Description
- Diese Erfindung bezieht sich auf ein Verfahren zur Herstellung von optisch aktiven γ-Butyrolacton-Derivaten, die wichtig sind als Insekten-Pheromone und Parfüms.
- Viele optisch aktive Lacton-Derivate der Formel (II):
- worin R¹ für eine Alkylgruppe mit 1 bis 10 Kohlenstoffatomen oder eine substituierte oder unsubstituierte Phenylgruppe steht und * ein asymmetrisches Kohlenstoffatom anzeigt, sind als solche wichtig als verschiedene Insekten- Pheromone oder Parfüms.
- Zu bekannten Verfahren zur Synthese dieser Verbindungen gehören (1) ein Verfahren, bei dem man von in der Natur vorkommenden optisch aktiven Substanzen ausgeht (vgl. "Agric. Biol. Chem.", Band 51, S. 635 (1987)), (2) ein Verfahren, bei dem γ-Ketosäuren unter Verwendung von Hefe reduziert werden (vgl. "Appl. Microbiol.", Band 11, S. 389 (1963)) und (3) ein Verfahren, das die Auftrennung von Racematen in die optischen Antipoden umfaßt (vgl. JP-A-55- 43053, wobei der hier verwendete Ausdruck "JP-A" für eine "ungeprüfte publizierte japanische Patentanmeldung" steht).
- Hinsichtlich des Verfahrens (3) ist es bekannt, daß racemisches γ-Butyrolacton beispielsweise erhalten wird durch Hydrierung von Maleinsäure und/oder Maleinsäureanhydrid in Gegenwart eines Rutheniumkatalysators, beispielsweise eines Phosphoniumchlorid-, primären bis tertiären Phosphinhydrochlorid- oder quaternären Phosphoniumhydrochlorid- Komplexes von Ruthenium (vgl. JP-A-2-200680).
- Das Verfahren (1), bei dem als Ausgangsmaterial eine in der Natur vorkommende Substanz verwendet wird, umfaßt viele Stufen und komplizierte Arbeitsgänge. Mit dem Verfahren (2), bei dem Mikroorganismen verwendet werden, ist es bis heute nicht gelungen, eine hohe optische Reinheit zu erzielen. Das Verfahren (3) hat den Nachteil, daß eine äquivalente Menge einer optisch aktiven Verbindung erforderlich ist und daß der erzielte Auftrennungs-Wirkungsgrad schlecht ist.
- Die Verwendung von chiralen Ruthenium-Phosphin-Komplexen für die homogene asymmetrische Hydrierung mehrerer funktionalisierter Ketone wurde bereits früher diskutiert, beispielsweise von Kitamura et al ["Journal of the American Chemical Society", Band 110, Nr. 2, Seiten 629-631]. Außerdem haben Noyori et al ["Journal of the American Chemical Society", Band 109, Nr. 19, Seiten 5856-5858] ein Verfahren zur Herstellung eines optisch aktiven Alkohols durch asymmetrische Hydrierung eines β-Ketoesters in Gegenwart eines solchen Ruthenium-Phosphin-Komplexes diskutiert, der auch von Fieser [in "Fieser and Fiesers Reagents for Organic Synthesis", Band 14, Seiten 38-44] diskutiert worden ist. Außerdem haben Kawano et al ["Journal of the Chemical Society, Chem. Con." 1988, Nr. 2, Seiten 87-88] die Ruthenium-BINAP-katalysierte Hydrierung von 1,3-Diketonen unter Bildung von 1,3-Diolen mit einer hohen Diastereo- und Enantio-Selektivität diskutiert. Außerdem haben Ikariya et al ["Journal of the Chemical Society", Chem. Con." 1985, Nr. 13, Seiten 299-293] diskutiert, daß neue chirale Ruthenium(II)-Komplexe Ru&sub2;Cl&sub4;(BINAP)&sub2;(NEt&sub3;) bzw. Ru&sub2;Cl&sub4;(p-Tolyl-(BINAP)&sub2;(NEt&sub3;) als Katalysatoren für die asymmetrische Hydrierung von Alkenen und einigen cyclischen Anhydriden dienen. Die nach den in diesem Abschnitt diskutierten Verfahren erhaltenen Verbindungen sind jedoch optisch aktive Alkohole und nicht optisch aktive Lactone, wie sie erfindungsgemäß erhalten werden.
