Description
- Die vorliegende Erfindung betrifft Chinazolinon-Derivate. Diese Erfindung betrifft weiterhin Verfahren zu ihrer Herstellung, ihre Verwendung als therapeutische Mittel und Arzneimittel, die sie enthalten. Die Verbindungen dieser Erfindung sind Inhibitoren einer nicht auf Calmodulin ansprechenden cyclo-GMP Phosphodiesterase und bei der Bekämpfung von Krankheitsbildern, bei denen eine solche Inhibierung als nützlich gilt, verwendbar. Sie sind Bronchodilatatoren und deshalb in der Bekämpfung von chronisch reversiblen obstruktiven Lungenkrankheiten, wie Asthma und Bronchitis verwendbar. Weiterhin handelt es sich bei ihnen um Vasodilatatoren, weshalb sie bei der Bekämpfung von Angina, Hypertonie und Stauungsinsuffizienz nützlich sind.
- Die japanische Patentanmeldung Nr. 52-51378 offenbart Verbindungen der allgemeinen Formel ( A ):
- in der R ein Alkylrest, eine Styrylgruppe, ein niederer Alkoxyrest, eine Nitrogruppe, ein Halogenatom oder eine substituierte Phenylgruppe ist und X und X¹, die gleich oder verschieden seien können, ein Wasserstoff - oder Halogenatom, eine Nitrogruppe oder ein niederer Alkylrest sind. Diese Verbindungen sollen in Form von pharmazeutischen Produkten, wie als bakterizide und mitizide Mittel oder als Zwischenprodukte bei deren Synthese nützlich sein. 2-(2-Ethoxyphenyl)chinazolin-4(3H)-on wird ausdrücklich offenbart.
- Das US-Patent 3169129 offenbart Verbindungen der allgemeinen Formel ( B ):
- in der R' ein Wasserstoffatom oder einen Substituenten, wie ein Halogenatom, eine Hydroxylgruppe, ein C&sub1;&submin;&sub4;Alkyl- oder C&sub1;&submin;&sub4;Alkoxyrest und R" ein Wasserstoffatom oder ein C&sub1;&submin;&sub4;Alkylrest bedeutet. Diese Verbindungen sollen günstige Fluoreszenseigenschaften aufweisen. Obwohl 2-(2-Methoxyphenyl)chinazolin-4(3H)-on ausdrücklich offenbart wird, wird pharmazeutische Wirksamkeit nicht erwähnt.
- In der vorliegenden Erfindung werden Verbindungen der Formel ( 1 ):
- und pharmazeutisch verträgliche Salze davon bereitgestellt, in der
- R¹ ein C&sub1;&submin;&sub6;Alkyl-, C&sub2;&submin;&sub6;Alkenyl-, C&sub3;&submin;&sub5;CycloalkylC&sub1;&submin;&sub4;alkyl-, PhenylC&sub1;&submin;&sub4; alkyl- oder ein mit 1 bis 6 Fluoratomen substituierter C&sub1;&submin;&sub4;Alkylrest ist,
- R² ein Wasserstoffatom, ein C&sub1;&submin;&sub6;Alkyl-, C&sub1;&submin;&sub6;Alkylthio-, C&sub1;&submin;&sub6;Alkoxyrest, eine Nitrogruppe oder ein Rest -NR³R&sup4; ist: und
- R³ und R&sup4; unabhängig voneinander Wasserstoffatome oder gegebenenfalls hydroxyl-substituierte C&sub1;&submin;&sub4;Alkylreste sind, mit der Maßgabe, daß das dem Stickstoffatom benachbarte Kohlenstoffatom nicht mit einer Hydroxylgruppe substituiert ist;
- mit der Maßgabe, daß R¹ keine Methyl- oder Ethylgruppe ist, wenn R² ein Wasserstoffatom ist.
- Geeigneterweise ist R¹ ein C&sub2;&submin;&sub5;Alkylrest, zum Beispiel eine Ethyl-, n-Propyl-, Isopropyl-, Butyl-, Isobutyl- oder Pentylgruppe.
- Geeigneterweise ist R¹ ein C&sub3;&submin;&sub5;Alkenylrest, zum Beispiel eine Allyl-, Butenyl- oder Pentenylgruppe.
- Geeigneterweise ist R¹ eine Cyclopropylmethyl- oder Benzylgruppe.
- Beispiele für mit 1 bis 6 Fluoratomen substituierte C&sub1;&submin;&sub4;Alkylreste beinhalten -CF&sub3;-, -CH&sub2;CF&sub3;- oder -CF&sub2;CHFCF&sub3;-Gruppen.
- Vorzugsweise ist R¹ eine n-Propylgruppe.
- Geeigneterweise ist R² ein Wasserstoffatom oder ein C&sub1;&submin;&sub6;Alkylrest, zum Beispiel eine Methyl- oder Ethylgruppe.
