Description
Die Erfindung betrifft Carbonsäurederivate, die eine Hemm
wirkung auf die Matrix-Metalloproteinasen (nachstehend als
MMPs abgekürzt) besitzen, oder die pharmazeutisch verträg
lichen Salze davon und ein Verfahren zu deren Herstellung,
ferner eine pharmazeutische Zusammensetzung, die die Car
bonsäurederivate oder die Salze davon enthält.
MMPs sind zinkabhängige, Calcium-benötigende Enzyme, die
am Abbau der extracellulären Matrix beteiligt sind. Unter
normalen physiologischen Bedingungen ist die Expression
der konstitutiven MMPS niedrig und wird durch natürlich
vorkommende Inhibitoren mit der Bezeichnung TIMPs
(Gewebsinhibitoren von Metalloproteinasen) reguliert. Je
doch kommt es bei pathologischen Zuständen, wie bei rheu
matoider Arthritis und Osteoarthritis, zu einer Fehlregu
lierung der MMP-Expression im Knorpel und zu einer Überex
pression der MMPs, die durch die konstitutiven TIMPs nicht
kontrolliert werden. Die MMP-Spiegel sind enzymatisch
hochaktiv und überschreiten die Menge der TIMPs. Dieser
Zustand führt zu einem Verlust von Proteoglykan und Colla
gen (J. Trzaskos et al., Acta. Onthopaedica Scandinavia,
66, 150 (1995)).
Zusätzlich sind MMP-Inhibitoren zur Behandlung von Horn
hautgeschwüren und Tumorprogression wirksam (R.P. Beckett
et al., D.D.T., 1, 16 (1996)), und MMPs spielen eine wich
tige Rolle bei der Pathogenese der Arteriosklerose und der
Restenose nach einer perkutanen transluminalen Coronar-An
gioplastie (PTCA) (C.M. Dollery et al., Circ. Res., 77,
863 (1995)). Es ist therapeutisch nützlich, erhöhte MMP-
Spiegel durch MMP-Inhibitoren bei diesen pathologischen
Zuständen zu kontrollieren. Jüngst wurde ein Übersichtsar
tikel über MMP-Inhibitoren veröffentlicht (R.P. Beckett et
al., D.D.T., 1, 16 (1996)).
Obwohl viele beschriebene MMP-Inhibitoren eine ausgezeich
nete in-vitro-Aktivität hatten, hatten diese Verbindungen
eine schlechte orale Bioverfügbarkeit. Beispielsweise wur
den diese Verbindungen für die intrapleurale Verabreichung
(Drug News & Perspectives, 8 (4), 247 (1995)) oder als Au
gentropfen (Drug of the Future, 18, 1101 (1993)) formu
liert.
Der vorliegenden Erfindung liegt daher die Aufgabe zu
grunde, pharmazeutische Präparate bereitzustellen, die als
oral verabreichbare MMP-Inhibitoren nützlich sind, sowie
ein Verfahren zu deren Herstellung.
Gegenstand der Erfindung sind neue Carbonsäurederivate der
allgemeinen Formel (I)

lung geeignet sind. Niedrigalkyl im Zusammenhang mit der
allgemeinen Formel (I) bedeutet geradkettiges oder ver
zweigtes C1-C5-Alkyl.
Die Verbindung der allgemeinen Formel (I) kann wie folgt
erhalten werden.
- (1) Im Falle, daß in den Verbindungen der allgemeinen For
mel (I) R1 die Bedeutung -OH hat und R3 die Bedeutung
-NHCH3 oder -NH-tBu hat, wird sie durch folgende Umsetzun
gen hergestellt
(worin R2, R3, R4 und A die oben angegebenen Be deutungen haben). - (2) Im Falle, daß in den Verbindungen der allgemeinen For
mel (I) R1 die Bedeutung -OH und R3 die Bedeutung
-NH(CH2)2SO2NH2 hat, wird sie durch die folgenden Umsetzun
gen hergestellt
(worin R2, R3, R4 und A die oben angegebenen Be deutungen haben.)
Die Estercarbonsäure (II) wird mit dem Amin (III) unter Verwendung von Kupplungsmitteln umgesetzt, um das Ester amid (IV) zu ergeben. Diese Reaktion wird als Yamada-Kupp lung bezeichnet. Bei dieser werden Diphenylphosphorylazid (DPPA) oder Diethylphosphorylcyanid (DEPC) als Kupplungs mittel in Gegenwart von Triethylamin in DMF verwendet (S. Yamada et al., J. Am. Chem. Soc., 94, 6203 (1972)). Zu sätzlich können Dicyclohexylcarbodiimid (DCC), Carbonyldi imidazol (CDI) oder 1-(3-Dimethylaminopropyl)-3-ethylcar bodiimid . HCl-Salz (WSCDI) etc. als Kupplungsmittel ver wendet werden.
Das Carbonsäureamid (V) wird durch Hydrolyse des Ester amids (IV) unter Verwendung von Trifluoressigsäure (TFA), Mineralsäure (HCl, H2SO4) oder Alkalimetallhydroxid (NaOH, KOH oder LiOH) erhalten. - (3) Im Falle, daß in den Verbindungen der allgemeinen For
mel (I) R1 die Bedeutung NHOH hat, wird sie durch folgende
Umsetzungen hergestellt
(worin R2, R3 und A die oben angegebenen Bedeu tungen haben.)
Das Carbonsäureamid (V) wird mit Hydroxylamin in Gegenwart von Kupplungsmitteln (DPPA, DEPC, DCC oder WSCDI) umge setzt, um Hydroxansäureamid (VI) zu ergeben.
Das Hydroxansäureamid (VI) wird auch durch Herstellung von o-Benzylhydroxansäureamid (VIII) unter Verwendung von o-Ben zylhydroxylamin (NH2OBn) anstelle von Hydroxylamin und nachfolgende Debenzylierung erhalten.
Die Verbindung (VI) wird auch durch Umsetzung von Hydroxylamin mit einem aktivierten Ester (IX), der sich von der Verbindung (V) herleitet, unter Verwendung von N-Hy droxysuccinimid (S.B. Singh, Tetra. Lett. 36, 2009 (1995)) erhalten.

