Description
- Die vorliegende Erfindung betrifft die Verwendung von:
- A) Phospholipidgemische, enthaltend N-Acyl-Phosphatidylethanolamine (NAPEs); und/oder
- B) Phospholipidgemische, enthaltend N-Acyl-Ethanolamine (NAEs) zusammen mit Phosphatidsäuren (PAs) und/oder Lysophosphatidsäuren (LPAs), zur Herstellung eines Medikaments mit appetithemmender bzw. anorektischer Aktivität zur Behandlung von Fettleibigkeit und Übergewicht.
- Die vorliegende Erfindung betrifft neue Phosphobioflavonkomplexe von NAPE oder NAE plus PA und/oder LPA, mit einem oder mehreren Bioflavonoid(en).
- Es ist bekannt, dass N-Acyl-Ethanolamine (NAEs) und N-Acyl-Phosphatidylethanolamine (NAPEs) in vielen Nahrungsmitteln tierischen und pflanzlichen Ursprungs vorhanden sind (H. H. Schmid et al., 1990, Prog. Lipid Res., 29, 1–43) und besonders überreich in Nahrungsmitteln, wie Soda, Eiern und Schokolade sind (K. D. Chapman et al., 1993, Arch. Biochem. Biophys, 301, 21–23; E. Di Tomaso et al., 1996, Nature, 382, 677–678).
- In vivo werden die NAEs durch Hydrolyse eines NAPE-Moleküls gebildet, was zur Entstehung eines Gemischs von NAE und eines Moleküls Phosphatidsäure (PA) führt, das wiederum gemäß dem nachstehenden Schema 1 zu Lysophosphatidsäure (LPA) hydrolysiert werden kann.
- Die

EP 604806 - Die
EP 604 806 - NAEs waren seit einiger Zeit auch wegen ihrer interessanten pharmakologischen Eigenschaften bekannt: Es ist in vitro gezeigt worden, dass N-Arachidonoyl-Ethanolamin ein Cannabinoidrezeptoragonist ist (L. Kanus, 1993, J. Med. Chem., 36, 3032–3034); N-Palmitoyl-Ethanolamin besitzt, wenn es Ratten intraperitoneal verabreicht wird, antiinflammatorische und antianaphylaktische Aktivität (L. Facci et al., 1995, Proc. Natl. Acad. Sci. USA, 92, 3376–3380); N-Palmitoyl-Ethanolamin und N-Stearoyl-Ethanolamin haben sich in der pharmakologischen Behandlung von inflammatorischen Erkrankungen bzw. Störungen, die aus der Degranulation der Mastzellen resultieren, als nützlich erwiesen (
EP-A-0550006 ); sie inhibieren auch die Peroxidation der Mitochondrienmembranen in vitro (N. M. Gulaya et al., 1998, Chem. Phys. Lipids, 97, 49–54); N-Oleoyl-Ethanolamin (NOE) besitzt eine signifikante anorektische Wirkung in der Ratte, wenn es auf dem intraperitonealen Weg verabreicht wird (F. Rodriguez de Fonseca et al., 2001, Nature, 414, 209–212). Da gut bekannt ist, dass NAEs im Gastrointestinaltrakt leicht zu freien Fettsäuren und Ethanolamin hydrolysiert werden, wird ihre Aktivität auf dem oralen Weg nicht erwartet. - Die vorliegende Erfindung betrifft die Verwendung von:
- A) Phospholipidgemische, enthaltend N-Acyl-Phosphatidylethanolamine (NAPEs); und/oder
- B) Phospholipidgemische, enthaltend N-Acyl-Ethanolamine (NAEs) zusammen mit Phosphatidsäuren (PAs) und/oder Lysophosphatidsäuren (LPAs),
- Die Strukturformeln von NAE, PA und LPA sind im Schema 2 gezeigt, worin R1, R2 und R4 Acylreste von langkettigen Fettsäuren, insbesondere Reste von Palmitin-, Stearin-, Öl-, Linol-, konjugierter Linol-, Linolen-, Gamma-Linolen-, Eicosapentaen- und Docosahexansäure sind.
