Die Erfindung betrifft ein Verfahren zur Herstellung von Propandiol(1,3)-phosphatiden
der Formel I

und R1 und R2 unabhängig voneinander je ein Wasserstoff
atom oder eine Alkylgruppe, vorzugsweise mit 1 bis 6 Kohlenstoffatomen, bedeuten.
Erfindungsgemäß werften die Verbindungen der Formel I hergestellt,
indem man gegebenenfalls in Stellung 2 alkylsubstituiertes Propandiol(1,3) mit einem geeigneten Acylierungsmittel
in an sich bekannter Weise zum Monofettsäureester der allgemeinen Formel II

in der n, R1 und R2 die oben angegebene Bedeutung besitzen,
umsetzt und den Monoester mit ß-Bromäthylphosphorsäuredichlorid zur Reaktion bringt unter Bildung einer Verbindung

welche anschließend mit Trimethylamin, Dimethylamin, Methylamin
oder Ammoniak direkt aminiert wird.
Geeignete Acylierungsmittel sind die Säurechloride und Säureanhydride
der Fettsäuren mit 10 bis 26 C-Atomen. Es können jedoch auch andere acylierende. Derivate dieser Fettsäuren
verwendet werden. Die Umsetzung erfolgt unter üblichen Acylierungsbedingungen,
zweckmäßig in einem wasserfreien polaren organischen Lösungsmittel oder Lösungsmittelgemisch. Bevorzugt
wird als Lösungsmittel eine Mischung von Chloroform und Pyridin im Verhältnis 3:1 biw 1:3. Die Umsetzung kann bei
Zimmertemperatur oder schwach erhöhter Temperatur erfolgen.
Die zweite Stufe des erfindungsgemäßen Verfahrens, die Umsetzung
einer Verbindung von Formel II mit ß-Bromäthylphosphorsäuredichlorid
erfolgt ebenfalls in einem organischen wasserfreien Lösungsmittel. Bevorzugt wird Chloroform. Die
Umsetzung erfolgt vorzugsweise in Gegenwart eines Amins in molarem .Überschuß, bezogen auf das PhosphorSäurederivat. Besonders
bevorzugt erfolgt die Umsetzung in Gegenwart von Triäthylamin.
In der dritten Stufe des erfindungsgemäßen Verfahrens, der Umsetzung des zentralen Zwischenproduktes der Formel III mit
Ammoniak oder einem der genannten Amine, erfolgt in einem polaren organischen Lösungsmittel oder Lösungsmittelgemisch
bei Temperaturen zwischen etwa 0° und der Siedetemperatur des Lösungsmittels bzw. der niedrigst siedenden Komponente
desselben. Bevorzugt wird ein Gemisch von Chloroform und Acetonitril oder Nitromethan. Zur Verbesserung der Löslichkeit
kann dieses noch einen niedrigen Alkanol und Wasser enthalten.

Aus Bull.Soc.CMm.Biol. 4j6, 895-903 (1964) ist bereits ein
Verfahren zur Herstellung von Palmitoylpropandiol(1,3)-phosphoryläthanolamin
bekannt. Die Herstellung erfolgte auf umständliche V/eise und unter Verwendung von Schutzgruppen
in einer vielstufigen Synthese mit sehr schlechten Ausbeuten. Die Erfindung ermöglicht es, diese Verbindung sowie
eine Reihe weiterer neuer Verbindungen entsprechend der obigen allgemeinen Formel I in einfacher Weise mit sehr guten
Ausbeuten herzustellen und macht daher diese Verbindungsgruppe auch technisch zugänglich.
Ein weiterer Gegenstand der Erfindung sind Verbindungen der

ein Wasserstoffatom oder eine Alkylgruppe, vorzugsweise mit
1 bis 6 Kohlenstoffatomen, bedeuten.
Die erfindungsgemäß erhältlichen Verbindungen sind weiße
mikrokristalline Pulver mit uncharakteristischem Schmelzverhalten. Die Charakterisierung erfolgte daher durch Dünnschichtchromatographie
und Elenientaranalyse.

