Description
Denne oppfinnelse angår fremstilling av nye 4-aryl-4-piperidincarbinoler som er anvendelige som;antidepres-siva og i enkelte tilfeller som anorektiske midler.
Sinnssykdommer omfatter både psykoser og neuroser. Symptomer som krever behandling, innbefatter depresjon, angst, opphisselse (agitatio) og hallusinasjoner. Blant de legemidler som særlig anvendes for behandling av såvel reaktive som endogene depresjoner, er inhibitorer for monoaminoxidase (MAO), såsom iproniazid, tranylcypromin, nialamid, phenelzin og pargylin, og de ikke-MAO-inhiberende tricykliske aroma-tiske dibenzazepiner, såsom imipramin og dibenzoecyclohepte-ner såsom amitriptylin.
Alle disse legemidler har uheldige bivirkninger
som begrenser deres anvendelighet. MAO-inhibitorer kan være gunstige ved mildere former for depresjon, men risikoen for alvorlige toksiske virkninger er et sterkt argument mot å bruke disse. De kan forårsake leverskade og akutt hypertensio, spesielt dersom de inntas sammen med ost, bananer eller andre aminholdige næringsmidler. MAO-inhibitorer kan også forårsake skjelvninger, søvnløshet, hyperhydrosis, agitatio, hypermanisk adferd, sinnsforvirring, hallusinasjoner, kramper og ortostatisk hypotensio. De forårsaker ofte uklarhet, svimmelhet, hodepine, hindret sæduttømning, problemer med vannlatningen, svakhet, tretthet, tørr munn, forstoppelse og uklart syn.
Imipramin kan forårsake uklart syn, tørr munn, forstoppelse, tilbakeholdelse av urinen, ortostatisk hyptensio, svekket åndedrett, myocardialt infarkt og hjertesvikt som følge av congestio. Tilsvarende problemer er erfart med amitriptylin.
Det foreligger stadig et behov for psykoterapeutiske midler som har færre bivirkninger enn de legemidler som anvendes idag, også for psykoterapeutiske midler som virker på annen måte enn de midler som for tiden anvendes, da ingen av disse er helt effektive.
I britisk patentskrift nr. 888 657 og canadisk patentskrift nr. 1 079 734 beskrives piperidincarbinoler som er anvendelige som analgetica og som hostestillende midler,
og som har den generelle formel:
hvor X er fenyl, Y er hydroxymethyl, 1-hydroxyethyl eller 1-hydroxypropyl, alk er alkylen med inntil 6 carbonatomer, og R er en oxygen- eller nitrogen-inneholdende heterocyklisk gruppe, tetrahydrofurfuryloxyethyl, aryl, aryloxy, aralkoxy, alkoxy med opp til 6 carbonatomer eller alkoxy substituert med hydroxy, ethoxy eller fenoxy.
Forbindelser av denne struktur, hvor Y er alkanoyl og de øvrige substituenter er definert på lignende måte, er dessuten kjent fra britisk patentskrift nr. 841 120.

Med den foreliggende oppfinnelse tilveiebringes det nu nye, terapeutisk virksomme 4-aryl-4-piperidincarbinoler med den generelle formel:

a) er H, alkyl med 1-4 carbonatomer eller benzyl,
2 3 b) hver av R og R uavhengig av hverandre er valgt blant H og lavere alkyl med 1-4 carbonatomer, idet

imidlertid R og R sammen kan være en uforgrenet alkylenbro med 3 eller 4 carbonatomer,
c) R er
1) fenyl eller 2-nafthyl, hvilke kan være substituert med én eller to substituenter som kan være like eller forskjellige, og som er valgt fra F, Cl, alkyl, perfluoralkyl, alkoxy, benzyloxy, alkylthic og dialkylamino, idet alkyl- og alkoxy-gruppene har fra 1 til 4 carbonatomer,
2) 2-, 3- eller 4-bifenylyl,
3) 2-pyrrolyl eller 2-pyrrolyl substituert med fra én til tre lavere alkylgrupper med 1-4 carbonatomer,
4) 2-, 3- eller 4-pyridyl, eller
5) 2-thienyl som i 5-stillingen er substituert med lavere alkyl med 1-4 carbonatomer,
d) hver av R"* og R^ uavhengig av hverandre er valgt blant alkyl med 1-4 carbonatomer,og R <5> og R <6>sammen
danner en forgrenet eller uforgrenet alkylenbro med fra 3 til 11 carbonatomer,

forutsatt at når R , R og R er methyl og R2 og R 3 er hydrogen, kan R<4> ikke være p-t-butylfenyl eller 2'-bifenylyl, samt deres estere med alifatiske mono- og dicarboxlsyrer med 1-8 carbonatomer og deres aminsalter med i farmasøytisk henseende forlikelige uorganiske syrer.