- Die Phospholipidgemische können in den Zusammensetzungen der Erfindung in Form ihrer Komplexe mit Bioflavonoiden vorliegen. Die Komplexe, hiernach mit „Phospobioflavonkomplexe” bezeichnet, sind ein weiterer Gegenstand der Erfindung.
- Komplexe von Phospholipiden, wie Lecithine, Phosphatidylcholin, Phosphatidylethanolamin und Phosphatidylserin, mit einer Anzahl von Pflanzenextrakten sind beschrieben worden (
US 4963527 ,US 4895839 ,EP 283713 - Die Komplexe, gebildet durch Aggregation der aktiven bzw. wirksamen Phospholipidkomponenten (NAPE und/oder NAE plus PA und/oder NAE plus LPA) mit einem oder mehreren Typ(en) von Bioflavonoiden, können erhalten werden, indem ein trockener Phospholipidrückstand unter starkem Rühren für einige Minuten bei einer Temperatur, vorzugsweise zwischen 40° und 65°C, in einer wässrigen alkoholischen Lösung (Alkohol vorzugsweise zwischen 70 und 90%), gepuffert auf einen sauren pH (pH vorzugsweise zwischen 3 und 5), enthaltend eine Fraktion von einem oder mehreren Typ(en) von Bioflavonoiden, vorzugsweise in einem prozentualen Anteil zwischen 0,5 und 15 Gew.-% der wässrigen alkoholischen Lösung, suspendiert wird. Wenn das Rühren unterbrochen wird, wird Ethanol partiell aus der resultierenden Emulsion unter Vakuum verdampft und anschließend erfolgt Entwässerung durch Sprühtrocknung, um einen trockenen körnigen Rückstand von Phosphobioflavonkomplexen herzustellen.
- Beispiele für Bioflavonoide, die verwendet werden können, um diese Phosphobioflavonkomplexe herzustellen, umfassen:
- a) einfache Polyphenole, wie Zimt-, Kumarin-, Kaffee- und Ferulasäure;
- b) Flavone, wie Hesperidin, Naringenin und Taxifolin;
- c) Flavonole, wie Kaempferolglycosid, Quercetin, Quercetinglycosid, Myricetin und Myricetinglycosid;
- d) Isoflavone, wie Genistein und Daidzein;
- e) Proanthocyanidine, wie Procyanidin B1, Procyanidin B2, Procyanidin B3 und Procyanidin C-1;
- f) Anthocyanidine, wie Pelargonidin, Delphinidin, Malvidin und Petunidin;
- g) Catechine, wie Epicatechin, Epicatechingallat, Epigallocatechin, Catechine und Gallocatechine;
- h) Tannine.
- Wie erwähnt sind Moleküle von NAPE, NAE, PA und LPA natürlicherweise in den Lipidfraktionen von vielen Nahrungsmitteln, die normalerweise in der Humanernährung verwendet werden (Sojalecithine, Eier, Kakao, Fleisch, ölige Extrakte aus verschiedenen Samen usw.), vorhanden und können bis zu verschiedenen Graden der Reinheit gemäß herkömmlicher Verfahren leicht extrahiert und isoliert werden. Alternativ können die NAPE- und NAE-Moleküle durch Synthesen gemäß chemischer Prozesse, die seit einiger Zeit bekannt sind, erhalten werden.
- NAE kann aus Ethanolamin und der entsprechenden Fettsäure hergestellt werden, zum Beispiel gemäß den Verfahren, die beschrieben sind in:
- – Roc E. T. et al. (1952), J. Am. Chem. Soc., 74, 3442
- – Chandrakumar N. S. et al. (1982), Biochim. Biophys. Acta, 711, 357.
