Oxazolidinonmethylhalogenidet med formel II' kan fremstilles ved omsætningen af det egnede isocyanat og epi-halogenhydrid ifølge US-PS nr. 4.128.654, hvortil der hen-15 vises. Det har vist sig, at forbedrede udbytter og lettelse af isolering af produktet kan opnås ved anvendelse af xylen som opløsningsmiddel og lithiumbromid-tri-n-butyl-phosphinoxid-kompleks som katalysator. Omsætningen udføres ved en temperatur i området mellem ca. 100°C og 145°C, for-20 trinsvis ca. 140-145°C.
Oxazolidinonmethylhalogenidet med formel II' kan oxideres til opnåelse af det tilsvarende sulfoxid (n=l) eller sulfon (n=2) med formel III' ved omsætning med persyrer, f.eks. m-chlorperbenzoesyre eller pereddikesyre.
25 Omsætningen udføres bedst ved en temperatur i området mellem 10°C og 60°C, fortrinsvis 20-30°C, i et opløsningsmiddel, såsom methylenchlorid eller chloroform. Produktet kan isoleres ved triturering af reaktionsblandingen med ether og frafiltrering af det faste produkt. Om nødvendigt kan on det renses yderligere ved omkrystallisation fra et egnet opløsningsmiddel.
Isocyanatet IV, der anvendes i skema A, er kommercielt tilgængeligt, når = CH^ eller C2H,-. Andre iso- cyanater kan fremstilles ved hjælp af fremgangsmåder, som er illustreret i skema B.

Derefter omsættes anilinet VII med phosgen i et egnet opløsnings-5 middel, såsom toluen eller xylen, ved en begyndelsestemperatur på ca. 40-70°C, efterfulgt af opvarmning til en temperatur på ca. 100-110°C. Produktet, isocyanat IV-a (R-^=CFg) , isoleres og renses ved destillering.
I omsætning 2 kontaktes p-acetamidothiophenol 10 VIII med et alkalimetalsalt (Na eller Li) af chlordi- fluoreddikesyre i 2-methoxyethylether som opløsningsmiddel ved en temperatur på ca. 130-165°C, fortrinsvis 155-165°C. Det resulterende difluormethyladditionspro-dukt IX omdannes til isocyanatet IV-b (R=HCF2) gennem 15 X i 2 trin, i lighed med ovennævnte fremgangsmåde.
I omsætning 3 omsættes p-acetamidothiophenol VIII med tetrafluorethylen i nærværelse af en organisk base, f.eks. diisopropylamin. Omsætningen udføres i en bombe i dimethylformamid eller dimethylsulfoxid ved en 20 temperatur i området fra ca. 0-50°C, fortrinsvis ved ca.
20-30°C. Produktet isoleres ved at hælde reaktionsblandingen i vand og ekstrahere med ether. Det resulterende tetrafluorderivat XI omdannes til IV-c (R^=-CF2-CF2H) gennem XII i 2 trin, i lighed med ovennævnte fremgangs-25 måde.

Omsætning a omfatter hydrolysen af II' til amino-diolerne XIV under basiske betingelser. Hydrolysen udføres bedst i en vand-alkohol (ethanol eller methanol) opløsning af natrium- eller kaliumhydroxid 5 ved en temperatur i området fra ca. 40-80°C, fortrinsvis ved 70-80°C. Produktet kan isoleres ved at fortynde reaktionsblandingen med vand og ekstrahere med methylen-chlorid. Den resulterende amino-diol XIV cycliseres derefter til XVI ved omsætning med diethyl- eller dimethyl-10 carbonat i ethanol eller 1,2-dimethoxyethan som opløsningsmiddel i nærværelse af en basisk katalysator, f.eks. natriumethoxid eller -methoxid. Produktet, som almindeligvis udskilles som et faststof, når reaktionsblandingen afkøles, isoleres ved filtrering.
15 Omsætning b omfatter ombytning af halogen i II' med carboxylat til opnåelse af XV. Denne omsætning udføres med kaliumacetat i dimethylformamid eller dimethyl-sulfoxid ved en temperatur i området fra ca. 80-140°C, fortrinsvis ved ca. 120-130°C. En katalysator, såsom 20 18-krone-6 kan anvendes til at lette omsætningshastig heden. Produktet isoleres ved at fortynde reaktionsblandingen med vand og ekstrahere med et egnet opløsningsmiddel, f.eks. ether. Den resulterende ester XV omdannes derefter til methanolet XVI under basiske hydrolysebetin-25 gelser. Denne omsætning udføres i en alkoholisk (methanol eller ethanol) kalium- eller natriumhdyroxidopløs-ning ved en temperatur på ca. 0-60°C, fortrinsvis ved ca. 25-30°C. Produktet, som almindeligvis udskilles som et faststof når, omsætningen er fuldstændig, isoleres 30 ved filtrering.
Fremgangsmåden til oxidering af sulfidet XVI
til det tilsvarende sulfoxid XVII (n=l) eller sulfon XVII (n=2) er den samme som for oxidationen af II til III, som er beskrevet i skema A.

