Die Verbindungen der Erfindung, in denen mindestens einer der Reste R2 oder R^ ein Wasserstoff atom bedeutet, bilden
basische Salze mit verschiedenen anorganischen und organischen Basen, die ebenfalls Gegenstand derjErfindung sind.
Solche Salze sind die Ammoniumsalze, Alkalimetallsälze, wie Lithium-, Natrium- und Kaliumsalze (die bevorzugt sind),
Erdalkalimetallsalze, wie Calcium- und Magnesiumsalze, Salze mit organischen Basen, wie das Dicyclohexylaminsalz,
Benzathin-, N-Methyl-D-glucamin- und Hydrabaminsalz, Salze
rait Aminosäuren, wie Arginin, Lysin und dergleichen. Die nichttoxischen pharmakologisch verträglichen Salze sind bevorzugt,
obwohl auch andere Salze wertvoll sein können, beispielsweise zur Gewinnung und Reinigung des Produktes. Die
Salze werden nach üblichen Verfahren hergestellt.
V/ie vorstehend gezeigt, liegt der Iminosäure- oder -ester-Teil

Die Verbindungen der Formel I und ihre physiologisch verträg-' liehen Salze sind blutdrucksenkende Wirkstoffe. Sie inhibieren
die Umwandlung des Decapeptids Angiotensin I in Angiotensind II und stellen deshalb wertvolle Stoffe zur Verminderung
oder Beseitigung der mit Angiotensin verbundenen Hypertension dar. Die Wirkung des Enzyms Renin auf Angiotensinogen, ein
Pseudoglobulin in der Blutbahn, erzeugt Angiotensin I. Angiotensin
I wird durch das Angiotensin Konverting-Enzym (ACE) in Angiotensin II umgewandelt. Letzteres ist ein wirksamer
druckerhöhender Stoff, der als auslösendes Agens für verschiedene Formen des Bluthochdrucks bei verschiedenen
Säugern, beispielsweise beim Menschen gilt. Die Verbindungen der Erfindung greifen in die Reaktionsfolge Angiotensinogen

deshalb die Angiotensin-abhängige Hypertension bei Säugern,

Eine Einzeldosis, oder vorzugsweise zwei bis vier unterteilte tägliche Dosierungen auf der Basis von etwa 0,1 bis 100
mg/kg Körpergewicht pro Tag ist zur Verminderung des Blutdrucks geeignet. Die Stoffe werden vorzugsv/eise oral verabreicht,
es können jedoch auch parenterale Wege, wie subkutane, intramuskuläre, intravenöse oder intraperitoneale Verabreichungen
in Betracht kommen.
Die Verbindungen der Erfindung können auch in Kombination mit einem Diuretikum zur Behandlung von Hypertension formuliert
werden. Ein Kombinationspräparat, das eine Verbindung der Erfindung und einen diuretischen Wirkstoff enthält, kann
in.einer wirksamen Menge verabreicht werden, die eine Gesamt-
* tagesdosia von etwa 30 bis 600 mg, vorzugsweise etwa 30 bis
330 mg der Verbindung der Erfindung und etwa 15 bis 300 mg,
vorzugsweise etwa 15 bis 200 mg des diuretischen Wirkstoffs
umfaßt. Beispiele für in Betracht kommende Diuretika, die in Kombination mit einer Verbindung der Erfindung verwendet werden
können, sind die Thiazid-Diuretika, wie Chlorthiazid, Hydrochlorthiazid, Flumethiazid, Hydroflumethiazid, Bendroflumethiazid,
Methylclothiazid, Trichlormethiazid, Polythiazid .oder Benzthiazid, sowie Äthacrinsäure, Ticrynafen, Chlorhalidon,
Furosemid, Musolimin, Bumetanid, Triamteren, Amilo-
• rid und Spironolacton, sowie Salze dieser Verbindungen.