- NAPE kann aus Phosphatidylethanolamin und dem entsprechenden Fettsäurechlorid oder -anhydrid gemäß den Verfahren hergestellt werden, die beschrieben sind in:
- – Schmid P. C. et al. (1988), J. Biol. Chem., 288 (6), 9802
- – Epps D. E. et al. (1980), Biochim. Biophys. Acta, 618, 420
- –
GB 2051069 B - Ein weiteres Verfahren zur Herstellung von NAPE mittels des Enzyms Phospholipase D, offenbart in der
US 4783402 , ist im unten stehenden Schema erläutert: Schema 2 worin R1, R2 und R4 für die Alkylkette von gesättigten, einfach oder mehrfach ungesättigten Fettsäuren mit 12–22 C-Atomen stehen;
R3 für einen Cholin-, Ethanolamin-, Inositol-, Glycerin-, Serinrest steht. - Die therapeutisch wirksamen Dosen von Zubereitungen, basierend auf NAPE und/oder NAE plus PA und/oder LPA variieren:
- a) im Falle von NAPE von 0,5 bis 50 mg und vorzugsweise von 1 bis 10 mg/Tag pro kg Körpergewicht;
- b) im Falle von NAE plus PA und/oder LPA von 0,5 bis 100 mg, vorzugsweise von 2 bis 20 mg/Tag/kg Körpergewicht. In diesem Gemisch von NAE + PA und/oder LPA kann der prozentuale Anteil von NAE zwischen 1 und 70% und vorzugsweise zwischen 25 und 50% der gesamten miteinander gemischten Lipide variieren.
- Die Zusammensetzungen der Erfindung können auch andere Nahrungs- bzw. Nährstoffkomponenten („nutritional components”) enthalten, die die therapeutischen Eigenschaften und Vorteile von NAPE und/oder den Gemischen von NAE + PA und/oder LPA weiter verwirklichen. Beispiele für diese Komponenten sind:
- a) Vitamine und vitaminähnliche Faktoren, wie Vitamin E, Vitamin C, β-Carotine, Vitamin A, Vitamin D, Liponsäure und CoQ;
- b) Extrakte von Gemüsen und/oder Medizinalpflanzen, basierend auf Mono- und Diterpenen, Saponinen und Phytosterolen;
- c) Proteine, Peptide oder Aminosäuren und deren Derivate, wie Glutathion, Carnosin, Arginin, Glutamin, Carnitin, Creatin und Taurin;
- d) Spurenelemente und Mineralsalze, wie Ca, Mg, Cr, Se, Va, Zn und Cu;
- e) Gemische von natürlichen amphiphilen Detergenzien, wie Phospholipide und Lysophospholipide; Glycolipide; amphiphile Proteine; Mono- und Diglyceride; Gallensäuren oder -salze, die dazu befähigt sind, NAPE und/oder Gemische von NAE + PA und/oder LPA in Lipidemulsionen verschiedener Typen, die dabei helfen, deren Absorption und Bioverfügbarkeit in vivo zu erhöhen, zu inkorporieren.
- Die aktiven Komponenten, gelagert als entwässerte bzw. dehydrierte Granulate oder Pulver, können als solche oder in Form von wässrigen oder öligen Lösungen verwendet werden, um verschiedene galenische Zubereitungen, wie Gelatinekapseln, Tabletten, Dragees, (Portions-)Beutel, brausende und nicht-brausende Oblatenkapseln („chachets”), Kaugummi usw. herzustellen.
- Die aktiven Komponenten in Form von entwässerten Granulaten oder Pulvern können auch verwendet werden, um verschiedene funktionelle Nahrungsmittel herzustellen:
- a) gemischt mit Ölen, um verschiedene Dressings, Soßen, Cremes, Mayonnaise usw. herzustellen;
- b) gemischt mit Mehl, um Brot, Nudeln bzw. Pasta, Cracker, Biskuits bzw. Kekse bzw. Brötchen und andere gebackene Produkte herzustellen;
- c) zugegeben zu Fruchtsäften und Fruchtsaftgetränken bzw. -konzentraten bzw. Fruchtmus („squashes”), Mineralwässern, Softgetränken und anderen Getränken;
- d) zugegeben zu Milch und Derivaten davon (Joghurt, Flans, Ricotta und Käse).
- Die pharmazeutischen oder diätetischen Zusammensetzungen der Erfindung haben sich als überraschend aktiv bzw. wirksam erwiesen beim:
- a) Kontrollieren von Übergewicht und folglich Verringern der Risiken, die mit Übergewicht und Fettleibigkeit verbunden sind;
- b) Verbessern der Funktionalität der Mitochondrien und der Produktion von Zellenergie;
- c) Erhöhen der antioxidativen Abwehrmechanismen („antioxidant defences”) in den verschiedenen Geweben;
- d) Verbessern der „Fluidität” der Zellmembranen und folglich der Funktionalität der Membranproteine (Enzyme, Rezeptoren, Transporter von essentiellen Nährstoffen und Spurenelementen usw.).
