Description
2. Arzneimittel nach Anspruch 1, dadurch gekennzeichnet, daß darin Glaucin bestehend aus 1-Glaucin
und einer höchstens äquimolaren Menge d-Glaucin und/oder deren von physiologisch unbedenklichen
Säuren abgeleiteten Salze oder quarternären Ammoniummethohalogenide enthalten sind.

Die Erfindung betrifft Arzneimittel mit antithrombotischer Wirkung. Es ist bekannt, daß Papaverin eine gewisse
antithrombotische Wirkung besitzt, jedoch bei der Injektion und bei den wirksamen Konzentrationen so
starke Gefäßnebenwirkungen zeigt, daß es für diese Indikation praktisch nicht verwendbar ist Bekannt ist ferner
das aus Glaucium flavum isolierbare Papaveraceen- und Fumariaceen-Alkaloid Glaucin, das rechtsdrehendes
1,2,9,10-Tetramethoxyaporphin der Formel:

ist und nach N. Donev (Farmatsiya, Sofia, 1962, Seiten
17 bis 21) antitussive, hypo tensive und adrenolytisehe
Wirkung besitzt. In Th. A. Henry »The Plant Alkaloids«, London, (1913), Seiten 258 bis 259 sind auch das
synthetisch hergestellte d,l-Glaucin und das aus diesem durch Spaltung gewonnene synthetische 1-Glaucin als
Stoffe beschrieben, über deren pharmakologische Eigenschaften jedoch bisher nichts bekannt war.
Die DE-OS 16 70 894 beschreibt schließlich ein Verfahren zur Herstellung von Aporphinen beispielsweise
2,3,5,6-Tetrahydroxyaporphin-hydrochlorid, die als Arzneimittel verwertbar sein sollen.
Aufgabe der Erfindung war es nun, Arzneimittel der eingangs genannten Art zu schaffen, die schon bei geringer
Wirkstoffkonzentration eine starke, von unerwünschten Nebenwirkungen weitgehend freie antithrombotische
Wirksamkeit besitzen.
Zur Lösung dieser Aufgabe sind die Arzneimittel der eingangs genannten Art erfindungsgemäß gekennzeichnet
durch einen Gehalt an
a) Glaucin mit einem Gehalt an I-Glaucin oder
b) deren von physiologisch unbedenklichen anorganischen oder organischen Säuren abgeleiteten Salzen
oder quarternären N-Alkylammoniumverbindun-
Die erfindungsgemäßen Arzneimittel hemmen die Thrombozytenaggregation schon bei Wirkstoffkonzentrationen,
die weit unter den für Papaverin erforderlichen Liegen und sind dabei von den beim Papaverin
auftretenden Nebenwirkungen weitgehend frei.
Nach einer bevorzugten Ausführungsform enthält das Arzneimittel Glaucin aus 1-Glaucin und einer höchstens
äquimolaren Menge d-Glaucin und/oder deren von physiologisch unbedenklichen Säuren abgeleitete
Salze oder quarternären Ammoniummethohalogenide.
Im folgenden werden die Arzneimittel und die Herstellung der darin verwendeten Wirkstoffe anhand von
Beispielen erläutert.:
Zur Herstellung von Laudanosin wurde zu 225 g (0,6 Mol) Papaverin-hydrochlorid in 2150%igein wäßrigem
Methylalkohol eine Lösung von 30 g Natriumhydroxid in 300 ml Wasser langsam zugegeben, der entstandene
Niederschlag abfiltriert und bei 6O0C für 20 Stunden getrocknet. Es wurden 201 g Papaverin-Base
vom Schmelzpunkt 144 bis 146° C erhalten (Ausbeute 98%).
200 g (0,6 Mol) der so erhaltenen Papaverin-Base wurden in 200 ml Methylalkohol gelöst, mit 120 ml Methyljodid
versetzt und die Mischung sechs Stunden zum Sieden erhitzt. Nach dem Abkühlen wurde das kristalline
Produkt abfiltriert und getrocknet. Dabei wurden 280 g Papaverin-methiodid vom Schmelzpunkt 127 bis
129° C erhalten (Ausbeute 97°).
Es wurden 200 g (0,4 Mol) Papaverin-Methiodid in 2 1 lO°/oigem wäßrigem Methylalkohol suspendiert und
langsam bis zur völligen Auflösung und dem Verschwinden der gelborangenen Färbung mit Natriumborhydrid
versetzt. Die Lösung wurde in 121 Wasser eingegossen und der entstehende weiße Niederschlag abfiltriert und