Die Verbindunden der Formel I können zum Gebrauch bei der
Verminderung des Blutdrucks zu Formulierungen, wie Tabletten, Kapseln oder Elixiere zur oralen Verabreichung, oder als
sterile Lösungen oder Suspensionen für parenterale Verabreichung hergerichtet werden. Etwa 10 bis 500 mg einer Verbindung
der allgemeinen Formel I wird dazu mit physiologisch verträglichen Träger-, Hilfs- oder Zusatzstoffen zu Dosierungsformen
verarbeitet.
Die Beispiele erläutern die Erfindung. Temperaturen sind in

a) (4~Phenylbutyl)-phosphonigsäure-diäthylester
4,8 g (0,2 Mol) Magnesiummetall werden in 50 ml Diäthyläther
auf ge schlämmt und tropf env/eise mit einer Lösung von 36,4- g
(0,22 Mol) 1-Chlor-4-phenylbutan in 100 ml Diäthyläther in
solcher Geschwindigkeit vernetzt, daß das Gemisch unter leichtem Rückfluß gehalten wird. Danach wird das Gemisch 1 Stunde
unter Rückfluß erhitzt und gerührt. Wach dem Abkühlen und
Filtrieren unter Argon wird die Grignard-Lösung (0,147 Mol
durch Titration) tropfenweise zu einer auf O0C abgekühlten
Lösung von 25,7 S (0,16 Mol) Diäthylchlorphosphit in 100 ml
Diäthyläther in einer Geschwindigkeit gegeben, mit der eine Innentemperatur von 0 bis 100C aufrechterhalten wird. Nach
der Zugabe wird das Gemisch 1,5 Stunden unter Rückfluß erhitzt. Nach dem Filtrieren und Eindampfen bei Normaldruck
unter Argon wird der Rückstand unter vermindertem Druck frak-' tioniert. Ausbeute: 29,7 g (4-Phenylbutyl)-phosphonigsäure-
25 diäthylester vom Kp. 110 bis 113°C bei 0,09 bis 0,1 mm.
b) /Äthoxy-(4~phenylbutyl)-phosphinyl7-eGsigsäure-methylester
Ein Gemisch von 16,9 g (0,11 Mol) Methylbromacetat und 5,0 g
(0,019 Mol) (4-Phenylbutyl)-phosphonigsäurediäthylester
wird'in einem 140 bis 150°C heißen Ölbad, erhitzt, bis Abdestillation
von Äthylbromid beobachtet wird. Danach werden weitere-16 g (0,063 Mol) (4-Phenylbutyl)-phosphonigsäurediäthylester
nach und nach zu dem Reaktionsgemisch gegeben, das dann weitere 45 Minuten erhitzt wird. Nach dem Abkühlen
auf 1000C wird überschüssiges Reagens unter vermindertem Druck
abdestilliert. Es worden 2lj g Rohprodukt vom R„-Wert 0,25
(Kieselgel/Äthylacetat) erhalten. Verunreinigungen mit höherem R„-Wert werden durch Flash-Chromatographie unter Verwendung
von Äthylacetat abgetrennt. Als Ergebnis werden 18,5 g /Athoxy-(4-phenylbutyl)-phosphinyl7-essigsäuremethylester
erhalten.
c) /Ä*thoxy-(zl~phenylbutyl)-phosphinyl7-essigsäure
Eine Lösung von 15 g (0,05 Mol) /Sthoxy-(4~phenylbutyl)-phosphinyl7-essigsäure-methylester
in 51 ml 0,99 N Natronlauge
(0,05 Mol) wird 30 Minuten bei Raumtemperatur gerührt.
Nach Extraktion mit Diäthyläther wird die Lösung mit 8,5 ml 5N SaIs säure behandelt. Das Produkt ..wird in Äthylacetat extrahiert,
getrocknet und das Lösungsmittel unter vermindertem Druck eingedampft. Ausbeute: '13,6 g /Äthoxy-(4—phenylbutyl)-phosphinyl7-essigsäure
als Öl vom R„-¥ert 0,75 an Kieselgel unter Verwendung von Dichlormethan/Essigsäure/Methanol im

mit 1,15 g (7,10 EiHoI) Carbonyldiimidazol behandelt und 1
Stunde bei O0C im Eisbad gerührt. Die erhaltene Lösung wird
dann mit 1,5 nil (11,3 mMol) Azidotrimethylsilan behandelt
und auf Raumtemperatur erwärmen gelassen. Nach 30 Minuten
wird- das Gemisch mit etwa 30 ml Toluol verdünnt und nacheinander
zweimal- mit 5$ Natriumbisulfätlösung und gesättigter
Kochsalslösung gewaschen, über Natriumsulfat getrocknet und eingedampft. Es v/erden 2,1 g rohes Acylazid erhalten. Dünnschichtchromatogramm
(Aceton/Dichlormethan im Verhältnis 1:2): Hauptpunkt beim Rf-Wert 0,57, IR (Film) 2150 cm"1 (N,).

