2. Verfahren zur Herstellung der Verbindungen der allgemeinen Formel (I), dadurch gekennzeichnet,
daß man in an sich bekannter Weise entweder
(1) ein Nitril der allgemeinen Formel (II)

Verbindungen reduziert und anschließend den Rest R2, Tür den Fall, daß dieser kein Wasserstoffatom
ist, abspaltet oder
(3) eine Verbindung der allgemeinen Formel (V)

Verbindung der allgemeinen Formel (I) in Form des Säureadditionssalzes isolieren kann.
3. Arzneipräparate, enthaltend Verbindungen der allgemeinen Formel (I) als Wirkstoff.
Gegenstand der Erfindung sind entsprechend den vorstehenden Patentansprüchen neue 5-;2-[(l-Melhyl-3-phenylpropyl)-amino]-äthyl|-salicylsäure-Derivate
der allgemeinen Formel (I)

in der R die Methoxy- oder die Aminogruppe ist. und ihre Säureadditionsalze, Verfahren zur Herstellung
dieser Verbindungen und Arzneipräparate, die diese Verbindungen enthalten.
Die Verhindungen der allgemeinen Formel (I) können eines oder mehrere asymmetrische Kohlenstollatomc
aufweisen. Die Erfindung umfaßt alle möglichen Enaiitiorncren und Diastereomeren der Verbindungen
der allgemeinen Formel (I). Die Racemule können in üblicher Weise, z. B. durch Salzbildung mit einer
ontisch aktiven Säure und anschließende fraktionierte
CH-CH, CH,-' \ (D
Kristallisation der diastereomeren Salze in ihre optischen Antipoden zerlegt werden.
Besonders bevorzugt sind die Verbindungen - ;2 - [(I - Methyl - 3-phcnylpropyl)-amino] -äthyljsalicylamid
- hydrochlorid und 5 - ',2 - [(I - Methyl- - phenylpropyl) - ammo] - äthyl! - salicylsäui emethylesler-hydrochlorid.
Die Verbindungen der allgemeinen Formel (1) können auf verschiedene, an sich bekannte Weisen,
hergestellt werden.
Verbindungen der ; !!gemeinen Formel (I) werden
durch Reduktion eines Nitrils der allgemeinen Formel

in der Z1 die Methoxycarboxy- oder die Carboxamidgruppe
ist oder einen Rest bedeutet, der sich in diese Reste umwandeln läßt, z. B. durch katalytische Hydrierung
in saurer Lösung, oder mit einem komplexen Metallhydrid, z. B. mil Lithiumaluminiumhydrid, her-■gestellt.
Die so erhaltenen primären Amine werden durch Kondensation mit 4-Phenyl-2-butanon und
anschließende Reduktion des entstandenen Azomethins, z. B. mit einem komplexen Meiallhydrid oder
mit Wasserstoff in Gegenwart eines Edelmetallkotalysators, umgesetzt.
In ähnlicher Weise wird eine Carbonyl verbindung der lllgemcinen Formel (111)

in der Z1 die vorgenannten Bedeutungen hat und R2
ein Wasserstoffatom, eine Ben/.ylgruppe, ein niederer
Alkylrest oder ein Acylrcst ist, mit 4-Phenyl-2-aminobutan kondensiert, wobei anschließend z. B. mit
Wasserstoff in Gegenwart eines Edelmetallkatalysators oder mit Natriumborhydrid reduziert wird.
Wenn R2 in der Verbindung der allgemeinen Formel (III) kein Wasserstoffatom ist, wird dieser Rest durch
Hydrolyse oder katalytische Hydrierung abgespalten. Die Verbindungen der allgemeinen Formel (III)
können auf verschiedene Weise hergesellt werden, z. B. indem man einen aromatischen Aldehyd der
allgemeinen Formel (IV)

haben, mit Nitromethan in Gegenwart einer Base zunächst /u einem Nitrostyrol der allgemeinen Formel