mel (I) R1 die (5-Methyl-2-oxo-1,3-dioxoren-4-yl)methylen
oxygruppe ist, wird sie durch folgende Umsetzungen herge
stellt

Nach der Methode von H.-M. Fischler et al. (Tetra. Lett., 1701 (1972)) wird die bei der obigen Reaktion verwendete Verbindung durch Halogenierung von Dimethylvinylcarbonat (XV), hergestellt aus Acyroin und Phosgen, erhalten (F. Sakamoto et al., Chem. Pharm. Bull., 32, 224 (1984)).
Die Verbindung der allgemeinen Formel (II) als Ausgangsma
terial wird durch eine asymmetrische Evans-Alkylierung er
halten (Evans, D.A., J.A.C.S., 104, 1737 (1982)).
Die Verbindung (X) wird mit (S)-(-)-4-Benzyl-2-oxazolidin
(XII) auf dem Wege über das Mischanhydrid (XI) umgesetzt,
um die Verbindung (XIII) zu ergeben. Dann wird die Alky
lierung durchgeführt, um die Verbindung (XIV) zu ergeben,
und der Hilfsstoff wird entfernt, um die Verbindung (II)
zu ergeben.
Die Herstellung der obigen Verbindung (X) als Ausgangsma
terial ist wie folgt.
- (1) Im Falle, daß R2 eine Alkylazulengruppe ist
Das Oxazolanon (XVI) wird mit dem Aldehyd (XVII) umge setzt, um den Azulendiester (XVIII) durch das Enaminver fahren zu ergeben (T. Yanagisawa et al., Chem. Pharm. Bull., 36 (1988)). Die Verbindung (XVIII) wird in den Azu lenmonoester (XIX) durch Demethoxycarbonylierung umgewan delt und diese nachfolgend hydrolysiert, um die Azulencar bonsäure (X) zu ergeben.
Der Aldehyd (XVII) wird durch die folgenden Umsetzungen erhalten. Das Lacton (XX) wird in die Verbindung (XXI) durch eine durch Säure hervorgerufene Ringöffnungsreaktion in Alkohol (ROH) umgewandelt. Danach wird der Aldehyd (XVII) durch PCC-(Pyridiniumchlorochromat)-Oxidation er halten
- (2) Im Falle, daß R2 die p-Alkoxyphenolethergruppe ist
Das p-Alkoxyphenol (XXII) wird in den Phenoxyester (XXIV) umgewandelt. Anschließend wird hydrolysiert, um die p-sub stituierte Phenoxycarbonsäure (X) zu ergeben. - (3) Im Falle, daß R2 die CH2-C(CH3)2(CH2OCH3)-Gruppe ist
Isobutyraldehyd (XXV) wird mit Acrylonitril umgesetzt, um
den Aldehyd (XXVI) zu ergeben. Dieser wird durch NaBH4 in
den Alkohol (XXVII) umgewandelt. Die anschließende Umset
zung mit CH3I ergibt das Methylderivat (XXVIII), und die
Carbonsäure (X) wird durch Hydrolyse erhalten.
Beispiele für Verbindungen der allgemeinen Formel (I) sind
die folgenden Verbindungen:
- 1. N-[(2R)-2-(1'-Carboxymethyl)-3-(azulen-1-yl)propio nyl]-L-phenylalanin-N-methylamid (Verbindung 1)
- 2. N-[(2R)-2-(1'-Carboxymethyl)-4-(azulen-1-yl)butyr oyl]-L-phenylalanin-N-methylamid (Verbindung 2)
- 3. (2S)-2-[(2R)-2-(1'-Carboxymethyl)-4-(azulen-1- yl)butyroyl]amino-3-(4-methoxy)phenylpropanoyl methylamid (Verbindung 3)
- 4. N-[(2R)-2-(1'-Carboxymethyl)-4-(azulen-1-yl)butyr oyl]-L-tert.-leucin-N-methylamid (Verbindung 4)

Die erfindungsgemäßen Carbonsäurederivate eignen sich als
pharmazeutische Präparate zur Behandlung und/oder Verhü
tung von Erkrankungen im Zusammenhang mit der MMP-indu
zierten Zerstörung der extrazellulären Matrix. Sie können
oral in Form von Tabletten, Kapseln, Granulaten und Siru
pen verabreicht werden und können auch intravenös verab
reicht werden. Eine wirksame Dosierung der Verbindungen
beträgt 10 bis 1000 mg einmal bis mehrmals täglich für Er
wachsene, obwohl die Dosierung in Abhängigkeit von dem Al
ter und den Symptomen angepaßt werden kann.
Die erfindungsgemäße Verbindung der allgemeinen Formel (I)
ist ein potenter Inhibitor der MMPs. Pharmakologische Ver
suche wurden wie folgt durchgeführt.
Die Hemmung der MMP-I-Aktivitäten (Typ-I-Collagenase)
wurde durch einen Enzymassay unter Verwendung eines von
menschlichen Fibroblasten abgeleiteten MMP-1 (Yagai Co.
Ltd.) und durch den Typ-I-Collagenase-Aktivität-Assaykit
(Yagai Co. Ltd., E.D. Harris, Jr. et al., Method Enzymol.,
82, 423 (1982) bestimmt. MMP-1 (0,01 E/ml), die Testver
bindung (Konzentrationen 10-10-10-5 M) und mit Fluores
ceinisothiocyanat (FITC)-markiertes Typ-I-Collagen (50 µg)
wurden in 500 µl 0,05 M Tris-HCl-Puffer (pH 7,5), der 200
mM NaCl und 5 mM CaCl2 enthielt, 4 h lang bei 37°C inku
biert. Die Reaktion wurde durch 0,05 M Tris-HCl-Puffer (pH
9,5), der 200 mM NaCl, 50 mM o-Fenantrolin und Ethanol
enthielt, gestoppt, und das Gemisch wurde 10 min lang mit
15.000 g zentrifugiert, und die Fluoreszensintensität (EX
495 nm, EM 520 nm) des so erhaltenen Überstands wurde ge
messen. Der prozentuale Abbau des Substrats wurde durch
Vergleich mit einer Fluoreszensintensität des hitzedenatu
rierten Substrats als 100% berechnet. Die Hemmung der MMP-
1-Aktivität wurde als IC50-Wert ausgedrückt, der die Kon
zentration der Testverbindung war, die notwendig war, um
eine 50%ige Hemmung des durch MMP-1 induzierten Abbaus von
Typ-I-Collagen zu ergeben.
Pro-MMP-2 wurde aus Kulturmedium, das auf mit menschlicher
pro-MMP-2-cDNA transfizierten COS-1-Zellen erhalten worden
war, abgetrennt. Es wurde durch 1 mM (p-Aminophenyl)queck
silberessigsäure aktiviert, und von einem menschlichen Fi
brosarkom abgeleitetes MMP-9 wurde von Yagai Co. Ltd. be
zogen. Die Hemmung beider Enzymaktivitäten wurde durch ei
nen Enzymassay unter Verwendung des Typ-IV-Collagenase-Ak
tivität-Assaykits (Yagai Co. Ltd., Biswaz, C. et al., J.
Cell. Biochem., 28, 39 (1984)) abgeschätzt. MMP-2 oder
MMP-9 (0,01 E/ml), die Testverbindung (mehrere Konzentra
tionen), und FITC-markiertes Typ-IV-Collagen (abgeleitet
von Rinderplazenta, 25 µg) wurden in 100 µl 0,05 M Tris-
HCl-Puffer (pH 7,5), der 200 mM NaCl und 5 mM CaCl2 ent
hielt, bei 42°C 4 h lang inkubiert. Die Hemmwirkung der
Verbindung wurde durch den gleichen Assay wie MMP-1 be
wertet. Unverändertes Typ-IV-Collagen wurde durch Zentri
fugation gefällt, und der prozentuale Abbau des Substrats
wurde durch Vergleich mit der Fluoreszensintensität von
hitzedenaturiertem Substrat als 100% berechnet. Die Hem
mung der Enzymaktivität wurde als IC50-Wert ausgedrückt,
der der Konzentration der Testverbindung entspricht, die
notwendig ist, um eine 50%ige Hemmung des Abbaus von Typ-
IV-Collagen durch das Enzym zu ergeben. Die Ergebnisse
sind in Tabelle 1 beschrieben.

