Description
- Die vorliegende Erfindung betrifft neue Derivate von N- Cyclohexylbenzamiden, die potente Stimulantien der gastrointestinälen Motilität sind.
- Zahlreiche Orthopamide sind für verschiedene pharmakologische Eigenschaften bekannt, wie beispielsweise die neuroleptischen Derivate des Sulpirids (US-A-3,342,826) oder die Anti-Emetika und Stimulantien der gastrischen Motilität, wie Metoclopramid (BE-A- 620,543). Von dem letztgenannten Produkt ist bekannt, daß es auf dopaminergen Wegen des zentralen Nervensystems wirkt.
- Neuere Verbindungen der Familie der Benzamide, wie das Cisaprid (EP-A-076,530) oder das Renzaprid (EP-A-094,742) sind für ihre Wirkung auf die Verdauungsmotilität in Abwesenheit zentraler antidopaminerger Wirkungen bekannt.
- Im übrigen sind Verbindungen, die der allgemeinen Formel
- entsprechen, in der R eine tertiäre Aminogruppe ist, insbesondere eine Piperidinogruppe, und R' eine Alkoxygruppe in para oder meta am Benzamidring ist, für ihre antikonvulsive Eigenschaft bekannt (GB-A-1,015,921).
- Wir haben nun entdeckt, daß die Verbindungen, die durch die nachstehend definierte allgemeine Formel (I) dargestellt werden und die sich von den vorstehenden Verbindungen insbesondere dadurch unterscheiden, daß sie einen polysubstituierten aromatischen Ring und insbesondere eine Alkoxygruppe in ortho umfassen, potente Stimulantien der gastrointestinalen Motilität sind und keine zentrale antagonistische dopaminerge Aktivität aufweisen, während die beschriebenen Verbindungen, die diese Substitution nicht besitzen, eine solche Aktivität nicht aufweisen. Beispielsweise zeigt eine Verbindung A des in der GB- A-1,015,921 beschriebenen Typs, die der allgemeinen Formel I entspricht, aber mit R&sub1; = R&sub3; = R&sub4; = H und Rz = 4-Pentyloxy, bei den höchsten Dosen, die verwendet wurden, um die erfindungsgemäßen Verbindungen zu testen, keinerlei Wirkung auf die intestinale Motilität.
- Gegenstand der vorliegenden Erfindung sind demgemäß neue Benzamide, die der allgemeinen Formel (I) entsprechen
- in der:
- R&sub1; aus (C&sub1;-C&sub4;)-Alkoxy-, Methoxy-(C&sub1;-C&sub3;)-alkoxy-, (C&sub3;-C&sub4;)- Alkenyloxy- und (C&sub5;-C&sub6;)-cycloalkoxygruppen ausgewählt ist
- oder R&sub1; und R&sub2; in Position 3 zusammen mit dem aromatischen Ring, an dem sie angebracht sind, einen 2,3-Dihydrobenzofuranzyklus bilden,
- R&sub2;, R&sub3;, R&sub4; unabhängig voneinander ausgewählt sind aus einem Wasserstoffatom, einem Halogenatom und Hydroxy-, (C&sub1;-C&sub3;)-Alkoxy-, Amino-, (C&sub1;-C&sub3;)-Alkylamino-, Di-(C&sub1;-C&sub3;)-alkylamino- und (C&sub1;-C&sub3;)- Alkylcarbonylaminogruppen,
- Z ausgewählt ist aus der Piperidinylgruppe und einer Piperidinylgruppe, die in 4 durch eine Hydroxy-, (C&sub1;-C&sub3;)-Alkoxy-, Hydroxyethyl-, (C&sub1;-C&sub3;)-Alkoxyethyl-, (C&sub1;-C&sub3;)-Alkoxymethylgruppe, eine (C&sub1;-C&sub4;)-Alkylgruppe oder zwei (C&sub1;-C&sub3;)-Alkylgruppen substituiert ist,
- ihre N-Oxide, insbesondere diejenigen, in denen die N-oxidgruppe durch Z getragen wird,
- und ihre Additionssalze mit pharmazeutisch annehmbaren Säuren.
- Die vorliegende Erfindung umfaßt die Stereoisomeren der relativen cis-Konfiguration und der relativen trans-Konfiguration, die durch die Substituenten des Cyclohexylringes gebildet werden, sowie die entsprechenden Enantiomeren.
- In der vorliegenden Erfindung bezeichnet man mit Alkyl- oder Alkoxygruppe gleichermaßen die linearen, verzweigten Gruppen oder diejenigen, die einen cyclischen Teil aufweisen.
- In der vorliegenden Erfindung bezeichnet man mit "pharmazeutisch annehmbare Salze" die Additionssalze mit Säuren, die die biologischen Eigenschaften der Verbindungen ergeben, ohne eine unerwünschte Wirkung aufzuweisen. Bei diesen Salzen kann es sich insbesondere um diejenigen handeln, die mit Mineralsäuren, wie Chlorwasserstoffsäure, Bromwasserstoffsäure, Schwefelsäure, Salpetersäure, Phosphorsäure, gebildet werden; die metallischen sauren Salze, wie Dinatriumorthophosphat und Monokaliumsulfat, und diejenigen von organischen Säuren, wie Ameisensäure, Essigsäure, Propionsäure, Glycolsäure, Oxalsäure, Fumarsäure, Milchsäure, Bernsteinsäure, Weinsäure, Äpfelsäure und Pamoasäure.
