Phosphor- und Thiophosphorsäureester substituierter Hydantoine, Verfahren zu deren Herstellung sowie deren
Verwendung
Die Erfindung betrifft neuartige Phosphor- und Thiophosphorsäureester
von Hydantoinen. Weiterhin betrifft die Erfindung ein Verfahren zur Herstellung von Phoephor- und Thiophosphorsäureesterderivaten
von sowohl in 1- als auch in 5~Stellung
substituierten Hydantoinen. Schließlich ist Gegenstand der
Erfindung die Verwendung der neuartigen Verbindungen als
Pestizide, insbesondere als Insektizide.

Eine Anzahl von Phosphor- und Thiophosphorsäureesterderivaten
von Hydantoinen ist bereits bekannt. So beschreibt beispielsweise die USA-Patentschrift 2 928 8Ί1 Verbindungen der folgenden

In der R ein niederer Alkylrest mit 1 bis H Kohlenstoffatomen
ist, X einmal Sauerstoff und einmal Sauerstoff oder Schwefel bedeutet und R1 ein Wasserstoffatom oder ein niederer Alkylrest mit 1 bis 4 Kohlenstoffatomen ist. Von diesen Verbindungen wird behauptet, daß sie als Insektizide brauchbar sind.Die
Ver-bindungen unterscheiden sich von denen der vorliegenden Erfindung durch die Stellung des Thiopttosphorsäurerestea
relativ zum Hydantoinring, nänlich durch die Substitution
in der !-Stellung statt der 3-Stellung; es wird weiter unten
gezeigt werden, daß eine Substitution ara Stickstoffatom 3
bei den vorbekannten Verbindungen tatsachlich zu einem Verlust der pestisiden Wirksamkeit dieser Verbindungen führt,
obwohl die Verbindungen dadurch strukturell denen der vorliegenden Erfindung noch näher kommen.
Aus der ebenfalls vorveröffentlichten französischen Patentschrift 1 093 728 ist die Synthese von insektizid wirksamen
Thiophosphorsäureester!! des Hydantoins bekannt. Die Verbindungen dieser Vorveröffentlichung entsprechen der folgenden allgemeinen Formel:

Im Rahmen der vorliegenden Untersuchung konnte gezeigt werden, daß bei diesen Verbindungen eine Substitution in
1-Stellung zu einer deutlichen Steigerung der pestiziden
Wirksamkeit führt. Darüber hinaus hat sich gezeigt, daft die pestizide Wirksamkeit gesteigert werden kann, wenn man am
Kohlenstoffatom 5 aliphatische Reste anstelle der Phenylreste
einführt.
Aus der kürzlich veröffentlichten holländischen Patentanmeldung
6 900 039 kann ebenfalls entnommen werden, dan 0,0-Dialkylphosphordithio- und -monothioester, welche den Hydantoinkem
enthalten, als Insektizide brauchbar sind. Diese Verbindungen entsprechen der allgemeinen Formel

in welcher T ein Sauerstoff- oder Schwefelatom ist, R* einen
CH,- oder C2Hp-ReSt und R2, R1 R und R ein Wasserstoffatom
oder Methylrest bedeuten, wobei mindestens einer der Rest· R
und R ein Methylrest sein muft, wAhrend die anderen Wasser·
f. « Stoffatome sind und wobei entweder R' od»r H ein Wasserntoff-
atom darstellen.

Diese Verbindungen sind für Säugetiere äußerst toxisch. Ee
wurde jedoch überraschend entdeckt, daß bei sorgfältiger Wahl
der Substituenten Ir und Rf,insbesondere wenn R* ein organischer Rest mit mindestens 2 Kohlenstoffatomen ißt, das
Toxizitätsniveau merklich gesenkt werden kann. Wenn darüber hinaus die Alkpxyreste in der vorstehenden Formel durch C1
bis Cg Alkylthio- und Alkylreste ersetzt werden, wird eine
weitere beachtliche Verminderung der ToxizitSt beobachtet.
Der Erfindung liegt demgemäss die Aufgabe zugrunde, neue
Thiophosphor- und Phosphorsäurederivate von Hydantoinen vorzuschlagen, welche hervorragende pestizide Wirksamkeit und
gleichzeitig eine geringe Toxizität für Säugetiere, verglichen
mit den bislang bekannten Verbindungen, besitzen.
Gegenstand der Erfindung sind somit Verbindungen der folgenden allgemeinen Formeln

In der vorstehenden Formel kann R1 ein beliebiger geeigneter
organischer Rest sein. Darunter werden einwertige,aliphatisehe, cycloaliphatische, aromatische und heterocyclische Rest sowie
deren substituierte Abkömmlinge verstanden. Die organischen Reste können durch beliebige Substituenten substituiert sein, ^
welche die pestizide Wirksamkeit der Verbindungen nioht stören, z.B. Halogenatome, wie Chlor oder Brom, Nitrogruppen,
Alkoxyreste, wie beispielsweise Methoxy-, Xthoxy- und ähnliehe
Rest·, Carboalkoxyreste, wie Carbomethoxy- oder Carboätnoxyreate,
Dialkylaminoreste, wie Dimethylamine)-, Diethylamino- oder
Hethyltthylaminoreete, Thloalkylreste, wie Methylthlo- oder
Xthylthioreste, Aoylreste, wie Aoetyl-, Propionyl- oder Bensoylreste und andere.

