Description
- Die vorliegende Erfindung betrifft neue Phenoxyethylamin- Derivate, dargestellt anhand der folgenden allgemeinen Formel (I), ihre optischen Isomere, pharmakologisch annehmbare Säureadditionssalze derselben, Verfahren zu deren Herstellung, sowie pharmazeutische Zusammensetzungen, die ausgezeichnete α&sub1;-blockierende Wirkung zeigen, welche dieselben als wirksamen Bestandteil enthalten und bei der Behandlung von Bluthochdruck und Dysurie verwendet werden können.
- Es ist bereits bekannt, daß ein α&sub1;-Blocker zur Behandlung von Bluthochdruck verwendet werden kann, und eine Reihe von α&sub1;-Blockern wurde bereits auf den Markt gebracht oder entwickelt
- Unlängst wurde gefunden, daß α&sub1;-Blocker auf den glatten Muskel des unteren Harntrakts wirken, und zur Zeit wird eine neue Verwendung für die α&sub1;-Blocker, nämlich die Behandlung von Dysurie, Hypertrophie der Prostata und Urinierhäufigkeit mit Interesse verfolgt. Die Verbindung YM-12617 [Japan Kokai 56-110665], ein Beispiel für Verbindungen mit ähnlicher Struktur wie die der vorliegenden Erfindung, ist ebenfalls ein α&sub1;-Blocker.
- Sie besitzt die Formel
- doch sind Medikamente wie YM-12617 wegen ungenügender Wirksamkeit und dem Auftreten von Nebenwirkungen für den praktischen Gebrauch nicht zufriedenstellend.
- Als Ergebnis umfangreicher Forschungen wurde nun gefunden, daß neue Phenoxyethylamin-Derivate, dargestellt durch die allgemeine Formel (I)
- worin R&sub1; und R&sub5; niederes Alkyl bedeuten, R&sub2; und R&sub3;, die gleich oder verschieden sein können, jeweils Wasserstoff oder niederes Alkyl bedeuten, oder
- einen 5- oder 6gliedrigen Ring bedeutet, der substituiert sein kann und ein Stickstoff-, Sauerstoff- oder Schwefel- Atom als Ringatom enthalten kann, R&sub4; Wasserstoff oder niederes Alkyl bedeutet, und n eine ganze Zahl bedeutet, ausgewählt aus 1 bis 3, ihre optischen Isomere und ihre pharmakologisch annehmbaren Säureadditionssalze ausgezeichnete α&sub1;-blockierende Wirkung zeigen.
- Gemäß vorliegender Erfindung werden auch Verfahren zur Herstellung der neuen Phenoxyethylamin-Derivate, dargestellt durch die allgemeine Formel (I), pharmazeutischer Zusammensetzungen derselben, sowie eine Behandlungsmethode damit verfügbar gemacht.
- In dieser Erfindung ist die in der allgemeinen Formel (I) durch R&sub1;, R&sub2;, R&sub3;, R&sub4; und R&sub5; dargestellte Niederalkyl-Einheit zum Beispiel Methyl, Ethyl, n-Propyl, Isopropyl, n-Butyl, Isobutyl, sek-Butyl, tert-Butyl und dergleichen. Der durch
- dargestellte 5- oder 6gliedrige Ring ist beispielsweise 1-Pyrrolidinyl, 1-Piperidinyl, 2-Methyl-1-piperidinyl, 3-Methyl-1-piperidinyl, 4-Methyl-1-piperidinyl, 1-Piperazinyl, 2-Methyl-1-piperazinyl, 3-Methyl-1-piperazinyl, 4-Methyl-1-piperazinyl, 4-Morpholinyl, 4-Thiomorpholinyl oxder dergleichen.

