Description
- Die vorliegende Erfindung betrifft kardiovaskulär aktive Verbindungen und insbesondere Derivate von N-Propyldopamin und ihre therapeutische Verwendung. Die EP-A 0 062 596 offenbart Aminoethoxyphenyl-Derivate, die therapeutisch nützliche kardiovaskuläre Mittel darstellen.
- Gegenstand der vorliegenden Erfindung sind Verbindungen der Formel:

- R und R&sub1;, die gleich oder verschieden sein können, Wasserstoffatome bedeuten oder Acylgruppen, die abgeleitet sind von wahlweise substituierten aliphatischen, aromatischen oder heteroaromatischen Carbonsäuren, von wahlweise substituierten Carbamin- oder Kohlensäuren, oder von Phosphorsäure der Formel
- worin R&sub4; ein Wasserstoffatom, eine wahlweise mit ein oder mebreren Gruppen, die ausgewahlt sind aus Hydroxy, Alkoxy, Acyloxy, Amino, Carboxy und Alkoxycarbonyl, substituierte C&sub1;-C&sub6;-Alkylgruppe, oder eine Phenylgruppe bedeutet, unter der Bedingung, daß wenn eine der Gruppen R und R&sub1; eine von Phosphorsäure abgeleitete Acylgruppe darstellt, die andere Gruppe ein Wasserstoffatom bedeutet;
- R&sub2; und R&sub3;, die gleich oder verschieden sein können, ein Wasserstoff- oder ein Halogenatom, eine Alkyl- oder eine Alkoxygruppe bedeuten;
- und ihre Salze mit pharmazeutisch annehmbaren Säuren.
- Die Verbindungen der Formel I, worin R und R&sub1; Wasserstoffatome darstellen, besitzen gefäßerweiternde und dopaminerge Wirkung und können in der kardiovaskulären Therapie als blutdrucksenkendes Mittel und zur Behandlung von Herzinsuffizienz eingesetzt werden.
- Die Verbindungen der Formel I, worin ein oder beide Substituenten R und R&sub1; von Wasserstoff verschieden sind, sind geeignete Vorläufer der aktiven Verbindungen und können für dieselben therapeutischen Zwecke verwendet werden.
- Der Grund für den obigen Vorbehalt in der Bedeutung von R und R&sub1; liegt in der Tatsache, daß nur die Mono-O-phosphatester geeignete Vorläufer der wirksamen Verbindungen sind.
- Wenn nicht anders ausgeführt bedeuten Alkyl oder Alkoxy in den Verbindungen der Formel I einen gerad- oder verzweigtkettigen Alkyl- oder Alkoxyrest mit 1 bis 4 Kohlenstoffatomen, wobei spezifische Beispiele Methyl, Ethyl, n-Propyl, i-Propyl, n-Butyl, i-Butyl, sec.-Butyl, tert.-Butyl, Methoxy, Ethoxy, n-Propoxy, i-Propoxy, und tert.-Butoxy umfassen; bevorzugt sind Methyl und Methoxy.
- Halogenatome umfassen Fluor, Chlor, Brom oder Iod, wobei die ersten drei bevorzugt sind.
- Von aliphatischen Carbonsäuren abgeleitete Acylgruppen bedeutet Acylreste, die von gerad- oder verzweigtkettigen aliphatischen Carbonsäuren mit 1 bis 6 Kohlenstoffatomen abgeleitet sind, wobei spezifische Beispiele die Acylgruppen sind, die von folgenden Säuren abgeleitet sind: Ameisensäure, Essigsäure, Propionsäure, Buttersäure, Isobuttersäure, Valeriansäure und Pivalinsäure; Acylgruppen von aromatischen oder heteroaromatischen Carbonsäuren leiten sich von wahlweise substituierter Benzoesäure oder Picolinsäure (2-, 3- oder 4- Picolinsäure), Pyrrolcarbonsäure, Isoxazolcarbonsäure und Chinolincarbonsäure ab. Sie können beispielsweise durch Alkyl oder Halogen substituiert sein. Spezifische Beispiele umfassen Benzoyl, 2-Pyridincarbonyl, 3-Pyridincarbonyl, 4- Pyridincarbonyl, 2-Chlorbenzoyl, 4-Chlorbenzoyl, 2-Methylbenzoyl, 3-Methylbenzoyl, 4-Methylbenzoyl, 2,4-Dimethylbenzoyl, 4-Nitrobenzoyl und 4-Isobutyrylbenzoyl.