Wenn in der obigen Formel die organischen Reste aliphatioche
Reste sind, dann kann es sich beispielsweise um Alkyl", Alkinyl-«
Aralkyl- und/oder Aralkenylreate sov/ie C, bis C- Cycloalkyl-
oder C^ bis C« Gycioalkylalkylreste handeln.
Ein beliebiger geeigneter Alkylrest kann beispielsweise der
organische Rest oein, z.B. ein Methyl-, Xthyl-, n-Propyl-,
Isopropyl-, n-Bufcyl-, Isobutyl-; see.-Butyl-, tert.-Butyl-,
n-Amyl- oder ein isomerer Rest, wie z.B. ein 1-Methylbutylrest,
2-Methylbutyl-, 1,1-Dimethylpropyl-, 1,2-Dimethylpropyl-,
2,2-Dimethylpropyl-, I,l-Dime1:hylpropyl-, 1,2-Dimethylpropyl-,
2,2-Dimethylpropyl-, 1-Äthylprcpylrast sowie die entsprechenden
gerad- und verzwedgtkettigen Isomeren des Hexylrestes.
Geeignete Alkenylreste sind beispielsweise der Kthenyl-,
1-Propenyl-, 2-Propenyl-, Isopropenyl-, 1-Butenyl-, 2-Butenyl-i
3-Butenylrest sowie die entsprechenden versweigtkettigen Isomeren
wie z.B. der 1-Isobutenyl-, 2-Ieobutenyl-, 2-sec-Butenyl-,
3-sec.-Butenylrest einschließlich des l-Methylen-2-propenylrestes,
der 1-Pentenyl-, 2-Pentenyl-, 3-Pentenyl- und ^-Pentenylrest
einschließlich der entsprechenden versweigtkettigen Isomeren^
der 1-Hexenyl-, 2-Hexenyl-, 3-Hexenyl-, 4-HeJcenyl- und 5-Hexenylrest
sowie die entsprechenden veruweigtkettigen Isomeren» b.B.

Auch geeignete Aralkylreste können den organischen Rest bilden,
z.B. ein Benzyl-,oC-Phenyläthyl-, ß-Phenyläthyl- oder |

Geeignete Aralkenylreste als organischer Rest sind beispielsweise dercC-Phenyläthenyl-, ß-Phenyläthenyl-, oL-Phenyl-1-propenyl-,
ß-Phenyl-1-propenyl-, i'-Phenyl-l-propenyl-,
o6-Phenyl-2-pΓopenyl-, ß-Phenyl-2-propenyl-, y-Phenyl-2-propenyl-
und der ß-Phenylisopropenylreet.
Als geeignete Cyeloalkylreste kommen für den organischen Rest
beispielsweise der Cyclopropyl-, Cyclobutyl-, Cycloamyl-, "
Cyclohexyl-, Cyclopropylmethyl-, Cyclopropyläthyl- und
Cyclopentylmethylreet infrage.
Geeignete Arylreste sind beispielsweise der Phenyl-,
<^-Naphthyl-, ß-Naphthyl- oder substituierte Phenylreste, wie
die 2,3- oder 4-Chlorphenylieomeren, die Dichlorphenylreste
und andere.

Als geeignete Allcarylreste sind beispielsweise der ο-ToIy 1-,
m~Tolyl- und p-Tolylreat zu nennen.
Die Alkylreete in den Stellungen 1 und 5 können darüber hinaus
miteinander verbunden sein, so daß ein 5- oder ßgliedricer
cyclischer Ring entsteht, ζ.B, ein 1,^-Teti'amethylen- oder
i,5-Pentaraethylenhydantoin.

