Abstract
Die Arzneistoffe 3-N-Formylhydroxylaminopropylphosphonsäure (Fosmidomycin) und 3-N-Acetylhydroxylaminopropylphosphonsäure (FR900098), die über stark polare funktionelle Gruppen verfügen, weisen eine unzureichende Resorption aus dem Gastrointestinal-Trakt auf. Dageben erweisen sich viele Ester dieser Wirkstoffe, und zwar die Verbindungen der Formel (I) DOLLAR F1 in der R¶1¶ für Wasserstoff und Methyl steht, und in der R¶2¶ und R¶3¶ aus der Gruppe ausgewählt sind, die aus Phenylestern, C¶1-9¶-Acyloxy-C¶1-9¶-alkylgruppen, Phenyl-C¶1-9¶-alkyloxy-C¶1-9¶-alkylestern, S-C¶1-9¶-Alkoxythio-C¶1-9¶-alkylestern, C¶1-9¶-Alkyloxybenzylestern und cyclischen Estern, bei denen die beiden esterbildenden Sauerstoffatome über eine Alkylkette mit 1 bis 5 Kohlenstoffatomen verbunden sind und die Alkylkette mit C¶1-9¶-Alkyl-, C¶1-9¶-Alkoxy-, C¶1-9¶-Acyl- und C¶1-9¶-Acyloxygruppen substituiert sein kann, besteht, als geeignete Prodrugs. Diese Verbindungen sind einerseits stabil genug, die Zeit im Gastrointestinaltrakt ohne nennenswerte Spaltung zu überdauern, andererseits werden sie in der Zielzelle weitestgehend gespalten.
Description
Die Arzneistoffe 3-N-Formylhydroxylaminopropylphosphonsäure
(Fosmidomycin) und 3-N-Acetylhydroxylaminopropylphosphonsäure
(FR 900098), die über stark polare funktionelle Gruppen verfü
gen, weisen eine unzureichende Resorption aus dem Gastrointe
stinal-Trakt auf. Diese erschwert oder verhindert die orale
Applikation dieser Substanzen. Dieser Applikationsweg ist aber
in allen Fällen, in denen es der Zustand des Patienten er
laubt, der parenteralen Applikation vorzuziehen. Dies ist ins
besondere bei Malaria-Therapeutika der Fall, die überwiegend
in Regionen mit wenig entwickeltem Gesundheitssystem ihren
Einsatz finden.
Aufgabe der vorliegenden Erfindung ist daher die Entwicklung
von sogenannten "Prodrugs", das sind Derivate des Wirkstoffes
in denen die polaren Partialstrukturen durch chemische Abwand
lung temporär blockiert sind. Das Ziel der Entwicklung dieser
Prodrugs muß es sein, solche Ester zu finden, die einerseits
stabil genug sind, die Zeit im Gastrointestinaltrakt mit sei
nen zum Teil extremen pH-Verhältnissen bis zum Übertritt in
das Blut ohne nennenswerte Spaltung zu überdauern. Auf der an
deren Seite müssen diese Ester so beschaffen sein, daß sie im
Plasma oder spätestens in der Zielzelle möglichst vollständig
von den dort vorkommenden Enzymen gespalten werden können, da
die zu markierenden polaren Strukturen in der Regel für die
Wirksamkeit essentiell sind. Darüber hinaus sind die Ester
funktionen so zu wählen, daß keine toxischen Spaltprodukte
entstehen.
Diese Aufgabe wird durch Arzneimittel mit den Merkmalen des
Anspruchs 1 gelöst. Weiterbildungen der Erfindung sind durch
die Unteransprüche definiert.
Durch Veresterung der polaren Strukturen der Phosphonsäure
gruppe läßt sich der polare Charakter dieser funktionellen
Gruppen deutlich herabsetzen. Zahlreiche unterschiedliche
Ester sind bereits als Prodrugs beschrieben worden [Krise, J.