Die erfindungsgemäßen Verbindungen sind neue, potente und
peroral aktive MMP-Inhibitoren.
Eine Lösung von δ-Valerolacton (14,9 g) in H2SO4 (5 ml)
und MeOH (200 ml) wurde 17 h lang am Rückfluß erhitzt.
Nach dem Abkühlen auf -10°C wurde NaHCO3 (1,5 g) zu dem Re
aktionsgemisch gegeben, und es wurde 10 min lang gerührt.
Der unlösliche Teil wurde abfiltriert. Nach Entfernung des
Lösungsmittels im Vakuum aus dem Filtrat wurde der Rück
stand in CH2Cl2 (300 ml) aufgelöst. Molekularsiebe (49 g)
und Pyridiniumchlorchromat (PCC) (49 g) wurden zu dem Ge
misch bei 0°C gegeben, und das Gemisch wurde 1 h lang bei
Raumtemperatur gerührt. Der unlösliche Teil wurde durch
Filtration durch Celite entfernt. Das Filtrat wurde einge
dampft, wodurch der Aldehyd (15 g) erhalten wurde. (M⁺ =
114).
3-Methoxycarbonylpropylaldehyd (7,0 g), hergestellt in Re
ferenzbeispiel 1(a) wurde in EtOH (100 ml) aufgelöst und
sodann mit Morpholin (5,4 ml) und Oxaazulanon (8,3 g) ver
setzt. Das Gemisch wurde 16 h lang am Rückfluß erhitzt.
Das Reaktionsgemisch wurde im Vakuum eingeengt. Nach Zu
gabe von Ethylacetat wurde die Ethylacetatschicht mit
Kochsalzlösung gewaschen, getrocknet (Na2SO4), filtriert
und eingeengt. Das resultierende Produkt wurde durch Sili
cagelsäulenchromatographie (SiO2, 250 g) unter Verwendung
von EtOAc:n-Hexan (1 : 2) gereinigt. Die Titelverbindung
wurde als violettes Öl (9,9 g) erhalten.

1-Methoxycarbonyl-3-(3-methoxycarbonylpropionyl)azulen
(9,9 g), hergestellt in Referenzbeispiel 1(b), wurde mit
100%iger Phosphorsäure (50 ml) versetzt. Das Gemisch wurde
20 min lang bei 110°C gerührt. Nach dem Abkühlen auf Raum
temperatur wurde das Gemisch in Eiswasser eingegossen. Das
Reaktionsgemisch wurde mit Ethylacetat (50 ml × 3) extra
hiert, und dann wurde die Ethylacetatschicht mit Kochsalz
lösung gewaschen, getrocknet (Na2SO4), filtriert und einge
engt. Das resultierende Produkt wurde durch Silicagelsäu
lenchromatographie (SiO2, 250 g) unter Verwendung von
EtOAc:n-Hexan (1 : 10) gereinigt. Die Titelverbindung wurde
als blaues Öl (6,5 g) erhalten.

Azulen-1-propionsäuremethylester (6,5 g), hergestellt in
Referenzbeispiel 1(c) wurde in MeOH (70 ml) aufgelöst. Es
wurde 10%ige NaOH (30 ml) zugegeben, und das Gemisch wurde
2 h lang bei Raumtemperatur gerührt. Nach dem Einengen des
Gemisches im Vakuum wurde der pH-Wert des resultierenden
Rückstands mit 10%iger HCl auf 3,0 eingestellt. Das Reak
tionsgemisch wurde mit Ethylacetat (50 ml × 3) extrahiert.
Die Ethylacetatschicht wurde mit Kochsalzlösung gewaschen
und getrocknet (Na2SO4), filtriert und im Vakuum eingeengt.
Das resultierende Produkt wurde durch Silicagelsäulenchro
matographie (SiO2, 200 g) unter Verwendung von EtOAc : n-
Hexan (1 : 4) gereinigt. Die Titelverbindung wurde in Form
von blauen Kristallen (4,2 g) erhalten.