Die unter die vorliegende Erfindung fallenden Verbindungen lassen sich durch Umsetzung eines in 1-Stellung substituierten
Hydantoins oder eines in 1*- und ^-Stellung disubstituierten
Hydantoins mit Formaldehyd in Gegenwart oder Abwesenheit eines Katalysators, wie Salzsäure, herstellen. Diese Hydroxymethylderivate des Hydantoins werden anschließend mit einem HaIogenierungsmittel, wie Thionylchlorid, Phosphortrichlorid oder
Phosphortribromid, umgesetzt, wobei die 3-Halogenmethylderivate
des Hydantoins erhalten werden.
Anschließend wird das Halogenderivat des substituierten Hydantoins mit einer Verbindung umgesetzt, welche das
Halogenatom in der nachstehend schematisch dargestellten Weis· zu ersetzen vermag.

beschrieben sind.
Die vorstehend wiedergegebenen Reaktionsschritte sind verallgemeinert und es lassen sich auch andere Aueganßsverbindungen und Umsetzungen anwenden, um die gleichen Endprodukte

In einen mit RUckflußkUhler versehenen Reaktionskolben wurden
42,1 g !-(p-Chlorphenyl)-hydantion, 60 ml 36iiger Formaldehyd

und 250 ml Wasser gegeben. Das Reaktionsgemiseh wurde 3 Stunden*
lang auf Rückflußtemperatur erhitzt, auf 2O0G abgekühlt und
mit 500 ml Wasser verdünnt, worauf sich das Produkt als Peststoff absetzte. Das Produkt wurde durch Filtration abgetrennt
und getrocknet, wobei 31,7 g l-Cp-GhIorphenyl)-3~hydroxymethylhydantoin,
Pp. 160 bis 1650C, erhalten wurden.

Herstellung von 3-Hydroxymethyl-i-phenylhydantoin
In einen mit Rückflußkühler versehenen Reaktionskolben wurden 100,6 g i-Pheny!hydantoin, 80 ml 36£iger wäßriger Formaldehyd,
300 ml Wasser und 600 ml Methanol gegeben. Das Reaktionsgemisch wurde 1,5 Stunden lang auf Rückflußtemperatur erhitst,
auf 200C abgekühlt und mit 700 ml Wasser verdünnt, wodweh

erhalten wurden.

In einen mit Rückflu&kOhler versehenen Reaktionskolben wurden
50,0 g 1-Methy!hydantoin, 50 ml 36?iger wäßriger Formaldehyd
und 450 ml Wasser gegeben. Das Reaktionsgemiseh vurde 2 Stunden
lang auf Ruckflufitemperatur erhitzt, auf Zimmertemperatur
abgekühlt und zur Trockne eingeengt.Der Rückstand wurde dreimal mit 250 ml Chloroform extrahiert. Die vereinigten Chloroformextrakte wurden aber Natriumsulfat getrocknet, abfiltriert
und nach Abziehen des Chloroforms wurden 35 g 3-Hydroxymethyl-1-methylhydantoin, Pp. 83 bis 850C, erhalten.

Ein Gemisch von 9,5 g S-Ammonium-O.O-diäthylphoephorthioat,
15,2 ε 3-Bromme&hyl-i-p-chlorphenyXhydantoin und 350 ml
Acetonitril wurde in einem mit Rückflusskühler ausgestatteten
Reakticnskolben 1 Stunde lanp; im Rückfluß erhitzt. Das ausfallende
Ainmoniurachlorid wurde abfiltriert und das Filtrat
wurde auf Eis pepoesen, worauf das erhaltene Gemisch mit
500 ml Benzol extrahiert, wurde. Der Benzolextrakt wurde an**
nchließend mit 1Ou ml Wasser sowie 100 ml einer gesättigten
Natriumbicarbonatlößung gewaschen und über wasserfreiem
Natriumsulfat getrocknet« Der Benzolextrakt wurde schließlich
filtriert und nach Abtrennen des Benzols wurden 17 g des
Produktes 5.n Form einer weißen Festsubstanz vom Fp.86 bie ■
880C erhalten.

Wie bereits oben ausgeführt, sind die Hydantointhiophosphoreäureester
der Erfindung wertvolle Pestizide, Insbesondere Insektizide. Bei Verwendung als Insektizide werden die
biologisch aktiven HydantoinfchiophosphorsSureester vorzugsweise
mit geeigneten inerten Zusätzen als Trägermittel vermischt, wie sie dem Faohstatm geläufig sind.
Allgemeine Verfahrensweise für die biologische Untersuchung der Verbindungen.
In den nachfolgenden Beispielen wurden die neuen Hydantointhiophosphatester im Gewächshaus sowie in Laboratorium untersuoht, um Ihr· biologische Aktivität su bestimmen.
Die Testverbindungen wurden in Form wäßriger Emulsionen
eingesetzt. Diese Emulsionen wurden duroh Auflösen der Verbindungen in Aceton und Dispergieren in destilliertem Wasser
■uaamnen mit Triton X-IOO9 einem Alkylarylpolyätheralkohol,
hergestellt duroh Umsetsung von Isoootylphenol mit Xthylenoxyd, erhalten, to da* sprühflhige Emulsionen gebildet wurden,
dl· die gewünschte Xonsentratlon an Testverbindung enthielten.
Diese Kmilaionen wurden anschließend den nachfolgend beschriebenen Standard-Laboratoriumstests unterworfen.