- Wenn R und R&sub1; substituierte aliphatische Säuren sind, sind die bevorzugten Substituenten Phenyl, ein, zwei oder drei Halogenatome, oder Alkoxy.
- Für den Fall, daß R und R&sub1; substituierte aromatische Säuren sind, sind die bevorzugten Substituenten ein, zwei oder drei Halogenatome, Alkoxy-, Nitro- und von aliphatischen Carbonsäuren abgeleitete Acylgruppen.
- Bevorzugte Substituenten für Carbamin- und Kohlensäuren sind Alkyl und Phenyl.
- Bei den Bedeutungen von R&sub4; bedeutet Acyloxy eine Acyloxygruppe mit 2 bis 4 Kohlenstoffatomen, wobei ein bevorzugtes Beispiel Acetoxy ist. Alkoxycarbonyl bedeutet eine Alkoxycarbonylgruppe mit 1 bis 4 Kohlenstoffatomen in der Alkoxyhälfte, wobei spezifische Beispiele Methoxycarbonyl und Ethoxycarbonyl sind. Pharmazeutisch annehmbare Säuren, die zur Herstellung von Salzen der Verbindungen der Formel I verwendet werden können, umfassen Salzsäure, Bromwasserstoffsäure, Iodwasserstoffsäure, Salpetersäure, Schwefelsäure, Phosphorsäure, Essigsäure, Benzoesäure, Maleinsäure, Fumarsäure, Bernsteinsäure, Weinsäure, Zitronensäure, Asparaginsäure und Methansulfonsäure.

- Die Herstellung der Verbindungen der Formel I kann nach unterschiedlichen alternativen Methoden erfolgen, die im folgenden beschrieben werden.
- Nützlich Zwischenprodukte für einige dieser Syntheseverfahren sind N-Propyldopamin und seine Derivate (II), in denen die aromatischen Hydroxygruppen, sofern gewünscht, geschützt sind,
- worin
- R&sub5; und R&sub6;, die gleich oder verschieden sein können, ein Wasserstoffatom oder eine
- Vorzugsweise sind R&sub5; und R&sub6; gleich, wenn das Ziel der Synthese die Herstellung von Verbindungen der Formel I ist, worin R und R&sub1; Wasserstoffatome sind, sie können aber verschieden sein, wenn die Synthese auf die Herstellung von Verbindungen der Formel I gerichtet ist, worin einer der Reste R und R&sub1; von Wasserstoff verschieden ist.
- Am Schluß des Syntheseverfahrens oder bevor man eine Reaktion an einem Zwischenprodukt durchführt, werden die wahlweisen Schutzgruppen mit üblichen Methoden entfernt.
- Wenn beispielsweise ein oder beide Reste R&sub5; und R&sub6; Methylgruppen sind, kann die Entfernung der Schutzgruppen von der Hydroxygruppe durch Reaktion mit Halogenwasserstoffsäuren oder mit Bortribromid erfolgen; wenn ein oder beide Reste R&sub5; und R&sub6; Benzylgruppen sind, kann die Entfernung der Schutzgruppen von der Hydroxygruppe durch Hydrogenolyse erfolgen.
- Die Verbindungen der Formel II sind nützliche Ausgangsverbindungen für drei verschiedene Syntheseverfahren:
- a) Das erste Verfahren besteht in der Reaktion zwischen Verbindung II und einem Halogenid einer geeigneten Phenoxyessigsäure der Formel
- worin
- R&sub2; und R&sub3; die Bedeutung wie in Formel I haben und X ein Chlor- oder Bromatom bedeutet.
