wie Arginin, Lysin und dergleichen. Die nichttoxischen, physiologisch verträglichen Salze sind bevorzugt. Andere
Salze sind jedoch ebenfalls nützlich, beispielsweise zur Isolierung oder Reinigung der Produkte. Die Salze werden
25 nach üblichen Verfahren hergestellt.
Das Symbol >k wird zur Bezeichnung verschiedener Asymmetriezentren
in den Verbindungen der Formel I verwendet. Die Verbindungen der Erfindung kommen in diastereomeren Formen
oder als Gemisch davon vor. In den vorstehend beschriebenen Verfahren können Razemate, Enantiomere oder Diastereomere

Die Verbindungen der vorstehenden allgemeinen Formel I und ihre Salze hemmen die Umwandlung des Decapeptids Angiotensin
I in Angiotensin II und vermindern oder beseitigen deshalb die durch Angiotensin II verursachte Hypertonie. Die erfindungsgemäßen
Verbindungen greifen in die Umsetzungsfolge
Angiotensinogen (Renin) ~* Angiotensin I ->
Angiotensin II ein, indem sie das ACE blockieren und dadurch die Entstehung der blutdruckerhöhenden Verbindung Angiotensin II vermindern
oder ausschalten.
Die Verabreichung von Verbindungen der allgemeinen Formel I
oder deren physiologisch verträglichen Salzen mildert die ■ durch Angiotensin II verursachte Hypertonie. Eine einzelne
Dosis oder vorzugsweise 2 bis 4 geteilte tägliche Gaben von etwa 0,1 bis 100 mg.
pro Tag sind geeignet zur Verminderung des Blutdrucks. Die Verbindungen werden vorzugsweise oral gegeben, sie können jedoch
auch parenteral, z.B. subcutan, intramuskulär, intravenös oder intraperetoneal in üblichen Darreichungsformen
verabfolgt werden.
Die Verbindungen der Erfindung können auch in Kombination mit einem DiuretUcuin zur Behandlung von Hochdruck konfektioniert
werden. Derartige Kombinationspräparate können in Dosen von etwa 30 bis βθθ mg, vorzugsweise 30 bis 300 rag einer
Verbindung der Erfindung und etwa 15 bis 300 mg, vorzugsweise etwa 15 bis 200 mg des Diuretikums pro Tag gegeben werden.
Beispiele für verwendbare Diuretika sind die Thiazid-Diuretika, wie Chlorthiazid, Hydrochlorthiazid, Flumethiazid,
Hydroflumethiazid, Bendroflumethiazid, Methychlothiazid,
Trichlormethiazid, Polythiazid und Benzthiazid, ferner Ethacrynsäure, Ticrynafen, Chlorthalidon, Fursosemid,
Musolimin, Buraetanid, Triamteren, Amilorid, Spironolacton
35 und Salze dieser Verbindungen.

Die Verbindungen der allgemeinen Formel I können zur oralen Gabe in Form von Tabletten, Kapseln oder Elixieren oder
zur parenteralen Gabe in Form von sterilen Lösungen oder Suspensionen konfektioniert werden. Etwa 10 bis 500 mg einer
Verbindung oder eines Gemisches von Verbindungen der allgemeinen Formel I wird mit einem physiologisch verträglichen
Träger, Excipiens, Bindemittel, Konservierungsmittel, Stabilisator
und Geschmackskorrigens In einer Einheitsdosis vermischt. Die Menge des Arzneistoffs in diesen Arzneipräparaten
ist derart, daß eine geeignete Dosis im angegebenen Bereich erhalten wird.
Gegenstand der Erfindung sind somit auch die Verbindungen der allgemeinen Formel I zur Verwendung bei der Behandlung
von Hypertonie, sowie Arzneimittel zur Behandlung von Hypertonie, die gekennzeichnet sind durch einen Gehalt an
einer Verbindung gemäß allgemeiner Formel I und übliche Träger-, Hilfs- und Zusatzstoffe,

