Den foreliggende opfindelse angår en særlig fremgangsmåde til adskillelse af olie og/eller phosphatidylethanolamin fra al kohol opløselige phosphatidylcholinprodukter indeholdende disse under udvinding af meget rent phosphatidylcholin ved kromatografi på silicagel.
5 De ved spiseoliefremstilling fremkomne vegetabilske råphosphatider indeholder udover phosphatidylcholin, phosphatidylethanolamin og mono-, di- og triglycerider (olie) såvel phophatidylinosit som andre phosphor-holdige glycerinestre og stoffer som peptider, aminosyrer, steroler, sterol estre, fri fedtsyrer og carbohydratderivater. Til anvendelse i 10 medicinalindustrien er der især behov for oliefrit og phosphatidyl-ethanol ami nfrit phosphatidylcholin med høj renhedsgrad.
Til adskillelse af de i råphosphatidet indeholdte ledsagestoffer er der allerede beskrevet flere fremgangsmåder. Som regel foretages der i et første trin en fjernelse af olie fra det vegetabilske råphosphatid 15 i acetone (DE-AS 1 053 299, US-PS 3 268 335, DE-AS 1 047 597), hvorefter der ekstraheres med ethanol i det andet trin (US-PS 2 724 649). I det tredje trin underkastes den ethanolopløselige phosphatidfraktion søjlekromatografi ved temperaturer, der ikke overstiger 35°C (US-PS 3 031 478). Ved oliefjernelsen med acetone dannes der acetonebiprodukter som 20 mesityloxid, di acetoneal kohol, phoron o.a. i små mængder. Fjernelsen af disse toxiske og p.g.a. deres karakteristiske lugt forstyrrende stoffer er meget kostbar og til dels endda umulig. Desuden er oliefjernelsen med acetone forbundet med en stigning i peroxiderne, hvis uheldige fysiologiske indflydelse er velkendt.
25 Den alkoholiske ekstraktion af vegetabilske råphosphatider fører til en olieholdig phosphatidfraktion (DE-OS 2 718 797, DE-PS 1 492 952, DE-AS 1 692 568), ud fra hvilken der efter kromatografisk behandling ved stuetemperatur kan udvindes et olieholdigt, phosphatidylethanolaminfrit phosphatidylcholin. Ifølge en af vore egne patentansøgninger (P 30 23 30 814.3) lykkes det ved tilsætning af en lille mængde vand at fraskille olien og udvinde et stærkt renset, oliefrit phosphatidylcholin. I alt udgør denne oliefjernelse med vand, der er velegnet til fremstilling af bestemte phosphatidfraktioner, imidlertid også en tretrinsproces (ekstraktion, kromatografi, oliefjernelse) ved udvinding af oliefrit phos-35 phatidylcholin, hvilken desuden er behæftet med oparbejdningen af vandige ethanol opløsninger.
Udvindingen af rent phosphatidylcholin forudsætter en kromatografisk adskillelse af de i den ethanolopløselige phosphatidfraktion

2 indeholdte ledsagestoffer. Som adsorptionsmiddel anvendes der hovedsagelig aluminiumoxid. Rensning sker både søjlekromatografisk og ved omrøring med skillemidlet, til enhver tid ved stuetemperatur.
I teknisk målestok anvendes basisk aluminiumoxid. Også her gennem-5 føres kromatografien altid ved stuetemperatur. Da aluminiumoxidet efter kromatografien er belagt med forureninger, navnlig med phosphatidyl -ethanolamin, må det kasseres. En yderligere ulempe består i dannelsen af lysophosphatidylcholin under kromatografien (0. Renkonen, J. Lipid. Res.
3, 181-3 (1962), D. Van Damme et al., Int. Symp. Chromatogr. Elektro-10 phoresis, 5th, 1968 (Pub. 1969), 268-78). Kromatografien over silicagel er hidtil kun tillagt analytisk og præparativ betydning. Således ad-sorberes phosphatider i hexanopløsninger ikke af silicagel (JA 77012202, US-PS 3 869 482), mens de i alkoholiske opløsninger tilbageholdes af silicagel (H. Richter et al., Pharmazie, 1977, 32 (3), 164). De for-15 skellige forhold kan forklares ved dannelsen af lipofile, polymolekylære phosphatidmiceller i hexan i modsætning til de molekylært foreliggende phosphatider i alkohol. Kromatografien gennemføres også her ved stuetemperatur. Adskillelsen og den selektive desorption af phosphatiderne sker ved anvendelse af alkoholiske phosphatidopløsninger enten med 20 al kohol/ammoni ak (JA 49093400) eller med blandinger af chloroform/ methanol (C.H. Lea et al., Biochem. J. 60, 353-63 (1965)) hhv. chloro-form/methanol/vand (DDR-PS 79 916). Denne fremgangsmåde har ikke hidtil haft nogen teknisk betydning, da der som elueringsmiddel altid er blevet anvendt blandinger af toksiske materialer, såsom ammoniak 25 eller chloroform, hvis genvinding og fjernelse fra lecithinet er meget bekostelig.
Sædvanlige, i handelen værende phosphatidylcholi nråprodukter, navnlig af sojabønnephosphatider, er produkter, der er udvundet ved ekstraktion med alkohol og de er al kohol opløselige og indeholder olie og 30 phosphatidylethanolamin som hovedforurening. Der findes imidlertid også phosphatidylcholi nråprodukter, der som beskrevet kun indeholder den ene eller den anden af de to hovedforureninger. Den foreliggende opfindelse er baseret på den opgave at udvikle en fremgangsmåde til adskillelse af olie og/eller phosphatidylethanolamin fra al kohol opløselige phosphat-35 idylcholinprodukter indeholdende disse under fremstilling af meget rent phosphatidylcholin, som er befriet for disse forureninger. Til alle de hidtil beskrevne fremgangsmåder er knyttet den ulempe, at de til fremstilling af et rent, olie- og phosphatidyl ethanol aminfrit produkt kræver