- Die Reaktion erfolgt in einem inerten Lösungsmittel in Gegenwart einer Base, die als Akzeptor für Halogenwasserstoffsäure fungiert, beispielsweise einem Alkalicarbonat oder -bicarbonat, oder einem tertiären Amin (Triethylamin oder Pyridin). In letztgenanntem Fall kann das Amin auch als Lösungsmittel dienen. Durch obige Reaktion erhält man die Zwischenprodukte der Formel

- R&sub2;, R&sub3;, R&sub5; und R&sub6; die obige Bedeutung haben.
- Die Reduktion des Amidcarbonyls in den Verbindungen der Formel IV, der gegebenenfalls die Entfernung der Schutzgruppen R&sub5; und R&sub6; von den Hydroxygruppen vorangeht oder nachfolgt, liefert die Verbindungen der Formel I, worin R und R&sub1; Wasserstoffatome sind.
- Die Reduktion der Verbindungen der Formel IV kann mit elektrophilen Reduktionsmittel, insbesondere Diboran, wahlweise komplexiert mit Methylsulfid, Tetrahydrofuran, aliphatischen Aminen wie Triethylainin oder aromatischen Aminen wie N,N-Diethylanilin oder Pyridin, erfolgen.
- Alternativ kann die Reduktion mit nucleophilen Reduktionsmitteln wie Metallhydriden, beispielsweise Lithiumaluminiumhydrid und Natrium-(2- methoxyethoxy)-aluminiumhydrid, erfolgen.
- Die Reaktion erfolgt in Gegenwart eines Lösungsmittels, das unter den Reaktionsbedingungen inert ist, beispielsweise Tetrahydrofuran, Dietliylether oder 1,2-Dimethoxyethan.
- b) Alternativ kann anstelle des Acylhalogenids III ein Phenoxyacetaldehyd der Formel
- verwendet werden, worin R&sub2; und R&sub3; die obige Bedeutung haben.
- Durch Umsetzung mit Verbindung II, die in einem inerten Lösungsmittel, gegebenenfalls in Gegenwart von Trocknungsmitteln, durchgeführt wird, und durch anschließende Reduktion nach üblichen Verfahren, erhält man die Verbindungen der Formel I, worin R und R&sub1; Wasserstoffatome oder, wenn R&sub5; und R&sub6; von Wasserstoff verschieden sind, ihre Vorläufer bedeuten.
- Die obige Reaktion erfolgt in einem inerten Lösungsmittel durch katalytische Hydrierung oder durch Reaktion mit Reduktions-mitteln wie Natriumborhydrid, Natriumcyanbor-hydrid und Lithiumaluminiumhydrid.
- c) Ein weiteres Verfahren zur Herstellung der Verbindungen der Formel I besteht in der Kondensation zwischen einer Verbindung der Formel II und einer Verbindung der Formel
- worin R&sub2; und R&sub3; die obige Bedeutung haben und X&sub1; eine aus tretende Gruppe wie ein Chlor- oder Bromatom oder eine Alkylsulfonyloxy- oder Arylsulfonyloxy- Gruppe bedeutet.
- Die Reaktion erfolgt in einem inerten Lösungsmittel und in Gegenwart einer Base und liefert direkt die Verbindungen der Formel I, worin R und R&sub1; Wasserstoffatome oder ihre Vorläufer sind, wenn in der verwendeten Verbindung der Formel II R&sub5; und R&sub6; von Wasserstoff verschieden sind.
- Zwei weitere alternative Verfahren zur Synthese der Verbindungen der Formel I unter Verwendung von Zwischenprodukten, die von N-Propyldopamin oder seinen Derivaten der Formel II verschieden sind, sind die folgenden:
- d) Ein Verfahren verwendet als Ausgangsprodukt eine Verbindung der Formel
- worin
- R&sub5; und R&sub6; die obige Bedeutung haben und R&sub7; eine Formylgruppe oder eine CH&sub2;Y-Gruppe bedeutet, worin Y ein Halogenatom oder eine Alkylsulfonyloxy- oder Arylsulfonyloxy-Gruppe darstellt; die mit einem sekundären Amin der Formel
- kondensiert wird, worin R&sub2; und R&sub3; die obige Bedeutung haben; indem man in ähnlicher Weise wie unter b) beschrieben verfährt.