- Die Zubereitungen der Erfindung können daher als Adjuvantien bei der Behandlung von Alterung und vielen Stoffwechselerkrankungen, die damit verbunden sind (Fettleibigkeit und Übergewicht; Diabetes; degenerative Gehirnerkrankungen, wie Alzheimer-Krankheit, Parkinson-Krankheit und senile Demenz; Stress, Depression; Tumore; Menopausensyndrome; Osteoporose; Prostatahypertrophie; Hautalterung; Panniculopathie (Cellulitis); und Alopezie), verwendet werden, möglicherweise in Kombination mit bekannten Wirkstoffen oder Nahrungsergänzungsmitteln.
- Die Erfindung betrifft daher auch die Verwendung von Phospholipidgemischen, enthaltend
- A) N-Acyl-Phosphatidylethanolamine (NAPEs); und/oder
- B) Phospholipidgemische, enthaltend N-Acyl-Ethanolamine (NAEs), zusammen mit Phosphatidsäuren (PAs) und/oder Lysophosphatidsäuren (LPAs)
- Die Erfindung wird in den folgenden Beispielen detaillierter erläutert.
- Beispiel 1
-
- 98 g N-Linoleoyl-Phosphatidylethanolamin + 1 g D-α-Tocopherol + 1 g Liponsäure.
- Die verschiedenen Komponenten werden in 10 Volumina Chloroform:Methanol (2:1, Vol/Vol) gelöst und gemischt. Das Lösungsmittel wird unter Vakuum verdampft und der resultierende trockene Rückstand wird in einer wässrigen Lösung, die auf physiologischem pH gepuffert ist, resuspendiert, um ein wässriges Gemisch einer Phospholipidemulsion, enthaltend die aktive Komponente (N-Linoleoyl-Phosphatidylethanolamin), zu bilden. Das wässrige Gemisch kann eingefroren und entwässert werden, um einen trockenen Rückstand der aktiven Phospholipidkomponente zu erhalten.
- Beispiel 2
-
- 20 g N-Eicosapentaenoylethanolamin + 60 g Phosphatidsäure + 80 g Soja-Phosphatidylcholin + 1 g D-α-Tocopherol + 1 g Liponsäure.
- Die verschiedenen Verbindungen werden in Chloroform-Methanol gelöst und behandelt, wie in Beispiel 1 beschrieben, um ein wässriges Gemisch einer Phospholipidemulsion, enthaltend die aktiven Komponenten (N-Eicosapentaenoylethanolamin und Phosphatidsäure), zu erhalten. Das wässrige Gemisch kann eingefroren und entwässert werden, um einen trockenen Phospholipidrückstand der aktiven Komponenten zu erhalten, wie in Beispiel 1 beschrieben.
- Beispiel 3
-
- 20 g N-Linolenoylethanolamin + 40 g Lysophosphatidsäure + 1 g D-α-Tocopherol + 1 g Liponsäure.
- Die verschiedenen Verbindungen werden in Chloroform-Methanol gelöst und behandelt, wie in Beispiel 1 beschrieben, um ein wässriges Gemisch einer Phospholipidemulsion, enthaltend die aktiven Komponenten (N-Linolenoylethanolamin und Phosphatidsäure), zu erhalten. Das wässrige Gemisch kann eingefroren und entwässert werden, um einen trockenen Phospholipidrückstand der aktiven Komponenten zu erhalten, wie in Beispiel 1 beschrieben.
- Beispiel 4
-
- 20 g N-gamma-Linolenoyl-Phosphatidylethanolamin + 80 g eines Gemisches von Lysophospholipiden (45% Lysophosphatidylcholin + 35% Lysophosphatidylethanolamin + 20% Lysophosphatidylinositol) + 1 g D-α-Tocopherol + 1 g Liponsäure.
