Abstract
Impfstoff zur Verwendung zur Vorbeugung und/oder Behandlung einer Krankheit, wobei der Impfstoff Folgendes umfasst:i. ein Viruskapsidprotein, das einen ersten Peptid-Tag umfasst, undii. ein Antigen, das an einen zweiten Peptid-Tag fusioniert ist, wobei das Antigen und das Viruskapsidprotein über eine Isopeptidbindung zwischen dem ersten und dem zweiten Peptid-Tag verbunden sind, und wobei i-ii ein virusähnliches Partikel bilden, das das Antigen darstellt.
Claims (51)
- Impfstoff zur Verwendung zur Vorbeugung und/oder Behandlung einer Krankheit, wobei der Impfstoff Folgendes umfasst: i. ein Viruskapsidprotein, das einen ersten Peptid-Tag umfasst, und ii. ein Antigen, das an einen zweiten Peptid-Tag fusioniert ist, wobei das Antigen und das Viruskapsidprotein über eine Isopeptidbindung zwischen dem ersten und dem zweiten Peptid-Tag verbunden sind, und wobei i-ii ein virusähnliches Partikel bilden, das das Antigen darstellt.
- Impfstoff nach
Anspruch 1 , wobei der erste Peptid-Tag einen SpyTag umfasst, und wobei der zweite Peptidtag einen SpyCatcher umfasst, wobei das Antigen und das Viruskapsidprotein über die Wechselwirkung zwischen dem SpyCatcher und einer SpyTag-Wechselwirkung verbunden sind, und wobei i-ii ein virusähnliches Partikel bilden, das das Antigen darstellt. - Impfstoff nach einem der vorhergehenden Ansprüche, wobei der erste Peptidtag einen SpyTag umfasst, und wobei der zweite Peptidtag einen KTag umfasst, und wobei der Impfstoff gegebenenfalls eine SpyLigase umfasst, und wobei das Antigen und das Viruskapsidprotein über die Wechselwirkung zwischen dem SpyTag und dem KTag verbunden sind, und wobei i-ii ein virusähnliches Partikel bilden, das das Antigen darstellt.
- Impfstoff nach einem der vorhergehenden Ansprüche, wobei der erste Peptidtag einen KTag umfasst, und wobei der zweite Peptidtag einen SpyTag umfasst, und wobei der Impfstoff gegebenenfalls eine SpyLigase umfasst, wobei das Antigen und das Viruskapsidprotein über die Wechselwirkung zwischen dem KTag und dem SpyTag verbunden sind, und wobei i-ii ein virusähnliches Partikel bilden, das das Antigen darstellt.
- Impfstoff nach einem der vorhergehenden Ansprüche, wobei das Viruskapsidprotein ein AP205-Kapsidprotein umfasst.
- Impfstoff nach einem der vorhergehenden Ansprüche, wobei das Viruskapsidprotein ein AP205-Kapsidprotein umfasst, und wobei der erste Peptidtag einen oder mehrere SpyCatcher umfasst, und wobei der SpyCatcher an das N-terminale Ende oder an das C-terminale Ende des AP205-Kapsidproteins fusioniert ist, gegebenenfalls durch einen Binder.
- Impfstoff nach einem der vorhergehenden Ansprüche, wobei das AP205-Kapsidprotein an seinem C-terminalen Ende an einen SpyCatcher und an seinem N-terminalen Ende an einen SpyCatcher fusioniert ist, gegebenenfalls durch einen Binder.
- Impfstoff nach einem der vorhergehenden Ansprüche, wobei das Viruskapsidprotein ein AP205-Kapsidprotein umfasst oder daraus besteht, und wobei der erste Peptidtag aus einem oder mehreren SpyTags besteht.