Ein heterogenes Gemisch von 7»0 g (41,5 mMol) L-0rnithinhydrochlorid
und 200 ml Äthanol wird bei O0C im Eisbad tropfenweise
mit 4,5 ml (1,5 Äquivalente) Thionylchlorid behandelt und dann 3*5 Stunden unter Argon als Schutzgas unter
Rückfluß erhitzt. Das Äthanol, Schwefeldioxid und überschüssiges Thionylchlorid werden unter vermindertem Druck ab-

destilliert und der erhaltene kristalline Feststoff dreimal mit Diäthyläther digeriert, durch Filtrieren abgetrennt und

5,0 g (21,4 mMol) L-Ornithin-äthylester-hydrochlorid (1:2)
werden mit 42,8 ml (2,0 Äquivalente) 1M Natriumäthoxid bei 25°C unter Argon als Schutzgas behandelt. Nach 20 Minuten
wird das Äthanol abdestilliert, der Rückstand in Essigsäureäthylester aufgenommen und das Natriumchlorid durch ein
Celite-Bett abfiltriert. Der Essigsäureäthylester wird abdestilliert
und der erhaltene weißliche Feststoff mit Isopropyläther
digeriert, wobei 2,2 g (S)-3-Amino-2-oxopiperidin als weißer kristalliner Feststoff erhalten werden. Eine
kleine Menge davon wird aus Isopropyläther umkristallisiert, wobei feine weiße Nadeln vom F. 1O3-1O9°C (sehr hygroskopisch)
erhalten werden.
c) (S)-3-^7rPhenylmethoxy)-carbonyl7-amino7-2-oxopiperidin
Ein heterogenes Gemisch aus 1,7 S (14,9 mMol) (S)-3-Amino-2-oxopiperidin,
10 ml wasserfreies Tetrahydrofuran und 3,6 ml (1,4 Äquivalente) Diisopropyläthylamin wird bei O0C
(Eisbad) unter Argon als Schutzgas mit 2,5 ml (1,2 Äquivalente) Benzylchloroformat behandelt. Nach 3 Stunden wird das
Reaktionsgemisch mit Essigsäureäthylester verdünnt und nacheinander mit Wasser, 5# Kaliumbisulfatlösung und Kochsalzlösung
gewaschen, über Magnesiumsulfat getrocknet und eingedampft. Es werden 3,9 g roher weißer Feststoff erhalten.

Ein Gemisch aus 8,0 g (47,5 mMol) 4~Phenylbutylchlorid und
15,0 ml (-72 mMol) Triäthylphosphit wird bei einer Badtemperatur
von 185°C 4-1,5 Stunden unter Argon als Schutzgas unter
Rückfluß erhitzt. Nach der Destillation des Gemisches werden 10,8 g Diäthyl-(4—phenylbutyl)-phosphonat als farblose Flüssigkeit
vom Kp. 152-154-0C (1,0 mm Hg) erhalten.
Ein Gemisch aus 0,73 S (2,6 mMol) Diäthyl-(4~phenylbutyl)-phosphonat,
10 ml Benzol und 1 Äquivalent Phosphorpentachlorid wird unter Argon als Schutzgas 30 Minuten unter Rückfluß
erhitzt. Dann wird das Benzol und das Phosphoroxichlorid unter vermindertem Druck entfernt. Es wird das Äthoxy~(4~phenylbutyl
)-phosphinylchlorid erhalten.