- Geeigneterweise ist R² ein C&sub1;&submin;&sub6;Alkylthio- oder C&sub1;&submin;&sub6;Alkoxyrest, zum Beispiel eine Methylthio-, Ethylthio-, Methoxy- oder Ethoxygruppe.
- Geeigneterweise ist R² eine Nitrogruppe oder ein Rest -NR³R&sup4;, zum Beispiel eine Methylamino-, Dimethylamino- oder 2-Hydroxyethylaminogruppe.
- Besondere Verbindungen dieser Erfindung sind:
- 2-(2-Propoxyphenyl)chinazolin-4(3H)-on,
- 7-Methylthio-2-(2-propoxyphenyl)chinazolin-4(3H)on,
- 7-Nitro-2-(2-propoxyphenyl)-4(3H)-chinazolinon,
- 7-Amino-2-(2-propoxyphenyl)-4(3H)-chinazolinon oder
- 7-Methylamino-2-(2-propoxyphenyl)-4(3H)-chinazolinon
- oder pharmazeutisch verträgliche Salze davon.
- Diese Erfindung umfaßt alle tautomeren und optisch isomeren Formen der Verbindungen der Formel ( 1 ).
- Verbindungen der Formel ( 1 ) können pharmazeutisch verträgliche Salze mit Metallionen, wie Alkalimetallen, zum Beispiel Natrium und Kalium oder einem Ammoniumion bilden.
- Verbindungen der Formel ( 1 ), in denen R² ein Rest -NR³R&sup4; ist, können mit Säuren, wie Salzsäure, Bromwasserstoffsäure Schwefelsäure und Phosphorsäure pharmazeutisch verträgliche Salze bilden.
- Um eine Verbindung der Formel ( 1 ) oder ein pharmazeutisch verträgliches Salz davon zur Behandlung von Menschen und anderen Säugern zu verwenden, wird sie üblicherweise gemäß pharmazeutischen Standard verfahren als Arzneimittel formuliert.
- Verbindungen der Formel ( 1 ) und ihre pharmazeutisch verträglichen Salze können zur Behandlung der angezeigten Krankheiten in standardisierter Weise verabreicht werden, zum Beispiel oral, sublingual, parenteral, transdermal, rektal, durch Inhalation oder durch buccale Verabreichung.
- Verbindungen der Formel ( 1 ) und ihre pharmazeutisch verträglichen Salze, die bei oraler oder buccaler Verabreichung wirksam sind, können geeigneterweise als Dosierungsformen, wie Flüssigkeiten, Sirupe, Tabletten, Kapseln und Pastillen formuliert werden. Eine Flüssigrezeptur zur oralen Verabreichung besteht im allgemeinen aus einer Suspension oder Lösung der Verbindung oder des Salzes in einem flüssigen Träger, zum Beispiel Ethanol, Glycerin oder Wasser mit einem Geschmacks- oder Färbemittel. Sofern die Zusammensetzung als Tablette vorliegt, kann ein beliebiger routinemäßig zur Herstellung von festen Rezepturen gebräuchlicher pharmazeutischer Träger verwendet werden. Beispiele solcher Träger beinhalten Stärke, Cellulosen, Lactose, Rohrzucker und Magnesiumstereat. Sofern die Zusammensetzung als Kapsel vorliegt, kann ein beliebiges routinemäßiges Verkapselungsverfahren geeignet sein, zum Beispiel unter Verwendung der vorstehend erwähnten Träger in einer Hartgelatinekapselumhüllung. Sofern die Zusammensetzung als Kapsel mit einer Umhüllung aus weicher Gelatine vorliegt, kann ein beliebiger pharmazeutischer Träger, der routinemäßig für die Herstellung von Dispersionen oder Suspensionen verwendet wird, verwendet werden, zum Beispiel wäßrige Gummis, Cellulosen, Silikate oder Öle, die in die Kapselumhüllung aus weicher Gelatine eingebracht werden.
- Typische parenterale Zusammensetzungen bestehen aus einer Lösung oder einer Suspension der Verbindung oder des Salzes in einem sterilen wäßrigen oder nicht wäßrigen Träger, der gegebenenfalls ein parenteral verträgliches Öl oder Lösungsvermittler enthält, zum Beispiel Polyethylenglycol, Polyvinylpyrrolidon, 2-Pyrrolidon, Cyclodextrin, Lecithin, Arachisöl oder Sesamöl.
- Eine typische Rezeptur für Zäpfchen umfaßt eine Verbindung der Formel ( 1 ) oder eines pharmazeutisch verträglichen Salzes davon, das auf diese Weise verabreicht wirksam ist, mit einem Binde- und/oder Gleitmittel, zum Beispiel polymere Glykole, Gelatinen, Kakaobutter oder andere niedrig schmelzende pflanzliche Wachse oder Fette oder ihre synthetischen Analoga.