Ein Gemisch aus p-Methoxyphenol (5 g), K2CO3 (8,35 g) und
Ethyl-6-bromhexanat (10,7 ml) in DMF (80 ml) wurde über
Nacht auf 60°C erhitzt. Es wurde H2O zugegeben, und das
Gemisch wurde mit Ether extrahiert. Die Etherschicht wurde
mit H2O und Kochsalzlösung gewaschen und getrocknet

Ethyl-[6-(4-methoxyphenyl)oxy]hexanat (10,7 g), herge

(Na2SO4), filtriert und im Vakuum eingeengt. Die Titelver
bindung wurde erhalten (8,51 g).

50%ige NaOH (10 ml) wurde zu einer Lösung von Isobutyral
dehyd (25 g) und Acrylonitril (73,5 g) mit 0°C gegeben,
und das Gemisch wurde 3 h lang bei 25 bis 35°C gerührt.
Das Reaktionsgemisch wurde in Wasser (150 ml) eingegossen
und mit Ethylacetat (50 ml × 3) extrahiert. Die Ethylace
tatschicht wurde mit Kochsalzlösung gewaschen und getrock
net (Na2SO4), filtriert und eingeengt. Das resultierende
Produkt wurde destilliert, wodurch die Titelverbindung (29
g) erhalten wurde. KP: 90 ∼ 100°C/2 mmHg.

4-Dimethyl-4-formylbutyronitril (27 g), hergestellt in Re
ferenzbeispiel 3(a), wurde in MeOH (100 ml) aufgelöst und
sodann mit NaBH4 (16,3 g) von 0°C versetzt. Das Gemisch
wurde 2,5 h bei Raumtemperatur gerührt. Das Reaktionsge
misch wurde in eine gesättigte NH4OH-Lösung (150 ml) ein
gegossen und mit Ethylacetat (50 ml × 3) extrahiert. Die
Ethylacetatschicht wurde mit Kochsalzlösung gewaschen und
getrocknet (Na2SO4), filtriert und eingeengt. Es wurde die
Titelverbindung (27,5 g) erhalten.

4-Hydroxymethyl-4-dimethylbutyronitril (27,5 g), herge
stellt in Referenzbeispiel 3(b), wurde in THF (100 ml)
aufgelöst und sodann mit 55%igem NaH (12,5 g) und CH3I
(36,7 g) von 0°C versetzt. Das Gemisch wurde 3 h lang bei
Raumtemperatur gerührt. Das Reaktionsgemisch wurde in Eis
wasser (300 ml) eingegossen und mit Ethylacetat (50 ml × 3)
extrahiert. Die Ethylacetatschicht wurde mit Kochsalz
lösung gewaschen, getrocknet (Na2SO4), filtriert und im Va
kuum eingeengt. Das resultierende Produkt wurde durch Si
licagelsäulenchromatographie unter Verwendung von
EtOAc:n-Hexan (5 : 1) gereinigt. Es wurde die Titelverbin
dung erhalten (22 g).

4-Dimethyl-5-methoxyvaleronitril (22 g), hergestellt in
Referenzbeispiel 3(c), wurde in EtOH (30 ml) aufgelöst
und hierauf mit 50%iger KOH (30 ml) versetzt. Das Gemisch
wurde 4 h lang am Rückfluß gekocht. Das Gemisch wurde im
Vakuum eingeengt, und der resultierende Rückstand wurde
mit H2O (20 ml) versetzt. Der pH-Wert wurde mit 10%iger
HCl auf 4,0 eingestellt. Das Reaktionsgemisch wurde mit
Ethylacetat (50 ml × 3) extrahiert. Die Ethylacetatschicht
wurde mit Kochsalzlösung gewaschen, getrocknet (Na2SO4),
filtriert und im Vakuum eingeengt. Es wurde die Titelver
bindung erhalten (22 g).

Azulen-1-propionsäure (2,1 g) wurde in THF (25 ml) aufge
löst, und die Lösung wurde mit NEt3 (1,65 ml) und an
schließend langsam tropfenweise mit Pivaloylchlorid (1,45
ml) von 0°C versetzt. Es wurde 2 h lang bei Raumtemperatur
gerührt. In einem getrennten Kolben wurde (S)-(-)-4-Ben
zyl-2-oxazolidin (2,66 g) in THF (20 ml) von -78°C aufge
löst, und die Lösung wurde mit n-Butyllithium in n-Hexan
(7,5 ml) versetzt. Es wurde 30 min lang gerührt. Diese Lö
sung wurde zu dem obigen Mischanhydrid von -78°C im Ver
lauf von 5 min gegeben. Dann wurde das Gemisch 1 h lang
bei Raumtemperatur gerührt. Das Reaktionsgemisch wurde in
eine gesättigte NH4Cl-Lösung (80 ml) gegossen und mit
Ethylacetat extrahiert. Die Ethylacetatschicht wurde mit
gesättigter NaHCO3-Lösung und Kochsalzlösung gewaschen,
getrocknet (Na2SO4), filtriert und im Vakuum eingeengt. Das
resultierende Produkt wurde durch Silicagelsäulenchromato
graphie (SiO2, 150 g) unter Verwendung von EtOAc : n-Hexan
(3 : 1) gereinigt. Die Titelverbindung wurde in Form von
blauen Kristallen (3,37 g) erhalten.

Eine 1,69-M-Lösung von n-Butyllithium in n-Hexan (7,0 ml)
wurde zu einer Lösung von Diisopropylamin (1,5 ml) in THF
(20 ml) von -78°C gegeben. Nach 30minütigem Rühren wurde
die in Beispiel 1(a) hergestellte Verbindung (3,37 g) zu
gesetzt. Dann wurde tert.-Butylbromacetat (5,21 ml) zuge
geben, und das Reaktionsgemisch wurde auf -20°C erwärmen
gelassen. Nach 20minütigem Rühren wurde das Reaktionsge
misch in eine gesättigte NH4Cl-Lösung (30 ml) eingegossen
und mit Ethylacetat (20 ml × 3) extrahiert. Die Ethylace
tatschicht wurde mit Kochsalzlösung gewaschen, getrocknet
(Na2SO4), filtriert und im Vakuum eingeengt. Das resultie
rende Produkt wurde durch Silicagelsäulenchromatographie
(SiO2, 150 g) unter Verwendung von EtOAc : n-Hexan (1 : 5) ge
reinigt. Die Titelverbindung wurde in Form von blauen Kri
stallen erhalten (2,8 g).