- Zu den pharmakologisch annehmbaren Säureadditionssalzen der der anhand der allgemeinen Formel (I) dargestellten Verbindungen gehören zum Beispiel Mineralsäursalze wie etwa Hydrochlorid, Hydrobromid, Hydroiodid, Nitrat, Sulfat, Phosphat, Metaphosphat und dergleichen, sowie Salze organischer Säuren wie etwa Acetat, Maleat, Fumarat, Citrat, Oxalat, Succinat, Malat, Tartarat, Lactat, Malonat, Propionat, Mandelat, p-Toluolsulfonat, Methansulfonat, DL-10-Camphersulfonat, Gluconat, und dergleichen.
- Unter den durch die allgemeine Formel (I) dargestellten Verbindungen sind auch Verbindungen mit einem asymmetrischen Kohlenstoff-Atom. Diese Verbindungen können in optisch aktiver Form vorliegen. Deshalb sind alle Racemate und optische R- und S-Isomere in dieser Erfindung eingeschlossen.
- Gemäß vorliegender Erfindung können die neuen, durch die allgemeine Formel (I) dargestellten Phenoxyethylamin- Derivate mit Hilfe verschiedener Methoden hergestellt werden.
- Bei der ersten Methode können die durch die allgemeine Formel (I) dargestellten Verbindungen hergestellt werden durch Umsetzung von Phenoxyethylaminen, dargestellt durch die allgemeine Formel (II)
- worin R&sub5; die gleiche Bedeutung wie oben beschrieben hat, mit Carbonyl-Verbindungen, dargestellt durch die allgemeine Formel (III)
- worin R&sub1;, R&sub2;, R&sub3;, R&sub4; und n die gleiche Bedeutung wie oben beschrieben haben, in Gegenwart eines Lösungsmittels, und anschließendes Hydrieren oder Behandeln mit einem Reduktionsmittel.
- Bei dem in dem Verfahren verwendeten Lösungsmittel kann es sich um irgendeine Art Lösungsmittel handeln, welches die Reaktion nicht hemmt. Beispiele für verwendbare Lösungsmittel sind Methanol, Ethanol, n-Butanol, Ether, Tetrahydrofuran und dergleichen.
- Als Reduktionsmittel für die vorliegende Reaktion kann beispielsweise Natriumborhydrid, Natriumcyanborhydrid, Lithiumaluminiumhydrid und dergleichen verwendet werden. Die Hydrierung ist unter atmosphärischem oder höherem Druck durchzuführen. Als Katalysator kann zum Beispiel Palladium/Kohle, Platinoxid, Raney-Nickel und dergleichen verwendet werden.
- Die oben erwähnte Reaktion ist bei einer Temperatur im Bereich von Raumtemperatur bis Rückflußtemperatur des verwendeten Reaktionslösungsmittels durchzuführen.
- Die durch die Formel (II) dargestellten Phenoxyethylamine mit einem Fluor-Atom sind neue Verbindungen und sind wichtige Zwischenstufen zur Herstellung der durch die Formel (I) dargestellten Verbindungen. Die Verbindungen (II) können hergestellt werden mit Hilfe eines Verfahrens, das durch folgendes Schema erläutert sei:
- worin X ein Halogen-Atom darstellt und R&sub5; die gleiche Bedeutung wie oben beschrieben hat. Das heißt, die Verbindungen (IV) werden umgesetzt mit einem Ethylenhalogenid oder Ethylenchlorhalogenid in einem organischen Lösungsmittel (Dimethylsulfoxid, N,N-Dimethylformamid, Toluol etc.) in Gegenwart einer Base (Pyridin, Triethylamin, Kaliumcarbonat, Natriumcarbonat etc.), um die Verbindungen (VI) zu ergeben, oder die Verbindungen (IV) werden umgesetzt mit Ethylencarbonat oder Ethylenchlorhydrin in einem organischen Lösungsmittel (Dimethylsulfoxid, N,N-Dimethylformamid, Toluol etc.) in Gegenwart einer Base (Kaliumcarbonat, Natriumcarbonat, Natriumhydroxid etc.), um die Verbindungen (V) zu ergeben, und dann werden diese V mit einem Halogenierungsmittel behandelt (Thionylchlorid, Phosphoroxychlorid, Phosphorpentachlorid, Thionylbromid, Triphenylphosphindibromid etc.) entweder in Gegenwart oder in Abwesenheit eines organischen Lösungsmittels (Chloroform, Benzol, Toluol etc.), um die Verbindungen (VI) zu ergeben.
- Die Verbindungen (VI) werden umgesetzt mit Phthalimid in einem organischen Lösungsmittel (N,N-Dimethylformamid, Dimethylsulfoxid, Toluol etc.) in Gegenwart einer Base (Kaliumcarbonat, Natriumcarbonat etc.), um die Verbindungen (VII) zu ergeben, und diese VII werden dann mit wäßrigem Alkali (Natriumhydroxid, Kaliumhydroxid etc.) hydrolysiert oder mit Hydrazin in einem organischen Lösungsmittel (Methanol, Ethanol etc.) behandelt, um die gewünschten Verbindungen (II) zu ergeben. All die oben erwähnten Reaktionen sind bei einer Temperatur im Bereich von Raumtemperatur bis Rückflußtemperatur des verwendeten Reaktionslösungsmittels durchzuführen.
- Die durch Formel (III) dargestellten Carbonyl-Verbindungen, von denen die meisten neue Verbindungen sind, lassen sich herstellen mit Hilfe eines Verfahrens, das durch folgendes Schema erläutert wird, und die Einzelheiten der Herstellung sind in den Bezugsbeispielen angegeben.