De nye 4-aryl-4-piperidincarbinoler fremstilles ved at et oxaazabicyclooctan med den generelle formel:
hvor R 1 - R6, har de: ovenfor angitte betydninger, reduseres, og at den erholdte piperidincarbinol, om ønkes, overføres til en ester med en alifatisk mono- eller dicarboxylsyre med 1-8 carbonatomer eller til et aminsalt med en i farmasøy-tisk henseende forlikelig uorganisk syre.
En flertrinns syntese av forbindelsene fra enkle ut-gangsmaterialer kan utføres som følger:
1) R<4>Br lithieres med n-butyl-lithium for dannelse av R<4>Li, som så omsettes med piperidinon med formelen:

som så kan omsettes med R^Li eller R^MgX, hvor R** er som ovenfor angitt og X er Cl, Br eller I, for dannelse av III. Som det vil forstås, kan denne alternative fremgangsmåte ikke benyttes dersom R 5 og R 6 sammen danner alkylen i forbindelsen med formel IV.
Nedenfor er de ovenfor skisserte reaksjoner beskrevet mer detaljert.
1) Fremstilling av I
Lithieringen av R 4Br kan utføres ved en temperatur fra -100° C til 50° C i et etheroppløsningsmiddeT, såsom for eksempel diethylether eller tetrahydrofuran. Kjente vanlig benyttede lithieringsmidler innbefatter for eksempel n-butyllithium, methyllithium og sek- og tert-butyllithium. Som kjent i faget kan det benyttes et magnesium-Grignard-reagens i steden for alkyllithiumforbindelsen, i hvilket tilfelle mellomproduktet som dannes, er en organomagnesium-forbindelse. Som likeledes kjent i faget, og som beskrevet for eksempel av Gschwend og Rodriguez i Organic Reactions, Vol. 26, kan visse forbindelser R 4 H, hvor R 4 som ovenfor angitt, lithieres direkte i stedet for via mellomproduktet R<4>Br. Tetramethylethylendiamin kan benyttes sammen med alkyllithiumforbindelsen. Slike direkte-lithierte forbindelser innbefatter alkoxybenzenene, hvor lithieringen finner sted i ortho-stilling til alkoxysubstituenten, foruten pyrrol og 5-alkylthiofener, hvor lithieringen finner sted i 2-stillingen.
I det andre trinn av fremstillingen av I' omsettes organolithium- eller organomagnesiumforbindelsen med det ovenfor omtalte piperidinon enten uten bruk av oppløsningsmiddel eller i et etheroppløsningsmiddel, såsom for eksempel diethylether eller tetrahydrofuran, ved temperatur fra -70°C til 50° C.
2) Fremstilling av II
Dehydratiseringen av I til II kan utføres i nærvær av en katalysator, såsom saltsyre, bromsyre, methansulfonsyre/ fosforpentoxyd, trifluoreddiksyre eller en arylsulfonsyre, enten i et oppløsningsmiddel, for eksempel et aromatisk hydro-carbon, eller i fravær av et oppløsningsmiddel. Avhengig av

arten av substituentene R , R , R og R kan dehydratiseringen utføres ved en passende temperatur i området fra 0° til 200° C. Etter dehydratiseringen nøytraliseres den sure katalysator for å overføre aminsaltet til det frie amin II.