3 omstændelige fremgangsmådetrin, ved hvilke der må regnes med dannelse eller forbi i ven af toxiske produkter. Det ved kromatografisk rensning anvendte aluminiumoxid har tilmed måttet kasseres.
Det har nu overraskende vist sig, at man ud fra al kohol opløselige, 5 olie- og/eller phosphatidylethanolaminholdige phosphatidfraktioner opnår et olie- og phosphatidyl ethanolaminfrit phosphatidylcholin med høj renhedsgrad ved hjælp af en sil icagelsøjlekromatografi ved højere temperaturer.
Fremgangsmåden ifølge opfindelsen til adskillelse af olie og/eller 10 phosphatidylethanolamin fra al kohol opløselige phosphatidylcholinprodukter indeholdende disse under fremstilling af olie- og phosphatidyl-ethanol aminfrit phosphatidylcholin er ejendommelig ved, at man ved en temperatur i området fra 60-90eC kommer opløsningen af det olie og/eller phosphatidylethanolaminholdige, al kohol opløselige phosphatidylcholin-15 produkt i en Cj^ lavere alkanol, eventuelt blandet med indtil 20 vol-% vand, eller en sådan blanding af flere Cj_4 lavere alkanoler, på en søjle af silicagel, eluerer søjlen ved denne temperatur med en Cj_4 lavere alkanol, eventuelt indeholdende indtil 20 vol-% vand, eller en sådan blanding af flere Cj_4 lavere alkanoler, og derved opsamler et 20 forløb indeholdende olien og/eller phosphatidyl ethanol aminen, som skal fraskilles, og adskilt herfra restløbet, som indeholder det rene phosphatidyl chol in, og adskiller opløsningsmidlet fra restløbet på sædvanlig måde.
Tilsætningen til si licagelsøjlen og elueringen af denne gennem-25 føres fortrinsvis ved en temperatur i området fra 60 til 70°C. Som elueringsmiddel foretrækkes det at anvende det benyttede opløsningsmiddel, hvilket gør fremgangsmåden særlig enkel. Som Cj_4 lavere alkanol foretrækkes ethanol.
Mængden af forløbet afhænger af det anvendte phosphatidylcholin-30 produkt. Ved simple analytiske metoder lader det sig fastslå, fra hvilket tidspunkt eluatet er fri for ledsageprodukterne, som skal fra-skilles og praktisk taget kun indeholder phosphatidylcholin. Erfaringsmæssigt udgør forløbet ca. 20 til 25% af eluatets samlede volumen. I forløbet befinder der sig alt efter det anvendte phosphatidylcholin-35 produkt også de andre, sædvanligvis tilstedeværende ledsagestoffer som sterol er, sterolderi vater, glycol ipider og phospholipider, som kan bringes til en videre anvendelse.