Mexikanischer Bohnenkäfer: Bohnenblätter wurden in die Emulsion
der su untersuchenden Verbindung eingetaucht und anschließend
trocknen gelassen. Die einseinen so behandelten Blätter wurden in Petrieohalen gelegt und Larven von fünf mexikanischen
Bohnenkäfern wurden in jede der beiden jeweils identischen Schalen eingesetzt.
Milben (Kontakt): In Töpfe eingesetzte Bohnenpflanzen, die von der zweifach gepunkteten Spinnreilbe befallen waren, wurden
auf einen Drehtisch gestellt und mit einer Zusammensetzung der zu untersuchenden Verbindung besprüht. Die Pflanzen wurden
7 Tage lang stehen gelassen und die Wirksamkeit gegenüber den Spinnmilben wurde ansehlieftend bestimmt.
Milben (systemisch): Bohnenpflanzen wurden behandelt, indem
20 mil der Testmisohung auf den Boden aufgebracht wurden. Die
Milben wurden naoh 2*. Stunden auf die Pflanzen Überführt. ^
Die Pflanzen wurden 7 weitere Tage lang stehen gelassen und
das Ausmaß der Milbenverniehtung wurde bewertet.
Blattläuse (Kontakt): Zn Töpfe eingesetzte Nasturtiuepflanzen,
die von Bohnenblattlftusen befallen waren, wurden auf einen Drehtisoh gestellt und mit einer Zusammensetzung der su unter*·
suchenden Verbindung besprüht. Die Pflanzen wurden 2 Tage

lang stehen gelassen, worauf die Wirksamkeit gegenüber Blattläusen bestimmt wurde.
Blattläuse (syatemisch): Nasturtiumpflansen wurden behandelt»
indem 20 ml der zn untersuchenden Mischung auf den Boden aufgebracht wurden. Die Insekten wurden nach 24 Stunden auf die
Pflanzen überführt. Anschließend wurden die Pflanzen 48 weitere
Stunden lang stehengelassen, worauf die Wirkung .bestimmt wurde.
Hausfliege: Eingesperrte Hausfliegen wurden mit der Zusammen-*
setsung besprüht, nach 2 Tagen wurde die Wirksamkeit ermittelt.
Baumwollkapaelkäfer: 5 erwachsene Baumwollkapselkäfer in einem
Drahtnetskäfig wurden mit der geeigneten Konzentration der Testzusammensetzung besprüht. Die Baumwollkapselkäfer wurden
mit einer Sucroselösung auf Filterpapier versehen. Die Käfige
wurden 24 Stunden lang bei einer Temperatur von ungefähr 21°C gehalten, worauf die Sterblichkeit nach 24 Stunden bestimmt wurde.
Erbsenblattläuse (Kontakt): Erbsenpflanzen wurden mit der
geeigneten Konzentration an der Testzusammensetzung besprüht,
worauf die Pflanzen mit 20 bis 30 erwachsenen Erbsenblattläusen
besetzt wurden. Die Pflanzen wurden 24 Stunden lang bei 210C

stehen gelassen, worauf die Wirksamkeit bestimmt wurde.
Erbsenblattläuae (systemisch): Erbsenpflanzen wurden behandelt,
indem 20 ml der zu untersuchenden Zusammensetzung? auf den Boden aufgebracht wurden. Die Erbsenblattläuse wurden anschließend
auf die Pflanzen überfahrt. Die Wirksamkeit gegenüber den
Erbsenblattläusen wurde nach 48 Stunden bestimmt.
Die Ergebnisse der vorstehenden Bestimmung sind in der nachfolgenden Tabelle IV zusammengefaßt. Einige der Verbindungen
wurden ferner gegenüber bestimmten anderen Coleopteren untersucht, z.B. gegenüber Mehlkäfern, Spinnenkflfern sowie
erwachsenen mexikanischen Bohnenkäfern. Weiterhin wurde die Wirksamkeit gegenüber deutschen Küchenschaben ermittelt.
Weitere Untersuchungen dienten der Bestimmung der oviciden Wirksamkeit, wobei gefunden wurde, daß die Verbindungen aueh

Zur Bestimmung der Toxizität der 1-substituierten Hydantoine
für Warmblüter wurden einige der i-AIkylhydantoinphoaphorsäureeeter auf ihre akute orale und dermale Toxizität untersucht. Die Ergebnisse sind in Tabelle VIX zusammengefaßt und
zeigen deutlich, daß ein Ersatz das Methylrestes in der
1-Stellung durch höhere Alkylreste sowie substituierte Alkylraste zu einer Verminderung der Toxizität für Säugetiere führt=