Das Äthoxy-(4~phenylbutyl)-phosphinylchlorid von Teil (f)
wird in 5 πύ. wasserfreies Methylenchlorid aufgenommen. Der
3-Amino-2-oxopiperidinessigsäure-äthylester von Teil (e) wird ebenfalls in 5 ml Methylenchlorid aufgenommen und zu
der erstgenannten Lösung hinzugefügt. Die erhaltene Lösung wird im Eisbad auf O0C abgekühlt und tropfenweise mit
1,0 ml (1,5 Äquivalente) Triäthylamin in 2 ml Methylenchlorid
5 Minuten unter Argon als Schutzgas behandelt. Nach 30 Minuten wird das Eisbad entfernt und das Gemisch wird
eine weitere Stunde gerührt. Dann wird das Reaktionsgemisch mit Essigsäureäthylester verdünnt, nacheinander mit gesättigter
Natriumbicarbonatlösung, Wasser und Kochsalzlösung
gewaschen, über Magnesiumsulfat getrocknet und eingedampft. Der Rückstand (2,3 g) wird an 100 g Kieselgel chromatographiert
und mit Aceton/Hexan im Verhältnis 2:1 eluiert.
Ausbeute: 1,1 g (S)-3-(/2Athoxy-(4-phenylbutyl)-phosphinyl7·«
amino7-2-oxo~1-piperidinessigsäure-äthylester als öl.

Ein Gemisch von 1,1 g (2,6 mMol) Ester-Produkt von Teil (g),
7 ml wasserfreies Methylenchlorid und 0,6 ml (1,7 Äquivalente) Bromtrimethylsilan wird unter Argon als Schutzgas 40
Stunden bei Raumtemperatur gerührt. Dann wird das Methylenchlorid und überschüssiges Bromtrimethylsilan unter vermindertem
Druck abdestilliert und der Rückstand in 10 ml Dioxan aufgenommen und mit 9,1 ml (3,5 Äquivalente) 1N Lithiumhydroxid
behandelt. Es entsteht ein weißer Niederschlag, von dem jedoch der größte Teil wieder in Lösung geht, die
eine milchige Trübung annimmt. Nach 2 Stunden wird das Di oxan abdestilliert. Die heterogene Lösung wird filtriert und
das Filtrat an 200 ml HP-20 säulenchromatographiert und mit einem linearen Gradienten Wasser —>
Acetonitril (0 — 90$)
eluiert. Die das gewünschte Produkt enthaltende Fraktionen werden vereinigt, auf ein geringes Volumen eingeengt, durch
Millipore filtriert und lyophilisiert. Ausbeute: 480 mg (S)-3-/7Hydroxy-(4-phenylbutyl)-phosphinyl7-amino7-2-oxo-1-piperidinessigsäure-Dilithiumsalz

das Eisbad entfernt und das Reaktionsgemisch weitere 1,5 Stunden gerührt. Das Reaktionsgemisch wird mit Essigsäureäthylester
verdünnt und nacheinander mit gesättigter Natriumbicarbonatlösung,
5% Kaliumbisulfatlösung und Kochsalzlösung

5 a) N-(6-Bromhexyl)-phthalimid
Ein Geraisch aus 11,7 g (0,1 Mol) kristallines 6-Aminohexanol
und 14-,8 g (0,1 Mol) phthalsäureanhydrid wird 1,5 Stunden unter
Argon als Schutzgas auf 170 C erhitzt. Dann wird das entwickelte
Wasser unter Erwärmen und im Argonstrom abdestilliert.
Das Reaktionsgemisch wird auf 1000C abgekühlt und absatzweise
über einen gasdichten Siphon mit 7»2 ml (0,086 Mol)
Phosphortribromid versetzt. Bei jeder Zugabe erfolgt eine hef
tige Reaktion. Nach vollständiger Zugabe wird das Reaktionsgemisch weitere 30 Minuten auf 100°0 erhitzt. Nach dem Abküh-
len wird das Gemisch mit 20 ml Äthanol verdünnt und dann auf
Eiswasser gegossen und über Nacht im Kühlschrank aufbewahrt. Ein gelber Feststoff wird abfiltriert und mehrmals mit kaltem
Wasser gewaschen, bis das Filtrat nahezu neutral reagiert. Das Rohprodukt wird aus Äthanol umkristallisiert. Ausbeute:
21,0 R N-(6-Bromhexyl)-phthalimid als blaßgelber Peststoff
vom F. 54-550C.