- Typische transdermale Rezepturen umfaßen einen üblichen wäßrigen oder nicht wäßrigen Träger, zum Beispiel eine Creme, Salbe, Lotion oder Paste oder sie sind in Form eines medizinischen Pflasters, einer Auflage oder einer Membran.
- Typische Inhalationszusammensetzungen sind in Form von Lösungen, Suspensionen oder Emulsionen, die in Form eines Aerosols unter Verwendung eines üblichen Treibmittels, wie Dichlordifluormethan oder Trichlorfluormethan oder in Form eines Pulvers für die Insufflation verabreicht werden können.
- Bevorzugt liegt die Zusammensetzung als Einheitsdosierform, zum Beispiel als Tablette, Kapsel oder als abgemessene Aerosoldosierung vor, so daß der Patient sich selber eine Einzeldosis verabreichen kann.
- Jede Einheitsdosis zur oralen Verabreichung enthält geeigneterweise 0,001 mg/kg bis 30 mg/kg und bevorzugt 0,005 mg/kg bis 15 mg/kg und jede Einheitsdosis zur parenteralen Veranreichung enthält geeigneterweise 0,001 mg/kg bis 10 mg/kg einer Verbindung der Formel ( 1 ) oder eines pharmazeutisch verträglichen Salzes davon, als freie Base berechnet.
- Die tägliche Dosierung bei oraler Verabreichung liegt geeigneterweise bei etwa 0,001 mg/kg bis 120 mg/kg einer Verbindung der Formel ( 1 ) oder eines pharmazeutisch verträglichen Salzes davon, als freie Base berechnet. Die tägliche Dosierung bei parenteraler Verabreichung liegt geeigneterweise bei etwa 0,001 mg/kg bis 40 mg/kg, zum Beispiel bei etwa 0,005 mg/kg bis 10 mg/kg einer Verbindung der Formel ( 1 ) oder eines pharmazeutisch verträglichen Salzes davon, berechnet als freie Base. Der wirksame Bestandteil kann den Erfordernissen gemäß verabreicht werden, zum Beispiel 1 bis 8 mal täglich oder durch Infusion. Die Zusammensetzungen der Erfindung sind Bronchodilatatoren und bei chronischen reversiblen obstruktiven Lungenkrankheiten, zum Beispiel Asthma und Bronchitis, nützlich. Die Zusammensetzungen der vorliegenden Erfindung zeigen vasodilatatorische Wirksamkeit auf und sind in der Behandlung von Angina, Bluthochdruck und Stauungsinsuffiziens nützlich. Solche Krankheitsbilder können durch orale, sublinguale, topische, rektale, parenterale Verabreichung oder durch Inhalation behandelt werden. Bei der Verabreichung durch Inhalation werden die Dosen durch ein Ventil reguliert wie erforderlich verabreicht und sind für einen Erwachsenen üblicherweise im Bereich von 0,1 - 5,0 mg einer Verbindung der Formel ( 1 ) oder eines pharmazeutisch verträglichen Salzes davon.
- Die Verbindungen dieser Erfindung können zusammen mit anderen pharmazeutisch wirksamen Verbindungen verabreicht werden, zum Beispiel in Kombination, gleichzeitig oder nacheinander. Geeigneterweise werden die Verbindungen dieser Erfindung und die andere wirksame Verbindung oder Verbindungen in einer einzigen pharmazeutischen Zusammensetzung formuliert. Beispiele für Verbindungen, die zusammen mit den Verbindungen der Formel ( 1 ) in Arzneimitteln enthalten sein können, sind Bronchodilatatoren, wie sympathomimetische Amine, wie Isoprenalin, Isoetharin, Salbutamol, Phenylephrin und Ephedrin oder Xanthin-Derivate, wie Theophyllin und Aminophyllin, antiallergische Mittel, wie Dinatriumchromglycat, Histamin H&sub1;-Antagonisten, Vasodilatoren, wie Hydralazin, Anginotensin umwandelnde Enzyminhibitoren, wie Captopril, Antianginalmittel, wie Isosorbidnitrat, Glycerintrinitrat und Pentaerythrittetranitrat, Antiarrhythmiemittel, wie Quinidin, Procainamid und Lignocain, Calciumantagonisten, wie Verapamil und Nifedipin, Diuretika, wie Thiazide und verwandte Verbindungen, wie Bendroflumethiazid ( Bendroflurazid ), Chlorthiazid, Chlorthalidon, Hydrochlorthiazid und andere Diuretika, wie Furosemid und Triamteren und Sedativa, wie Nitrazepam, Flurazepam und Diazepam.