- Es besteht daher ein Bedarf für ein Verfahren zur Synthese eines gewünschten optisch aktiven γ-Butyrolacton-Derivats mit einer höheren optischen Reinheit unter Anwendung einer verminderten Anzahl von Stufen.
- Unter Berücksichtigung der obengenannten Umstände haben die Erfinder der vorliegenden Erfindung umfangreiche Untersuchungen durchgeführt und dabei gefunden, daß ein Lacton-Derivat der Formel (II) mit einer hohen optischen Reinheit durch einfache Verfahrensmaßnahmen und eine verminderte Anzahl von Stufen erhalten werden kann durch enantioselektive Hydrierung einerγ-Ketosäure oder eines Esters derselben in Gegenwart eines verhältnismäßig billigen optisch aktiven Ruthenium-Katalysators.
- Das erfindungsgemäße Verfahren wird durch das folgende Reaktionsschema erläutert: H&sub2;/Ruthenium-Phosphin:Komplex
- worin R¹ die oben angegebe Bedeutung hat, R² für ein Wasserstoffatom oder eine niedere Alkylgruppe (z.B. eine solche mit 1 bis 4 Kohlenstoffatomen) steht und * ein asymmetrisches Kohlenstoffatom anzeigt.
- Die γ-Ketosäure oder ihr Ester (I), die erfindungsgemäß als Ausgangsverbindung verwendet wird, kann in der Regel leicht synthetisiert werden nach dem Verfahren, wie es von Jean Mathieu und Jean Weill-Rayal in "Formation of C-C Bonds", S. 374, Georg Thieme-Verlag, Stuttgart (1979), beschrieben ist.
- Spezifische Beispiele für die γ-Ketosäuren oder ihre Ester (I) sind Lävulinsäure, 4-Oxohexansäure, 4-Oxoheptansäure, 4-Oxodecansäure, 4-Oxododecansäure, 4-Phenyl-4-oxobuttersäure, 4-p-Methoxyphenyl-4-oxobuttersäure, 4-(3,4-Dimethoxyphenyl)-4-oxobuttersäure, 4-(3,4,5-Trimethoxyphenyl)-4- oxobuttersäure, 4-p-Chlorophenyl-4-oxobuttersäure und 4- Phenyl-4-oxobuttersäure; und ihre niederen Alkylester, deren Alkylrest vorzugsweise 1 bis 4 Kohlenstoffatome aufweist, beispielsweise ihre Methyl- und Ethylester.
- Zu Beispielen für optisch aktive Ruthenium-Phosphin- Komplexe, die verwendet werden können, gehören:
- (1) Verbindungen der Formel (III):
- RuxHyClz:(R³-BINAP)&sub2;(Q)P (III)
- worin R³-BINAP für ein tertiäres Phosphin der Formel (IV) steht:
- worin bedeuten:
- R³ ein Wasserstoffatom, eine Methylgruppe oder eine tert-Butylgruppe;
- Q ein tertiäres Amin;
- y die Zahl 0 oder 1;
- wobei dann, wenn y = 0, x 2, z 4 und p 1 darstellen; und dann, wenn y = 1, x 1, z 1 und p 0 darstellen;
- (2) Verbindungen der Formel (V):
- [Ru(R³-BINAP)](O -R&sup4;)&sub2; (V)
- worin R³-BINAP wie oben definiert ist; R&sup4; steht für eine niedere Alkylgruppe (z.B. eine solche mit 1 bis 4 Kohlenstoffatomen), eine halogenierte niedere Alkylgruppe (z.B. eine solche mit 1 bis 4 Kohlenstoffatomen) oder eine Phenylgruppe; und
- (3) Verbindungen der Formel (VI):
- worin X steht für ein Halogenatom; R&sup5; und R&sup6;, die gleich oder verschieden sein können, jeweils stehen für eine substituierte oder unsubstituierte Phenylgruppe und R&sup7;-BINAP steht für ein tertiäres Phosphin der Formel (VII):
- worin R&sup7; ein Wasserstoffatom, eine Methylgruppe, eine tert-Butylgruppe oder eine Methoxygruppe darstellt.
