- hamburg Substituierte 4-Aryl-2, 5-dihydro-furan-2-one und 4-Aryl-1 , 5-dihydro-2H-pyrrol-2-one und Verfahren zu ihrer Herstellung Gegenstand der Erfindung sind neue substituierte 4-Aryl-2,5-dihydrofuran-2-one und 4-Aryl-1,5-dihydro-2H-pyrrol-2-one der allgemeinen Formel (I) in der R1 und R2, die gleich oder verschieden sein können, ein Wasserstoff- oder Halogenatom, eine Alkyl- oder Acylaminogruppe mit 1 bis 6 Kohlenstoffatomen oder eine Carbamoylmethylgruppe, und R4, die gleich oder verschieden sein können, ein Wasserstoffatom oder eine Methylgruppe, ein Wasserstoffatom, eine Dimethoxyphenylgruppe oder eine Acylaminogruppe .-der allgemeinen Formel (II) R6-CONH- (II) worin R6 für eine Alkylgruppe mit 1 bis 4 Kohlenstoffatomen steht, und A ein Sauerstoffatom, eine Imino- oder Alkyliminogruppe mit 1 bis 4- Kohlenstoffatomen bedeutet und deren physiologisch verträgliche Säureadditionssalze sowie Verfahren zu ihrer Herstellung und ihre Verwendung in pharmazeutischen Präparaten.
- Ferner betrifft die Erfindung neue substituierte 4-Aryl-2,5-dihydro-furan-2-one und 4-Aryl-1,5-dihydro-2H-pyrrol-2-one der allgemeinen Formel (III) in der R7 eine Wasserstoffatom, eie Methyl- oder Benzylgruppe, den Rest oder bedeutet und worin X für ein Chlor- oder Bromatom steht und Verfahren zu ihrer Herstellung sowie ihre Verwendung als Zwischenprodukte für die Herstellung der 4-Aryl-2,5-dihydrofuran-2-one und 4-Aryl-1,5-dihydro-2H-2-one der Formel (I).
- Die erfindungsgemäßen Verbindungen der allgemeinen Formel (I) und ihre Säureadditionssalze besitzen wertvolle therapeutische Eigenschaften. Sie zeichnen sich durch ß-adrenolytische und blutdrucksenkende Wirkung aus und können daher zur Behandlung von Angina pectoris, Hypertonie und Arrhythmien eingesetzt werden. Die Dosierung beträgt beim Menschen etwa 15 -200 mg pro Tag.
- Die Verbindungen können als solche oder in Form ihrer Säureadditionssalze, gegebenenfalls mit geeigneten festen oder flüssigen pharmakologisch verträglichen Trägerstoffen und/oder Verdünnungsmitteln gebräuchlicher Art vermischt, zur Herstellung von Lösungen für Injektionszwecke und insbesondere von peroral zu verabreichenden pharmazeutischen Präparaten, wie Dragees, Tabletten oder Liquida, verwendet werden. Geeignete Trägerstoffe sind beispielsweise Milchzucker (Lactose), Gelatine, Maisstärke, Stearinsäure, äthanol, Propylenglykol, Äther des Tetrahydrofurfurylalkohols und Wasser.
- Das Verfahren zur Herstellung der Verbindungen der Formel (I) ist dadurch gekennzeichnet, daß man die neuen Verbindungen der Formeln (1V) oder (V) worin R1, R², A und ,X die obengenannte Bedeutung haben, mit einem Amin der allgemeinen Formel (VI) in der R3, R4 und R5 die obengenannte Bedeutung haben, umsetzt, indem man eine Lösung der Verbindungen (IV) bzw. (V) in einem Alkohol, bevorzugt t-Butanol, mit einem Oberschuß des Amins bei erhöhter Temperatur behandelt und gegebenenfalls die freien Basen mit Säuren in die entsprechenden Säureadditionssalze umwandelt. Geeignete Säuren sind beispielsweise Chlor- oder Bronwasserstoff-, Schwefel-, Oxal-, Fumar- oder Maleinsäure.
