Die Messung des Desoxyribonukleinsäuregehaltes (DNS) von Zellkernen ist für viele Untersuchungen as
der Biologie und Medizin notwendig. Meist wird die Absorption von UV-Strahlung durch die DNS oder
die Absorption oder Fluoreszenz von sichtbarem Licht durch an die DNS angelagerte Farbstoffe
(Feulgen-Reaktion) mit geeigneten Cytophotometern 3» gemessen.
Dazu werden die Zellen auf der Oberfläche einer Glasplatte fixiert, entsprechend gefärbt, einzeln im
Mikroskop aufgesucht, eingestellt und die optischen Messungen durchgeführt. Dieser Meßvorgang erfordert
pro Zelle einige Minuten, so daß Messungen an größeren Zellpopulationen nicht durchgeführt worden
sind.
Einen wesentlichen Fortschritt brachten neuartige Durchflußverfahren, bei denen die Zellen als Suspension
durch eine fest eingestellte Meßkammer fließen, so daß dort an einigen tausend Zellen pro Sekunde
selbsttätig die optischen Messungen durchgeführt werden können. Hierfür werden vorzugsweise Fluoreszenzmethoden
verwendet.
Hierdurch ist es möglich geworden, die Verteilung der DNS-Menge in einer Zellpopulation mit geringem
Zeitaufwand und hoher statistischer Sicherheit zu bestimmen. Es ist möglich, daraus eine maligne
Entartung der Zellpopulation mit einer geänderten Prolieferationskinetik zu erkennen und so zahlreiche,
sehr wichtige Probleme der Krebsdiagnose und -therapie zu untersuchen.
Die zur spezifischen und quantitativen Färbung benutzte Feulgenreaktion besteht aus einer Hydrolyse
der DNS durch Salzsäure unter Bildung von Aldehydgruppen und einer Reaktion der Aldehydgruppen mit einem Schiffschen Reagenz, wässerigen Lösungen von Farbstoffen, die durch Schwefeldioxyd
entfärbt wurden. Als Farbstoffe werden vorzugsweise
Fuchsin für Absorptionsmessungen und Akridinfarbstoffe
für Fluoreszenz benutzt.
Die Hydrolyse ist dabei die für die DNS spezifische Reaktion, die aber nur schwer zu beherrschen
ist Geringe Abweichungen der Dauer, Temperatur und Säurekonzentratioa führen zu nicht auswertbaren
Ergebnissen. Insbesondere ist das Resultat der Hydrolyse von der Art und dem Alter der Zellen abhängig.
Verzichtet man auf die Hydrolyse und färbt die Zellen direkt mit einem Fluoreszenzfarbstoff, so
werden im allgemeinen außer der DNS auch die Ribonukleinsäuren und andere Zellinhaltssloffe gefärbt. Eine quantitative DNS-Bestimmung ist dann
nicht möglich. .
Die gestellte Aufgabe besteht also darm, durch ein geeignetes Verfahren die DNS der Zellen reproduzierbar
und isoliert anzufärben und die oben beschriebenen Nachteile der bekannten Verfahren zu
vermeiden.
Diese Aufgabe wird durch die im Anspruch 1 angegebene
Erfindung gelöst. Dabei wird die Tatsache ausgenutzt, daß die DNS der Zelle vorzugsweise im
Zellkern (mehr als 99 °/o) und die ebenfalls angefärbten
Zeilinhaltsstoffe wie Ribonukleinsäuren im Zellplasma konzentriert sind.
Das folgende Ausführungsbeispiel sowie die Figuren der Zeichnung sollen die Erfindung veranschaulichen:
Die Zellen werden 15 Minuten in einer Lösung von 0,2 g Pepsin in 100 ml 0,2% Salzsäure bei
37° C inkubiert. Die dadurch isolierten, aber in ihrer Färbbarkeit nicht beeinträchtigten Zellkerne werden
in physikalischer Kochsalzlösung gewaschen und in einer nukleinsäurespezifischen Farbstofflösung (Ethidiumbromid,
Akridinorange, Coriphosphin usw.) in bekannter Weise gefärbt.
Die Zeichnung zeigt ein Ergebnis der Fluoreszenzmessung an Epithelzellen aus der Scheide nach
Feulgenfärbung mit Akriflavin (a), nach Färbung mit Ethidiumbromid ohne (b) und mit (c) vorhergehender
Pepsinbehandlung. Man erkennt, daß nur das vorgeschlagene Verfahren ein präzises und für die
Krebsdiagnostik geeignetes Ergebnis mit scharfer Auflösung ergibt. Überdies ist das vorgeschlagene
Verfahren schneller (30 Minuten gegenüber 2 Stunden für Feulgenfärbung) und besser reproduzierbar.

Claims (2)
1. Verfahren zur Präparation von suspendierten Zellen für eine quantitative Bestimmung der
Desoxyribonukleinsäure mit einem Durchflußverfahren durch Anfärbung mit einem nukleinsäurespezifischen
Farbstoff, dadurch gekennzeichnet, daß die Zellen vor der Anfärbung erst mit einem die Zellmembran auflösenden
Reagenz behandelt werden, wodurch die Zellkerne isoliert werden, und daß anschließend nur
diese der Anfärbung unterzogen werden.
2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, daß als die Zellmembran auflösendes
Reagenz Pepsin in salzsaurer Lösung verwendet wird.