- Die optisch aktiven Ruthenium-Phosphin-Komplexe (1) der Formel (III) können beispielsweise nach dem in "J. Chem. Soc., Chem. Commun.", S. 922 (1985), oder in JP-A-61-63690 beschrieben Verfahren erhalten werden.
- Spezifische Beispiele für die Komplexe (1) sind Ru&sub2;Cl&sub4;- (BINAP)&sub2;(NET&sub3;) (worin BINAP steht für 2,2'-Bis-(di phenylphosphino)-1,1'-binaphthyl und Et steht für eine Ethylgruppe; das gilt auch nachstehend), Ru&sub2;Cl&sub4;(p-Tol- BINAP)&sub2;(NEt&sub3;) (worin p-Tol-BINAP steht für 2,2'-Bis-(di-p- tolylphosphino)-1,1'-binaphthyl, das gilt auch nachstehend), RuHCl(BINAP)&sub2; und RuHCl(p-Tol-BINAP)&sub2;.
- Die optisch aktiven Ruthenium-Phosphin-Komplexe (2) der Formel (V) können beispielsweise nach dem Verfahren erhalten werden, wie es in JP-A-62-265293 beschrieben ist.
- Spezifische Beispiele für die Komplexe (2) sind:
- Ru(BINAP)(O CH&sub3;)&sub2;, Ru(BINAP)(O CF&sub3;)&sub2;,
- Ru(p-Tol-BINAP)(O CH&sub3;)&sub2;, Ru(p-Tol-BINAP)(O CF&sub3;)&sub2;,
- Ru(BINAP) (O t-Bu)&sub2;
- (worin t-Bu steht für eine tert-Butylgruppe, das gilt auch nachstehend) und
- Ru(BINAP)(O Ph)&sub2; (worin Ph steht für eine Phenylgruppe, das gilt auch nachstehend).
- Die optisch aktiven Ruthenium-Phosphin-Komplexe (3) der Formel (VI) können beispielsweise aus einem Ruthenium-Phosphin-Komplex der Formel (VIII) synthetisiert werden:
- worin R&sup5;, R&sup6; und X wie oben definiert sind und dma steht für N,N-Dimethylacetamid.
- In den Ruthenium-Phosphin-Komplexen der Formeln (VI) und (VIII) stehen R&sup5; und R&sup6;, die gleich oder verschieden sein können, jeweils für eine substituierte oder unsubstituierte Phenylgruppe, wobei zu spezifischen Beispielen dafür gehören eine Phenylgruppe; eine Alkyl-substituierte Phenylgruppe, z.B. 2-Methylphenyl, 3-Methylphenyl und 4- Methylphenyl; eine Alkoxy-substituierte Phenylgruppe, z.B. 2-Methoxyphenyl, 3-Methoxyphenyl und 4-Methoxyphenyl; und eine Dialkylaminophenylgruppe, z.B. 2-Dimethylaminophenyl, 3-Dimethylaminophenyl und 4-Dimethylaminophenyl.
- Unter den Verbindungen der Formel (VIII) kann beispielsweise die Verbindung, in der R&sup5; und R&sup6; beide eine Phenylgruppe darstellen und X für ein Chloratom steht, d.h. [RuCl(PPh&sub3;)&sub2;-dma)]&sub2;(µ-Cl)&sub2;, quantitativ wie folgt hergestellt werden:
- [RuCl&sub3;(PPh&sub3;)&sub2;(dma)](dma), das beispielsweise nach dem Verfahren von I.S. Thorburn, S.J. Rettig und B.R. James, "Inorg. Chem.", Band 25, S. 234-240 (1986), erhalten wird, wird in Hexan bei 60 bis 90ºC 1 bis 20 h lang reagieren gelassen und nach dem Abkühlen auf Raumtemperatur wird die Reaktionsmischung durch ein Glasfilter filtriert, mit Hexan gewaschen und getrocknet.
