Description
Alkoxymethyl- bzw. alkylthiomethylsubstitutierte
Pyrazolyl(thiono)(thiol)-phosphor(phosphön)-säureester
bzw. -esteramide, Verfahren zu ihrer Herstellung und ihre Verwendung als Insektizide und Akarizide
Die vorliegende Erfindung betrifft neue alkoxymethyl- bzw. alkylthiomethylsubstituierte Pyrazolyl(thiono)(thiol)-phosphor-(phosphön)-säureester
bzw. -esteramide, welche insektizide und akarizide Eigenschaften haben und ein Verfahren zu ihrer Herstellung.
Es ist bereits bekannt, daß 0,0-Dialkyl-O-pyrazolyl-thionophosphorsäureester,
wie 0,0-Diäthyl-O- [3-methyl-pyrazol(5)yiD-thionophosphorsäureester,
insektizide und akarizide Eigenschaften haben (vergleiche US-Patentschrift 2 754 244).
Es wurde nun gefunden, daß die neuen alkoxymethyl- bzw. alkylthiomethylsubstituierten
Pyrazolyl(thiono)(thiol)-phosphor-(phosphon)-säureester
bzw. -esteramide der Formel

Weiterhin wurde gefunden, daß die neuen aikoxymethyl- und alkylthiomethylsubstituierten
Pyrazolyl(thiono)(thiol)-phosphor-(phosphon)-säureester
bzw -esteramide der Formel (I) erhalten werden- wenn man (Thiono)(Thiol)Phosphor(phosphon)-säureester-
bzw. -esteramidhalogenide der Formel

Überraschenderweise zeigen die erfindungsgemäßen alkoxymethyl- bzw. alkylthiomethylsubstituierten Pyrazolyl(thiono)(thiol)-phosphor(phosphon)-säureester
bzw. -esteramide eine bessere insektizide und akarizide Wirkung als die vorbekannten 0,0-Dialkyl-O-pyrazolylthionophosphorsäureester
analoger Konstitution und gleicher Wirkungsrichtung. Die Stoffe gemäß vorliegender
Erfindung stellen somit eine echte Bereicherung der Technik dar.
Verwendet man beispielsweise S-sec.-Butyl-thionothioläthanphosphonsäureesterchlorid
und l-Äthyl^-methylthiomethyl-S-hydroxi-pyrazol
als Ausgangsmaterialien, so kann der Reaktionsverlauf durch das folgende Formelschema wiedergegeben werden:

(III) können nach literaturbekannten Verfahren hergestellt werden, indem man Hydrazinderivate mit alkoxy- bzw. alkylthiosubstituierten
Acetessigsäurealkylestern umsetzt. Als Beispiele dafür seien im einzelnen genannt:

Das Verfahren zur Herstellung der erfindungsgemäßen Verbindungen wird bevorzugt unter Mitverwendung geeigneter Lösungsader Verdünnungsmittel durchgeführt. Als solche kommen praktisch
alle inerten organischen Solventifin infrage. Hierzu gehören
insbesondere aliphatisehe und aromatische, gegebenenfalls
chlorierte, Kohlenwasserstoffe, wie Benzol, Toluol, Xylol, Benzin, Methylenchlorid, Chloroform, Tetrachlorkohlenstoff,
Chlorbenzol, oder Äther, z.B. Diäthyl- und Dibutyläther, Dioxan, ferner Ketone, beispielsweise Aceton, Methyläthyl-, Methylisopropyl-
und Methylisobutylketon, außerdem Nitrile, wie Aceto- und Propionitril.
Als Säureakzeptoren können alle üblichen Säurebindemittel Verwendung
finden. Besonders bewährt haben sich Alkalicarbonate und -alkoholate, wie Natrium- und Kaliumcarbonat, -methylat bzw.
-äthylat, ferner aliphatische, aromatische oder heterocyclische Amine, beispielsweise Triäthylamin, Trimethylamin, Dimethylanilin,
Dimethylbenzylamin und Pyridin.
Die Reaktionstemperatur kann innerhalb eines größeren Bereichs variiert werden. Im allgemeinen arbeitet man zwischen 0 und
loo°C, vorzugsweise bei 2o bis 6o°C.
Die Umsetzung läßt man im allgemeinen bei Normaldruck ablaufen.