- Die bevorzugten Verbindungen der Formel I sind diejenigen, die am aromatischen Ring einen Alkoxy-Substituenten in 2, insbesondere eine Methoxy-, Ethoxy- oder Cyclopropylmethoxygruppe, einen Amino-Substituenten in 4 und Chlor in 5 aufweisen.
- Unter diesen Benzamiden bevorzugt man spezieller die Verbindungen der Formel (I), in der Z eine Piperidinogruppe ist, die vorzugsweise in 4 durch eine oder zwei Methylgruppen oder durch eine Hydroxygruppe substituiert ist.
- Die bevorzugten Verbindungen weisen demgemäß die Struktur (IV) oder (V) auf.
- worin R&sub7; = CH&sub3;, R&sub1;&sub0; = H oder R&sub7; = CH&sub3; = R&sub1;&sub0; oder R&sub7; = OH, R&sub1;&sub0; = H und R&sub1; = OCH&sub3;, OC&sub2;H&sub5; oder Cyclopropylmethoxy.
- Die Verbindungen der Formel (I) können durch Kondensation zwischen einer Benzoesäure der Formel (VI), worin R&sub1;, R&sub2;, R&sub3;, R&sub4; die gleichen Bedeutungen wie vorstehend aufweisen, und einem Cyclohexylamin der Formel (VII) der cis- oder trans- Konfiguration, wobei Z die gleichen Definitionen wie in der Formel (I) aufweist, gemäß dem Schema
- erhalten werden.
- Bei dem Kupplungsmittel kann es sich um Carbonyldiimidazol oder um Dicyclohexylcarbodiimid handeln, das in Lösungsmitteln wie Methylenchlorid oder Tetrahydrofuran oder auch Pyridin verwendet wird.
- In dem Fall, in dem die Benzoesäure der Formel (VI) keine basische Amino- oder Alkylaminofunktion trägt, kann sie in Form des Säurechlorids durch Einwirkung von Thionylchlorid in einem Lösungsmittel wie Toluol vor der Reaktion mit dem Amin der Formel (VII) aktiviert werden.
- Das Amin der Formel VII kann auf mehreren verschiedenen Wegen erhalten werden, die entweder zum Diastereoisomer der cis- Konfiguration der Formel VII' oder zum Diastereoisomer der trans- Konfiguration der Formel VII" führen.
- Die Verbindung der Formel VII' kann durch Umsetzung des cis-Amins der Formel VIII' mit Hilfe eines Hydrids, wie Lithiumaluminiumhydrid, in Tetrahydrofuran erhalten werden.
- Gleichermaßen kann die Verbindung der Formel VII" durch Reduktion des Amids der Formel VIII" erhalten werden
- Die Verbindungen VIII werden in drei Stufen ausgehend von ß- Aminosäuren entsprechend dem folgenden Schema erhalten:
- Die Stufe 1 besteht darin, das primäre Amin durch eine Acylgruppe, wie Trifluoracetyl, wie bereits in der Literatur beschrieben (Act. Chim. Acad. Scient. Hung 99 (2), 175 - 192 (1979)), oder durch eine Urethangruppe, wie Benzyloxycarbonyl oder t-Butoxycarbonyl, oder durch eine andere Gruppe, die üblicherweise in der Synthese von Peptiden verwendet wird, zu schützen.
- Die Stufe 2 besteht darin, die Gruppe Z entweder durch eines der für die Herstellung von (I) beschriebenen Verfahren oder auch durch die intermediäre Bildung eines gemischten Anhydrids, das ausgehend von Chlorformiaten erhalten wurde, zu kondensieren.
- Die Stufe 3 besteht darin, die Schutzgruppe von der Aminfunktion zu entfernen, indem man ein alkalisches Medium im Fall von Trifluoracetyl, einen Hydrierungskatalysator im Fall einer Benzyloxycarbonylgruppe oder Trifluoressigsäure in Methylenchlorid im Fall einer t-Butoxycarbonylgruppe verwendet.
- Die Ausgangs-ß-Aminosäuren der relativen cis- oder trans- Konfiguration werden durch die Verfahren erhalten, die bereits in der Literatur beschrieben sind.