Die Verbindungen der allgemeinen Formel I sind stark grenzflächenaktiv
und weisen eine lytische Aktivität gegenüber natürlichen Membranen (z.B. Erythrocyten) auf. Sie lassen
sich daher in vielfältiger Weise zur Lyse derartiger Membranen verwenden. Außerdem ist eine pharmakologische Wirksamkeit
zu erwarten.
Die folgenden Beispiele erläutern die Erfindung weiter. Beispiel 1
A. Palmitoyl-2,2-dimethylpropandiol(1,3) (Formel II)
Eine Lösung von 100 g (0,96 Mol) 2,2-Dimethylpropandiol(i,3)
in einem Gemisch aus 200 ml absolutem Chloroform und 100 ml Pyridin wird tropfenweise mit 100 g (0,36 Mol) Palmitinsäurechlorid
in 400 ml absolutem Chloroform versetzt. Nach dem Eintropfen läßt man noch eine Stunde weiterrühren und
zieht das Lösungsmittelgemisch am Rotationsverdampfer ab. Das erhaltene noch pyridinhaltige öl wird unter Rühren auf
ein Gemisch aus 1,5 1 0,3 N H2SO, und 1 kg Eis gegeben. Der
weiße Niederschlag wird abgesaugt, getrocknet und aus Hexan umkristallisiert. Man erhält 80 g (65 $>
bezogen auf Palmitinsäurechlorid) analysenreines Palmitoyl-2,2-dimethylpropandiol(1,3).

B. Palmitoyl-2,2-dimethylpropandiol(1,3)-phosphorsäure-ßbromäthylester
(Formel III) ·
Man stellt unter Eiskühlung eine Lösung von 49 g ß-Bromäthyl-

phosphorsäuredichlorid (0,2 Mol) und 40 g abs. Triäthylamin
(0,4 Mol) in 150 absolutem Chloroform her. Bei Raumtemperatur wird in dieses Phosphorylierungsgemisch eine Lösung
von 50 g Palmitoyl-2,2-dimethylpropandiol(1,3) (0,14
Mol) in 250 ml abs. Chloroform unter Rühren eingetropft. Nach 2 bis 3 Std. weiterem Rühren ist das Palmitoyl-2,2-dimethylpropandiol(1,3)
dünnschichtchromatographisch nicht mehr nachzuweisen. Zur Hydrolyse der Phosphorsäurechloride
wird das Reaktionsgemisch 1:1 (v/v) mit Eis versetzt und 20 min gerührt. Die im Scheidetrichter abgetrennte Chloroformphase
wird über Natriumsulfat getrocknet und danach zur Trockne eingeengt. Die Ausbeute beträgt 52 g und stellt
ein Rohprodukt, das zur weiteren Umsetzung mit den Basen Trimethylamin, Dimethylamin, Methylamin oder Ammoniak geeignet
ist, dar.
Zur Analyse wird eine Probe der Substanz säulenchromatographisch
gereinigt.
Analysendaten: bezogen auf das Monohydrat

gefunden: C 50,73 # H 9,16 £ Br 13,8 £ P 5,66 £
C. Palmitoyl-2,2-dimethylpropandiol(1,3)-phosphoryl-äthanol-,·
amin (Formel I)
5 g (10 mMol) des zentralen Zwischenproduktes III (Palmitoyl-2,2-dimethylpropandiol(
1,3)-phosphorsäure-ß-bromäthyleeter) werden in 50 ml Chloroform gelöst und nacheinander
mit 100 ml Acetonitril, 100 ml Methanol und 100 ml 25 #iger wässriger Ammoniaklösung versetzt. Man läßt das Gemisch
24 Std. bei 490C im gut verschlossenen Kolben reagieren.

Danach wird das Lösungsmittelgemisch am Rotationsverdampfer
abgezogen und der Rückstand in 100 ml Chloroform, 120 ml
Methanol und 100 ml 2 5&iger Ameisensäure gelöst und gut geschüttelt.
Die Chloroformphase wird abgetrennt und zur Neutralisation der Ameisensäure mit 100 ml 0,1 M Natriumacetat
(pH 5»6) und 120 ml Methanol versetzt, geschüttelt und nach nochmaliger Phasentrennung zu 10 g Natriumsulfat gegeben und
eine Stunde gerührt. Nach Filtration und Abziehen des Lösungsmittels
wird der Rückstand aus Methyläthylketon umkristallisiert. Die endgültige Reinigung erfolgt durch Chromatographie
an Kieselgel.
Man erhält 3,6 g (57 # bezogen auf Palmitoyl-2,2-dimethylpropandiol)
weißes analysenreines Palmitoyl-2,2-dimethylpropandiol( 1,3) -phosphoryläthanolamin.

berechnet: C 59,33 % H 10,39 # N 3,01 $>
P 6,65 1° gefunden: C 58,97 $> H 10,47 $>
N 3,07 $> P 6,49 %
Palmitoyl-2,2-dimethylpropandiol(i,3)-phosphoryl-N-methyläthanolamin
Eine Lösung von 5 g (10 mMol) des ß-Bromäthylesters von Beispiel
1 B. wird wie in Beispiel 1 C. beschrieben mit Methylamin umgesetzt, aufgearbeitet und chromatographisch gereinigt.
Man erhält 3,5 g (52 # bezogen auf 2,2-Dimethylpropandiol-(1,3)-monopalmitinsäureester)
analysenreines Produkt.

