Description
- Die vorliegende Erfindung betrifft 1-[2-(Phenylmethyl)phenyl)-piperazin-Verbindungen und ein Verfahren zu ihrer Herstellung. Insbesondere betrifft sie 1-[2-(Phenylmethyl)-phenyl]piperazin-Verbindungen der Formel (I):
- worin R&sub1; ein Wasserstoffatom, eine Niedrigalkylgruppe mit 1-4 Kohlenstoffatomen und ein Halogenatom ist, R&sub2; ist ein Wasserstoffatom, eine Niedrigalkylgruppe mit 1-4 Kohlenstoffatomen, ein Halogenatom und eine Niedrigalkoxygruppe mit 1-4 Kohlenstoffatomen ist und R&sub3; ein Wasserstoffatom oder eine Methylgruppe ist, und die nichttoxischen Säureadditionssalze davon, sowie ein Verfahren zur Herstellung dieser Verbindungen und pharmazeutische Mittel, die diese Verbindungen enthalten. In der Formel I bedeutet "Halogen" Fluor, Chlor, Brom oder Jod, wobei Chlor besonders bevorzugt ist.
- Die Verbindungen der vorliegenden Erfindung werden nach folgendem Schema hergestellt:
- In den Ausgangsverbindungen der allgemeinen Formel (II) besitzen R&sub1; und R&sub2; die gleichen, für (I) angegebenen Bedeutungen. Bei (III) steht X für Chlor oder Brom.
- Die Herstellung der Ausgangsverbindungen der allgemeinen Formel (II) wird, ausgehend von, entweder a)den entsprechenden 2-Aminobenzophenonen durchgeführt, entweder a) durch direkte Reduktion unter Verwendung von Hydrazin unter den Bedingungen der Wolff-Kishner Reaktion (M.Gall et al: J.Med.Chem., 19 (8), 1057, 1976; W.D. Zahler et al: Ber., 96, 765. 1963; H.H. Oelschläger et al: Arz. Forsch., 23(6), 802, 1973) oder Natrium und Ethanol (E.J. Engels et al: J.Chem.Soc., 2694, 1959), oder b) durch Reduktion mit Zink und Salzsäure aus den entsprechendenden Carbinolen (Hoffmann-La Roche: US Patent Nr. 4 194 049) oder c) direkt von den substituierten Anilinen (P.S. Gassman et al: J.Am.Chem.Soc., 100(24), 7600, 1978; P.G. Gassmann et al: J.C.S.Chem.Comun., 488, 1973).
- Das Piperazin wird nach dem obigen Schema hergestellt, ausgehend von den jeweiligen 2-(Phenylmethyl)anilinen (II) und Bis-(2-haloethyl)aminen (III) in Gegenwart von basischen Coadjuvantien. Geeignete basische Coadjuvantien sind Alkali- oder Erdalkalimetallcarbonate oder Bicarbonate. Alkohole, die 1-4 Kohlenstoffatome aufweisen, oder Alkyloxyethanole mit 4-6 Kohlenstoffatomen werden als geeignete Lösungsmittel verwendet.
- Die N-methylierten Homologen (I, R&sub3;=CH&sub3;) werden aus (I,R&sub3;=H) durch Eschweiler-Clarke Reaktion, unter Verwendung von Formaldehyd und Ameisensäure erhalten.
- Desweiteren bezieht sich die Erfindung auf pharmazeutische Zusammensetzungen, die mindestens eine Verbindung gemäß Anspruch 1 enthalten, gegebenenfalls in Kombination mit pharmazeutisch verträglichen Trägern und/oder Adjuvantien.
- Die Verbindungen der vorliegenden Erfindung sind in der Humantherapie sehr brauchbar aufgrund ihrer hervorragenden antidepressiven Aktivität. Die Verbindungen können mit geeigneten Trägern vermischt, oral in Form von Tabletten, als beschichtete Tabletten, Kapseln, Puder, Syrup, Lösungen, etc. und als Injektionen, mit einer täglichen Dosis im Bereich von 0.1 bis 10 mg/kg verabreicht werden.
- In einem Kolben, der mit einem Rührer und einem Kühler versehen ist, werden 18,33 g (0,1 Mol) 2-Benzylanilin und 17.85 g (0.1 Mol) Bis-(2-chlorethylamin) Hydrochlorid, in 100 ml n- Butanol gelöst und 8 Stunden in einem Ölbad unter Rückfluß erhitzt. Man läßt die Mischung abkühlen und gibt 6.91 g (0.05 Mol) wasserfreies Kaliumcarbonat unter 8-stündigem Kochen bei Rückflußtemperatur zu; die Zugabe von 6.91 g des wasserfreien Kaliumcarbonats unter 8-stündigem Rückfluß wird zweimal wiederholt. Danach läßt man die Mischung abkühlen, die unlöslichen Salze filtriert man ab und engt die Butylalkohollösung zur Trockene ein. Man löst den harzartigen Rückstand in 100 ml Methylenchlorid, leitet Chlorwasserstoff unter Kühlung ein und fällt das Hydrochlorid durch Zugabe von Ethyläther aus; dann wird es abfiltriert und getrocknet, wobei man 28 g eines schwach gefärbten Feststoffs erhält. 10.36 g (37%) des Endprodukts in Form eines weißen Feststoffes, Smp. 192-194ºC werden durch Umkristallisation aus Acetonitril erhalten.

