Description
Die vorliegende Erfindung betrifft
die Verwendung von substituierten 1,4-Benzochinon bei der
Prophylaxe oder Behandlung von Fibrose in Säugern.
Im Zusammenhang mit den außerordentlich hohen Abla
gerungen von Collagen in den Geweben von Menschen und
Tieren im Verlauf solcher Krankheiten wie Leber- und
Lungenfibrosen wurden die Inhibitoren der Collagen-
Biosynthese untersucht und auf ihre mögliche Verwen
dung als Arzneimittel für die Prophylaxe und Behand
lung solcher Krankheiten überprüft. Allerdings haben
die bisher aufgefundenen Inbibitoren der Collagen-
Biosynthese den Nachteil, daß sie aufgrund der ihnen
eigenen Toxizität, so z. B. der Toxizität aufgrund
ihres Gehaltes an β-Aminopropionitril, nicht wieder
holt verabreicht werden können.
Das Forschungsvorhaben, das zur Entwicklung einer die
Collagen-Biosynthese inhibierenden Verbindung durch
geführt wurde, die sich gut für die wiederholte Verab
reichung eignet, führte zu dem Befund, daß Verbindun
gen der folgenden allgemeinen Formel (I) derartige die
Collagen-Biosynthese inhibierende Aktivität und eine
niedrige Toxizität haben:
In der Formel (I) ist A eine Methylengruppe oder eine
Carbonylgruppe, n ist eine ganze Zahl von 2 bis ein
schließlich 21 und R bedeutet ein Wasserstoffatom oder
eine niedere Alkylgruppe.
Aus der EP-A 0 031 727 ist die Verwendung von Verbin
dungen der Formel (I) zur Behandlung von ischämischen
Krankheiten bekannt. Darüber hinaus ist aus der DE-OS-
25 19 730 bekannt, daß diese Verbindungen eine Wirkung
auf die Lysosomalmembranen der Zellen und insbesondere
immunitätsverstärkende Wirkung haben. Ferner ist ihre
Zellmetabolismus verbessernde Aktivität aus der EP-A-0 021 841 bekannt.
Die Erfindung betrifft daher die Verwendung von
Verbindungen der allgemeinen Formel (I) bei der Pro
phylaxe oder Behandlung von Fibrose aufgrund einer
übermäßig hohen Collagen-Akkumulation in Säugern, wo
bei man einem Säuger eine wirksame Menge einer Ver
bindung der Formel (I) verabreicht.
In der oben angegebenen allgemeinen Formel (I) ist die
niedere Alkylgruppe R ein Rest mit 1 bis 3 C-Atomen,
wie z. B. Methyl, Ethyl usw., n ist bevorzugt eine gan
ze Zahl von 7 bis einschließlich 11, und A ist bevor
zugt eine Methylengruppe. Wenn A eine Methylengruppe
ist, kann die endständige Hydroxygruppe, die durch OR
symbolisiert wird, durch eine Schutzgruppe wie eine
Acetylgruppe geschützt werden. Als Beispiele für Ver
bindungen der Formel (I), in der A eine Methylengruppe
und OR eine geschützte Hydroxygruppe sind, werden ge
nannt: 6-(10-Acetoxydecyl-)2,3-dimethoxy-5-methyl-1,4-
benzochinon, 6-(9-Acetoxynonyl-)2,3-dimethoxy-5-methyl-
1,4-benzochinon und dergleichen.
Die Verbindungen der allgemeinen Formel (I) zeigen
Aktivitäten in der Inhibierung von Protocollagen-Pro
lin-Hydroxylase und Aktivitäten in der Inhibierung der
Collagen-Biosynthese. Dies wird im Folgenden beschrie
ben.
Die inhibierende Wirkung wurde getestet nach dem Ver
fahren von R.E. Rhoads et al. (Methods in Enzymology
XVII B, 306 (1971)) unter Benutzung einer teilweise
gereinigten Enzymzubereitung von Hühnerembryos in
Übereinstimmung mit den Methoden von K.I. Kivirriko et
al. und J. Halme et al. (J. Biol. Chem. 242, 4007
(1967) und Biochem. Biophys. Acta 198, 460 (1967)) mit
(Pro-Pro-Gly)5 · 4H2O (Protein Research Foundation,
Osaka) als Substrat. In diesem Test wurde die teil
weise gereinigte Enzymzubereitung als Protein in einer
Menge von 100 µg verwendet.
Im Grunde in Übereinstimmung mit der Methode von R.A.
Salvador et al. (Arch. Biochem. Biophys. 174, 382
(1976)), wurden 0,2 mg/kg jeder Testverbindung intra
peritoneal an Ratten eines SD Stammes (weiblich, 3
Wochen alt) einmal täglich an 6 aufeinanderfolgenden
Tagen verabreicht und der Collagengehalt des Uterus
mit dem Kontrollwert verglichen. Das Ergebnis zeigt
Tabelle 2.
Die Verbindung (I) zeigt eine sehr niedrige Toxizität.
Beispielsweise verursachte die Verabreichung einer
Verbindung (I) (n=9, A=CH2, R=H) durch Wasserverabrei
chung mit der Magensonde an Ratten mit einer Aufnahme
von 2500 mg/kg und Tag innerhalb von 5 Wochen, keine
Abweichungen in Körpergewicht, Nahrungsaufnahme, Harn
analyse und Bluttests.