Die Verbindung (2,8 g), hergestellt in Beispiel 1(b),
wurde in THF (20 ml) und H2O (10 ml) aufgelöst und auf 0°C
abgekühlt. Es wurde langsam 30%iges H2O2 (2,7 g) zugegeben,
und das Gemisch wurde gerührt. Nach 10 Minuten wurde
LiOH.H2O (0,5 g) zugesetzt, und das Gemisch wurde 1 h lang
bei Raumtemperatur gerührt. Danach wurde 10%ige Na2SO3-Lö
sung (20 ml) zu dem Reaktionsgemisch gegeben. Nach 20minü
tigem Rühren wurde das Lösungsmittel im Vakuum abgedampft.
Der resultierende Rückstand wurde mit gesättigter NaHCO3-Lö
sung behandelt, um den pH auf 8 einzustellen. Das Reak
tionsgemisch wurde mit Ethylacetat (20 ml × 3) extrahiert.
Dann wurde die Ethylacetatschicht mit Kochsalzlösung gewa
schen, getrocknet (Na2SO4), filtriert und im Vakuum einge
engt. Freies Benzyloxazolidinon konnte umkristallisiert
und für die weitere Verwendung zurückgeführt werden. Das
wäßrige Gemisch wurde mit 10%iger HCl auf einen pH von 2
angesäuert. Es wurde mit Ethylacetat (20 ml × 3) extra
hiert, und die Ethylacetatschicht wurde mit Kochsalzlösung
gewaschen, getrocknet (Na2SO4), filtriert und im Vakuum
eingeengt. Das resultierende Produkt wurde durch Silica
gelsäulenchromatographie (SiO2, 150 g) unter Verwendung
von EtOAc : n-Hexan (1 : 3) gereinigt. Die Titelverbindung
wurde in Form von blauen Kristallen (1,5 g) erhalten.

Die in Beispiel 1(c) hergestellte Verbindung (500 mg)
wurde in DMF (10 ml) aufgelöst und dann mit HOBt.H2O (0,24
g) und N-Ethylmorpholin (0,18 g) versetzt. Das Gemisch
wurde 30 min lang bei 0°C gerührt, und hierauf wurden L-
Phenylalanin-N-methylamid (0,28 g) und WSCDI (0,3 g) zu
dem Gemisch gegeben. Das Gemisch wurde 4 h lang bei Raum
temperatur gerührt. Das Reaktionsgemisch wurde in 10%ige
Na2CO3-Lösung (20 ml) eingegossen, und es wurde mit Ethyl
acetat (20 ml × 3) extrahiert. Die Ethylacetatschicht
wurde mit Kochsalzlösung gewaschen, getrocknet (Na2SO4),
filtriert und im Vakuum eingeengt. Das resultierende Pro
dukt wurde durch Silicagelsäulenchromatographie (SiO2, 50
g) unter Verwendung von EtOAc : n-Hexan (1 : 3) gereinigt. Die
Titelverbindung wurde in Form von blauen Kristallen erhal
ten (650 mg).

Die in Beispiel 1(d) hergestellte Verbindung (650 mg)
wurde in CH2Cl2 (5 ml) aufgelöst und mit Trifluoressigsäure
(2 ml) von 0°C versetzt. Das Gemisch wurde 2 h lang bei
Raumtemperatur gerührt. Das Reaktionsgemisch wurde in Eis
wasser (50 ml) eingegossen und mit CHCl3 (20 ml × 3) ex
trahiert. Die CHCl3-Schicht wurde mit Kochsalzlösung gewa
schen, getrocknet (Na2SO4), filtriert und im Vakuum einge
engt. Das resultierende Produkt wurde durch Silicagelsäu
lenchromatographie (SiO2, 50 g) unter Verwendung von CHCl3 : MeOH
(7 : 1) gereinigt. Die Titelverbindung wurde in Form
von blauen Kristallen (400 mg) erhalten.

Die in Beispiel 1 hergestellte Verbindung (200 mg) wurde
in DMF (10 ml) und HOBt.H20 (73,0 mg) und N-Ethylmorpholin
(54,9 mg) versetzt. Das Gemisch wurde 30 min lang bei 0°C
gerührt und hierauf mit Hydroxylammoniumchlorid (33,2 mg),
NEt3 (48,3 mg) und WSCDI (191,2 g) versetzt. Das Gemisch
wurde 3 h lang bei Raumtemperatur gerührt. Das Reaktions
gemisch wurde in gesättigte NH4Cl (30 ml) eingegossen und
mit Ethylacetat (20 ml × 3) extrahiert. Die Ethylacetat
schicht wurde mit Kochsalzlösung gewaschen, getrocknet
(Na2SO4), filtriert und im Vakuum eingeengt. Das resultie
rende Produkt wurde durch Silicagelsäulenchromatographie
(SiO2, 50 g) unter Verwendung von CHCl3 : MeOH (5 : 1) gerei
nigt. Die Titelverbindung wurde in Form von weißen Kri
stallen (83 mg) erhalten.

4-(4-Methoxyphenyl)oxybutansäure (3,0 g) wurde in THF (50
ml) aufgelöst. Die Lösung wurde mit NEt3 (2,4 ml) und an
schließend langsam tropfenweise mit Pivaloylchlorid (2,1
ml) von 0°C versetzt. Es wurde 2 h lang bei Raumtemperatur
gerührt. In einem getrennten Kolben wurde (S)-(-)-4-Ben
zyl-2-oxazolidin (3,7 g) in THF (25 ml) von -78°C aufge
löst. Zu dieser Lösung wurde n-Butyllithium in n-Hexan (10
ml) gegeben, und es wurde 30 min lang gerührt. Die Lösung
wurde sodann zu dem obigen Mischanhydrid von -78°C im Ver
lauf von 5 min gegeben. Dann wurde das Gemisch 1 h lang
bei Raumtemperatur gerührt. Das Reaktionsgemisch wurde in
eine gesättigte NH4Cl-Lösung (80 ml) eingegossen und mit
Ethylacetat extrahiert. Die Ethylacetatschicht wurde mit
gesättigter NaHCO3-Lösung und Kochsalzlösung gewaschen,
getrocknet (Na2SO4), filtriert und im Vakuum eingeengt. Das
resultierende Produkt wurde durch Umkristallisation in
Ethylacetat gereinigt. Die Titelverbindung wurde in Form
von weißen Kristallen (4,0 g) erhalten.

