Abstract
Es wird ein neues Verfahren zur Herstellung von Benzonitrilverbindungen, die als Herbizide nützlich sind, und ihrer Intermediate bereitgestellt. DOLLAR A Das Verfahren betrifft die Herstellung von Verbindungen der Formel (1), welches das Umsetzen einer Verbindung der Formel (1a) mit einem oder mehreren Cyaniden, die unter Kaliumcyanid, Natriumcyanid und Zinkcyanid ausgewählt sind, in Anwesenheit eines Metallkatalysators umfaßt. DOLLAR F1 In dem Schema ist X Wasserstoff, ein Fluor- oder Chloratom; Z eine Nitro-, Amino- oder C¶1¶-C¶4¶-Alkylsulfonylaminogruppe oder dergleichen; Y ein Fluor-, Chlor- oder Bromatom oder dergleichen; und Q ein Wasserstoffatom, eine Nitro- oder Aminogruppe oder dergleichen.
Description
Die vorliegende Erfindung betrifft ein Verfahren zur
Herstellung von Benzonitrilverbindungen, die im US Patent Nr.
5,127,935 und JP-5-186436A beschrieben sind und als Herbizide
nützlich sind, und ihrer Intermediate.
In JP-9-48761A wird ein Verfahren zur Herstellung von
Benzonitrilverbindungen beschrieben. Da das Verfahren die
Verwendung von Kupfercyanid als Cyanisierungsmittel umfaßt,
hat seine Verwendung im industriellen Maßstab die folgenden
Probleme. Die Behandlung der Abfallflüssigkeit ist problema
tisch, da die Abfallflüssigkeit Kupfer enthält, das eines der
Metalle ist, die unter die Wasserschutzbestimmungen fallen.
Zudem ist Kupfercyanid teuer. Dementsprechend bestand ein Be
darf zur Verbesserung des vorstehend genannten Verfahrens.
Die vorliegende Erfindung betrifft die Entwicklung eines
weiteren Verfahrens zur Herstellung von Benzonitrilverbindun
gen, die als Herbizide nützlich sind, und ihrer Intermediate,
um die vorstehend genannten Aufgaben zu lösen. Es ergab sich,
daß die Benzonitrilverbindung durch Synthetisieren in Anwe
senheit eines Metallkatalysators in hoher Ausbeute erhalten
werden kann, selbst wenn Kalium-, Natrium- oder Zinkcyanid
als Cyanisierungsmittel eingesetzt wird, das eine niedrigere
Reaktivität als Kupfercyanid hat. Somit betrifft die
vorliegende Erfindung das im folgenden dargelegte Her
stellungsverfahren.
- 1. [1] Ein Verfahren zur Herstellung von Benzonitrilverbindun
gen, die durch die Formel (1) dargestellt sind:

X Wasserstoff, ein Fluor- oder Chloratom ist;
Z eine Nitro-, Amino-, C1-C4-Alkylsulfonylamino-, Bis(C1-C4-alkylsulfonyl)amino-, (C2-C5-Alkoxycarbonyl)amino-, (C2-C5-Alkylcarbonyl)amino-, (gegebenenfalls substituiertes Benzoyl)amino-, (C2-C5-Alkylcarbonyl) (C1-C4-Alkylsulfonyl) amino- oder eine (gegebenenfalls substituiertes Benzoyl) (C1- C4-Alkylsulfonyl)aminogruppe darstellt; und
Q ein Wasserstoffatom oder eine Nitro-, Amino-, (C2-C5- Alkoxycarbonyl)amino-, 6-Trifluormethyl-2,4(1H,3H)pyrimidin dion-3-yl-, 1-Methyl-6-trifluormethyl-2,4(1H,3H)pyrimidin dion-3-yl- oder eine 1-Amino-6-trifluormethyl-2,4-(1H,3H) pyrimidindion-3-ylgruppe darstellt, mit der Maßgabe, daß wenn Z eine Nitro- oder Aminogruppe ist, Q eine (C2-C5-Alkoxy carbonyl)amino-, 6-Trifluormethyl-2,4(1H,3H)-pyrimidindion-3- yl-, 1-Methyl-6-trifluormethyl-2,4(1H,3H)-pyrimidindion-3-yl- oder eine 1-Amino-6-trifluormethyl-2,4(1H,3H)pyrimidindion-3- ylgruppe ist;
dadurch gekennzeichnet, daß das Verfahren das Umsetzen einer halogenierten Benzolverbindung, die durch die Formel (1a) dargestellt ist:
worin X, Z und Q wie vorstehend definiert sind und Y ein Fluor-, Chlor-, Brom- oder Iodatom ist, mit der Maßgabe, daß wenn X ein Fluoratom ist, Y ein Chlor-, Brom- oder Iodatom ist, und wenn X ein Chloratom ist, Y ein Brom- oder Iodatom ist;
mit einem oder mehreren Cyaniden, die unter Kaliumcyanid, Natriumcyanid und Zinkcyanid ausgewählt sind, in Anwesenheit eines Metallkatalysators umfaßt. - 2. [2] Ein Verfahren, das vorstehend in Punkt [1] beschrieben wird, wobei Z eine C1-C4-Alkylsulfonylamino-, Bis(C1-C4-alkyl sulfonyl)amino-, (C2-C5-Alkylcarbonyl)(C1-C4-alkyl sulfonyl)amino- oder eine (gegebenenfalls substituiertes Benzoyl)(C1-C4-alkylsulfonyl)aminogruppe ist.
