Description
3-Oxo-benzisoxazolmethyl(thiono)thiolphosphor(phosphon)-säureester,
Verfahren zu ihrer Herstellung sowie ihre Verwendung als Insektizide und Acarizide
Die vorliegende Erfindung betrifft neue 3-0xo-benzisoxazolmethyl(thiono)thiolphosphor(phosphon)-säureester,
welche insektizide und akarizide Eigenschaften haben sowie ein Verfahren zu ihrer Herstellung.
Es ist bereits bekannt, daß Phthalimido-methylthionothiolphosphorsäureester,
wie z.B. der 0,0-Dimethyl-S-phthalimidomethyl-dithiophosphorsäureester,
insektizide Eigenschaften aufweisen (vgl. Deutsche Patentschrift 930.446).
Es wurde nun gefunden, daß die neuen 3-0xo-benzisoxazolmethyl(thiono)-thiolphosphor(phosphon)-säureester
der Formel (I)

in welcher R und R. für gleiche oder verschiedene, geradkettige
oder verzv/eigte Alkylreste mit 1 bis 6 Kohlenstoff -

atomen atehen, R1 außerdem einen Alkoxyrest mit 1 bis 6 Kohlenstoffatomen
bedeutet, Y und Y1 für Wasserstoff, Halogenatome,
Nitro- oder Alkylgruppen mit 1 bis 6 Kohlenstoffatomen stehen und X ein Sauerstoff- oder Schwefelatom ist, starke
insektizide und akarizide neben tickiziden Eigenschaften aufweisen.
Weiterhin wurde gefunden, daß die neuen 3-0xo-benzisoxazolmethyl(thiono)-thiolphosphor(phosphon)-säureester
der Konstitution (I) erhalten werden, wenn man (Thiono)Thiolphosphor(phosphon)-säureester
der Formel (II)

in welchen Formeln R, R1, X, Y und Y1 die oben angegebene
Bedeutung haben und Hai ein Halogen-, vorzugsweise Chloroder Bromatom bedeutet, umsetzt.
Überraschenderweise zeichnen sich die erfindungsgemäßen
3-0xo-benzisoxazolmethyl(thiono)-thiolphosphor(phosphon)-säureester
durch eine erheblich höhere insektizide und akarizide Wirkung aus als die bekannten Phthalimidomethyl-

thionothiolphosphorsäureecter analoger Konstitution und
gleicher Wirkungsrichtung. Die erfindungsgemäßen Stoffe
stellen somit eine echte Bereicherung der Technik dar.
Verwendet man beispielsweise 3-Oxo-chlormethylbenzisoxazol
und das Ammoniumsalz des Ο,Ο-Diathylthionothiolphosphorsäureesters
als Ausgangsmaterialien, so kann der Reaktionsablauf durch das folgende Formelschema wiedergegeben werden:

Die zu verwendenden Ausgangsstoffe sind durch die Formeln (II) und (III) eindeutig allgemein definiert. Vorzugsweise stehen
R und R1 für geradkettige oder verzweigte Alkylreste mit
1 bis 4 Kohlenstoffatomen, wie z.B. für Methyl, Äthyl, n- oder iso-Propyl, n-, see-, tert.- oder iso-Butyl, R1 bedeutet
außerdem bevorzugt einen Alkoxyrest mit 1 bis 4 Kohlenstoffatomen, wie z.B. Methoxy, Äthoxy, iso-Propoxy, n-Propoxy,
n-, see-, tert.- oder iso-Butoxy, Y und Y1 bedeuten
bevorzugt Wasserstoff, Chlor, Brom und/oder Alkyl mit 1 bis 4 Kohlenstoffatomen.
Als Beispiele für verfahrensgemäß einzusetzende (Thiono)-Thiolphosphor(phosphon)-säureester
seien im einzelnen genannt: Die Alkali-, Erdalkali- oder Ammoniumsalze von 0,0-Dimethyl-,
0,0-Diäthyl-, O,O-Dipropyl-, O^-Diisopropyl-, 0,0-Dibutyl-,