80-120°C, fortrinsvis ved ca. 90-100°C. Fjernelsen af sulfonylgruppen i XXI opnås ved omsætning af XXI med naphthalen-natrium ved en temperatur på ca. 10-40°C, fortrinsvis ved 25-30°C. Egnede opløsningsmidler omfatter 35 1,2-dimethoxyethan og tetrahydrofuran.
o 10 150603 På den anden side kan XIV opnås direkte fra anilinet XIX og glycidol i en kogende methanolopløsning, som vist i omsætning b. Produktet isoleres ved at fortynde reaktionsblandingen med vand og ekstraheres med 5 methylenchlorid.
Forskellige simple estere af XVI og XVII kan fremstilles som vist i skema D ved omsætning med et egnet syrechlorid eller -anhydrid i et indifferent opløsningsmiddel, f.eks. methylenchlorid, dimethylformamid 10 eller 1,2-dimethoxyethan. Omsætningen udføres fortrins vis ved en temperatur på ca. 0-60°C, eventuelt i nærværelse af en base, f.eks. pyridin eller triethylamin. Når det omsatte.anhydrid er cyclisk, såsom ravsyreanhydrid er produktet ravsyremonoesteren XIII-a (R^-CI^-CE^-CX^H) .

Produktet XVII (n=2), som opnås ved hjælp af ovennævnte metoder, er en racemisk blanding, dvs. en 50:50-blanding af (+)- og (-)-enantiomere. Syntesen af 35 optisk aktivt XVII (R-^CH^, n=2) kan opnås gennem spalt- 150803

12 ning af mellemproduktet XIV, idet man benytter sig af dets basiske nitrogen. Almindelige spaltningssyrer, f.eks.D- eller L-mandelsyre, kan anvendes. Således omsættes XIV (R^CH^) med L-mandelsyre i en vandig ethanolopløsning. Derefter om-5 krystallis_eres det resulterende salt gentagne gange fra acetonitril, indtil der opnås et konstant smeltepunkt. Derefter gøres saltet basisk ved behandling med ammoniumhydroxid til opnåelse af det optisk aktive (+)-XIV (R^CH^) . Derefter omdannes (+)-XIV til det optisk aktive (-)-XVII 10 (R^CHg, n=2) ved hjælp af den tidligere beskrevne fremgangsmåde. (+)-XVII (R^=CH3, n=2) kan også fremstilles ved anvendelse af D-mandelsyre.
Fremstilling af udgangsmaterialer.
15 Eksempel 1 5-Chlormethyl-3-(4-methylthiophenyl)-2-oxazolidinon (II, R^CHj, X=C1) .
Til en 130°C varm opløsning af 105 g (0,64 mol) 4-methylthiophenylisocyanat og 93 g (1 mol) epichlorhy-20 drin i 600 ml xylen sættes 200 ml af et tri-n-butylphos-phinoxid-lithiumbromidkompleks. (Komplekset er fremstillet ved at opvarme en blanding af 45 g tri-n-butylphos-phinoxid, 12,7 g lithiumbromid og 550 ml xylen til tilbagesvalingstemperatur, indtil der opnås en klar opløsning 25 ved fjernelse af vand). Der sker en exotermisk omsætning.
Når hele katalysatoren er tilsat, opvarmes blandingen under tilbagesvaling i 1 time og afkøles. Opløsningen dekanteres over i en kolbe og koncentreres til halvdelen af volumen. Samme volumen petroleumsether sættes til remanen- 30 sen. Det dannede faststof opsamles ved filtrering og omkrystalliseres fra acetonitril til opnåelse af 102 g af titelforbindelsen (udbytte: 62%); smp.: 106,5-107,5°C.

Når blandingen er tilbagesvalet i 5 timer, afdampes opløsningsmidlet ved koncentration, og faststofremanensen tritureres med ether og filtreres til op-.nåelse af 2,3 g af titelforbindelseny smp.: 121-123°C.

Anvende1ighed 20 Denne type af hidtil ukendte kemiske forbindelser er aktive, in vitro, overfor mange forskellige bakterielle organismer, herunder de gramnegative enterobacteriaceae, betalactamase-producerende staphylococcer og anaerobe bakterier. 5 Kulturer af Staphylococcus aureus eller Escherichia coli dyrkes fra opbevarede, frosne lagre i trypticase--soyamedium (BBL), hvilket er et gængst bakteriologisk vækstmedium, under omrystning, indtil der opnås en uklarhed med optisk tæthed (650 yarn) på 0,4-0,5. Disse kulturer 10 centrifugeres og resuspenderes i enten 50%’s gastrisk mucin fra svin til Staphylococcus aurerus eller fysiologisk saltopløsning til Escherichia coli.
