Description
Die Erfindung betrifft den Gegenstand des Patentanspruchs.
Zu den Derivaten des Pyrrolidin-2-ons der allgemeinen
Formel I
und den entsprechenden Pyrrolidinen der allgemeinen
Formel II
worin R1-5 Wasserstoff (außer bei R₄) oder (ggf.
substituierte) Aryl- bzw. Alkylgruppen sind, gehören
viele bekannte Verbindungen, die interessante
ZNS-Aktivität entwickeln (Archivum Immunologiae et
Therapiae Experimentalis 1975, 23, 733-751). Viele
Pyrrolidin-2-on (I) zeigen prostaglandinähnliche
Aktivität (DE-OS 25 27 989).
Die Pyrrolidin-2-on-Derivate (I) wurden üblicherweise
durch Ringschluß der entsprechenden 4-Aminobuttersäure
oder -Ester der Formel III
und 4-Halogen-Butyrylamid, durch Reaktion der Laktone
mit Aminen oder Hydrolyse von 2-Imino-pyrrolidinen
(GB-PS 13 50 582 und US-PS 40 12 495) gewonnen.
Diese Verfahren haben meist nur sehr beschränkte
Ausbeute, sind hinsichtlich der Substituenten und
der verschiedenen Stellungen beschränkt, und zeitigen
Schwierigkeiten in der Herstellung reiner Stereo-
und optisch-aktiver Isomere.
Die Bechaffung geeigneter Ausgangsmaterialien ist
schwierig.
In einfacher, unaufwendiger Weise sind 3,4-cis-
oder trans-substituierte, wahlweise optisch aktive
Pyrrolidin-2-on-Derivate und entsprechende Pyrrolidine
durch Hydrierung von 3-Pyrrolin-2-on-Derivaten
der allgemeinen Formel V
erhältlich, wobei R³, R⁴, R⁵ sowie R⁵′ in gleicher
Weise wie obenstehend definiert sind, wobei ein geeigneter
Katalysator und während der Hydrierung
nach Wahl ein optisch aktives (Angew. Chem. 83, 956
(1971)) oder eingebautes asymmetrisches Zentrum von
C-5 von (V) verwendet werden (DE-OS 29 23 553).
Eine der Hauptschwierigkeiten bei der Herstellung
der Pyrrolidin-2-on-Derivate (I) durch Hydrierung
der 3-Pyrrolin-2-on-Derivate (V) besteht in der
Schwierigkeit, solche 3-Pyrrolin-2-on-Derivate
(V) zu bilden.
Einige 3-Pyrrolin-2-on-Derivate (V) werden gemäß
US-PS 32 72 842 in folgender Weise präpariert:
Dabei sind R⁵ und R⁵′ nicht Wasserstoff und R⁴ ist
stets S-CH₂-(S=Substituent). Der Ringschluß gemäß
der US-PS 32 72 842 läuft, wie Versuche der Anmelderin
ergaben, bei R₁=H und R₃=H, Alkyl oder substituiertes
Alkyl nicht ab, was als Ergebnis der unterschiedlichen
Substituenten betrachtet werden
kann; dies Verfahren ist daher auf ein ganz bestimmtes
Substitionsmuster beschränkt.
Nach G. Stork und R. Matthews, Chemical Communications
1970, 445-6 wurden 3-Pyrrolin-2-on-Derivate
(V) nach folgender Reaktion erzeugt.
Die Verwendung von Dicyclohexylcarbodiimid (DCC)
für die Kondensation des Aminoketon-Derivats (VI)
mit der instabilen Phosphonessigsäure (VII) ist für
eine Groß-Synthese ungeeignet. Außerdem ist DCC
sehr teuer und der N, N′-Dicyclohexyl-Harnstoff ist
ein Nebenprodukt, dessen Beiseitigung schwierig ist,
so daß das Verfahren unwirtschaftlich ist.
Eine ähnliche Ringschluß-Reaktion wurde von T. W.
Guntert et al. in Helv. Chim. Acta 60, 334-9 (1977)
vorgeschlagen.
Jedoch erfordert dieses Verfahren eine wiederholte
chromatografische Trennung zur Isolierung des
3-Pyrrolin-2-on-Derivats (V) mit R⁴=17 β-Steroidrest
mit 10%iger Ausbeute, so daß dieses Verfahren
für eine Groß-Synthese ungeeignet ist, wobei außerdem
noch darauf hinzuweisen ist, daß die Autoren
die Ausgangs-Bromverbindung von (IX) nicht herstellen
konnten.
Aufgabe der Erfindung ist es, ein wirtschaftliches
Verfahren zur Herstellung am Stickstoff durch Wasserstoff
substituierter 3-Pyrrolin-2-on-Derivate
anzugeben.