- In einer weiteren Ausführungsform der vorliegenden Erfindung wird ein Verfahren zur Herstellung von Verbindungen der Formel ( 1 ) und pharmazeutisch verträglicher Salze davon zur Verfügung gestellt, wobei das Verfahren das Cyclisieren einer Verbindung der Formel ( 2 ):
- in der R¹ wie vorstehend definiert ist, und R&sup5; ein wie vorstehend definierter Rest R² oder ein Vorläufer davon ist, umfaßt und anschließend falls notwendig:
- - Überführen des Restes R&sup5; in einen Rest R²;
- - Gegebenenfalls Bilden eines pharmazeutisch verträglichen Salzes davon.
- Geeigneterweise wird die Cyclisierung einer Verbindung der Formel ( 2 ) in Gegenwart einer Base, zum Beispiel wäßrige Natron- oder Kalilauge, gegebenenfalls mit einem Co-Lösungsmittel, zum Beispiel Pyridin, Tetrahydrofuran, Dioxan oder Dimethylformamid, bei Umgebungs- oder erhöhten Temperaturen, zum Beispiel von 20 bis 150ºC, bevorzugt bei der Rückflußtemperatur des Reaktionsgemisches, durchgeführt.
- Ein Beispiel dafür, daß R&sup5; ein Vorläufer des Restes R² ist, ist gegeben wenn R&sup5; eine Nitrogruppe ist. Eine solche Gruppe kann durch Behandlung mit einem Reduktionsmittel, zum Beispiel durch katalytische Hydrierung, in eine Aminogruppe überführt werden.
- Eine Verbindung der Formel ( 1 ), in der R² eine Aminogruppe ist, kann unter reduktiven Aminierungsbedingungen, zum Beispiel durch Umsetzung mit einem geeigneten Aldehyd oder Keton, wie Acetaldehyd oder Aceton in Gegenwart eines Reduktionsmittels, wie Natriumborhydrid oder durch Umsetzung mit einem geeigneten Orthoameisensäureester, wie Trimethylorthoameisensäureester, gefolgt von einer Umsetzung mit einem Reduktionsmittel, wie Natriumborhydrid, in einen C&sub1;&submin;&sub4;Alkylaminorest überführt werden.
- Eine Verbindung der Formel ( 2 ) kann durch Umsetzung einer Verbindung der Formel ( 3 ):
- in der X ein Halogenatom und R¹ wie vorstehend definiert ist, mit einer Verbindung der Formel ( 4 ):
- in der L eine Aminogruppe oder ein C&sub1;&submin;&sub4;Alkoxyrest und R&sup5; wie vorstehend definiert ist und anschließend, falls L ein C&sub1;&submin;&sub4;Alkoxyrest ist, durch Umsetzung mit Ammoniak, hergestellt werden.
- Geeigneterweise wird die Umsetzung der Verbindungen der Formeln ( 3 ) und ( 4 ) bei Umgebungs- oder erhöhter Temperatur, zum Beispiel 20 bis 100ºC in einem Lösungsmittel, wie Aceton, Toluol oder Tetrahydrofuran in Gegenwart einer Base, wie Triethylamin, Pyridin, wäßriger Natronlauge oder wäßriger Natriumacetatlösung, durchgeführt. Bevorzugt wird die Umsetzung bei Umgebungstemperatur in Aceton oder Tetrahydrofuran, in Gegenwart eines Gemisches aus Essigsäure und wäßriger Natriumacetatlösung, durchgeführt. Geeigneterweise ist X ein Brom- oder Chloratom.
- Eine Verbindung der Formel ( 3 ) kann durch Umsetzung einer Verbindung der Formel ( 5 ):
- in der R¹ wie vorstehend definiert ist, mit einem geeigneten Halogenierungsmittel hergestellt werden. Geeignete Halogenierungsmitttel beinhalten Thionylchlorid oder Phosphortribromid.
- Pharmazeutisch verträgliche Basenadditionssalze der Verbindungen der Formel ( 1 ) können durch Standardverfahren, zum Beispiel durch Umsetzung einer Lösung der Verbindung der Formel ( 1 ) mit einer Lösung der Base hergestellt werden.
- Pharmazeutisch verträgliche Säureadditionssalze der Verbindungen der Formel ( 1 ), in denen R² ein Rest -NR³R&sup4; ist, können auf übliche Weise aus der korrespondierenden Base der Verbindungen der Formel ( 1 ) hergestellt werden. Die Base kann zum Beispiel mit einer Säure in einem C&sub1;&submin;&sub4;Alkanol umgesetzt werden oder ein Ionenaustauscherharz kann verwendet werden. Die Salze der Verbindungen der Formel ( 1 ) können unter Verwendung von Ionenaustauscherharzen in einander überführt werden. Pharmazeutisch nicht verträgliche Salze sind daher von Nutzen, da sie in pharmazeutisch verträgliche Salze überführt werden können.