- Die Herstellung von optisch reinen Verbindungen VII kann durch stereoselektive Kristallisation der N-geschützten ß-Aminosäuren, zum Beispiel der N-benzoylierten, in Form eines Salzes bewirkt werden, das mit Hilfe eines optisch aktiven Amins, wie Chinchonidin, erhalten wurde. Eine solche Auftrennung ist in J. Chem. Soc. (1970), 1597 - 1600 beschrieben. Die optisch reine ß- Aminosäure wird anschließend durch Aneinanderreihen der folgenden Reaktionen in das Diamin VII überführt:
- Die Stufe 1 besteht darin, die Gruppe Z durch die bereits beschriebenen Verfahren zu kondensieren. Die Stufe 2 besteht darin, selektiv die tertiäre Amidfunktion durch ein Hydrid wie AlLiH&sub4; zu reduzieren. Die Stufe 3 besteht darin, die Benzoylgruppe in einem sauren wäßrigen Medium zu hydrolysieren. Die Folge der Reaktionen wird mit dem einen oder dem anderen der Enantiomere bewirkt.
- Ein anderes Verfahren zum Erhalt von optisch reinen Verbindungen besteht darin; eine stereoselektive Hydrolyse von Dimethyl-2,2- cyclohex-4-endicarboxylat mit Hilfe einer Lipase, wie der Lipase von Schweineleber, durchzuführen. Man erhält nach einer Curtius- Umlagerung, die mit dem Säureester durchgeführ wird, optisch reine ß-Aminosäuren.
- Ein derartiges Verfahren ist in Tetrahedron Letters (1984), 25, 2557 - 2560 beschrieben. Die Doppelbindung von Cyclohexen wird durch Hydrierung in Gegenwart eines Katalysators wie Palladium auf Kohle reduziert. Dieser Schritt kann in jedem Stadium in der Folge der Verfahrensschritte als Funktion der verwendeten Schutzgruppen durchgeführt werden. Diese ß-Aminosäuren werden durch die vorstehend beschriebenen Verfahren in das Diamin VII überführt.
- Die Verbindungen der Formel VII können auch durch eine Mannich- Reaktion mit Cyclohexanon, gefolgt von einer Reaktion der reduktiven Aminierung, die entweder über das Oxim oder über ein Imin, wie demjenigen, das mit Benzylamin erhalten wird, verläuft, gemäß dem Schema:
- erhalten werden.
- In diesem Fall wird eine Mischung aus cis- und trans-Verbindung erhalten, deren Verhältnis mit dem in der letzten Stufe verwendeten Reduktionsmittel variiert. Beispielsweise liefert ein Oxim, das ausgehend von dem Keton der Formel (IX) erhalten wurde, zum größten Teil ein trans-Amin VII" durch Reduktion mit Hilfe von Natrium in Isoamylalkohol und größtenteils ein cis-Amin VII' nach Reduktion mit dem Hydrid AlLiH&sub4;.
- Die Verbindungen der allgemeinen Formel VII" können auch ausgehend von dem Keton IX durch Reduktion mittels beispielsweise K-Selektrid (der Formel K(s-Bu)-3BH) in Tetrahydrofuran erhalten werden.
- Der erhaltene cis-Alkohol wird anschließend aktiviert, beispielsweise durch ein Mesylat-Zwischenprodukt, dann durch eine Funktion substituiert, die ein primäres Amin erzeugen kann, wie Azid oder Phthalimid. Die Vorstufe wird anschließend durch in der Literatur bekannte Verfahren in das Amin überführt. Das Reaktionsschema ist das folgende:

- Die folgenden Beispiele erläutern die Herstellung von Verbindungen der Formel I.
- Beispiel 1
- N-[cis-2-(4-Methylpiperidinornethyl)cyclohexyl]-4-amino-5-chlor-2- methoxybenzamid.
- Zu einer Lösung von 4-Amino-5-chlor-2-methoxybenzoesäure (3,67 g; 18,2 mMol) in THF (50 ml) wird Carbonyldiimidazol (2,95 g; 18,2 mMol) gegeben.
- Nach einstündigem Rühren bei Umgebungstemperatur wird cis-2-(4- Methylpiperidinomethyl)cyclohexylamin (3,83 g; 18,2 mMol) in THF (15 ml) gelöst dazugegeben. Die Mischung wird über Nacht bei Umgebungstemperatur gerührt. Die Mischung wird unter reduziertem Druck eingedampft. Der Rückstand wird mit Methylenchlorid aufgenommen, mit einer 8%-igen wäßrigen Bicarbonat-Lösung, dann mit Wasser gewaschen.
- Die organische Phase wird über Natriumsulfat getrocknet, und das Lösungsmittel wird unter reduziertem Druck abgezogen, um Kristalle zu ergeben (5,87 g, 83%), die aus einer Mischung Isopropylether/Aceton (8/2) umkristallisiert werden.
- Schmelzpunkt 141 - 142ºC.
- IR: (C = O): 1638 cm&supmin;¹ (CHCl&sub3;)
- N-[cis-2-(4-Methylpiperidinomethyl)cyclohexyl]-4-amino-5-chlor-2- methoxybenzamid, Hydrochlorid, Hydrat.
- Das im vorstehenden Beispiel erhaltene Produkt (4,56 g) wird in absolutem Ethanol (15 ml) gelöst und mit einer 4 N Lösung von Chlorwasserstoff in Ethylether (4,3 ml) behandelt. Das Medium wird konzentriert, und das Hydrochlorid wird durch Zugabe von Wasser (30 ml) in Form von Hydrat (5,4% Wasser) kristallisiert.