P., Stella, V. J. Advanced Drug Delivery Reviews 1996, 19,
287-310., Freeman, S., Ross, K. C. Progess in Medicinal Chemi
stry 1997, 34, 112-147]. Dabei wurden bei einfachen Alkyl
estern sowie bei Benzylestern häufig unzureichende Freiset
zungsraten der Wirkform beobachtet. Generell hat die Struktur
des Wirkstoffes einen großen Einfluß auf die Eigenschaften des
Ester-Prodrugs, so daß die Brauchbarkeit einzelner Ester kaum
vorherzusagen ist. Die Eignung der verschiedenen denkbaren
Ester ist letztlich nur durch die Darstellung der entsprechen
den Verbindungen und deren Untersuchung zu beurteilen.
Überraschend hat sich nun gezeigt, daß Arzneimittel mit einem
Gehalt an mindestens einer Verbindung der Formel (I)
in der R1 aus der Gruppe ausgewählt ist, die aus Wasserstoff
und Methyl besteht,
und
in der R2 und R3 unabhängig voneinander aus der Gruppe ausge wählt sind, aus Phenylestern, C1-9-Acyloxy-C1-9-alkylgruppen, Phenyl-C1-9-alkyloxy- C1-9-alkylestern, S-C1-9-Alkoxythio-C1-9--alkylestern, C1-9-Alkyloxybenzylestern und cyclischen Estern, bei denen die beiden esterbildenden Sauerstoffatome über eine Alkylkette mit 1 bis 5 Kohlenstoffatomen verbunden sind und die Alkylkette mit C1-9-Alkyl-, C1-9-Alkoxy-, C1-9-Acyl- und C1-9-Acyloxygruppen substituiert sein kann, besteht, wobei jede Phenylgruppe mit C1-9-Alkylgruppen, C1-9-Alkoxygruppen, C1-9-Acyloxygruppen oder einer Trifluormethylgruppe substituiert sein kann und jede Alkyl-, Alkoxy- und Acylgruppe verzweigt oder unverzweigt sein kann, sich ausgezeichnet als Prodrugs eignen, da sie die Hydroxylaminophosphonsäuren gut freisetzen.

in der R2 und R3 unabhängig voneinander aus der Gruppe ausge wählt sind, aus Phenylestern, C1-9-Acyloxy-C1-9-alkylgruppen, Phenyl-C1-9-alkyloxy- C1-9-alkylestern, S-C1-9-Alkoxythio-C1-9--alkylestern, C1-9-Alkyloxybenzylestern und cyclischen Estern, bei denen die beiden esterbildenden Sauerstoffatome über eine Alkylkette mit 1 bis 5 Kohlenstoffatomen verbunden sind und die Alkylkette mit C1-9-Alkyl-, C1-9-Alkoxy-, C1-9-Acyl- und C1-9-Acyloxygruppen substituiert sein kann, besteht, wobei jede Phenylgruppe mit C1-9-Alkylgruppen, C1-9-Alkoxygruppen, C1-9-Acyloxygruppen oder einer Trifluormethylgruppe substituiert sein kann und jede Alkyl-, Alkoxy- und Acylgruppe verzweigt oder unverzweigt sein kann, sich ausgezeichnet als Prodrugs eignen, da sie die Hydroxylaminophosphonsäuren gut freisetzen.
Besonders bevorzugte Verbindungen sind im folgenden aufge
führt:
Im Fall der Beispiele 7-9 (bzw. 16-18) erfolgt primäre enzyma
tische Spaltung in einigem Abstand zu der Phosphonsäurestruk
tur. Die Freisetzung der Phosphonsäure erfolgt dann durch ei
nen spontanen Zerfall der gebildeten labilen Spaltprodukte.
Im Fall des Beispiels 9 (bzw. 18) entsteht 4-Hydroxyzimtsäure
als Nebenprodukt, die auch als Konservierungsmittel in der
Nahrungsmittelindustrie eingesetzt wird.
