blevet identificeret inkluderende et 33-aminosyre-hormon og forskellige carboxylterminus-15 fragmenter af dette peptid (f.eks. octapeptidet CCK2(._33 og tetrapeptidet CCK^^gg) . (Se G. J. Dockrav, Br. Med. Bull., 38, (Nr. 3), 253-258 (1982)).
De forskellige CCK-peptider menes at være involveret i styringen af glat muskel-kontraktilitet, exo-20 crin og endocrin kirtelsekretion, sensorisk nervetransmission og adskillige hjernefunktioner. Indgift af de native peptider medfører galdeblærekontraktion, amylase-sekretion, aktivering af centrale neuroner, spiseinhi-bition, antikonvulsive aktioner og andre adfærdseffekter.

De biologiske aktiviteter af de omhandlede for-2_5 bindeiser er vurderet ved anvendelse af en indledende screening-test, der hurtigt og nøjagtigt målte bindingen af den testede forbindelse til kendte CCK-receptorste-der. Specifikke CCK-receptorer er påvist at eksistere i centralnervesystemet. (Se Hays et al., Neuropeptides, 2Q 1: 53-62 (1980), og Sauter et al., Science, 208: 1155-1156 (1980)).
Ved screening-testen blev de cerebrale cortexer fra CFLP-hanmus med en vægt på 30-40 g dissekeret på is, vejet og homogeniseret i 10 voluminer 50 mM Tris-HCl-'25 puffer (pH 7,4..yed 0-4°C), Den resulterende suspension blev centrifugeret, supernatanten blev bortkastet, og den vundne pellet blev vasket ved gensuspension i Tris-rHCl-puffer efterfulgt af gencentrifugering.
Den endelige pellet blev gensuspenderet i 10 volu-3Q miner 10 mM Hepes-puffer (pH 7,2 ved 23°C) indeholdende 130 mM NaCl, '4,7 mM KC1, 1 mM EDTA, 5 mg/ml kvægalbumin og bacitracin (0,25 mg/ml).
Ved mætnings-undersøgelser blev cerebrale cortica-le membraner inkuberet 120 minutter i et slutvolumen 35 på 500 μϋΐβΓ af Hepes-inkubationspuffer (pH 7,2) sammen med 0,1-^0,5 nM tritieret-pentagastrin (Amersham Chemical, England).

Ved udvekslingsforsøgene blev membraner inkuberet med en enkelt koncentration (2 nM) af ligand, sam-men med stigende koncentrationer (10 til 10 M) af konkurrerende testforbindelse. I hvert enkelt tilfælde 5 blev den ikke-specifikke binding defineret som den, der var vedvarende i nærværelse af det ikke-mærkede octapep-tid CCK26_33 (10-6 M).
Efter inkubation blev radioaktivitet, der var bundet til membraner, skilt fra den, der forelå frit i 10 opløsning, ved hurtig filtrering gennem Whatman GF/B-filtre og vasket tre gange med 4 ml' s portioner af iskold Tris-HCl-puffer. Filtre fra prøver inkuberet med tritieret pentagastrin blev anbragt i polyethylen-medi-cinglas med 8 ml scintillations-cocktail, og radioakti-15 viteten blev bestemt ved scintillations-spektrometri (effektivitet 47-52%).
Den specifikke binding til CCK-receptorsteder blev defineret som det totale bundne tritierede pentagastrin minus den mængde tritieret pentagastrin, der var 20 bundet i nærværelse af 10 M octapeptid, CCK2g_33.
Mætningskurver for specifik tritieret-pentaga-strin-binding til corticale muse-membraner blev analyseret ved statistiske metoder for at tilvejebringe vurderinger vedrørende det maksimale antal bindingssteder 25 (Bmax) 0<3 ligevægts-dissociationskonstanten (K^) .

