Abstract
Verfahren zur Inhibierung einer MUC1-positiven Tumorzelle in einem Subjekt, wobei das Verfahren umfasst das Verabreichen dem Subjekt eines MUC1-Peptids von wenigstens 4 aufeinander folgenden MUC1-Resten und nicht mehr als 20 aufeinander folgenden MUC1-Resten, das die Sequenz CQC umfasst (SEQ ID NO:4), wobei das aminoterminale Cystein von CQC an seinem NH2-Terminus von wenigstens einem Aminosäurerest abgedeckt ist, der nicht der nativen MUC1-Transmembransequenz entsprechen braucht.
Claims (50)
- Verfahren zur Inhibierung einer MUC1-positiven Tumorzelle in einem Subjekt, wobei das Verfahren umfasst das Verabreichen dem Subjekt eines MUC1-Peptids von wenigstens 4 aufeinander folgenden MUC1-Resten und nicht mehr als 20 aufeinander folgenden MUC1-Resten, das die Sequenz CQC umfasst (SEQ ID NO:4), wobei das aminoterminale Cystein von CQC an seinem NH2-Terminus von wenigstens einem Aminosäurerest abgedeckt ist, der nicht der nativen MUC1-Transmembransequenz entsprechen braucht.
- Verfahren nach Anspruch 1, wobei das Peptid wenigstens 5, 6, 7 oder 8 aufeinander folgende MUC1-Reste umfasst.
- Verfahren nach Anspruch 1, wobei das Peptid nicht mehr als 10 aufeinander folgende Reste, 11 aufeinander folgende Reste, 12 aufeinander folgende Reste, 13 aufeinander folgende Reste, 14 aufeinander folgende Reste, 15 aufeinander folgende Reste, 16 aufeinander folgende Reste, 17 aufeinander folgende Reste, 18 aufeinander folgende Reste oder 19 aufeinander folgende Reste von MUC1 enthält.
- Verfahren nach Anspruch 1, wobei die MUC1-positive Tumorzelle eine Karzinomzelle, eine Leukämiezelle oder eine Myelomzelle ist.
- Verfahren nach Anspruch 4, wobei die Karzinomzelle eine Prostata- oder Brustkarzinomzelle ist.
- Verfahren nach Anspruch 1, wobei das Peptid mit einer Domäne für die Einbringung in Zellen fusioniert ist.
- Verfahren nach Anspruch 6, wobei die Domäne für die Einbringung in Zellen Poly-D-R, Poly-D-P oder Poly-D-K ist.
- Verfahren nach Anspruch 1, wobei das Verabreichen eine intravenöse, intraarterielle, intratumorale, subkutane, topische oder intraperitoneale Verabreichung umfasst.
- Verfahren nach Anspruch 1, wobei das Verabreichen eine lokale, regionale, systemische oder kontinuierliche Verabreichung umfasst.
- Verfahren nach Anspruch 1, wobei das Inhibieren das Induzieren eines Wachstumsstopps der Tumorzelle, einer Apoptose der Tumorzelle und/oder einer Nekrose eines die Tumorzelle umfassenden Tumorgewebes umfasst.
- Verfahren nach Anspruch 1, das ferner das Verabreichen einer zweiten Anti-Krebs-Therapie an das Subjekt umfasst.
- Verfahren nach Anspruch 11, wobei die zweite Anti-Krebs-Therapie eine Operation, Chemotherapie, Radiotherapie, hormonelle Therapie, Toxintherapie, Immuntherapie und Kryotherapie ist.
- Verfahren nach Anspruch 11, wobei die zweite Anti-Krebs-Therapie vor dem Peptid verabreicht wird.
- Verfahren nach Anspruch 11, wobei die zweite Anti-Krebs-Therapie nach dem Peptid verabreicht wird.
- Verfahren nach Anspruch 11, wobei die zweite Anti-Krebs-Therapie gleichzeitig mit dem Peptid verabreicht wird.
- Verfahren nach Anspruch 1, wobei das Subjekt ein Mensch ist.
- Verfahren nach Anspruch 1, wobei das Peptid in einer Dosierung von 0,1–500 mg/kg/Tag verabreicht wird.
- Verfahren nach Anspruch 1, wobei das Peptid in einer Dosierung von 10–100 mg/kg/Tag verabreicht wird.
- Verfahren nach Anspruch 1, wobei das Peptid täglich verabreicht wird.
- Verfahren nach Anspruch 19, wobei das Peptid täglich über 7 Tage, 2 Wochen, 3 Wochen, 4 Wochen, einen Monat, 6 Wochen, 8 Wochen, zwei Monate, 12 Wochen oder 3 Monate verabreicht wird.
- Verfahren nach Anspruch 1, wobei das Peptid wöchentlich verabreicht wird.
- Verfahren nach Anspruch 21, wobei das Peptid wöchentlich über 2 Wochen, 3 Wochen, 4 Wochen, 6 Wochen, 8 Wochen, 10 Wochen oder 12 Wochen verabreicht wird.
- Verfahren nach Anspruch 1, wobei das Peptid nur L-Aminosäuren umfasst.
- Verfahren nach Anspruch 1, wobei das Peptid nur D-Aminosäuren umfasst.
- Verfahren nach Anspruch 1, wobei das Peptid eine Mischung aus L- und D-Aminosäuren umfasst.
- Verfahren nach Anspruch 1, das ferner den Schritt des Bestimmens der Expression von MUC1 in einer Tumorzelle des Subjekts vor dem Verabreichen des Peptids umfasst.
- Verfahren nach Anspruch 1, das ferner den Schritt des Bestimmens der Wirkung des Peptids auf die Expression von MUC1 in einer Tumorzelle des Subjekts umfasst.