- Eine Mischung von 3.46 g 1-[2-(Phenylmethyl)-phenyl]-piperazin-Hydrochlorid, 2.5 ml 38%igem Formaldehyd und 1 ml Ameisensäure wird in einem Ölbad bei 110-120ºC 2 Stunden erhitzt. Dann wird sie am Rotationsverdampfer konzentriert, wobei nach dem Trocknen, 3.64 g eines reinen, gelblichen Feststoffes mit einem Smp. von 202-204ºC und einer korrekten Analyse erhalten werden.

- a) 42.14 g 2-Benzyl-4-chloranilin, 35.52 g Bis-(2-chlorethylamin) Hydrochlorid in 190 ml n-Butanol und wiederholte Zusatzgaben von 13.35 g wasserfreiem Kaliumcarbonat werden nach der in Beispiel 1 beschriebenen Methode zur Reaktion gebracht. Nach dem Abkühlen wird das unlösliche Material abfiltriert und über den gleichen Filter, mit 4·50 ml Methylenchlorid gewaschen (der unlösliche Rückstand, der aus anorganischen Salzen besteht, wir verworfen). Das Lösungsmittel wird verdampft, wobei 30.8 g (50%) eines Feststoffes als Hydrochlorid erhalten werden. Die Umkristallisation aus Isopropanol ergibt 21 g (34%) eines weißen Feststoffes mit einem Smp. von 209-210ºC mit einer korrekten Analyse.

- b) In einen Kolben, der mit einem Rührer und Kühler versehen ist, gibt man 27.60 g 2-Benzyl-4-Chloranilin, 22.70 g Bis-(2-chlorethylamin), 80 ml n-Butoxyethanol und 17.60 g wasserfreies Kaliumcarbonat. Die Mischung wird für 30 Stunden in einem Ölbad unter Rückfluß erhitzt. Danach läßt man sie abkühlen, gießt sie auf 300 ml Wasser und extrahiert viermal mit 4·250 ml Ethylacetat. Die organischen Extrakte werden mit 3x100 ml 1M Natriumhydroxidlösung und salzgesättigtem Wasser zur Neutralisierung gewaschen, dann getrocknet und eingedampft, wobei man einen öligen Rückstand (39 g) erhält. Durch Kiesegel Säulenchromatography (Methylenchlorid/Methanol mit zunehmender Polarität) werden 12.4 g (34%) eines rosafarbenen, dem Base entsprechenden Feststoffs bei 4% CH&sub3;OH erhalten, Smp. 68-70ºC mit korrekter Analyse.

- Aus 6,47 g 1-[4-Chlor-2-(phenylmethyl)phenyl]piperazin- Hydrochlorid, 4,18 ml 38%igem Formaldehyd und 1,68 ml Ameisensäure erhält man unter Befolgung des Beispieles 2 6,65 g eines Feststoffes; Smp. 207,5-209ºC, Analysenwerte korrekt.