3) F remstilling av III
Forbindelse II kan metalleres som beskrevet ovenfor i forbindelse med fremstillingen av I. Lithieringen kan ut-føres ved en temperatur fra -70° C til 70° C, fortrinnsvis ved temperaturer fra -10° C til -20° C, i fravær av både oxygen og vann. Det således dannede metallsalt i oppløsning kan omsettes med ovennevnte keton R 5 COR 6 i et etheroppløs-ningsmiddel, såsom for eksempel diethylether eller tetrahydrofuran, ved temperaturer fra -100° C til 50° C. Etter hydrolyse av det erholdte lithium- eller magnesiumsalt fås forbindelsen III.
4) Fremstilling av IV
Ved reduksjon av III fås IV. Reduksjonen kan utfø-res av borhydridraagenser såsom natriumborhydrid i alkohol eller natriumcyanoborhydrid i en alkohol/eddiksyreblanding, eller ved katalytisk hydrogenering, f.eks. under anvendelse av en palladiumkatalysator, i et eddiksyre- eller alkohol-oppløsningsmiddel, i nærvær av en mineralsyre.
Ved den ovenfor omtalte alternative fremgangsmåte kan den metallerte forbindelse II, i oppløsning, omsettes med 5 7 6 esteren R COOR for dannelse av V, som omsettes med R Li eller R^MgX i et etheroppløsningsmiddel ved temperaturer fra -20° C til 50° C. Ved hydrolyse av det resulterende lithium- eller magnesiumsalt fås III.
Egnede salter med i farmakologisk henseende aksep-terbare syrer, såsom saltsyre, svovelsyre, fosforsyre og maleinsyre, kan fremstilles ut fra de frie baser IV. Slike salter kan være å foretrekke når de frie baser er oljer. Salter av baser IV kan også være mer lagringsstabile og mer absorberbare oralt enn de frie baser.
I den etterfølgende tabell 1 er det gitt en oversikt over de forbindelser som er fremstilt i de etterfølgende eksempler. "Me" er CH3, "Et" er C2H5, "Ph" er CgH5, "PhCH2" er benzyl og "Cp" er cyclopentyl i tabellen.
De nye antidepressiva kan administreres for å be-handle psykiatriske depresjoner av den reaktive og den endogene type på en hvilken som helst måte som gir kontakt mellom det aktive middel og det sted i et pattedyrs legeme hvor det aktive middel skal gjøre sin virkning. De kan admninistreres på en hvilken som!helst konvensjonell måte som benyttes for legemidler, enten som individuelle terapeutiske midler eller 1 en kombinasjon av terapeutiske midler. De kan administreres alene, men blir vanligvis gitt sammen med en farmasøytisk bærer som velges på grunnlag av administreringsmåten og vanlig farmasøytisk praksis.
Vanligvis kan en daglig dose av aktiv bestanddel være fra 0,001 til 50 milligram pr. kg. kroppsvekt. Vanligvis vil en total mengde av 0,01 til 20 mg, fortrinnsvis 0,1 til 10 mg, pr. dag pr. kg kroppsvekt, gitt i oppdelte doser fra 2 til 4 ganger pr. dag eller i en form hvor den aktive bestanddel frigjøres på forsinket måte, gi de ønskede terapeutiske resultater.
En standard metode til å påvise og sammenligne den antidepressive virkning av de nye forbindelser,
som gir god korrelasjon med virkningen på mennesker, er den metode som går ut på å forhindre tetrabenazin-frembrakt pas-sivitet og depresjon hos mus (Eversett, "The Dopa Response Potentiation Test and Its Use in Screening for Antidepressant Drugs", sider 164 - 16 7 i Antidepressant Drugs [Proceedings of the First International Symposium], S. Garattini og M.N.G. Dukes (editors), 1967).