- Als Ausgangsverbindungen zur erstellung der Verbindungen (IV) und (V) sind insbesondere die neuen Verbindungen der Formel (III) geeignet, in der R7 ein Wasserstoffatom bedeutet.
- Sie lassen sich mit Epichlor- oder Epibromhydrin umsetzen.
- Das Verfahren zur erstellu von Verbindungen der Formel (III), in der A für ein Sauerstoffatom steht und R1, R2 und R7 die angegebene bedeutung haben, ist dadurch gekennzeichnet, daß man ein 9-Halogen-acetophenon der allgemeinen Formel (VII), mit der angegebenen 3edeutung von R1 R2 und worin X ein Chlor- oder Bromatom ist, mit dem Alkali-Salz einer 2-Dialkylphosphono-essigsäure zu a-Ketoacylestern der allgemeinen Formel (VIII) umsetzt, in der R1, R2 und R7 die oben angegebene Bedeutung haben und R8 für eine Methyl- oder Äthylgruppe steht, und diese anschließend durch intramolekulare Kondensation in die 2,5-Dihydrofuran-2-one der Formel (III) überführt.
- Bevorzugte Ausgangsverbindungen sind solche 2-Halogenacetophenone der allgemeinen Formel (VII), bei denen R7 eine Nethyl- oder Benzylgruppe bedeutet. Die Ausgangsverbindungen der allgemeinen Formel (VII) sind nach bekannten, in der Literatur beschriebenen Verfahren zugänglich.
- Vorzugsweise verwendet man das Kaliumsalz der 2-Diäthylphosphono-essigsäure, mit dem sich die IJmoetzung bei Raumtemperatur durchführen läßt. Die α-Ketoacylester (VIII) lassen sich entweder mit Kalium-tert. butylat in Dimethoxyäthan bei niedrigen Temperaturen oder vorteilhafter durch Phasentransferkatalyse beispielsweise mit 3enzyltributylammoniumbromid oder Tetrabutylammoniumbromid zum Butenolid cyclisieren.
- Verbindungen der Formel (III), in der A eine Imino- oder Alkyliminogruppe bedeutet, werden durch Umsetzung von Verbindungen der Formel (III), in denen A für ein Sauerstoffatom steht, mit Ammoniak oder Aminen der allgemeinen Formel R9-NH2, 2' wobei R9 eine Alkylgruppe mit 1 bis 4 Eohlenstoffatomen bedeutet, gewonnen. Dabei entstehen zunächst Verbindungen der allgemeinen Formel (IX) die durch Erhitzen in einem inerten Lösungsmittel wie beispielsweise Toluol oder Chlorbenzol, gegebenenfalls in Anwesenheit eines Eatalysators, beispielsweise p-Toluolsulfonsäure, durch Dehydratisierung in die 3-Pyrrolin-2-one der Formel (III) übergeführt werden.
- Die Methyläther gemäR der allgemeinen Formel (III) werden vorzugsweise durch Erhitzen mit Bromwasserstoff;Eisessig gespalten, während sich die 3enzyläther schon bei Raumtemperatur mit Bromwasserstoff/Eisessig oder auch durch Hydrierung mit 'Vasserstoff in Gegenwart von Edelmetallkatalysatoren wie Palladium/Aktivkohle in die Phenole umwandeln lassen Mit der säurekatalysierten Ätherspaltung erhält man aus Verbindungen mit Cyanomethyl-Substituenten der Phenylgruppe erfindungsgemäße Verbindungen der allgemeinen Formel (I), in denen R¹bzw. R² die Carbamoylmethylgruppe bedeuten.