Die Darstellung der Verbindungen erfolgt ausgehend von dem li
teraturbekannten 3-Brompropylphosphonsäurediethylester [He
witt, D. G., Teese, M. W. Aust. J. Chem. 1984, 37, 205.], der
in prinzipiell bekannter Weise über die freie Phosphonsäure in
das Phosponsäuredichlorid überführt wird. Aus diesem werden
mit den entweder kommerziell erhältlichen oder nach literatur
bekannten Verfahren hergestellten Alkoholen die entsprechenden
3-Brompropylphosphonsäureester erhalten. Diese werden in prin
zipiell bekannter Weise mit N,O-bis-Boc-hydroxylamin [Staszak,
M. A., Doecke, C. W. Tetrahedron Lett. 1993, 34, 7043] umge
setzt. Nach säurekatalysierter Abspaltung der Schutzgruppen
und N-selektiver Acylierung werden dann die Zielverbindungen
erhalten.
(I) a) Me3SiBr, CH2Cl2, 0°C → RT, 16 h, b) H2O, RT,
2,5 h; (II) PCl5, CHCl3, 70°C, 2 h; (III) R-OH, Pyridin, Die
thylether, 0°C → RT, 5 h; (IV) a) N,O-bis-Boc-Hydroxylamin,
NaH, DMF, 1 h, b) dann das Produkt aus dem vorangegangenen
Schritt, DMF, RT, 20 h; (V) Trifluoressigsäure, CH2Cl2, 0°C → RT,
2 h; (VI) Acetylchlorid, Triethylamin, CH2Cl2, 0°C → RT,
12 h.
Die Synthese der Beispiele 10-18 verläuft in den ersten 5 Stu
fen analog. In der letzten Stufe wird mit O-Formylacetat an
stelle von Acetylchlorid acyliert.
Alle Arbeiten werden in zweimal evakuierten und mit Argon ge
fluteten Reaktionsgefäßen ausgeführt. Alle Lösungsmittel wur
den nach üblichen Methoden vor Gebrauch getrocknet.
Zu einer Lösung von 3-Brompropylphosphonsäurediethylester
(12,95 g, 50 mmol) in CH2Cl2 (40 ml) wurden bei 0°C Trimethyl
bromsilan (38,27 g, 250 mmol) während 15 Min. getropft. An
schließend wurde 16 h gerührt, wobei die Lösung RT erreichte.
Anschließend wurde das überschüssige Reagenz im Vakuum ent
fernt und der Rückstand in Wasser (50 ml) aufgenommen und 2 h
lang gerührt. Dann wurde mit Wasser verdünnt, die organische
Phase abgetrennt und die wäßrige Phase dreimal mit Diethyle
ther extrahiert. Die wäßrige Phase wurde vollständig vom Lö
sungsmittel befreit und der erhaltene Feststoff getrocknet.
Ausbeute: 6,56 g (65%)

Zu einer Suspension von 3-Brompropylphosphonsäure (6,09 g, 30 mmol)
in CHCl3 (40 ml) wurden PCl5 (12,49 g, 60 mmol) während
10 Min. gegeben. Anschließend wurde 2 h auf 70°C erhitzt. Nach
Abdestillieren der flüchtigen Bestandteile wurde das Rohpro
dukt durch Destillation im Kugelofen gereinigt.
Ausbeute: 4,07 g (57%)

Zu einer Lösung von 3-Brompropylphosphonsäuredichlorid (2,4 g,
10 mmol) in Diethylether (10 ml) wurde bei 0°C eine Lösung von
2-Hydroxyessigsäuremethylester (1,54 ml, 20 mmol) und Pyridin
(1,64 ml, 20 mmol) in Diethylether (10 ml) getropft. Anschlie
ßend wurde 5 h gerührt, wobei die Lösung RT erreichte. Nach
Abfiltrieren des ausgefallenen Feststoff wurde das Lösungsmit
tel im Vakuum abdestilliert und der Rückstand in Ethylacetat
(50 ml) aufgenommen. Diese Lösung wurde mit 1 N HCl, ges.