- 3. [3] Ein Verfahren, das im vorstehenden Punkt [1] beschrieben wird, wobei Z eine C1-C4-Alkylsulfonylaminogruppe ist.
- 4. [4] Ein Verfahren, das im vorstehenden Punkt [1] beschrieben wird, wobei Q eine 6-Trifluormethyl-2,4(1H,3H)pyrimidindion- 3-yl-, 1-Methyl-6-trifluormethyl-2,4(1H,3H)pyrimidindion-3- yl- oder eine 1-Amino-6-trifluormethyl-2,4(1H,3H)pyrimidin dion-3-ylgruppe ist.
- 5. [5] Ein Verfahren, das im vorstehenden Punkt [2] beschrieben wird, wobei Q eine 6-Trifluormethyl-2,4(1H,3H)pyrimidindion- 3-yl-, 1-Methyl-6-trifluormethyl-2,4(1H,3H)pyrimidindion-3- yl- oder eine 1-Amino-6-trifluormethyl-2,4(1H,3H)pyrimidin dion-3-ylgruppe ist.
- 6. [6] Ein Verfahren, das im vorstehenden Punkt [3] beschrieben wird, wobei Q eine 6-Trifluormethyl-2,4(1H,3H)pyrimidindion- 3-yl-, 1-Methyl-6-trifluormethyl-2,4(1H,3H)pyrimidindion-3- yl- oder eine 1-Amino-6-trifluormethyl-2,4(1H,3H)pyrimidin dion-3-ylgruppe ist.
- 7. [7] Ein Verfahren, das in den vorstehenden Punkten [1] bis [6] beschrieben wird, wobei Palladium, Nickel oder Platin als der Metallkatalysator eingesetzt wird.
- 8. [8] Ein Verfahren, das in den vorstehenden Punkten [1] bis [6] beschrieben wird, wobei Palladium als der Metall katalysator eingesetzt wird.
- 9. [9] Ein Verfahren, das in den vorstehenden Punkten [7] und [8] beschrieben wird, wobei ein Metall, das unter Zink, Eisen, Mangan, Zinn, Kupfer, Magnesium, Chrom, Titan und Aluminium ausgewählt ist, oder ein Halogenid des Metalls als Cokatalysator eingesetzt wird.
- 10. [10] Ein Verfahren, das in den vorstehenden Punkten [7] und [8] beschrieben wird, wobei ein Halogenid eines Metalls als Cokatalysator eingesetzt wird, das unter Zink, Zinn und Kupfer ausgewählt ist.
Das erfindungsgemäße Verfahren wird im folgenden einge
hend beschrieben. Für das erfindungsgemäße Verfahren gilt
folgendes Schema:

benzoyl(n-propylsulfonyl)amino-, 4-i-Propyloxybenzoyl(n-
propylsulfonyl)amino-, 2-n-Butoxybenzoyl(n-propylsulfonyl)
amino-, 3-n-Butoxybenzoyl(n-propylsulfonyl)amino- und die 4-
n-Butoxybenzoyl(n-propylsulfonyl)aminogruppe und dergleichen.