- Schmelzpunkt: 58 - 145ºC (Zersetzung).
- IR: (C = O): 1627 cm&supmin;¹ (KBr).
- N-(trans-2-(4-Methylpiperidinomethyl)cyclohexyl]-4-amino-5-chlor- 2-methoxybenzamid, Dihydrochlorid.
- Das Produkt wird wie in Beispiel 1 unter Verwendung von trans-2- (4-Methylpiperidinomethyl) cyclohexylamin hergestellt. Das Produkt wird durch Chromatographie über einer Kieselgelsäule gereinigt, wobei man mit einem System Methylenchlorid/Ethylacetat/Methanol/Ammoniak (85/15/5/0,3) eluiert. Das erhaltene Öl wird mit Chlorwasserstoffsäure in Ether behandelt, das Lösungsmittel wird verdampft, und der Rückstand wird in Aceton aufgenommen, um Kristalle zu liefern.
- Fp.: 182 - 184ºC
- IR: (C = O): 1646 cm&supmin;¹ (KBr).
- N-[cis-2-(4,4-Dimethylpiperidinomethyl)cyclohexyl]-4-amino-5- chlor-2-methoxybenzamidhydrochlorid, Hemihydrat.
- Das Produkt wird wie in Beispiel 3 ausgehend von cis-2-(4,4- Dimethylpiperidinomethyl) cyclohexylamin hergestellt.
- Fp.: 132 - 153ºC (Zers.)
- IR: (C = O): 1617 cm&supmin;¹ (CHCl&sub3;).
- Die anderen Verbindungen werden durch identische Verfahren unter Verwendung von Benzoesäure und dem entsprechenden Cyclohexylamin- Derivat erhalten. Bei 3-Chlor-2, 4, 6-trimethoxybenzoesäure wird das Säurechlorid als aktiviertes Zwischenprodukt verwendet. Die Eigenschaften dieser Verbindungen sind in der Tabelle I zusammengefaßt. - Die folgenden Beispiele erläutern die Herstellung von Cyclohexylaminen der Formel VII.
- cis-2-(4-Methylpiperidinomethyl)cyclohexylamin.
- a) 1-[cis-(2-Trifluoracetamidocyclohexylcarbonyl)]-4- methylpiperidin.
- cis-2-Trifluoracetamidocyclohexancarbonsäurechlorid (8,61 g; 33 mMol), gelöst in Benzol (50 ml), wird tropfenweise mit 4- Methylpiperidin (8,3 g; 83 mMol), gelöst in Benzol (20 ml), behandelt. Das Reaktionsmedium wird 1 Std. 30 Min. bei Umgebungstemperatur gehalten, bevor man das Lösungsmittel verdampft. Der Rückstand wird durch Chloroform aufgenommen, die organische Phase wird mit Wasser gewaschen, über Natriumsulfat getrocknet, dann unter reduziertem Druck eingedampft. Das erhaltene Produkt wird aus einer Mischung Benzol/Petrolether umkristallisiert.
- Fp.: 135,5ºC
- IR: (C= O): 1718 - 1621,5 cm&supmin;¹ (CHCl&sub3;).
- b) 1-[cis-(2-Aminocyclohexylcarbonyl]-4-methylpiperidin.
- 1-[cis-(2-Trifluoracetamidocyclohexyl)carbonyl]-4-methylpiperidin (8,8 g; 0,027 Mol) wird in 95%-igem Ethanol (250 ml) gelöst, dann wird eine Lösung von 1 N Natriumhydroxid in 95%-igem Ethanol (50 ml; 0,05 Mol) dazugegeben.
- Das Reaktionsmedium wird 48 Std. bei Umgebungstemperatur gerührt, dann unter reduziertem Druck eingedampft. Der Rückstand wird in Chloroform aufgenommen, mit 1 N NaOH gewaschen, die organische Phase wird über Na&sub2;SO&sub4; getrocknet und eingedampft, um ein gelbliches Öl zu ergeben.
- IR (C = O): 1621 cm&supmin;¹ (CHCl&sub3;).
- c) cis-2-(4-Methylpiperidinomethyl)cyclohexylamin.
- 1-[cis-(2-Aminocyclohexyl)carbonyl]-4-methylpiperidin (6,57 g; 0,0293 Mol) wird in Tetrahydrofuran (25 ml) gelöst und tropfenweise zu einer Suspension von AlLiH&sub4; (2,22 g; 0,0586 Mol) in Tetrahydrofuran (50 ml) gegeben. Am Ende der Zugabe wird das Reaktionsmedium 24 Std. zum Rückfluß gebracht, dann unter vorsichtiger Zugabe von Wasser (2,3 ml), dann einer 15%-igen Natriumhydroxid-Lösung (2,3 ml), dann von Wasser (5 ml) hydrolysiert.