NaH-CO3-Lösung und ges. NaCl-Lösung gewaschen und über MgSO4 ge
trocknet. Das nach Abdestillieren des Lösungsmittels erhaltene
Produkt wurde ohne weitere Reinigung weiter verwendet.
Ausbeute: 2,18 g (63%)

Zu einer Lösung von 3-N,O-Bis-tert.-butyloxycarbonyl
hydroxylamin (1,75 g, 7,5 mmol) in DMF (15 ml) wurde NaH (60%
Suspension, 0,33 g, 8,25 mmol) gegeben. Nach 1 h bei RT wurde
eine Lösung von 3-Brompropylphosphonsäure-bis-(methoxycarbonylmethyl)-ester
(2,08 g, 6 mmol) zugetropft und
anschließend 20 h bei RT gerührt. Dann wurden Diethylether (50 ml)
und ges. NH4Cl-Lösung (30 ml) zugegeben, die organische
Phase abgetrennt und diese mit Wasser und NaCl-Lösung gewa
schen. Nach dem Trocknen über MgSO4 wurde das Lösungsmittel ab
destilliert.
Ausbeute: 2,3 g (77%)

Zu einer Lösung von 3-N,O-Bis-tert.-butyloxycarbonyl-
hydroxylaminopropylphosphonsäure-bis-(methoxycarbonylmethyl)-ester
(2,3 g, 4,62 mmol) in CH2Cl2 (10 ml) wurde bei 0°C
Trifluoressigsäure (5 ml) gegeben und dann 2 h gerührt. Anschlie
ßend wurden die flüchtigen Bestandteile im Vakuum abdestil
liert und der Rückstand in Wasser aufgenommen. Die neutrali
sierte Lösung wurde mit Ethylacetat extrahiert. Die vereinig
ten Extrakte wurden mit NaCl-Lösung gewaschen, über MgSO4 ge
trocknet. Anschließend wurde das Lösungsmittel im Vakuum abde
stilliert.
Ausbeute: 1,27 g (92%)

Zu einer Lösung von 3-Hydroxylamino-propylphosphonsäure-bis-(methoxycarbonylmethyl)-ester
(1,27 g, 4,25 mmol) und Triethylamin
(0,37 ml, 5 mmol) in CH2Cl2 (20 ml) wurde bei 0°C Ace
tylchlorid (0,36 ml, 5 mmol) gelöst in CH2Cl2 (5 ml) getropft.
Die Lösung wurde 12 h gerührt, wobei sie Raumtemperatur er
reicht. Die mit CH2Cl2 verdünnte Lösung wurde mit 1 N HCl, ges.
NaHCO3-Lösung und ges. NaCl-Lösung gewaschen und über MgSO4 getrocknet.
Der nach dem Abdestillieren des Lösungsmittels er
haltene Rückstand wurde säulenchromatographisch (Ethylacetat)
gereinigt.
Ausbeute: 0,84 g (58%)

Eine Lösung von 3-Hydroxylaminopropylphosphonsäure-bis-
(methoxycarbonylmethyl)-ester (1,20 g, 4 mmol) O-Formylacetat
(10 ml) wurde 12 h gerührt. Der nach Abdestillieren der flüch
tigen Bestandteile im Vakuum erhaltente Rückstand wurde in
CH2Cl2 gelöst und mit 1 N HCl, ges. NaHCO3-Lösung und ges. NaCl-
Lösung gewaschen und über MgSO4 getrocknet. Der nach dem Abde
stillieren des Lösungsmittels erhaltene Rückstand wurd säulen
chromatographisch (Ethylacetat) gereinigt.
Ausbeute: 0,85 g (64%)

Ausbeute: 1,27 g (49%)
1H-NMR (CDCl3): δ = 1,22 (m, 6H), 2,08-2,39 (m, 10H), 2,69 (m, 4H), 3,72 (m, 2H), 4,12 (m, 4H), 5,79 (m, 2H), 7,09 (m, 4H), 7,42 (m, 4H), 8,39 (s, 1H).