Unter diesen sind die C1-C4-Alkylsulfonylamino-, Bis(C1-C4-
Alkylsulfonyl)amino-, (C2-C5-Alkylcarbonyl) (C1-C4-
Alkylsulfonyl)amino- und die (gegebenenfalls substituiertes
Benzoyl)(C1-C4-Alkylsulfonyl)aminogruppe bevorzugt, wobei die
C1-C4-Alkylsulfonylaminogruppe stärker bevorzugt ist.
Beispiele von Q umfassen das Wasserstoffatom, die
Nitro-, Amino-, Methoxycarbonylamino, Ethoxycarbonylamino-,
n-Propoxycarbonylamino-, i-Propoxycarbonylamino-, n-Butoxy
carbonylamino-, i-Butoxycarbonylamino-, s-Butoxycarbonyl
amino-, tert-Butoxycarbonylamino-, 6-Trifluormethyl-
2,4(1H,3H)pyrimidindion-3-yl-, 1-Methyl-6-trifluormethyl-
2,4(1H,3H)pyrimidindion-3-yl- und die 1-Amino-6-trifluor
methyl-2,4(1H,3H)pyrimidindion-3-ylgruppe und dergleichen.
Unter diesen sind die 6-Trifluormethyl-2,4(1H,3H)pyrimidin
dion-3-yl-, 1-Methyl-6-trifluormethyl-2,4(1H,3H)pyrimidin
dion-3-yl- und die 1-Amino-6-trifluormethyl-2,4(1H,3H)
pyrimidindion-3-ylgruppe bevorzugt.
Bezogen auf 1 Mol der halogenierten Benzolverbindung
(1a) als Ausgangssubstanz wird Kalium- oder Natriumcyanid im
allgemeinen in einer Menge von 0,5 bis 10 Mol, vorzugsweise
0,8 bis 3 Mol eingesetzt, und Zinkcyanid wird im allgemeinen
in einer Menge von 0,25 bis 10 Mol, vorzugsweise von 0,4 bis
1,5 Mol eingesetzt. Diese Metallcyanide können in Kombination
eingesetzt werden.
Beispiele des Metallkatalysators umfassen Eisen, Kobalt,
Nickel, Ruthenium, Rhodium, Palladium, Iridium, Platin,
Osmium und dergleichen. Nickel, Palladium und Platin sind
bevorzugt, wobei Palladium stärker bevorzugt ist. Genauer
gesagt ist der Nickelkatalysator ein getragener Katalysator,
wie von Siliciumdioxid getragenes Nickel, von Aluminiumoxid
getragenes Nickel und von Kohlenstoff getragenes Nickel. Ein
Komplexkatalysator, wie Dichlor-bis-(triphenylphosphin)
nickel, Dichlor[1,2-bis(diphenylphosphino)ethan]nickel,
Dichlor[1,3-bis(diphenylphosphino)propan]nickel, Dichlor[1,4-
bis(diphenylphosphino)butan]nickel, Dichlor[1,1'-(diphenyl
phosphino)ferrocen]nickel, Dibrombis(triphenylphosphin)
nickel, Dibrom[1,2-bis(diphenylphosphin)ethan]nickel,
Dibrom[1,3-bis(diphenylphosphin)propan]nickel, Dibrombis[1,4-
bis(diphenylphosphin)butan]nickel, Dibrombis[1,1'-(diphenyl
phosphin)ferrocen]nickel, Brombis(triphenylphosphin)nickel,
Chlorbis(triphenylphosphin)nickel, Tris(triphenylphosphin)
nickel, Bis(triphenylphosphin)dicarbonylnickel, Tetrakis
(triphenylphosphin)nickel, Tetrakis(triphenylphosphit)nickel,
Bis(cyclooctadien)nickel, und Acetylacetonatnickel; und
Nickelchlorid, Nickelbromid, Nickeliodid, Nickelnitrat,
Nickelacetat, Nickeloxid, Nickelcyanid und dergleichen.