- Die Phenole der Formel (III) (R7 = H) lassen sich mit Epichlor- oder Epibromhydrin zu den Verbindungen der allgemeinen Formeln (IV) und (V) umsetzen, die als Gemische anfallen. Eine Trennung der Gemische ist durch Säulenchromatografie möglich, aber meist nicht erforderlich. Die Reaktion erfolgt in einem Überschuß des Epihalogenhydrins in Gegenwart einer katalytischen Menge einer organischen Base bei Temperaturen von etwa 100°C.
- Die zur Umsetzung mit den Verbindungen der allgemeinen Formeln (IV) und (V) geeigneten Amine der allgemeinen Formel (VI) sind in der Literatur beschrieben oder nach bekannten Methoden zu erhalten.
- Die folgenden Beispiele dienen zur Erläuterung der Erfindung: Beispiel 1 4-[2-(2-Hydroxy-3-tert.butylamino-propoxy)-r-chlor-phenyl]-2,5-dihydro-furan-2-on 6,8 g 4-(2-Hydroxy-4-chlor-phenyl)-2,5-dihydro-furan-2-on werden mit 70 ml Epibromhydrin und 2 Tropfen Piperidin 2 Stunden auf 1000 C erwärmt. Nach Abdampfen des uberschüssigen Bromhydrins wird das Gemisch aus Epoxi- und Bromhydrin-Verbindung durch Hochdruckflüssigchromatografie getrennt. Die Epoxi-Verbindung kristallisiert nach Anreiben mit Diisopropyläther, Ausbeute: 3,0 g 4-[2-(2,3-Epoxi-propoxy)-4-chlor-phenyl]-2,5-dihydro-furan-2-on. Fp. 155-57 C Eine Lösung von 3,0 g 4-[2-(2,3-Epoxi-propoxy)-4-chlorphenyl]-2,5-dihydro-furan-2-on und 20 ml tert. -Butylamin in 400 ml tert. Butanol und 50 ml Methylenchlorid wird 5 Tage bei Raumtemperatur stehengelassen, dann eingeengt. Der Rückstand wird zwischen Methylenchlorid und wäßriger, verdünnter Salzsäure verteilt, die wäßrige Schicht mit Natriumhydrogencarbonat alkalisch gestellt und mit CH2C12 extrahiert. Die CH2C12-Phase wird abgetrennt, eingedampft, der Rückstand in Essigester aufgenommen unt athanol. HCl versetzt. Nach Umkristallisation aus Äthanol/tert. Butanol erhält man 2,4 g 4-[2-(2-Hydroxy-3-tert.
- butylamino-propoxy)-4-chlor-phenyl]-2,5-dihydro-furan-2-on als Hydrochlorid.
- Fp. 21445O C (z.) Beispiel 2 4-[2-(2-Hydroxy-3-isopropylamino-propoxy)-phenyl] 1,5-dihydro-2H-pyrrol-2-on 21,5 g 4- (2-Hydroxy-phenyl )-1 , 5-dihydro-2H-pyrrol-2-on werden mit 150 ml Epibromhydrin und einigen Tropfen Piperidin 3,5 Stunden auf 75° C erwärmt. Nach Säulenchromatografie an Kieselgel erhält man 4,5 g Epoxi-Verbindung und 43 g Bromhydrin-Verbindung als Öle.
- 2,5 g des 3romhydrins werden in 30 ml tert Butanol mit 30 ml Isopropylamin 15 Stunden bei Raumtemperatur gerührt.
- Danach wird im Vakuum eingedampft und der Rückstand nach Ansäuern mit 2n HCl mit Methylenchlorid extrahiert. Nach Alkalisieren der wäßrigen Phase wird erneut mit Methylenchlorid ausgeschüttelt und die CH2Cl2-Phase nach Ansäuern mit äthanol. HCl eingeengt. Nach Zugabe von wenig Äthanol kristallisieren 1,76 g des Hydrochlorids von 4-[2-(2-Hydroxy-3-isopropylamino-propoxy)-phenyl]-1,5-dihydro-2H-pyrrol-2-on aus.