Anspruch 8 , wobei das AP205-Kapsidprotein an seinem C-terminalen Ende an einen SpyTag und an seinem N-terminalen Ende an einen SpyTag fusioniert ist. - Impfstoff nach einem der vorhergehenden Ansprüche, wobei das Viruskapsidprotein, das einen ersten Peptidtag umfasst, die Aminosäuresequenz von SEQ ID NO.76 oder eine biologisch aktive Sequenzvariante umfasst, die wenigstens 70 %, sowie 75 %, sowie 80 %, sowie 85 %, sowie 90 %, sowie 95 %, sowie 96 %, sowie 97 %, sowie 98 %, sowie 99 %, sowie 99,5%, sowie 100 % Sequenzidentität mit der Aminosäuresequenz von SEQ ID NO.76 aufweist.
- Impfstoff nach einem der vorhergehenden Ansprüche, wobei die Wechselwirkung eine Wechselwirkung auf Grundlage einer Isopeptidbindung umfasst.
- Impfstoff nach einem der vorhergehenden Ansprüche, wobei der SpyTag und der KTag nach einem der vorhergehenden Ansprüche mittels einer SpyLigase verbunden sind.
- Impfstoff nach einem der vorhergehenden Ansprüche, wobei die Krankheit Krebs, eine Herz-Kreislauf-Erkrankung, eine Immuninflammationskrankheit, eine chronische Krankheit, eine neurologische Krankheit und/oder eine Infektionskrankheit ist.
- Impfstoff nach einem der vorhergehenden Ansprüche, wobei der Krebs ausgewählt ist aus der Gruppe umfassend Brustkrebs, Magenkrebs, Eierstockkrebs und/oder seröses Gebärmutterkarzinom.
- Impfstoff nach einem der vorhergehenden Ansprüche, wobei die Krankheit ausgewählt ist aus der Gruppe umfassend eine Lipidstörung, wie Hyperlipidämie Typ I, Typ II, Typ III, Typ IV oder Typ V Hyperlipidämie, sekundäre Hypertriglyceridämie, Hypercholesterinämie, familiäre Hypercholesterinämie, Xanthomatose, Cholesterin-Acetyltransferase-Mangel, ein arterioskleroses Leiden (z. B. Arteriosklerose), eine koronare Herzkrankheit, eine Herz-Kreislauf-Erkrankung und die Alzheimererkrankung.
- Impfstoff nach einem der vorhergehenden Ansprüche, wobei die Herz-Kreislauf-Erkrankung ausgewählt ist aus der Gruppe bestehend aus Dyslipidämie, Arteriosklerose und/oder Hypercholesterinämie.
- Impfstoff nach einem der vorhergehenden Ansprüche, wobei die Asthma- oder Immuninflammationserkrankung ausgewählt ist aus der Gruppe umfassend allergisches Asthma, Allergie, Nasenpolypose, atopische Dermatitis, allergische Ösophagitis, hypereosinophiles Syndrom und Churg-Strauss-Syndrom.
- Impfstoff nach einem der vorhergehenden Ansprüche, wobei die Infektionskrankheit ausgewählt ist aus der Gruppe umfassend Malaria und/oder Tuberkulose und/oder HIV und/oder Grippe.
- Impfstoff nach einem der vorhergehenden Ansprüche, wobei der SpyTag die Aminosäuresequenz SEQ ID NO.36 umfasst und/oder der KTag die Aminosäuresequenz SEQ ID NO.38 umfasst.
- Impfstoff nach einem der vorhergehenden Ansprüche, wobei das AP205-Kapsidprotein die Aminosäuresequenz SEQ ID NO.58 umfasst.
- Impfstoff nach einem der vorhergehenden Ansprüche, wobei der erste Peptidtag an das N- und/oder das C-Ende des AP205-Kapsidproteins fusioniert ist.
- Impfstoff nach einem der vorhergehenden Ansprüche, wobei das Antigen ein Polypeptid, ein Peptid und/oder ein Antigenfragment eines Polypeptids ist, das mit einer anormalen physiologischen Reaktion assoziiert ist.
- Impfstoff nach einem der vorhergehenden Ansprüche, wobei die anormale physiologische Reaktion eine Autoimmunkrankheit, eine allergische Reaktion und/oder ein Krebs ist.