Der Palladiumkatalysator ist ein getragener Katalysator,
wie von Siliciumdioxid getragenes Palladium, von Aluminium
oxid getragenes Palladium und von Kohlenstoff getragenes
Palladium; ein Komplexkatalysator, wie (2,2-Bipyridin)
dichlorpalladium, Bis(acetonitril)palladiumchlorid,
Bis(benzonitril)palladiumchlorid, Bis[1,2(diphenyl
phosphin)ethan]palladium, Bis[1,2(diphenylphosphin)
ethan]palladiumchlorid, Dichlorbis(triphenylphosphin)
palladium, Dichlorbis(trimethylphosphin)palladium,
Dichlorbis(tributylphosphin)palladium, Tetrakis(triethyl
phosphit)palladium, Bis(cycloocta-1,5-dien)palladium,
Tetrakis(triphenylphosphin)palladium, Dicarbonylbis(tri
phenylphosphin)palladium, Carbonyltris(triphenyl
phosphin)palladium, Acetylacetonatpalladium, Dichlor(1,5-

Der Platinkatalysator ist ein getragener Katalysator,
wie von Siliciumdioxid getragenes Platin, von Aluminiumoxid
getragenes Platin und von Kohlenstoff getragenes Platin; ein
Komplexkatalysator, wie Bis(acetonitril)dichlorplatin,
Bis(benzonitril)dichlorplatin, Dichlorbis(triphenyl
phosphin)platin, Dichlorbis(trimethylphosphin)platin,
Dichlorbis(tributylphosphin)platin, Tetrakis(triphenyl
phosphin)platin, Tetrakis(triphenylphosphit)platin,
Tris(Triphenylphosphin)platin, Dicarbonylbis(triphenyl
phosphin)platin, Bis(triphenylphosphin)(ethylen)platin,
Carbonyltris(triphenylphosphin)platin, Bis(1,5-cyclo
octadien)platin, (1,5-Cyclooctadien)dichlorplatin und (1,5-
Cyclooctadien)dibromplatin; und Platinchlorid, Platinbromid,
Platiniodid, Platinoxid und dergleichen.
Die vorstehenden Katalysatoren können einzeln oder in
Kombination eingesetzt werden.
Der Katalysator wird im allgemeinen in einer Menge von
0,0001 bis 100 Mol%, vorzugsweise von 0,01 bis 50 Mol%
eingesetzt, bezogen auf die Verbindung der allgemeinen Formel
(1a).
Falls erforderlich, kann ein Ligand zu dem
Metallkatalysator zugegeben werden, um den Katalysator zu
stabilisieren und zu aktivieren.
Beispiele für Liganden sind ein- oder mehrzähnige
tertiäre Phosphine, Bis(dialkylphoshino)alkane, wie 1,1-
Bis(dimethylphosphino)methan, 1,1-Bis(Diethylphosphino)
methan, 1,2-Bis(dimethylphosphino)ethan, 1,2-(Diethyl
phosphino)ethan, 1,3-Bis(dimethylphosphino)propan und 1,4-
Bis(dimethylphosphino)butan; Bis(diphenylphosphino)alkane wie
1,1-Bis(diphenylphosphino)methan, 1,2-Bis(diphenylphosphino)
ethan, 1,3-Bis(diphenylphosphino)propan, 1,4-Bis(diphenyl
phosphino)butan, 1,5-Bis(diphenylphosphino)pentan und 1,6-
Bis(diphenylphosphino)hexan; und 1,1'-Bis(diphenyl
phosphino)ferrocen, Tris(3-sulfonatphenyl)phosphinnatrium
salz, Bis(diphenylphosphino)binaphthyl, Trimethylphosphin,
Triethylphosphin, Tributylphosphin, Triphenylphosphin,
Tris(para-tolyl)phosphin, Tris(para-fluorphenyl)phosphin,
Tris(2,6-dimethylphenyl)phosphin, Natriumdiphenylphosphin
benzol-3-sulfonat, Bis(3-sulfonatphenyl)phosphinobenzol-
Natriumsalz, 17-Diphenylphosphino-2,5,8,11,14-pentaoxo
dicyclo-[13.4.0]-nonadeca-15,17,19-trien und 20-Diphenyl
phosphino-2,5,8,11,14,17-hexaoxobicyclo-[16.4.0]-docosa-
18,20,22-trien und dergleichen.