N 2,92 # P 6,46 % gefunden: C 59,85 $ H 10,58 # N 2,95 $ P 6,35 $>

Palmitoyl-2,2-dimethylpropandiol( 1,3)-phosphoryl-N,N-dimethyläthanolamin
Setzt man 5 g des ß-Bromäthylesters (10 mMol) von Beispiel
1 B wie in Beispiel 1 C beschrieben mit Dirnethylamin um,
so erhält man nach chromatographischer Reinigung 4,2 g (60 °/o
bezogen auf 2,2-Dimethylpropandiol(1,3)-palmitinsäureester)
analysenreines Produkt.

berechnet: C 60,82 % H 10,62 % N 2,84 $ P 6,27 #
gefunden: - N 2,51 f> P 5,91 %
Palmitoyl-2,2-dimethylpropandiol(1,3)-phosphoryl-cholin
Eine Lösung des ß-Bromäthylesters aus Beispiel 1 B. (5 g in
50 ml Chloroform (10 mMol)) wird in schon bekannter Weise mit Irimethylamin umgesetzt, aufgearbeitet und chromatographiert.
Die Ausbeute beträgt 4 g (57 $> bezogen auf 2,2-Dimethylpropandiol(1,3)-palmitinsäureester)
an analysenreinem leeithinanalogen Produkt.

Claims (8)
- PatentansprücheVerfahren zur Herstellung einer Verbindung der allge ieinen Formel I0 H2C-O-C-GnH2n+1R1-C-R2 (I)H9C-O-P-O-CH5-CH9-N - Xο-worin η eine ganze Zahl zwischen 9 und 25, X1, X2 und X, unabhängig voneinander je ein Wasserstoffatom oder eine Methylgruppe und R1 und R2 unabhängig voneinander je ein Wasserstoff atom oder eine Alkylgruppe, vorzugsweise mit 1 bis 6 Kohlenstoffatomen, bedeuten, dadurch gekennzeichnet, daß man gegebenenfalls in Stellung 2 Alkyl-substituiertes Propandiol-(1,3) mit einem geeigneten Acylierungsmittel in an sich bekannter Welse zum Monofettsäureester der allgemeinen Formel II0 H2C-O-S-CnH2n+1R1-C-R2 (II)H2C-OHin der n, R1 und R2 die oben angegebene Bedeutung besitzen, umsetzt und den Monoester mit ß-Bromäthylphosphorsäuredichlorid zur Reaktion bringt unter Bildung einer Verbindung5 09813/1058der Formel III(III)HoC-O-P-O-CH5-CH9-Br OHwelche anschließend mit Trimethylamin, Dimethylamin, Methylamin oder Ammoniak direkt aminiert wird.
- 2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, daß die Acylierung mit einem Säurechlorid oder Anhydrid in einer Mischung von Chloroform und Pyridin durchgeführt wird.
- 3. Verfahren nach Anspruch 1, dadurch gekennzeichnet, daß die Phosphorylierung in einem wasserfreien polaren organischen Lösungsmittel durchgeführt wird.
- 4. Verfahren nach Anspruch 3» dadurch gekennzeichnet, daß die Phosphorylierung in Gegenwart von überschüssigem Amin, bezogen auf Phosphorylierungsmittel, erfolgt.
- 5. Verfahren nach einem der vorhergehenden Ansprüche, dadurch gekennzeichnet, daß die weitere Umsetzung der Verbindung der allgemeinen Formel III in einem Gemisch organischer polarer Lösungsmittel, die gegebenenfalls Wasser enthalten, erfolgt.509813/ 1058
- 6. Verfahren nach Anspruch 5» dadurch gekennzeichnet, daß ein Gemisch von Chloroform und Acetonitril oder Nitromethan verwendet wird.
- 7. Verfahren nach Anspruch 6, dadurch gekennzeichnet, daß das Lösungsmit is elgemisch zusätzlich einen niedrigen Alkanol und Wasser enthält.
- 8. Verbindungen der allgemeinen Formel IVH2C-O-3-CmH2m+1(IV)worin m eine ganze Zahl von 9 bis 14 und von 16 bis 25, X^, X2 und X™ unabhängig voneinander je ein Wasserstoffatom oder eine Methylgruppe und R^ und R2 unabhängig voneinander je ein Wasserstoffatom oder eine Alkylgruppe, vorzugsweise mit 1 bis 6 Kohlenstoffatomen, bedeuten.509813/1058