25 b) (6-Phthalimidohexyl)-phosphonsäure-diäthylester
Ein, Gemisch aus 5»5 g (17»7 mMol) N-(6-Bromhexyl)-phthalimid
und 10,0 ml (58,4 mMol) Triäthy3phosphitwird unter Argon als
Schutzgas 16 Stunden unter Rückfluß erhitzt (Badtemperatur 160-165°C). Die flüchtigen Bestandteile werden durch Destillation
bei 100°C (Badtemperatur) und 0,5 mm Hg Druck entfernt, wobei ein blaßgelbes viskoses Öl zurückbleibt. Das
Rohprodukt wird durch Flash-Chromatographie an 100 g Kieselgel gereinigt und mit Aceton/Hexan im Verhältnis 1:2
eluiert. Ausbeute: 6,00 g (6-Phthalimidohexyl)~phosphonsäurediäthylester
als farbloses viskoses Öl. TLC (Aceton/Hexan 1:1): einzelner Fleck bei Rf = 0,40.

das Äthoxy-(6-phthalimidohexyl)-phosphinylchlorid erhalten.

Äthoxy-(6-phthalimidohexyl)-phosphinylChlorid und (S)-3-Amino-2-oxopiperidinessigsäure-äthylester
werden nach dem Verfahren von Beispiel 1 (g) umgesetzt. Es wird der (S)-3-/2Sthoxy-(6-phthalimidohexyl)-phosphinyl7-amino7-2-oxo-1-piperidinessigsäure-äthylester

abdekantiert. Der Rückstand wird mit Methylenchlorid digeriert und filtriert. Das vereinigte Piltrat
wird zur Trockne eingedampft. Es wird (S)-3-//T6-Aminohexyl)
äthoxyphosphinyl7-amino7-2-oxo-1-piperidinessigsäure-äthylester
erhalten.

Eine Lösung von 7,3 g (52,9 mMol) Diäthylphosphit in 100 ml wasserfreies Tetrahydrofuran wird mit 2,20 g (4-5,8 mMol) 50$ Dispersion von Natriumhydrid in öl in kleinen Mengen unter Argon als Schutzgas behandelt. Dann wird das Gemisch 30 Minuten unter Rückfluß erhitzt, auf Raumtemperatur abgekühlt und mit 6,9 g (15 mMol) 4-Methylbenzolsulfonsäure-2-^2r4--methylphenyl) -sulf onyl7-amino7-3-phenylpropyle st er behandelt. Nach 15 Minuten scheidet sich ein weißer Feststoff ab. Es werden weitere 75 ml Tetrahydrofuran zugegeben und weitere 15 Stunden gerührt. Danach wird das Gemisch 1 Stunde unter Rückfluß erhitzt, wieder abgekühlt und zwisehen 75 ml Essigsäureäthylester und 50 ml 5$ Kaliumbisulfatlösung verteilt. Die Essigsäureäthylesterphase wird nacheinander mit 5% Kaliumbisulfatlösung, gesättigte Natriumbicarbonatlösung und gesättigte Kochsalzlösung gewaschen, über Natriumsulfat getrocknet und eingedampft. Der Rückstand wird mit Hexan digeriert, wobei 5,9 g ^-^r^-Methylphenyl)-sulfonyl7-amino7~3-phenylpropyl7-phosphonsäurediäthylester als weißlicher Feststoff vom F. 86-89 erhalten
Eine Lösung von 7,3 g (52,9 mMol) Diäthylphosphit in 100 ml wasserfreies Tetrahydrofuran wird mit 2,20 g (4-5,8 mMol) 50$ Dispersion von Natriumhydrid in öl in kleinen Mengen unter Argon als Schutzgas behandelt. Dann wird das Gemisch 30 Minuten unter Rückfluß erhitzt, auf Raumtemperatur abgekühlt und mit 6,9 g (15 mMol) 4-Methylbenzolsulfonsäure-2-^2r4--methylphenyl) -sulf onyl7-amino7-3-phenylpropyle st er behandelt. Ein Gemisch von 5,9 g (13,9 mMol) </2-/7I>-Methylphenyl)-sulfonyl7-amino7-3~phenylpropyl7~phosphonsäurediäthylester,
8,0 g (85,1 mMol) Phenol und 50 ml 4-8$ wäßrige Bromwasserstoffsäure
wird 5,5 Stunden unter Rückfluß erhitzt. Nach dem Abkühlen wird das Gemisch mit 50 ml Wasser verdünnt und mit
zweimal 50 ml Essigsäureäthylester gewaschen. Die wäßrige Phase wird zur Trockne eingedampft, in 30 ml Wasser aufgenommen
und erneut eingedampft. Dieser Vorgang wird noch zweimal wiederholt. Schließlich wird der Rückstand in Wasser aufgenommen
und auf eine Säule mit AG 50 W - X2 in der H -Form aufgebracht (60 ml Bettvolumen) und zunächst mit Wasser und
dann mit 5$ Pyridin-Wasser eluiert. Die das gewünschte Produkt
enthaltenden Fraktionen werden vereinigt und zur Trockne eingedampft. Der feste Rückstand wird mit Acetonitril digeriert,
wobei 2,55 g (2-Amino~3-phenylpropyl)-phosphonsäure als weißlicher kristalliner Feststoff vom F. 34-7 C (Zers.)
erhalten werden.