- Impfstoff nach einem der vorhergehenden Ansprüche, wobei das Antigen ein Protein, ein Peptid und/oder ein Antigenfragment aus der Gruppe umfassend krebsspezifische Polypeptide, Polypeptide, die mit Herz-Kreislauf-Erkrankungen assoziiert sind, Polypeptide, die mit Asthma assoziiert sind, Polypeptide, die mit nasaler Polypose assoziiert sind, Polypeptide, die mit atopischer Dermatitis assoziiert sind, Polypeptide, die mit allergischer Ösophagitis assoziiert sind, Polypeptide, die mit dem Hypereosinophilem Syndrom assoziiert sind, Polypeptide, die mit Churg-Strauss-Syndrom assoziiert sind und/oder Polypeptide, die mit krankheitserregenden Organismen assoziiert sind, ist.
- Impfstoff nach einem der vorhergehenden Ansprüche, wobei das Antigen ausgewählt ist aus der Gruppe bestehend aus Interleukin-17, Hämagglutinin, GD2, EGF-R, CEA, CD52, CD21, menschlichem Melanomprotein gp100, menschlichem Melanomprotein Melan-A/MART1, Tyrosinase, NA17-A nt, MAGE-3, HPV 16 E7, HPV L2, PD-1, PD-L1, CTLA-4, p53, hCG, Fel d1 und (IHNV) G-Protein oder einem Antigenfragment hiervon.
- Impfstoff nach einem der vorhergehenden Ansprüche zur Verwendung in einer Behandlung einer Infektionskrankheit, wobei das Antigen ein Protein, ein Peptid oder ein Fragment hiervon von dem krankheitserregenden Organismus ist, der die Infektionskrankheit verursacht.
- Impfstoff nach einer der vorhergehenden Ansprüche, wobei das Antigen in der Lage ist, ein Einsetzen einer Immunreaktion bei einem Tier, wie einem Säugetier, wie einem Homo sapiens, einer Kuh, einem Schwein, einem Pferd, einem Schaf, einer Ziege, einem Lama, einer Maus, einer Ratte, einem Affen und/oder einem Vogel, wie einem Huhn und/oder einem Fisch, wie einem Lachs auszulösen.
- Impfstoff nach einem der vorhergehenden Ansprüche, wobei das Antigen, das an einen SpyCatcher fusioniert ist, ferner einen Polyhistidintag umfasst.
- Impfstoff nach einem der vorhergehenden Ansprüche, wobei der SpyCatcher die Aminosäuresequenz SEQ ID NO.37, SEQ ID NO.60 und/oder SEQ ID NO.61 umfasst.
- Impfstoff nach einem der vorhergehenden Ansprüche, wobei der SpyCatcher in einer Position an das Antigen fusioniert ist, ausgewählt aus der Gruppe umfassend das N-terminale Ende, das C-terminale Ende und/oder insertiert im Leserahmen in die Codierungssequenz des Antigens.
- Impfstoff nach einem der vorhergehenden Ansprüche, wobei der SpyCatcher, der an das Antigen fusioniert ist, Folgendes umfasst: i. eine Polypeptidsequenz ausgewählt aus der Gruppe bestehend aus SEQ ID NO.18, SEQ ID NO.19, SEQ ID NO.20, SEQ ID NO.21, SEQ ID NO.22, SEQ ID NO.23, SEQ ID NO.24, SEQ ID NO.25, SEQ ID NO.26, SEQ ID NO.27 und SEQ ID NO.28 oder ii. eine Sequenzvariante der Polypeptidsequenz, wobei die Sequenz wenigstens 95 % Sequenzidentität mit den Sequenzen umfassend SEQ ID NO.18, SEQ ID NO.19, SEQ ID NO.20, SEQ ID NO.21, SEQ ID NO.22, SEQ ID NO.23, SEQ ID NO.24, SEQ ID NO.25, SEQ ID NO.26, SEQ ID NO.27 und SEQ ID NO.28 aufweist.