Beispiele anderer Liganden sind Phosphonsäureester, wie
Triethylphosphit, Tributylphosphit, Triphenylphosphit und
Tris(2,6-dimethylphenyl)phosphit, Phosphoniumsalze, wie
Triphenylmethylphosphoniumiodid, Triphenylmethyl
phosphoniumbromid, Triphenylmethylphosphoniumchlorid,
Triphenylallylphosphoniumiodid, Triphenylallyl
phosphoniumbromid, Triphenylallylphosphoniumchlorid,
Tetraphenylphosphoniumiodid, Tetraphenylphosphoniumbromid und
Tetraphenylphosphoniumchlorid; Phosphorsäureester, wie
Triphenylphosphat, Trimethylphosphat, Triethylphosphat und
Triallylphosphat; Phosphinoxide, wie Triphenylphosphinoxid
und Diphenylmethylphosphinoxid; Phosphinsulfide, wie
Triphenylphosphinsulfid und Diphenylmethylphosphinsulfid;
ungesättigte Kohlenwasserstoffe, wie Cyclooctadien und
Cyclopentadien; Nitrile, wie Benzonitril und Acetonitril;
Pyridinanaloga, wie 2,2'-Bipyridin, 1,10-Phenanthrolin und 8-
Hydroxychinolin; Acetylacetone und dergleichen.
Der Ligand wird im allgemeinen in einer Menge von 0 bis
10000 Mol%, vorzugsweise von 0 bis 1000 Mol% eingesetzt, be
zogen auf den Metallkatalysator.
Erfindungsgemäß werden die Metallkatalysatoren und die
Phosphine in Kombination oder getrennt zu einem
Reaktionssystem gegeben, oder alternativ dazu können sie als
ein Komplex eingesetzt werden, der vorher gebildet wurde.
Für die Reduktion eines zweiwertigen Metallkomplexes zum
korrespondierenden nullwertigen Komplex können Zinkpulver,
Zinnpulver, Manganpulver, Magnesiumpulver, Cobaltpulver und
Eisenpulver, Eisen(II)-Salz, Zinn(II)-Salz, Mangan(III)-Salz,
Cobalt(II)-Salz, Ameisensäure und Oxalsäure und Gemische da
von und Legierungspulver davon, und ein Grignard-Reagens, wie
Ethylmagnesiumbromid, ein Alkyllithiumreagens, wie Butyl
lithium, ein metallisches Hydrierungsreagens, wie Natrium
borhydrid, und eine elektrolytische Reduktion eingesetzt wer
den.
Beispiele des organischen Lösungsmittels, das in dem
erfindungsgemäßen Verfahren eingesetzt wird, umfassen
aromatische Kohlenwasserstoffe, wie Benzol, Toluol, Xylol und
Chlorbenzol; halogenierte Kohlenwasserstoffe, wie Chloroform
und Methylenchlorid; Nitrile, wie Acetonitril und
Propionitril; Ketone, wie Aceton, Methylethylketon und
Methylisobutylketon; Ether, wie Diethylether, Dioxan,
Tetrahydrofuran und Dimethoxyethan; tertiäre Amine, wie
Pyridin, 5-Ethyl-2-methylpyridin, Chinolin und N,N'-
Dimethylanilin; Säureamide, wie N,N'-Dimethylacetamid, N,N'-
Dimethylformamid, N-Methylpyrrolidon und Tetramethyl
harnstoff; Schwefel enthaltende Verbindungen, wie Dimethyl
sulfoxid und Sulfolan; Phosphorsäureverbindungen, wie
Hexamethylphosphorsäuretriamid; und dergleichen. Zwei oder
mehr dieser Verbindungen können für die Verwendung gemischt
werden.
Um die Reaktion glatter ablaufen zu lassen, ist die
Verwendung eines Cokatalysators wirksam. Beispiele des
Cokatalysators umfassen Metallsalze, wie Kupfer(I)-chlorid,
Kupfer(I)-bromid, Kupfer(I)-iodid, Kupfer(II)-chlorid,
Kupfer(II)-oxid, Kupfer(II)-sulfat, Kupfer(I)-cyanid,
Kupferpulver, Zink(II)-chlorid, Zink(II)-bromid, Zink(II)-
iodid, Zink(II)-cyanid, Zinkpulver, Zinn(II)-chlorid,
Zinn(II)-bromid, Zinn(II)-iodid, Zinnpulver, Aluminiumoxid,
Aluminiumchlorid, Eisen(II)-chlorid, Eisen(III)-chlorid,
Eisenpulver, Magnesium(II)-chlorid, Chrom(II)-chlorid,
Mangan(II)-chlorid, Blei(IV)-acetat, Titan(IV)-chlorid,
Kaliumhexachloroplatinat(IV) und Kaliumhexacyanoferrat(III);
Kronenether, wie 18-Krone-6-ether, 15-Krone-5-ether und
Dicyclohexyl-18-Krone-6-ether; und Siliciumtetrachlorid.