- Bei der zweiten Methode können die durch die allgemeine Formel (I) dargestellten Verbindungen hergestellt werden durch Umsetzung von Phenoxyacetaldehyden, dargestellt durch die allgemeine Formel (VIII)
- worin R&sub5; die gleiche Bedeutung wie oben beschrieben hat, mit Amin-Derivaten, dargestellt durch die allgemeine Formel (IX)
- worin R&sub1;, R&sub2;, R&sub3;, R&sub4; und n die gleiche Bedeutung wie oben beschrieben haben, oder diesen optisch aktiven Aminen, in Gegenwart eines Lösungsmittels, und anschließendes Hydrieren oder Behandeln mit einem Reduktionsmittel.
- Bei dem in dem Verfahren verwendeten Lösungsmittel kann es sich um irgendeine Art Lösungsmittel handeln, welches die Reaktion nicht hemmt. Beispiele für verwendbare Lösungsmittel sind Methanol, Ethanol, n-Butanol, Ether, Tetrahydrofuran und dergleichen.
- Als Reduktionsmittel für die vorliegende Reaktion kann beispielsweise Natriumborhydrid, Natriumcyanborhydrid, Lithiumaluminiumhydrid und dergleichen verwendet werden. Die Hydrierung ist unter atmosphärischem oder höherem Druck durchzuführen. Als Katalysator kann zum Beispiel Palladium/Kohle, Platinoxid, Raney-Nickel und dergleichen verwendet werden.
- Die oben erwähnte Reaktion ist bei einer Temperatur im Bereich von Raumtemperatur bis Rückflußtemperatur des verwendeten Reaktionslösungsmittels durchzuführen.
- Die durch die Formel (VIII) dargestellten Phenoxyacetaldehyde mit einem Fluor-Atom sind neue Verbindungen und sind wichtige Zwischenstufen zur Herstellung der durch die Formel (I) dargestellten Verbindungen. Die Verbindungen (VIII) können hergestellt werden mit Hilfe eines Verfahrens, das durch folgendes Schema erläutert sei:
- worin R&sub5; die gleiche Bedeutung wie oben beschrieben hat.
- Das heißt, die Verbindungen (IV) werden umgesetzt mit Chloracetaldehyddiethylacetal in einem organischen Lösungsmittel (Tetrahydrofuran, N,N-Dimethylformamid, Dimethylsulfoxid etc.) in Gegenwart einer Base (Kaliumcarbonat, Natriumcarbonat, Triethylamin, Natriumhydrid, Natriumamid etc.), um die Verbindungen (X) zu ergeben. Bei dieser Reaktion kann Kaliumiodid oder Natriumiodid zugesetzt werden. Die erhaltenen Verbindungen (X) werden mit einer organischen Säure (Oxalsäure, Maleinsäure etc.) oder einer Mineralsäure (Schwefelsäure, Salzsäure etc.) in einem organischen Lösungsmittel (Aceton, Tetrahydrofuran etc.) oder einem wäßrigen organischen Lösungsmittel hydrolysiert, um die Phenoxyacetaldehyde (VIII) zu ergeben.
- All die oben erwähnten Reaktionen sind bei einer Temperatur im Bereich von Raumtemperatur bis Rückflußtemperatur des verwendeten Reaktionslösungsmittels durchzuführen.
- Die durch Formel (IX) dargestellten Amin-Verbindungen, von denen die meisten neue Verbindungen sind, lassen sich herstellen mit Hilfe eine Verfahrens, das durch folgendes Schema erläutert wird, und die Einzelheiten der Herstellung sind in den Bezugsbeispielen angegeben.
- worin R&sub1;, R&sub2;, R&sub3;, R&sub4; und n die gleiche Bedeutung wie oben beschrieben haben und R&sub6; niederes Alkyl oder niederes Halogenalkyl darstellt.
- Bei der dritten Methode können die durch Formel (I) dargestellten Verbindungen hergestellt werden durch Umsetzung von Fluorphenoxyalkan-Derivaten, dargestellt durch Formel (VI), mit Amin-Derivaten, dargestellt durch die Formel (IX), ohne Lösungsmittel oder in einem organischen Lösungsmittel, falls notwendig, in Gegenwart einer Base und Kaliumiodid oder Natriumiodid.
- Bei dem in dem Verfahren verwendeten Lösungsmittel kann es sich um irgendeine Art Lösungsmittel handeln, welches die Reaktion nicht hemmt. Beispiel für verwendbare Lösungsmittel sind Methanol, Ethanol, Isopropanol, n-Butanol, Dimethylsulfoxid, N,N-Dimethylfomamid, Benzol, Toluol, Tetrahyfrofuran und dergleichen.
- Als Base kann bei der vorliegenden Reaktion zum Beispiel Pyridin, Triethylamin, Kaliumcarbonat, Natriumcarbonat und dergleichen verwendet werden. Die Reaktion ist bei einer Temperatur im Bereich von Raumtemperatur bis 200ºC durchzuführen.
- Bei der vierten Methode können die durch Formel (I) dargestellten optisch aktiven Verbindungen hergestellt werden durch Bildung eines Salzes der Racemate mit einem optischen Trennungsreagens und anschließendes Umkristallisieren des erhaltenen Salzes.
- Als optisches Trennungsreagens kann in der vorliegenden Reaktion beispielsweise D-10-Camphersulfonsäure, L-10-Camphersulfonsäure, (+)-Dibenzoyl-D-weinsäure, (-)-Dibenzoyl- L-weinsäure, L-(+)-Weinsäure, D-(-)-Weinsäure, L-(+ )-Mandelsäure, D-(-)-Mandelsäure, D-Camphercarbonsäure, D-Äpfelsäure, L-Äpfelsäure, (+)-Di-p-toluoyl-D-weinsäure, (-)-Di-p- toluoyl-L-weinsäure, (-)-Menthyloxyessigsäure, (-)-Diacetyl- L-weinsäure, (+)-Monomethyl-D-weinsäure, (-)-Monomethyl-L- weinsäure, (-)-Diaceton-2-ketogulonsäure, (-)-Chinasäure, D-Glutaminsäure, L-Glutaminsäure, (S)-(-)-Pyrrolidon-5-carbonsäure, (R)-(-)-2-Phenylpropionsäure, (S)-(+ )-2-Phenylpropionsäure, (S)-1-(2-Naphthylsulfonyl)pyrrolidin-2-carbonsäure, (S)-1-(4-Toluolsulfonyl)pyrrolidin-2-carbonsäure und dergleichen verwendet werden.
- Als Lösungsmittel für die optische Trennung können zum Beispiel Wasser, niedere Alkohole wie etwa Methanol, Ethanol und Isopropanol, Halogenkohlenwasserstoffe wie etwa Chloroform, Dichlormethan, Dichlorethan und Tetrachlorkohlenstoff, Ketone wie etwa Aceton und Methylethylketon, Ether wie etwa Diethylether, Diisopropylether und Dioxan, aromatische Kohlenwasserstoffe wie etwa Benzol, Toluol, Xylol, Kohlenwasserstoffe wie etwa Hexan, Pentan und Cyclohexan, Nitrile wie etwa Acetonitril, Ester wie etwa Ethylacetat und Ethylformiat, Amide wie etwa N,N-Dimethylformamid und N,N-Dimethylacetamid, aprotische organische Lösungsmittel wie etwa Dimethylsulfoxid und Nitromethan sowie eine Mischung oben beschriebenr Lösungsmittel verwendet werden.
- Die optische Trennung ist bei einer Temperatur durchzuführen, die von Kühlen mit Eiswasser bis zu Erhitzen reicht.