- Man erhält die Verbindungen der Formel I, worin R und R&sub1; Wasserstoffatome oder, wenn R&sub5; und R&sub6; von Wasserstoff verschieden sind, ihre Vorläufer sind.
- e) Alternativ können Doparnin oder ein Derivat der Formel
- worin R&sub5; und R&sub6; die obige Bedeutung haben, verwendet werden, um dann nach einem der in Abschnitt a), b) oder c) beschriebenen Syntheseverfahren das Zwischenprodukt der Formel
- zu erhalten, worin R&sub2;, R&sub3;, R&sub5; und R&sub6; die obige Bedeutung haben.
- Dies Zwischenprodukt (X) liefert durch nachf olgende Alkylierung mit einem Propylhalogenid in einem geeigneten organischen Lösungsmittel und wahlweise in Gegenwart einer Base die Verbindungen der Formel I, worin R und R&sub1; Wasserstoffatome oder ihre Vorläufer sind, wenn R&sub5; und R&sub6; von Wasserstoff verschieden sind.
- die Verbindungen der Formeln II, III, V, VI, VII, VIII und IX sind bekannt oder können nach bekannten Methoden hergestellt werden.
- Die oben beschriebenen verschiedenen Syntheseverfahren sind im wesentlichen alle gleichwertig, was ihre vergleichbaren Ausbeuten und ihre Vielseitigkeit betrifft. Die Verbindungen der Formel I, worin R und R&sub1; Wasserstoffatome darstellen oder in denen eine oder beide aromatische Hydroxygruppen in geschützter Form (beispielsweise als Methylether oder Benzylether) vorliegen, werden hergestellt, indem man nach den oben beschriebenen Verfahren arbeitet.
- Die Verbindungen der Formel I, in denen beide aromatischen Hydroxygruppen in freier Form vorliegen (R=R&sub1;=H) oder in denen eine Hydroxygruppe geschützt ist (vorzugsweise in Form von Benzylether), sind auch die Ausgangsverbindungen zur Herstellung von Verbindungen der Formel I, in denen ein oder beide Reste R und R&sub1; von Wasserstoff verschieden sind.
- Die Herstellung der Verbindungen der Formel I, in denen ein oder beide Reste R und R&sub1; von aliphatischen oder aromatischen Carbonsäuren abgeleitete Acylgruppen sind, erfolgt durch Acylierung nach üblichen Verfahren, beispielsweise mit einem geeigneten Acylhalogenid oder mit einem Anhydrid, oder, wenn einer der Reste R und R&sub1; eine Gruppe der Formel
- bedeutet, gemäß der in der EP-A 167 204 im Namen der Simes S.p.A. beschriebenen Methode der Monophosphorylierung der Hydroxygruppen des Catechins, auf die, soweit es die Reagentien und die experimentellen Bedingungen betrifft, Bezug genommen wird.
- Wenn die Verbindungen der Formel I ein oder mehrere Substituenten R, R&sub1;, R&sub2; und R&sub3; mit asymmetrischen Kohlenstoffatomen haben, können die erfindungsgemäßen Verbindungen in Form von Stereoisomeren vorliegen.
- Die vorliegende Erfindung betrifft die möglichen Mischungen der Stereoisomeren der Verbindungen der Formel I ebenso wie die einzelnen Stereoisomeren, die durch optische Trennung oder durch stereospezifische oder stereoselektive Synthese erhalten werden.
- Die Herstellung der Salze der Verbindungen der Formel I mit pharmazeutisch annehmbaren Säuren erfolgt nach üblichen Methoden.
- Eine zweckmäßige Methode besteht in der Zugabe der ausgewählten Säure zu einer Lösung von Verbindung I in einem Lösungsmittel, aus dem das erhaltene Salz ausfällt und von dem es durch Filtration abgetrennt werden kann.
- Wie oben ausgeführt besitzen die Verbindungen der Formel I dopaminerge gefäßerweiternde Wirkung und können in der Therapie als blutdrucksenkende Mittel und gegen Herzinsuffizienz eingesetzt werden.
