Description
2. Verfahren zur Herstellung der Verbindungen nach Anspruch I, bei denen R2 Wasserstoff bedeutet,
dadurch gekennzeichnet, daß man eine Verbindung gemäß Anspruch 1, bei der R2 niederes Alkyl bedeutet,
mit einem Ätherspaltungsmitte! wie Alumlnlumchiorld oder Pyrldlnhydrochlorld, behandelt.
Die Verbindung betrifft die anspruchsgemäß definierten 5-Hydroxy- bzw. 5-Alkoxy-l-lndanone der allgemeinen Formel I, die als Zwischenprodukte bei der Herstellung von 2,2-disubstltulerten l-Oxo-5-lndanyioxy-carbon-
3« säuren verwendet werden können. Letztere sind wirksame Dluretlca und Saluretlca, die zur Behandlung von
Krankheiten verwendet werden können, bei denen Elektrolyt und Flüssigkeit Im Körper zurückgehalten wird.
Sie werden in der DE-OS 23 51 412 beschrieben.
In den Verbindungen der allgemeinen Formel I bedeutet R beispielsweise Methyl, Äthyl, n-Propyl, Isopropyl,
N-Butyl, Isobutyl, teri.-Butyl oder n-Pentyl, Cyclopropyl, Cyclobutyl, Cyclopentyl oder Cyclohexyl; R1 bedeutet
.'5 beispielsweise Methyl, Äthyl, N-Propyl, Isopropyl, η-Butyl, Isobutyl, tert.-Butyl oder n-Pentyl, Allyl, 1-, 2- oder
3-Butenyl, 1-, 2-, 3- oder 4-Penthenyl, Benzyl, Phenetyl oder Phenylpropyl, Cyclopropylmethyl oder Cyclopentylmethyl. R und R' können mit dem Kohlenstoffatom, an das sie gebunden sind, zu einem Cycloalkylrest
mit 3 bis 7 Kernkohlenstoffatomen zusammengeschlossen sein, z. B. Cyclopropyl, Cyclobutyl, Cyclopentyl,
Cyclohexyl oder Cycloheptyl.
■S' Die erfindungsgemäßen Zwischenprodukte der allgemeinen Formel I können nach dem In der DE-OS
23 51 412 beschriebenen Verfahren zu den 2,2-dlsubstltulerten l-Oxo-5-lndanyloxy-carbonsäuren welterverarbeltet werden. Beispielsweise nach einer Verätherungsmethode, die darin besteht, daß man eine Halogencarbonsäure oder einen Ester derselben der allgemeinen Formel

Die crilndungsgcmallcn 2,2-dlsubstltulcrien 5-Ilydroxy-l-lndanone werden hergestellt. Indem man das
entsprechend substituierte, 2,2-dlsubstltulerte 5-nled.Alkoxy-l-lndanon mit einem Ätherspaltungsmittel, wie
Alumlnlumchlorld, Pyrldlnhydrochlorld oder Natrium In flOssigem Ammoniak behandelt. Wenn man mit
Alumlnlumchlorld arbeitet, kann man ais Lösungsmittel beispielsweise Heptan, Schwefelkohlenstoff oder
Methylenchlorid verwenden, und wenn man mit Pyridin-hydrochlorld arbeitet, braucht man überhaupt kein
Lösungsmittel. Diese Umsetzung wird durch die folgende Gleichung erläutert:

Die erfindungsgemaßen 2,2-dlsubstitulerten 5-nled.Alkoxy-l-indanone werden hergestellt. Indem man ein 2-substltulertes 5-nled-Alkoxy-l-lndanon (III) mit einem geeigneten Alkyllerungsmlttel der allgemeinen Formel
R1Z umsetzt, worin R' und Z die obigen Bedeutungen haben. Diese Umsetzung wird durchgeführt, Indem man
zunächst das 2-substituierte 5-nied.Alkoxy-l-lndanon (III) mit einer geeigneten Base, z.B. einem Alkallhydrid,
wie Natriumhydrid, oder einem Alkallalkoholat, z. B. Kallum-tert.-butylat, umsetzt. Man kann auch andere
Basen verwenden, wie Natriumarnld und Llthlumamld. Die baslfizlerte Verbindung wird dann mit dem Alkyllerungsmlttel R1Z umgesetzt. Hierfür kann man jedes Lösungsmittel verwenden, welches sich gegenüber den
Reaktionsteilnehmern Inert oder praktisch Inert verhält. Geeignete Lösungsmittel sind z. B. 1,2-Dlmethoxyäthan, tert. Butanol, Benzol und Dimethylformamid. Die Umsetzung kann bei etwa 25 bis 1500C durchgeführt
werden. Im allgemeinen wird sie bei Temperaturen Im Bereich von etwa 75 bis 90" C durchgeführt. Sie wird
durch die folgende Gleichung erläutert:

Die bei dem obigen Verfahren verwendeten, 2-substltulerten 5-nled.Alkoxy-l-lndanone (III) können nach
mehreren Verfahren hergestellt werden. Ein Verfahren besteht darin, daß man das 2-substltulerte 5-Hydroxy-l-Indanon mit einem Alkyllerungsmittel, wie Dimethylsulfat oder Dläthylsulfat, In Gegenwart einer Base, wie
Natriumhydroxid oder Kaliumhydroxid, behandelt. Man kann auch andere Alkyllerungsmittel verwenden, wie
Methyljodld, Äthyljodld und dergleichen, wobei man Dimethylformamid als bevorzugtes Lösungsmittel und
Kaliumcarbonat als Base verwendet. Die bei diesem Verfahren verwendeten 2-substltuierten 5-Hydroxy-l-lndanone sind aus der US-PS 36 68 241 bekannt.
Eine zweite Methode zur Herstellung der 2-substltulerten 5-nled.Alkoxy-l-lndanone (HI) 1st die Cycllalkylierung eines Im Kern durch eine niedere Alkoxygruppe substituierten (2-Alkylldenalkanoyl)-benzols (IV) durch
Behandlung mit einer als Elektronenakzeptor wirkenden Säurre, z. B. einer Lewis-Säure, wie konzentrierter
Schwefelsäure, Polyphosphorsäure und Bortlfluorld. Die Umsetzung kann bei etwa 0 bis 6O0C durchgeführt
werden und wird Im allgemeinen bei Raumtemperatur durchgeführt. Sie wird durch die folgende Gleichung

Die 2-Splro-5-nled.alkoxy-l-lndanone (IUa) werden hergestellt, Indem man ein 2-(to-Halogenalkyl)-5-nied.alkoxy-1-lndanon
(III b) mit einer Base, z. B. einem Alkalihydrid, wie Natriumhydrid, in einem geeigneten
Inerten Lösungsmittel, wie 1,2-Dimethoxyäthan, bei der Rückflußtemperatur des betreffenden Lösungsmittels
behandelt. Diese Umsetzung wird durch die folgende Gleichung erläutert:

In der obigen Gleichung bedeutet y eine ganze Zahl von 3 bis 6.
Die in der obigen Gleichung verwendeten. Im Kern durch eine niedere Alkoxygruppe substituierten (2-AlkylidenalkanoyO-benzole
(IV) können nach verschiedenen Methoden hergestellt werden. Eine Methode, die auf die
Herstellung von Im Kern In 4-Stellung durch eine niedere Alkoxygruppe substituierten (2-MethylenalkanoyD-benzolen
(IV a) beschränkt Ist, besieht darin, daß man ein durch eine niedere Alkoxygruppe kernsubstituiertes
4-Alkanoylbenzol (V) mit Dlmethylamln-hydrochlorld und Paraformaldehyd umsetzt und das so erhaltene
Mannich'sche Zwischenprodukt (V a) mit wäßriger Natrlumblcarbonatlösung oder wasserfreiem Dimethylformamid
mit oder ohne Erhitzen behandelt. So erhält man die gewünschte Verbindung IV a. Diese Reaktion wird
durch die folgende Gleichung erläutert:

Eine zweite Methode zur Herstellung der durch dnc niedere Alkoxygruppe kcmsubsiluilcrlcn 2-(AlkylldetuilkanoyD-benzole
(IV b), die auf die Herstellung derjenigen Verbindungen beschrankt Ist, hol denen R die
Methylgruppe bedeutet, besteht darin, daß man ein durch eine niedere Alkoxygruppe kernsubstllulertes 2-Brom-2-methylproplonylbenzol
(VI) mit einem Bromwasserstoffabspaltungsmltiel, wie Llthlumbromld oder Lithlumchlorld
behandelt. Ein geeignetes Lösungsmittel für diese Reaktion Ist beispielsweise Dimethylformamid. Diese
Umsetzung wird vorteilhaft bei etwa 50 bis 120" C Im Verlaufe von I bis 6 Stunden durchgeführt. Sie wird
durch die folgende Gleichung erläutert:

Eine dritte Methode zur Herstellung der Verbindungen der Formel IV, nämlich der Homologen (IV c), besteht
darin, daß man ein durch eine niedere Alkoxygruppe kernsubstituiertes Benzol (VIII) mit einem geeigneten
verzwelgtkettlgen Alkanoylhalogenid, In Gegenwart eines Frledel-Crafts-Katalysators zu dem entsprechenden 4-nled.alkoxy-kernsubstltulerten
Alkanoylbenzol (VI a) umsetzt, welches halogeniert wird, worauf man Halogenwasserstoff
abspaltet und das 4-(2-Alkylldenalkanoyl)-benzol (IVc) erhält. Diese Umsetzung wird durch die
folgende Gleichung erläutert:

Die In der Stellung Nr. 4 durch eine niedere Alkoxygruppe substituierten Alkanoylbenzole (V) sind entweder
bekannte Verbindungen oder können durch Umsetzung eines Alkanoylhalogenids mit einem durch eine niedere
Alkoxygruppe kernsubstituierten Benzol (VIII) in Gegenwart eines Friedei-Crafts-Katalysators, wie Aluminiumchlorid,
hergestellt werden. Lösungsmittel und Temperatur für diese Reaktion sind nicht besonders ausschlaggebend;
man kann jedes Lösungsmittel mit guten Ergebnissen verwenden, das sich gegenüber dem Acylhalogenld
und den durch die niedere Alkoxygruppe kernsubstituierten Benzolen indifferent verhält. Methylenchlorid ist
ein besonders geeignetes Lösungsmittel. Diese Umsetzung verläuft folgendermaßen:

Die bei der obigen Reaktion verwendeten, durch eine niedere Alkoxygruppe kernsubstituierten (2-Brom-2-methylproplonyD-benzole werden durch Bromleren der entsprechenden, durch eine niedere Alkoxygruppe kernsubstltulerten (2-Methylproplonyl)-benzole unter bekannten Bromlerungsbedlngungen hergestellt, wie es die
folgende Gleichung erläutert:

Das bei der obigen Reaktion verwendete, durch eine niedere Alkoxygruppe kernsubstiiuierte (2-MethyiproplonyD-benzol (IX) wird, wie oben beschrieben, unter Verwendung eines Isobutyrylhalogenldes als Alkanoylhalogenld hergestellt, wie es die folgende Gleichung erläutert:

Ein Gemisch aus 18,3 g (0,075 Mol) 2-Äthyl-5-hydroxy-6,7-dlchlor-l-lndanon, 23 g Kaliumcarbonat und 15 ml
Methyljodld in 100 ml Dimethylformamid wird unter Rühren 2 Stunden auf 55° C erwärmt und in 300 ml
Wasser gegossen. Es scheiden sich 18 g 2-Äthyl-5-methoxy-6,7-dlchlor-l-lndanon ab, welches nach dem Umkristallisieren aus Butylchlorld bei 146 bis 147° C schmlla.

Stufe B:
2-ÄthyI-2-methyl-5-methoxy-6,7-dlchlor-l -lndanon
2,1 g einer 57%lgen Öldlsperslon (0,05 Mol) von Natriumhydrid werden mit Petroläther gewaschen und unter
Stickstoff mit 500 ml 1,2-Dlmethoxyäthan und 13 g (0,05 MoI) 2-Äthyl-5-methoxy-6,7-dlchIor-l-lndanon In
einen Kolben überführt. Das Reaktionsgemisch wird In einer Inerten Atmosphäre 2 Stunden auf RuckfluBtemperatur erhitzt, dann auf 250C gekühlt, mit 7 ml Methyljodld versetzt, 10 Minuten auf ROckflußtemperatur
eThltzt und In 1,51 Eiswasser gegossen. Es scheiden sich 12,0 g 2-Äthyl-2-methyl-5-methoxy-6,7-dlchlor-l-lndanon ab, welches nach dem Umkristallisieren aus Methyicyclohexan bei 121° C schmilzt.

Aluminiumchlorid In 180 ml Heptan wird 1 Stunde auf RückfluBtemperatur erhitzt und dann gekühlt. Das
Heptan wird dekantiert und das Produkt durch Abkratzen In eine Lösung von 10 ml konzentrierter Salzsäure In
100 ml Eiswasser überführt, mit 200 ml Äther extrahiert und der Extrakt mit Wasser gewaschen und über
Magnesiumsulfat getrocknet. Der Äther wird abgedampft und das Produkt aus 500 ml Buiylchlorld umkrlstalllslcrt.
Man erhalt 3,7 g 2-Alhyl-2-melhyl-5-hydroxy-(>,7-dlchlor-l-lndanon; Γ. 215 his 217" Γ.

Ein Gemisch aus 100 g (0,565 Mol) 2,3-Dlchloranlsol und 66 g (0,62 Mol) Isobutyrylchlorld In 400 ml Methylenchlorid
wird unter Rühren auf 5° C gekühlt und Im Verlaufe einer Stunde mit 83 g (0,62 Mol) Alumlnlumchlorld
versetzt. Man laßt das Reaktionsgemisch eine Temperatur von 25° C annehmen und gießt es nach 24 ;<
Stunden In 400 ml Eiswasser, welches 30 ml Salzsäure enthält. Die organische Phase wird mit 5prozentlger
Natronlauge und mit Wasser gewaschen, über Magnesiumsulfat getrocknet und unier vermindertem Druck
destilliert. Man erhält 68 g 2',3'-Dlchlor-4'-methoxy!sobutyrophenon, welches bei 0,7 hPa bei 120 bis 130cC
überdestilliert.

Natriumhydrid in 500 ml wasserfreiem 1,2-Dlmethoxyäthan wird unter Rühren 1 Stunde in einer Inerten Atmo
sphäre auf 80 bis 85° C erhitzt, dann auf 30° C gekohlt und mit 8 ml Methyljodld versetzt. Das Reaktionsge
misch wird 15 Minuten auf RückfluBtemperatur erhitzt, worauf man das Lösungsmittel unter vermindertem
Druck abdestilliert. Das Rohprodukt wird In 500 ml Wasser gegossen, mit Äther extrahiert und der Extrakt mit
Wasser gewaschen und über Magnesiumsulfat getrocknet. Nach dem Abdampfen des Äthers wird das Produkt
aus Methylcyclohexan umkrlstalllslert. Man erhält 8,2 g 2,2-Dlmethyl-5-methoxy-6,7-dlchlor-l-Indanon;

gekühlt. Das Heptan wird von dem Reaktionsgemisch abdekantiert und der feste Rückstand auf ein Gemisch
aus 500 g Eis und 50 ml konzentrierter Salzsäure gegossen. Es scheiden sich 7,6 g Produkt ab, weiches nach dem
Umkristallisieren aus Nltromethan bei 273° C schmilzt.