- Eine durch die allgemeine Formel (I) dargestellte Verbindung der vorliegenden Erfindung kann per os verabreicht werden, z.B. in Form von Pillen oder Tabletten, worin sie zusammen mit irgendeinem der üblichen pharmazeutischen Carrier vorliegen kann, in herkömmlicher Weise bereitet durch Vermischen einer Verbindung dieser Erfindung zusammen mit einem üblichen Träger oder Adjuvans wie etwa Talk, Magnesiumstearat, Stärke, Lactose, Gelatine, irgendeinem der zahlreichen Gumen oder dergleichen. In ihrer vorteilhaftesten Form enthalten die erfindungsgemäßen Zusammensetzungen also neben dem wirksamen Bestandteil der vorliegenden Erfindung einen nichttoxischen pharmazeutischen Carrier. Beispielhafte feste Träger sind Lactose, Magnesiumstearat, Calciumstearat, Stärke, Terra alba, Dicalciumakazin oder dergleichen.
- Typische flüssige Träger sind Erdnußöl, Sesamöl, Olivenöl, Wasser oder dergleichen. Die erfindungsgemäßen wirksamen Bestandteile können in herkömmlicher Weise in derartigen, den wirksamen Bestandteil enthaltenden Zusammensetzungen verabreicht werden, so daß sie schließlich innerhalb des Dosierbereichs liegen, der naachstehend erläutert wird. Somit kann eine breite Vielfalt pharmazeutischer Formen eingesetzt werden, die für viele Arten der Verabreichung und Dosierung geeignet sind. Zur oralen Verabreichung können wirksamer Bestandteil und pharmazeutischer Carrier zum Beispiel die Form eines Pulvers, einer Granalie, Pille, Tablette, Kapsel, Pastille, eines Elixiers, Sirups oder einer anderen flüssigen Suspension oder Emulsion annehmen, wogegen die Zusammensetzung zur parenteralen Verabreichung in Form einer sterilen Lösung vorliegen kann. Zur intrarektalen Verabreichung kann die Zusammensetzung in Form eines Suppositoriums vorliegen.
- Die Methode der Anwendung der erfindungsgemäßen Verbindungen umfaßt die innerliche oder äußerliche Verabreichung einer erfindungsgemäßen Verbindung, vorzugsweise oral oder parenteral, und vorzugsweise vermischt mit dem pharmazeutischen Träger, zum Beispiel in der Form irgendeiner der obigen Zusammensetzungen oder eingefüllt in eine Kapsel, zur Linderung von zu behandelnden Zuständen und deren Symptomen in einem lebenden animalischen Körper. Zur Erläuterung kann sie oral dosiert in einer Menge von etwa 0,01 bis etwa 100 mg pro Tag (geteilt in drei Teile) verwendet werden, während parenterale Dosierungen normalerweise niedriger sind und gewöhnlich etwa die Hälfte der oralen Dosis betragen. Die Einheitsdosis wird vorzugsweise geeignet oft täglich gegeben, typischerweise dreimal.
- Die Einheitsdosis kann je nach Verabreichungshäufigkeit schwanken. Natürlich muß eine geeignete klinische Dosis gemäß Zustand, Alter und Gewicht des Patienten eingestellt werden, und es braucht nicht weiter ausgeführt zu werden, daß aufgrund der erhöhten Wirkung der erfindungsgemäßen Verbindungen, zusammen mit ihren verminderten Nebenwirkungen, diese auch für einen breiten Variationsbereich geeignet sind, und daher sollte diese Erfindung nicht durch die angegebenen exakten Bereiche eingeschränkt werden. Die genaue Dosierung, sowohl Einheitsdosierung als auch tägliche Dosierung, muß natürlich anhand gesicherter medizinischer Grundlagen bestimmt werden.
- Die folgenden Versuche zeigen die ausgezeichnete Wirkung der vorliegenden Verbindungen (Verbindungsnummer bedeutet Beispiel-Verbindungsnummer), zum Beispiel α&sub1;-Adrenozeptorblockierende Wirkung und beim Dopamin-D&sub2;-Rezeptor-Radioligandenbindungstest unter Verwendung von 5-{2-[[2-(2-Ethoxyphenoxy)ethyl]amino]propyl}-2-methoxybenzolsulfonamidhydrochlorid: YM-12617 als Bezugsverbindung.
- Es besteht die Möglichkeit, daß Verbindungen mit Affinität zu Dopamin-D&sub2;-Rezeptoren Nebenwirkungen hervorrufen wie etwa Übelkeit, Erbrechen oder anomale Prolaktin-Sekretion. Deshalb ist die Verbindung am brauchbarsten, die selektiv als α&sub1;-Adrenozeptor wirkt.
- Methode: Aus männlichen Ratten (Japanese white, etwa 2,5 kg Körpergewicht) wurde jeweils die Brustaorta isoliert und helikal geschnitten. Die Präparate wurden in Krebs-Henseleit-Lösung suspendiert, bei 37ºC gehalten und mit einer Gasmischung von 95% O&sub2; + 5% CO&sub2; begast. Reaktionen auf Medikamente wurden unter einer Spannung von 2 g isometrisch aufgezeichnet. Nach einer Äquilibrierungsphase von etwa 60 min wurden die Konzentration/Reaktion-Summenkurven für Noradrenalin konstruiert. Die Präparate wurden 30 min lang den Testverbindungen ausgesetzt, ehe nochmals Noradrenalin zur Einwirkung kam. Das Dosisverhältnis wurde erhalten aus dem Verhältnis des ED&sub5;&sub0;-Werts (benötigte Konzentration, um die Hälfte der Maximalreaktion hervorzurufen) für Noradrenalin in Gegenwart und Abwesenheit der Testverbindung. Die Dissoziationskonstanten (KB) der Testverbindungen wurden gemäß folgender Gleichung erhalten.
- KB = [Konzentration der Testverbindung (M) / (Dosisverhältnis-1)] Die pA&sub2;-Werte wurden dann als negativer Logarithmus von KB ausgedrückt.
- Dieser Wert stellt die blockierende Wirkung einer Testverbindung für α&sub1;-Adrenozeptoren dar. In dieser experimentellen Phase wurde 10&supmin;&sup6; M Propranolol gegeben, um ß-Adrenozeptoren zu blockieren. Aus den in Tabelle 1-a gezeigten Ergebnissen geht hervor, daß die α&sub1;-Adrenozeptor-blockierende Wirkung der Testverbindungen fast die gleiche ist wie die von YM-12617.
- Methode: Diese Experimente wurden gemäß der Methode von Coward D., et al. [Naunyn-Schmiedeberg's Archives of Pharmacology (1987) 335, 115-122] durchgeführt. In Kürze, es wurde Gehirn-Striatum aus männlichen Wistar-Ratten (etwa 200 g Körpergewicht) isoliert. Das Gewebe wurde in eiskaltem 50 mM Tris-Puffer (pH 7,4) mit einem Polytron homogenisiert und 15 min lang mit 50 000 g zentrifugiert. Die Pellets wurden im gleichen Tris-Puffer homogenisiert und 5 min lang bei 37ºC inkubiert. Die Homogenisate wurden zentrifugiert, und es wurden Fraktionen synaptischer Membranen erhalten. Die Membranfraktionen wurden 50 min lang mit ³H-Spiperon und verschiedenen Konzentrationen an Testverbindungen bei 25ºC inkubiert. Die Reaktion wurde beendet durch Filtration durch einen GF/B-Glasfilter unter Vakuum. Die Radioaktivität auf dem Filter wurde unter Verwendung eines Flüssigszintillationszählers gemessen. Die Dissoziationskonstanten (Ki) der Testverbindungen wurden gemäß folgender Gleichung bestimmt.