- Fine weitere Ausführungsform der vorliegenden Erfindung stellt eine Verbindung der Formel ( 1 ) wie vorstehend definiert oder 2-(2-Methoxyphenyl)chinazolin-4(3H)-on oder ein pharmazeutisches Salz davon zur Verwendung als Arzneimittel, besonders zur Verwendung als Bronchodilatator und/oder Vasodilatator, zur Verfügung.
- In einer weiteren Ausführungsform stellt die vorliegende Erfindung die Verwendung einer Verbindung der Formel ( 1 ) wie vorstehend definiert, von 2-(2-Methoxyphenyl)chinazolin-4(3H)-on oder von 2-(2-Ethoxyphenyl) chinazolin-4(3H)-on oder eines pharmazeutischen Salzes davon, zur Herstellung eines Arzneimittels mit bronchodilatatorischer und/oder vasodilatatorischer Wirksamkeit, zur Verfügung.
- 2-(2-Methoxyphenyl)chinazolin-4(3H)-on und 2-(2-Ethoxyphenyl)chinazolin-4(3H)-on können pharmazeutisch verträgliche Salze bilden, als Arzneimittel formuliert und als Arzneimitel wie vorstehend für die Verbindungen der Formel ( 1 ) beschrieben, verwendet werden.
- Die folgenden biologischen Testverfahren, Daten und Beispiele dienen der Erläuterung dieser Erfindung.
- Männliche Meerschweinchen des Dunkin Hartley Stammes ( 500 - 600 g ) wurden mit Sagatal ( Natriumpentobarbital ) ( 60 mg/kg ) anästhesiert. Der Luftwegwiderstand wurde unter Verwendung einer Modifikation der klassischen Konzett-Rossler Technik ( Versuchsanordnung zu Untersuchungen an der Bronchialmuskulatur, Naunyn-Schmiedebergs Arch. Exp. Path. Pharmak., Vol. 195 (1940 ), 71-74 ) gemessen. U 46619 ( 9,11-Methanepoxy-PGH&sub2; ) wurde mit einer Zugabegeschwindigkeit von 2,5 nmol/min i.v. infundiert, wodurch eine zeitlich konstante ( steady state ) Bronchialverengung ( annähernd 120% Zunahme des basalen Luftwiderstandes ) erzeugt wird. Die zu testende Verbindung wurde durch eine i.v. Bolusinjektion verabreicht und anschließend die Peakhöhe der Inhibierung der Bronchialverengung aufgezeichnet.
- Die durch U 46619 hervorgerufene Bronchialverengung wurde durch Verabreichung der Verbindung von Beispiel 1 ( 10 umol/kg ) um 34% verringert. Dieses Ergebnis zeigt in vivo die Wirksamkeit gegen Bronchialverengung.
- Die Wirksamkeit der Verbindungen der vorliegenden Erfindung als Inhibitoren einer nicht auf Calmodulin ansprechenden cyclo-GMP Phosphodiesterase wurde unter Verwendung des in der Europäischen Patentanmeldung Nr. 293063 beschriebenen Verfahrens gemessen. Die Verbindungen der Beispiele 1 bis 5 hatten IC&sub5;&sub0;-Werte ( die Konzentration des Inhibitors, die für eine 50% Inhibierung der Enzymaktivität benötigt wird ) im Bereich von 0,71 bis 5,30 uM. Die Verbindungen der vorliegenden Erfindung haben den Vorteil, daß sie cyclo-AMP Phosphodiesterase ( Typ III ) selektiv nicht inhibieren.
- a) Ein Gemisch aus 2-Propoxybenzoesäure ( 5 g ) in Thionylchlorid ( 20 ml ) wurde drei Stunden unter Rückfluß erhitzt. Das Thionylchlorid wurde unter vermindertem Druck entfernt und der Rückstand mit Toluol azeotrop destilliert, wodurch das Säurechlorid als gelbes Öl ( 5,5 g ) erhalten wurde. Eine Lösung von Anthranilamid ( 3,77 g ) in einem Gemisch aus gesättigtem wäßrigem Natriumacetat ( 20 ml ) und Essigsäure ( 20 ml ) wurde dem gelben Öl in Aceton ( 4 ml ) zugesetzt. Die entstandene Lösung wurde eine Stunde bei Umgebungstemperatur gerührt, wobei man 3,78 g 2-(2-Propoxybenzamido)benzamid als hellbraune Abscheidung erhielt. Eine aus Ether umkristallisierte Probe wies einen Schmelzpunkt von 149 - 150ºC auf.