Gegebenenfalls kann Natriumiodid, Natriumbromid, Kaliumiodid,
Kaliumbromid oder dergleichen zu dem Reaktionssystem gegeben
werden. Unter den vorstehend genannten Cokatalysatoren sind
die Halogenide der Metalle bevorzugt, die unter Zink, Eisen,
Mangan, Zinn, Kupfer, Magnesium, Chrom, Titan und Aluminium
ausgewählt sind. Die stärker bevorzugten Cokatalysatoren sind
die Halogenide des Metalls, das unter Zink, Zinn und Kupfer
ausgewählt ist. Die Menge des einzusetzenden Cokatalysators
ist im allgemeinen 0 bis 10000 Mol%, vorzugsweise 0 bis 500
Mol%, bezogen auf den Metallkatalysator.
Im allgemeinen wird die Reaktion bei einer Temperatur
von Raumtemperatur bis 300°C, vorzugsweise von 50 bis 200°C
durchgeführt. Alternativ dazu kann sie bei der
Rückflußtemperatur des einzusetzenden Lösungsmittels
durchgeführt werden. Im allgemeinen ist die Reaktionszeit in
einem Bereich von 30 Minuten bis 72 Stunden, vorzugsweise von
1 bis 24 Stunden.
Das erfindungsgemäße Verfahren wird unter Bezugnahme auf
die folgenden Beispiele, die den Umfang der Erfindung nicht
einschränken sollen, eingehender beschrieben.
Für die Reaktion in den Beispielen gilt folgendes
Reaktionsschema.
1,2 g (2,5 mMol) 3-(4-Brom-5-ethylsulfonylamino-2-
fluorphenyl)-1-methyl-6-trifluormethyl-2,4(1H,3H)pyrimidin
dion, 0,15 g Natriumcyanid und 0,04 g Zinkchlorid wurden zu
5 ml 5-Ethyl-2-picolin gegeben, und das Gemisch wurde 1
Stunde bei 120°C unter einer Argonatmosphäre gerührt. Danach
wurde 0,6 g Tetrakis(triphenylphosphin)palladium zu dem
Gemisch gegeben, das dann 10 Stunden unter Rühren erhitzt
wurde. Nach dem vollständigen Ablauf der Reaktion wurde die
Titelverbindung mit einer Ausbeute von 90% erhalten, gemessen
durch Flüssigchromatographie durch ein Verfahren mit einem
inneren Standard.
1,2 g (2,5 mMol) 3-(4-Brom-5-ethylsulfonylamino-2-
fluorphenyl)-1-methyl-6-trifluormethyl-2,4(1H,3H)pyrimidin
dion und 0,11 g 55%iges Natriumhydrid in Öl wurden zu 20 g 5-
Ethyl-2-picolin gegeben, und das Gemisch wurde 30 Minuten bei
Raumtemperatur gerührt. Danach wurde 0,07 g Kupfer(II)-
chlorid und 0,15 g Natriumcyanid zu dem Gemisch gegeben, das
dann 1 Stunde bei 140°C unter einer Argonatmosphäre gerührt
wurde. Danach wurde 0,3 g Tetrakis(triphenylphosphin)
palladium zu dem Gemisch gegeben, das dann 3 Stunden unter
Rühren erhitzt wurde. Nach dem vollständigen Ablauf der
Reaktion wurden 79% der Titelverbindung als Wert der
relativen Fläche in der Flüssigchromatographie nachgewiesen.
0,5 g (1,0 mMol) 3-(4-Brom-5-ethylsulfonylamino-2-
fluorphenyl)-1-methyl-6-trifluormethyl-2,4(1H,3H)pyrimidin
dion wurde in 10 ml 5-Ethyl-2-picolin gelöst, dazu wurde 0,04
g Natriumhydrid unter Eiskühlung gegeben, und das Gemisch
wurde 5 Minuten gerührt. Danach wurde 0,04 g Palladiumacetat
und 0,16 g 1,4-Bis(diphenylphosphino)ethan zu dem Gemisch
gegeben, das dann 1 Stunde bei 90°C unter einer Argon
atmosphäre gerührt wurde. Nach dem Abkühlen des Gemisches
wurden 0,03 g Zinkchlorid und 0,06 g Natriumcyanid zu dem
Gemisch gegeben, das dann weitere 4 Stunden gerührt wurde.