Zur Durchführung des Verfahrens setzt man die Ausgangsstoffe meist in äquivalentem Verhältnis ein. Ein Überschuß der einen
oder anderen Reaktionskomponente bringt keine wesentlichen Vorteile. Vorzugsweise legt man das 5-Hydroxy-pyrazol(III)
und den Säureakzeptor in einem Lösungsmittel vor und rührt diese Suspension bei erhöhter Temperatur. Nach dem Abkühlen
fügt man das Phosphorsäureesterderivat(II) zu der Mischung und arbeitet diese nach beendeter Reaktion in üblicher Weise
auf, indem man die Reaktionslösung mit Wasser und einem organischen Lösungsmittel versetzt, die organische Phase abtrennt,
trocknet und das Lösungsmittel abdampft.
Die neuen Verbindungen fallen in Form von Ölen an, die sich zum Teil nicht unzersetzt destillieren lassen, jedoch durch
sogenanntes "Andestillieren", d.h. durch längeres Erhitzen
unter vermindertem Druck auf mäßig erhöhte Temperaturen von den letzten flüchtigen Anteilen befreit und auf diese Weise
gereinigt werden. Zu ihrer Charakterisierung dient der Brechungsindex .
Wie bereits mehrfach erwähnt, zeichnen sich die erfindungsgemäßen alkoxymethyl- bzw. alkylthiomethylsubstituierten PyrazolylCthionoKthiolJ-phosphorCphosphonJ-säureester
bzw. -esteramide durch eine hervorragende insektizide und akarizide Wirksamkeit aus. Sie wirken gegen Pflanzen-, Hygiene- und Vorratsschädlinge, sowie auf dem veterinär-medizinischen Sektor. Sie
besitzen bei geringer Phytotoxizität sowohl eine gute Wirkung gegen saugende als auch fressende Insekten und Milben.
Aus diesem Grunde können die erfindungsgeaäßen Verbindungen
mit Erfolg im Pflanzenschutz sowie auf dem Hygiene-, Vorratsschutz- und Veterinärsektor als Schädlingsbekämpfungsmittel
eingesetzt werden.

Die Wirkstoffe eignen sich bei guter Pflanzenvertraglicnkeit und günstiger Warmblütertoxizität 7nv Bekämpfung von tierischen
Schädlingen, insbesondere Insekten und Spinnentieren, die in der Landwirtschaft, in Forsten, im Vorrats- und Materialschutz
sowie auf dem Hygienesektor vorkommen. Sie sind gegen normal sensible und resistente Arten sowie gegen alle oder

bereiteten Anwendungsformen kann in weiten Bereichen variieren. Die Wirkstoffkonzentration der Anwendungsformen kann von
0,0000001 bis zu 100 Gew.-% Wirkstoff, vorzugsweise zwischen
0,01 und 10 Gew.-% liegen.
Die Anwendung geschieht in einer den Anwendungsformen angepaßten
üblichen Weise.
Bei der Anwendung gegen Hygiene- und Vorratsschädlinge zeichnen sich die Wirkstoffe durch eine hervorragende Residualwirkung auf Holz und Ton sowie durch eine gute Alkalistabilität
auf gekalkten Unterlagen aus.

Zur Herstellung einer zweckmäßigen Wirkstoffzubereitung vermischt man 1 Gewichtsteil Wirkstoff mit der angegebenen
Menge Lösungsmittel und der angegebenen Menge Emulgator und verdünnt das Konzentrat mit Wasser auf die gewünschte
Konzentration.
Mit der Wirkstoffzubereitung werden Kohlpflanzen (Brassica
oleracea), welche stark von der Pfirsichblattlaus (Myzus persicae) befallen sind, tropfnaß besprüht.
Nach den angegebenen Zeiten wird die Abtötung in % bestimmt. Dabei bedeutet 100 %, daß alle Blattläuse abgetötet
wurden; 0 % bedeutet, daß keine Blattläuse abgetötet wurden.

Test mit parasitierenden Fliegenlarven
Lösungsmittel: 35 Gewichtsteile Aethylenpolyglykolmonomethy-
äther Emulgator: 35 Gewichtsteile Nonylphenolpolyglykoläther
Zur Herstellung einer zweckmäßigen Wirkstoffzubereitung vermischt
man 30 Gewichtsteile der betreffenden aktiven Substanz mit der angegebenen Menge Lösungsmittel, das den oben genannten
Anteil Emulgator enthält und verdünnt das so erhaltene Konzentrat mit Wasser auf die gewünschte Konzentration.
Etwa 20 Fliegenlarven (Lucilia cuprina) werden in ein Test-
röhrchen gebracht, welches ca. 2 cm Pferdemuskulatur enthält. Auf dieses Pferdefleisch werden 0,5 ml der Wirkstoffzubereitung
gebracht. Nach 24 Stunden wird der Abtötungsgrad in % bestimmt. Öabei bedeuten 100 %, daß alle, und
0 %, daß keine Larven abgetötet worden sind.

Herstellungsbeispiele:

Eine Suspension aus 9,3 g (5o m Mol) l-Methyl-3-n-propylthiomethyl-5-hydroxipyrazol,
8,4 g (60 m Mol) gemahlener Pottasche und 2oo ml Acetonitril läßt man eine Stunde bei 5o C rühren,
kühlt anschließend auf Raumtemperatur ab und fügt 8,6 g (5o m Mol) O-Äthyl-äthanthionophosphonsäureesterchlorid hinzu. Nach
einstündigem Rühren bei 5o°C wird die Reaktionslösung mit 2oo ml Wasser und mit 3oo ml Toluol ausgeschüttelt. Die organische
Phase wird über Magnesiumsulfat getrocknet und nach der Filtration das Toluol am Rotationsverdampfer im Vakuum abgezogen.
Es verbleiben 14 g (87 % der Theorie) O-Äthyl-O-QL-methyl-3-npropylthiomethyl-pyrazol(5)yl]-thionoäthanphosphonsäureester
in Form eines gelben Öls mit dem Brechungsindex nß :l,522o.