Die Verbindungen (I) werden erfindungsgemäß
verwendet für die
Prophylaxe und Behandlung von Fibrose der Organe bei
Tieren und insbesondere Säugetieren und Säugern, z. B.
Laboratoriumstieren wie Kaninchen, Ratten, Mäusen
usw., Haustieren wie Hunden, Katzen usw. und bei Men
schen. Fibrose ist der allgemeine Name der Krankheit
und bezeichnet alle Erkrankungen, die aufgrund einer
übermäßig hohen Akkumulation von Collagen in Geweben
auftreten. Es schließt solche Krankheiten wie Lungen
fibrose, Hepatozirrhose, Arteriosklerose, Skleroderma,
Myelofibrose, chronische Arthritis, rheumatoide
Arthritis usw. ein, ist jedoch nicht auf diese be
schränkt.
Für die Verwendung als prophylaktisches Mittel oder
Heilmittel bei Fibrose können die Verbindungen (I)
gemäß dieser Erfindung oral oder auch in anderer Form
verabreicht werden, entweder in Substanz oder in For
mulation mit geeigneten physiologisch verträglichen
Trägern oder Verdünnungsmitteln in solchen Dosier-For
men wie Pudern, Körnchen, Tabletten, Kapseln, Injek
tionslösungen usw. Die Dosierung ist abhängig von Fak
toren wie Art der Krankheit, den Symptomen, Versuchs
person, Art der Verabreichung oder Formen der Dosie
rung. Im Falle der parenteralen Verabreichung wie z. B.
der Injektion ist die tägliche Dosis der Verbindung
ca. 25 bis 500 mg (0,5 bis 10 mg/kg), bevorzugt 50
bis 250 mg (1,0 bis 5 mg/kg) für einen erwachsenen
Menschen. Im Falle der oralen Verabreichung ist die
tägliche Dosis ca. 500 bis 5000 mg (10 bis 100 mg/kg),
bevorzugt 1000 bis 2500 mg (20 bis 50 mg/kg) für einen
erwachsenen Menschen.
Die erfindungsgemäß zu verwendende Arzneimittelzubereitung
enthält das Arzneimittel in Form einer Dosiereinheit.
Unter einer Dosiereinheit ist das Medikament zu ver
stehen, das eine Tagesdosis der Verbindung (I) ent
hält, wie sie oben beschrieben ist, oder ein Viel
faches davon (bis vierfaches) oder ein Bruchteil davon
(bis zu 1/40), das in Form physikalisch voneinander
getrennter einzelner Einheiten vorliegt und für die
Verabreichung als Medizin geeignet ist. Jede Dosie
rungseinheit enthält generell 0,3 bis 250 mg der Ver
bindung (I). Dabei enthält eine Injektionsampulle be
vorzugt 0,3 bis 30 mg und jede andere Form enthält be
vorzugt 10 bis 250 mg der Verbindung (I).
Die Verbindungen der allgemeinen Formel (I), in denen
n eine ganze Zahl von 2 bis 9 ist, wurden beschrieben
in der US-PS 4 139 545. Die Verbindungen (I), in denen
n eine ganze Zahl von 10 bis 21 ist, wurden beschrie
ben in der EP-Patentveröffentlichung 21 841.

Nach einem gängigen pharmazeutischen Verfahren wurden
die obigen Komponenten gemischt und in Gelatinekapseln
gefüllt. Man erhielt eine Arzneimittelzubereitung in
Kapseln.

Die oben angegebenen Komponenten wurden gemischt und
die Lösung wurde nach einem gängigen pharmazeutischen
Verfahren in Weichkapseln eingefüllt.
Claims (7)
1. Verwendung von Verbindungen der allgemeinen Formel (I)
in der A eine Methylen- oder Carbonylgruppe, n eine ganze Zahl
von 2 bis einschließlich 21 und R Wasserstoff oder ein niederer
Alkylrest sind, bei der Prophylaxe oder Behandlung von Fibrose
aufgrund übermäßig hoher Akkumulation von Collagen in Säugern.
2. Verwendung nach Anspruch 1, wobei n eine ganze Zahl
von 7 bis einschließlich 11 ist.
3. Verwendung nach Ansprüchen 1 bis 2, wobei A eine
Methylengruppe und n 9 ist.
4. Verwendung nach Ansprüchen 1 bis 3, wobei die Ver
bindung der Formel (I) 2,3-Dimethoxy-5-methyl-6-(10′-
hydroxydecyl-)1,4-benzochinon ist.
5. Verwendung nach Ansprüchen 1 bis 4, wobei die Fi
brose Lungenfibrose, Hepatozirrhose, Arteriosklerose,
Skleroderma oder rheumatoide Arthritis ist.
6. Verwendung nach Ansprüchen 1 bis 5, wobei die Ver
bindungen parenteral in einer Dosis von 0,5 bis 10
mg/kg Körpergewicht und Tag verabreicht werden.
7. Verwendung nach Ansprüchen 1 bis 5, wobei die Ver
bindungen oral in einer Dosis von 10 bis 100 mg/kg
Körpergewicht und Tag verabreicht werden.