5 Forbindelserne med en K^-værdi på 10 ymolær eller mindre blev betragtet som aktive og blev derefter underkastet en anden in vivo-screeningstest, ved hvilken forbindelsens evne til at undertrykke appetit hos standard-laboratorierotter blev vurderet.
10 Ved denne screening blev voksne Wistar-hanrotter med en vægt på 250-400 g anbragt individuelt og trænet til at konsumere deres daglige foderindtagning i løbet af en seks timers periode. Foder blev derefter fjernet i den resterende 18 timers periode af hvert døgn. Drik-15 kevand var til rådighed hele tiden, og dyrene holdtes på en 12 timer lys/12 timer mørke-cyclus. Denne træning foregik i en periode på syv dage. Pulveriseret snarere end pelleteret foder anvendtes for at begrænse spild til et minimum og lette måling af foder indtagning.
20 På hver testdag anvendtes en gruppe på 12 rotter, seks der fik en enkelt dosis af testforbindelse, og seks der tjente som kontrolgruppe. Straks efter dosering fik rotterne adgang til foder. Foderindtagning fulgtes derefter over den næste 6 ti-mers-periode. Fodertragtene 25 blev vejet med mellemrum på 30 minutter og 1, 2, 3, 4, 5 og 6 timer efter indgift af testforbindelsen. Den procentuelle repression af foderindtagning hos laboratorierotter under disse betingelser for henholdsvis det kendte octapeptid CCK26_33, °9 for en række forbindelser ifølge 30 opfindelsen er vist i Tabel II.