- Vektor umfassend wenigstens ein Polynukleotid, das Folgendes codiert i. ein AP205-Kapsidprotein, das einen N-terminalen und/oder einen C-terminalen SpyCatcher umfasst, nach einem der vorhergehenden Ansprüche und/oder ii. ein Antigen, das an einen SpyTag und/oder einen KTag fusioniert ist, nach einem der vorhergehenden Ansprüche.
- Vektor nach einem der vorhergehenden Ansprüche, wobei das AP205-Kapsidprotein einen SpyCatcher umfasst, der von einem Polypeptid codiert wird, das ausgewählt ist aus der Gruppe bestehend aus SEQ ID NO.76 und/oder SEQ ID NO.78 oder einer Sequenzvariante die wenigstens 70 %, sowie 75 %, sowie 80 %, sowie 85 %, sowie 90 %, sowie 95 %, sowie 96 %, sowie 97 %, sowie 98 %, sowie 99 %, sowie 99,5 %, sowie 100 % Sequenzidentität mit SEQ ID NO.76 und/oder SEQ ID NO.78 aufweist.
- Vektor nach einem der vorhergehenden Ansprüche, wobei ein SpyTag die Nukleotidsequenz SEQ ID NO.39 umfasst.
- Vektor nach einem der
Ansprüche 34 bis36 , wobei das mit einem SpyTag fusionierte Antigen eine Polynukleotidsequenz aufweist, die Folgendes umfasst: i. eine Nukleotidsequenz, die ein Polypeptid kodiert, das ausgewählt ist aus der Gruppe bestehend aus Interleukin-17, Hämagglutinin, GD2, EGF-R, CEA, CD52, CD21, menschlichem Melanomprotein gp100, menschlichem Melanomprotein Melan-A/MART1, Tyrosinase, NA17-A nt, MAGE-3, HPV 16 E7, HPV L2, PD-1, PD-L1, CTLA-4, p53, hCG, Fel d1 und (IHNV) G-Protein, und/oder ii. einer Sequenzvariante des Polynukleotids, wobei die Kodon-Verwendung verändert ist. - Wirtszelle, die wenigstens ein Polypeptid exprimiert, das von einem Polynukleotid nach einem der
Ansprüche 34 bis37 codiert wird. - Wirtszelle, wobei die Wirtszelle Folgendes exprimiert: i. ein erstes Polypeptid; ein AP205-Kapsidprotein, das einen N-terminalen und/oder einen C-terminalen SpyCatcher nach einem der vorhergehenden Ansprüche umfasst, und ii. ein zweites Polypeptid; ein Antigen, das an einen SpyTag und/oder einen KTag fusioniert ist nach einem der vorhergehenden Ansprüche, und iii. gegebenenfalls eine SpyLigase, wobei die Zelle ausgewählt ist aus der Gruppe umfassend Bakterien-, Hefe-, Pilz-, Pflanzen-, Säugetier- und/oder Insektenzellen.
- Wirtszelle nach einem der vorhergehenden Ansprüche, wobei die Wirtszelle Folgendes exprimiert i. ein erstes Polypeptid, das eine Sequenz aufweist, die zu wenigstens 95 % identisch ist mit SEQ ID NO.76 und/oder SEQ ID NO.78 und/oder ii. ein zweites Polypeptid, das eine Sequenz aufweist, die zu wenigstens 95 % identisch ist mit SEQ ID NO.38, und iii. ein drittes Polypeptid, das eine Sequenz aufweist, die zu wenigstens 95 % identisch ist mit SEQ ID NO.82, SEQ ID NO.79, SEQ ID NO.80 und/oder SEQ ID NO.81, wobei die Zelle ausgewählt ist aus der Gruppe umfassend Bakterien-, Hefe-, Pilz-, Pflanzen-, Säugetier- und/oder Insektenzellen.