- b) 2-(2-Propoxybenzamido)benzamid ( 2,73 g ) wurde einem unter Rückfluß kochendem Gemisch aus 2 n wäßriger Natronlauge ( 55 ml ) und Pyridin ( 2 ml ) zugesetzt. Die resultierende Lösung wurde 15 Minuten unter Rückfluß gerührt, auf Eis ( 150 ml ) gegeben und mit konzentrierter Salzsäure angesäuert, wobei man 2,28 g einer Probe der Titelverbindung mit einem Schmelzpunkt von 88 - 89ºC erhielt. Diese und eine weitere vergleichbar hergestellte Probe ( 0,77 g ) wurden aus Ethanol/Wasser umkristallisiert, wobei man 2,57 g der reinen Titelverbindung als weißen Feststoff mit einem Schmelzpunkt von 89 - 90ºC erhielt.
- a) Durch eine Lösung von Kupfersulfatpentahydrat ( 50 g ) in Wasser ( 200 ml ) wurde 30 Minuten Methylmercaptan durchgeleitet, so daß Kupfer(I)thiomethoxid ( 11,70 g ) als Abscheidung erhalten wurde. Dieses wurde angesammelt und nacheinander mit Wasser, Methanol und Ether gewaschen.
- Unter Kühlung ( 3 - 4ºC ) wurde eine Lösung von Natriumnitrit ( 3,70 g ) in Wasser ( 15 ml ) einer gerührten Suspension von 4-Amino-2-nitrobenzoesäure ( 9,11 g ) in Wasser ( 50 ml ) und konzentrierter Schwefelsäure ( 10 ml ) zugesetzt um ein Diazoniumsalz herzustellen. Die entstehende Lösung wurde 30 Minuten tropfenweise zu einer gekühlten ( 4ºC ) Aufschlämmung von Kupfer(I)thiomethoxid ( 11,7 g ) in Wasser gegeben und das Reaktionsgemisch 1,5 Stunden bei Umgebungstemperatur gerührt.
- Das Reaktionsgemisch wurde mit Chloroform ( insgesamt 650 ml ) gut extrahiert und die Chloroformextrakte wurden mit 2 n Salzsäure ( 2*25 ml ) gewaschen, getrocknet ( Magnesiumsulfat ) und unter vermindertem Druck eingeengt. Der Rückstand wurde in verdünnter wäßriger Natronlauge gelöst und 2 n Salzsäure wurde zum Ausfällen von 4-Methylthio-2-nitrobenzoesäure ( 3,71 g ) mit einem Schmelzpunkt von 172,5ºC zugesetzt.
- b) Eine gerührte Lösung von 4-Methylthio-2-nitrobenzoesäure ( 4,56 g ) und Thionylchlorid ( 2,77 g ) in Benzol ( 65 ml ) wurde 1,5 Stunden unter Rückfluß erhitzt. Wäßrige Ammoniaklösung ( 10 ml ) wurde zu dem gerührten und gekühlten ( < 5ºC ) Reaktionsgemisch tropfenweise zugesetzt, das dann unter Kühlung 45 Minuten gerührt wurde. Das Benzol wurde unter vermindertem Druck entfernt und der feste Rückstand mit Wasser gewaschen und umkristallisiert, wobei man 4-Methylthio-2-nitrobenzamid ( 2,35 g ) mit einem Schmelzpunkt von 176 - 178ºC erhielt.
- c) Ein gerührtes Gemisch aus 4-Methylthio-2-nitrobenzamid ( 2,47 g ), Zinnchloriddihydrat ( 13,13 g ) und Ethanol ( 90 ml ) wurde eine Stunde unter Rückfluß und einer Stickstoffatmosphäre erhitzt. Das Reaktionsgemisch wurde auf Eis gegeben und mit einer 5%igen wäßrigen Natriumbicarbonatlösung auf einen Wert von pH 7 neutralisiert. Das entstehende Gemisch wurde mit Ethylacetat:Methanol ( 9:1, 6*100 ml ) extrahiert. Die vereinigten Extrakte wurden mit Kochsalzlösung gewaschen, getrocknet ( Magesiumsulfat ) und unter vermindertem Druck eingeengt wobei man 2-Amino-4-methylthiobenzamid ( 1,84 g ) mit einem Schmelzpunkt von 172,5 - 173,5ºC erhielt.
- d) Eine Lösung von 2-Propoxybenzoylchlorid ( 0,76 g ) in Acetonitril ( 6 ml ) wurde einem eiskalten gerührten Gemisch aus 2-Amino-4-methylthiobenzamid ( 0,70 g ) und Triethylamin ( 0,39 g ) in Acetonitril ( 6 ml tropfenweise zugesetzt. Das Reaktionsgemisch wurde 17 Stunden bei Umgebungstemperatur gerührt. Das Acetonitril wurde unter vermindertem Druck entfernt und der zurückbleibende Feststoff mit Wasser gewaschen und aus Ethanol umkristallisiert, wobei man 4-Methylthio-2-(2-propoxybenzamido)benzamid ( 0,49 g ) mit einem Schmelzpunkt von 178,5 - 181ºC erhielt. Eine weitere Probe ( 0,40 g, Schmelzpunkt 182 - 184ºC ) wurde durch Einengen der ethanolischen Mutterlauge und Umkristallisieren des Rückstandes aus Ethanol hergestellt.