- Pharmazeutische Zusammensetzung, die umfasst a) ein MUC1-Peptid von wenigstens 4 aufeinander folgenden MUC1-Resten und nicht mehr als 20 aufeinander folgenden MUC1-Resten, das die Sequenz CQC umfasst, wobei das aminoterminale Cystein von CQC an seinem NH2-Terminus von wenigstens einem Aminosäurerest abgedeckt ist, der nicht der nativen MUC1-Transmembransequenz entsprechen braucht, und b) einen pharmazeutisch annehmbaren Träger, Puffer oder ein pharmazeutisch annehmbares Verdünnungsmittel.
- Zusammensetzung nach Anspruch 28, wobei das Peptid aus wenigstens 5, 6, 7 oder 8 aufeinander folgenden MUC1-Resten besteht.
- Zusammensetzung nach Anspruch 28, wobei das Peptid nicht mehr als 10 aufeinander folgende Reste, 11 aufeinander folgende Reste, 12 aufeinander folgende Reste, 13 aufeinander folgende Reste, 14 aufeinander folgende Reste, 15 aufeinander folgende Reste, 16 aufeinander folgende Reste, 17 aufeinander folgende Reste, 18 aufeinander folgende Reste oder 19 aufeinander folgende Reste von MUC1 enthält.
- Zusammensetzung nach Anspruch 28, wobei das Peptid mit einer Domäne für die Einbringung in Zellen oder einer Domäne für die Transduktion von Zellen fusioniert ist.
- Zusammensetzung nach Anspruch 31, wobei die Domäne für die Transduktion von Zellen eine HIV-Tat-Domäne für die Transduktion von Zellen ist.
- Zusammensetzung nach Anspruch 31, wobei die Domäne für die Einbringung in Zellen Poly-D-R, Poly-D-P oder Poly-D-K ist.
- Zusammensetzung nach Anspruch 28, wobei das Peptid eine Länge von wenigstens 8 Resten hat und wenigstens zwei nicht benachbarte Reste über ihre Seitenketten eine Brücke bilden.
- Zusammensetzung nach Anspruch 34, wobei die Brücke einen Linker, chemisch modifizierte Seitenketten oder ein Hydrocarbon-Stapling umfasst.
- Zusammensetzung nach Anspruch 34, wobei der Linker Modifikationen umfasst, die eine alpha-helikale Struktur des Peptids stabilisieren.
- Zusammensetzung nach Anspruch 28, wobei der Puffer β-Mercaptoethanol, Glutathion oder Ascorbinsäure umfasst.
- Verfahren zur Hemmung der Oligomerisierung und des nuklearen Transports von MUC1 in eine Zelle, das umfasst das In-Kontakt-Bringen einer MUC1-exprimierenden Zelle mit einem MUC1-Peptid von wenigstens 4 aufeinander folgenden MUC1-Resten und nicht mehr als 20 aufeinander folgenden MUC1-Resten, das die Sequenz CQC umfasst, wobei das aminoterminale Cystein von CQC an seinem NH2-Terminus von wenigstens einem Aminosäurerest abgedeckt ist, der nicht der nativen MUC1-Transmembransequenz entsprechen muss.
- Verfahren nach Anspruch 38, wobei das Peptid aus wenigstens 5, 6, 7 oder 8 aufeinander folgenden Resten von MUC1 besteht.
- Verfahren nach Anspruch 38, wobei das Peptid nicht mehr als 10 aufeinander folgende Reste, 11 aufeinander folgende Reste, 12 aufeinander folgende Reste, 13 aufeinander folgende Reste, 14 aufeinander folgende Reste, 15 aufeinander folgende Reste, 16 aufeinander folgende Reste, 17 aufeinander folgende Reste, 18 aufeinander folgende Reste oder 19 aufeinander folgende Reste von MUC1 enthält.
- Verfahren nach Anspruch 38, wobei das Peptid mit einer Domäne für die Einbringung in Zellen fusioniert ist.
- Verfahren nach Anspruch 41, wobei die Domäne für die Einbringung in Zellen Poly-D-R, Poly-D-P oder Poly-D-K ist.
- Verfahren nach Anspruch 43, wobei die MUC1-exprimierende Zelle eine Tumorzelle ist.
- Verfahren nach Anspruch 43, wobei die MUC1-positive Tumorzelle eine Karzinomzelle, eine Leukämiezelle oder eine Myelomzelle ist.
- Verfahren nach Anspruch 44, wobei die Karzinomzelle eine Prostata- oder Brustkarzinomzelle ist.
- Verfahren nach Anspruch 43, wobei die Tumorzelle in einem lebenden Subjekt vorliegt.
- Verfahren nach Anspruch 46, wobei das lebende Subjekt ein Mensch ist.
- Peptidomimetikum, das die Struktur und MUC1-Bindungsfähigkeit eines MUC1-Peptids von wenigstens 4 aufeinander folgenden MUC1-Resten und nicht mehr als 20 aufeinander folgenden MUC1-Resten nachahmt, das die Sequenz CQC umfasst, wobei das aminoterminale Cystein von CQC an seinem NH2-Terminus von wenigstens einem Aminosäurerest abgedeckt ist, der nicht der nativen MUC1-Transmembransequenz entsprechen braucht.
- MUC1-Peptid von wenigstens 4 aufeinander folgenden MUC1-Resten und nicht mehr als 20 aufeinander folgenden MUC1-Resten, das die Sequenz CQC umfasst, wobei alle Aminosäurereste des Peptids D-Aminosäuren sind.
- Peptid nach Anspruch 49, das die Sequenz KRRCQC umfasst.