HCl 23 4-CH3 H CH3 CH3 H NH 256-57 (Z.) HCl 24 H 5-nC3H7 CH3 CH3 H NH 194-98 HCl - 11 ethanol zugetropft. Nach 2 Stunden Rückfluß wird eingedampft, der Rückstand mit Wasser versetzt und mit Methylenchlorid extrahiert. Die organische Phase wird mehrfach mit 2n NaOH und dann mit Wasser ausgeschütte. Nach Abdampfen des Lösungsmittels erhält man 100 g des 3enzyläthers als Öl, aus dem nach Verreiben mit n-Hexan 75 g 2-Benzyloxy-4-chlor-acetophenon vom Fp. 640 C auskristallisieren.
- b) Zu 68 g (0,26 mol) 2-Benzyloxy-4-chlor-acetophenon in 700 ml Methanol werden bei 0 100 C langsam 41,7 g (0,26 mol) Brom zugetropft und anschließend wird noch 15 Stunden bei Raumtemperatur gerührt. Dabei kristallisiert das #-Bromacetophenon zusammen mit dem Ketal dieser Verbindung aus. Zur Spaltung des Ketals wird die Substanz in 300 ml Dioxan gelöst und eine Stunde bei 400 S mit 300 ml 6n HCl behandelt. Nach Filtration erhält man 77,8 g 2-Benzyloxy-4-chlor-phenacylbromid.
- Fp. 75 - 780 C c) Eine Lösung von 40,5 g (0,12 mol) 2-Benzyloxy-4-chlorphenacylbromid in 400 ml Acetonitril werden mit 33,7 g (0,145 mol) des Ealiumsalzes der 2-Diäthylphosphono-essigsäure 15 Stunden bei Raumtemperatur gerührt. Anschließend saugt man das Kaliumbromid ab und entfernt das Lösungsmittel. Dann nimmt man den Rückstand in Methylenchlorid auf, schüttelt mit Wasser aus und engt im Vakuum ein. Man erhält 52,5 g rohen a-Ketolphosphono-essigester, der nach Anreiben mit Diisopropyläther kristallisiert.
- Ausbeute: 45,4 g Fp. 84 - 850 C d) 45,4 g (0,1 mol) des obigen Phosphonesters werden zusammen mit 3,25 g (0,01 mol) Tetrabutylammonium-bromid in 350 ml Methylenchlorid gelöst und in einer Zweiphasenreaktion mit 7,9 g (0,12 mol) KOH (85 %ig) in 250 ml Wasser eine Stunde bei 20° C kräftig gerührt. Die organische Phase wird mit Wasser neutral gewaschen und eingedampft. Durch Verreiben mit Diisopropyläther erhält man 28 g 4-(2-Benzyloxy-4-chlor-phenyl)-2,5-dihydro-furan-2-on.
- Fp. 117 - 220 C e) 17 g des obigen Benzyläthers werden in 80 ml 40 zeiger Bromwasserstoffsäure in Eisessig eine Stunde bei Raumtemperatur gerührt. Nach Eindampfen wird der Rückstand mit Äther/Diisopropyläther verrieben und abgesaugt.
- Ausbeute: 7,5 g 4-(2-Hydroxy-4-chlor-phenyl)-2,5-dihydro-furan-2-on Fp. 184-185° C 3eispiel 26 4-(2-.Rydroxy-5-acetamido-phenyl)-2,5-dihydro-furan-2-on 8,5 g 4-(2-Benzyloxy-5-acetamido-phenyl )-2, 5-dihydro-furan-2-on werden in 300 ml Xthanol/Essigester (1:1) mit Wasserstoff unter leichtem Überdruck bei Raumtemperatur hydriert (Katalysator 10 Oio! Pd-C).