Nach dem vollständigen Ablauf der Reaktion wurden 61% der
Titelverbindung als Wert der relativen Fläche in der
Flüssigchromatographie nachgewiesen.
2,0 g (4,2 mMol) 3-(4-Brom-5-ethylsulfonylamino-2-
fluorphenyl)-1-methyl-6-trifluormethyl-2,4(1H,3H)pyrimidin
dion, 0,13 g Zinkcyanid und 0,5 g Tetrakis(triphenylphosphin)
palladium wurden zu 10 g 5-Ethyl-2-picolin gegeben, und das
Gemisch wurde 3,5 Stunden bei 80°C unter einer Argon
atmosphäre gerührt. Nach dem vollständigen Ablauf der
Reaktion wurden 94% der Titelverbindung als Wert der
relativen Fläche in der Flüssigchromatographie nachgewiesen.
In Tabelle 2 werden weitere Beispiele zusammen mit den
vorstehenden Beispielen gezeigt. Jegliche Verbindung, worin
der Benzolring an der 2-Position cyanisiert war, wurde in
allen Beispielen 1 bis 9 kaum nachgewiesen.

Mit dem erfindungsgemäßen Verfahren können
Benzonitrilverbindungen, die als Herbizide nützlich sind, und
ihre Intermediate in hoher Ausbeute erhalten werden.
Claims (10)
1. Verfahren zur Herstellung von Benzonitrilverbindungen,
die durch die Formel (1) dargestellt sind:

X Wasserstoff, ein Fluor- oder Chloratom ist;
Z eine Nitro-, Amino-, C1-C4-Alkylsulfonylamino-, Bis(C2-C4-alkylsulfonyl)amino-, (C2-C5-Alkoxycarbonyl)amino-, (C2-C5-Alkylcarbonyl)amino-, (gegebenenfalls substituiertes Benzoyl)amino-, (C2-C5-Alkylcarbonyl) (C1-C4-Alkyl sulfonyl)amino- oder eine (gegebenenfalls substituiertes Benzoyl)(C1-C4-Alkylsulfonyl)aminogruppe darstellt; und
Q ein Wasserstoffatom oder eine Nitro-, Amino-, (C2-C5- Alkoxycarbonyl)amino-, 6-Trifluormethyl- 2,4(1H,3H)pyrimidindion-3-yl-, 1-Methyl-6-trifluormethyl- 2,4(1H,3H)pyrimidindion-3-yl- oder eine 1-Amino-6-trifluor methyl-2,4-(1H,3H)pyrimidindion-3-ylgruppe darstellt, mit der Maßgabe, daß wenn Z eine Nitro- oder Aminogruppe ist, Q eine (C2-C5-Alkoxycarbonyl)amino-, 6-Trifluormethyl-2,4(1H,3H) pyrimidindion-3-yl-, 1-Methyl-6-trifluormethyl-2,4(1H,3H) pyrimidindion-3-yl- oder eine 1-Amino-6-trifluormethyl- 2,4(1H,3H)pyrimidindion-3-ylgruppe ist,
dadurch gekennzeichnet, daß das Verfahren das Umsetzen einer halogenierten Benzolverbindung, die durch die Formel (1a) dargestellt ist:
worin X, Z und Q wie vorstehend definiert sind und Y ein Fluor-, Chlor-, Brom- oder Iodatom ist, mit der Maßgabe, daß wenn X ein Fluoratom ist, Y ein Chlor-, Brom- oder Iodatom ist, und wenn X ein Chloratom ist, Y ein Brom- oder Iodatom ist,
mit einem oder mehreren Cyaniden, die unter Kaliumcyanid, Natriumcyanid und Zinkcyanid ausgewählt sind, in Anwesenheit eines Metallkatalysators umfaßt.