1,3 g (0,055 Mol) Natriumhydrid, 500 ml 1,2-Dlmethoxyäthan und 6 ml n-Propyljodld. Bei diesem Verfahren
erhält man 6,0 g 2-Äthyl-2-n-propyl-5-methoxy-6,7-dlchlor-l Indanon, welches nach dem Umkristallisieren aus
Äthanol bei 92° C schmilzt.

chlorld versetzt. Man läßt das Reaktlonsgemlsch sich auf 25° C anwärmen und gießt es nach 24 Stunden in eine so
Lösung von 600 ml Salzsäure In 31 Eiswasser. Die organische Phase wird mit lOprozentiger Natronlauge und
mit Wasser gewaschen und über Magnesiumsulfat getrocknet. Nach dem Abdampfen des Lösungsmittels wird
das Produkt aus Hexan umkristallisiert. Man erhält 295 g 2\3'-Dlchlor-4'-methoxylsovalerophenon; F. 49 bis
54° C.

Ein Gemisch aus 261,6g (1,0 Mol) 2',3'-Dlehlor-4'-meihoxylscivalerophcnon, 75,Og (2.5 Mol) Paral'ormaldehyd,
327 g (4,0 Mol) Dlmethylamln-hydrochlorld und 26 ml Essigsaure wird 18 Stunden unter Rühren auf
dem Dampfbad erhitzt, mit 500 ml Dimethylformamid versetzt, weitere 3 Stunden erhitzt und dann In 1,7 1 <■-Eiswasser
gegossen. Das sich abscheidende Rohprodukt wird In 600 ml Benzol gelöst und die Lösung über
Natriumsulfat getrocknet. Nach dem Abdampfen des Benzols erhält man 237 g 2-Methylen-2'.3'-dlchlor-4'-methoxylsovalerophenon
(F. 46 bis 51° C), welches ohne weitere Reinigung In Stufe C eingesetzt wird.
Stufe C:
2-Isopropyl-5-methoxy-6,7-dlchlor-l-indanon
25° C gerührt und dann langsam zu einer reichlichen Menge Eiswasser zugesetzt. Das sich abscheidende
getrocknet. Durch Umkristallisieren aus einem Gemisch aus Benzol und Hexan erhält man 134 g 2-Isopropyl-S-
methoxy-6,7-dlchlor-l-lndanon; F. 118 bis 1190C.

2-Methyl-2-lsopropyl-5-methoxy-6,7-dlchloΓ-l-lndanon
Eine Suspension von 7,3 g (0,025 Mol) 2-Isopropyl-5-methoxy-6,7-dlchlor-l-lndanon und 810 mg (0,028 Mol)
Natriumhydrid In 250 ml wasserfreiem 1,2-Dlmethoxyäthan wird 1 Stunde unter Rühren in einer Inerten Atmosphäre auf 80 bis 85° C erhitzt, dann auf 30° C gekühlt und mit 6 ml Methyljodld versetzt. Das Reaktlonsge-
-° misch wird auf 80° C erhitzt, worauf man das Lösungsmittel unter vermindertem Druck abdestlllleit und den
Rückstand in Eiswasser gießt. Es scheiden sich 7,0 g 2-Methyl-2-lsopropyl-5-methoxy-6,7-dlchlor-l-lndanon ab,
welches nach dem Umkristallisieren aus einem Gemisch aus Äthanol und Wasser bei 143° C schmilzt.

Eine Suspension von 7,0 g (0,0244 Mol) 2-Methyl-2-lsopropyl-5-methoxy-6,7-dlchlor-I-lndanon und 9,0 g
(0,068 Mol) Aluminiumchlorid In 400 ml Heptan wird unter Rühren 1 Stunde auf Rückflußtemperatur erhitzt
und dann gekühlt. Das Heptan wird von dem Reaktlonsgemlsch abdekantiert und der feste Rückstand In eine
Lösung von 20 ml konzentrierter Salzsäure In 300 ml Wasser gegossen. Das Rohprodukt wird mit 300 ml Äther
JS extrahiert, der Extrakt mit Wasser gewaschen, über Magnesiumsulfat getrocknet, bis auf ein Volumen von
100 ml destilliert und mit 100 ml Hexan versetzt. Es scheiden sich 6,5 g Σ-Με^Ι-Σ-Ιδορη^Ι-δ-Ι^Γί^-ό,?-
dlchlor-1-lndanon ab; F. 215° C.