- Zusammensetzung umfassend einen Impfstoff nach einem der
Ansprüche 1 bis33 . - Verfahren zur Herstellung einer pharmazeutischen Zusammensetzung, umfassend einen Impfstoff nach einem der vorhergehenden Ansprüche, wobei das Verfahren die folgenden Schritte umfasst i. Erhalten eines ersten Polypeptids; eines AP205-Kapsidproteins, das einen SpyCatcher, einen SpyTag oder einen KTag umfasst nach einem der vorhergehenden Ansprüche, und ii. Erhalten eines zweiten Polypeptids; eines Antigens, das an einen SpyTag, einen KTag oder einen SpyCatcher fusioniert ist nach einem der
Ansprüche 1 bis33 , und iii. Exponieren des ersten Polypeptids gegenüber Bedingungen, die ein Bilden von virusähnlichen Partikeln ermöglichen, und iv. Erhalten eines Impfstoffs durch ein Verbinden des zweiten Polypeptids und der virusähnlichen Partikel über eine Wechselwirkung zwischen dem SpyCatcher und dem SpyTag- oder dem KTag-Polypeptid der virusähnlichen Partikel, und v. Erzeugen einer Zusammensetzung, die den Impfstoff nach einem derAnsprüche 1 bis33 umfasst, dadurch Erhalten einer pharmazeutischen Zusammensetzung. - Verfahren zum Verabreichen eines Impfstoffs zur Behandlung und/oder zum Verhindern eines klinischen Zustandes bei einem behandlungsbedürftigen Subjekt, wobei das Verfahren die folgenden Schritte umfasst i. Erhalten wenigstens eines Impfstoffs nach einem der
Ansprüche 1 bis33 , und ii. wenigstens einmaliges Verabreichen der Zusammensetzung an ein Subjekt zur Vorbeugung und/oder Behandlung einer Krankheit nach einem derAnsprüche 1 bis33 . - Verfahren zur Verabreichung eines Impfstoffs nach
Anspruch 43 , wobei der Impfstoff verstärkt wird durch ein Verabreichen in einer Weise oder ein Körperteil, die/das sich von der vorhergehenden Verabreichung unterscheidet. - Verfahren zum Verabreichen eines Impfstoffs nach einem der
Ansprüche 43 bis44 , wobei der Impfstoff in einen Bereich verabreicht wird, der am wahrscheinlichsten der Träger einer gegebenen Krankheit ist. - Verfahren zum Verabreichen eines Impfstoffs nach einem der
Ansprüche 43 bis45 , wobei das Subjekt ein Tier ist, wie ein Säugetier, wie eine Kuh, ein Schwein, ein Pferd, ein Schaf, eine Ziege, ein Lama, eine Maus, eine Ratte, ein Affe, am meisten bevorzugt wie ein Mensch; oder ein Vogel, wie ein Huhn, oder ein Fisch, wie ein Lachs. - Verfahren zum Verabreichen eines Impfstoffs nach einem der
Ansprüche 43 bis46 , wobei der Impfstoff in Kombination mit jeglichem anderen Impfstoff verabreicht wird. - Verfahren zum Verabreichen eines Impfstoffs nach einem der
Ansprüche 43 bis47 , wobei der Impfstoff einen Teil eines Impfstoffcocktails bildet. - Satz von Teilen, umfassend i. eine Zusammensetzung, umfassend einen Impfstoff nach einem der
Ansprüche 1 bis33 , und ii. ein medizinisches Instrument oder andere Mittel zum Verabreichen des Impfstoffs, und iii. Anweisungen darüber, wie der Satz von Teilen zu verwenden ist. - Satz von Teilen nach
Anspruch 49 , umfassend einen zweiten Wirkstoff. - Verfahren zum Hervorrufen einer Immunreaktion bei einem Subjekt, wobei das Verfahren die folgenden Schritte umfasst i. Erhalten einer Zusammensetzung, die wenigstens einen Impfstoff nach einem der
Ansprüche 1 bis33 umfasst, und/oder ii. wenigstens einmaliges Verabreichen der Zusammensetzung an ein Subjekt zur Vorbeugung und/oder Behandlung einer Krankheit nach einem derAnsprüche 1 bis33 , wodurch eine Immunreaktion bei dem Subjekt hervorgerufen wird.