- Die erfindungsgemäßen Verbindungen induzieren nach intravenöser Verabreichung in spontan hypertensiven Ratten bei einer Dosis von 0,033-0,53 um/kg eine bedeutsame systemische arterielle Druckerniedrigung zwischen 20 und 90 mmHg.
- Insbesondere zeigte die Verbindung N-[2-(2-Methoxyphenoxy)-ethyl]-N-propyldopaminhydrochlorid (nachfolgend als Verbindung A bezeichnet), verabreicht in Dosen von 12,5 ug/kg und 200 ug/kg, eine Verringerung des systemischen Arteriendrucks von 51 bzw. 89 mmHG verglichen mit dem Basalwert.
- Ferner wurden die dopaminerge Aktivität sowie die α&sub1;-antagonistische Wirkung der erfindungsgemäßen Verbindungen ausgewertet.
- Die dopaminerge Aktivität von Verbindung A wurde beispielsweise als Prozentsatz Rezeptorbindung an isolierten Membranen von gestreiftem Rattenmuskel berechnet; der IC&sub5;&sub0; ergab sich zu 0,3uM für den D&sub2;-Rezeptor und zu 3uM für den D&sub1; Rezeptor. Die α1-antagonistische Wirkung wurde als antagonistisclie Wirkung (pA&sub2;) auf die durch Phenylephrin (α&sub1;-Agonist) induzierte Gefäßverengung an isolierter Kaninchenaorta bestimmt. Der für Verbindung A erhaltene pA&sub2;-Wert betrug 7,7.
- Zur praktischen Verabreichung können die Verbindungen der Formel I vorzugsweise in Form einer pharmazeutischen Zusammensetzung hergestellt werden, die für den gewählten Verabreichungsweg geeignet ist.
- Die pharmazeutischen Zusammensetzungen, die die Verbindungen der Formel I oder ihre pharmazeutisch annehmbaren Salze zusammen mit ein oder mehreren festen oder flüssigen, organischen oder anorganischen pharmazeutischen Bindemitteln und wahlweise weiteren Zusätzen wie Verdünnungsmitteln, Konservierungsmitteln, Befeuchtungsmitteln, Farb- und Geschmacksstoffen enthalten, sind ein weiterer Gegenstand der vorliegenden Erfindung.
- Die erfindungsgemäßen pharmazeutischen Zusammensetzungen können in Form fester pharmazeutischer Präparate, wie Tabletten, Dragees, Kapseln, Granulaten und Suppositorien, verabreicht werden, oder in Form flüssiger pharmazeutischer Präparate, wie Sirup, Suspensionen, Emulsionen und Lösungen, die zur oralen oder parenteralen Verabreichung geeignet sind.
- Die erfindungsgemäßen Verbindungen könne auch in Form pharmazeutischer Präparationen mit langsamer und verzögerter Freisetzung formuliert werden.
- Die Herstellung der erfindungsgemäßen pharmazeutischen Zusammensetzungen kann nach üblichen Techniken erfolgen.
- Die folgenden Beispiele sollen die vorliegende Erfindung besser erläutern.
- Zu einer Lösung von N-Propyldopamin (5,52 g; 20 mMol) und Triethylamin (6,12 ml) in wasserfreiem Dimethylformamid (100 ml) wurde (2,6-Dimethylphenoxy)- acetylchlorid (3,97 g; 20 mMol) in wasserfreiem Dimethylformamid (10 ml) bei Raumtemperatur und unter Stickstoff hinzugegeben.
- Die Reaktionsmischung wurde 3 Stunden gerührt und das Lösungs-mittel wurde unter reduziertem Druck abgedampft.
- Der erhaltene Rückstand wurde in Ethylacetat gelöst, mit verdünnter HCl und anschließend mit Wasser bis zu einem neutralen pH gewaschen.
- Nach dem Trocknen über Natriumsulfat und Abdampfen des Lösungsmittels wurde das Rohprodukt in Methanol gelöst.
- Zu der so erhaltenen Lösung wurde p-Toluolsulfonsäure (0,5 g) hinzugefügt und die Mischung wurde unter Rückfluß 8 Stunden erhitzt.