d) (2-Phthalimido-3-phenylpropyl)-phosphonsäure
Ein Gemisch von 2,0 g (9,3 mMol) (2~Amino-3-phenylpropyl)-
phosphonsäure und 1,55 & (10,5 mMol) Phthalsäureanhydrid
wird in einem Kolben unter Argon als Schutzgas bei 195-20O0C
(Badtemperatur) 1,5 Stunden zusammengeschmolzen. Der glasartige dunkle Rückstand wird dann mit 25 ml Essigsäureäthylester
unter Rückfluß gekocht, bis er sich aufgelöst hat und ein flaumiger kristalliner Feststoff ausfällt. Das
abgekühlte Gemisch wird mit 25 ml Diäthyläther verdünnt und filtriert. Der Feststoff wird sorgfältig mit Diäthyläther

zugesetzt. Das Gemisch wird bei Raumtemperatur mehrere Stunden gerührt und dann unter vermindertem Druck
über Nacht eingeengt. Der Rückstand wird in Methylenchlorid gelöst und mit Triethylamin und Wasser behandelt und erneut
unter vermindertem Druck eingedampft. Der Rückstand wird in

Chloroform aufgenommen und mit Triäthylamin, 1-Chlorisobutylpropanoat,
Natriumchlorid und Tetrabutylammoniumjodid behandelt.
Das Gemisch wird bei Rückflußtemperatur etwa 15 Stunden gerührt. Dann wird das Reaktionsgemisch unter vermindertem
Druck eingedampft und der Rückstand mit Diäthyläther versetzt. Die wasserlöslichen Peststoffe, die sich aus

(o —> 90$) eluiert. Die das gewünschte Produkt enthaltenden
Fraktionen werden vereinigt, unter vermindertem Druck eingedampft und der Rückstand in Wasser gelöst, filtriert und
lyophilisiert. Es wird das Monolithiumsalz von (S)-Hexahydro-5-/772-methyl-1

Das Dinatriumsalz der (S)-3-^Sydroxy-(4-phenylbutyl)-phosphinyl7-amino7-2-oxo-1-piperidinessigsäure

Es werden 1000 Tabletten aus folgenden Bestandteilen hergestellt:
Dinatriumsalz von (S)-Hexahydro-3-
/7Hydroxy-( 4~phenylbutyl)-phosphinyl7-
amino7-2-oxo-1H-azepin-1-essigsäure 50
Lactose 25
Avicel 38
Maisstärke 15
Magnesiumstearat 2