anschließend, gegebenenfalls nach Umwandlung des Restes —OR2 in eine Hydroxylgruppe, zu einer Verbindung
der allgemeinen Formel (1) durch reduzierende Alkylierung mit 4-Phenyl-2-butanon umgesetzt
werden.
Die erfindungsgemäßen Verbindungen der allgemeinen Formel (1) besitzen eine ausgeprägte pharntakologische
Wirksamkeit auf das kardiovaskuläre System.
ίο So verursacht z. B. die Verbindung 5-!2-[(l-Methyl-3
- phenylpropyl) - amino] - äthyl | - salicylamid - hydrochlorid
eine dosisabhängige Konstriktion bei isolierten Arterienpräparaten innerhalb eines Dosisbereiches
von 1 ng/ml bis 1 μ&τηΐ. Darüber hinaus wird bei
diesen Konzentrationen auch die anschließende Konstriktoraktivität
bei Verabreichung von Spasmogenen, wie Noradrenalin und vHydroxytryptamin, verhindert.
Die Verwendung der vorgenann'en Verbindung ist deshalb zur Behandlung von vaskulärem Kopfschmetz,
z. B. Migräne, geeignet. Bei Hunden mit renalei Hypertonie wird bei intravenöser Verabreichung
von 1 mg kg der Blutdruck um etwa 45 Torr ernied igt. Hierbei wird der Puls nicht beeinflußt.
Die vergenannte Verbindung ist deshalb zur Behänd lung ν >η ausgeprägter Hypertonie geeignet.
Die Verbindung 5-',2-[(1-Mcthyl-3-phenylpropyl)-aminoj-äthyll-salicylsäure-methylcster-hydrochlorid
hat be; Hunden mit renaler Hypertonie bei einer intravenösen Dosis von 1 mg;kg eine Erniedrigung des
Blutdrucks um etwa 40 Torr und des Pulses um etwa 20 Schläge/Minute zur Folge.
Die Verbindungen der allgemeinen Formel (1) und ihre Saureadditionssalzc können als Wirkstoffe in
Arzneimitteln Tür die Human- oder \ cicrinärmedizin
zu therapeutischen oder prophylaktischen Zwecken verwendet werden. Bevorzugte Salze sind die Salze der
Chlorwasserstoff-, Schwefel-, Malein-. Essig-, Fumar-. Milch und Zitronensäure. Die den Wirkstoff enthaltenden
Arzneimittel können in üblicher Weise unter Verwendung von Trägerstoffen und anderen Hilfsmittelin,

von physiologisch geeigneten Trägerstoffen und Hilfsmitteln in Form von Lösungen. Suspensionen
oder in Form von trockenen Präparaten, die
unmittelbar vor der Verabreichung in flüssige Form gebracht werden, angewendet werden. Die Dosis des
aktivin Wirkstoffes kann innerhalb eines großen Bereiches variieren und wird nach der Starke des
gewünschten Effekts und nach dem Alter und dem Gewicht des Patienten ausgewählt.

I ine Lösung von 7,8 g >(2-Aminoäthyl)-salicylsäure-mcthylcster
wird mit 6.7 g 4-Phenyl-2-hulanon !Stunde bei OC in 150ml Äthanol', die 0.62g
Natriumhydroxid enthalten, gerührt. Danach werden 0,6 g Natriumborhydrid portionsweise während 30 Minuten
zugegeben, und nach einer weiteren Stunde wird das Lösungsmittel abgedampft. Der ölige Rückstand
wird mit 2n-HCl und Äther behandelt und wasserstoffsäure und mit Äthyfacetat gewaschen und
getrocknet. Man erhält 7,9 g5-i2-[(l-Methyl-3-phenylfiltriert.
Der erhaltene weiße Feststoff wird mit Chlorpropyl)
- amino] - äthylj - salicylsäure - methylesterhydrochlorid nach dem Umkristallisieren aus Methanol/Äthylacetat
vom Fp. 199 bis 200 C
5-l2-[( 1 -Mcthyl-3-phenylpropyl)-amino]-äthyl ;-salicylamid-hydrochlorid
1,4 g des vorgenannten Esters in 50 ml Methanol und 20 ml Ammoniaklösung (d = 0,880) werden bei
Raumtemperatur 5 Tage stehengelassen. Nach dem Eindampfen des Gemisches wird der Rückstand mit
verdünnter Salzsäure aufgeschlämmt. Hierbei erhall
ίο man 1,0 g des Amidhydrochlorids vom Fp. 168 bis
169 C.