- Unter den Verbindungen der Formel (VI) kann der Komplex, in dem R&sup5; und R&sup6; beide eine Phenylgruppe, R&sup7; ein Wasserstoffatom und X ein Chloratom darstellen, d.h. [RuCl(PPh&sub3;)-(BINAP)]&sub2;(µ-Cl)&sub2;, quantitativ erhalten werden durch Umsetzung des auf diese Weise erhaltenen [RuCl(PPh&sub3;)&sub2;(dma)&sub2;(µ-Cl)&sub2;-Komplexes mit BINAP in einem Lösungsmittel, z.B. in Chlorbenzol, o-Dichlorbenzol, Methylenchlorid und 1,2-Dichlorethan, bei 50 bis 100ºC für 5 bis 20 h, und anschließende Entfernung des Lösungsmittels durch Destillation unter vermindertem Druck.
- Die folgenden optisch aktiven Ruthenium-Phosphin-Komplexe, die zu den Verbindungen (3) gehören, können ebenfalls auf die gleiche Weise wie vorstehend beschrieben hergestellt werden, wobei jedoch das Ausgangsmaterial entsprechend geändert wird:

- [RuBr(PPh&sub3;)(p-MeO-BINAP)]&sub2;(µ-Br)&sub2;
- [RuI(PPh&sub3;)(BINAP)]&sub2;(µ-I)&sub2;

- In den obigen Formeln steht Ph für eine Phenylgruppe; Me steht für eine Methylgruppe; p-Tol-BINAP steht für 2,2'- Bis(di-p-tolylphosphino)-1,1'-binaphthyl; p-MeO-BINAP steht für 2,2'-Bis(di-p-methoxyphenylphosphino)-1,1'- binaphthyl; und p-t-Bu-BINAP steht für 2,2'-Bis(di-p-tert- butylphenylphosphino)-1,1'-binaphthyl.
- Bei der Durchführung des erfindungsgemäßen Verfahrens wird eine γ-Ketosäure oder ein Ester derselben (I) in einem Lösungsmittel gelöst, die Lösung wird in einen Autoklaven eingeführt und der Lösung wird ein optisch aktiver Ruthenium-Phosphin-Komplex in einer Menge von 0,01 bis 0,001 mol pro mol der γ-Ketosäure oder ihres Esters (I) zugegeben zur Durchführung einer enantioselektiven Hydrierung unter Rühren bei einer Temperatur von 5 bis 50ºC und vorzugsweise von 25 bis 30ºC unter einem Wasserstoffdruck von 5 bis 100 kg/cm² für 5 bis 300 h. Das Produkt kann aus der Reaktionsmischung isoliert werden durch Entfernung des Lösungsmittels und Destillieren des Rückstandes unter vermindertem Druck und/oder durch Verdünnen des Rückstandes mit einem Entwicklungs-Lösungsmittel und anschließende Silicagel-Kolonnenchromatographie.
- Die vorliegende Erfindung bietet industrielle Vorteile, sie schafft ein Verfahren zur Herstellung eines optisch aktiven γ-Butyrolacton-Derivats, das verwendbar ist als Insekten-Pheromon oder als Parfüm, in einer zufriedenstellenden optischen Reinheit durch Anwendung einfacher Verfahrensmaßnahmen und einer verminderten Anzahl von Stufen.
- Die vorliegende Erfindung wird nachstehend unter Bezugnahme auf Beispiele näher erläutert, es ist jedoch klar, daß die Erfindung darauf nicht beschränkt ist.