- Die verschiedenen Verbindungen werden in Chloroform-Methanol gelöst und behandelt, wie in Beispiel 1 beschrieben, um ein wässriges Gemisch einer Phospholipidemulsion, enthaltend den aktiven Bestandteil (N-gamma-Linolenoyl-Phosphatidylethanolamin), zu erhalten. Das wässrige Gemisch kann eingefroren und entwässert werden, um einen trockenen Phospholipidrückstand des aktiven Bestandteils zu erhalten, wie in Beispiel 1 beschrieben.
- Beispiel 5

- 20 g eines trockenen Phospholipidrückstands, erhalten wie in den Beispielen 1–4 oben beschrieben + 200 g einer öligen Lösung (Olivenöl, Soja-, Maiskeim-, Sonnenblumen-, Borretsch-, schwarze Johannisbeer-, Fisch- oder Meeresalgenöl(e) oder Gemische davon).
- 20 g trockener Phospholipidrückstand wird langsam in 200 g öliger Lösung unter langsamem, kontinuierlichem Rühren gelöst. Die Phospholipide der trockenen Rückstände werden in den öligen Lösungen restrukturiert, um eine in Öl dispergierte mizelläre Organisation, enthaltend die aktiven Komponenten, zu bilden.
- Beispiel 6
- 100 g eines trockenen Phospholipidrückstands von N-Docosahexanoyl-Phosphatidylethanolamin, erhalten wie in Beispiel 1 beschrieben, wird unter starkem Rühren für 5 Minuten bei 45°C in 900 ml einer wässrigen alkoholischen Lösung (75% Alkohol), gepuffert auf pH 4,5, enthaltend 5 Gew.-% Grünteecatechine und Epicatechine, resuspendiert. Die resultierende Emulsion wird dann auf Raumtemperatur abgekühlt und durch Sprühtrocknung entwässert, wodurch ein trockener körniger Rückstand von Phosphobioflavonkomplexen von N-Docosahexanoyl-Phosphatidylethanolamin und Grünteecatechinen gebildet wird.
- Beispiel 7
- 50 g N-Linolenoylethanolamin und 50 g Lysophosphatidsäure (CLPA) werden langsam unter starkem Rühren bei 60°C zu 900 ml einer wässrigen alkoholischen Lösung (85% Alkohol), gepuffert auf pH 4,0, enthaltend 10 Gew.-% eines Gemisches von Catechinen, Epicatechinen und Proanthocyanidinen, die aus Traubenkernen extrahiert sind, gegeben und darin für 10 Minuten emulgiert. Wenn das Rühren angehalten wird, wird die resultierende Emulsion auf Raumtemperatur abgekühlt und durch Sprühtrocknung entwässert, um einen trockenen körnigen Rückstand von Phosphobioflavonkomplexen von N-Linolenoylethanolamin und Traubenkernbioflavonoiden zu bilden.
- Pharmakologische und/oder diätetische Tests
- Eine Reihe von experimentellen Tests an Ratten und klinische Tests an Menschen sind durchgeführt worden, um die pharmakologischen und/oder diätetischen Eigenschaften der Zusammensetzung der Erfindung zu studieren.
- In den experimentellen Tests erhielten die Ratten eine hochkalorienreiche, hochtriglyceridreiche, hochcholesterinreiche Nahrung bzw. Diät. Die folgenden Parameter wurden nach 20-tägiger Behandlung bewertet:
- 1) Wirkung der Zusammensetzungen auf die Lipoperoxidspiegel in Plasma, in der Leber, im Gehirn und im Herzen;
- 2) Wirkung der Zusammensetzungen auf Änderungen im Körpergewicht;
- 3) Wirkung der Zusammensetzungen auf Änderungen der Membranfluidität von Erythrozytenghosts und Blutplättchen im Plasma („plasma platelets”);
- 4) Wirkung der Zusammensetzungen auf die Funktionalität der Lebermitochondrien, bewertet durch Messung: a) des O2-Verbrauchs; b) des reduzierten Glutathions; c) des Potentials der Mitochondrienmembranen;
- 5) Wirkung der Zusammensetzungen auf die Plasmaspiegel von Gesamtcholesterin und HDL-Cholesterin;
- 6) Wirkung der Zusammensetzungen auf die Plasmaspiegel der Gesamttriglyceride.