- Das Lösungsmittel wurde abgedampft und der Rückstand wurde durch Säulenchromatographie an Silicagel (Eluens Methylenchlorid: Methanol = 97:3) gereinigt.
- N-[2-(3,4-Dihydroxyphenyl)-ethyl]-N-propyl-(2,6-dimethyl-phenoxy)-acetamid wurde als chromatographisch reines Öl erhalten (Dünnschichtchromatograpie - Eluens Methylenchlorid:Methanol 90:10).
- ¹H-NMR (60 MHz-CDCl&sub3;): delta (ppm): 0,8 (3H, t); 2,2 (3H, s); 2,3 (3H, s); 4,0 (2H, s); 4,6 (2H, s); 6,8-7,0 (6H, m).

- Zu einer Lösung von N-[2-(3,4-Dihydroxyphenyl)-ethyl]-N-propyl-(2,6- dimethylphenoxy)-acetamid (5,5 g; 15,4 mMol), hergestellt wie in Beispiel 1 beschrieben, in Tetrahydrofuran (55 ml), wurde BH&sub3; S(CH&sub3;)&sub2; (5,583 ml; 61,5 mMol) unter Stickstoff hinzugegeben, wobei die Temperatur bei 0-5ºC gehalten wurde. Die Reaktionsmischung wurde unter Rückfluß 30 Minuten erhitzt und nach Abkühlen wurde bei 5-10ºC 6N HCl (55 ml) hinzugefügt und die Mischung unter Rückfluß 30 Minuten erhitzt.
- Das Lösungsmittel wurde abgedampft und der Rückstand in Wasser gelöst, mit Natriumcarbonat neutralisiert und mit Methylenchlorid extrahiert.
- Nach Trocknen über Natriumsulfat und Abdampfen des Lösungsmittels erhielt man ein Rohprodukt, das in Ethanol gelöst wurde.
- Aus der Lösung, die mit HCl-gesättigtem Ethylether angesäuert wurde, kristallisierte N-[2-(2,6-Dimethylphenoxy)-ethyl]-N-propyl-dopamin als Hydrochlorid mit einem Schmelzpunkt von 104-105ºC (Ethanol/Ethylether). Auf ähnliche Weise wurden die folgenden Verbindungen hergestellt:
- Schmp. = 107-109ºC (Ethanol/Ethylether)
- Schmp. = 126-129ºC (Ethanol/Ethylether)
- Schmp. = 137-138ºC (Ethanol/Ethylether)
- Schmp. = 124-126ºC (Ethanol/Ethylether)
- Schmp. = 141-143ºC (Ethanol/Ethylether)
- Schmp = 187-188ºC (Ethanol/Ethylether)
- Zu einer Lösung von N-[2-(2-Methoxyphenoxy)-ethyl]-N-propyl-dopamin-hydrochlorid (4 g; 11,7 mMol), hergestellt wie in Beispiel 2, in Pyridin (30 ml), wurde Isobutyrylchlorid (4 ml) hinzugegeben.
- Nach 4 Stunden bei Raumtemperatur wurde die Mischung in Wasser gegossen und mit Methylenchlorid extrahiert.
- Die organische Phase wurde bis zum Rückstand abgedampft, mit Ethylether behandelt, mit einer verdünnten Natriumbicarbonat-Lösung gewaschen und über Natriumsulfat getrocknet.
- Nach Abdampfen des Lösungsmittels wurde ein Öl erhalten, das in Ethylether gelöst wurde.
- Durch Ansäuern der Lösung mit einer mit Maleinsäure gesättigten ethanolischen Lösung wurde 3,4-O-Diisobutyryl-N-[2-(2-methoxyphenoxy)-ethyl]-N-propyl-dopamin- maleat abgetrennt; Schmp. 100-102ºC (Ethylacetat).