getrocknet. Es wurden 230 g Laudanosin vom Schmelzpunkt 113 bis 1150C erhalten (Ausbeute 78%).
Zur Herstellung von Laudanosolin-hydrobromid wurden 121 g (0,34 Mol) Laudanosin in 600 ml 40%iger
Bromwasserstoffsäure etwa 10 Stunden bis zur vollständigen
Abgabe von Methylbromid erhitzt. Das nach Kühlen der Lösung auskristallisierende Produkt wurde
abgetrennt. Es wurden 100 g Laudanosolin-hydrobromid vom Schmelzpunkt 23O0C erhalten (Ausbeute
77%).
Zur Herstellung von Tetrahydroxyaporphin-hydrochlorid wurden 98 g (0,26 Mol) Laudanosolin-hydrobromid
in 1,2150%igem wäßrigen Methylalkohol bei 8O0C
gelöst. Nach vollständiger Lösung wurde durch Zugabe von Eis auf 60C abgekühlt und dann eine auf 60C gekühlte,
filtrierte Lösung von 100 g (0,62 Mol) Eisen(III)-chIorid in 500 ml 50%igem wäßrigem Methylalkohol
zugegeben. Nach einer Minute wurden 1,5 1 konzentrierter Salzsäure hinzugefügt und die Lösung bei
Zimmertemperatur stehengelassen. Die dabei ausfallenden graubraunen Kristalle wurden abfiltriert, mit Aceton
gewaschen und getrocknet. Es wurden 43 g Tetrahydroxyaporphin-hydrochlorid vom Schmelzpunkt 242
bis 244° C erhalten (Ausbeute 50%).
Zur Herstellung von d.l-Glaucin-hydrobromid wurden
6 g (0,018 Mol) Tetrahydroxyaporphin-hydrochlorid unter Erwärmen in 840 ml Methylalkohol gelöst. Getrennt
davon wurden 21,6 g (0,126 Mol) Trimethylphenylamoniumchlorid in Methylakohol mit einer Lösung
von 8,5 g (0,15MoI) KOH in Methylalkohol versetzt.
Nach Abfiltrieren vom ausgeschiedenen Kaliumchlorid wurde das Filtrat mit Methylalkohol auf 840 ml aufgefüllt
Nachfolgend wurden beide Lösungen langsam unter Schutzgas gemischt und dann innerhalb von 6 Stunden
langsam zu auf 110°C erhittzem Anisol zugefügt, wobei der Methylalkohol abdestillierte. Nach vollständiger
Zugabe wurde die Lösung gekühlt und von einem schwarzen, amorphen Rückstand abfiltriert Das dunkelgrüne
Filtrat wurde im Vakuum bis zur Trockne eingedampft Der Rückstand wurde mit 5 ml Allylacetat
versetzt Das aus der erhaltenen Mischung auskristallisierte Produkt wurde abfiltriert und getrocknet Es wurden
5,5 g djl-Glaucin-hydrobromid vom Schmelzpunkt
235° C (unter Zersetzung) erhalten (Ausbeute 67%).
Es wurden 60 g dJ-Glaucin-hydrobromid, das durch
die vorstehend beschriebenen Verfahrensstufen erhalten wurde und gemäß Dünnschichtchromatogramm etwa
10% l-(N,N-Dimethylaminoäthyl)-3,4,6,7-tetramethoxyphenanthren enthielt in 200 ml 50%igem wäßrigem
Äthanol gelöst und mit einem Überschuß von 8,5 g Kaliumhydroxid in 50 ml Wasser versetzt
Aus der erhaltenen Mischung wurde durch Ausschütteln
mit etwa 500 ml Chloroform die freie d,l-Glaucinbase extrahiert. Die abgetrennte organische Phase wurde
über wasserfreiem Natriumsulphat getrocknet und filtriert. Nach Verdampfen des Lösungsmittels wurden
50 g eines öligen Rückstandes erhalten, der aus 75 ml Äthylacetat umkristallisiert wurde. Nach dem Abfiltrieren
und Trocknen wurden 30,4 g d,l-GIaucinbase vom Schmelzpunkt 128 bis 130° C erhalten. Nach wiederholter
Kristallisation aus weiteren Msngen 100 ml Äthylacetat wurden 25,15 g d,l-Glaucinbase vom Schmelzpunkt
138 bis 14O0C erhalten, die gemäß Dünnschichtchromatogramm
weniger als 0,1% Verunreinigungen enthielt.
Nach Kristallisation der vereinigten Rückstände aus Äthylalkohol wurden etwa 5 g 1-(N,N-Dime\:hylaminoäthyl)-3,4,6,7-tetramethoxyphenantren
mit einem Schmelzpunkt von 248 bis 250° C und einem massenspektrometrisch ermittelten Molekulargewicht von 269
erhalten.
NMR-Spektrum:
9,13 (s,l), 7,88 (d,l), 7,66 (d,l), 7,46 (s,2), 3,97 (s,3), 3,93 (s,6), 3,86 (s,3), 3,44-3,14 (m,4), 2,73 (s,3), 2,52 (s,3).

