Die Erfindung betrifft ein Verfahren zur Herstellung von Hexahydrobenzopyran-Derivaten der allgemeinen
Formel I

in welcher R einen niederen Alkylrest darstellt, das dadurch gekennzeichnet ist, daß man ein Alkylvinylketon

in Gegenwart eines Inertgases bei Raumtemperatur bis 60° C gegebenenfalls in Anwesenheit eines polaren
Lösungsmittels an Cyclohexan-l^-dion anlagert, dann
das erhaltene 2-(3-Oxoalkyl)-cyclohexan-l,3-dion mit einem zur Reduktion von Ketogruppen üblichen Mittel
in alkalischer Lösung partiell zum entsprechenden 2-(3-Hydroxyalkyl)-cycIohexan-l,3-dion reduziert und bo
anschließend in Gegenwart einer Säure in üblicher Weise aufarbeitet.
Als Alkylreste R kommen vorzugsweise der Methyloder Äthylrest in Frage.
Die Addition von Alkylvinylketon an CycIohexan-1,3-dion
ist an sich bekannt, sie wird jedoch nach den bekannten Arbeitsmethoden stets in Gegenwart basischer
Katalysatoren durchgeführt. So beschreiben I. N.
N a ζ a r ο ν und S. I. Zav'yalor (IsvesL Akd. Nauk
SSSR, Otdel. Khim. Nauk 1957, 207; C. A. 51, [1957],
11 302 a) die Umsetzung von Methylvinylketon mit Cyclohexan-l,3-dion in alkalischer Lösung. Hierbei
bildet sich als Hauptprodukt das 2,2-Bis-(3-oxobutyl)-cyclohexan-l,3-dion,
während das gewünschte 2-(3-Oxobutyl)-cyclohexan-l,3-dion nur in einer Ausbeute von ca.
30% isoliert wird.
Nach der ersten Stufe des erfindungsgemäßen Verfahrens gelingt es, das betreffende Alkylvinylketon
auch ohne Zusatz von Katalysatoren glatt an Cyclohexan-l,3-dion anzulagern, wobei in praktisch quantitativer
Ausbeute nur das 2-(3-Oxoalkyl)-cyclohexan-l,3-dion gebildet wird. Darüber hinaus werden aufwendige
Reinigungsoperationen, wie sie beispielsweise bei dem oben beschriebenen bekannten Verfahren notwendig
sind, vermieden.
Dieser glatte Ablauf der ersten Reaktionsstufe des erfindungsgemäßen Verfahrens ist überraschend. Der
Fachmann weiß nämlich, daß die Addition von Vinylketonen an 1,3-Diketone in der Weise erfolgt, daß
die Methylgruppe des 1,3-Diketons in Gegenwart von Basen ein Carbanion bildet; an dieses Carbanion lagert
sich dann die positiv polarisierte terminale Methylengruppe des Vinylketons an. Man konnte also nicht
erwarten, daß sich die Alkylvinylketone in Abwesenheit basischer Katalysatoren an Cyclohexan-l^-dion anlagern
würden. Noch weniger konnte erwartet werden, daß man bei der Umsetzung der betreffenden
Alkylvinylketone mit Cyclohexan-l^-dion die Ausbeute
an 2-(3-Oxoalkyl)-cyclohexan-l,3-dion etwa verdreifachen würde, wenn die Reaktion in Abwesenheit von
Katalysatoren durchgeführt wird.
Über die in der zweiten Stufe des erfindungsgemäßen Verfahrens durchzuführende Reduktion des betreffenden
2-(3-OxoalkyI)-cyclohexan-l,3-dions ist bisher nichts bekannt. Da diese Verbindungen drei Ketogruppen
enthalten, war zu erwarten, daß man bei der Reduktion Gemische unterschiedlicher Reduktionsprodukte
oder die entsprechenden Trihydroxyverbindungen erhalten würde.
Überraschenderweise ist dies aber nicht der Fall, sondern die betreffenden 2-(3-OxoaIkyI-cyclohexan-1,3-dione
werden mit den zur Reduktion von Ketogruppen üblichen Mitteln selektiv zu den entsprechenden
2-(3-Hydroxyalkyl)-cyclohexan-l,3-dionen reduziert. Die gebildeten 2-(3-Hydroxyalkyl)-cyclohexan-l,3-dione
lagern sich dann in Gegenwart von Säuren direkt in die Hexahydrobenzopyran-Derivate der Formel I um.
Die erste Stufe des erfindungsgemäßen Verfahrens wird beispielsweise so durchgeführt, daß man beide
Reaktionskomponenten in Wasser löst, die Lösung bei Raumtemperatur etwa 5 bis 30 Stunden aufbewahrt und
das Reaktionsgemisch in üblicher Weise aufarbeitet, indem man beispielsweise die wäßrige Phase mit einem
organischen Lösungsmittel, wie Chloroform, extrahiert und die organische Phase im Vakuum einengt.
Man erhält dann einen Destillationsrückstand von fast reinem 2-(3-Oxoalkyl)-cyclohexan-l,3-dion. Diese
einfache Arbeitsweise läßt sich selbstverständlich auch variieren, indem man die Reaktion bei einer Temperatur
bis 60° C durchführt und dem Reaktionsgemisch zusätzlich noch polare Lösungsmittel wie Methanol
oder Aceton als Lösungsvermittler zusetzt. Zur Vermeidung unerwünschter Nebenreaktionen wird die
Reaktion unter Ausschluß von Luftsauerstoff in Gegenwart eines Inertgases wie Stickstoff, durchgeführt.
