Den foreliggende opfindelse angår en analogifrem-gangsmåde til fremstilling af hidtil ukendte 4-aryl-4-piperi-dincarbinoler med nedenstående formel (IV) eller farmaceutisk acceptable salte deraf. Disse forbindelser er værdifulde 5 som antidepressive midler og i nogle tilfælde som anorektiske midler.
Mental sygdom omfatter både psykoser og neuroser.
Symptomer, der kræver behandling, omfatter depression, angst, sindsoprør og hallucinationer. Blandt de lægemidler, der 10 især anvendes til behandling af både reaktive og endogene depressioner, er monoaminoxidase-(MAO)-inhibitorer, såsom iproniazid, tranylcypromin, nialamid, phenelzin og pargylin, og de ikke-MAO-inhiberende tricycliske aromatiske diben-zazepiner, såsom imipramin, og dibenzocycloheptener, såsom 15 amitriptylin.
Alle disse lægemidler har alvorlige bivirkninger, som begrænser ders anvendelighed. MAO-inhibitorer kan gavne mildere former for depression, men risikoen for alvorlige toksiske virkninger er et kraftigt argument imod deres anven-20 delse. De kan forårsage leverskade og akut hypertension, især hvis de gives i forbindelse med ost, bananer eller anden aminholdig føde. MAO-inhibitorerne kan også forårsage skælven, søvnløshed, hyperhydrosis, sindsoprør, hypermanisk opførelse, forvirring, hallucinationer, krampetrækninger og 25 orthostatisk hypotension. De forårsager ofte svimmelhed, vertigo, hovedpine, inhibering af ejakulation, urinerings-besvær, svaghed, træthed, tør mund, forstoppelse og sløret syn.
Imipramin kan forårsage sløret syn, tørhed i munden, 30 forstoppelse, urineringsretension, orthostatisk hypotension, åndedrætsdepression, myocardieinfarktion og hjertelammelse ved forøget blodtilførsel. Lignende vanskeligheder kendes ved amitriptylin.
Der er stadigt behov for psykoterapeutiske midler, 35 som har færre bivirkninger end de lægemidler, der anvendes i dag. Der er også behov for psykoterapeutiske midler, der 2

har andre virkningsmåder end de midler, der for tiden anvendes, da ingen af disse er fuldstændig effektive.

Det følgende er mere specikke detaljer om de omsætninger, der er beskrevet ovenfor. 1
Fremstilling af (I)
20 Lithiumbehandling af R4Br kan udføres ved -100°C
til 50°C i et etherisk opløsningsmiddel, f.eks. diethyl-ether eller tetrahydrofuran. Kendte alment anvendte midler til lithiumbehandling omfatter f.eks. n-butyl-lithium, methyllithium og see.- og tert.-butyllithium.
25 Som det er kendt i teknikken7kan et magnesiumreagens ifølge Grignard anvendes i stedet for alkyllithiumforbin-delsen, i hvilket tilfælde det fremstillede mellemprodukt er en organomagnesiumforbindelse. I teknikken er det også kendt, som f.eks. beskrevet af Gschwend og Rodriguez, 30 Organic Reactions, bind 26, at nogle forbindelser R4H, 4 hvori R er som defineret ovenfor, kan lithiumbe- 4 handles direkte fremfor via mellemproduktet R Br. På det andet trin i fremstillingen af (I) omsættes organolithium- eller organomagnesiumforbindelsen med ! 5 ovennævnte piperidinon enten rent eller i et etherisk opløsningsmiddel, f.eks. diethylether eller tetrahydro-furan^ved -70°C til 50°C.
2) Fremstilling af (II) 10 Dehydratiseringen af (I) til (II) kan udføres i nær værelse af en katalysator, såsom saltsyre, hydrogenbromid-syre, methansulfonsyre/phosphorpentoxid, trifluoreddikesyre eller en arylsulfonsyre, enten i et opløsningsmiddel, såsom et aromatisk carbonhydrid, eller i fraværelse af 15 et opløsningsmiddel. Afhængigt af arten af substituenterne 12 3 4 R , R , R og R kan dehydratiseringen udføres ved en egnet temperatur i området fra 0 til 200°C. Efter dehydratiseringen neutraliseres syrekatalysatoren til omdannelse af aminsaltet til den fri amin (II).
20 3) Fremstilling af (III)
Forbindelse (II) kan metalbehandles som beskrevet ovenfor i forbindelse med fremstillingen af (I). Lithium-behandling kan udføres ved -70°C til 70°C, fortrinsvis 25 ved -10°C til -20°C, i fraværelse af både oxygen og vand.
Det således fremstillede metalsalt i opløsning kan omsættes med ovennævnte keton R^COR^ i et etherisk opløsningsmiddel, f.eks. diethylether eller tetrahydrofuran, ved -100°C til 50°C. Ved hydrolyse af det resulterende lithium-30 eller magnesiumsalt opnås (III).
4) Fremstilling af (IV)
Reduktion af (III) tilvejebringer (IV). Reduktionen kan udføres ved hjælp af borhydridreagens.er, såsom natrium-35 borhydrid i en alkohol eller natriumcyanoborhydrid i en alkohol/eddikesyreblanding, eller ved hjælp af kataly-