Zur Trennung der Isomeren wurden 5,09 g (0,014 Mol) d,l-Glaucin in 70 ml Äthylalkohol gelöst und
mit einer Lösung von 2,2 g (0,014 MoI) d-Weinsäure in 70 ml Äthylalkohol (500C) versetzt. Das sich beim langsamen
Abkühlen ausscheidende, feinkristalline Produkt wurde abfiltriert, mit Äther gewaschen und getrocknet.
Es wurden 3,6 g 1-Glaucin-d-bitatrat mit einem Schmelzpunkt von 210 bis 212°C und einer spezifischen Drehung
in Wasser von -26° erhalten (Ausbeute 93%).
Das noch unreine 1-Glaucin-d-bitatrat wurde mit einer wäßrigen Natriumyhdroxidlösung versetzt und mit
Äther extrahiert. Nach dem Verdampfen des Lösungsmittels wurde der Rückstand in 50 ml Äthylalkohol gelöst
und mit einer Lösung von 1,15 g d-Weinsäure in 50 ml Äthylalkohol versetzt. Nach dem Abtrennen wurden
3,39 g 1-Glaucin-d-bitatrat mit einem Schmelzpunkt von 212 bis 215°C und einer spezifischen Drehung in
Wasser von —32° erhalten. Die spezifische Drehung
der 1-Glaucinbase in Äthylalkohol betrug —101° (94%
optische Reinheit).
2,48 g 1-Glaucin wurden in 15 ml Äthylalkohol mit einem
kleinen Oberschuß an 48%iger Bromwasserstoffsäure versetzt Nach der Abtrennung wurden 2,78 g 1-Glaucin-hydrobromid
vom Schmelzpunkt 235° C (unter Zersetzung) erhalten (Ausbeute 98,5%). Aus den Mutterlaugen der ersten und zweiten d-Bitatrat-Kristallisation
wurde nach dem Eindampfen ein grünlicher Rückstand erhalten, der in 20 ml Wasser gelöst
mit wäßrigem Natriumhydroxid behandelt und dann mit 250 ml Äther extrahiert wurde. Nach dem
Trocknen wurde Abfiltriert und das Lösungsmittel abgedampft Dabei wurden 1,67 g d-GIaucin vom
Schmelzpunkt 120° C erhalten. Die spezifische Drehung betrug in Äthylalkohol 104,6°. Das hieraus durch Umsetzung
mit Bromwasserstoffsäure erhaltene d-Glaucinhydrobromid zeigte einen Schmelzpunkt von 235° C
(unter Zersetzung). Das durch Methylierung aus d-Boline erhaltene d-Glaucin zeigt eine spezifische Drehung
in Äthylalkohol von +115°.
Zur Herstellung von 1-Glaucin-hydrochlorid wurde 1-Glaucin
in wenig Methylalkohol gelöst und ein geringer Überschuß an konzentrierter Salzsäure zugegeben. Aus
dem erhaltenen, voluminösen Niederschlag wurden durch Zugabe von Äthylacetat weißlich bis rosa gefärbte
Kristalle erhalten. Nach dem Abfiltrieren, Waschen mit Aceton und Trocknen wurde weißlich bis rosa gefärbtes,
feinkristallines 1-GIaucin-hydrochlorid vom
Schmelzpunkt 232 bis 233° C erhalten.
Zur Herstellung von 1-Glaucin-hydrojodid wurde 1-Glaucin
in 2 η Salzsäure gelöst und mit gesättiger KaIiumjodidlösung versetzt. Der erhaltene kristalline Niederschlag
wurde aus einer Mischung von Methylalkohol und Äther umkristallisiert Das dabei auskristallisierende,
gelbliche Glaucin-hydrojodid zeigte einen Schmelzpunkt von 238° C.
Es wurden 3,92 g (0,01 Mol) Glaucin, 1,5 g (0,011 Mol)
so Kaliumcarbonat und 5 ml Methyljodid 6 Stunden in Methanol erhitzt. Die noch heiße Lösung wurde filtriert
und das Lösungsmittel im Vakuum entfernt, wobei 4,2 g d-Glaucinmethiodid vom Schmelzpunkt 218—220°C erhalten
wurden (Ausbeute 85%).

Stattdessen können auch mit einer Bruchrille versehene Tabletten mit einem Wirkstoffgehalt von 400 mg
hergestellt werden, die eine Teilung der Dosierung gestatten.

Zur Herstellung von Kapseln mit die Thrombozytenaggregation hemmender Wirkung wurden Hartgelatinekapseln

1. Arzneimittel mit antithrombotischer Wirkung, gekennzeichnet durch einen Gehalt an
a) Glaucin mit einem Gehalt an 1-Glaucin oder
b) deren von physiologisch unbedenklichen anorganischen oder organischen Säuren abgeleiteten
Salzen oder quarternären N-Alkylammoniumverbindungen.