Grupper å 10 Carworth CF-^S hunnmus, hver av vekt 18-21 g, fikk faste i 1,5 timer og ble inkubert med antagonistforbindelser i orale doser, såsom 0, 1, 3, 9, 27 og 81 mg/kg i 0,20 ml 1 %
/r)
Methocel^ (methylcellulose). Musene ble 30 minutter etter gitt intraperitonealt 32 mg/kg tetrabenazin (i form av methan-sulfonatet) (oppløst i 0,2 0 ml 0,05 %-ig vandig KC1 ved pH 2,0). 1 time etter administreringen av antagonisten (30 minutter etter administreringen av tetrabenazin) ble musene undersøkt med henblikk på tegn til utforskningsaktivitet og ptosis (nedsynkning av øyelokket). Normal utforskningsaktivitet (opphør av pasiviteten) ble registrert når en mus, som ble løftet ut etter halen fra en gruppe på 10 mus i en forsøkskasse og anbragt på et forsøkskasselokk av rustfritt stål (31,8 cm x 20,3 cm med maskevidde 8,4 mm), enten snudde hodet horisontalt 30° i begge retninger eller beveget seg til enden av sikten innen 10 sekunder etter at den var blitt plassert på sikten. Opphør av ptosis ble registrert når nedsyk-ningen av øyelokket var mindre enn 5 0 % for begges øynes ved-kommende nøyaktig 2 sekunder etter at musen var blitt plassert med hodet vendende mot iakttageren. Den følgende tabell viser dataene som ble oppnådd ved testing av forbindelsene ifølge de ovenstående eksempler.
A norexia- data
Hunnmus av vekt 16 - 20 g, som har fastet natten over i 17 - 21 timer, benyttes ved denne undersøkelse. 30 minutter etter administrering av graderte orale doser av testforbindelsen, utblandet i vandig Methocel (methylcellulose av viskositet 100 cP, kvalitet MC, Dow Chemical Co.) og 2,8 % "Tween 80" og avpasset til 0,1 ml pr, 10 g kroppsvekt, overføres hver mus til en lys enkeltcelle av Lucite4* (13,3 cm x 12,7 cm x 12,7 cm) med et nettinggulv av maskevidde 0,64 cm x 0,64. Fem celler er anordnet på rekke i hver bur-enhet. Hver celle inneholder en seksjon av en svart stang av Lucite (13 cm x 1,2 cm x 1,2 cm), i hvis overside det er anordnet ti fordypninger av diameter 0,8 cm, hver inneholdende 0,05 ml 50 %-ig søt kondensert melk (Borden<1>s Eagle Brand). 30 minutter senere flyttes musene tilbake til "skoeske"-beholderne, og antall melkeporsjoner som er blitt fortært av hver enkelt mus, noteres. Fraksjoner av melkeporsjoner som er blitt fortært, blir likeledes anslått og notert. 5 mus pr. legemiddeldose kan drikke maksimalt 5 0 melkeporsjoner (2,5 ml melk). Fortæring av 9 eller færre melkeporsjoner betraktes som en indikasjon på anorexia. ED,-Q-verdier beregnes på grunnlag av de oppnådd responser.
Dataene som er oppført i Tabell 3,viser at enkelte av de nye forbindelser er anvendelige som anorektiske midler og således kan være anvendelige ved behandling av fettsyke.
Den beste av de nye forbindelser ansees å være forbindelsen ifølge Eksempel 2 anvendt som antidepressivt middel.
Som det vil fremgå av det ovenstående er de nye antidepressive og anorektiske midler anvendelige på det medi-sinske område, spesielt for behandling av sinnsforstyrrelser og fettsyke.
Eksempel 1
4- Fenyl- a, a, l- trimethyl- 4- piperldlnmethanol
15 ml av en 1,5 M oppløsning av n-butyllithium i hexan ble tilsatt under nitrogenatmosfære til en oppløsning av 4,7 g kommersielt tilgjengelig l-methyl-4-fenyl-l,2,3,6-tetrahydropyridin (II; R<1> = Me; R<2>, R<3> = H; R<4> = fenyl) i 5 0 ml tetrahydrofuran, som ble holdt ved -10° C. Den dyp-røde oppløsning ble omrørt ved -10° C i 15 minutter og deretter overført som en langsom strøm til en omrørt blanding av 15 ml aceton og 25 ml tetrahydrofuran, som ble holdt ved
-70° C. Overskudd av 10 %-ig saltsyre ble tilsatt etter om-røring ved -70° C i 5 minutter. Blandingen ble tilsatt å
anta romtemperatur og ble deretter vasket med toluen. Det vandige skikt ble gjort alkalisk med natriumhydroxyd og ekstrahert med methylenklorid. Ved fjerning av oppløsnings-midlet fra den tørrede oppløsning og kortbanedestillasjon av
o -4
residuet (badtemperatur 90 - 135 C; trykk 5 x 10 mm Hg [0,07 Pa]) ble det oppnådd 2,79 g 5-fenyl-2,6,6-trimethyl-7-oxa-2-azabicyclo-[3.2.1]-octan (III: R<1>, R<5>, R<6> = Me; R2, R3 = H, R 4 = fenyl) som en olje av renhet ca. 85 %.