- IC&sub5;&sub0;: benötigte Konzentration an Testverbindung um 50% der ³H-Spiperon-Bindung zu hemmen, berechnet mit Hilfe eines Pseudo-Hill-Diagramms.
- L: ³H-Spiperon-Konzentration
- Kd: ³H-Spiperon-Dissoziationskonstante, berechnet mit Hilfe eines Scatchard-Diagramms.
- Die pKi-Werte wurde dann als negativer Logarithmus von Ki ausgedrückt. Dieser Wert gibt die Affinität der Testverbindung gegenüber Dopamin-D&sub2;-Rezeptoren an. Unspezifische Bindung von ³H-Spiperon wurde in Gegenwart von 10&supmin;&sup5; M Sulpirid bestimmt. Die Protein-Konzentration weurde mit Hilfe der Methode von Lowry bestimmt. Aus den in Tabelle 1-b gezeigten Ergebnissen geht hervor, daß die Affinitäten der Testverbindungen gegenüber Dopamin-D&sub2;-Rezeptoren niedriger sind als die von YM-12617. - Die Selektivität der Testverbindungen gegenüber α&sub1;-Adrenozeptoren im Vergleich zu Dopamin-D&sub2;-Rezeptoren wird gemäß folgender Gleichung bestimmt.
- α&sub1;-Adrenozeptor-Selektivität = 10(pA&sub2; - pKi)
- Aus den in Tabelle 2 gezeigten Ergebnissen geht hervor, daß die Selektivität der Testverbindungen gegenüber α&sub1;-Adrenozeptoren der von YM-12617 überlegen ist. Tabelle 2 α&sub1;-Adrenozeptor-Selektivität Testverbindung Beispiel (als HCl-Salz)
- Die folgenden Bezugsbeispiele und Beispiele sind nur der Erläuterung halber gegeben und sollten nicht als einschränkend ausgelegt werden.
- Eine Suspension von 10,0 g 5-Fluor-2-methoxyphenol, 52,9 g Ethylenbromid und 14,6 g Kaliumcarbonat in 50 ml N,N-Dimethylformamid wurde 11 h lang auf 80ºC erhitzt. Nach Abkühlen wurde die Reaktionsmischung in Wasser gegossen und mit Ethylacetat extrahiert. Der Extrakt wurde mit Wasser gewaschen, getrocknet und verdampft. Der Rückstand wurde unter Verwendung von 50% Chloroform/n-Hexan als Laufmittel an Kieselgel chromatographiert, um 11,6 g der gewünschten Verbindung als farbloses Öl zu ergeben.