Z eine Nitro-, Amino-, C1-C4-Alkylsulfonylamino-, Bis(C2-C4-alkylsulfonyl)amino-, (C2-C5-Alkoxycarbonyl)amino-, (C2-C5-Alkylcarbonyl)amino-, (gegebenenfalls substituiertes Benzoyl)amino-, (C2-C5-Alkylcarbonyl) (C1-C4-Alkyl sulfonyl)amino- oder eine (gegebenenfalls substituiertes Benzoyl)(C1-C4-Alkylsulfonyl)aminogruppe darstellt; und
Q ein Wasserstoffatom oder eine Nitro-, Amino-, (C2-C5- Alkoxycarbonyl)amino-, 6-Trifluormethyl- 2,4(1H,3H)pyrimidindion-3-yl-, 1-Methyl-6-trifluormethyl- 2,4(1H,3H)pyrimidindion-3-yl- oder eine 1-Amino-6-trifluor methyl-2,4-(1H,3H)pyrimidindion-3-ylgruppe darstellt, mit der Maßgabe, daß wenn Z eine Nitro- oder Aminogruppe ist, Q eine (C2-C5-Alkoxycarbonyl)amino-, 6-Trifluormethyl-2,4(1H,3H) pyrimidindion-3-yl-, 1-Methyl-6-trifluormethyl-2,4(1H,3H) pyrimidindion-3-yl- oder eine 1-Amino-6-trifluormethyl- 2,4(1H,3H)pyrimidindion-3-ylgruppe ist,
dadurch gekennzeichnet, daß das Verfahren das Umsetzen einer halogenierten Benzolverbindung, die durch die Formel (1a) dargestellt ist:
worin X, Z und Q wie vorstehend definiert sind und Y ein Fluor-, Chlor-, Brom- oder Iodatom ist, mit der Maßgabe, daß wenn X ein Fluoratom ist, Y ein Chlor-, Brom- oder Iodatom ist, und wenn X ein Chloratom ist, Y ein Brom- oder Iodatom ist,
mit einem oder mehreren Cyaniden, die unter Kaliumcyanid, Natriumcyanid und Zinkcyanid ausgewählt sind, in Anwesenheit eines Metallkatalysators umfaßt.
2. Verfahren nach Anspruch 1, wobei Z eine C1-C4-
Alkylsulfonylamino-, Bis(C1-C4-alkylsulfonyl)amino-, (C2-C5-
Alkylcarbonyl)(C1-C4-alkylsulfonyl)amino- oder eine
(gegebenenfalls substituiertes Benzoyl)(C1-C4-
alkylsulfonyl)aminogruppe ist.
3. Verfahren nach Anspruch 1, wobei Z eine C1-C4-Alkyl
sulfonylaminogruppe ist.
4. Verfahren nach Anspruch 1, wobei Q eine 6-Trifluor
methyl-2,4(1H,3H)pyrimidindion-3-yl-, 1-Methyl-6-trifluor
methyl-2,4(1H,3H)pyrimidindion-3-yl- oder eine 1-Amino-6-
trifluormethyl-2,4(1H,3H)pyrimidindion-3-ylgruppe ist.
5. Verfahren nach Anspruch 2, wobei Q eine 6-Trifluor
methyl-2,4(1H,3H)pyrimidindion-3-yl-, 1-Methyl-6-trifluor
methyl-2,4(1H,3H)pyrimidindion-3-yl- oder eine 1-Amino-6-
trifluormethyl-2,4(1H,3H)pyrimidindion-3-ylgruppe ist.
6. Verfahren nach Anspruch 3, wobei Q eine 6-Trifluor
methyl-2,4(1H,3H)pyrimidindion-3-yl-, 1-Methyl-6-trifluor
methyl-2,4(1H,3H)pyrimidindion-3-yl- oder eine 1-Amino-6-
trifluormethyl-2,4(1H,3H)pyrimidindion-3-ylgruppe ist.
7. Verfahren nach Anspruch 1 bis 6, wobei Palladium, Nickel
oder Platin als Metallkatalysator eingesetzt wird.
8. Verfahren nach Anspruch 1 bis 6, wobei Palladium als
Metallkatalysator eingesetzt wird.
9. Verfahren nach Anspruch 7 oder 8, wobei ein Metall, das
unter Zink, Eisen, Mangan, Zinn, Kupfer, Magnesium, Chrom,
Titan und Aluminium ausgewählt ist, oder ein Halogenid des
Metalls als Cokatalysator eingesetzt wird.
10. Verfahren nach Anspruch 7 oder 8, wobei ein Halogenid
eines Metalls als Cokatalysator eingesetzt wird, das unter
Zink, Zinn und Kupfer ausgewählt ist.