deren Thionoanalogen, ferner die entsprechenden Salze von O-Methyl-methan-, O-Methyl-äthan-, 0-Methyl-propan-, 0-Methylisopropan-,
O-Methyl-butan-, O-Äthyl-inethan-, O-Äthyl-äthan-,
0-Äthyl-propan-, 0-Äthyl-butan-, O-iso-Propyl-methan-τ 0-iso-Propyl-äthan-,
O-Butyl-propan- und O-Butyl-tert.-butanthiolphosphonsäureester
und deren Thionoanalogen.
Die ale Ausgangsstoffe benötigten Salze der (Thiono)-Thiolphosphor(phosphon)-säureeeter
der Konstitution (III) sind aus der Literatur bekannt und ebenso wie die 3-0xobenzisoxazolderivate
der Konstitution (II) auch in technischem Maße zugänglich.
Das Herstellungsverfahren wird bevorzugt unter Mitverwendung geeigneter Lö'sungs- bzw. Verdünnungsmittel durchgeführt.
Als solche kommen praktisch alle inerten organischen SoI-ventien infrage. Hierzu gehören vor allem aliphatische und
aromatische, gegebenenfalls chlorierte Kohlenwasserstoffe, wie Benzol, Toluol, Xylol, Benzin, Methylenchlorid^ Chloroform,
Tetrachlorkohlenstoff, Chlorbenzol, Äther, wie z.B. Diäthyl- und Dibutyläther, Dioxan, ferner Ketone, beispielsweise
Aceton, Methyläthyl-, Methyl-iso-propyl- und Methylisobutylketon,
außerdem Nitrile, wie Acetonitril und Propionitril, außerdem Alkohole, wie Äthanol oder Propanol.
Als Säureakzeptoren können alle üblichen Säurebindemittel Verwendung finden. Besondere bewährt haben sich Alkalicarbonate
und -alkoholate, wie Natrium- und Kaliumcarbonat, -methylat bzw. -athylat, ferner aliphatische, aromatische
oder heterocyclische Amine, beispielsweise Triethylamin, Dimethylamin, Dimethylanilin, Dimethylbenzylamin und Pyridin.

Die Reaktionstemperatur kann innerhalb eines größeren Bereiches variiert werden. Im allgemeinen arbeitet man
zwischen 0 und 1000C, vorzugsweise bei 30 bis 4O0O.
Die Umsetzung wird im allgemeinen bei Normaldruck durchgeführt
.
Zur Durchführung des Verfahrens setzt man die Ausgangsstoffe meist in äquimolarem Verhältnis ein. Ein Überschuss der
einen oder anderen Reaktionskomponente bringt keine wesentlichen
Vorteile. Die Umsetzung wird bevorzugt in Gegenwart eines der oben genannten Lösungsmittel sowie in Anwesenheit
eines Säureakzeptors bei den angegebenen Temperaturen vorgenommen und das Reaktionsgemisch nach ein- bis mehrstündigem
Rühren - gegebenenfalls unter Erwärmen - in üblicher Weise aufgearbeitet.
Die erfindungsgemäßen Stoffe fallen entweder in Form farbloser bis schwach gelb gefärbter, viskoser, wasserunlöslicher
Öle an, die sich nicht unzersetzt destillieren lassen, jedoch durch sogenanntes "Andestillieren", d.h.
durch längeres Erhitzen unter vermindertem Druck auf mäßig erhöhte Temperaturen von den letzten flüchtigen Anteilen
befreit und auf diese Weise gereinigt werden können. Zu ihrer Charakterisierung dient vor allem der Brechungsindex,
oder sie stellen kristalline Substanzen dar; dann werden sie durch ihren Schmelzpunkt charakterisiert.
Wie bereits mehrfach erwähnt, zeichnen sich die neuen 3-0xobenzisoxazolmethyl-(thiono)-thiolphosphor(phosphon)-säureester
durch eine hervorragende insektizide und akarizide Wirksamkeit

/gegenüber Pflanzen-» Hygiene- und Vorratsschädlingen aus.
Sie besitzen dabei sowohl eine gute Wirkung gegen saugende als auch fressende Insekten und Milben (Acarina). Gleichzeitig
weisen sie eine geringe Phytotoxizität und z.T. auch tickizide Eigenschaften nuf.
Aua diesen Gründen werden die erfindungsgemäßen Verbindungen
mit Erfolg als Schädlingsbekämpfungsmittel im Pflanzen- und Vorratsschutz sowie auf dem Hygiene- und
Veterinärsektor eingesetzt.