In den nach dem erfindungsgemäßen Verfahren herstellbaren
Verbindungen hat der Alkylrest C₁-C₁₆
Kohlenwasserstoffatome und die Substituenten (bis
zu 5 gleichen oder gemischten) bedeuten
Alkyl, Halogenalkyl, (z. B. F₃C), O-Alkyl,
N-Dialkyl (gleich oder verschieden), S-Alkyl,
Halogen, O-Benzyl, N-Dibenzyl, N-Alkyl/Benzyl,
S-Benzyl, N-Alkyl, OH, NH₂ und SH (wobei die
letzten drei während des basischen Ringschlusses
geschützt sind).
Für die Herstellung von Verbindungen
der allgemeinen Formel (V) durch Ringschluß-Reaktion für
den Fall, daß R¹=Wasserstoff und R³=Wasserstoff,
Alkylrest oder substituierter Alkylrest ist,
hat es sich als am besten erwiesen, eine Reaktion
mit 2-Amino-Acetophenon gemäß der allgemeinen
Formel XI:
mit einem 2-Brom-Acylchlorid oder -bromid zur Bildung
der entsprechenden Bromverbindung (XII) der
allgemeinen Formel
(in fast quantitativer Ausbeute) durchzuführen. Die
Bromverbindung (XII) reagiert im Gegensatz zur
Chlorverbindung (IX) zwischen Raumtemperatur und
50°C mit Triphenylphosphin, z. B. in Benzol oder
Chlorbenzol-Lösung, ohne Zersetzung und bildet das
entsprechende Phosphoniumbromid (XIII) der allgemeinen
Formel:
das in Alkoholen und Wasser für die Ringschluß-Reaktion
zur Bildung der entspechenden Verbindung (V)
beispielsweise mit 2n NaOH-Lösung ausreichend löslich
ist.
Somit sind entsprechend diesem Verfahren die Ausgangs-Aminoketone
(XI) im allgemeinen leicht verfügbare
Verbindungen, die in praktisch quantitativer
Ausbeute in schwach basischem Medium, z. B. Natriumbicarbonat
in einer wäßrigen Äthermischung, mit
2-Bromacyl-Chloriden oder -Bromiden zu dem entsprechenden
N-(2-Bromacyl)-Aminoketon (XII) acyliert
werden können, wobei R₃ und R₅ die genannten Bedeutungen
haben.
Die Ketone gemäß obiger Formel (XII) reagieren bei
Raumtemperatur mit Triphenylphosphin z. B. in Benzollösung
und bilden das entsprechende Phosphoniumbromid
(XIII), das ausfällt und durch einfaches Filtrieren
gewonnen werden kann. Die Lösung von (XIII)
in Alkohol (vorzugsweise Methanol) oder Wasser und
die Behandlung mit mehr als einem Äquivalent einer
Base (vorzugsweise 2n NaOH-Lösung) bei Raumtemperatur
bildet schnell das gewünschte 3-Pyrrolin-2-
on-Derivat (V) und Triphenylphosphinoxid in praktisch
quantitativer Ausbeute, das aus der Lösung
ausfällt. Nach Verdünnnen mit Wasser und Verdampfen
des Alkohols wird die Mischung gesammelt und durch
Extraktion z. B. mit Methylenchlorid separiert, das
das Triphenylphosphinoxid entfernt.
Das auf diese Weise hergestellte 3-Pyrrolin-2-on-Derivat
(V) kann leicht mit beispielsweise Pd/C, Pt
oder chiralen Rhodiumkomplexen als Katalysator in
Alkohol (vorzugsweise Methanol-Lösung) zu dem ZNS-
aktiven substituierten Pyrrolidin-2-on-Derivat
(I-cis) hydriert werden. Diese Reaktionsfolge hat
den Vorteil, daß Derivate erhalten werden, bei denen
R³ und R⁴ ausschließlich als cis-Substituenten
vorliegen. Die (I-cis) Verbindungen können in die
entsprechenden (I-trans) Verbindungen durch Basen-
oder Säurebehandlung umgesetzt werden.
Ein zusätzlicher Vorteil der Erfindung besteht darin,
daß die Reduktion der Pyrrolidin-2-on-Derivate
(I) z. B. mit B₂H₆ oder LiAlH₄ zur Bildung neuer Pyrrolidine
(II-cis (R³ und R⁴ cis) oder II-trans (R³
und R⁴ trans)) führt, je nach dem Ausgangsmaterial.
Die nachfolgenden Beispiele einer erfindungsgemäßen
Synthese eines 3-Pyrrolin-2-on-Derivates und seiner
Weiterverarbeitung zum entsprechenden Pyrrolidin
erläutern den Erfindungsgedanken.