Ved terapeutisk anvendelse som appetit-undertrykkende midler bliver forbindelserne givet til patienten ved dosisniveauer på fra ca. 200 til ca. 2800 mg pr. dag. De specifikke doser kan imidlertid 5 varieres alt efter patientens tilstand, alvoren af den behandlede tilstand og aktiviteten af den anvendte forbindelse. Fastlæggelse af optimale doser for en speciel patient ligger inden for fagmandens kunnen.
Til fremstilling af farmaceutiske præparater 10 ud fra de omhandlede forbindelser kan indifferente, farmaceutisk acceptable bærere være enten faste eller flydende. Præparater i fast form omfatter pulvere, tabletter/ dispergerbare granuler, kapsler, oblatkapsler og suppositorier.
15 En fast bærer kan være ét eller flere stoffer, der også kan fungere som fortyndingsmidler, smagsmidler, opløsningsformidlere, smøremidler, suspenderingsmidler, bindemidler eller tablet-disintegrationsmidler. Den kan også være et indkapslende materiale.
20 X pulvere er bæreren et findelt fast stof, der foreligger i blanding med den findelte aktive komponent.
I tabletter er den aktive komponent blandet med bæreren, der har de nødvendige bindende egenskaber, i passende forhold og komprimeret i ønsket form og størrelse.
25 Til fremstilling af suppositorie-præparater bli ver et lavtsmeltende voks, såsom en blanding af fedt-syreglycerider og kakaosmør, først smeltet, og den aktive bestanddel dispergeres deri ved for eksempel omrøring, Den smeltede homogene blanding hældes derefter i 30 forme af passende størrelse og henstilles til afkøling og størkning.
Pulvrene og tabletterne indeholder fortrinsvis fra ca. 5 til ca. 70 vægt% af den aktive komponent. Egnede bærere er magnesiumcarbonat, magnesiumstearat, 16 DK 168006 B1 talkum, lactose, sukker, pectin, stivelse, tragacanth, methylcellulose, natriumcarboxymethylcellulose, et lavtsmeltende voks, kakaosmør og lignende.
Angivelsen "farmaceutisk præparat" skal omfatte 5 en komposition af den aktive komponent med indkapslende materiale som en bærer tilvejebringende en kapsel, hvori den aktive komponent (med eller uden andre bærere) er omgivet af en bærer, der på denne måde er kombineret dermed. På tilsvarende måde er også inkluderet 10 oblatkapsler.
Tabletter, pulvere, oblatkapsler og kapsler kan anvendes som faste dosisformer egnede til oral indgift.
Flydende præparater omfatter opløsninger, suspensioner og emulsioner. Sterile vand- og vand/propy-15 lenglycol-opløsninger af de aktive forbindelser er eksempler på flydende præparater, der er egnede til parenteral anvendelse af de omhandlede forbindelser. Flydende præparater kan også foreligge i opløsning i vandig polyethylenglycolopløsning.
20 Vandige opløsninger til oral anvendelse af de omhandlede forbindelser kan fremstilles ved at opløse den aktive forbindelse eller et salt deraf i vand og tilsætte egnede farvemidler, smagsstoffer, stabilisatorer og fortykningsmidler, som ønsket. Vandige suspen-25 sioner til oral anvendelse Tcan fremstilles ved at disperge-re den findelte aktive komponent i vand sammen med et viskost materiale, såsom naturlige eller syntetiske gummiarter, harpikser, methylcellulose, natriumcarboxymethylcellulose og andre suspenderingsmidler, der er 30 kendt til farmaceutiske præparater.
Det farmaceutiske præparat er fortrinsvis i enhedsdosisform. I sådan form er præparatet opdelt i enhedsdoser indeholdende passende mængder af den aktive komponent. Enhedsdosisformen kan være et emballeret 35 præparat, idet emballagen indeholder separate mængder . af præparatet som for eksempel emballerede tabletter, DK 168006 B1 17 kapsler og pulvere i medicinglas eller ampuller. Enhedsdosisformen kan også være en kapsel, oblatkapsel eller tablet i sig selv, eller den kan være et passende antal af enhver af disse emballerede former.
5 Opfindelsen beskrives nærmere gennem følgende eksempler.
Eksempel 1
Fremstilling af N-[(l,l-dimethylethoxyicarbonyl] 10 thyltryptophyl—L-rphenylalaninamid - Blanding af dia- stereomere.
N-tert-rButyloxycarbonyl-a •'-methyl tryptophan (2,29 g, 7,19 mmol) blev opløst i 50 ml tørt, destilleret ethylacetat, og der blev tilsat 1,38 g (9,0 mmol) 15 hydroxybenzotriazol og 0,05 g 4^dimethylaminopyridin. Blandingen blev afkølet til 5°C, og en opløsning af 1,65 g (8,0 mmol). dicyclohexylcarboddimid i 10 ml ethylacetat blev tilsat. 30 Produktet fra Eksempel 11 blev behandlet med en 20% opløsning af piperidin i dimethylformamid ved 0°C i 30 minutter. En opløsning af 0,14 g (1,2 mmol) tert-butyleddi-kesyre i 5 ml ethylacetat blev behandlet med 0,367 g 15 (2,4 mmol) hydroxybenzothiazolhydrat og 0,247 g (1,2 mmol) dicyclohexylcarbodiimid. 30 En opløsning af trichlorbutoxycarbonyl-DL-a- methyl-tryptophan (0,422 a, 1,0 mmol) i 10 ml dichlor-methan blev behandlet med 0,203 mg (1,1 mmol) penta-fluorphenol efterfulgt af 0,216 mg (1,05 mmol) dicvclo-hexvlcarbodiimid. Den resulterende blanding blev om-35 rørt ved stuetemperatur i 30 minutter. En opløsning af 0,10 g (0,43 mmol) DL-a-methyl- DK 168006 B'l 27 tryptophan i 5 ml vand blev behandlet med 0,095 g (0,9 mmol) natriumcarbonat og omrørt samtidigt med, at der langsomt blev tilsat en opløsning af 0,106 g (0,5 mmol) trichlorethoxycarbonylchlorid i 3 ml dioxan. Den 5 resulterende blanding blev omrørt i tre timer, hvorefter dioxanet blev fjernet i vakuum, og inddampningsresten blev fortyndet med 5 ml vand. Eksempel 32
Fremstilling af [1-(lH-indol-3-ylmethyl)-l-methyl-2-20 oxo-2- [ [2- (2-pyridinyl) ethyl] amino] ethyl] carbaminsyre- 2.2.2- trifluor-1,1-dimethylester.
Trin 1: Fremstilling af lH-imidazol-l-carboxylsyre- 2,2,2-trif luor-1,1-dimethylethylester.
25 1,1-Carbonyldiimidazol (6,3 g, 39 mmol) blev op løst i 100 ml tørt toluen opvarmet til tilbagesvaling.
En opløsning af l,l,l-trifluor-2-methyl-2-propanol (2 g, 15,5 mmol) i 80 ml toluen blev tilsat dråbevis. Blandingen blev omrørt i to timer.
30 Efter dette tidsrum blev toluenet fordampet, hvorved vandtes 9,6 lH-imidazol-l-carboxylsvre-2,2,2-trifluor-1,1-dimethylethylester som et hvidt fast stof.
Trin 2: Fremstilling af N-(2,2,2-trifluor-1,1-dimethyl-3 5 ethoxycarbonyl) -α-methyltryptophan-methylester.
Et halvt gram af det rå reaktionsprodukt fra Trin 1 ovenfor (indeholdende ca. 0,17 g, 0,77 mmol af DK 168006 B1 32 det ønskede materiale) blev opløst i 3 ml dioxan, og opløsningen blev opvarmet til tilbagesvaling. En opløsning af a-methyltryptophan-methylester (0,35 g, 1,45 mmol) i 5 ml dioxan blev tilsat dråbevis i løbet af en 5 halv time. Blandingen blev opvarmet og omrørt under tilbagesvaling i to dage, hvorefter der blev tilsat yderligere 0,175 g (0,77 mmol) a-methyltryptophan-methylester. Omrøring og opvarmning af denne blanding blev fortsat i yderligere 24 timer.
10 Blandingen blev derefter inddampet, og der blev tilsat 30 ml ethylacetat, og den brunf arvede opløsning blev vasket successivt med portioner af mættet natrium-bicarbonatopløsning og brine. Det organiske lag blev fraskilt, tørret over vandfrit magnesiumsulfat og ind-15 dampet, hvorved vandtes en brun olie.
Det rå reaktionsprodukt blev chromatograferet på en silicagelsøjle, idet der blev elueret med 25% ethylacetat i hexan, hvorved vandtes 100 mg N-(2,2,2-tri-f luor-1,1-dimethylethoxycarbonyl) -a-methyltryptophan-20 methylester som en stråfarvet olie.
PMR-spektret af materialet viste spidser ved 1,25 (singlet), 1,7 (dublet), 3,0-3,5 (multiplet), 3,70 (singlet) og 6,9-7,7 (multiplet) ppm nedstrøms fra te-tramethylsilan-signalet.