2,1 g rohes Acylazid werden in 15 DiI wasserfreies Toluol aufgenommen
und auf 90°C (Badtemperatur) erhitzt, bis die Stick- ; stoffentwicklung aufhört (etwa 30 Minuten). IR-Spektrum (Film)
2250" cm (-NCO). Das Gemisch wird nach dem Abkühlen mit
1,75 S (7,25 mMol) L-Prolin-phenylmethylester-hydrochlorid

Gasförmiges Äthylamin wird durch eine Lösung von 9»0 ml (60
mMol) Chlorameisensäurebenzylester in 200 ml wasserfreies Tetrahydrofuran 1 Stunde lang geleitet. Das heterogene Gemisch
wird dann mit 10 ml (72 ml-lol) Triäthylamin behandelt und bei
Raumtemperatur 20 Minuten gerührt. Das Reaktionsgemisch wird hierauf mit Äthyiacetat verdünnt und mit 5# Kaliumbisulfat,
Wasser und Kochsalzlösung gewaschen, über Magnesiumsulfat getrocknet und eingedampft. Der Rückstand (10,2 g) kristallisiert
beim Stehen bei O0C. Der weiße Peststoff wird mit x^rasserfreiem
Hexan digeriert und dann durch Filtration gesammelt. Ausbeute: 9?3 g Äthylcarbaminsäure-phenylmethylester als weißer
Feststoff vom F. etwa Raumtemperatur. Dünnschichtchromato-
gramm (Äthylacetat): einziger Fleck beim R--Wert 0,9.

b) Äthyl-(hydroxymethyl)-carbaminsäure-phenylmethylester Ein Gemisch aus 8,5 g (4-7,4- mMol) Äthylcarbaminsäure—phenyl methylester,
$8,0 ml (0,47 Mol) wäßriges Formaldehyd, 6,6 g
• Kaliumcarbonat und 40 ml Dioxan' wird 3,5 Stunden unter Argon
als Schutzgas unter Rückfluß gekocht. Das abgekühlte .Reaktionsgemisch v/ird mit Äthylacetat verdünnt und mit Wasser, gesättigter
Katriumbicarbonatlösung und Kochsalzlösung gewaschen,

exotherme Reaktion statt. Nach 1 Stunde wird das Dichlormethan und das Phosphoroxichlorid unter vermindertem Druck
abdestilliert. Der Rückstand v/ird mit Toluol verdünnt und dann zur Entfernung von restlichem Phosphoroxichlorid eingedampft.
Der Rückstand wird in 10 ml wasserfreies Toluol aufgenommen und mit 5,8 g (1,2 Äqu.) Diäthyl-(4-phenylbutyl)-phosphonit
bei 25°C in einer Ärgon-Aümosphäre behandelt.
Das Reaktionsgemisch v/ird 1 Stunde unter Rückfluß gekocht
und dann zur Trockne eingedampft. Der Rückstand (etwa 11 g) wird an 30 g Kieselgel chromatographiert und mit Hexan/

Ausbeute: 3,2 g (10,0 mMol) ^Äthylamino)-methyl7-(4-phenylbutyl)-phosphinsäure-äthylester-hydrochlorid
als Öl, das bei längerem Stehen fost v/ird. Dünnschichtchromatograiam

12,0 ml (etwa 15 mMol) 12,5prozentige Lösung von Phosgen in Benzol und 20 ml wasserfreies Dichlormethan werden in
Argon als Schutzgas bei -200C (Tetrachlorkohlenstoff/Trokkeneis-Bad)
tropfenweise innerhalb von 10 Minuten mit einem Gemisch von 1,8 ml (16,6 mMol) E-Methylmorpholin, 2,0 g
(8,3 mMol) L-Prolin-phenylmethylester-hydrochlorid und 10 ml
Dichlormethan behandelt. Nach 30 Minuten Rühren bei -200C
wird das Bad entfernt und das Umsetzungsgemisch weitere
■ Minuten gerührt. Benzoldichlormethan und überschüssiges
Phosgen werden im Vakuum abdestilliert, wobei ein weißer Feststoff zurückbleibt. Der Rückstand wird in 15 ml wasserfreies
Dichlormethan aufgenommen und mit 2,6 ml (25 mMol) N-Methylmorpholin und 2,6 g (8,3 mMol) /rÄthylamino)-methyl7
(4-phenylbutyl)-phosphinsäure-äthylester-hydrochlorid bei
Raumtemperatur unter Argon behandelt. Nach 16 Stunden wird
das Reaktionsgeniisch mit Äthylacetat verdünnt, mit gesättigter
Natriumbicarbonatlösung, 5$ Kaliumbisulfatlösung und
Kochsalzlösung gewaschen, über Magnesiumsulfat getrocknet
■ und eingedampft. Der Rückstand (2,2 g) wird an 100 g Kie-25
selgel chromatographiert und mit einem Gemisch aus Hexan und
Aceton im Verhältnis 12:1 gefolgt von Hexan/Aceton im Verhältnis 1:1 eluiert. Ausbeute durch Eindampfen: 800 mg