Als Reduktionsmittel für die selektive Reduktion in der zweiten Verfahrensstufe eignen sich beispielsweise
Wasserstoff in Gegenwart üblicher Metallkatalysatoren, wie Raney-Nickel, Platinoxyd oder Palladiumkatalysatoren
sowie gebräuchliche Metallhydride, wie Natriumborhydrid oder Lithiumaluminiumhydrid.
Die Reduktion wird in alkalischer Lösung, beispielsweise in wäßriger oder alkoholischer Natronlauge,
durchgeführt
In fast quantitativer Ausbeute werden die Verbindungen
der Formel I erhalten, wenn man die betreffenden 2-(3-Oxoalkyl)-cyclohexan-13-dione in wäßriger Alkalihydroxydlösung,
beispielsweise in Natronlauge mit Wasserstoff und Raney-Nickel, bei 200C 100° C und 1 bis
100 atm. Druck reduziert und anschließend das Reaktionsgemisch
ansäuert Eine weitere Möglichkeit mit fast quantitativer Ausbeute besteht darin, daß die
Reduktion der betreffenden 2-(3-OxoalkyI)-cyclohexan-1,3-dione
mit Natriumborhydrid bei -20° C bis +5O0C
durchgeführt wird.
Man kann zur Reduktion auch andere Metallhydride, beispielsweise Lithiumaluminiumhydrid verwenden, jedoch
ist eine solche Arbeitsweise aufwendiger und kostspieliger als die vorstehend genannten Reduktionsmethoden.
Die Hexahydrobenzopyran-Derivate der angegebenen Formel I sind wertvolle Zwischenprodukte zur
Synthese pharmakologisch wirksamer Substanzen, insbesondere von Steroidhormonen der Androstan- und
Pregnanreihe sowie der 19-Nor-steroide.
1,3-Cyclohexandion das aus 250 g Resorcin durch
Hydrierung gemäß (Org Synth. Coil. Voll. Ill, S. 278
[1955]) erhalten worden ist wird in 2400 ecm Wasser gelöst Man gibt zu der Lösung 225 ml Methylvinylketon
in 1000 ml Wasser zu und läßt die klare Lösung 24 Stunden bei Raumtemperatur unter Stickstoff stehen.
Zum Entfernen des überschüssigen Methylvinylketons wird die Lösung im Vakuum bei 30—40°C gerührt, dann
mit Natriumchlorid gesättigt, mit Chloroform extrahiert und die organische Phase mit Natriumsulfat getrocknet.
Nach Abdestillieren des Lösungsmittels im Vakuum wird in quantitativer Ausbeute als Rückstand ein öl
erhalten, das leicht kristallisiert.
Nach Umkristallisation aus Äther erhält man 2-(3-Oxobutyl)-cyclohexandion-l,3 vom F. 100- 1040C.
4,2 g des oben erhaltenen rohen 2-(3-Oxobutyl)-cycIohexandions-1,3
werden mit 80 ml Methanol und 20 ml in Natriumhydrid versetzt und das Ganze wird auf — 100C
abgekühlt und innerhalb einer Stunde mit 0,4 g Natriumborhydrid portionsweise versetzt Man rührt
danach eine Stunde bei — 1O0C und hierauf eine Stunde
bei Raumtemperatur. Dann säuert man unter Kühlung mit Eisessig langsam auf pH 5—6 an, destilliert das
Lösungsmittel im Vakuum weitgehend ab und stellt nach Aufnehmen in Wasser mit konzentrierter Salzsäure
auf pH 1 —2 ein. Die Lösung wird mit Natriumchlorid gesättigt, mit Chloroform ausgeschüttelt und die
organische Phase mit Natriumbicarbonatlösung und Wasser gewaschen. Nach Trocknen mit Natriumsulfat
und Abdestillieren des Lösungsmittels im Vakuum erhält man ein Rohprodukt, das nach einmaliger
Destillation im Vakuum beim Kp.o.01118—1200C 3,5 g
2-Methyl-2,3,4,6,7,8-hexahydro-[5H]-benzopyran-5-on vom F. 29-300C liefert.
Reduktion durch katalytische Hydrierung
Man löst 330 g des oben erhaltenen rohen 2-(3-Oxobutyl)-cyclohexandions-l,3
in 750 ml 10%iger Natronlauge und hydriert in Anwesenheit von 75 g Raney-Nikkel
bei 46° C und 130 atü. Nach 4 Stunden ist die Wasserstoffaufnahme praktisch beendet, doch rührt
man das Reaktionsgemisch noch weitere 4 Stunden unter den angegebenen Bedingungen. Nach Abtrennen
des Katalysators versetzt man mit konzentrierter Salzsäure bis zu einem pH-Wert von 1 —2, sättigt mit
Natriumchlorid und extrahiert mit Chloroform. Die organische Phase wird mit Natriumbicarbonatlösung
und Wasser gewaschen, mit Natriumsulfat getrocknet und eingedampft. Nach Destillation des Rückstandes im
Vakuum beim Kp.0,01 118—1200C werden 290 g 2-Methyl-2,3,4,6,7,8-hexahydro-[5H]-benzopyran-5-on
erhalten, das beim Stehen kristallisiert und bei 29—300C
schmilzt.
Claims (1)
- Patentanspruch:Verfahren zur Herstellung von Hexahydrobenzopyran-Derivaten der allgemeinen Formel I(1)in welcher R einen niederen Alkylrest darstellt, dadurch gekennzeichnet, daß man ein Alkylvinylketon der allgemeinen Formel II