En oppløsning av 1,47 g av det ovenfor erholdte produkt III i en blanding av 10 ml methanol og 2 ml eddiksyre ble kjølt med is og behandlet med 0,76 g natriumcyanoborhydrid. Blandingen ble omrørt ved romtemperatur i 2 timer, og overskuddet av borhydrid ble spaltet ved tilsetning av 8 ml konsentrert saltsyre under kjøling med is. Etter omrøring ved romtemperatur i 30 minutter ble blandingen gjort alkalisk med vandig natriumhydroxydoppløsning og ekstrahert med methylenklorid. Ved fjerning av oppløsningsmidlet fra den tørrede oppløsning og krystallisasjon av residuet fra ethylacetat ble det oppnådd 0,98 g 4-fenyl-a,a,1-trimethyl-4-piperidinmethanol, med smeltepunkt 146 - 147° C. NMR (220 MHz i CDC13): t 2,7 - 2,8 (m, 5H); 7,0 - 8,2 (m, 9H); 8,0 (s, 3H) og 8,9

Eksempel 2
4-(3<1->Trifluormethylfenyl)-a,a,l-trimethyl-4-piperidinmethanol og dens hydroklorld
En 1,5 M oppløsning av n-butyllithium i 150 ml hexan ble tilsatt under' nitrogenatmosfære og ved -70° C til en omrørt oppløsning av 50 g 3-brombe.nzotrifluorid i 300 ml tetrahydrofuran. Blandingen ble omrørt ved -70° C i 15 minutter og deretter tillatt å oppvarmes til -20° C. En oppløs-ning av 30 g friskt destillert l-methyl-4-piperidon i 50 ml tetrahydrofuran ble tilsatt langsomt, mens temperaturen ble holdt ved -20° C. Blandingen ble så omrørt ved 0° C i 30 minutter og ved romtemperatur i 3 timer. Vann og methylenklorid ble tilsatt, og det vandige skikt ble ekstrahert flere ganger med methylenklorid. Ved fjerning av oppløsnings-midlet fra de sammenslåtte og tørrede methylenkloridoppløs-ninger og kortbanedestillasjon av residuet (badtemperatur 110 - 130° C og trykk 5 x 10~<4> mm Hg [0,07 Pa]) ble det oppnådd 41,6 g l-methyl-4-(3'-trifluormethylfenyl)-4-piperidinol (I: R<1> = Me; R<2>, R<3> = H; R<4> = m-trifluormethylfenyl) i form av et fast stoff.
En blanding av 28,1 g av dette produkt I og 100 ml trifluoreddiksyre ble kokt med tilbakeløpskjøling i 24 timer. Overskuddet av syren ble fjernet i vakuum, residuet ble opp-løst i methylenklorid og oppløsningen ble gjort basisk med 10 %-ig vandig natriumcarbonatoppløsning. Fjerning av opp-løsningsmidlet fra den tørrede organiske fase og kortbane-destillas jon av residuet ga 24,06 g l-methyl-4-(3'-trifluor-1 2 methylfenyl)-l,2,3,6-tetrahydropyridin (Ila: R = Me; R ,
R<3> = H; R<4> = m-trifluormethylfenyl).