- Eine Suspension von 15,0 g 1-(4-Hydroxyphenyl)-2-propanon, 24,6 g 1-Brompropan und 13,8 g Kaliumcarbonat in 100 ml N,N-Dimethylformamid wurde 4 h lang auf 65-70ºC erhitzt. Nach Abkühlen wurde die Reaktionsmischung in Wasser gegossen und mit Ether extrahiert. Der Extrakt wurde mit Wasser gewaschen, getrocknet und verdampft. DerRückstand wurde destilliert, um 15,2 g der gewünschten Verbindung als blaßgelbes Öl zu ergeben, Sdp. 136-138ºC (7 mmHg).

- Zu 125 g Chlorsulfonsäure wurden unter Rühren und Eiskühlung tropfenweise 25,0 g 4-Methoxyphenylaceton gegeben. Die Reaktionsmischung wurde 2 h lang bei Raumtemperatur gerührt, in Eiswasser gegossen und mit Ethylacetat extrahiert. Der Extrakt wurde mit Wasser gewaschen, getrocknet und verdampft, um 19,0 g brauner Kristalle zu ergeben. Zu einer Lösung der erhaltenen Kristalle in 200 ml Tetrahydrofuran wurden unter Rühren und Eiskühlung tropfenweise 25 ml Ammoniakwasser gegeben, wonach die Reaktionsmischung 2 h lang bei Raumtemperatur gerührt wurde. Das Lösungsmittel wurde entfernt, es wurde mit Wasser gewaschen, um 13,0 g der gewünschten Verbindung als blaßbraune Kristalle zu ergeben, die aus Methanol als blaßbraune Prismen umkristallisiert wurden, Schmp. 194-196ºC.

- (1) Zu einer Lösung von 14,9 g Trifluor-N-[2-(4-methoxyphenyl)ethyl]acetamid in 45 ml Dichloromethan wurden unter Rühren und Eiskühlung tropfenweise 21,1 g Chlorsulfonsäure gegeben. Die Reaktionsmischung wurde 2 h lang am Rückfluß gehalten, in Eiswasser gegossen und mit Chloroform extrahiert.
- Der Extrakt wurde mit Wasser gewaschen, getrocknet und verdampft, um 13,5 g eines gelben Öls zu ergeben. Zu 30 ml Ammoniakwasser wurden unter Rühren und Eiskühlung tropfenweise 13,5 g des gelben Öls in 20 ml Tetrahydrofuran gegeben. Die Reaktionsmischung wurde 30 min lang bei Raumtemperatur gerührt. Das Lösungsmittel wurde entfernt, und es wurde mit Wasser gewaschen, um 10,6 g 5-[2-(Trifluoracetylamino)ethyl]-2-methoxybenzolsulfonamid als farblose Kristalle zu ergeben, die aus Ethanol als farblose Kristalle umkristallisiert wurden, Schmp. 165-166ºC.

Die Reaktionsinischung wurde mit 13 ml Salzsäure angesäuert. Der Niederschlag wurde filtriert und aus Wasser umkristallisiert, um 4,22 g der gewünschten Verbindung als farblose Nadeln zu ergeben, Schmp. 263-266ºC.

Der Rückstand wurde destilliert, um 9,15 g 2-(5-Fluor-2-methoxyphenoxy)ethanol als farbloses Öl zu ergeben, Sdp. 138-140ºC (8 mmHg)

- (2) Zu einer Lösung von 9,00 g 2-(5-Fluor-2-methoxyphenoxy)ethanol in 4,5 ml Pyridin wurden unter Rühren und Eiskühlung tropfenweise 3,9 ml Thionylchlorid gegeben. Die Reaktionsmischung wurde 2 h lang auf 80-90ºC erhitzt, in 40 ml 10% Salzsäure gegossen und mit Chloroform extrahiert. Der Extrakt wurde mit Wasser gewaschen, getrocknet und verdampft. Der Rückstand wurde destilliert, um 50 g 2-(2-Chlorethoxy)-4-fluoranisol als farbloses Öl, Sdp. 118-120ºC (6 mmHg) zu ergeben, das zu Kristallen verfestigt wurde, Schmp. 35-38ºC.

Der Niederschlag wurde filtriert, um 10,4 g N-[2-(5-fluor-2-methoxyphenoxy)ethyl]phthalimid als farblose Kristalle zu ergeben, die aus Ethanol umkristallisiert als farblose Nadeln wurden, Schmp. 134,5-136ºC.