- In einen 100 ml-Autoklaven wurden 2,14 g (14,8 mmol) Ethyllävulinat und 14 ml Ethanol eingeführt und es wurden 21 mg (0,02 mmol) [RuCl&sub3;(PPh&sub3;)((R)-(BINAP)]&sub2;(µ-Cl)&sub2; in einer Stickstoffatmosphäre zu der Lösung zugegeben zur Durchführung einer Hydrierung bei 25ºC und einem Wasserstoffdruck von 100 kg/cm² für 112 h. Nach der Entfernung des Lösungsmittels durch Destillation wurde der Rückstand durch Silicagel-Kolonnenchromatographie unter Verwendung von Hexan/Diethylether als Entwicklungslösungsmittel gereinigt und das Eluat wurde 4 h lang auf 130ºC erhitzt. Nach dem Abkühlen auf Raumtemperatur wurde die resultierende Lösung mit 4 ml Diethylether verdünnt, mit 3 ml einer gesättigten wäßrigen Natriumhydrogencarbonatlösung gewaschen und über wasserfreiem Natriumsulfat getrocknet. Nach dem Abtrennen des wasserfreien Natriumsulfats durch Filtrieren wurde das Filtrat unter vermindertem Druck eingeengt und der Rückstand wurde unter vermindertem Druck destilliert, wobei man 1,3 g (Ausbeute: 88 %) (R)-4- Methyl-γ-butyrolacton erhielt,

- Durch Gaschromatographie unter Verwendung einer optisch aktiven Kapillar-Kolonne unter den nachstehend angegebenen Bedingungen (dies gilt auch weiter unten) wurde gefunden, daß das Produkt eine optische Reinheit von 99,46 %ee hatte:
- Kolonne: Lipodex B (hergestellt von der Firma Macherey- Nagel Co.); 25 m geschmolzenes Siliciumdioxid
- Kolonnen-Temperatur: 150ºC
- Kolonnen-Einlaßtemperatur: 190ºC
- Trägergas: Helium
- In einen 100 ml-Autoklaven wurden 1,18 g (10 mmol) Methyllävulinat und 15 ml Methanol eingeführt und es wurden 169 mg (0,1 mmol) [Ru&sub2;Cl&sub4;((R)-BINAP)&sub2;NEt&sub3; in einer Stickstoffatmosphäre zugegeben zur Durchführung einer Hydrierung bei 50ºC und bei einem Wasserstoffdruck von 100 kg/cm² für 20 h. Die Reaktionsmischung wurde durch Destillation von dem Lösungsmittel befreit und einer Silicagel- Kolonnenchromatographie unterworfen unter Verwendung von Hexan/Diethylether als Entwicklungslösungsmittel, um den Katalysator zu entfernen. Das Eluat wurde 3 h lang auf 130ºC erhitzt, danach auf Raumtemperatur abgekühlt. Die reultierende Lösung wurde mit 4 ml Diethylether verdünnt, mit 3 ml einer gesättigten wäßrigen Natriumhydrogencarbonatlösung gewaschen und über wasserfreiem Natriumsulfat getrocknet. Nach dem Abtrennen des wasserfreien Natriumsulfats durch Filtrieren wurde das Filtrat unter vermindertem Druck eingeengt und der Rückstand wurde unter vermindertem Druck destilliert, wobei man 0,7 g (Ausbeute: 70 %) (R)-4-Methyl-γ-butyrolacton erhielt.
- Optische Reinheit: 98 %ee

- Auf die gleiche Weise wie in Beispiel 2, wobei jedoch Ru&sub2;Cl&sub4;((R)-BINAP)&sub2;NEt&sub3; durch Ru&sub2;Cl&sub4;((S)-BINAP)&sub2;NEt&sub3; ersetzt wurde, erhielt man 0,7 g (Ausbeute: 70 %) (S)-4-Methyl-γ- butyrolacton.