- e) Eine gerührte Lösung von 4-Methylthio-2-(2-propoxybenzamido)benzamid ( 0,85 g ) und Pyridin ( 1 ml ) in 2 n wäßriger Natronlauge wurde 2 Stunden unter Rückfluß erhitzt. Das gekühlte Reaktionsgemisch wurde mit konzentrierter Salzsäure angesäuert und das entstehende Gemisch mit Chloroform ( 3*25 ml ) extrahiert. Die vereinigten Extrakte wurden mit Wasser und Kochsalzlösung gewaschen, getrocknet ( Magnesiumsulfat ) und dann unter vermindertem Druck eingeengt, wobei man einen Feststoff erhielt, der aus Ethanol umkristallisiert 0,61 g der Titelverbindung mit einem Schmelzpunkt von 156,5 - 158,5ºC ergab.
- 4-Nitroanthranilsäuremethylester ( 2,0 g; hergestellt aus 4-Nitroanthranilsäure durch Umsetzung mit trockenem Chlorwasserstoff in Methanol unter Rückfluß ) wurde einer Lösung aus 2-Propoxybenzoylchlorid ( 2,0 g ) und Triethylamin ( 1,0 g ) in trockenem Tetrahydrofuran ( 5 ml ) bei 0ºC zugesetzt. Nachdem das Gemisch 60 Minuten bei Raumtemperatur gerührt wurde, wurde Wasser zugesetzt und das Gemisch mit Wasser und Dichlormethan ausgeschüttelt. Die organische Phase wurde abgetrennt, getrocknet ( Magnesiumsulfat ) und eingeengt, wobei sich ein Feststoff ergab ( 3,6 g ). Dieses Material ( 1.0 g ) wurde in 50 ml gesättigter methanolischer Ammoniaklösung gelöst und die Lösung 18 Stunden in einem Druckgefäß bei 100ºC erhitzt. Abdampfen der Lösungsmittel und Umkristallisation des Rückstandes aus Dichlormethan - Petrolether ergab 0,9 g 7-Nitro-(2-propoxyphenyl)-4(3H)-chinazolinon mit einem Schmelzpunkt von 138ºC.
- Eie gerührte Lösung von 7-Nitro-2-(2-propoxyphenyl)-4(3H)-chinazolinon ( 3,0 g ) in trockenem Methanol ( 450 ml ) wurde nacheinander unter einer Kohlendioxidatmosphäre mit 5%igem Pd/C ( 1,5 g ) und wasserfreiem Ammoniumformiat ( 2,8 g ) behandelt. Nach 60 Minuten wurden die Feststoffe durch Filtration entfernt und das Filtrat bis zur Trockne eingeengt. Der Rückstand wurde mit Wasser und Dichlormethan ausgeschüttelt, die organische Phase abgetrennt, getrocknet ( Magnesiumsulfat ) und eingeengt. Der Rückstand wurde aus Diethylether - Petrolether umkristallisiert, wobei man 2,7 g 7-Amino-2-(2-propoxyphenyl)-4(3H)-chinazolinon mit einem Schmelzpunkt von 185ºC erhielt.
- 7-Amino-2-(2-propoxyphenyl)-4(3H)-chinazolinon ( 0,5 g ) wurde in Trimethylorthoameisensäureester ( 5 ml ) 22 Stunden unter Rückfluß erhitzt. Das Reaktionsgemisch wurde zur Trockne eingeengt. Der Rückstand wurde in trockenem Tetrahydrofuran gelöst und 24 Stunden mit Natriumborhydrid ( 0,3 g ) und 4 Å Molekularsieb behandelt. Das Reaktionsgemisch wurde mit Wasser und Dichlormethan ausgeschüttelt, die organische Phase abgetrennt, getrocknet ( Magesiumsulfat ) und eingeengt. Der Rückstand wurde aus Dichlormethan - Diethylether umkristallisiert, wobei man 0,17 g 7-Methylamino-2-(2-propoxyphenyl)-4(3H)-chinazolinon mit einem Schmelzpunkt von 204 - 205ºC erhielt.
- Arzneimittel zur oralen Verabreichung werden durch Kombination des Folgenden hergestellt: 2-(2-Propoxyphenyl)chinazolin-4(3H)-on 2% w/w Soyalecithin in Soyabohnenöl Hydriertes pflanzliches Speisehartfett und Bienenwachs Die Rezepturen werden dann in einzelne Weichgelatinekapseln gefüllt.