- Nach üblicher Aufarbeitung erhält man 5,3 g 4-(2-Hydroxy-5-acetamido-phenyl)-2,5-dihydro-furan-2-on.
- Fp. 248 - 490 C (Z.) Beispiel 27 4-(2-Hydroxy-5-carbamoylmethyl-phenyl)-2,5-dihydro-furan-2-on 4,4 g 4-(2-Benzyloxy-5-cyanomethyl-1-phenyl)-2,5-dihydrofuran-2-on werden in 3O ml HBr-Eisessig (40 %) bis zur vollständigen Lösung der Substanz gerührt. Nach Abdampfen des Eisessigs wird der Rückstand mit Essigester/Äther verrieben und abgesaugt. Ausbeute: 3,2 g. Zur Entfernung von Bromwasserstoffsäure wird die Substanz in gesättigter Natriumhydrogencarbonat-Lösung suspendiert und nach Filtration mit Äthanol gewaschen.
- Ausbeute: 1,6 g 4-(2-Hydroxy-5-carbamoylmethyl-phenyl)-2,5-dihydro-furan-2-on Fp. 2250 C (Z.) Beispiel 28 4-(2-Hydroxy-phenyl)-1,5-dighydro-2H-pyrrol-2-on 22,0 g 4-(2-Benzyloxy-phenyl)-2,55-dihydro-furan-2-on werden in einem Autoklaven mit 300 ml flüssigem NR3 3 Stunden auf 1000 C erhitzt. Nach Abdampfen des NH3 nimmt man den öligen Rückstand in Isopropanol auf und saugt 15 g eines kristallinen.
- Niederschlags ab. Fp. 163 - 66°C. Aufgrund der NMR-spektroskopischen Daten handelt es sich um 3-Hydroxy-4-(2-benzylotyphenyl)-pyrrolidin-2-on (Formel IX).
- 14 g dieser Verbindung werden durch halbstündiges Erhitzen in 50 ml Toluol unter Zusatz katalytischer Mengen p-Doluolsulfolsäure dehydratisiert. Nach Erkalten der Lösung kristallisieren 8,5 g 4-(2-Benzyloxy-phenyl)-1,5-dihydro-2H-pyrrol-2-on aus.
- Fp. 453 - 540 C 8,0 g des Benzyläthers werden in 40 ml HBr (40 %ig) in Eisessig suspendiert und bei Raumtemperatur gerührt. Nach ca.
- 15 Minuten ist eine klare Lösung entstanden, aus der sich das Phenol langsam abscheidet. Das Lösungsmittel wird- abgedampft, der Rückstand mit Toluol versetzt und abgesaugt. Das Produkt wird anschließend 5 Minuten in 50 ml Eiswasser gerührt, abgesaugt und getrocknet.
- Ausbeute: 4,7 g 4-(2-Hydroxy-phenyl)-1,5-dighydro-2H-pyrrol-2-on Fp. 190 - 940 C In gleicher Weise werden die in folgender Tabelle aufgeführten Verbindungen erhalten: Tab. 2 3eispiel Nr. Claims (9)
- Patent ansprüche (1.) Substituierte 4-Aryl-2,5-dihydro-furan-2-one und 4-Aryl-1,5 dihydro-2H-pyrrol-2-one der allgemeinen Formel (1) in der R¹ und 22, die gleich oder verschieden sein können, ein Wasserstoff- oder Halogenatom, eine Alkyl- oder Acylaminogruppe mit \ bis 6 Kohlenstoffatomen oder eine Carbamoylmethylgruppe, und R , die gleich oder verschieden sein können, ein Wasserstoffatom oder eine Methylgruppe, R5 ein Wasserstoffatom, eine Dimethoxyphenylgruppe oder eine Acylaminogruppe der allgemeinen Formel (II) R6-CONH- (II) worin R6 für eine Alkylgruppe mit 1 bis 4 Kohlenstoffatomen steht, und A ein Sauerstoffatom, eine Imino- oder Alkyliminogruppe mit 1 bis 4 Sohlenstoffatomen bedeutet, sowie deren physiologisch verträgliche Säureadditionssalze.