Zu den saugenden Insekten gehören im wesentlichen Blattläuse (Aphidae) wie die grüne Pfirsichblattlaus (Myzus persicae),
die schwarze Bohnen- (Doralis fabae), Hafer- (Rhopalosiphum padi), Erbsen- (Macrosiphum pisi) und Kartofellaus (Macrosiphum
solanifolii), ferner die Johannisbeergallen- (Cryptomyzus korschelti), mehlige Apfel- (Sappaphis mali), mehlige Pflaumen-(Hyalopterus
arundinis) und schwarze Kirschenblattlaus (Myzus cerasi), außerdem Schild- und Schmierläuse (Coccina), z.B. die
Efeuschild- (Aspidiotus hederae) und Napfschildlaus (Lecanium
hesperidum) sowie die Schmierlaus (Pseudococcus maritimus); Blasenfüße (Thysanoptera) wie Hercinothrips fermoralis und Wanzen,
beispielsweise die Rüben- (Piesma quadrata), Baumwoll-(Dysdercus intermedius), Bett- (Cimex lectularius), Raub- (Rhodnius
prolixus) und Chagaswanze (Triatoma infestans), ferner Zikaden, wie Euscells bilobatus und Nephotettix bipunctatus.
Bei den beißenden Insekten wären vor allem zu nennen Schmetterlingsraupen
(Lepidoptera) wie die Kohlschabe (Plutella maculipennis),
der Schwammapinner (Lymantria dispar), Goldafter (Euproctis chrysorrhoea) und Ringelspinner (Malacosoma neustria),
weiterhin die Kohl- (Marnestra brassicae) und die Saateule
(Agrotis segetum), der große Kohlweißling (Pieris brassicae), kleine Frostspanner (Cheimatobia brumata), Eichenwickler
(Tortrix viridana), der Heer- (laphygma frugiperda) und aegyptische
Baumwollwurm (Prodenia litura), ferner die Gespinst-(Hyponomeuta padella), Mehl- (Ephestia kühniella) und große
Wachsmotte (Galleria meHonella),

Weiterhin zählen zu den beißenden Insekten Käfer (Coleoptera) z.B. Korn- (Sitophilus granarius = Calandra granaria),
Kartoffel- (Leptinotarsa decemlineata), Ampfer- (Gastrophysa viridula), Meerrettichblatt- (Phaedon cochleariae), Rapsglanz-(Meligethes
aeneus), Himbeer- (Byturus tomentosua), Speisebohnen-(Bruchidius
= Aeanthoscelides obtectus), Speck- (Dermestes frisch!), Khapra- (Trogoderma granarium), rotbrauner Reismehl-(Tribolium
castaneum), Mais- (Calandra oder Sitophilus zeamais),
Brot- (Stegobium paniceum), gemeiner Mehl- (Tenebrio molitor) und Getreideplattkäfer (Oryzaephilus surlnamensis), aber auch

melanogaster), Mittelmeerfrucht- (Ceratitis capitata), Stuben- (Musca domestica), kleine Stuben- (Fannia ca'nicularis),
Glanz- (Phormia aegina) und Schmeißfliege (Calliphora erythrocephala) sowie den Wadenstecher (Stomoxys calcitrans);
ferner Mücken, z.B. Stechmücken wie die Gelbfieber- (Aedes aegypti), Haus- (Culex pipiens) und Malariamücke (Anopheles
stephensi).

Zu den Milben (Acari) zählen besonders die Spinnmilben (Tetranychidae) wie die Bohnen- (Tetranychus telarius =
Tetranychus althaeae oder Tetranychus urticae) und die Obstbaumspinnmilbe (Paratetranychus pilosus = Panonychus ulmi),
Gallmilben, z.B. die Johannisbeergallmilbe (Eriophyes ribis) und Tarsonemiden beispielsweise die Triebspitzenmilbe
(Hemitarsonemus latus) und Cyclamenmilbe (Tarsonemus pallidus)}
schließlich Zecken wie die lederzecke (Ornithodorus moubata).
Bei der Anwendung gegen Hygiene- und Vorratsschädlinge, besonders Fliegen und Mücken, zeichnen sich die Verfahrensprodukte außerdem durch eine hervorragende Residualwirkung
auf Holz und Ton sowie eine gute Alkalistabilität auf gekalkten Unterlagen aus.