Nach dem Abkühlen wurde der Niederschlag abfiltriert und das Filtrat wurde verdampft. Der Rückstand wurde mit Wasser verdünnt und mit Chloroform extrahiert. Der Extrakt wurde mit Wasser gewaschen, getrocknet und verdampft. Der Rückstand wurde destilliert, um 3,81 g der gewünschten Verbindung als farbloses Öl zu ergeben, Sdp. 126-127ºC (9 mmHg)

- Eine Suspension von 10,8 g 5-Fluor-2-methoxyphenol und 12,6 g Kaliumcarbonat in 45 ml N,N-Dimethylformamid wurde 45 min lang auf 100ºC erhitzt. Dieser Reaktionsmischung wurden 18,5 ml Chloracetaldehyddiethylacetal und 12,7 g Kaliumiodid zugesetzt, und die Reaktionsmichung wurde 4 h lang auf 140ºC erhitzt. Nach dem Abkühlen wurde die Reaktionsmischung in Eiswasser gegossen und mit Ether extrahiert. Der Extrakt wurde mit wäßriger Natriumhydroxid-Lösung und Wasser gewaschen, getrocknet und verdampft. Der Rückstand wurde unter Verwendung von Chloroform als Laufmittel an Kieselgel chromatographiert, um 11,8 g der gewünschten Verbindung als farbloses Öl zu ergeben.

- (2) 2-(5-Fluor-2-methoxyphenoxy)acetaldehyd
- Einer Lösung von 23,0 g 2-(5-Fluor-2-methoxyphenoxy)acetaldehyddiethylacetal in 130 ml Aceton wruden 100 ml 10% wäßrige Oxalsäure zugesetzt und die Mischung wurde 2 h lang am Rückfluß gehalten. Nach dein Abkühlen wurde die Reaktionsmischung verdampft, und der Rückstand wurde mit Wasser verdünnt und mit Ether extrahiert. Der Extrakt wurde mit Wasser gewaschen, getrocknet und verdampft. Dem Rückstand wurde Isopropylether zugesetzt, und der Niederschlag wurde filtriert, um 4,99 g der gewünschten Verbindung als blaßbraune Kristalle zu ergeben, die aus einer Mischung von Isopropanol und Isopropylether als blaßbraune Kristalle umkristallisiert wurden, Schmp. 92,5-95ºC.

- Eine Lösung von 0,92 g 2-Methoxy-5-(2-oxopropyl)benzolsulfonamid und 0,70 g 2-(5-Fluor-2-methoxyphenoxy)ethylamin in 30 ml Methanol wurde 2 h lang am Rückfluß gehalten. Dieser Lösung wurden unter Eiskühlung 0,23 g Natriumborhydrid zugesetzt, und die Lösung wurde 1 h lang bei Raumtemperatur gerührt. Die Reaktionsmischung wurde verdampft und mit 30 ml 10% Salzsäure angesäuert. Der Niederschlag wurde filtriert und aus einer Mischung von Methanol und Ether umkristallisiert, um 0,69 g der gewünschten Verbindung als blaßgelbe Nadeln zu ergeben, Schmp. 258-261ºC.

- Eine Suspension von 1,50 g 5-(2-Aminoethyl)-2-methoxybenzolsulfonamid-hydrochlorid und 0,8 ml Triethylamin in 50 ml Methanol wurde 30 min lang auf 75ºC erhitzt. Der Reaktionsmischung wurde 1,00 g 2-(5-Fluor-2-methoxyphenoxy)acetaldehyd zugesetzt, und die Reaktionsmischung wurde 10 min lang auf 75ºC erhitzt. Unter Eiskühlung wurde die Lösung mit 0,45 g Natriumborhydrid versetzt, und die Lösung wurde 1 h lang bei Raumtemperatur gerührt. Die Reaktionsmischung wurde verdampft, mit 10% Salzsäure angesäuert und mit Ether gewaschen. Die wäßrige Schicht wurde mit Kaliumcarbonat alkalisch gemacht und mit Ethylacetat extrahiert. Der Extrakt wurde mit Wasser gewaschen, getrocknet und verdampft. Der Rückstand wurde mit einer Mischung aus Methanol und Ether verfestigt, und der Niederschlag wurde filtriert, um 0,46 g ger gewünschten Verbindung als blaßbraune Kristalle zu ergeben, die aus N,N-Dimethylformamid als blaßgelbe Kristalle umkristallisiert wurden, Schmp. 201-205ºC.

- Eine Suspension von 1,00 g R-(-)-5-(2-Aminopropyl)-2-methoxybenzolsulfonamid {[α]D²&sup8; -13,8º (c=1, MeOH), Schmp. 156,5- 160,5ºC (H&sub2;O)}, 11,00 g 2-(2-Bromethoxy)-4-fluoranisol und 0,20 g Kaliumiodid in 40 ml N,N-Dimethylformamid wruden 10 h lang auf 85ºC erhitzt. Nach dein Abkühlen wurde die Reaktionsmischung in Wasser gegossen, mit 10% wäßriger Natriumhydroxid-Lösung alkalisch gemacht und mit Ethylacetat extrahiert. Der Extrakt wurde mit Wasser gewaschen, getrocknet und verdampft. Der Rückstand wurde unter Verwendung von Chloroform/Methanol (9:1) als Laufmittel an Kieselgel chromatographiert, um 0,81 g der freien Base als farblose Kristalle zu ergeben, die aus Methanol als farblose Nadeln umkristallisiert wurden, Schmp. 144-145ºC.

- Eine Suspension von 25 g (±)-5-{2-[2-(5-Fluor-2-methoxyphenoxy)ethylamino]propyl}-2-methoxybenzolsulfonamid und 15,2 g L-10-Camphersulfonsäure {[α]D²&sup0; -20 23 (c=5, H&sub2;O)} in 450 ml Methanol wurde erhitzt, um Lösung zu ergeben. Die Lösung wurde auf Raumtemperatur abkühlen lassen, und der Niederschlag wurde abfiltriert.
- Das Filtrat wurde verdampft, und dieser Vorgang wurde dreimal für den Rückstand wiederholt. Der erhaltene Rückstand wurde aus Ethanol umkristallisiert, um die gewünschte Verbindung als farbloses Schuppen zu ergeben, Schmp. 184-184ºC.

