Description
Lipase-Präparate haben technische Bedeutung und können beispielsweise zur Fettspaltung verwandt werden.
Ebenfalls können Lipase-Präparate zum Beizen bei der Gerbereivorbereitung Verwendung finden.
Die Handelsprodukte sind zum Zwecke der Standardisierung mit indifferenten Substanzen wie beispielsweise
Zucker, Stärke, Kieselgur usw. vermischt Diese Zusätze müssen als integrierende Bestandteile der Präparate
betrachtet werden. Es hat sich nun gezeigt daß Lipase häufig durch Zugabe solcher Zusatzstoffe insbesondere
dann, wenn es sich um oberflächenaktive Stoffe handelt, eine starke Aktivitätsverminderung erfährt
Es wurde nun gefunden, daß diese Aktivitätsverminderung
nicht beziehungsweise nur in ganz geringfügigem Maße eintritt wenn solche lipasehaltigen Präparate
zusätzliche 10—50 Gewichtsteile, vorzugsweise 20—40 Gewichtsteile (bezogen auf 1 Gewichtsteil Lipase)
einer Mischung enthalten, die sich aus folgenden Bestandteilen zusammensetzt: 70—90%, vorzugsweise
82-86% Lactose; 8-20%, vorzugsweise U —13%
Molkenprotein; 0,1—3%, vorzugsweise 0,2—0,5%
Vnkdifett; 0,1 —5%, vorzugsweise 1—2% Molkenmineralstoffe
und 0,1—3%, vorzugsweise 0,5—2% Wasser (% = Gewichtsprozente).
Unter Molkenprotein oder Serumprotein versteht man die Proteine, die sich in der unerhitzten Milch nach
Ausfällung des Caseins (durch Elektrolyts, besonders Wasserstoffionen oder Lab) befinden. Dieses Milchserumprotein
hat beispielsweise folgende Zusammensetzung: 55-65%^,-Lactoglobulin, 13—22% «-Lactalbumin,
16—25% Immunglobuline und 3—4% Serumalbumin.
Milchfett besteht aus den Triglyceriden gesättigter und ungesättigter Fettsäuren. Die Fettsäurezusammensetzung
(in Gewichtsprozent) ist beispielsweise folgende:
Buttersäure 2—8, vorzugsweise 3,6—5,5; Capronsäure
1 —3, vorzugsweise 1,5—2; Caprylsäure 0,1—2, vorzugsweise
04— 1: Caprinsäure 0,1 —5, vorzugsweise 03—23;
Laurinsäure 1 —5, vorzugsweise 2,0—2,6; Myristinsäure 5—15, vorzugsweise 9,5—12; Palmitinsäure 25—40, vorzugsweise
28—38; Stearinsäure 1 — 15, vorzugsweise 13—10; gesättigte Säuren über Cie 1 —4, vorzugsweise
2-3; Decensäure 0,05-03; Dodecensäure 0,05—0,5; Tetradecensäure 0,1 —3; Hexadecensäure 1 —7; Octadecensäure
20—40, vorzugsweise 25—35; Octaclecadiensäure 1 — 5, ungesättigte Säuren über Ci81 —3.
Unter Molkenmineralstoffe wird eine Mischung von
Phosphaten, Citraien, Chloriden, Sulfaten und Carbonaten
des Natriums, Kaliums, Calciums, Magnesiums und spurenweise des Eisens verstanden. Der Gehall der Metalle
in der Salzmischung in Gewichtsprozenten ist beispielsweise folgender: Natrium 2—8, vorzugsweise
4—5 Gewichtsprozent; Kalium 10—30, vorzugsweise 15—20 Gewichtsprozent; Calcium 2—30, vorzugsweise
4—15 Gewichtsprozent; Magnesium 0,5—3, vorzugsweise
0,8—13 Gewichtsprozent; Eisen 0,01— 0,1, vorzugsweise
0,02—0,08 Gewichtsprozent Innerhalb der Mischung kommen, bezogen auf ein Gewichtsteil Magnesium,
beispielsweise 3—8, vorzugsweise 5—7 Gewichtsteile Natrium, 2—15, vorzugsweise 4—8 Gewichtsteile
Calcium und 15—30, vorzugsweise 18—25 Gewichtsteile Kalium.