Das Dinatriumsalz von (S)~Hexahydro-3-^7Eydroxy-(4--phenylbutyl)-phosphinyl7-amino7-2-oxo-1H-azepin-1-essigsäure,
die Lactose und das Avicel werden vermischt. Dann wird die Maisstärke untergemischt. Das Magnesiumstearat wird zugegeben
und das trockne Gemisch in einer Tablettenpresse zu 1000 Tabletten verpreßt, die jeweils 50 mg Wirkstoff enthalten.
Die Tabletten werden mit einer Lösung von Methocel E 15
(Methylcellulose) beschichtet, die als Farbstoff einen Yellow Nr. 6 enthaltenden Lack aufweist.

In ähnlicher Weise können Tabletten mit einem Gehalt von
50 mg Produkt von einem der Beispiele 1 und 3 bis 45 hergestellt
werden.
1S Beispiel 49
Zwei Gelatinekapseln Nr. 1 werden mit einem Gemisch folgender Bestandteile gefüllt:
Dinatriumsalz von (S)-3-^ZZ""2-(Benzoylamino)-3-phenylpropyl7-hydroxyphosphinyl7~amino7-
20 2-oxo-1-piperidinessigsäure 100 mg
Magnesiumstearat 7 mg
Lactose 193 mg
In ähnlicher Weise können Kapseln, die 100 mg eines Produktes von irgendeinem der Beispiele 1 bis 27 und 29 bis 46
enthalten, hergestellt werden.
Beispiel 50 Es wird eine injizierbare Lösung folgender Zusammensetzung

Dinatriurasalz von (S)-3-{/7Sydroxy-(4-phenylbutyl)-phosphinyl7-amino7-2-oxo-1-piperidinessigsäure
500 g
Methylparaben 5 g
35 Propylparaben 1 g
Natriumchlorid 25 g
Wasser zur Injektion 5 Liter

Der Wirkstoff, die Konservierungsmittel und das Natriumchlorid werden in 3 Liter Wasser zur Injektion gelöst.
Dann wird das Volumen auf 5 Liter aufgefüllt. Die Losung
wird durch ein steriles Filter filtriert und aseptisch in vorsterilisierte Ampullen abgefüllt, die mit vorsterilisier
ten Gummistöpseln verschlossen werden. Jede Ampulle enthält 5 ml Lösung in einer Konzentration von 100 mg Wirkstoff pro
ml Lösung zur Injektion.
In ähnlicher Weise können in^izierbare Lösungen mit einem
Gehalt von 100 mg Wirkstoff pro ml Lösung aus den Produkten irgendeines der Beispiele 2 bis 46 hergestellt werden.
Es werden 1000 Tabletten aus folgenden Bestandteilen hergestellt:

Avicel 100
Hydrochlorothiazid 12,5

Maisstärke 17,5
25 Stearinsäure 7
Zur Herstellung wird das Dinatriumsalz von (S)-Hexahydro-3-/7ßydroxy-(4-phenylbutyl)-phosphinyl7-amino7-2-oxo-1H-azepin-1-essigsäure,
das Avicel und ein Teil der Stearinsäure vermischt. Das Gemisch wird gemahlen und durch ein
Sieb Nr. 2 geführt,dann mit dem Hydrochlorothiazid, der Lactose, der Maisstärke und dem Rest der Stearinsäure vermischt.
Das Gemisch wird in einer Tablettenpresse zu 350 mg schweren kapseiförmigen Tabletten verpreßt. Die Tabletten
werden gekerbt, um sie in Hälften zu teilen.

In ähnlicher Weise können Tabletten mit einem Gehalt von 100 mg eines der Produkte von irgendeinem der Beispiele 1
und 3 bis 4-6 hergestellt werden.