- Die Aluminiumsalze werden durch Filtration entfernt, und das Tetrahydrofuran wird im Vakuum abgedampft. Der Rückstand wird durch eine Säurelösung aufgenommen, die mit einer 15%-igen Natriumhydroxid-Lösung alkalisch gemacht wird, mit Chloroform extrahiert wird. Die organische Phase wird über Na&sub2;SO&sub4; getrocknet und im Vakuum eingedampft. Der Rückstand wird durch Destillation gereinigt.
- Sp. (27 Pa): 86 - 87ºC.
- cis-2-(4,4-Dimethylpiperidinomethyl)cyclohexylamin.
- a) 1-[cis-2-Trifluoracetamidocyclohexyl)carbonyl]-4,4- dimethylpiperidin.
- Das Produkt wird gemäß dem in Beispiel A a) beschriebenen Verfahren erhalten, indem man 4,4-Dimethylpiperidin verwendet. Das verbleibende Öl wird aus einer Mischung von Petrolether/Ethylacetat (80/20) kristallisiert.
- Fp.: 97 - 106ºC, IR (C = O): 1717,8 - 1622,0 cm&supmin;¹ (CHCl&sub3;).
- b) 1-(cis-(2-Aminocyclohexyl)carbonyl]-4,4-dimethylpiperidin.
- Das in a) erhaltene Produkt wird gemäß dem in Beispiel A b) beschriebenen Verfahren detrifluoracetyliert, um ein Öl zu liefern.
- IR (C = O): 1621 cm&supmin;¹ (CHCl&sub3;).
- C) cis-2-(4,4-Dimethylpiperidinomethyl)cyclohexylamin.
- 1-[cis-(2-Aminocyclohexyl)carbonyl]-4,4-dimethylpiperidin wird gemäß dem in Beispiel A c) beschriebenen Verfahren mit AlLiH&sub4; reduziert. Das erhaltene Öl wird destilliert.
- Sp. (53 Pa): 112ºC.
- Die anderen cis-Cyclohexylamin-Derivate werden gemäß den gleichen Verfahren erhalten und sind in der Tabelle II aufgeführt. TABELLE II

- Trans-2-(4-methylpiperidinomethyl)cyclohexylamin.
- a) 2-(4-Methylpiperidinomethyl)cyclohexylamin.
- Cyclohexanon (47,5 g; 0,484 Mol) wird in 95%-igem Ethanol gelöst, das Formaldehyd (36 g einer 37%-igen wäßrigen Lösung), Chlorwasserstoffsäure (1,7 ml einer 36%-igen wäßrigen Lösung) und 4-Methylpiperidin (40 g; 0,4 Mol) enthält. Die Lösung wird 4 Stunden zum Rückfluß gebracht. Das Ethanol wird im Vakuum verdampft, und der Rückstand wird durch 2 N Salzsäure aufgenommen. Die wäßrigen Phase wird mit Ether gewaschen, mit einer 15%-igen wäßrigen Natriumhydroxid-Lösung alkalisch gemacht, dann mit Methylenchlorid extrahiert. Die organische Phase wird über Na&sub2;SO&sub4; getrocknet, dann wird das Lösungsmittel im Vakuum abgezogen.
- Das erhaltene Öl wird destilliert.
- Sp. (27 Pa): 85 - 90ºC
- IR (C = O): 1706 (CHCl&sub3;).
- b) 2-(4-Methylpiperidinomethyl)cyclohexanonoxim.
- 2-(4-Methylpiperidinomethyl)cyclohexanon (20,8 g; 0,1 Mol) wird in Ethanol (100 ml) gelöst. Zu dieser Lösung wird Hydroxylaminhydrochlorid (7,54 g; 0,109 Mol), gelöst in Wasser (15 ml), dazugegeben. Die Reaktionsmischung wird 1 Std. bei Umgebungstemperatur gerührt. Der Rückstand wird in Chloroform aufgenommen und mit einer 2 N wäßrigen Natriumhydroxid-Lösung gewaschen. Die organische Phase wird über Na&sub2;SO&sub4; getrocknet und eingedarnpft, um ein weißes Pulver zu liefern.
- Fp.: 113 - 116,5ºC.
- c) 2-(4-Methylpiperidinomethyl)cyclohexylamin.
- Das im vorangehenden Beispiel erhaltene Oxim (13,5 g; 0,06 Mol) wird in Amylalkohol (200 ml) gelöst. Natrium (8,33 g; 0,36 Mol) wird in kleinen Mengen so dazugegeben, daß das Sieden sanft bleibt. Am Ende der Zugabe wird das Reaktionsmedium abgekühlt, dann mit einer wäßrigen 2 N HCl-Lösung angesäuert. Die wäßrige Phase wird mit Ethylacetat extrahiert, mit einer 15%-igen Natriumhydroxid-Lösung alkalisch gemacht, dann mit Methylenchlorid extrahiert. Die organische Phase wird über Na&sub2;SO&sub4; getrocknet, dann eingedampft. Das verbleibende Öl wird destilliert.
- Sp. (27 Pa): 94 - 98ºC. Das erhaltene Produkt enthält ungefähr 75% trans-2-(4-Methylpiperidinomethyl)cyclohexylamin. Das reine Diastereoisomer wird auf der Ebene des Benzamids isoliert.