als Katalysator wird in einer Parr-Apparatur bei einem Druck von 3»5 bar 1,5 Stunden hydriert. Der Katalysator
wird durch !Filtration im Celite-Bett abgetrennt und
das Methanol abgestreift. Es werden 600 mg Rückstand erhalten, der durch ein Siliciumdioxidkissen geschickt und mit
Dichlormethan/Methanol/Essigsäure im Verhältnis 100:5:5 zur Entfernung polarer Verunreinigungen eluiert wird. Nach dem
Eindampfen und der azeotropen Entfernung (!Toluol) restlicher Essigsäure werden 550 mg 1-^7Äthyl/7äthoxy(4-phenyrbutyl)-phosphinyl7-methyl7-amino7-carbonyl7-L-prolin
als Öl erhalten. Dünnschichtchronatogramm (Dichlormethan/Methanol/Essigsäure
im Verhältnis 100:5:5): einziger Fleck beim R„-Wert 0,6.

Ein Gemisch aus 550 mg (1,18 mMol) 1-/7Äthyl/2athoxy(4~phenylbütyl)-phosphinyl7-methyl7-amino7-carbonyl7-L-prolin
in 4 ml wasserfreies Dichlormethan wird mit 0,36 ml (2,0 £qu.) Tri-
2^ methylsilylbromid durch eine gasdichte Spritze unter Argon
als Schutzgas bei Raumtemperatur behandelt. Nach 2,5 Stunden wird Dichlormethan und überschüssiges Trimethylsilylbromid
unter vermindertem Druck abdestilliert. Der'Rückstand wird in Äthylacetat \md Wasser aufgenommen. Die Schichten werden
getrennt und die Äthylacetatschicht zweimal mit Wasser und mit Kochsalzlösung gewaschen, über Magnesiumsulfat getrocknet
und eingedampft. Der Rückstand (450 .mg) wird durch eine
30 ml Säule mit AG-50W-X2 (Li+) geschickt und mit V/asser
eluiert. Die gevrünschten !Traktionen v/erden vereinigt und
lyophilisiert. Das weiße Lyophilisat (400 mg) wird an einer Säule mit 200 ml HP-20 chromatographiert und mit Wasser —$
Acetonitril (0—>90#) eluiert. Das Wasser und Acetonitril
werden abgestreift und der Rückstand in Wasser aufgenommen, filtriert (Millipore) und lyophilisiert. Ausbeute: 340 mg

Ein GemiGch aus 5,2 g (25 mMol) Äthyl(hydroxymethyl)-carbaininsäure-phenylraethylester
in 20 ml wasserfreies Dichlormethan wird bei Raumtemperatur unter Argon als Schutzgas mit 5,7 S
(27,5 mMol) Phosphorpentachlorid behandelt. Es erfolgt eine exotherme Reaktion. Nach 1-' Stunde wird das Dichlormethan
und das Phosphoroxichlorid unter vermindertem Druck abdestilliert.
Der Rückstand wird mit Toluol verdünnt und dann zur Entfernung des restlichen Phosphoroxichlorids eingedampft.
Der Rückstand wird hierauf in 20 ml wasserfreies Toluol auxgenommen
und bei 250C unter Argon als Schutzgas mit 6,2 g (1,1 Äqu.) Diäthyl-(2-phenyläthyl)-phosphonit behandelt. Das
Reaktionsgemisch wird 3 Stunden unter Rückfluß gekocht und dann zur Trockne eingedampft. Der Rückstand (10,2 g) wird
an 350 g Kieselgel chromatographiert und mit Äthylacetat eluiert.
Ausbeute: 6,9 g (17,7 mMol) /7Äthoxy(2-phenyläthyl)-phosphinyl7-meth7/l7-äthylc^J?baminsäure-phenylmethylester.