7 tisk hydrogenering, f.eks. under anvendelse af en palladium-katalysator, i et eddikesyre- eller alkoholopløsningsmiddel i nærværelse af en mineralsyre.
I en hvilken som helst af ovennævnte fremgangsmåder, 5 hvori tilstedeværende forbindelser har en piperidindel, kan N-substituenten R1 være benzyl. Det N-benzylerede produkt er også et værdifuldt mellemprodukt, og det kan omdannes ved katalytisk hydrogenolyse under anvendelse af en palladium-katalysator i et eddikesyreopløsningsmiddel til den 10 sekundære amin, dvs. (IV), hvori R1 er H. Den sekundære amin kan alkyleres ved hjælp af det egnede alkylhalogenid eller cycloalkylhalogenid til fremstilling af (IV), hvor R1 er alkyl med 1-12 carbonatomer eller cycloalkyl med 3-8 carbonatomer. Nogle af R^—grupperne kan indføres ved en 15 alternativ fremgangsmåde, hvor den sekundære amin først acyleres med et acylchlorid, hvis acylgruppe, når den er reduceret, svarer til den ønskede R1-gruppe. Amidet, der fremstilles ved acylering, kan reduceres med et hydridreduk-tionsmiddel, såsom BH3 eller LiAlH4, til tilvejebringelse 20 af den ønskede amin (IV).