Eine 1,69-M-Lösung von n-Butyllithium in n-Hexan (8,1 ml)
wurde zu einer Lösung von Diisopropylamin (1,8 ml) in THF
(40 ml) von -78°C gegeben. Nach 30minütigem Rühren wurde
die in Beispiel 3(a) hergestellte Verbindung (4,0 g) zu
gesetzt. Sodann wurde tert.-Butylbromacetat (5,2 ml) zuge
geben, und das Reaktionsgemisch wurde 30 min lang bei
-78°C gerührt. Es wurde im Verlauf von 1 h auf -20°C er
wärmen gelassen und dann 1 h bei 0°C gerührt. Das Reakti
onsgemisch wurde in eine gesättigte NH4Cl-Lösung (30 ml)
eingegossen und mit Ethylacetat extrahiert. Die Ethylace
tatschicht wurde mit gesättigter NaHCO3-Lösung und mit
Kochsalzlösung gewaschen, getrocknet (Na2SO4), filtriert
und im Vakuum eingeengt. Das resultierende Produkt wurde
durch Silicagelsäulenchromatographie unter Verwendung von
EtOAc : n-Hexan (1 : 5) gereinigt. Die Titelverbindung wurde
als farbloses Öl (2,53 g) erhalten.

Die in Beispiel 3(b) hergestellte Verbindung (2,53 g)
wurde in THF/H2O (4 : 1) aufgelöst (25 ml) und auf 0°C abge
kühlt. Langsam, tropfenweise und unter Rühren wurde
30%iges H2O2 (2,2 ml) zugesetzt. Nach 10 min wurde LiOH.H2O
(0,43 g) zugegeben, und es wurde 1 h lang bei Raumtem
peratur gerührt. Hierauf wurde eine 10%ige Na2SO3-Lösung
(30 ml) tropfenweise zugegeben. Nach 15minütigem Rühren
wurde das Lösungsmittel im Vakuum abgedampft. Der resul
tierende Rückstand wurde mit gesättigter NaHCO3-Lösung be
handelt, um den pH auf 10 einzustellen. Das Reaktionsge
misch wurde mit CHCl3 (20 ml × 3) extrahiert. Die CHCl3-
Schicht wurde mit Kochsalzlösung gewaschen, getrocknet
(Na2SO4), filtriert und im Vakuum eingeengt. Freies Benzyl
oxazolidinon, das umkristallisiert werden könnte, wurde
für weitere Verwendung recyclisiert. Die basische Schicht
wurde sodann abgekühlt und mit 10%iger HCl auf einen pH
von 2 angesäuert. Das Gemisch wurde mit Ethylacetat extra
hiert (3 × 20 ml), und die Ethylacetatschicht wurde mit
Kochsalzlösung gewaschen, getrocknet (Na2SO4), filtriert
und im Vakuum eingeengt. Das resultierende Produkt wurde
durch Silicagelsäulenchromatographie unter Verwendung von
EtOAc : n-Hexan (1 : 3) gereinigt. Die Titelverbindung wurde
in Form von weißen Kristallen (1,13 g) erhalten.

Die in Beispiel 3(c) hergestellte Verbindung (500 mg)
wurde in THF/DMF (3 : 1) (8 ml) aufgelöst. L-Phenylalanin-N-
methylamid (329 mg), DEPC (293 mg) und Et3N (182 mg) wur
den bei 0°C zugesetzt. Das Gemisch wurde 3,5 h bei Raum
temperatur gerührt. Das Reaktionsgemisch wurde in 10%ige
Na2CO-Lösung (20 ml) eingegossen und mit Ethylacetat (20
ml × 3) extrahiert. Die Ethxylacetatschicht wurde mit
Kochsalzlösung gewaschen, getrocknet (Na2SO4), filtriert
und im Vakuum eingeengt. Das resultierende Produkt wurde
durch Silicagelsäulenchromatographie unter Verwendung von
EtOAc:n-Hexan (1 : 3) gereinigt. Die Titelverbindung wurde
in Form von weißen Kristallen (0,64 g) erhalten.

Die in Beispiel 3(d) hergestellte Verbindung (600 mg)
wurde in CH2Cl2 (10 ml) aufgelöst und mit Trifluoressig
säure (3 ml) von 0°C versetzt. Das Gemisch wurde 2 h lang
bei Raumtemperatur gerührt. Das Reaktionsgemisch wurde in
Eiswasser (50 ml) eingegossen und mit CHCl3 (20 ml × 3)
extrahiert. Die CHCl3-Schicht wurde mit Kochsalzlösung ge
waschen, getrocknet (Na2SO4), filtriert und im Vakuum ein
geengt. Das resultierende Produkt wurde durch Umkristalli
sation in Ethylacetat gereinigt. Die Titelverbindung wurde
in Form von weißen Kristallen (470 mg) erhalten.

Die in Beispiel 3 hergestellte Verbindung (51 mg) wurde in
DMF (2 ml) aufgelöst. Es wurden K2CO3 (18 mg) und 4-Bromme
thyl-5-methyl-2-oxo-1,3-dioxol (31 mg) bei 0°C zugegeben.
Das Gemisch wurde 1 h lang bei 0°C gerührt. Das Reaktions
gemisch wurde in gesättigte NH4Cl-Lösung eingegossen und
mit Ethylacetat (20 ml × 3) extrahiert. Die Ethylaceat
schicht wurde mit Kochsalzlösung gewaschen, getrocknet,
(Na2SO4), filtriert und im Vakuum eingeengt. Das resultie
rende Produkt wurde durch Silicagelsäulenchromatographie
unter Verwendung von EtOAc : n-Hexan (1 : 1) gereinigt. Die
Titelverbindung wurde in Form von weißen Kristallen (45
mg) erhalten.

oxy]pentansäure (250 mg), hergestellt in Beispiel 3, wur
de in THF/DMF (3 : 1) (6 ml) aufgelöst. 2(S)-2-Amino-3-(4-
methyl)phenylpropanoylmethylamid (250 mg), DEPC (140 mg)
und Et3N (90 mg) wurde bei 0°C zugegeben. Das Reaktions
gemisch wurde 3,5 h lang bei Raumtemperatur gerührt. Das
Reaktionsgemisch wurde in 10%ige Na2CO3-Lösung (20 ml) ein
gegossen und mit Ethylacetat (20 ml × 3) extrahiert. Die
Ethylacetatschicht wurde mit Kochsalzlösung gewaschen, ge
trocknet (Na2SO4), filtriert und im Vakuum eingeengt. Das
resultierende Produkt wurde durch Silicagelsäulenchromato
graphie (SiO2, 50 g) unter Verwendung von EtOAc : n-Hexan
(2 : 1) gereinigt. Die Titelverbindung wurde in Form eines
farblosen Öls (250 mg) erhalten.