57.8 g (0,327 Mol) 2,3-Dlchloranlsol werden In 300 ml Dlchlormethan gelöst, und die Lösung wird mit 52,7 g
(0.367 Mol) Cyclopentylacetylchlorld versetzt. Die Lösung wird auf+50C gekühlt und allmählich Im Verlaufe
^1 einer Stunde bei +5° C mit 48,0 g (0,36 Mol) Alumlnlumchlorld versetzt. Das Gemisch wird 2 Stunden bei +5° C
und dann 16 Stunden bei 20 bis 25° C gerührt und hierauf In eine Lösung von 150 ml 12 η-Salzsäure In 1 1
Eiswasser gegossen. Die organische Phase wird abgetrennt und die wäßrige Phase mit Dlchlormethan extrahiert.
Die vereinigten organischen Phasen werden mit Kochsalzlösung, 10%lger Natronlauge und wieder mit Kochsalzlösung
gewasche.i und über Magnesiumsulfat getrocknet. Nach dem Abdampfen des Lösungsmittels hinterbleibt
« ein brauner fester Stoff, der aus Hexan umkristallisiert wird. So erhält man 53,2 g 2',3'-Dlchlor-4'-methoxy-2-cyclopentylacetophenon;
F. 60 bis 61,5° C.

man 21,6g (0.72 Mol) Paraformaldehyd und 9,65 g konzentrierte Schwefelsäure zusetzt. Das Gemisch wird 20
Stunden auf 80 bis 850C erhitzt. Das Dioxan wird unter vermindertem Druck abgedampft, und zu dem als
Rückstand hinterbleibenden Harz setzt man Wasser zu und extrahiert sodann mit Äther. Der Ätherextrakt wird
mit Wasser gewaschen und über Magnesiumsulfat getrocknet. Der Äther wird abgedampft, und beim Verrühren
des Rückstandes mit 5 ml Hexan erhält man einen festen Stoff, den man aus Llgroln umkristallisiert. Man
erhält 33,3 g 4*-<2-Cyclopentyl-2-methylenacetyl)-2\3'-dlchloranlso!·, F. 59 bis 63° C. Durch Umkristallisieren aus
Butylchlorld gewinnt man eine analysenreine Probe (F. 66 bis 67,5° C).

Stufe C: ι»
2-Cyclopentyl-5-methoxy-6,7-d!chlor-l-Indanon
33,3 g 4'-<2-Cyclopentyl-2-methylenacety!V2\3'-dlchloranlsol werden in 150 ml 98%lger Schwefelsäure gelöst,
und die Lösung wird 1,5 Stunden bei 20° C gerührt. Dann wird die Lösung unter Rühren zu Eiswasser zugetropft. Die wäßrige Phase wird von dem harzigen Produkt dekantiert, worauf man frisches Wasser zusetzt. Nach
20 Stunden erstarrt das Hare und wird aus einem Gemisch aus 3 Teilen Hexan und 1 Teil Benzol umkristallisiert. So erhält man 2-Cyclopenty!-5-methoxy-6,7-dlchlor-l-lndanon; F. 116 bis 117° C.

Stufe D:
2-Cyclopentyl-2-methyl-5-methoxy-6,7-dlchlor-1 -Indanon
7,5 g (0,025 Mol) 2-Cyclopentyl-5-methoxy-6,7-dlchlor-l-lndanon werden unter Stickstoff In 200 ml trockenem
1.2-Dlmethoxyäthan gelöst. Die Lösung wird mit Natriumhydrid (57% In Mineralöl; 1,16 g, 0,0275 Mol) versetzt
und das Gemisch bei 80° C gerührt, bis die Wasserstoffentwicklung aufhört (2 Stunden). Dann wird die Lösung
gekühlt, mit 7,5 ml Methyljodld versetzt und das Gemisch wieder auf Rückflußtemperatur erhitzt und sodann
gekühlt. Der größte Teil des 1,2-Dlmethoxyäthans wird abgedampft, und nach dem Zusatz von Wasser zu dem ■'"
Rückstand erstarrt dieser bald und wird aus Methylcyclohexan und dann aus einem Gemisch aus 4 Teilen Äthanol und 1 Teil Wasser umkristalllslert. So erhält man 3,4 g 2-Cyclopentyl-2-methyl-5-methoxy-6,7-dlchlor-llndanon; F. 109 bis 111,5° C.

2-Cyclopentyl-2-methyl-5-hydroxy-6,7-dlchlor-l -Indanon «>
3,4 g (0,0109 Mol) 2-Cyclopentyl-2-methyl-5-methoxy-6,7-dlchlor-l-Indanon werden zu 180 ml trockenem
Heptan zugesetzt, worauf man 4,36 g (0,0327 Mol) Alumlnlumchlorld zusetzt. Das Gemisch wird 1 Stunde auf
Rückflußtemperatur erhitzt und das Heptan dann von dem harzartigen Rückstand abdekantiert. Der Rückstand
wird zu einer Lösung von 15 ml 12 η-Salzsäure In 200 ml Eiswasser zugesetzt. Der sich abscheidende feste Stoff ·»?
wird aus Benzol umkristalllslert. Man erhält 2,77 g 2-Cyclopentyl-2-methyl-5-hydroxy-6,7-dlchlor-l-lndanon;
F. 19ObIs 1940C.