Til en oppløsning av dette produkt Ila i 150 ml tetrahydrofuran ble det ved -10° C tilsatt 80 ml av en 1,5 M oppløsning av n-butyllithium i hexan. Den røde oppløsning ble omrørt ved -10° C i 15 minutter og deretter overført i en langsom strøm til en omrørt blanding av 60 ml aceton og 60 ml tetrahydrofuran, som ble holdt ved -20° C. Blandingen ble tilsatt å oppvarmes til 10° C og ble deretter behandlet med 10%-ig vandig natriumkloridoppløsning. Skiktene ble skilt fra hverandre, og den vandige fase ble ekstrahert flere ganger med methylenklorid. Fjerning av oppløsningsmidlet fra de sammenslåtte, tørrede organiske faser ga 34,35 g urent 5-(3'-trifluormethylfenyl)-2,6,6-trimethyl-7-oxa-2-azabicyclo-[3.2.1]-octan (III: R<1>, R<5>, R<6> = Me; R<2>, R<3> = H; R<4> = m-trifluormethylfenyl). Dette produkt III ble oppløst i en blanding av 150 ml methanol og 2 5 ml eddiksyre, og oppløsningen ble, under kjøling med is, behandlet med 9 g natriumcyanoborhydrid. 50 ml konsentrert saltsyre ble tilsatt etter om-røring ved romtemperatur i 2 timer, og etter ytterligere 30 minutter ble blandingen gjort basisk med vandig natrium-hydroxydoppløsning og ekstrahert med methylenklorid. Fjerning av oppløsningsmidlet fra de tørrede organiske faser ga 31,74 g produkt, som ble fordelt mellom fortynnet saltsyre og ether/toluen. Etter fornyet alkalinisering av den vandige fase og påfølgende ekstraksjon med methylenklorid ble det oppnådd 27,40 g produkt. Kortbanedestillasjon (til badtemperatur 210° C, 10 mm Hg [0,1 Pa]) og krystallisasjon av destillatet fra cyclohexan ga 7,59 g 4-(3<1->trifluormethylfenyl)-2,2,l-trimethyl-4-piperidinmethanol, sm.p. 111 -

Eksempel 3
1,[l'-methyl-4'-(2'',3'<1->dimethoxyfenyl)-4'-piperidinyl]-cyclopentanol
En oppløsning av 138 g veratrol i 400 ml tørt tetrahydrofuran ble kjølt i et bad av tørr is og aceton og behandlet med 500 ml av en 1,6 M oppløsning av n-butyl-lithium med en slik hastighet at reaksjonsblandingens temperatur holdt seg ved 0° C. Etter fullført tilsetning ble reaksjonsblandingen omrørt ved romt4emperatur i 3 timer. Deretter ble den resulterende hvite oppslemning kjølt i et bad av tørris og aceton, og det ble tilsatt 90,4 g l-methyl-4-piperidon med en slik hastighet at reaksjonsblandingens temperatur holdt seg under -10° C. Etter fullført tilsetning ble reaksjonsblandingen omrørt ved 0° C i 2 timer og bråkjølt med 400 ml vann og fortynnet med 400 ml ether.
Det organiske skikt ble fraskilt, og det vandige skikt ble ekstrahert med 3 x 400 ml methylenklorid. De sammenslåtte, organiske skikt ble tørket over K-jCO^ og konsentrert under redusert trykk, hvorved det ble oppnådd 270,9 g av en olje. Denne olje ble oppvarmet under redusert trykk for å fjerne alt materiale med kokepunkt lavere enn 150°/0,025 mm Hg (3,3 Pa). Det således erholdte produkt ble destillert under redusert trykk, hvorved man fikk 73,56 g (84,8 % utbytte) 4-(2<1>,3<*->dimethoxyfenyl)-1-methyl-l,2,3,6-tetrahydropyridin (Ila: R<1> = Me; R<2>, R<3> = H; R<4> = 2,3-dimethoxyfenyl ) i form av en farveløs olje med kokepunkt 100 - 120°C/C,05 mmi;Hg (7 Pa).