Die in Beispiel 5(a) hergestellte Verbindung (250 mg)
wurde in CH2Cl2 (4 ml) aufgelöst. Trifluoressigsäure (1,5
ml) wurde bei 0°C zugegeben. Das Gemisch wurde 2 h lang
bei Raumtemperatur gerührt. Das Reaktionsgemisch wurde in
Eiswasser (30 ml) eingegossen und mit CHCl3 (20 ml × 3)
extrahiert. Die CHCl3-Schicht wurde mit Kochsalzlösung ge
waschen, getrocknet (Na2SO4), filtriert und im Vakuum ein
geengt. Das resultierende Produkt wurde durch Umkristalli
sation in 50%igem EtOH gereinigt. Die Titelverbindung
wurde in Form von weißen Kristallen erhalten (175 mg).

wurde in THF/DMF (3 : 1) (5 ml) aufgelöst. Hydroxylammonium
chlorid (25 mg), DEPC (61 mg) und Et3N (72 mg) wurden bei
0°C zugegeben. Das Gemisch wurde 3,5 h lang bei Raumtempe
ratur gerührt. Das Reaktionsgemisch wurde in 10%ige
Na2CO3-Lösung (10 ml) eingegossen und mit Ethylacetat (20
ml × 3) extrahiert. Die Ethylacetatschicht wurde mit Koch
salzlösung gewaschen, getrocknet (Na2SO4), filtriert und im
Vakuum eingeengt. Das resultierende Produkt wurde durch
Silicagelsäulenchromatographie (SiO2, 50 g) unter Verwen
dung von CHCl3 : MeOH (5 : 1) gereinigt. Die Titelverbindung
wurde in Form von weißen Kristallen (62 mg) erhalten.

Die in Beispiel 3(c) hergestellte Verbindung (500 mg)
wurde in CH2Cl2/DMF (4 : 1) (5 ml) aufgelöst. HOBt (271 mg),
WSCDI (339 mg) und N-Methylmorpholin (0,19 ml) wurden bei
0°C zugegeben. Das Gemisch wurde 1 h bei Raumtemperatur
gerührt. 2-Aminoethansulfonamid (481 mg) und N-Methylmor
pholin (0,19 ml) wurden zu dem Gemisch gegeben. Das Reak
tionsgemisch wurde 3 h lang bei Raumtemperatur gerührt.
Das Reaktionsgemisch wurde in Eiswasser (50 ml) eingegos
sen und mit Ethylacetat (20 ml × 3) extrahiert. Die Ethyl
acetatschicht wurde mit Kochsalzlösung gewaschen, getrock
net (Na2SO4), filtriert und im Vakuum eingeengt. Das resul
tierende Produkt wurde durch Silicagelsäulenchromatogra
phie (SiO2, 50 g) unter Verwendung von CHCl3 : MeOH (5 : 1)
gereinigt. Die Titelverbindung wurde erhalten.

wurde in CH2Cl2 (6 ml) aufgelöst und mit Trifluoressigsäure
(1 ml) bei 0°C versetzt. Das Gemisch wurde 3 h lang bei
Raumtemperatur gerührt. Das Reaktionsgemisch wurde im Va
kuum eingeengt. Die Titelverbindung wurden in Form von
Kristallen (54%) erhalten.

Die in Beispiel 7 hergestellte Verbindung (363 mg) wurde
in CH2Cl2/DMF (1 : 1) (4 ml) aufgelöst. HOBt (125 mg), WSCDI
(156 mg) und N-Methylmorpholin (89 µg) wurden bei 0°C zu
gegeben. Nach 1stündigem Rühren wurden N-Methylmorpholin
(0,11 ml) und Hydroxylammoniumchlorid (70 ml) zugegeben,
und es wurde über Nacht bei Raumtemperatur gerührt. Gesät
tigte NaHCO3-Lösung und Ether wurden in das Reaktionsge
misch eingegossen. Der resultierende Niederschlag wurde
durch Filtration gesammelt. Es wurde die Titelverbindung
erhalten (43%).

Im Referenzbeispiele 3(d) hergestellte 4-Dimethyl-5-me
thoxypentansäure (16,0 g) wurde in THF (150 ml) aufgelöst.
Die Lösung wurde mit NEt3 (12 ml) und dann langsam trop
fenweise mit Pivaloylchlorid (14,5 ml) bei 0°C versetzt.
Es wurde 2 h lang bei Raumtemperatur gerührt. In einem ge
trennten Kolben wurde (S)-(-)-4-Benzyl-2-oxazolidin (26,6
g) in THF (150 ml) bei -70°C aufgelöst. Die Lösung wurde
mit n-Butyllithium in n-Hexan (75 ml) versetzt, und sie
wurde 30 min lang gerührt. Die Lösung wurde sodann zu dem
obigen Mischanhydrid von -78°C im Verlauf von 5 min gege
ben. Dann wurde das Gemisch 1 h lang bei Raumtemperatur
gerührt. Das Reaktionsgemisch wurde in eine gesättigte
NH4Cl-Lösung (150 ml) eingegossen und mit Ethylacetat (50
ml × 3) extrahiert. Die Ethylaceatschicht wurde mit gesät
tigter NaHCO3-Lösung und Kochsalzlösung gewaschen, ge
trocknet (Na2SO4), filtriert und im Vakuum eingeengt. Das
resultierende Produkt wurde durch Silicagelsäulenchromato
graphie unter Verwendung von EtOAc : n-Hexan (5 : 1) gerei
nigt. Die Titelverbindung wurde in Form eines farblosen
Öls (18,9 g) erhalten.