- 80 männliche Ratten mit einem Gewicht von jeweils 150–200 g, wurden verwendet. Die Tiere wurden in acht Gruppen von zehn Tieren aufgeteilt:
- 1. Gruppe: Kontrolle (C); 10 Tiere (Kontrolle zum Zeitpunkt 0) wurden verwendet wie sie sind und 10 erhielten eine hochkalorienhaltige, hochfetthaltige, hochcholesterinhaltige Standardnahrung für 20 Tage, bestehend aus: Casein: 20%; Gemisch von Spurenelementen und Mineralsalzen: 3,5%; Gemisch von Vitaminen: 0,1%; Cholinbitartrat: 0,2%; Cellulose: 2%; Cholesterin: 0,5%; Natriumcholat: 0,25%; Saccharose: 58,44%; Schmalz: 10,0% und Olivenöl: 4,9%.
- 2. Gruppe: behandelt mit N-Oleoyl-Ethanolamin als solches (NOE); die Tiere erhielten für 20 Tage die gleiche Nahrung wie die Kontrollen, abgesehen davon, dass 50 mg NOE die gleiche Menge an Olivenöl ersetzten (verwendetes Olivenöl: 4,85%).
- 3. Gruppe: behandelt mit N-Oleoyl-Phosphatidylethanolamin, zubereitet wie in Beispiel 1 beschrieben (NOPE); die Tiere erhielten für 20 Tage die gleiche Nahrung wie die Kontrollen, abgesehen davon, dass 50 mg NOPE (zubereitet wie in Beispiel 1 beschrieben) die gleiche Menge an Olivenöl ersetzten (verwendetes Olivenöl: 4,85%).
- 4. Gruppe: behandelt mit N-Oleoyl-Ethanolamin + Phosphatidsäure, zubereitet wie in Beispiel 2 beschrieben: (NOE + PA); die Tiere erhielten für 20 Tage die gleiche Nahrung wie die Kontrollen, abgesehen davon, dass 400 mg der in Beispiel 2 beschriebenen Zubereitung (enthaltend ~ 50 mg NOE und 150 mg PA) die gleiche Menge an Olivenöl ersetzten (verwendetes Olivenöl: 4,50%).
- 5. Gruppe: behandelt mit N-Oleoyl-Ethanolamin + Lysophosphatidsäure, zubereitet wie in Beispiel 3 beschrieben: (NOE + LPA); die Tiere erhielten für 20 Tage die gleiche Nahrung wie die Kontrollen, abgesehen davon, dass 150 mg der in Beispiel 3 beschriebenen Zubereitung (enthaltend ~ 50 mg NOE und 100 mg LPA) die gleiche Menge an Olivenöl ersetzten (verwendetes Olivenöl: 4,75%).
- 6. Gruppe: behandelt mit „Phosphobioflavonkomplexen” von N-Oleoyl-Phosphatidylethanolamin und Grünteebioflavonen (B.F.), zubereitet wie in Beispiel 6 beschrieben (NOPE + B.F.). Die Tiere erhielten für 20 Tage die gleiche Nahrung wie die Kontrollen, abgesehen davon, dass 50 mg NOPE und 25 mg B.F. (entsprechend ~ 75 mg der in Beispiel 6 beschriebenen Zubereitung) die gleiche Menge an Olivenöl ersetzten (verwendetes Olivenöl: 4,825%).
- 7. Gruppe: behandelt mit Grünteebioflavonen (B.F.). Die Tiere erhielten für 20 Tage die gleiche Nahrung wie die Kontrollen, abgesehen davon, dass 25 mg B.F. die gleiche Menge an Olivenöl ersetzten (verwendetes Olivenöl: 4,875%).
- Wenn die Membranfluidität der Erythrozytenghosts und Blutplättchen im Plasma gemessen wird, wird TMA-DPH gemäß dem von Caimi F. et al., 1999, Thromb. Hoemost., 82, S. 149 beschriebenen Verfahren als die Fluoreszenzsonde verwendet.
- Malonyldialdehyd wird gemäß der von K. Yagi et al., 1982, in „Lipid Peroxides in Biology and Medicine”, Academic Press, New York, 99, 324–340 beschriebenen Verfahrensweise getestet.