Claims (6)
1. Verbindung der Formel
worin
R und R&sub1;, die gleich oder verschieden sein können, Wasserstoffatome bedeuten
oder Acylgruppen, die abgeleitet sind von wahlweise substituierten
aliphatischen, aromatischen oder heteroaromatischen Carbonsäuren, von
wahlweise substituierten Carbamin- oder Kohlensäuren, oder von
Phosphorsäure der Formel
worin
R&sub4; ein Wasserstoffatom, eine wahlweise mit ein oder mehreren Gruppen,
die ausgewählt sind aus Hydroxy, Alkoxy, Acyloxy, Amino, Carboxy und
Alkoxycarbonyl, substituierte C&sub1;-C&sub6;-Alkylgruppe, oder eine Phenylgruppe
bedeutet, unter der Bedingung, daß, wenn eine der Gruppen R und R&sub1; eine
von Phosphorsäure abgeleitete Acylgruppe darstellt, die andere Gruppe
ein Wasserstoffatom bedeutet;
R&sub2; und R&sub3;, die gleich oder verschieden sein können, ein Wasserstoff- oder ein
Halogenatom, eine Alkyl- oder eine Alkoxygruppe bedeuten;
und ihre Salze mit pharmazeutisch annehmbaren Säuren.
2. Verbindung nach Anspruch 1, worin R und R&sub1;, die gleich oder verschieden sein
können, Acylgruppen bedeuten, die sich von Carbonsäuren ableiten, die
ausgewählt sind aus der Gruppe Ameisensäure, Essigsäure, Propionsäure,
Buttersäure, Isobuttersäure, Valeriansäure, Pivalinsäure, wahlweise
substituierter Benzoesäure oder wahlweise substituierter 2-Picolinsäure, 3-
Picolinsäure und 4-Picolinsäure.
3. Verbindung nach Anspruch 1, worin eine der Gruppen R und R&sub1; ein
Wasserstofatom bedeutet und die andere eine Acylgruppe, abgeleitet von
Phosphorsäure der Formel
worin
R&sub4; ein Wasserstoffatom oder eine Methyl- oder Ethylgruppe bedeutet, wahlweise
substituiert mit ein oder mehreren Gruppen, die ausgewählt sind aus
Hydroxy, Methoxy, Acetoxy, Amino, Carboxy, Methoxycarbonyl,
Ethoxycarbonyl und Phenyl.
4. Verbindung nach Anspruch 1, worin R und R&sub1; Wasserstoffatome bedeuten.
5. Verfahren zur Herstellung von Verbindungen nach Anspruch 1, das umfaßt:
a) die Reaktion zwischen N-Propyldopamin oder einem seiner Derivate der
Formel
worin
R&sub5; und R&sub6;, die gleich oder verschieden sein können, ein Wasserstoffatom oder
eine Schutzgruppe, ausgewählt aus Methyl und Benzyl, bedeuten;
und einem Halogenid einer Phenoxyessigsäure der Formel
worin
R&sub2; und R&sub3;, die gleich oder verschieden sein können, ein Wasserstoffatom oder
ein Halogenatom, eine Alkyl-oder eine Alkoxygruppe bedeuten und X
ein Chlor- oder Bromatom bedeutet;
in einem inerten Lösungsmittel in Gegenwart einer Base, die als Akzeptor für
Halogenwasserstoffsäure fungiert und ausgewählt ist aus einem Alkalicarbonat
oder -bicarbonat oder einem tertiären Amin, ausgewählt aus Triethylamin oder
Pyridin.
b) die Reduktion des so erhaltenen Zwischenprodukts der Formel
worin
R&sub2;, R&sub3;, R&sub5; und R&sub6; die obige Bedeutung haben; mit elektrophilen oder
nucleophilen Reduktionsmitteln, ausgewählt aus Diboran, wahlweise
komplexiert mit Dimethylsulfid, Tetrahydrofuran und aliphatischen oder
aromatischen Aminen, Lithiumaluminiumhydrid und Natrium-(2-
methoxyethoxy)-aluminiumhydrid, in einem unter den Reaktionsbedingungen
inerten Lösungsmittel.
6. Pharmazeutische Zusammensetzung, die eine therapeutisch wirksame Menge einer
Verbindung nach Anspruch 1 zusammen mit ein oder mehreren pharmazeutisch
annehmbaren Bindemitteln enthält.