- Zu dem so erhaltenen L-10-Camphersulfonsäure-Salz wurde wäßrige Natriumhydroxid-Lösung gegeben, und die Mischung wurde mit Chloroform extrahiert. Der Extrakt wurde mit Wasser gewaschen, getrocknet und verdampft. Der Rückstand wurde aus Ethanol umkristallisiert, um die freie Base als farblose Nadeln zu ergeben, die mit denen von Beispiel 35 übereinstimmten.
- R-(-)-5-{2-[2-(5-Fluor-2-methoxyphenoxy)ethylamino]propyl}- 2-methoxybenzolsulfonamid-D-10-camphersulfonat wurde unter Verwendung von (±)-5-{2-[2-(5-Fluor-2-methoxyphenoxy)ethylamino]propyl}-2-methoxybenzolsulfonamid und D-10-Camphersulfonsäure {[α]D²&sup0; +20-23º (c=5, H&sub2;O)} in gleicher Weise wie in Beispiel 36 beschrieben erhalten. Die erhaltenen Kristalle stimmten mit denen von Beispiel 34 überein. - Es sollte klar sein, daß diese Erfindung nicht beschränkt ist auf die genauen Einzelheiten der Arbeitsweise oder auf genaue Verbindungen, Zusammensetzungen oder gezeigte und beschriebene Verfahrensweisen, da dem Fachmann naheliegende Änderungen und Entsprechungen augenfällig sein dürften.
Claims (16)
1. Phenoxyethylamin-Derivat, ausgewählt aus den durch
Formel (I),
dargestellten, worin R&sub1; und R&sub5; niederes Alkyl bedeuten,
R&sub2; und R&sub3; gleich oder verschieden sein können und
jeweils Wasserstoff oder niederes Alkyl bedeuten, oder
den Rest eines 5- oder 6gliedrigen Ringsystems bedeuten
kann, das ein Stickstoff-, Sauerstoff- oder Schwefel-
Atom als Ringatom enthalten kann, R&sub4; Wasserstoff oder
niederes Alkyl bedeutet, und n eine ganze Zahl bedeutet,
ausgewählt aus 1 bis 3, sowie einschließlich dessen
pharmazeutisch annehmbare Säureadditionssalze.

umfassend die Umsetzung eines Phenoxyethylamins,
dargestellt durch die Formel (II)
worin R&sub5; die gleiche Bedeutung wie oben beschrieben hat,
mit einer Carbonyl-Verbindung, dargestellt durch die
Formel (III)
worin R&sub1;, R&sub2;, R&sub3;, R&sub4; und n die gleichen Bedeutungen wie
oben beschrieben haben, und anschließende Reduktion der
so erhaltenen Verbindung.
11. Verfahren zur Herstellung von
Phenoxyethylamin-Verbindungen, dargestellt durch die allgemeine Formel (I), und
ihrer pharmazeutisch annehmbaren Säureadditionssalze,
umfassend die Umsetzung eines Phenoxyacetaldehyds,
dargestellt durch die Formel (VIII)
worin R&sub5; die gleiche Bedeutung wie oben beschrieben hat,
mit einer Amin-Verbindung, dargestellt durch die Formel
(IX)
worin R&sub1;, R&sub2;, R&sub3;, R&sub4; und n die gleichen Bedeutungen wie
oben beschrieben haben, und anschließende Reduktion der
so erhaltenen Verbindung.
12. Verfahren zur Herstellung von
Phenoxyethylamin-Verbindungen, dargestellt durch die allgemeine Formel (I), und
ihrer pharmazeutisch annehmbaren Säureadditionssalze,
umfassend die Umsetzung einer
Fluorphenoxyalkan-Verbindung dargestellt durch die Formel (VI)
worin X für ein Halogen-Atom steht, und R&sub5; die gleiche
Bedeutung wie oben beschrieben hat, mit einer
Amin-Verbindung, dargestellt durch die Formel (IX)
worin R&sub1;, R&sub2;, R&sub3;, R&sub4; und n die gleichen Bedeutungen wie
oben beschrieben haben.
13. Verfahren zur Herstellung einer optisch aktiven
Phenoxyethylamin-Verbindung, dargestellt durch die allgemeine
Formel (I), oder deren pharmazeutisch annehmbarem
Säureadditionssalz, umfassend die Trennung eines Racemats
davon mittels eines Trennungsreagens.
14. α&sub1;-Blocker, vorteilhaft bei der Behandlung von
Hypertonie oder Dysurie, umfassend eine wirksame Menge einer
Phenoxyethylamin-Verbindung, dargestellt durch die
allgemeine Formel (I), oder eines pharmazeutisch
annehmbaren Säureadditionssalzes davon, und einen
pharmazeutisch annehmbaren Carrier oder ein solches
Verdünnungsmittel.
15. Pharmazeutische Zusammensetzung, vorteilhaft bei der
Behandlung von Hypertonie oder Dysurie, umfassend eine
wirksame Menge eines Phenoxyethylamin-Derivats,

zur Behandlung von Hypertonie oder Dysurie.