Trin 3: Fremstilling af N-(2,2,2-trifluor-1,1-dimethyl-ethoxycarbonyl)-a-methyltryptophan.
N- (2,2,2-trif luor-1,1-dimethylethoxycarbonyl) -a-methyltryptophan-methylester (0,1 g, 0,29 mmol) blev op-30 løst i 3 ml dioxan. Til denne opløsning blev der sat 0,04 g (0,95 mmol) lithiumhydroxid opløst i 2 ml vand.
Denne blanding blev omrørt ved stuetemperatur i 24 timer.
Ved afslutningen af denne periode blev blandingen 35 inddampet, og der blev tilsat 5 ml vand. Opløsningen blev syrnet ved tilsætning af 2M saltsyre, og den syrnede blanding blev ekstraheret fem gange med ethylacetat.

Trin 4: Fremstilling af [1-(lH-indol-3-ylmethyl)-1-me-thyl-2-oxo-2-[[2-(2-pyridinyl)ethyl]amino]-ethyl]carbaminsyre-2,2,2-trifluor-1,1-dimethyl-ester.
15 En opløsning af N-(2,2,2-trifluor-l,l-dimethyl- ethoxycarbonyl)-a-methyltryptophan (0,06 g, 0,16 mmol) i 2 ml ethylacetat blev afkølet i et isbad og derefter behandlet successivt med 30 ul (0,17 mmol) triethylamin og 28 yl (0,17 mmol] isobutylformiat.
20 Efter 45 minutter blev blandingen yderligere be handlet med en opløsning af 25,7 yl (0,17 mmol) 2-(2-pyridinyl)ethylamin i 2 ml ethylacetat. Efter 1 time fik reaktionsblandingen lov at opvarme til stuetemperatur og blev omrørt ved denne temperatur i 18 timer.
25 Derefter blev de udfældede faste stoffer frafil treret, og opløsningen blev vasket successivt med portioner af mættet natriumbicarbonat og brine, tørret og inddampet, hvorved vandtes 40 mg af en grøn olie. Dette råprodukt blev renset ved søjlechromatografi på silica— 30 gel, idet der blev elueret med 25% ethylacetat i hexan, hvorved vandtes 15 mg af en gul olie, der blev krystalliseret af dichlormethan/hexan, hvorved vandtes [1-(1H-indol-3-ylmethyl]-l-methyl-2-oxo-2-[[2-(2-pyridinyl)-ethyl]amino]ethyl]carbaminsyre-2,2,2-trifluor-1,1- 35 dimethylester som et amorft gult fast stof.
PMR-spektret af dette materiale viste spidser ved 1,6 (singlet), 1,62 (singlet), 2,8 (triplet), 3,35 (singlet), 3,60 (triplet), 6,9-7,6 (multiplet), 8,15 34 DK 168006 B1 (singlet) og 8,4 (dublet) ppm nedstrøms fra tetrame-thylsilan-signalet.

Fremstilling af (i) - [2-[2 (cyclohexylethyl) amino] -1-(lH-indol-3-yl) methyl) -l-methyl-2-oxoethyl] carbaminsyre- 1,1-dimethylpropylester.
20 tert-Amyloxycarbonyl-a-methyl tryptophan (1 mmol) blev opløst i 3 ml ethylacetat, og der blev tilsat 1,9 mmol hydroxybenzotriazol. Denne blanding blev omrørt i tre timer ved stuetemperatur. Til denne blanding blev der sat 1 mmol 2-(cyclohexyl)ethylamin (1 mmol), og 25 blandingen blev omrørt ved stuetemperatur i to dage.