Stufe A:
2-Cyclopentyl-2-methyl-5-methoxy-6,7-dlchlor-l -Indanon
Eine Lösung von 7,35 g (0,03 Mol) 2-Methyl-5-methoxy-6,7-dlchlor-l-lndanon In 100 ml tert.Butylalkohol
wird In einer Inerten Atmosphäre auf Rückflußtemperatur erhitzt und mit 4,48 g (0,04 Mol) Kallum-tert.butylat
versetzt. Nach 1 '/2stündlgem Erhitzen am Rückflußkühler setzt man 9,9 g (0,06 Mol) Cyclopentylbromld zu und «'
erhitzt noch eine weitere Stunde auf Rückflußtemperatur. Das Reaktionsgemisch wird In 1 1 Wasser gegossen,
und man erhält 2-Cyciopentyl-2-methyl-5-methoxy-6,7-dlchlor-l-!ndanon; F. 109 bis IH0C.
Stufe B:
2-Cyclopentyl-2-methyl-5-hydroxy-6,7-dlchlor-l -Indanon ^
3,4g (0,0109 Mol) 2-Cyclopentyl-2-methyl-5-methoxy-6,7-dlchlor-l-lndanon werden zu 180ml trockenem
Heptan zugesetzt, worauf man 4,36 g (0,0327 Mol) Alumlnlumchlorld zusetzt. Das Gemisch wird 1 Stunde auf
Rückflußtemperatur erhitzt und das Heptan dann von dem harzartigen Rückstand abdekantlert. Der Rückstand
wird zu 200 ml Eiswasser, die 15 ml 12 η-Salzsäure enthalten, zugesetzt. Der feste Niederschlag wird aus Benzol
umkristallisiert. So erhält man 2,77 g 2-Cyclopentyl-2-methyl-5-hydroxy-6,7-dlchlor-l-lndanon; F. 190 bis
194° C.

Stufe C:

Ein Gemisch aus 2,98g (0,01 Mol) 2-CyclopentyI-2-methyl-5-hydroxy-6,7-dlchlor-l-lndanon, 2,15 g (0,011
Mol) Bromesslgsäure-tert-butylester, 276 g (0,02 Mol) Kaliumcarbonat und 15 ml Dimethylformamid wird 1
Stunde bei 25° C gerührt und dann In 50 ml Wasser gegossen. Der sich abscheidende (l-Oxo^-cyclopentyl-oJ-dlchlor-5-lndanyloxy)-esslgsäure-tert.butylester wird abfiltriert, mit Wasser gewaschen und getrocknet.
Stufe D:
(l-Oxo^-cyclopentyl^-methyl-öJ-dlchlor-S-lndanyloxyJ-essigsäure
Eine Lösung von 4,13 g (0,01 Mol) (l-Oxo-2-cyclopentyl-2-methyl-6,7-dlchlor-5-lndanyloxy)-esslgsaure-
^5 tert.butylester und 0,3 g p-Toluolsulfonsäure In 50 ml Benzol wird 10 Minuten auf Rückflußtemperatur erhitzt
und dann auf 250C gekühlt. So erhalt man (l-Oxo-2-cyclopentyl-2-methyl-6,7-dlchlor-5-lndanyloxy)-esslgs!lure,
die nach dem Umkristallisieren aus Essigsäure bei 113 bis 114° C schmilzt.
Stufe A:
2-Benzyl-2-methy!-5-methoxy-6,7-dlchlor-l-lndanon
1,68 g (0,015 Mol) Kallum-tert.butylat In 40 ml trockenem tert. Butylalkohol werden zu einer am Rückflußkühler siedenden Losung von 2-Methyl-5-methoxy-6,7-dlchlor-l-indanon In 50 ml Benzol zugesetzt. Nach 30
Minuten langem Erhitzen auf Rückflußtemperatur tropft man eine Lösung von 1,72 g (0,01 Mol) Benzylbromid
In 10 ml Benzol zu. Nach weiterem 30 Minuten langem Erhitzen auf Rückflußtemperatur wird das Gemisch
gekühlt, mit 10 ml Wasser versetzt und zur Trockne eingedampft. Der Rückstand wird mit Wasser gewaschen
4<> und In Äther aufgenommen. Die Ätherlösung wird mit Wasser und mit gesättigter Kochsalzlösung gewaschen,
über Natriumsulfat getrocknet und eingedampft. Der sirupartige Rückstand von 2-Benzyl-2-methyl-5-methoxy-6,7-dlchlor-l-lndanon (3 g) wird unmittelbar ohne Reinigung In der nächsten Verfahrensstufe eingesetzt.
Stufe B:
2-Benzyl-2-methyl-5-hydroxy-6,7-dlchlor-1 -Indanon
6 g 2-Benzyl-2-methy!-5-methoxy-6,7-dlchlor-l-Indanon werden mit 60 g Pyrldln-hydrochlorld gemischt, und
das Gemisch wird 3 Stunden auf 180°C erhitzt. Die dunkle Flüssigkeit wird In 1 I Wasser gegossen und das dunkle pulverförmlge Produkt gesam.nelt und aus Essigsäure umkrlstalllslert. So erhält man 2 g 2-Benzyl-2-
*'· methyl-S-hydroxy-oJ-dlchlor-l-indanon; F. 219 bis 221°C. Das Produkt wird durch sein kernmagnetisches

