Claims (3)
1. Verfahren zur Synthese einer N-acetylierten Verbindung, ausgehend von Heparinen, die teilweise
oder vollständig N-desulfatiert sind, beruhend auf Titration der freien Aminogruppen in den
N-desulfatierten Heparinen, umfassend die Zugabe einer stochxometrischen Menge Acetanhydrid in einem
Verhältnis von 1 bis 1,1 Molen Acetanhydrid pro Mol freie Aminogruppen bei einem pH, der zwischen 9,3 und
9,5 liegt, und einer Temperatur zwischen 25 und 3O0C.
2. Produkt, erhalten aus Heparin durch das in Anspruch beschriebene Verfahren, das die folgenden
analytischen Charakteristika aufweist:
- Antigerinnungsaktivität (ABTT-Anti-X): weniger als 10 IU/mg;
- N-Acetylgehalt (ausgedrückt als CH3CO-): 3 bis 7 %
G/G, entsprechend einem N-Acetylierungsgrad von 40
bis 90 %;
- Uronsäuren (nach Hydrolyse) : 31 +. 1 %;
- Hexosamine (nach Hydrolyse): 32 + 1 %;
- organisches Sulfat (als S) : 8 ± 1 %;
- Molverhältnis Uronsäure:Hexosamin:S:Acetyl:
1:1:1,5-1,7:0,4-0,9;
- spezifische optische Drehung: > + 35°.
3. Pharmazeutische Zusammensetzungen, enthaltend das in
Anspruch 2 definierte Produkt als einzigen aktiven Bestandteil mit antithrombotischen und
fibrinolytischen Eigenschaften, mit niedriger Gerinnungsaktivität, geeignet für orale Verwendung
und Injektionsverwendung.