- (5,5)- und (R,R)-2-(4-Methylpiperidinomethyl)cyclohexylamin.
- a) 1-[2-(R)-Benzamido-(S)-cyclohexylcarbonyl]-4- methylpiperidin.
- 2-(R)-Benzamido-(S)-cyclohexylcarbonsäure (8,87 g) wird in 200 ml Tetrahydrofuran gelöst. Nach Zugabe von Carbonyldiimidazol (5,82 g) wird das Reaktionsmedium 1 Std. 30 Min. bei Umgebungstemperatur gerührt. Eine Lösung von 4-Methylpiperidin in 20 ml Tetrahydrofuran wird dazugegeben, und das Reaktionsmedium wird noch 48 Stunden gerührt. Das Lösungsmittel wird im Vakuum abgezogen, der Rückstand wird in Methylenchlorid aufgenommen. Die organische Phase wird 2 mal mit 50 ml 1 N HCl, 2 mal mit 50 ml 1 N NaOH, dann 2 mal mit 50 ml Wasser gewaschen, über Na&sub2;SO&sub4; getrocknet und unter reduziertem Druck eingedampft.
- Man erhält 11,16 g Öl (94%) [a]²&sup0;&sub5;&sub7;&sub8; 578 = 47,9º (c = 1, MeOH); IR: ν = 1731 schwach, 1648, 1620 cm&supmin;¹ (CHCl&sub3;).
- a') Ausgehend von der entgegengesetzten Säure erhält man durch das gleiche Verfahren 1-[2-(5)-Benzamido-(R)-cyclohexylcarbonyl]- 4-methylpiperidin.
- [α]²&sup0;&sub5;&sub7;&sub8; = +49,2º; (c = 1, MeOH); IRν = 1730 (schwach), 1648, 1619 cm&supmin;¹ (CHCl&sub3;).
- b) N-Benzoyl-2-(R)-4-methylpiperidinomethyl-(R)- cyclohexylamin.
- Das in Stufe a) erhaltene Produkt (10,38 g), gelöst in 20 ml Tetrahydrofuran, wird tropfenweise zu einer Suspension von AlLiH&sub4; (2,4 g) in 130 ml Tetrahydrofuran, gekühlt bei 0ºC, gegeben. Am Ende der Zugabe wird die Mischung 45 Minuten auf 60ºC gebracht. Das Medium wird abgekühlt, dann durch eine wäßrige 1 N NaOH- Lösung (12 ml) hydrolysiert. Der unlösliche Feststoff wird durch Filtration entfernt, und das Filtrat wird im Vakuum eingedampft. Der Rückstand wird in 1 N Salzsäure gelöst. Die wäßrige Phase wird mit Ethylacetat gewaschen, dann durch 2 N Natronlauge alkalisch gemacht. Das Produkt wird mit Ethylacetat extrahiert. Die organische Phase wird über Na&sub2;SO&sub4; getrocknet, dann unter reduziertem Druck eingedampft. Das gesuchte Produkt wird aus einer Mischung Petrolether-Ethylacetat (95-5) kristallisiert und durch Filtration gesammelt = 6,53 g (65%).
- [α]²&sup0;&sub5;&sub7;&sub8; = - 67,10 (c = 1, MeOH).
- Fp. = 89 - 91ºC.
- b') Ausgehend von dem in a') erhaltenen Produkt erhält man durch das gleiche Verfahren N-Benzoyl-2-(5)-(4-methylpiperidinomethyl)- (S)-cyclohexylamin.

- c) (R,R)-2-(4-Methylpiperidinomethyl)cyclohexylamin.
- Das in Stufe b) erhaltene Produkt (6,43 g) wird in 55 ml 6 N HCl gelöst. Die Mischung wird 5 Tage zum Rückfluß gebracht. Das Medium wird mit Ethylacetat extrahiert, mit einer konzentrierten Natriumhydroxid-Lösung alkalisch gemacht und dreimal mit 100 ml Ethylacetat extrahiert. Die organische Phase wird über Na&sub2;SO&sub4; getrocknet, dann unter reduziertem Druck eingedampft. Das Diamin wird in Form eines Öls erhalten (4,17 g; 97%).
- [α]²&sup0;D = 25,20 (C = 1, MeOH).
- c') Ausgehend von dem in b') erhaltenen Produkt liefert das gleiche Verfahren (S,S)-2-(4- Methylpiperidinomethyl) cyclohexylamin.
- [α]²&sup0;D = +25,20 (c = 1, MeOH).
- Nachstehend werden die Ergebnisse aufgeführt, die die Eigenschaften der Verbindungen der Formel I in zwei Tests, nämlich die Magenentleerung von Feststoffen und den Antagonismus von Stereotypen, die durch Apomorphin induziert wurden, belegen. Der erste Test gestattet, die Aktivität der Produkte auf die Verdauungsmotilität zu belegen, der zweite Test gestattet, die Abwesenheit von zentralen dopaminergen antagonistischen Wirkungen nachzuweisen.