Je nach ihrem Anwendungszweck können die neuen Wirkstoffe in die
üblichen Formulierungen übergeführt werden, wie Lösungen, Emulsionen,'
Suspensionen, Pulver, Pasten und Granulate. Diese werden in bekannter V/eise hergestellt, z.B. durch Vermischen der Wirkstoffe
mit Streckmitteln, d.h. flüssigen Lösungsmitteln und/ oder Trägerstoffen gegebenenfalls unter Verwendung von oberflächenaktiven
Mitteln also Emulgier- und/oder Dispergiermitteln, wobei 25.B. im Falle der Benutzung von Wasser als Streckmittel
gegebenenfalls organische Lösungsmittel als Hilfslößungsraittel
verwendet werden können. Als flüssige Lösungsmittel kommen im wesentlichen infrage: Aromaten (z.B. Xylol,
Benzol), chlorierte Aromaten (z.B. Chlorbenzole), Paraffine (z.B. Erdölfraktionen), Alkohole ("z.B. Methanol, Butanol),
stark polare Lösungsmittel wie Dimethylformamid und Dimethylsulfoxyd sowie V/asser; als feste Trägerstoffe: natürliche
Gesteins&ehle (z.B. Kaoline, Tonerden, Talkum, Kreide) und
synthetische Gesteinsmehle (z.B. hochdisperse Kieselsäure, Silikate); als Emulgiermittel: nichtionogene und anionische
Emulgatoren wie Polyoxyäthylen-Fettsäure-Ester, Polyoxyäthylen-Fettalkohol-Äther,
z.B. Alkylarylpolyglykoläther, Alkylsulfonate und Arylsulfonate; als Dispergiermittel: z.B. Lignin,
Sulfitablaugen und Methylcellulose.
Die erfindungsgemäßen Wirkstoffe können in den Formulierungen in Mischung mit anderen bekannten Wirkstoffen vorliegen.

Die Formulierungen enthalten im allgemeinen zwischen 0,1 und
95 Gewichtsprozent Wirkstoff, vorzugsweise zwischen 0,5 und 90 %.
Die Wirkstoffe können als solche, in Form ihrer Formulierungen
oder in den daraus bereiteten Anwendungsformen, wie gebrauchsfertige
Lösungen, emulgierbare Konzentrate, Emulsionen, Suspensionen, Spritzpulver, Pasten, lösliche Pulver, iJtäubmittel
und Granulate angewendet werden. Die Anwendung geschieht in üblicher Weise, z.B. durch Verspritzen, Versprühen, Vernebeln, Verstäuben, Verstreuen, Verräuchern, Vergasen, Gießen,
Beizen oder Inkrustieren.
Die Wirkstoffkonzentrationen in den anwendungsfertigen Zubereitungen
können in größeren Bereichen variiert werden. Im allgemeinen liegen sie zwischen 0,0001 und 10 96, vorzugsweise
zwischen 0,01 und 1 %.
Die Wirkstoffe können auch mit gutem Erfolg im Ultra-Low-Volume-Verfahren
(ULV) verwendet werden, wo es möglich ist, !-Ormulierungen
bis zu 95 % oder sogar den 100%igen Wirkstoff allein auszubringen.