- a) 39,52 g 2-Benzyl-4-methylanilin, 36,43 g Bis(2-chlorethylamin) in 130 ml n-Butoxyethanol und 28,5 g wasserfreies Kaliumcarbonat setzt man gemäß dem in Beispiel 3 (b) beschriebenen Verfahren um. Durch Eindampfen der organischen Phase erhält man einen öligen Rückstand (58 g), den man dann in trockenem Methylenchlorid auflöst und unter Kühlen in einem Eis-Wasser-Bad einen Chlorwasserstoff-Gasstrom einleitet. Das gebildete Hydrochlorid fällt durch Zugabe von trockenem Ethyläther (50 ml) aus und wird, nachdem man es über Nacht im Kühlschrank stehen läßt, abfiltriert, mit Ethyläther gewaschen und getrocknet. 34,0 g (55%) Monohydrochlorid in Form eines weißen Feststoffes mit Smp. 202-204ºC und in chromatographisch reiner Form erhält man hierbei. Durch Umkristallisation in Ethanol erhält man weiße Kristalle mit Smp. 209-210ºC und korrekten Analysenwerten.

- b) 22,0 g 1-[5-Methyl-2-(phenylmethyl)phenyl)piperazin, die man durch Neutralisation des Monohydrochlorids mit IN Natriumhydroxid, gelöst in Toluol, erhält, löst man in 130 ml absolutem Ethanol. In diese Lösung leitet man Chlorwasser-stoffgas ein, wobei man in einem Eis-Wasser-Bad kühlt, bis Sättigung erreicht ist und eine Ausfällung erfolgt. Das Gemisch wird auf Rückfluß gekocht und in weiteren 50 ml Ethanol gelöst. Nach Abkühlen und Filtrieren erhält man 25 g weißer Kristalle, die dem Hydrochlorid entsprechen; Smp. 210-212ºC, Analysenwerte korrekt.

- a) Ein Gemisch aus 37,24 g 2-Benzyl-4-methylanilin in 215 ml absolutem Ethanol und 61,98 g Bis-(2-brom-ethylamin)- Hydrobromid erhitzt man 8 Stunden auf Rückflußtemperatur. Nach Abkühlen gibt man 20,0 g wasserfreies Natriumcarbonat hinzu und kocht weitere 8 Stunden auf Rückflußtemperatur. Nach 24-stündiger Aufbewahrung im Kühlschrank filtriert man den Nieder-schlag ab, wäscht wiederholt mit Methylenchlorid (insgesamt 500 ml Volumen); die resultierende Lösung dampft man im Vakuum ein, wobei man 37,0 g (58%) eines gelblichen Feststoffes erhält, der dem Hydrobromid entspricht. Smp. 220-222ºC; chromatographisch rein mit korrekter Bromidanalyse. Durch Umkristallisieren in Ethanol erhält man weiße Kristalle (Ausbeute 80%), Smp. 227- 229ºC, Analysenwerte korrekt.

- b) 10,0 g des gemäß a) hergestellten Hydrobromids suspendiert man in 100 ml Toluol und neutralisiert mit 1N Natriumhydroxid, während man 30 Minuten rührt. Die wäßrige Phase dekantiert man und wäscht die Toluolphase mit Wasser und neutralisiert. Nach Eindampfen verbleibt ein Rückstand von 8 g, der in 80 ml Hexan umkristallisiert wird, wobei man 5,07 g weißer Kristalle erhält, Smp. 64-67ºC; der Base entsprechend; Analysenwerte korrekt.

- Aus 5,20 g 1-[5-Methyl-2-(phenylmethyl)phenyl]piperazin- Hydrochlorid, 3,60 ml 37%igem Formaldehyd und 1,45 ml Ameisensäure erhält man bei Durchführung des Verfahrens gemäß Beispiel 2 5,25 g eines weißen Feststoffes, Smp. 208-211ºC, chromatographisch rein und Analysenwerte korrekt.