- Die Magenentleerung wird bei männlichen Sprague-Dawley-Ratten von ungefähr 150 g untersucht. Am Tag 0 wird eine postdiaphragmatische Vagotomie so durchgeführt, daß ein Modell der verlangsamten Magenentleerung geschaffen wird. Am Tag 7 erhalten die Tiere auf ösophagialem oder intraperitonealem Weg das zu testende Produkt oder Placebo (TO). Bei T + 15 Minuten werden unverdauliche n Teilchen mit 1 mm Durchmesser auf intragastrischem Weg verabreicht. Die Tiere werden bei T + 3 Std. euthanasiert, und die Teilchen, die im Magen verbleiben, werden gezählt. Die Magenentleerung wird als Prozentsatz der Teilchen, die den Magen verlassen haben, ausgedrückt.
- Sie ist unterhalb von 30% bei den Ratten, die einer Vagotomie unterzogen wurden; zum Vergleich ist sie oberhalb von 50% bei den Tieren, die nur eine Laparotomie erhalten hatten.
- Bei jeder Verbindung werden mindestens 3 Dosen getestet (Minimum 10 Tiere pro Dosis), ein positiver Vergleichstest (Metoclopramid 5 mg kg&supmin;¹) wird immer parallel durchgeführt. Die statistischen Vergleiche beruhen auf dem nicht-parametrischen Test von Mann und Whitney.
- Die erhaltenen Ergebnisse sind in Tabelle III aufgeführt. Wenn sich ein Produkt als aktiv erweist, bestimmt man seine minimale wirksame Dosis (DME) und die Potentialisierung (in Form eines Vielfachen des Vergleichswertes), die bei dieser Dosis beobachtet wird. Wenn das Produkt bei der geringsten getesteten Dosis aktiv ist, präzisiert man, daß die DME kleiner oder gleich (< ) dieser Dosis ist.
- Stereotypien des Mundbereiches werden bei männlichen Sprague- Dawley-Ratten von ungefähr 150 g durch die subkutane Injektion von Apomorphinhydrochlorid (1 mg kg&supmin;¹) induziert. Die zu untersuchenden Produkte werden auf intraperitonealem Weg 30 Minuten vor der Injektion von Apomorphin verabreicht. Die Tiere werden 15, 30, 45, 60, 75 und 90 Minuten nach dieser Injektion beobachtet.
- Die Intensität der Stereotypien wird durch ein Bewertungssystem bewertet, das von 0 (Abwesenheit von anormalen Bewegungen) bis 3 (Lecken und/oder intensives oder permanentes Kauen) geht, und in Form einer kumulierten Skala innerhalb von 90 Minuten ausgedrückt.
- Bei jedem Produkt werden mindestens 2 Dosen getestet (minimal 6 Tiere pro Dosis), ein positiver Vergleich (Metoclopramid 3 mg kg&supmin;¹) wird immer parallel durchgeführt. Die statistischen Vergleiche beruhen auf dem nicht-parametrischen Test von Mann und Whitney. Die erhaltenen Ergebnisse sind in Tabelle III aufgeführt. Wenn die Produkte inaktiv (IN) sind, präzisiert man die stärkste getestete Dosis; wenn sich ein Produkt als aktiv erweist, präzisiert man die wirksame Dosis (D.E.) und den Prozentsatz der beobachteten Hemmung. - Die Verbindungen der Formel (I) können verwendet werden, um die gastrointestinale Motilität zu stimulieren. Sie können verwendet werden, um Krankheiten des Magenbereiches zu behandeln, wie Gastroparesie, gastro-ösophagialer Reflux, Dyspepsien, Verstopfung und bestimmte Formen des Syndroms des reizbaren Colons oder des Reizdarms.
- Man kann diese Verbindungen oder ihre Additionssalze, die mit pharmazeutisch annehmbaren Säuren erhalten wurden, beim Menschen in einer großen Vielfalt von pharmazeutischen Zusammensetzungen verabreichen.
- Gegenstand der vorliegenden Erfindung sind demgemäß ebenfalls therapeutische Zusammensetzungen, die als Wirkstoff eine Verbindung der Formel (I) oder eines ihrer Additionssalze mit pharmazeutisch annehmbaren Säuren umfassen.
- Der Wirkstoff kann auf oralerm, intravenösem, subkutanem oder parenteralem Weg auf einmal oder mehrere Male bei einer täglichen Dosis von 0,05 mg bis 50 mg verabreicht werden.
- Die therapeutischen Zusammensetzungen können 0,05 bis 50 mg Wirkstoff pro Einheit enthalten. Bei der Verabreichung auf oralem Weg können sie in Form von Tabletten oder Kapseln vorliegen, die einen der gegenwärtig auf dem pharmazeutischen Gebiet verwendeten Träger enthalten.