- Ein Arzneimittel zur parenteralen Verabreichung wird durch Lösen der Titelverbindung das Beispiels 4 ( 0,02 g ) in Polyethylenglycol 300 ( 25 ml ) unter Erhitzen hergestellt. Diese Lösung wird dann mit Wasser für Injektionen Ph. Eur. verdünnt ( auf 100 ml ). Die Lösung wird durch Filtration durch einen 0,22 um Membranfilter sterilisiert und in sterilen Behältern verschlossen.
Claims (12)
1. Verbindung der Formel (1):
oder ein pharmazeutisch verträgliches Salz davon, in der
R¹ ein C&sub1;&submin;&sub6;Alkyl-, C&sub2;&submin;&sub6;Alkenyl-, C&sub3;&submin;&sub5;CycloalkylC&sub1;&submin;&sub4;alkyl-, PhenylC&sub1;&submin;&sub4;-
alkyl-, oder ein mit 1 bis 6 Fluoratomen substituierter C&sub1;&submin;&sub6;Alkylrest
ist;
R² ein Wasserstoffatom, ein C&sub1;&submin;&sub6;-Alkyl-, C&sub1;&submin;&sub6;Alkylthio-,
C&sub1;&submin;&sub6;Alkoxyrest, eine Nitrogruppe oder ein Rest -NR³R&sup4; ist; und
R³ und R&sup4; unabhängig voneinander Wasserstoffatome oder
gegebenenfalls hydroxyl-substituierte C&sub1;&submin;&sub4;Alkylreste sind, mit der Maßgabe, daß
das dem Stickstoffatom benachbarte Kohlenstoffatom nicht mit einer
Hydroxylgruppe substituiert ist;
mit der Maßgabe, daß R¹ keine Methyl- oder Ethylgruppe ist, wenn R²
ein Wasserstoffatom ist.
2. Verbindung nach Anspruch 1, in der R¹ ein C&sub2;&submin;&sub5;Alkylrest ist.
3. Verbindung nach Anspruch 1, in der R¹ ein C&sub3;&submin;&sub5;Alkenylrest ist.
4. Verbindung nach einem der Ansprüche 1 bis 3, in der R² ein
Wasserstoffatom oder ein C&sub1;&submin;&sub6;Alkylrest ist.
5. Verbindung nach einem der Ansprüche 1 bis 3, in der R² ein
C&sub1;&submin;&sub6;Alkylthio- oder C&sub1;&submin;&sub6;Alkoxyrest ist.
6. Verbindung nach einem der Ansprüche 1 bis 3, in der R² eine
Nitrogruppe oder ein Rest -NR³R&sup4; ist.
7. Verbindung nach Anspruch 1, nämlich
2-(2-Propoxyphenyl)chinazolin-4(3H)-on,
7-Methylthio-2-(2-propoxyphenyl)chinazolin-4(3H)-on,
7-Nitro-2-(2-propoxyphenyl)-4(3H)-chinazolinon,
7 Amino-2-(2-propoxyphenyl)-4(3H)-chinazolinon oder
7-Methylamino-2-(2-propoxyphenyl)-4(3H)-chinazolinon oder
ein pharmazeutisch verträgliches Salz davon.
8. 2-(2-Methoxyphenyl)chinazolin-4(3H)-on oder eine Verbindung nach
einem der Ansprüche 1 bis 7 oder ein pharmazeutisch verträgliches Salz
davon, zur Verwendung als Arzneistoff.
9. Verbindung nach Anspruch 8 zur Verwendung als Bronchodilatator
und/oder Vasodilatator.
10. Arzneimittel, umfassend eine Verbindung nach einem der Ansprüche 1
bis 9 und einen pharmazeutisch verträglichen Träger.
11. Verfahren zur Herstellung einer Verbindung der Formel (1) oder eines
pharmazeutisch verträgliches Salzes davon, wie in Anspruch 1 definiert,
wobei das Verfahren das Cyclisieren einer Verbindung der Formel (2):
in der R¹ wie in Anspruch 1 definiert ist und R&sup5; ein wie in Anspruch 1
definerter Rest R² oder ein Vorläufer davon ist, umfasst und anschließend,
falls notwendig:
- Überführen des Restes R&sup5; in einen Rest R²;
- gegebenenfalls Bilden eines pharmazeutisch verträglichen Salzes davon.
12. Verwendung einer wie in Anspruch 1 definierten Verbindung der
Formel (1), von 2-(2-Methoxyphenyl)chinazolin-4(3H)-on oder von
2-(2-Ethoxyphenyl)chinazolin-4(3H)-on oder eines pharmazeutisch
verträglichen Salzes davon zur Herstellung eines Arzneimittels mit
bronchodilatatorischer und/oder vasodilatatorischer Wirksamkeit.