- 2. Substituierte 4-Aryl-2,5-dihydro-furan-2-one und 4-Aryl-1,5-dihydro-2H-pyrrol-2-one der allgemeinen Formel (III) in der R7 ein Wasserstoffatom, eine Methyl- oder Benzylgruppe und den Rest oder bedeutet, worin X für ein Chlor- oder Bromatom steht.
- 3. Verfahren zur Herstellung der Verbindungen nach Anspruch 1, dadurch gekennzeichnet, daß man Verbindungen der Formeln (IVr oder (V) worin R1, R2, A und X die oben angegebene Bedeutung haben, oder ein Gemisch dieser Verbindungen mit einem Amin der allgemeinen ormel (VI) in der R³, R4 und R5 obengenannte Bedeutung haben, umsetzt, und gegebenenfalls die freien 3asen mit Säuren in die entsprechenden Säureadditionssalze umwandelt.
- 4. Verfahren zur Herstellung der Verbindungen nach Anspruch 2, bei denen R1 R2 und R7 in der allgemeinen Formel (III) die angegebene 3edeutung haben und A für ein Sauerstoffatom steht, dadurch gekennzeichnet, daß man ein 2-Halogenacetophenon der allgemeinen Formel (VII) mit der angegebenen Bedeutung von R1, R2 und R7 und worin X ein Chlor- oder Bromatom bedeutet, mit dem Salz einer 2-Dialkylphosphono-essigsäure zu -PLetoacylestern der allgemeinen formel (VIII) umsetzt, in der R', R2 und R7 die angegebene 3edeutung haben und R8 für eine Nethyl- oder Äthylgruppe steht, und diese anschlie-Pend durch intramolekulare Kondensation in die obengenannten 2,5-Dihydro-furan-2-one der allgemeinen Formel (III) überführt.
- 5. Verfahren zur tierstellung der Verbindungen nach Anspruch 2, bei denen R7 in der Formel (III) die angegebene Bedeutung hat und A eine Imino- oder Alkyliminogruppe bedeutet, dadurch gekennzeichnet, daß man Verbindungen der allgemeinen Formel (III), in denen A ein Sauerstoffatom bedeutet, mit Aminen der allgemeinen Formel R9-NH2 umsetzt, wobei R9 ein Wasserstoffatom oder eine Alkylgruppe mit 1 bis 4 Kohlenstoffatomen bedeutet.
- 6. Verfahren zur Herstellung der Verbindungen nach Anspruch 2, bei denen R7 in der allgemeinen Formel (III) ein Wasserstoffatom bedeutet, dadurch gekennzeichnet, daß man Verbindungen der allgemeinen Formel (III), in denen R7 eine Methyl- oder Benzylgruppe bedeutet, einer säurekatalysierten Ätherspaltung unterwirft oder falls R7 eine Benzylgruppe bedeutet, hydriert.
- 7. Verfahren zur fIerstellung der Verbindungen nach Anspruch 2, bei denen R7 in der allgemeinen Formel (III) den Rest bedeutet und worin X ein Chlor- oder Bromatom ist, dadurch gekennzeichnet, daß man eine Verbindung der allgemeinen Formel (III), in der R7 ein Wasserstoffatom bedeutet, mit Epichlor- oder Epibromhydrin umsetzt.
- 3. Verwendung der Verbindungen nach Anspruch 2 als Zwischenprodukte für die Herstellung der Verbindungen der allgemeinen Formel (I) nach Anspruch 1.
- 9. Pharmazeutisches Eräparat, dadurch gekennzeichnet, daß es eine oder mehrere der Verbindungen gemäß Anspruch 1 und gegebenenfalls bleiche Trägerstoffe und/oder Verdünnungsmittel enthält.