Für die auf Seite 3 aufgeführten Verbindungen 3 und 6 wurde
die Wirksamkeit an P. vinckei-infizierten BALB/c-Mäusen gete
stet. Die Mäuse wurden am Tag 0 durch i. p. Injektion von
5×107 infizierten Erythrozyten von einer Donor-Maus infiziert.
Am Tag 1 wurde der Erfolg der Infektion an Giemsa-gefärbten
Blutausstrichen kontrolliert. Die Parasitämie betrug dabei ca.
1%. Die Behandlung erfolgte an den Tagen 1 und 2 durch p. o.
Applikation von jeweils 75 mg/kg der Prodrogen. Parallel wurde
eine Gruppe mit der selben Dosis FR 900098 behandelt. Als Kon
trolle blieb eine dritte Gruppe unbehandelt. Zu verschiedenen
Zeitpunkten zwischen den Tagen 4 und 11 wurde die Parasitämie
bestimmt. In der Tabelle ist jeweils der Mittelwert der Para
sitämie von 3 Mäusen angegeben.
Claims (3)
1. Arzneimittel mit einem Gehalt an mindestens einer Verbin
dung der Formel (I)
in der R1 aus der Gruppe ausgewählt ist, die aus Wasser stoff und Methyl besteht,
und
in der R2 und R3 unabhängig voneinander aus der Gruppe aus gewählt sind, die aus Phenylestern, C1-9-Acyloxy-C1-9-alkylgruppen, Phenyl-C1-9-alkyloxy- C1-9-alkylestern, S-C1-9- Alkoxythio-C1-9-alkylestern, C1-9-Alkyloxybenzylestern und cyclischen Estern, bei denen die beiden esterbildenden Sau erstoffatome über eine Alkylkette mit 1 bis 5 Kohlenstoffa tomen verbunden sind und die Alkylkette mit mit C1-9-Alkyl-, C1-9-Alkoxy-, C1-9-Acyl- und C1-9-Acyloxygruppen substituiert sein kann, besteht,
wobei jede Phenylgruppe mit C1-9--Alkylgruppen, C1-9-Alkoxygruppen, C1-9-Acyloxygruppen oder einer Trifluorme thylgruppe substituiert sein kann und jede Alkyl-, Alkoxy- und Acylgruppe verzweigt oder unverzweigt sein kann, zur oralen Applikation.
in der R1 aus der Gruppe ausgewählt ist, die aus Wasser stoff und Methyl besteht,
und
in der R2 und R3 unabhängig voneinander aus der Gruppe aus gewählt sind, die aus Phenylestern, C1-9-Acyloxy-C1-9-alkylgruppen, Phenyl-C1-9-alkyloxy- C1-9-alkylestern, S-C1-9- Alkoxythio-C1-9-alkylestern, C1-9-Alkyloxybenzylestern und cyclischen Estern, bei denen die beiden esterbildenden Sau erstoffatome über eine Alkylkette mit 1 bis 5 Kohlenstoffa tomen verbunden sind und die Alkylkette mit mit C1-9-Alkyl-, C1-9-Alkoxy-, C1-9-Acyl- und C1-9-Acyloxygruppen substituiert sein kann, besteht,
wobei jede Phenylgruppe mit C1-9--Alkylgruppen, C1-9-Alkoxygruppen, C1-9-Acyloxygruppen oder einer Trifluorme thylgruppe substituiert sein kann und jede Alkyl-, Alkoxy- und Acylgruppe verzweigt oder unverzweigt sein kann, zur oralen Applikation.
2. Arzneimittel nach Anspruch 1 zur prophylaktischen und
therapeutischen Behandlung von Infektionen, die durch Bakteri
en, Protozoen, mehrzellige Parasiten, Viren oder Pilze her
vorgerufen werden.