4 ter af forskellig kornstørrelse eller også presset silicagel, som kan være aktiveret eller desaktiveret. Der anvendes fortrinsvis neutrale silicagelprodukter.
Fremgangsmåden ifølge opfindelsen kan gennemføres ved normalt tryk 5 eller også ved overtryk. En ganske særlig fordel ved fremgangsmåden ifølge opfindelsen består i, at man stadig kan genanvende silicagelen efter udført kromatografi. Samtlige forureninger opfanges i forløbet og efter hovedløbet bliver der kun en ringe restbelægning af phosphatidyl -cholin tilbage.
10 En yderligere fordel ved silicagelen ligger i den meget høje be lastning, dette sorptionsmiddel kan udsættes for. Således adskilles der ca. 60 dele faststof fra den al kohol opløselige phosphatidfraktion ved en søjlekromatografisk adskillelse med 100 dele silicagel.
Som phosphatidylcholinprodukter kan der anvendes sådanne, som fås 15 ved ekstraktion med alkohol fra fx. soja, jordnød, solsikke eller raps. Phosphatidet opløses med en laveremolekylær alifatisk alkohol, såsom fx. methanol, ethanol, n-propanol eller isopropanol, men især med 94 til 96 vol-%ig ethanol. Opløsningen befris for bundfald, og den klare alkoholfase overføres således eller i form af et koncentrat til fremgangsmåden 20 ifølge opfindelsen. Det ved ekstraktionen anvendte opløsningsmiddel kan fjernes helt, og det opnåede produkt kan igen opløses i en eller flere Cj_4 lavere al kano!er, eventuelt indeholdende 1-20 vol-% vand. Udvindingen af rent phosphatidylcholin fra såvel olie- som phosphatidyl-ethanol ami nhol dige phosphatidylcholinråprodukter forenkles i forhold til 25 de kendte fremgangsmåder, specielt ved sammenlægningen af oliefjernelses- og rensningsfremgangsmådetrinnene. En yderligere fordel, der også opnås ved anvendelse af phosphatidylcholinprodukter, fra hvilke et af de to følgestoffer allerede er adskilt på anden måde, opnås ved genanvendelsen af silicagelen.

Phosphatiderne analyseredes ved hjælp af tyndtlagskromatografi.
35 Olieindholdet blev sat lig med det dialyserbare. Vandet blev bestemt efter Karl Fischer og restethanolindholdet ved gaskromatografi.

5 Sø.i le kromatografi
Der anvendtes en doppeltkappekromatografisøjle (indvendig diameter 4,5 cm, højde 37 cm). Før søjlen var der tilsluttet en varmeveksler for at sikre en ensartet søjle- og tilsætningstemperatur. Søjlen fyldtes med 5 en opslæmning af 200 g silicagel (Merck) i det til enhver tid angivne opløsningsmiddel. Efter udført kromatografi kan silicagelen anvendes igen.
Udgangsmateriale 10 Sojaråphosphatid ekstraheres ved 35°C med 95 vol-%ig ethanol i vægtforholdet 1:2,5 (råphosphatid:ethanol). Bundfaldet adskilles fra den overstående ethanol fase ved stuetemperatur. Ethanol fasen inddampes.
Det derved opnåede faste stof har følgende sammensætning: 15 Phosphatidylcholin (PC) 43%
Phosphatidyl ethanolamin (PE) 12%
Olie 21%
Dette faste stof anvendtes til eksemplerne 1-4.

Sojaråphosphatid befris for olie med acetone og ekstraheres derefter med 95 vol-%ig ethanol. Den ethanol opløselige phosphatidfraktion inddampes og analyseres: 25 PC-indhold 52% PE-indhold 20%
Det i eksempel 6 anvendte olieholdige phosphatidyl ethanolaminfri phosphatidylcholin fremstilledes ifølge DE-OS 2 718 797 (eksempel 3).

Claims (3)
1. Fremgangsmåde til adskillelse af olie og/eller phosphatidyl - ethanolamin fra al kohol opløselige phosphatidylcholinprodukter indeholdende disse under fremstilling af olie- og phosphatidyl ethanol aminfrit phosphatidylcholin, KENDETEGNET ved, at man ved en temperatur i området fra 60-90°C kommer opløsningen af det olie- og/eller phosphatidyl-30 ethanol aminholdige, al kohol opløselige phosphatidylcholinprodukt i en Cj_4 lavere alkanol, eventuelt blandet med indtil 20 vol-% vand, eller en sådan blanding af flere C14 lavere alkanoler, på en søjle af si 1 i - cagel, eluerer søjlen ved denne temperatur med en Cj_4 lavere alkanol, eventuelt indeholdende indtil 20 vol-% vand, eller en sådan blanding af 35 flere Cj_^ lavere alkanoler, og derved opsamler et forløb indeholdende olien og/eller phosphatidyl ethanol aminen, som skal fraskilles, og adskilt herfra restløbet, som indeholder det rene phosphatidylcholin, og adskiller opløsningsmidlet fra restløbet på sædvanlig måde.