Claims (12)
1. Verbindungen der Formel
in der:
R&sub1; aus (C&sub1;-C&sub4;)-Alkoxy-, Methoxy- (C&sub1;-C&sub3;)-alkoxy-, (C&sub3;-C&sub4;)-
Alkenyloxy- und (C&sub5;-C&sub6;)-Cycloalkyloxygruppen ausgewählt
ist oder R&sub1; und R&sub2; in der Position 3 zusammen mit dem
aromatischen Ring, an dem sie angebracht sind, einen 2,3-
Dihydrobenzofuranzyklus bilden,
R&sub2;, R&sub3;, R&sub4; unabhängig voneinander ausgewählt sind aus einem
Wasserstoffatom, einem Halogenatom und Hydroxy-, (C&sub1;-C&sub3;)-
Alkoxy-, Amino-, (C&sub1;-C&sub3;)-Alkylamino-, Di-(C&sub1;-C&sub3;)-
alkylamino- und (C&sub1;-C&sub3;)-Alkylcarbonylaminogruppen,
Z ausgewählt ist aus der Piperidinylgruppe und einer
Piperidinylgruppe, die in 4 mit einer Hydroxy-, (C&sub1;-C&sub3;)-
Alkoxy-, Hydroxyethyl-, (C&sub1;-C&sub3;)-Alkoxyethyl-, (C&sub1;-C&sub3;)-
Alkoxymethylgruppe, einer (C&sub1;-C&sub4;)-Alkylgruppe oder zwei
(C&sub1;-C&sub3;)-Alkylgruppen substituiert ist,
ihre N-Oxide
und ihre Additionssalze mit pharmazeutisch annehmbaren
Säuren.
2. Verbindungen nach Anspruch 1, worin:
R&sub1; ausgewählt ist aus (C&sub1;-C&sub4;)-Alkoxy-, Methoxy-(C&sub1;-C&sub3;)-
alkoxy-, (C&sub3;-C&sub4;)-Alkenyloxy- und (C&sub5;-C&sub6;)-
Cycloalkyloxygruppen
oder R&sub1; und R&sub2; in Position 3 zusammen mit dem aromatischen
Ring, an dem sie angebracht sind, einen 2,3-
Dihydrobenzofuranzyklus bilden,
R&sub2;, R&sub3;, R&sub4; unabhängig voneinander ausgewählt sind aus einem
Wasserstoffatom, einem Halogenatom und Hydroxy-, (C&sub1;-C&sub3;)-
Alkoxy-, Amino-, (C&sub1;-C&sub3;)-Alkylamino-, Di-(C&sub1;-C&sub3;)-
alkylamino- und (C&sub1;-C&sub3;)-Alkylcarbonylaminogruppen,
Z ausgewählt ist aus der Piperidinylgruppe und einer
Piperidinylgruppe, die in 4 durch eine (C&sub1;-C&sub3;)-Alkoxy-,
(C&sub1;-C&sub3;)-Alkoxyethyl-, (C&sub1;-C&sub3;)-Alkoxymethylgruppe, eine
lineare oder verzweigte (C&sub1;-C&sub4;)-Alkylgruppe oder zwei (C&sub1;-
C&sub3;)-Alkylgruppen substituiert ist,
ihre N-Oxide
und ihre Additionssalze mit pharmazeutisch annehmbaren
Säuren.
3. Verbindungen nach Anspruch 1 der Formel
in der R&sub1; eine (C&sub1;-C&sub4;)-Alkoxygruppe ist.
4. Verbindungen nach Anspruch 3, worin R&sub1; eine Methoxygruppe
ist.
5. Verbindungen nach Anspruch 3, worin R&sub1; eine Ethoxy- oder
Cyclopropylmethoxygruppe ist.
6. Verbindungen nach irgendeinem der Ansprüche 3, 4 oder 5,
worin Z eine Piperidino-, 4-Methylpiperidino-, 4,4-
Dimethylpiperidino- oder 4-Hydroxypiperidinogruppe ist.
7. Verbindungen nach Anspruch 6 der Formel
in der R&sub7; eine Methylgruppe ist und R&sub1;&sub0; ein Wasserstoffatom
ist.
8. Verbindungen nach Anspruch 6 der Formel
in der R&sub7; und R&sub1;&sub0; Methylgruppen sind.
9. Verbindungen nach Anspruch 6 der Formel
in der R&sub7; eine Hydroxygruppe ist und R&sub1;&sub0; ein
Wasserstoffatom ist.
10. Verfahren zur Herstellung einer Verbindung der Formel I
nach Anspruch 1, in dem man eine Kondensation zwischen
einer Benzoesäure der Formel
in der R&sub1;, R&sub2;, R&sub3; und R&sub4; die in Anspruch 1 angegebene
Bedeutung aufweisen,
mit einem Cyclohexylamin der Formel
in der Z die in Anspruch 1 angegebene Bedeutung aufweist,
bewirkt.
11. Verfahren nach Anspruch 10, in dem man die Reaktion in
Anwesenheit eines Kupplungsmittels bewirkt, das aus
Carbonyldiimidazol und Dicyclohexylcarbodiimid ausgewählt
ist.
12. Therapeutische Zusammensetzung, die als Wirkstoff eine
Verbindung nach irgendeinem der Ansprüche 1 bis 9 umfaßt.