Die in Beispiel 9(a) hergestellte Verbindung (11,5 g)
wurde in THF (100 ml) aufgelöst. Dann wurden 2M Lithiumdi
isopropylamid (LDA) in Hexan (19,5 ml) von -78°C zuge
setzt. Nach 1stündigem Rühren bei -78°C wurde tropfenweise
tert.-Butylbromacetat (26 ml) zugesetzt. Nach 1stündigem
Rühren wurde das Gemisch in eine NH4Cl-Lösung (150 ml)
eingegossen. Das Gemisch wurde mit Ethylacetat (150 ml)
extrahiert. Die organische Schicht wurde mit Kochsalzlö
sung gewaschen, getrocknet (Na2SO4), filtriert und im Va
kuum eingeengt. Das resultierende Produkt wurde durch Si
licagelsäulenchromatographie unter Verwendung von EtOAc : n-Hexan
(1 : 5) gereinigt. Die Titelverbindung wurde in Form
von gelben Kristallen erhalten (10,8 g).

Die in Beispiel 9(b) hergestellte Verbindung (10,0 g)
wurde in THF (30 ml) und H2O (10 ml) aufgelöst, und es
wurde auf 0°C abgekühlt. Langsam, tropfenweise und unter
Rühren wurde 30%iges H2O2 (20 ml) zugesetzt. Nach 10 min
wurde LiOH.H2O (1,9 g) zugegeben, und es wurde 1 h lang
bei Raumtemperatur gerührt. Dann wurde tropfenweise 10%ige
Na2SO3-Lösung (40 ml) zugegeben. Nach 20minütigem Rühren
wurde das Lösungsmittel im Vakuum abgedampft. Der resul
tierende Rückstand wurde mit gesättigter Na2CO3-Lösung be
handelt, um den pH auf 10 einzustellen. Das Reaktionsge
misch wurde mit Ethylacetat (20 ml × 3) extrahiert. Die
Ethylacetatschicht wurde mit Kochsalzlösung gewaschen und
getrocknet (Na2SO4), filtriert und im Vakuum eingeengt.
Freies Benzyloxazolidinon, das umkristallisiert werden
konnte, wurde für den weiteren Gebrauch recyclisiert. Die
basische Schicht wurde sodann abgekühlt und mit 10%iger
HCl auf einen pH von 2 angesäuert. Das Gemisch wurde mit
Ethylacetat (20 ml × 3) extrahiert. Die Ethylacetatschicht
wurde mit Kochsalzlösung gewaschen, getrocknet (Na2SO4),
filtriert und im Vakuum eingeengt. Das resultierende Pro
dukt wurde durch Silicagelsäulenchromatographie unter Ver
wendung von EtOAc : n-Hexan (1 : 3) gereinigt. Die Titelver
bindung wurde in Form eines farblosen Öls (4,0 g) erhal
ten.

Die in Beispiel 9(c) hergestellte Verbindung (1,0 g)
wurde in DMF (15 ml) aufgelöst. L-Phenylalanin, N-Methyl
amid (0,94 g) und NaHCO3 (5,0 g) wurden zugegeben. Das Ge
misch wurde 30 min lang bei 0°C gerührt und dann mit DPPA
(1,5 g) bei 0°C versetzt. Das Reaktionsgemisch wurde 1 h
bei 0°C gerührt und im Verlauf von 8 h auf Raumtemperatur
erwärmen gelassen. Das Reaktionsgemisch wurde in gesättig
te NH4Cl-Lösung (50 ml) eingegossen, und es wurde mit
Ethylacetat (30 ml × 3) extrahiert. Die Ethylacetatschicht
wurde mit Kochsalzlösung gewaschen, getrocknet (Na2SO4),
filtriert und im Vakuum eingeengt. Das resultierende Pro
dukt wurde durch Silicagelsäulenchromatographie unter Ver
wendung von EtOAc : n-Hexan (3 : 1) gereinigt. Die Titelver
bindung wurde in Form eines farblosen Öls erhalten (0,9
g).

Raumtemperatur gerührt. Das Reaktionsgemisch wurde in ge
sättigte NH4Cl-Lösung (10 ml) eingegossen. Es wurde mit
Ethylacetat (20 ml × 3) extrahiert. Die Ethylacetatschicht
wurde mit Kochsalzlösung gewaschen, getrocknet (Na2SO4),
filtriert und im Vakuum eingeengt. Das resultierende Pro
dukt wurde durch Silicagelsäulenchromatographie unter Ver
wendung von EtOAc gereinigt. Die Titelverbindung wurde in
Form eines farblosen Öls (400 mg) erhalten.

Die in Beispiel 9 hergestellte Verbindung (300 mg) wurde
in DMF (20 ml) aufgelöst. HOBt (144 mg) und N-Ethylmorpho
lin (215 mg) wurden zugegeben. Das Gemisch wurde 30 min
lang bei 0°C gerührt und sodann mit o-Benzylhydroxyamin
(152 mg) und WSCDI (179 g) versetzt. Das Gemisch wurde 3,5
h lang bei Raumtemperatur gerührt. Das Gemisch wurde in
eine 10%ige Na2CO3-Lösung (30 ml) eingegossen und es wurde
mit Ethylacetat (20 ml × 3) extrahiert. Die Ethylacetat
schicht wurde mit Kochsalzlösung gewaschen, getrocknet
(Na2SO4), filtriert und im Vakuum eingeengt. Das resultie
rende Produkt wurde durch Silicagelsäulenchromatographie
unter Verwendung von EtOAc gereinigt. Die Titelverbindung
wurde in Form eines farblosen Öls (380 mg) erhalten.

Ein Gemisch aus der in Beispiel 10(a) hergestellten Ver
bindung (350 mg) und 5% Pd-auf-Kohle (5 mg) in MeOH (10
ml) wurde 2 h lang unter einer Wasserstoffatmosphäre ge
rührt. Dann wurde es filtriert und eingeengt. Das resul
tierende Produkt wurde durch Silicagelsäulenchromatogra
phie unter Verwendung von CHCl3 : MeOH (7 : 1) gereinigt.
Die Titelverbindung wurde in Form von weißen Kristallen
(273 mg) erhalten.