Stufe A:
2,3-Dlchlor-4-(6-bromhexanoyl)-anlsol
Ein Gemisch aus 89 g (0,50 Mol) 2,3-Dlchloranisol und 120 g (0,59 Mol) 6-Bromhexanoylchlorld In 500 ml
<■< Methylenchlorid wird unter Rühren auf 5° C gekühlt und Im Verlaute von V2 Stunde antellwelsc mit 74 g (0,56
Mol) Alumlnlumchlorld versct/t. Das Rcakllonsgemlsch wird IX Stunden auC 25" C gehalten und dann In eine
lösung von 100 ml Sal/silure In I I lilswasser gegossen. I)Ic organische Phase wird abgetrennt und mit Wasser,
mit T'Uger Natronlauge sowie mil verdünnter Sal/silure gewaschen. Das Methylcnchlorid wird unter verminder-
tem Druck abgedampft. Das Rohprodukt wird In 200 ml Äther gelöst, die Lösung Ober Magnesiumsulfat
getrocknet, filtriert und mit 600 ml Hexan versetzt. So erhalt man 2,3-Dlchlor-4-(6-bromhexanoyl)-anlsol; F. 52

2-(4-Chlorbutyl)-5-methoxy-6,7-dlchlor-l-!ndanon
Ein Gemisch aus 10 g 2,3-Dlchlor-4-(6-bromhexanol)-anisol, 4 g Dlmethylamln-hydrochlorld, 2 g Paraformaldehyd und 0,5 ml Essigsäure wird 2 Stunden auf dem Dampfbad erhitzt, dann mit 30 ml Dimethylformamid
versetzt und noch 2*/j Stunden erhitzt. Das Reaktionsgemisch wird in Wasser gegossen, mit Äther extrahiert is
und der Extrakt mit Wasser gewaschen und über Magnesiumsulfat getrocknet. Nach dem Abdampfen des
Äthers erhät man 9 g rohes 2,3-Dlchlor-4-(6-chlor-2-methylenhexanoyi)-anlsoi, weiches durch Behandein mit
50 ml konzentrierter Schwefelsäure cycllalkyllert wird. Die Schwefelsäurelösung wird In 300 ml Wasser gegossen,
worauf man 5,8 g (2-(4-Chlorbutyl)-5-methoxy-6,7-dlchlor-l-lndanon gewinnt, welches nach dem Umkristallisieren aus Cyclohexan bei 92° C schmilzt.
Analyse
Cl = 33,07%;
gefunden: C = 52,25%; H = 4,50%;
Cl = 33,03%.
Stufe C:
S'-Methoxy-o'J'-dlchlorsplro-icyclopentan-l^'-lndanon)
Eine Suspension von 370 mg (0,0155 Mol) Natriumhydrid In 250 ml 1,2-Dlmethoxyäthan wird unter Rühren
In einer Inerten Atmosphäre auf Rückflußtemperatur erhitzt. Nach Zusatz einer Lösung von 4,5 g (0,014 Mol) 2-(4-Chlorbutyl)-5-methoxy-6,7-dlchlor-l-lndanon In 50 ml 1,2-Dlmethoxyäthan Im Verlaufe von 20 Minuten
erhitzt man noch 3 Stunden auf Rückflußtemperatur. Dann wird das Lösungsmittel bis auf ein Volumen von
50 ml abdestilllert und der Rückstand In 300 ml Wasser gegossen. So erhält man 2,6 g 5'-Methoxy-6',7'-dlchlorsplro-(cyclopentan-l,2'-indanon), welches nach dem Umkristallisieren aus einem Gemisch aus Äthanol und
Wasser bei 170° C schmilzt.
Analyse
C = 59,34%; H = 5,08%.
Stufe D:
S'-Hydroxy-o'J'-dlchlorsplro-icyclopentan-l^'-lndanon)
Ein Gemisch aus 2,6 g (0,0091 MoI) S'-Methoxy-o'J'-dlchlorsplro-icycjopentan-l^'-lndanon) und 26 g Pyridin-hydrochlorld wird 1 Stunde unter Rühren auf 185° C erhitzt und dann in 200 ml Wasser gegossen. Es scheiden sich 2,3 g S'-Hydroxy-o'J'-dlchlorsplro-icyclopentan-l^'-indanon) ab, welches nach dem Umkristallisieren
aus Nltromethan bei 236° C schmilzt.
Analyse
gefunden: C = 57,77%; H = 4,54%.
Beispiel 11
Stufe A:
2,3-Dichlor-4-(7-brornheptanoyl)-anlsol
Die obige Verbindung wird nach dem Verfahren der Stufe A des Beispiels 10 unter Verwendung der folgenden Stoffe hergestellt: 89 g (0,5 Mol) 2,3-Dlchloranlsol, 127 g (0,56 Mol) 7-Bromheptanoylchlorid, 74 g (0,56
Mol) Alumlnlumchlorld und 500 ml Methylenchlorid. Das Verfahren liefert 107 g 2,3-Dlchlor-4-(7-bromheptanoyl)-anlsol (Ausbeute 58%), welches nach dem Umkristallisieren aus Cyclohexan bei 57° C schmilzt.
Analyse
gefunden: C = 45,53%; H = 4,66%.
Stufe B:
2-(5-Chlorpentyl)-5-methoxy-6,7-dlchlor-l-lndanon
2-(5-Chlorpentyl)-5-methoxy-6,7-dlchlor-l-lndanon
Die obige Verbindung wird gemäß Stufe B des Beispiels 10 unter Verwendung der folgenden Stoffe herge-
" stellt: 22,5 g (0,06 Mol) 2,3-Dlchlor-4-(7-bromheptanoyl)-anlsol, 22,5 g Dlmethylamln-hydrochlorld, 4,7 g Paraformaldehyd
und 1,8 ml Essigsäure. Das rohe a\/?-ungesäülgte Keton wird In 100 ml Schwefelsäure cycllalkyllert.
Dieses Verfahren liefert 15g 2-(5-Chlorpentyl)-5-methoxy-6,7-dlchlor-l-lndanon, welches nach dem
Umkristallisieren aus einem Gemisch aus Benzol und Hexan bei 115° C schmilzt.