- In einen 100 ml-Autoklaven wurden 1,78 g (10 mmol) 4- Phenyl-4-oxobuttersäure und 14 ml Ethanol eingeführt. Eine aus 44,9 mg (0,05 mmol) Ru((R)-p-Tol-BINAP)(O CH&sub3;)&sub2;, der doppelten molaren Menge an konzentrierter Chlorwasserstoffsäure und 5 ml Ethanol getrennt hergestellte Lösung wurde zu der oben hergestellten Lösung in einer Stickstoffatmosphäre zugegeben zur Durchführung einer Hydrierung bei 35ºC und bei einem Wasserstoffdruck von 70 kg/cm² für 230 h. Zu der Reaktionsmischung wurden 8 ml einer 10 %igen wäßrigen Natriumhydroxidlösung zugegeben, danach wurde 3 h lang bei 40ºC gerührt. Nach dem Abkühlen auf Raumtemperatur wurde die resultierende Lösung zweimal mit 30 ml-Portionen Methylenchlorid extrahiert. Die organische Schicht wurde entfernt und die wäßrige Lösung wurde mit einer 10 %igen wäßrigen Chlorwasserstoffsäure-Lösung auf pH 4 eingestellt und dann erneut dreimal mit 20 ml-Portionen Methylenchlorid extrahiert. Der vereinigte Extrakt wurde eingeengt und der Rückstand wurde durch Silicagel-Kolonnenchromatographie gereinigt unter Verwendung von Hexan/Diethylether als Entwicklungslösungsmittel, wobei man 1,02 g (Ausbeute: 63 %) (S)-4-Phenyl-γ-butyrolacton erhielt.

- 1.8-3.0 (m, 4H, CH&sub2;CH&sub2;), 5.57 (t, 1H, J=7 Hz, CH), 7.45 (s, 5H, Ph)
- Die in der nachstehenden Tabelle 1 angegebenen Verbindungen wurden auf die gleiche Weise wie in Beispiel 4 synthetisiert, wobei jedoch die angegebene γ-Ketosäure oder ihr Ester verwendet wurde, die Reaktionsbedingungen (Molverhältnis-substrat/Katalysator, Temperatur, Zeit und Lösungsmittel) wie in Tabelle 1 angegeben geändert wurden und der Wasserstoffdruck von 70 kg/cm² in 100 kg/cm² geändert wurde. Claims (3)
1. Verfahren zur Herstellung eines optisch aktiven γ-
Butyrolacton-Derivats der allgemeinen Formel (II):
worin R¹ steht für eine Alkylgruppe mit 1 bis 10
Kohlenstoffatomen oder eine substituierte oder unsubstituierte
Phenylgruppe und * ein asymmetrisches Kohlenstoffatom
anzeigt, das umfaßt die enantioselektive Hydrierung einer γ-
Ketosäure oder eines Esters derselben der folgenden
allgemeinen Formel (I)
worin R¹ und * wie oben definiert sind und R² steht für ein
Wasserstoffatom oder eine niedere Alkylgruppe,
in Gegenwart eines optisch aktiven Ruthenium-Phosphin-
Komplexes.
2. Verfahren nach Anspruch 1, worin der optisch aktive
Ruthenium-Phosphin-Komplex ausgewählt wird aus den
Verbindungen der nachstehend angegebenen allgemeinen Formeln
(III), (V) oder (VI):
RuxHyClz(R³-BINAP)&sub2;(Q)p (III)
worin R³-BINAP steht für ein tertiäres Phosphin der Formel
(IV):
worin R³ ein Wasserstoffatom, eine Methylgruppe oder eine
tert-Butylgruppe darstellt; Q steht für ein tertiäres
Amin; y steht für 0 oder 1; wobei dann, wenn y = 0, x 2, z
4 und p 1 darstellen; und dann, wenn y = 1, x 1, z 1 und p
0 darstellen;
[Ru(R³-BINAP)](O -R&sup4;)&sub2; (V)
worin R³-BINAP wie oben definiert ist und R&sup4; steht für eine
niedere Alkylgruppe, eine halogenierte niedere Alkylgruppe
oder eine Phenylgruppe; und
worin X steht für ein Halogenatom; R&sup5; und R&sup6;, die gleich
oder verschieden sein können, jeweils stehen für eine
substituierte oder unsubstituierte Phenylgruppe; R&sup7;-BINAP
steht für ein tertiäres Phosphin der Formel (VII):
worin R&sup7; ein Wasserstoffatom, eine Methylgruppe, eine
tert-Butylgruppe oder eine Methoxygruppe darstellt.
3. Verfahren nach Anspruch 1 oder 2, worin R¹ für eine
Alkylgruppe mit 1 bis 4 Kohlenstoffatomen steht.
