Den foreliggende opfindelse angår hidtil ukendte kemiske mellemprodukter, der er nyttige til fremstilling af terapeutisk aktive forbindelser.
5 Det er kendt fra Journal of Medicinal Chemistry, 1973, bind 16, side 168-169, at optisk aktive 1-aryloxy-2,3-epoxypropan-derivater kan fremstilles og kan bringes til at reagere med en amin, såsom isopropy1 am in til dannelse af et optisk aktivt 1-aryloxy-3-amino-2-propanolderivat. Det således beskrevne epo-10 xyderivat er imidlertid en venstredrejende forbindelse, som har en (R)-absolut konfiguration og 1-aryloxy-3-amino-2-pro-panolderivatet fremkommet deraf er en højredrejende forbindelse, der også har (R)-absolut konfigurationen. Det er velkendt, at mange 1-ary 1oxy-3-am i no-2-propano1 der ivater har værdifuld 15 β-adrenergi sk blokerende virkning og derfor er nyttige til behandling af b.a. hjertesygdomme, og det er endvidere kendt, at hvis en sådan racemisk forbindelse opspaltes i sine optisk aktive enantiomorfe, er den /5-adrenergisk blokerende virkning i reglen overvejende i den venstredrejende isomer, som har (S)-20 absolut konfiguration.
Den foreliggende opfindelse angår l-aryloxy-2-propanolderiva-ter med (S)-absolut konfiguration, der kan anvendes som mellemprodukter til fremstilling af 1-aryloxy-3-amino-2-propanol-25 derivater, som har j3-adrenerg i sk blokerende virkning.

Sådanne udgangsforbindelser kan anvendes til fremstilling af 1-aryloxy- 3-amino-2-propanolderivater med (R)-absolut konfiguration.
10 Selv om de tilsvarende racemiske l-aryloxy~3-amino-2-propanol-derivater har værdifulde medicinske egenskaber som /3-adrener-giske blokerende midler, ligger denne aktivitet overvejende hos (S)-isomeren og er til stede i kun ganske ringe grad hos (R)-isomeren.
15 (R) ~giycerinacetonid er meget let til gængel i g udfra D-mannit, men den tilsvarende (S)-isomer er ikke tilnærmelsesvis så let tilgængelig. L-mannit, som ville give forbindelsen, er meget sjældnere og meget dyrere end D-mannit, og andre fremgangsmå- 20 der til fremstilling af (S)-glycerinacetonid er tilsvarende dyre. Den foreliggende opfindelse er baseret på anvendelse af det let tilgængelige (R)-glycerinacetonid til fremstilling af (S) -1-ary1oxy-3-amino-2-propanolderi vater, og nøglemellemprodukterne ved fremgangsmåden er de, der er beskrevet for den 25 foreliggende opfindelse, nemlig (S)-l-aryloxy-3-"Y-0"-2-propa-nolderivaterne. Eksempel 1.
4,5 g p-carbamoylmethylphenol sættes til en opløsning af 0,7 g natri= um i 15 ml methoxyethanol, og blandingen omrøres i 15 minutter. En 5 opløsning af 10 g (+)-3-benzyloxy-l-tolueη-p-sulphonyloxypropan-2-ol i 25 ml methoxyethanol tilsættes, og blandingen opvarmes under tilbagesvaling i 1½ time, afkøles og hældes i iskoldt vand. Blandingen filtreres, og den faste rest krystalliseres af ethylacetat. Derved fås 5 g (+)-3-benzyloxy-l-p-carbamoylmethylphenoxypropan-2-ol, smp.

Eksempel 2.
En opløsning af 5,5 g (+)-3-benzyloxy-l-p-carbamoylmethylphenoxypropan-2-ol i 150 ml methanol rystes i en atmosfære af hydrogen ved atmosfæretryk i nærværelse af en 30% palladium-på-trækul katalysator, indtil den nødvendige mængde hydrogen er blevet absorberet. Blandingen filtreres og filtratet inddampes til tørhed. Den faste rest krystalliseres af methanol og derved fås (+)-l-carbamoylmethylphenoxypropan-2,3- 2o diol, smp. 182,5 - 184°C, /«7^ + 7,4° (c = 0,5% i methanol).
Eksempel 5.
2,8 g toluen-p-sulfonylchlorid sættes til en afkølet opløsning af 25 3,3 g (+)-l-p-carbamoylmethylphenoxypropan-2,3-diol i 33 ml pyridin, og blandingen får lov at henstå i 20 timer ved +4°C. Blandingen fortyndes med 25 ml ethylacetat og hældes i iskold vandig svovlsyre (12,8 ml koncentreret svovlsyre i 78 ml vand). Det organiske lag fraskilles, og det sure, vandige lag ekstraheres tre gange med ethylace= tat (25 ml hver gang). De forenede ethylacetatopløsninger tørres over 30 vandfri magniumsulfat og inddampes til tørhed, og den tilbageblevne olie tritureres med ether. Det derved fremkomne faste stof krystalliseres af ethylacetat, og derved fås 1,35 g (-)-1-p-carbamoylmethyl= phenoxy-3-toluen-p-sulfonyloxypropan-2-ol, /57q - 7,4° (c = 0,5% i methanol).