Eine Lösung von 51,16 g (0,609 Mol) Natriumcarbonat
in 550 ml Wasser wurde unter Stickstoff bei 0°C bis
10°C einer zweiphasigen Lösung aus 48,5 g (0,203 Mol)
2-Amino-4′-trifluormethylacetophenon-Hydrochlorid
in 250 ml Wasser und 250 ml Äther zugegeben. Eine
zweite Lösung von 18,41 ml (0,223 Mol) Bromacetylchlorid
in 200 ml trockenem Äther wurde bei 0
bis 5°C innerhalb 15 Minuten zu der obigen umgerührten
Suspension gegeben. Die zu Beginn dicke Suspension
wurde dünner. Nach zweistündigem Rühren wurde Äthylacetat
zugegeben, bis eine klare zweiphasige Lösung
erhalten wurde. Das Wasser wurde abgetrennt und die
Äthylacetat-Ätherlösung mit Wasser bis zur Neutralität
gewaschen. Eindampfen im Vakuum ergab 62,3 g
(95%) N-[(4'Trifluormethylbenzoyl)methyl]-2-bromacetamid.
Eine rekristallisierte Probe besaß einen
Schmelzpunkt von 201-11°C.
70,5 g (0,269 Mol) Triphenylphosphin wurden einer
Suspension von 62,36 g (0,192 Mol) des Produktes
von Beispiel 1 in 600 ml Benzol zugegeben. Nach 4
Tagen Rühren bei Raumtemperatur wurde der Feststoff
aufgesammelt und in 250 ml Aceton 3 Stunden lang
suspensiert. Filtrieren ergab 77,66 g (69%) N-[(4′-
Trifluormethylbenzoyl)-methyl]-2-(triphenylphosphonium-bromid)-
acetamid mit einem Schmelzpunkt von
250-1°C.
72,5 ml einer 2n NaOH-Lösung wurden langsam unter
Stickstoff einer Lösung von 77,6 g (0,132 Mol) des
Produktes von Beispiel 2 in 780 ml Methanol zugegeben,
wobei die Temperatur unter 40°C gehalten wurde.
Nach Umrühren während einer Stunde wurden 15 ml
einer 2n Salzsäurelösung hinzugegeben, so daß ein
pH-Wert von etwa 6,5 erreicht wurde. Das Methanol
wurde teilweise im Vakuum verdampft und nach Zugabe
von 20 ml Wasser vollständig entfernt. Der Niederschlag
wurde aufgenommen und mit Wasser gewaschen.
Nach Trocknen wurde der Rest, gewogen 67,25 g, in
200 ml Methylenchlorid 2 Stunden lang suspendiert.
Eine Filtration ergab 27,3 g (91%) 4-(4′-Trifluormethylphenyl)-
3-pyrrolin-2-on mit einem Schmelzpunkt
von 208-20°C (Zersetzung).
Eine Lösung von 27,2 g (0,12 Mol) des Produktes von
Beispiel 3 in 500 ml Methanol (teilweise suspendiert)
wurde 10 Stunden lang mit 4,1 g 10% Pd auf
Holzkohle als Katalysator hydriert. Filtration und
Verdampfung im Vakuum ergab 27,5 (100%) 4-(4-Trifluormethylphenyl)-
pyrrolidin. Eine rekristallisierte
(Aceton-Äther/1 : 2) Probe besaß einen Schmelzpunkt
von 121-2°C. Analyse für C₁₁H₁₀F₃NO:
experimentell:
C 57,65‰ H 4,38% N 6,35%;
berechnet:
C 57,64% H 4,40% N 6,11%.

Claims (1)
1. Verfahren zur Herstellung von 3-Pyrrolin-2-onen
der allgemeinen Formel
dadurch gekennzeichnet, daß man ein Phosphonium-bromid
der allgemeinen Formel
in dem
R³ ein Wasserstoffatom, Alkylrest oder substituierter Alkylrest;
R⁴ ein Phenylrest oder substituierter Phenylrest;
R⁵ und
R⁵′ ein Wasserstoffatom, Alkylrest oder substituierter Alkylrest bedeuten
und wobei R⁵ und/oder R⁵′ ein Wasserstoffatom ist, einer Ringschluß-Reaktion in basischer Umgebung unter Stickstoff unterwirft.
R³ ein Wasserstoffatom, Alkylrest oder substituierter Alkylrest;
R⁴ ein Phenylrest oder substituierter Phenylrest;
R⁵ und
R⁵′ ein Wasserstoffatom, Alkylrest oder substituierter Alkylrest bedeuten
und wobei R⁵ und/oder R⁵′ ein Wasserstoffatom ist, einer Ringschluß-Reaktion in basischer Umgebung unter Stickstoff unterwirft.