- 18,18 g 2-Benzyl-5-chloranilin, 14,90 g Bis-(2-chlorethylamin) Hydrochlorid setzt man unter wiederholten Zugaben von 5,78 g wasserfreiem Kaliumcarbonat gemäß dem in Beispiel 1 beschriebenen Verfahren um. Das Hydrochlorid bildet sich in Methylenchlorid aus und ein Niederschlag bildet sich bei Zugabe von Ethyläther. Diesen filtriert man, wäscht wiederholt mit Ethyläther und trocknet, wobei man 17 g eines hygroskopischen Produktes erhält. Durch Umkristallisation in Ethanol erhält man 8,4 g (25%) eines weißlichen Feststoffes, Smp. 256-258ºC, chromatographisch rein, korrekte Elementaranalysenwerte.

- Aus 2,10 g 1-[5-Chlor-2-(phenylmethyl)phenyl)piperazin- Hydrochlorid, 1,36 ml 37%igem Formaldehyd und 0,55 ml Ameisensäure erhält man bei Durchführung des Verfahrens gemäß Beispiel 2 2,2 g eines weißlichen Feststoffes, Smp. 195-197ºC, chromatographisch rein, Analysendaten korrekt.

- 53,8 g 2-(4'-Methoxybenzyl)-4-methylanilin, 42,25 g Bis- (2-chlorethylamin) Hydrochlorid in 185 ml n-Butanol setzt man bei wiederholten Zugaben von 16,36 g wasserfreiem Kalium-carbonat gemäß dem in Beispiel 1 beschriebenen Verfahren um. N-Butanol dampft man ab und rührt den resultierenden harzartigen Rückstand mit 300 ml Toluol. Das Hydrochlorid wird in Form eines weißen Feststoffes unlöslich gemacht, abfiltriert und getrocknet. Die Ausbeute beträgt 19,25 g (25%). Smp. 180- 182ºC, chromatographisch rein. Das wäßrige Toluol wird auf das halbe Volumen aufkonzentriert und eine zweite Menge des Produkts, verunreinigt durch die Base, wird isoliert. Das Hydrochlorid kristallisiert man in einer Ausbeute von 75% aus Ethanol um, Smp. 183-185ºC, Analysenwerte korrekt.
- IR-Spektrum (KBr), cm&supmin;¹: 3400, 3100-2440, 1620, 1520, 1250, 1190, 1050, 830. ¹H-NMR-Spektrum (CDCl&sub3;) ppm: 2,25 ( , 3H; CH&sub3;-), 3,12 (zwei symmetrische Bandgruppen, 8H; Piperazin), 3,60 ( , 3H; -OCH&sub3;), 8,25 (breit, 2H; ).
- Aus 5,0 g 1-[4-Methyl-2-[(4'-methoxyphenyl)methyl)phenyl]-piperazin-Hydrochlorid, 3,12 ml 37%igem Formaldehyd und 1,25 ml Ameisensäure isoliert man bei Durchführung des Verfahrens gemäß Beispiel 2 5,1 g eines weißlichen Feststoffes, Smp. 195-197ºC, chromatographisch rein, Analysenwerte korrekt.
- IR-Spektrum (KBr), cm&supmin;¹: 3450, 3040-2400, 1520, 1255, 1040, 1000, 865, 825.
- ¹H-NMR-Spektrum (CDCl&sub3;), ppm: 2,26 ( , 3H; CH&sub3;-), 2,80 ( , 3H; N-CH&sub3;), 3,18 (breit, 8H; Piperazin), 3,75 ( , 3H; -OCH&sub3;), 3,91 ( , 2H; Ar- -Ar), 6,7-7,2 ( , 7H; Ar-) und 9,6 (breit, 1H; ).
- 38,12 g 2-(2'-Chlorbenzyl)-4-chloranilin, 28,23 g Bis-(2- chlorethylamin) Hydrochlorid in 150 ml n-Butanol setzt man bei wiederholten Zugaben von 10,51 g wasserfreiem Kaliumcarbonat gemäß dem in Beispiel 1 beschriebenen Verfahren um. N-Butanol dampft man ab und löst den harzartigen Rückstand in Toluol, neutralisiert mit einer 1N Natriumhydroxidlösung, wäscht mit Wasser und dampft das Toluol (21 g) ab. Den Rückstand löst man in Ethanol. Chlorwasserstoffgas leitet man in diese Lösung unter Kühlen ein, wobei das Hydrochlorid ausfällt. Das Produkt wird abfiltriert, getrocknet und in Isopropanol umkristallisiert, wobei man 8,15 g des Endproduktes erhält, Smp. 171,5- 173ºC, Analysenwerte korrekt.