25 ml wasserfreies Dichlormethan wird unter Argon als Schutzgas
bei -20°C (Trockeneis/Tetrachlorkohlenstoff) tropfenweise in 10 Minuten mit einem Gemisch von 4,4 ml (39 mMol)
N-Methylmorphoiin, 4,7 g (20 mMol) L-Prolin-phenylmethylester-hydrochlorid
und 10 ml Dichlormethan behandelt. Danach wird das Eisbad entfernt und das Gemisch 30 Minuten gerührt.
Das Dichlormethan, das Benzol und überschüssiges Phos gen werden unter vermindertem Druck abdestilliert. Der Rückstand
wird in Dichlormethan aufgenommen und mit 3,8 g (13 mMol) /CAthylamino)-methyl7~(2-phenyläthyl)-phosphinsäure-äthylester-hydrochlorid
und 4/1 ml (39 mMol) N-Methylmorpholin
unter Argon alü Schutzgas boii 2[)°C behandelt.

in 15 ml wasserfreies Tetrahydrofuran wird mit 0,24- g
(5 mMol) 50/0 Natriumhydrid-öldispersion behandelt und bei
Raumtemperatur 1 Stunde gerührt. Das Gemisch wird, dann mit
0,34-ml (5i5 mMol) Methyl j ο did behandelt und bei liaumtemperatur
v/eitere 3 Stunden gerührt. Das Gemisch wird hierauf zwischen Athylacetat und 5$ Kaliumbisulfatlösung verteilt.
Die organische Phase wird nacheinander mit gesättigter Katriumbicarbonatlösvmg
und gesättigter Kochsalzlösung gewaschen, über Natriumsulfat getrocknet und eingedampft. Der
Eückstand wird durch Entspannungs-Chromatographie an Kieselgel gereinigt und mit Aceton/Dichlormethan im Verhältnis
2:5 eluiert. Es wird der 1-/£Hethyl/7athoxy(4-phenylbutyl)-phosphinyl7-methyl7-amino7-carbonyl7-L-pI>olin-phenylmethyl-

Die Tabletten werden mit einer Lösung von Methocel E .15 (Methylcellulose) beschichtet, die als Farbstoff einen
gelb Nr. 6 enthaltenden Lack enthält.
In' ähnlicher V/eisc können Tabletten mit einem Gehalt von
50 mg an einer der Verbindungen gemäß Beispiele 1 bis 64-
und 66 bis 71 hergestellt werden.
Es werden zweiteilige Kr. 1 Gelatinekapseln, von denen jede
100 mg Dinatriumuals von 1-f/7Äthyl/7hydroxy(2-phenyläthyl)-ph.osphin3rl7-methyl7-cimino7-carbonyl7-L-prolin
enthält, mit

Wasser zur Injektion 5 Liter
Der Wirkstoff, die Konservierungsstoffe und das Natriumchlorid
werden in 3 Liter V/asser zur Injektion gelöst. Das Volumen wird dann auf 5 Liter gebracht. Die Lösung wird durch
ein keimfreies Filter filtriert und aseptisch in vorsterilisierte Fläschchen gefüllt, die mit vorsterilisierten Guonistopfen
verschlossen werden. Jedes Pläschchen enthält 5 eil
Lösung mit einer Konzentration von 100 mg Wirkstoff pro ml
30 Lösung zur Injektion.
In ähnlicher Weise können injizierbare Lösungen mit einem Gehalt von 100 mg Wirkstoff pro ml Lösung aus den Produkten
der Beispiele 2 bis 7^ hergestellt werden. 35

werden durch Verkluinpen des Dinatriumsalises von 1-/7Äthyl-/7Hydroxy(7I--pheny
lbutyl)-phosphiny^-methy^-amincy'-carbonyl/-L-prolin,
Avicel und eines Teils der Stearinsäure hergestellt. Die Klümpchen werden gemahlen und durch ein
Hr. 2-Öieb geschickt, dann mit dem Hydrochlorothiazid, der
Lactose, Maisstärke und dem Rest der Stecirinsäure vermischt
Das Gemisch wird in einer Tablettenpresse zu kapseiförmigen Tabletten der Kasse 550 mg verpreßt. Die Tabletten werden
zur Halbierung markiert.
In gleicher Weise können Tabletten mit einem Gehalt von
100 mg einer der Verbindungen gemäß Beispiele 1 bis 70 hergestellt werden.