Eksempel 1 4-Pfrenyl-a,α-1-trlmethy1-4-piperidinmethanol n-Butyllithium (15 ml af en 1,5 molær opløsning i hexan) sættes under nitrogen til en opløsning af 4,7 g af et 5 kommercielt tilgængeligt l-methyl-4-phenyl-l,2,3,6-tetra- 1 2 3 4 hydropiperidin (II: R = Me; R , R = H, R = phenyl) i 50 ml tetrahydrofuran og holdes ved -10°C. En opløsning af 1,47 g af ovennævnte produkt (III) i en blanding af 10 ml methanol og 2 ml eddikesyre afkøles med is og behandles med 0,76 g natriumcyanoborhydrid. Blandingen omrøres ved stuetemperatur i 2 timer, og over-25 skuddet af borhydrid sønderdeles ved tilsætning, med isafkøling, af 8 ml koncentreret saltsyre. Efter omrøring ved stuetemperatur i 0,5 timer gøres blandingen basisk med en vandig natriumhydroxidopløsning og ekstraheres med methylenchlorid. Eksempel 2 4-(31-Trifluormethylphenyl)-g, a,l-trimethyl-4-piperidin-methanol og hydrochloridet deraf
En 1,5 molær opløsning af n-butyllithium i 150 ml hexan sættes under nitrogen ved -70°C til en omrørt opløsning 10 af 50 g 3-brombenzotrifluorid i 300 ml tetrahydrofuran. Blandingen omrøres ved -70°C i 15 minutter og får derefter lov til at opvarmes til -20°C. En opløsning af 30 g frisk destilleret l-methyl-4-piperidon i 50 ml tetrahydrofuran tilsættes langsomt, medens temperaturen holdes ved -20°C. Derefter omrøres blandingen ved 0°C i 0,5 timer og ved stuetemperatur i 3 timer. Vand og methylenchlorid tilsættes, og det vandige lag ekstraheres flere gange med methylenchlorid. Eksempel 3 1-[1'-Methyl-41-(211,311-dlmethoxyphenyl)-41-piperidinyl]--cyclopentanol 5 En opløsning af 130 g veratrol i 400 ml tørt tetra- hydrofuran afkøles i et tøris/acetonebad og behandles med 500 ml af en 1,6 molær opløsning af n-butyllithium ved en sådan hastighed, at reaktionsblandingens temperatur forbliver ved 0°C. Efter at tilsætningen er afslutttet, 10 omrøres reaktionsblandingen ved stuetemperatur i 3 timer.
Ved afslutningen af denne periode afkøles den resulterende hvide opslæmning i et tøris/acetonebad, og der tilsættes 90j4 g l-methyl-4-piperidon ved en sådan hastighed, at reaktionsblandingens temperatur forbliver under -10°C.
15 Efter at tilsætningen er afsluttet, omrøres reaktionsblandingen ved 0°C i 2 timer, blandes hurtigt med 400 ml vand og fortyndes med 400 ml ether. Den aktive ingrediens kan indgives oralt på faste 10 dosisformer, såsom kapsler, tabletter og pulvere, eller på flydende dosisformer, såsom eliksirer, sirupper og suspensioner. Den kan også indgives parenteralt, på sterile, flydende dosisformer, eller rektalt i form af suppositorier.
15 Gelatinekapsler kan indeholde den aktive ingrediens og pulveriserede bærestoffer, såsom lactose, saccharose, mannitol, stivelse, cellulosederivater, magnesiumstearat og stearinsyre. Lignende fortyndingsmidler kan anvendes til fremstilling af sammenpressede tabletter. Både tabletter 20 og kapsler kan fremstilles som produkter med forsinket frigivelse til tilvejebringelse af kontinuerlig frigivelse af medicin i løbet af timer. Sammenpressede tabletter kan være overtrukkede med sukker eller folie for at skjule ubehagelig smag og beskytte tabletten mod atmos-25 færen, eller de kan være overtrukket enterisk til selektiv disintegration i den gastrointestinale kanal.
Flydende dosisformer til oral indgivelse kan indeholde farve- og smagsstoffer for at forøge patientens godtagelse .
30 Almindeligvis er vand, en egnet olie, en fysiologisk saltopløsning, vandige dextrose- (glucose-) og beslægtede sukkeropløsninger og glycoler, såsom propylenglycol eller polyethylenglycoler, egnede bærestoffer til parenterale opløsninger. Opløsninger til parenteral indgivelse indeholder 35 fortrinsvis et vandopløseligt salt af den aktive ingrediens, egnede stabiliseringsmidler, og om nødvendigt pufferstoffer.

En standardfremgangsmåde til opdagelse og sammenligning af den antidepressive aktivitet af de her omhandlede forbindelser, for hvilke der er god korrelation med human virkning, er forebyggelsen af tetrabenazinfremkaldt sedation og depres-5 sion hos mus. (Everett, "The Dopa Response Potentation Test and Its Use in Screening for Antidepressant Drugs", side 164-167 i Antidepressant Drugs (Proceedings of the First International Symposium), S. Garattini & M.N.G. Dukes (udgivere), 1967).
10 Grupper på 10 Carworth-CF^-hunmus, der vejer 18-21 g, fastes i 1,5 timer og intuberes med antagonistiske forbindelser i orale doser, såsom 0, 1, 3, 9, 27 og 81 mg/kg i 0,20 ml 1%1 s "Methocel®" (methylcellulose). Protokol over anorexia-data fra CNS-forsøg
Til disse forsøg anvendes hunmmus, der vejer 16-20 g og som har fastet natten over i 17-21 timer. 30 minutter efter indgivelse af graduerede orale doser af en for-5 bindelse fremstillet i vandigt "Methocel®" (methyl- cellulose, viskositet 100 cps, grad MC , Dow Chemical Co.) og 2,8% "Tween 80" og doseret med 0,1 ml pr. 10 g legemsvægt, overføres hver mus til et individuelt, lyst, "Lucite®"rum (13,3 x 12,7 x 12,7 cm) med et gulv af trådnet 10 med 0,64 x 0,64 cm tråde. I hvert bur anbringes 5 run lineært. Inden i hvert rum er der en del af- en sort "Lucite^'-stang (13 x 1,2 x 1,2 cm) , på hvis top, der er pletformede fordybninger (0,8 cm i diameter), som hver især indeholder 0,05 ml 50%'s sødet, kondenseret mælk.