Der Gehalt an Säuren in Gewichtsprozenten in der Salzmischung ist beispielsweise folgender: Phosphatanion
(PO«3~) 10—50, vorzugsweise 15—25 Gewichtsprozent;
Citratanion 10—30, vorzugsweise 15—25 Gewichtsprozent;
Chioridanion 10—20, vorzugsweise 12—16 Gewichtsprozent; Sulfatanion 2—8, vorzugsweise
3—6 Gewichtsprozent; Carbonatanion 2—15,
vorzugsweise 5—10 Gewichtsprozent; bezogen auf 1 g Sulfatanion kommen innerhalb der Mischung beispielsweise
2—5, vorzugsweise 23—3 Gewichtsteiie Chioridanion,
3—10, vorzugsweise 4—6 Gc α ichtsteile Phosphatanion. 3—10, vorzugsweise 4—6 Gewichtsteile
Citratanion und 03—2, vorzugsweise 03—13 Carbonatanion.
Vorzugweise werden als erfindungsgemäße Stabilisierungsmittel
10—50 Gewichtsteiie, insbesondere 20—40 Gewichtsteüe (bezogen auf 1 Gewichtsteil Lipase)
SQBmolkenpulver verwendet Süßmolkenpulver ist ein gängiges Handelsprodukt und wird beispielsweise
durch Sprühtrocknung von Molke (Süßmolke) hergestellt 'siehe Ullmanns Encyclopädie der technischen

Das verwendete Süßmolkenpulver stellt ein Gemisch aus 70—90%, vorzugsweise 82—86% Lactose, 8—20%,
vorzugsweise 11 —13% Molkenprotein, 0,1—3%, vorzugsweise
0,2—03% Müchfett 0,1—5%, vorzugsweise 1 —2% Molkenmineralstoffe und 0,1 —3%, vorzugsv/eise
03—2% Wasser dar (0Zo bedeutet stets Gewichtsprozente).
Die zu stabilisierende Lipase kann tierischen oder nicht-tierischen Ursprungs sein. Beispielsweise handelt
es sich um Lipase aus Pankreas oder um Lipase aus Pflanzen, beispielsweise aus Ricinusbohnen oder Lipase
aus Mikroorganismen, beispielsweise aus Pilzen wie Rhizopus arrhizus, Rhizopus nigricans, Rhizopus oryzae,
Rhizopus delemar, Aspergillusarten wie Aspergillus niger, Aspergillus oryzae oder Welchia perfringens, Mycotorula
lipolytica, Candida cyclindrav'ea, Geotrichum candidum. Vorzugsweise handelt es sich um Lipase
nicht-tierischen Ursprungs.
Die Gewinnung der in Betracht kommenden Lipase erfolgt in der hierfür bekannten Weise und ist beispielsweise
angegeben in Ullmanns Encyclopädie der technischen Chemie, Band 7 (1956), Seite 391-397, 406-411
oder in Bulletin de la Societe de Chimie Biologique 1966,
48 Nr. 6, Seite 747-770 und 1968, 50 Nr. 11, Seite 2179—2182. Beispielsweise wird eine Pilzlipase entsprechend
Bulletin de la Societe de Chimie Biologique 1966, 48 Nr. 6, Seite 747 ff. wie folgt erhalten: Das nach Abtrennung
des Myzels aus Kulturen von Pilzsporen erhaltene Filtrat wird im Vakuum bei 300C konzentriert, zentrifugiert
und aus der oberen Schicht nach Verdünnung das Enzym mit Aceton bei 0°C gefällt und im Vakuum