Die obige Verbindung wird gemäß Stufe A des Beispiels 9 unter Verwendung der folgenden Stoffe hergestellt:
11 g (0,0289 Mol) 2-(5-Chlorpentyl)-5-methoxy-6,7-dlchlor-l-lndanon, 150ml Benzol, 100ml tert. Butanol und
4,86 g Kallum-tert-butylat. Das Verfahren liefert 8,6g S'-Methoxy-o'J'-dlchlorsplro-icyclohexan-l^'-lndanon)
-" (Ausbeute 100%), welches nach dem Umkristallisieren aus Butylchlorld bei 2100C schmilzt.

Berechnet für C15H16CI2O2: C = 60,21%; H = 5,39%;
gefunden: C = 60,37%; H = 5,46%.
Stufe D:
S'-Hydroxy-o'J'-dlchlorsplro-fcyclohexan-l^'-lndanon)
S'-Hydroxy-o'J'-dlchlorsplro-fcyclohexan-l^'-lndanon)
Die obige Verbindung wird gemäß Stufe D des Beispiels 10 unter Verwendung der folgenden Stoffe hergestellt:
9,0g S'-Methoxy-o'J'-dichlorsplro-icyclohexan-l^'-lndanon) und 90g Pyrldln-hydrochlorld. Das Verfahren
liefert 8,0g S'-Hydroxy-o'J'-dlchlorsplro-Ccyclohexan-l^'-lndanon) (Ausbeute 93%), welches nach dem
Umkristallisieren aus Äthanol bei 268 bis 270° C schmilzt.
Analyse

Beispiel 12

2'-Chlor-3'-methy!-4'-methoxy-2-cyclopentylacetophenon
Man arbeitet nach Stufe A des Beispiels 6 unter Verwendung der folgenden Stoffe: 36,5 g (0,234 Mol) 2-Methyl-3-chloranlsol,
37,7g (0,257 Mol) Cyclopentylacetylchlorld, 34,3 g (0,257 Mol) Alumlnlumchlorld und
-< 250 ml Methylenchlorid. So erhält man 50 g 2'-Chlor-3'-methyl-4'-methoxy-2-cyclopenlylacetophenon (Ausbeute
80%). welches bei 0,6 mm Hg bei 145 bis 1650C überdestilliert.

Berechnet für C15H15ClO2: C = 67,54%; H = 7,18%;

Stufe B:

2'-Chlor-3'-methyl-4'-methoxy-2-cyclopentylacrylophenon
Man arbeitet nach Stufe B des Beispiels 6 unter Verwendung der folgenden Stoffe: 45 g (0,169 Mol) 2'-Chlor-S'-methyW-methoxy^-cyclopentylacetophenon,
12,6 g (0,42 Mol) Paraformaldehyd, 61 g (0,75 Mol) Dlmethylamin-hydrochiorld
und 4,2 ml Essigsäure. So erhält man 36,3 g 2'-Chlor-3'-methy!-4'-methoxy-2-cyclopentylacrylophenon
(Ausbeute 78%), welches nach dem Umkristallisieren aus Petroiather bei 45 bis 46,5° C schmilzt.

oxy-o-melhyl^-chlor-l-lndanon (Ausbeute 85%); F. nach dem Umkristallisieren aus Hexan 73 bis 74° C.

Berechnet für C1JInClO2: C = 68,93%; H = 6,87%;
gefunden: C = 69,08%; H = 6,76%.
Stufe D: Z-Cyclopentyl^.o-dlmeihyl-S-meihoxy^-chlor-l-indanon
Man arbeitet nach Stufe D des Beispiels 6 unter Verwendung der folgenden Stoffe: 2,79 g (0,01 Mol) 2-Cyclopentyl-S-methoxy-ö-methyl^-chlor-l-lndanon,
0,264g (0,011 Mol) Natriumhydrid, 100 ml 1,2-Dlmethoxyäthan
und 3,11 ml (0,05 Mol) Methyljodld. So erhält man 2,8 g 2-Cyclopentyl-2,6-dlmethyl-5-methoxy-7-chlor-l-indanon
(Ausbeute 96%), welches nach der Reinigung durch Chromatographie bei 48 bis 52° C schmilzt.

7,36g (0,552 Mol) Alumlnlumchlorld und 200 ml Heptan. Man erhält 3,3g 2-Cyclopentyl-2,6-dlmethyl-5-hydroxy-7-chlor-l-lndanon; F. nach Umkristallisieren aus einem
Gemisch aus Äthanol und Wasser 175 bis 177° C.
Analyse
Berechnet für C16H^ClO2: C = 68,93%; H = 6,87%;
gefunden: C = 69,07%; H = 6,71%.