3. Arzneimittel nach Anspruch 1 oder Anspruch 2, dadurch ge
kennzeichnet, daß es einen oder mehrere Wirkstoffe aus der
Gruppe enthält, die aus
3-N-Acetylhydroxylamino-propylphosphonsäure-bis-(methoxy carbonylmethyl)-ester, (S)-3-N-Acetylhydroxylamino propylphosphonsäure-bis-[1-(methoxycarbonyl)ethyl]-ester, 3-N-Acetyl-hydroxylamino-propylphosphonsäurediphenylester, 3-N-Acetyl-hydroxylanino-propylphosphansäure-bis-(4-methyl phenyl)ester, 3-N-Acetyl-hydroxylamino-propylphosphonsäure- bis-(4-trifluormethylphenyl)ester, 3-N-Acetyl-hydroxyl amino-propylphosphonsäure-bis-(4-methoxyphenyl)ester, 3-N-Acetyl-hydroxylamino-propylphosphonsäure-bis-[2-(S-benzoyl thio)ethyl]ester, 3-N-Acetyl-hydroxylamino-propylphosphon säure-bis-[2-(S-pivaloylthio)ethyl]ester, 3-N-Formyl- hydroxylamino-propylphosphonsäure-bis-(methoxycarbonyl methyl)-ester, (S)-3-N-Formylhydroxylamino-propylphosphon säure-bis-[1-(methoxycarbonyl)ethyl]-ester, 3-N-Formyl- hydroxylamino-propylphosphonsäurediphenylester, 3-N-Formyl- hydroxylamino-propylphosphonsäure-bis-(4-methylphenyl)ester, 3-N-Formyl-hydroxylamino-propylphosphonsäure-bis-(4- trifluormethylphenyl)ester, 3-N-Formyl-hydroxylamino- propylphosphonsäure-bis-(4-methoxyphenyl)ester, 3-N-Formyl- hydroxylamino-propylphosphonsäure-bis-[2-(S-benzoyl thio)ethyl]ester und 3-N-Formyl-hydroxylamino- propylphosphon-säure-bis-[2-(S-pivaloylthio)ethyl]ester besteht.
3-N-Acetylhydroxylamino-propylphosphonsäure-bis-(methoxy carbonylmethyl)-ester, (S)-3-N-Acetylhydroxylamino propylphosphonsäure-bis-[1-(methoxycarbonyl)ethyl]-ester, 3-N-Acetyl-hydroxylamino-propylphosphonsäurediphenylester, 3-N-Acetyl-hydroxylanino-propylphosphansäure-bis-(4-methyl phenyl)ester, 3-N-Acetyl-hydroxylamino-propylphosphonsäure- bis-(4-trifluormethylphenyl)ester, 3-N-Acetyl-hydroxyl amino-propylphosphonsäure-bis-(4-methoxyphenyl)ester, 3-N-Acetyl-hydroxylamino-propylphosphonsäure-bis-[2-(S-benzoyl thio)ethyl]ester, 3-N-Acetyl-hydroxylamino-propylphosphon säure-bis-[2-(S-pivaloylthio)ethyl]ester, 3-N-Formyl- hydroxylamino-propylphosphonsäure-bis-(methoxycarbonyl methyl)-ester, (S)-3-N-Formylhydroxylamino-propylphosphon säure-bis-[1-(methoxycarbonyl)ethyl]-ester, 3-N-Formyl- hydroxylamino-propylphosphonsäurediphenylester, 3-N-Formyl- hydroxylamino-propylphosphonsäure-bis-(4-methylphenyl)ester, 3-N-Formyl-hydroxylamino-propylphosphonsäure-bis-(4- trifluormethylphenyl)ester, 3-N-Formyl-hydroxylamino- propylphosphonsäure-bis-(4-methoxyphenyl)ester, 3-N-Formyl- hydroxylamino-propylphosphonsäure-bis-[2-(S-benzoyl thio)ethyl]ester und 3-N-Formyl-hydroxylamino- propylphosphon-säure-bis-[2-(S-pivaloylthio)ethyl]ester besteht.