En opløsning af 1,25 g (-)-l-p-carbamoylmethylphenoxy-3-toluen-p-sulfonyloxypropan-2-ol i 20% vandig natriumhydroxidopløsning omrøres ved stuetemperatur i 15 minutter. En blanding af 0,5 g (+)-l-p-carbamoylmethylphenoxy-2,3-epoxypropan 5 ml isopropylamin og 5 ml isopropanol opvarmes under tilbagesvaling i li time og inddampes så til tørhed. Remanensen krystalliseres af ethylacetat og derved fås (-)-1-p-carbamoylmethylphenoxy-3-isopropyl= 15 aminopropan-2-ol, smp. 151,5-153°C, - 13,6° (c = 1% i vandig N-saltsyre).

Eksempel 11.
25 5,95 g o-cyanophenol sættes til en opløsning af 1,15 g natrium i 20 ml 2-methoxyethanol, blandingen omrøres i 5 minutter, og der tilsættes en opløsning af 15,1 g 3-t-butoxy-l-tolueh-p-sulfonyloxypropan- 2-ol i 30 ml 2-methoxyethanol. Blandingen opvarmes under tilbagesva-30 ling i 90 minutter, afkøles og hældes i en blanding af is og vand (150 ml), og blandingen gøres basisk med vandig 2N-natriumhydroxid-opløsning og ekstraheres to gange med chloroform (100 ml hver gang).
De forenede ekstrakter vaskes med vand, tørres og inddampes til tørhed og remanensen kromatograferes på silicagel under anvendelse af chloro-35 form som eluant. De ønskede fraktioner af eluatet inddampes til tørhed og derved fås som olieagtig remanens l-o-cyanophenoxy-3-t-butoxy= propan-2-ol, hvis struktur bekræftes ved protonmagnetisk resonans-spektroskopi.

Den som udgangsmateriale anvendte 3-t-butoxy-l-toluen-p-sulfonyloxy= propan-2-ol kan fås på følgende måde:
Isobutylen bobles i 3 timer ind i en opløsning af 12,0 g 2,3-o-iso= 5 propylidenglycerin i 175 ml methylenchlorid indeholdende 1 ml svovlsyre og blandingen får lov at henstå i 18 timer og vaskes så 3 gange med 5% vandig natriumbicarbonatopløsning (100 ml hver gang). Methylen= chloridopløsningen tørres og inddampes til tørhed og remanensen destilleres under reduceret tryk. Derved fås l-0-t-butyl-2,3-0-isopro= 10 pylidenglycerih, kogepunkt 110-ll6°C/50 mm.
En blanding af 22,5 g af ovenstående forbindelse og 100 ml 5% vandig svovlsyre omrøres ved stuetemperatur i 30 minutter, der tilsættes fast natriumcarbonat, indtil pH-værdien af opløsningen er 10, og blandingen ekstraheres to gange med chloroform (100 ml hver gang).

De forenede ekstrakter vaskes med vand, tørres og inddampes til tørhed, og·remanensen destilleres under reduceret tryk. Derved fås 1-0-t-butylglycerin, kogepunkt 84-85°C/l,4 mm.
14,4 g toluen-p-sulfonylchlorid sættes til en omrørt opløsning af 20 11,2 g af ovenstående forbindelse i 170 ml pyridin, som holdes ved -15°C, og blandingen omrøres ved nævnte temperatur, indtil alt chlo- ridet er opløst^og får så lov at opvarmes til 0°C i løbet af 18 timer.
Blandingen filtreres for at fjerne pyridinhydrochloridjog filtratet fortyndes med 100 ml ethylacetat og hældes i en afkølet blanding af 25 66,2 ml svovlsyre og 380 ml vand. Ethylacetatlaget fraskilles, og det vandige sure lag ekstraheres tre gange med ethylacetat (100 ml hver gang). De forenede ethylacetatopløsninger vaskes med saltvand, tørres og inddampes til tørhed, og remanensen opløses i chloroform og kromatograferes på en silicagelsøjle under anvendelse af chloro- 30 form i begyndelsen og derefter stigende koncentrationer af ethylacetat i chloroform som eluant. Eluatet fremkommet ved anvendelse af en 30% v/v opløsning af ethylacetat i chloroform inddampes til tørhed og derved fås som olieagtig remanens 3-t-butoxy-l-toluen-p-sulfonyl= oxypropan-2-ol, hvis struktur bekræftes ved protonmagnetisk resonans- spektroskopi.