Die zur Herstellung der erfindungsgemäßen Verbindungen benötigten
Ausgangsstoffe werden z.B. wie im folgenden beschrieben
hergestellt:
a) C2H5O-CH2-CO-CH2-CO-OC9H5
1. Stufe
In eine Lösung von 84 g (1 Mol) Diketen in trockenem Tetrachlorkohlenstoff
leitet man bei -20°C 72 g (1 Mol) Chlor ein, tropft die Reaktionslösung anschließend unter Rühren in 200 iul
Äthanol, wobei die Temperatur 0 C nicht übersteigen sollte, zieht anschließend das Solvens im Vakuum am Rotationsverdampfer
ab und destilliert den Rückstand. Es werden 155 g (94 % der Theorie) 5^ -Chloracetessigsäureäthylester mit dem
Siedepunkt 90 C/7 Torr gewonnen.
2. Stufe
2,2 Mol Natriumäthylat in Äthanol - hergestellt aus 55,2 g (2,2 Mol) Natrium und 500 ml Äthanol - werden mit 500 ml
Tetrahydrofuran verdünnt. Man versetzt die Lösung bei Raumtemperatur so schnell mit 164,5 g (1 Mol) Pf-Chloracetessigsäureäthylester,
daß die Reaktxonstemperatur auf 50 C ansteigt kühlt die Mischung anschließend auf Raumtemperatur ab, fügt
72 g (1,2 Mol) Eisessig hinzu, dampft das Lösungsmittel am Rotationsverdampfer im Vakuum ab, schüttelt den Rückstand
mit 250 ml Wasser, extrahiert 2 χ mit je 250 ml Methylenchlorid und trocknet die vereinigten organischen Phasen über
Magnesiumsulfat. Das Lösungsmittel wird im Vakuum abgezogen und der Rückstand destilliert. Man erhält so 123,7 (71 %
der Theorie) X -Äthoxyacetessigsäureäthylester mit dem Siedepunkt von 75°C/3 Torr.

Analog wird der X -Methoxyacetessigsäureinethylester in einer
Ausbeute von 63 % der Theorie und dem Kp. "^ 76 C/7 Torr hergestellt:
b) 11-C3H7-S-CH-CO-CH2-CO-OC2H5
Zu einer Lösung von 300 g (2,3 Mol) Acetessigsäureäthylester in 350 ml Äther tropft man bei 00C innerhalb von 90 Minuten
367 g (2,3 Mol) Brom, rührt die Mischung anschließend 1 Stunde bei Raumtemperatur nach, versetzt sie unter Eiskühlung mit
500 ml Wasser, trennt die Phasen und wäscht die Ätherphase einmal mit 100 ml einer TO %igen Natriumbicarbonatlösung.
Die über Magnesiumsulfat getrocknete Ätherphase tropft man bei Raumtemperatur in eine äthanolische Lösung von
2 Mol Natriumpropylmercaptid-hergestellt aus 46 g (2 Mol)
Natrium, 600 g Äthanol und 152 g (2 Mol) Propy!mercaptan
- rührt den Ansatz noch 1 Stunde bei Raumtemperatur nach, schüttelt ihn anschließend mit 2 Mol Natronlauge (1334 g
einer 6 %igen Lösung) aus, verwirft die Ätherphase, säuert die wäßrige Phase mit konzentrierter Salzsäure bis
pHv2 an, schüttelt sie 3 χ mit je 300 ml Methylenchlorid
aus, trocknet die vereinigten organischen Phasen über Magnesiumsulfat und destilliert das Lösungsmittel ab.
Nach der Destillation des Rückstandes werden 200 g (50 % der Theorie) ^^Propylmercaptoacetessigester mit dem Kp.
—85°C/O,7 Torr gewonnen.
Analog werden hergestellts
if -Methylmercaptoacetessigester in 49 %iger Ausbeute und
einem Kochpunkt von 100°C/2 Torr
y -Ä'thylmercaptoacetessigester in 56 %iger Ausbeute mit
einem Kochpunkt von 99°C/1 Torr

Zu einer Lösung von 38 g (ο,2 Mol) X -Propylthioacetessigsäuremethylester
in 15o ml Äthanol tropft man 9,2 g (o,2 Mol) Methylhydrazin in der Weise, daß die Temperatur auf 55-6O°C ansteigt,
und rührt die Mischung anschließend noch 1 Stunde bei 60°c nach. Zur Aufarbeitung der Reaktionslösung wird das Lösungsmittel
am Rotationsverdampfer im Vakuum vollständig abdestilliert, der kristalline Rückstand mit 25o ml Äther verrührt,
abgesaugt und getrocknet. Es verbleiben 24 g (65 % der Theorie) l-Methyl^-n-propylthiomethyl-S-hydroxi-pyrazol in Form farbloser
Kristalle mit dem Schmelzpunkt lo8-llo°C.