- Der/Das hepatozelluläre O2-Verbrauch, Mitochondrienmembranpotential und Gehalt an reduziertem Glutathion werden gemäß den von T. M. Hagen et al., 1999, FASEB J., 13, 99, 411 beschriebenen Verfahren getestet.
- Die in den Tabellen I, II, III und IV dargelegten Daten zeigen, dass eine Verabreichung von Zusammensetzungen, enthaltend die aktiven Komponenten (NOPE; NOE + PA; NOE + LPA; NOPE + B.F.):
- 1) die Membranfluidität von Ghost und Blutplättchen wiederherstellt;
- 2) die antioxidativen Abwehrmechanismen von Plasma, Leber, Gehirn und Herz verbessert;
- 3) übermäßige Erhöhungen des Körpergewichts limitiert;
- 4) übermäßige Erhöhungen von Plasmacholesterin- und Triglyceridspiegeln limitiert;
- 5) die Funktionalität der Mitochondrien verbessert.
- Diese Wirkungen, die durch orale Verabreichung der gemäß der Erfindung hergestellten Formulierungen (NOPE; NOE + PA; NOE + LPA; NOPE + B.F.) erhältlich sind, sind immer statistisch signifikant. Es ist wichtig zu bemerken, dass kein statistisch signifikanter Vorteil erhalten werden kann, indem äquivalente orale Dosen von N-Oleoylethanolaminen als solche verabreicht werden.
- Die oben dargelegten Daten zeigen die überraschende Synergie der Wirkung, die zwischen NAPE und/oder NAE + PA und den verschiedenen Bioflavonoidmolekülen beobachtet wird; die durch Verabreichung der „Phosphobioflavonkomplexe” von NAPE (siehe Daten, dargelegt in den Tabellen I, II, III und IV) und NAE plus PA und/oder LPA erhältlichen therapeutischen Ergebnisse sind immer weit höher als die Summe der Vorteile, die bei einzelnen, getrennten Verabreichungen von äquivalenten Dosen von NAPE (oder NAE) und Bioflavonoiden erhältlich sind.
- In allen Diätbehandlungstests, die am Menschen durchgeführt wurden, ergaben die Wirkungen, die durch orale (Verabreichung) der durch die Erfindung beanspruchten Formulierungen (NAPE; NAE + PA; NAE + LPA; NAPE + B.F. und NAE plus PA und/oder LPA + B.F.) erhältlich waren, immer im hohen Maße signifikante Ergebnisse und Vorteile, sowohl bei der Verhinderung von biologischen Anzeichen des Alterns (Verbesserung der Mitochondrienaktivität, bessere Membranfluidität, Verbesserung von antioxidativen Abwehrmechanismen in Plasma und limitierte Gewichtserhöhung) als auch der Verbesserung der klinischen Parameter, die in Bezug auf die Verhinderung des Alterns getestet wurden, und vieler der Stoffwechselerkrankungen, die damit assoziiert sind. Es ist bemerkenswert, dass auch bei Menschen kein signifikanter Vorteil durch Verabreichung äquivalenter oraler Dosen von N-Oleoylethanolamin als solches erhalten werden kann.

Claims (3)
- Verwendung von Phospholipidgemischen, enthaltend: a) N-Acyl-Phosphatidylethanolamine (NAPE); und/oder b) Phospholipidgemische, enthaltend N-Acyl-Ethanolamine (NAE) zusammen mit Phosphatidsäuren (PA) und/oder Lysophosphatidsäuren (LPA), zur Herstellung eines Medikaments mit anorektischer Aktivität oder zur Behandlung von Fettleibigkeit und Übergewicht.
- Phosphobioflavonkomplexe von Bioflavonoiden mit einem aktiven Phospholipidkomponent, ausgewählt aus N-Acyl-Phosphatidylethanolaminen (NAPE) und/oder N-Acyl-Ethanolaminen (NAE) plus Phosphatidsäure (PA) und/oder N-Acyl-Ethanolaminen plus Lysophosphatidsäure (LPA).
- Zusammensetzung, umfassend die Phosphobioflavonkomplexe nach Anspruch 2.