Nach den angegebenen Zeiten wird der Abtötungsgrad in %
bestimmt. Dabei bedeutet 100 ^, daß alle Raupen getötet
wurden, während 0 # angibt, daß keine Raupen getötet wurden.

der gemeinen Spinnmilbe (Tetranychus urticae) befallen.
Nach den angegebenen Zeiten wird die Wirksamkeit der V/irkstoffzubereitung
bestimmt, indem man die toten Tiere auszählt. Der 80 erhaltene Abtötungsgrad wird in $>
angegeben. 100 f^ bedeutet, dass alle Spinnmilben abgetötet wurden, 0 fo bedeutet, dass
keine Spinnmilben abgetötet wurden.

vermischt man 30 Gewichtsteile der betreffenden aktiven Substanz mit der angegebenen Menge Lösungsmittel, das den
oben genannten Anteil Emulgator enthält und verdünnt das so erhaltene Konzentrat mit Wasser auf die gewünschte
Konzentration.
Etwa 20 Fliegenlarven (Lucilia cuprina) werden in ein Teströhrchen gebracht, welches ca. 2 cm Pferdemuskulatur
enthält. Auf dieses Pferdefleisch werden 0,5 ml der Wirkstoff zubereitung gebracht. Nach 24 Stunden wird der Abtötungsgrad
in 'fo bestimmt. Dabei bedeuten 100 fa, daß alle,
und 0 fo, daß keine Larven abgetötet worden sind.
Die erhaltenen Versuchsergebnisse gehen aus Tabelle 5 hervor:

Beispiel F; Zeckentest
Lösungsmittel: 35 Gewichtsteile Äthylenglykolmonomethyläther
35 Gewichtsteile Nonylphenolpolykoläther
Zwecks Herstellung einer geeigneten Formulierung vermischt man drei Gewichtsteile Wirkstoff mit sieben Teilen des
oben angegebenen Lösungsmittel-Emulgator-Gemisches und verdünnt das so erhaltene Emulsionskonzentrat mit Wasser auf
die jeweils gewünschte Konzentration.
In diese Wirkstoffzubereitungen werden adulte, vollgesogene
Zeckenweibchen der Arten Boophilus microplus (sensibel bzw. resistent) eine Minute lang getaucht. Nach dem Tauchen von
je 10 weiblichen Exemplaren der verschiedenen Zeckenarten überführt man diese in Petrischalen, deren Boden mit einer
entsprechend großen Filterscheibe belegt ist.
Nach 10 Tagen wird die Wirksamkeit der Wirkstoffzubereitung
bestimmt durch Ermittlung der Hemmung der Eiablage gegenüber unbehandelten Kontrollzecken. Die Wirkung drückt man
in Prozent aus, wobei 100 $ bedeutet, daß keine Eier mehr
abgelegt wurden und 0 % besagt, daß die Zecken Eier in normaler Menge ablegten.
Untersuchter Wirkstoff, geprüfte Konzentration, getestete Parasiten und erhaltene Befunde gehen aus der folgenden
Tabelle 5 hervor:

Die benötigten Ausgangsprodukte können beispielsweise wie

329 g (2,21 Mol) 3-Oxo-7-methyl-benzisoxazol (hergestellt
nach Böshagen, Chem. Ber. 100, 959 /J9&7_7) werden mit 987 ecm
30$igein Formaldehyd auf dem Wasserbad in Lösung gebracht.
Nach halbstündigem Stehen der Mischung bei 80°C läßt man dieselbe erkalten, wobei das 3-0xo-N-hydroxymethyl-7-methylbenzisoxazol
auskristallisiert. Nach dem Absaugen wird das Kristalli8at mit Wasser gewaschen und an der Luft getrocknet.
Man erhält ein gelbliches, körniges Pulver vom Schmelzpunkt 112°C. Die Ausbeute beträgt 324 g (81,9 & der Theorie).
322 g (1,8MoI) 3~Oxo-N-hydroxymethyl-7-methyl-1,2-benzisoxazol
werden in 1,3 1 Methylenchlorid suspendiert. Nach Zugabe von 5 ml Dimethylformamid tropft man unter gelegentlicher Außenkühlung
257 g (2,16 Mol) Thionylchlorid zu der Mischung. Nach kurzer Zeit gehen die festen Anteile in Lösung. Man
rührt den Ansatz 1 Stunde nach und wäscht ihn anschließend im Scheidetrichter mit Wasser kongoneutral.
Nach dem Trocknen über Natriumsulfat wird das Lösungsmittel abdestilliert. Man erhält das 3-Oxo-N-chlormethyl-7-methylbenzisoxazol
in rosafarbenen Nädelchen vom Pp. 1220C. Die