- IR-Spektrum (KBr), cm&supmin;¹: 3500, 3100-2400, 1590, 1480, 1450, 1240, 1100, 1050, 925, 740. ¹H-NMR-Spektrum (d&sub6;DMSO), ppm: 3,08 (breit, 8H; Piperazin). 4.12 ( , 2H; Ar- -Ar), 6,8-7,5 ( , 7H; Ar-) und 9,6 (breit, 2H; ).
- 20,81 g 2-(4'-Methylbenzyl)-4-chloranilin, 16,03 g Bis-(2- chlorethylamin) in 85 ml n-Butanol setzt man bei wiederholten Zugaben von 6,13 g wasserfreiem Kaliumcarbonat gemäß dem in Beispiel 1 beschriebenen Verfahren um. Die Butanolschicht dampft man ab, nimmt in Toluol auf, macht mit einer 1N NaOH- Lösung basisch, wäscht in Wasser und dampft zur Trockene ein. Den Rückstand löst man in 60 ml Ethanol. 15 ml konzentrierte Salzsäure gibt man hinzu und filtriert den Niederschlag (Ausgangs-Anilin Hydrochlorid) ab. Das Filtrat dampft man ein, neutralisiert und löst in 90 ml trockenem Methylenchlorid. Chlorwasserstoffgas leitet man in diese Lösung unter Kühlen ein, wobei das Hydrochlorid ausfällt. Nach 24-stündiger Aufbewahrung im Kühlschrank wird der Niederschlag abfiltriert, getrocknet und in Methanol umkristallisiert, wobei man 7,87 g (26%) des Endproduktes erhält, Smp. 216-218ºC, chromatographisch rein, Analysenwerte korrekt.
- IR-Spektrum (KBr), cm&supmin;¹: 3500, 3100-2400, 1480, 1450, 1230, 1110, 1040, 820. ¹H-NMR-Spektrum (d&sub6;-DMSO), ppm: 2,25 ( , 3H; CH&sub3;-), 3,08 (breit, 8H; Piperazin), 9,96 ( , 2H; Ar- -Ar), 7,2 ( , 7H; Ar-) und 9,75 (breit, 2H; ).
Claims (4)
1. 1-[2-(Phenylmethyl)phenyl]piperazinverbindungen der Formel
(I):
worin R&sub1; für ein Wasserstoffatom, eine Alkylgruppe mit 1-4
Kohlenstoffatomen oder ein Halogenatom steht, R&sub2; für ein
Waserstoffatom, eine Alkylgruppe mit 1-4 Kohlenstoffatomen, ein
Halogenatom oder eine Alkoxygruppe mit 1-4 Kohlenstoffatomen
steht und R&sub3; ein Wasserstoffatom oder eine Methylgruppe
darstellt, und die nicht-toxischen Säureadditionssalze davon.
2. Verfahren zur Herstellung der Verbindungen nach Anspruch
1, wobei man die Verbindungen der allgemeinen Formel (II)
worin R&sub1; und R&sub2; wie für (I) definiert sind, mit den bis-(2-
Halogenethyl)aminen der allgemeinen Formel (III)
(XCH&sub2;CH&sub2;)&sub2;NH (III)
worin X für ein Chlor- oder Bromatom steht, in einem Medium aus
einem Alkohol mit 1-4 Kohlenstoffatomen oder einem
Alkoxyethanol mit 4-6 Kohlenstoffatomen und in Gegenwart eines
basischen Co-adjuvans bei einer Temperatur zwischen
Zimmertemperatur und Siedetemperatur umsetzt und, falls gewünscht,
nachfolgend eine Methylierung mit einem Gemisch aus Formaldehyd und
Ameisensäure bei einer Temperatur im Bereich von 80 bis 130ºC
durchführt.
3. Verfahren nach Anspruch 2, wobei das basische Co-adjuvans
ein Alkali- oder Erdalkalicarbonat oder -bicarbonat ist.
4. Pharmazeutisches Mittel, enthaltend mindestens eine
Verbindung nach Anspruch 1, gegebenenfalls in Kombination mit
pharmazeutisch akzeptablen Trägern und/oder Adjuvantien.