En oppløsning av 4,7 g av dette produkt Ila i 50 ml tetrahydrofuran ble behandlet med n-butyllithium som beskrevet i Eksempel 1, og oppløsningen av lithiumsaltet ble tilsatt til en blanding av 20 ml cyclopentanon og 30 ml tetra-hydrof uran ved -70° C. Isolering som beskrevet i Eksempel 1 og krystallisasjon av det urene produkt fra ethylacetat ga 3,2 9 g 2'-methyl-5'-(2'',3''-dimethoxyfenyl)-spiro-[cyclo-pentan-1,6'-[7]-oxa-[2]-azabicyclo-[3.2.1]-octan] (III: R1 = Me; R2, R<3> = H; R<4> = 2,3-dimethoxyfenyl; R<5>, R<6> =

4-( 3'- methoxyfenyl)- g, a, l, 3- tetramethyl- 4- piperidinmethanol
4-(3'-methoxyfenyl)-a,a,1,3-tetramethyl-4-piperidinmethanol ble fremstilt ut fra 3-bromanisol etter fremgangsmåten beskrevet i Eksempel 2, bortsett fra at 1,3-dimethyl-4-piperidon ble benyttet isteden for l-methyl-4-piperidon ved fremstillingen av II. Produktet ble erholdt i form av en olje, som destillerte ved en badtemperatur på 130 - 160° C under et vakuum på 5 x 10 -4 Torr (0,07 Pa). Det var en blanding (mengdeforhold 83/17) av to isomere (R 3i cis- eller trans-stilling i forhold til arylgruppen), hvilket ga seg til kjenne ved 360 MHz ved NMR-spektroskopi. Isomeren som forekommer i størst mengde, har methylgruppen C-3 som en dublett (J = 7,2 Hz) ved t 8,5 og de to a-methylgrupper ved 8,7 og 8,9. Molekylvekt beregnet for C17H27N02: m/z 277,204.

gangsmåten beskrevet i Eksempel 2, bortsett fra at (-)-3-methylcyclopentanon ble benyttet ved fremstillingen av III.
Sm.p. 103 - 104° C. [a] -6,7° (c=l,07, CHCI3). NMR-spek-
trene var identiske med spektrene for produktet ifølge Eksempel 46.
Claims (2)
1. Analogifremgangsmate ved fremstilling av terapeutisk virksomme 4-aryl-4-piperidincarbinoler med formelen
hvor a) R^" er H, alkyl med 1-4 carbonatomer eller benzyl, 2 3 b) hver av R og R uavhengig av hverandre er valgt blant H og lavere alkyl med 1-4 carbonatomer, idet 1 2
imidlertid R og R sammen kan være en uforgrenet alkylenbro med 3 eller 4 carbonatomer, 4 c) R er 1) fenyl eller 2-nafthyl, hvilke kan være substituert med én eller to substituenter som kan være like eller forskjellige, og som er valgt fra F, Cl, alkyl, perfluoralkyl, alkoxy, benzyloxy,
alkylthio og dialkylamino, idet alkyl- og alkoxy-gruppene har fra 1 til 4 carbonatomer, 2) 2-, 3- eller 4-bifenylyl, 3) 2-pyrrolyl eller 2-pyrrolyl substituert med fra én til tre lavere alkylgrupper med 1-4 carbonatomer , 4) 2-, 3- eller 4-pyridyl, eller 5) 2-thienyl som i 5-stillingen er substituert med lavere alkyl med 1-4 carbonatomer, d) hver av R og R^ uavhengig av hverandre er valgt blant alkyl med 1-4 carbonatomer, eller R 5 og R 6 sarnnen danner en forgrenet eller uforgrenet alkylenbro med fra 3 til 11 carbonatomer,
forutsatt at når R^", R^ og R^ er methyl og R ' og R er hydrogen, kan R 4 ikke være p-t-butylfenyl eller 2'-bifenylyl, samt deres estere med alifatiske mono- og dicarboxlsyrer med 1-8 carbonatomer og deres aminsalter med i farmasøytisk henseende forlikelige uorganiske syrer, karakterisert ved at et oxaazabicyclooctan med den generelle formel:
hvor R<1> - R<6> har de ovenfor angitte betydninger, reduseres, og at den erholdte piperidincarbinol, om ønkes, overføres til en ester med en alifatisk mono- eller dicarboxylsyre med 1-8 carbonatomer eller til et aminsalt med en i farmasøy-tisk henseende forlikelig uorganisk syre.
2. Analogifremgangsmåte ifølge krav 1, for fremstilling av 4-(3<1->trifluormethylfenyl)-a,a,l-trimethyl-4-piperidinmethanol og dens hydroklorid,
karakterisert ved at de tilsvarende utgangs-materialér benyttes.
